Abstract:Salt stress is a major abiotic stress factor affecting crop production, and understanding of the response mechanisms of seed germination to salt stress can help to improve crop tolerance and yield. The differences in regulatory pathways during germination in different salt-tolerant barley seeds are not clear. Therefore, this study investigated the responses of different salt-tolerant barley seeds during germination to salt stress at the proteomic and metabolic levels. To do so, the proteomics and metabolomics of two barley seeds with different salt tolerances were comprehensively examined. Through comparative proteomic analysis, 778 differentially expressed proteins were identified, of which 335 were upregulated and 443 were downregulated. These proteins, were mainly involved in signal transduction, propanoate metabolism, phenylpropanoid biosynthesis, plant hormones and cell wall stress. In addition, a total of 187 salt-regulated metabolites were identified in this research, which were mainly related to ABC transporters, amino acid metabolism, carbohydrate metabolism and lipid metabolism; 72 were increased and 112 were decreased. Compared with salt-sensitive materials, salt-tolerant materials responded more positively to salt stress at the protein and metabolic levels. Taken together, these results suggest that salt-tolerant germplasm may enhance resilience by repairing intracellular structures, promoting lipid metabolism and increasing osmotic metabolites. These data not only provide new ideas for how seeds respond to salt stress but also provide new directions for studying the molecular mechanisms and the metabolic homeostasis of seeds in the early stages of germination under abiotic stresses.
Salt stress affects the biomass of industrial crop Jerusalem artichoke by affecting sugar transport and metabolism

Abstract:Even though Jerusalem artichoke (Helianthus tuberosus L.) has strong resistance to abiotic stresses, salinity can still reduce the biomass of Jerusalem artichoke. The purpose of this study was to elucidate the differences in the development of Jerusalem artichoke and the dynamics of sugar throughout the growth period under high (7.23-8.15 g/kg) and low (3.20-4.32 g/kg) salinity stress in the field in Jiangsu Province, China. This study confirmed that high salinity promoted the conversion of reducing sugars to non-reducing sugars (fructans) in Jerusalem artichoke tubers, but significantly reduced the biomass of Jerusalem artichoke and advanced the peak time of the dry matter accumulation of aerial parts. In addition, in the early and late stages of tuberization, the total sugar content of tubers under low salinity conditions (786 ± 8 mg/g and 491 ± 8 mg/g) was 93.3% and 1.15 times than those under high salinity conditions, respectively. Moreover, the total sugar content in stems was consistently greater under high than low salinity conditions in the same period. The accumulation rate and the amount of dry matter were significantly higher in stems than in other tissues. Therefore, the aerial parts of "Nanyu No. 1" could be harvested before mid-to-early October, and the tubers after mid-November. This study revealed the internal reasons for the decreased yield of Jerusalem artichoke under salt stress, and provided theoretical basis and guidance for the cultivation and utilization of Jerusalem artichoke in saline-alkali soil.

Abstract:Salinity is a major abiotic stress that affects plant growth and development. Plant roots are the sites of salt uptake. Here, an isobaric tag for a relative and absolute quantitation based proteomic technique was employed to identify the differentially expressed proteins (DEPs) from seedling roots of the salt-tolerant genotype Han 12 and the salt-sensitive genotype Jimai 19 in response to salt treatment. A total of 121 NaCl-responsive DEPs were observed in Han 12 and Jimai 19. The main DEPs were ubiquitination-related proteins, transcription factors, pathogen-related proteins, membrane intrinsic protein transporters and antioxidant enzymes, which may work together to obtain cellular homeostasis in roots and to determine the overall salt tolerance of different wheat varieties in response to salt stress. Functional analysis of three salt-responsive proteins was performed in transgenic plants as a case study to confirm the salt-related functions of the detected proteins. Taken together, the results of this study may be helpful in further elucidating salt tolerance mechanisms in wheat.

