Abstract:Uroliths are uncommon findings in rats. This report describes a case of a neutered male rat with dysuria due to an infectious cystitis as well as an urolith of the urinary bladder consisting of struvite and whewellit. Following cystotomy as well as treatment with antibiotics and analgetics the rat recovered rapidly.

Abstract:Struvite urinary calculi, which are composed of magnesium, ammonium, and phosphate, can cause complications including sepsis and renal failure. Struvite calculi were identified within the urinary bladder and renal pelvis of 2 Long-Evans rats that died within days after arrival from a commercial vendor. The remaining rats in the shipment were screened by physical examination, radiography, and ultrasonography, revealing an additional 2 animals that were clinically affected. These rats were euthanized, necropsied, and yielded similar findings to those from the first 2 rats. In addition, urine samples had an alkaline pH and contained numerous bacteria (predominantly Proteus mirabilis), leukocytes, and crystals. All calculi were composed completely of struvite. Another 7 rats in the shipment had alkaline urine with the presence of blood cells; 6 of these rats also had abundant struvite crystals, and P. mirabilis was cultured from the urine of 3 rats. Further investigation by the vendor identified 2 of 100 rats with struvite calculi from the same colony. Although no specific cause could be implicated, the fact that all the affected rats came from the same breeding area suggests a genetic or environmental triggering event; a contribution due to diet cannot be ruled out. Our findings suggest that the affected rats had metabolic disturbances coupled with bacterial infection that predisposed them to develop struvite calculi. During sudden increases of struvite urinary calculi cases in rats, urine cultures followed by appropriate surgical intervention and antibiotic therapy is warranted. Additional factors, including diet, merit attention as well.
Rat strains differ in susceptibility to Ureaplasma parvum-induced urinary tract infection and struvite stone formation

Abstract:Individuals with struvite uroliths are susceptible to recurrent urinary tract infections (UTI), sepsis, and renal disease. Unfortunately, little is known about the host-specific factors that predispose to this disease. In order to develop a rodent model that can address this problem, we inoculated female Fischer 344 (F344), Lewis (LEW), Sprague-Dawley (SD), and Wistar (WIS) rats with a host-adapted strain of Ureaplasma parvum. Animals were necropsied at 2 weeks postinoculation; 100% of F344, 42% of SD, 10% of LEW, and 10% of WIS rats remained infected. Severe bladder lesions and struvite calculi were seen in 64% of F344 rats; in other rat strains, bladder lesions were mild or absent. F344 rats with struvite uroliths had the highest urinary levels of proinflammatory cytokines, such as