Abstract:Jerusalem artichoke ( Helianthus tuberosus L.), a vegetable with medical applications, has a strong adaptability to marginal barren land, but the suitability as planting material in saline land remains to be evaluated. This study was envisaged to examine salt tolerance in Jerusalem artichoke from the angle of photosynthetic apparatus stability by dissecting the photosynthetic electron transport process. Potted plants were exposed to salt stress by watering with a nutrient solution supplemented with NaCl. Photosystem I (PSI) and photosystem II (PSII) photoinhibition appeared under salt stress, according to the significant decrease in the maximal photochemical efficiency of PSI (△MR/MR 0 ) and PSII. Consistently, leaf hydrogen peroxide (H 2 O 2 ) concentration and lipid peroxidation were remarkably elevated after 8 days of salt stress, confirming salt-induced oxidative stress. Besides photoinhibition of the PSII reaction center, the PSII donor side was also impaired under salt stress, as a K step emerged in the prompt chlorophyll transient, but the PSII acceptor side was more vulnerable, considering the decreased probability of an electron movement beyond the primary quinone (ETo/TRo) upon depressed upstream electron donation. The declined performance of entire PSII components inhibited electron inflow to PSI, but severe PSI photoinhibition was not averted. Notably, PSI photoinhibition elevated the excitation pressure of PSII (1-qP) by inhibiting the PSII acceptor side due to the negative and positive correlation of △MR/MR 0 with 1-qP and ETo/TRo, respectively. Furthermore, excessive reduction of PSII acceptors side due to PSI photoinhibition was simulated by applying a specific inhibitor blocking electron transport beyond primary quinone, demonstrating that PSII photoinhibition was actually accelerated by PSI photoinhibition under salt stress. In conclusion, PSII and PSI vulnerabilities were proven in Jerusalem artichoke under salt stress, and PSII inactivation, which was a passive consequence of PSI photoinhibition, hardly helped protect PSI. As a salt-sensitive species, Jerusalem artichoke was recommended to be planted in non-saline marginal land or mild saline land with soil desalination measures.

Effect of soil salinity on fructan content and polymerization degree in the sprouting tubers of Jerusalem artichoke (Helianthus tuberosus L.)

Abstract:In addition to their role as reserve carbohydrates, fructans have been recognized as compounds that are protective against adverse environments. The aim of this study was to identify changes in the content and the degree of polymerization (DP) of fructan in sprouting tubers of Jerusalem artichoke under salt stress. Fructan was extracted from tubers at 1, 3, 5, and 7 days after planting in sandy loam soil irrigated with NaCl solution. Fructan accumulation and polymerization and the expression of genes encoding enzymes for fructan synthesis and degradation were evaluated. No significant differences between the control and treatment groups were observed until 5 days after sowing. The highest level of salinity (250 mM) not only inhibited sprouting and root growth but also decreased the level of fructan in the tubers. The proportion of fructan at DP 2-5 rapidly increased one day after sowing and then decreased over time. Under various NaCl treatments, at 7 days after sowing, all fructans except fructan at DP 6-10 were present in proportions less than or equal to the control. The variation in the DP of fructan was related to the transcription level of fructan metabolism genes. Fructan may support sprouting or resistance to salt stress by changing the DP of fructan molecules through hydrolysis without changing the total amount of fructan. The low-molecular-weight oligosaccharides (DP < 5) may be the major carbohydrates that support tuber sprouting or that are involved in protection from salt stress.

The following results were achieved: 1) Serious lesions were discovered in ileal mucous membrane. 2) Mucus IgA content was reduced markedly and the reduction was related to bacterial translocation.
Identification of Up-Regulated Genes Provides Integrated Insight into Salt-Induced Tolerance Mechanisms in Sesuvium Portulacastrum Roots

Abstract:Sesuvium portulacastrum, a promising facultative halophyte species, has a remarkable ability to survival at severe salt of ~1000 mM NaCl. However, genes encoding proteins involved in the process of salt tolerance in S. portulacastrum are limited. In this study, suppression subtractive hybridization together with reverse northern blot analysis and RT-PCR was used to identify up-regulated genes in roots after treatment with seawater for 2–24 h. We identified a total of 148 unigenes, and most of these genes were predominantly associated with ‘signal transduction and transcription’, ‘stress/defense response’, ‘transport’ and ‘metabolism’. Comparative analysis between physiological response and differentially expressed genes highlighted that sodium (Na+) transport, compartmentation, reactive oxygen species (ROS) scavenging, and sugar-metabolizing regulation play important roles in salt tolerance of S. portulacastrum, whereas inhibition of root elongation induced by high salinity may be related to lignin deposition and disruption of calcium homeostasis.

Abstract:The germination, growth, total phenolics, glucosinolate, myrosinase and antioxidant activity of radish sprouts germinated under 0 (control), 10, 50 and 100mM of NaCl were investigated. The glucoraphasatin (4-methylthio-3-butenyl-glucosinolate), total glucosinolate and total phenolic contents of 5- and 7-day-old sprouts treated with 10 and 50mM of NaCl were significantly decreased. However, the antioxidant activity of sprouts treated with 10 and 50mM of NaCl was not affected. The glucoraphasatin and total glucosinolate contents of 5- and 7-day-old sprouts, total phenolic contents of 3- and 5-day-old sprouts were significantly increased and myrosinase activities of 3- and 5-day-old sprouts were inhibited, although the germination was dramatically inhibited by 100mM of NaCl treatment. These results indicated that salt stress (100mM of NaCl treatment) could improve the nutritional value of radish sprouts, and germination of sprouts under adequate salt stress could be one useful way to enhance health-promoting compounds of plant food.

Abstract:Jerusalem artichokes are a perennial crop with high drought tolerance and high value as a raw material to produce biofuels, functional feed, and food. However, there are few comprehensive metabolomic studies on Jerusalem artichokes under drought conditions. Methods: Ultra-performance liquid chromatography and tandem mass spectrometry were used to identify differential metabolites in Jerusalem artichoke seedling leaves under polyethylene glycol (PEG) 6000-simulated drought stress at 0, 18, 24, and 36 h. Results: A total of 661 metabolites and 236 differential metabolites were identified at 0 vs. 18, 18 vs. 24, and 24 vs. 36 h. 146 differential metabolites and 56 common were identified and at 0 vs. 18, 24, and 36 h. Kyoto Encyclopedia of Genes and Genomes enrichment identified 236 differential metabolites involved in the biosynthesis of secondary metabolites and amino acids. Metabolites involved in glycolysis, phenolic metabolism, tricarboxylic cycle, glutamate-mediated proline biosynthesis, urea cycle, amino acid metabolism, unsaturated fatty acid biosynthesis, and the met salvage pathway responded to drought stress. Conclusion: A metabolic network in the leaves of Jerusalem artichokes under drought stress is proposed. These results will improve understanding of the metabolite response to drought stress in Jerusalem artichokes and develop a foundation for breeding drought-resistant varieties.

Abstract:Soil salinity is a very serious abiotic stressor that affects plant growth and threatens crop yield. Thus, it is important to explore the mechanisms of salt tolerance of plant and then to stabilize and improve crop yield. Asparagus is an important cash crop, but its salt tolerance mechanisms are largely unknown. Full-length transcriptomic and metabolomic analyses were performed on two asparagus genotypes: ‘jx1502’ (a salt-tolerant genotype) and ‘gold crown’ (a salt-sensitive genotype). Compared with the distilled water treatment (control), 877 and 1610 differentially expressed genes (DEGs) were identified in ‘jx1502’ and ‘gold crown’ under salt stress treatment, respectively, and 135 and 73 differentially accumulated metabolites (DAMs) were identified in ‘jx1502’ and ‘gold crown’ under salt stress treatment, respectively. DEGs related to ion transport, plant hormone response, and cell division and growth presented differential expression profiles between ‘jx1502’ and ‘gold crown.’ In ‘jx1502,’ 11 ion transport-related DEGs, 8 plant hormone response-related DEGs, and 12 cell division and growth-related DEGs were upregulated, while 7 ion transport-related DEGs, 4 plant hormone response-related DEGs, and 2 cell division and growth-related DEGs were downregulated. Interestingly, in ‘gold crown,’ 14 ion transport-related DEGs, 2 plant hormone response-related DEGs, and 6 cell division and growth-related DEGs were upregulated, while 45 ion transport-related DEGs, 13 plant hormone response-related DEGs, and 16 cell division and growth-related DEGs were downregulated. Genotype ‘jx1502’ can modulate K + /Na + and water homeostasis and maintain a more constant transport system for nutrient uptake and distribution than ‘gold crown’ under salt stress. Genotype ‘jx1502’ strengthened the response to auxin (IAA), as well as cell division and growth for root remodeling and thus salt tolerance. Therefore, the integration analysis of transcriptomic and metabolomic indicated that ‘jx1502’ enhanced sugar and amino acid metabolism for energy supply and osmotic regulatory substance accumulation to meet the demands of protective mechanisms against salt stress. This work contributed to reveal the underlying salt tolerance mechanism of asparagus at transcription and metabolism level and proposed new directions for asparagus variety improvement.

Abstract:Metabolic changes were investigated in overwintering Jerusalem artichoke (Helianthus tuberosus L.) tubers using proton nuclear magnetic resonance ((1)H NMR) metabolomics. Three varieties were studied; as a result of overwintering, the amount of inulin was found to decrease in Jerusalem artichoke tubers. This was mainly due to its conversion to sucrose and, at the same time, formation of inulin with a lower degree of polymerization. Major effects on the concentration of citric acid, malic acid, γ-aminobutyric acid (GABA), and adenosine were also found. Intriguingly, malic acid concentration increased and citric acid concentration decreased. These changes, together with an increase in sucrose and GABA concentrations, were ascribed to mobilization of nutrients prior to sprouting, suggesting that malic acid and GABA serve as carbon and nitrogen sources during sprouting of Jerusalem artichokes.
Effect of Salt Stress on Growth and Physiological Properties of Asparagus Seedlings

Abstract:Salt stress could inhibit the growth and development of crops and negatively affect yield and quality. The objective of this study was to investigate the physiological responses of different asparagus cultivars to salt stress. Twenty days old seedlings of salt-tolerant Apollo and salt-sensitive cultivar JL1 were subjected to 0 (CK) and120 mM NaCl stress for 20 d. Their changes in growth, ion contents, antioxidant enzyme activities and gene expression were analyzed. Salt stress significantly inhibited the growth of both cultivars, and JL1 showed a greater decrease than Apollo. The root development of Apollo was promoted by 120 mM NaCl treatment. The Na+ content in roots, stems, and leaves of both cultivars was increased under salt stress, while K+ content and K+/Na+ decreased. The salt-tolerant cultivar Apollo showed less extent of increase in Na+ and decrease in K+ content and kept a relatively high K+/Na+ ratio to compare with JL1. The contents of proline, soluble sugar and protein increased in Apollo, while these substances changed differently in JL1 under salt stress. Activities of superoxide dismutase (SOD), peroxidase (POD), and catalase (CAT) were gradually increased under salt stress in Apollo, while the corresponding enzyme activities in JL1 were decreased at the late stage of salt stress. The expression of SOD, POD, and Cat genes of both cultivars changed in a similar way to the enzyme activities. Malondialdehyde (MDA) content was increased slightly in Apollo, while increased significantly in JL1. At the late stage of salt stress, Apollo maintained a relatively high K+/Na+, osmotic adjustment ability and antioxidant defense capability, and therefore exhibited higher tolerance to salt stress than that of JL1.
In Situ Study of Metabolic Response of Arabidopsis thaliana Leaves to Salt Stress by Neutral Desorption-Extractive Electrospray Ionization Mass Spectrometry

Abstract:Salt stress is one of the most common factors limiting plant cultivation. In this study, metabolic responses to salt stress in Arabidopsis thaliana (A. thaliana) leaves were analyzed in situ by neutral desorption-extractive electrospray ionization mass spectrometry (ND-EESI-MS) without any sample pretreatment. Metabolic changes of A. thaliana leaves were observed in response to salt stress conditions, including the levels of serine, glutamic acid, arginine, cinnamic acid, ferulic acid, caffeic acid, protocatechuic acid, epicatechin, morin, myricetin, apigravin, and β-cotonefuran. The content of serine increased under 50, 100, and 200 mM NaCl salt stress, reaching the highest level at 200 mM NaCl, but decreased under the maximum concentration of 300 mM NaCl. A similar phenomenon was observed for arginine, glutamic acid, cinnamic acid, caffeic acid, ferulic acid, and epicatechin, respectively, involved in the metabolic pathway of shikimate-phenylpropanoid. Both principal component analysis (PCA) and partial least-squares discrimination analysis (PLS-DA) showed that the salt stress treatment groups of the higher concentrations (200 and 300 mM) could be well distinguished from those of the lower concentrations (50 and 100 mM) and the control. Marker metabolites, like m/z 261 (apigravin) and m/z 305 (β-cotonefuran), were assistantly selected from the fingerprints by variable importance for the projection (VIP). Our results indicated the potential of the ND-EESI-MS method for the rapid recognition of metabolic conditions in plant leaves under salt stress.
Ionomic Responses and Correlations Between Elements and Metabolites under Salt Stress in Wild and Cultivated Barley.

Abstract:A thorough understanding of ionic detoxification and homeostasis is imperative for improvement of salt tolerance in crops. However, the homeostasis of elements and their relationship to metabolites under salt stress have not been fully elucidated in plants. In this study, Tibetan wild barley accessions, XZ16 and XZ169, differing in salt tolerance, and a salt-tolerant cultivar CM72 were used to investigate ionomic profile changes in tissues in response to 150 and 300 mM NaCl at the germination and seedling stages. At the germination stage, the contents of Ca and Fe significantly decreased in roots, while K and S contents increased, and Ca and Mg contents decreased in shoots, after 10 d of treatment. At the seedling stage, the contents of K, Mg, P and Mn in roots and of K, Ca, Mg and S in shoots decreased significantly after 21 d of treatment. Moreover, Na had a significant negative correlation with metabolites involved in glycolysis, α-ketoglutaric acid, maleic acid and alanine in roots, and metabolites associated with the tricarboxylic acid (TCA) cycle, sucrose, polyols and aspartate in leaves. The salt-tolerant genotypes XZ16 and CM72 showed a lower Na content in tissues, and less reduction in Zn and Cu in roots, of Ca, Mg and S in leaves, and shoot DW than the sensitive genotype XZ169, when exposed to a higher salt level. The results indicated that restriction of Na accumulation and rearrangement of nutrient elements and metabolites in barley tissues are possibly attributable to development of salt tolerance.

Abstract:Salt stress is an important environmental limiting factor. Water dropwort ( Oenanthe javanica ) is an important vegetable in East Asia; however, its phenotypic and physiological response is poorly explored. For this purpose, 48 cultivars of water dropwort were grown hydroponically and treated with 0, 50, 100, and 200 mm NaCl for 14 days. Than their phenotypic responses were evaluated, afterward, physiological studies were carried out in selected sensitive and tolerant cultivars. In the present study, the potential tolerant (V11E0022) and sensitive (V11E0135) cultivars were selected by screening 48 cultivars based on their phenotype under four different levels of salt concentrations (0, 50, 100, and 200 mm). The results depicted that plant height, number of branches and leaves were less effected in V11E0022, and most severe reduction was observed in V11E0135 in comparison with others. Than the changes in biomass, ion contents, accumulation of reactive oxygen species, and activities of antioxidant enzymes and non-enzymatic antioxidants were determined in the leaves and roots of the selected cultivars. The potential tolerant cultivar (V11E0022) showed less reduction of water content and demonstrated low levels of Na + uptake, malondialdehyde, and hydrogen peroxide (H 2 O 2 ) in both leaves and roots. Moreover, the tolerant cultivar (V11E0022) showed high antioxidant activities of ascorbate peroxidase (APX), superoxide dismutase, peroxidase, catalase (CAT), reduced glutathione (GSH), and high accumulation of proline and soluble sugars compared to the sensitive cultivar (V11E0135). These results suggest the potential tolerance of V11E0022 cultivar against salt stress with low detrimental effects and a good antioxidant defense system. The observations also suggest good antioxidant capacity of water dropwort against salt stress. The findings of the present study also suggest that the number of branches and leaves, GSH, proline, soluble sugars, APX, and CAT could serve as the efficient markers for understanding the defense mechanisms of water dropwort under the conditions of salt stress.

Salt Stress Affects the Redox Status of Arabidopsis Root Meristems

Abstract:We report the redox status (profiles) for specific populations of cells that comprise the Arabidopsis root tip. For recently germinated, 3-5-day-old seedlings we show that the region of the root tip with the most reduced redox status includes the root cap initials, the quiescent center and the most distal portion of the proximal meristem, and coincides with (overlays) the region of the auxin maximum. As one moves basally, further into the proximal meristem, and depending on the growth conditions, the redox status becomes more oxidized, with a 5-10 mV difference in redox potential between the two borders delimiting the proximal meristem. At the point on the root axis at which cells of the proximal meristem cease division and enter the transition zone, the redox potential levels off, and remains more or less unchanged throughout the transition zone. As cells leave the transition zone and enter the zone of elongation the redox potentials become more oxidized. Treating roots with salt (50, 100, and 150 mM NaCl) results in marked changes in root meristem structure and development, and is preceded by changes in the redox profile, which flattens, and initially becomes more oxidized, with pronounced changes in the redox potentials of the root cap, the root cap initials and the quiescent center. Roots exposed to relatively mild levels of salt (<100 mM) are able to re-establish a normal, pre-salt treatment redox profile 3-6 days after exposure to salt. Coincident with the salt-associated changes in redox profiles are changes in the distribution of auxin transporters (AUX1, PIN1/2), which become more diffuse in their localization. We conclude that salt stress affects root meristem maintenance, in part, through changes in redox and auxin transport.

Abstract:Salt stress is a major abiotic stress with detrimental effects on plants. Sucrose, the main product of photosynthesis in plants, is used as a source of energy for the development of Chinese rose ( Rosa chinensis ). Hua Xianzi, a variety of Chinese rose, was investigated in this study. Phenotypic traits such as digital biomass, greenness average, leaf angle and leaf area, as well as physiological characters such as proline (Pro), malondialdehyde (MDA), H 2 O 2 , O 2 - and antioxidant enzymes such as SOD (Superoxide Dismutase), POD (Peroxidase), CAT (Catalase) and APX (Ascorbate Peroxidase), changed significantly in response to salt stress (150 mmol/L NaCl for 72 h). The physiological changes were examined based on photosynthetic and fluorescence indicators. Salt stress increased the concentration of glucose, fructose and sucrose, upregulated the expression of Rc-SS1 , Rc-SS2 , Rc-SPS1 , Rc-SPS2 , Rc-αA1 , Rc-αA2 , Rc-αA3 , Rc-βA1 and Rc-βA3 genes. Finally, it decreased the starch content in leaves. This study revealed that salt stress disrupted the equilibrium of sucrose metabolism in leaves. This finding provides the basis for further research into the mechanism of salt tolerance adaption.

Abstract:Background and aimsSoil alkalinity and salinity are two of the most common environmental stress factors that impact plant growth and productivity. S. corniculata is native to the saline-alkali soil of Northeast China which shows a higher alkali tolerance than other suaeda plants. It will be very important to identify the effect of salt and alkali stress on S. corniculata through proteomics and metabolomics analysis to improve understanding of the resistance of plants.MethodsS. corniculata seedlings were exposed to salt and alkali stress for 72 h, respectively. Metabolic changes were quantified by conducting MS-based proteomics and NMR-based metabolomics analysis.ResultsThe response of S. corniculata to salt stress was distinct from that of plants subjected to alkali stress at both physiological and molecular levels. The integrated-omic studies identified 22 and 19 differentially expressed proteins and metabolites mainly involved in carbohydrate metabolism and amino acid metabolism.ConclusionsOn account of the up-regulation of energy metabolism and higher accumulation of organic osmolytes, S. corniculata shows high pH resistance when it suffers alkali stress. These findings provide insights into the different regulatory mechanisms in halophyte S. corniculata response to salt and alkali stresses.

Abstract:Abstract Background Beta vulgaris L. is one of the main sugar-producing crop species and is highly adaptable to saline soil. This study explored the alterations to the carbon and nitrogen metabolism mechanisms enabling the roots of sugar beet seedlings to adapt to salinity. Results The ionome, metabolome, and transcriptome of the roots of sugar beet seedlings were evaluated after 1 day (short term) and 7 days (long term) of 300 mM Na + treatment. Salt stress caused reactive oxygen species (ROS) damage and ion toxicity in the roots. Interestingly, under salt stress, the increase in the Na + /K + ratio compared to the control ratio on day 7 was lower than that on day 1 in the roots. The transcriptomic results showed that a large number of differentially expressed genes (DEGs) were enriched in various metabolic pathways. A total of 1279 and 903 DEGs were identified on days 1 and 7, respectively, and were mapped mainly to 10 Kyoto Encyclopedia of Genes and Genomes (KEGG) pathways. Most of the genes were involved in carbon metabolism and amino acid (AA) biosynthesis. Furthermore, metabolomic analysis revealed that sucrose metabolism and the activity of the tricarboxylic acid (TCA) cycle increased in response to salt stress. After 1 day of stress, the content of sucrose decreased, whereas the content of organic acids (OAs) such as L-malic acid and 2-oxoglutaric acid increased. After 7 days of salt stress, nitrogen-containing metabolites such as AAs, betaine, melatonin, and (S)-2-aminobutyric acid increased significantly. In addition, multiomic analysis revealed that the expression of the gene encoding xanthine dehydrogenase (XDH) was upregulated and that the expression of the gene encoding allantoinase (ALN) was significantly downregulated, resulting in a large accumulation of allantoin. Correlation analysis revealed that most genes were significantly related to only allantoin and xanthosine. Conclusions Our study demonstrated that carbon and nitrogen metabolism was altered in the roots of sugar beet plants under salt stress. Nitrogen metabolism plays a major role in the late stages of salt stress. Allantoin, which is involved in the purine metabolic pathway, may be a key regulator of sugar beet salt tolerance.

Abstract:Utilizing salt-tolerant plants is a cost-effective strategy for agricultural production on salinized land. However, little is known about the mechanism of dandelion (Taraxacum mongolicum Hand.-Mazz.) in response to saline stress and caffeic acid biosynthesis. We investigated the morphological and physiological variations of two dandelions, namely, "BINPU2" (dandelion A) and "TANGHAI" (dandelion B) under gradient NaCl concentrations (0, 0.3%, 0.5%, 0.7%, and 0.9%), and analyzed potential mechanisms through a comparison analysis of transcriptomes in the two dandelions. Dandelion A had a high leaf weight; high ρ-coumaric acid, caffeic acid, ferulic acid, and caffeoyl shikimic acid contents; and high activities of POD and Pro. The maximum content of four kinds of phenolic acids mostly occurred in the 0.7% NaCl treatment. In this saline treatment, 2468 and 3238 differentially expressed genes (DEGs) in dandelion A and B were found, of which 1456 and 1369 DEGs in the two dandelions, respectively, showed up-regulation, indicating that more up-regulated DEGs in dandelion A may cause its high salt tolerance. Further, Gene Ontology (GO) analysis and Kyoto Encyclopedia of Genes and Genomes (KEGG) analysis showed that dandelion salt response and caffeic acid metabolism were mainly enriched in the phenylpropanoid biosynthesis pathway (ko00940) and response to ethylene (GO: 0009723). The caffeic acid biosynthesis pathway was reconstructed based on DEGs which were annotated to PAL, C4H, 4CL, HCT, C3′H, and CSE. Most of these genes showed a down-regulated mode, except for parts of DEGs of 4CL (TbA05G077650 and TbA07G073600), HCT (TbA03G009110, TbA03G009080, and novel.16880), and COMT (novel.13839). In addition, more up-regulated transcription factors (TFs) of ethylene TFs in dandelion A were found, but the TFs of ERF104, CEJ1, and ERF3 in the two dandelions under saline stress showed an opposite expression pattern. These up-regulated genes could enhance dandelion salt tolerance, and down-regulated DEGs in the caffeic acid biosynthesis pathway, especially CSE (TbA08G014310) and COMT (TbA04G07330), could be important candidate genes in the synthesis of caffeic acid under saline stress. The above findings revealed the potential mechanisms of salt response and caffeic acid metabolism in dandelion under saline stress, and provide references for salt-tolerant plant breeding and cultivation on saline–alkali land in the future.
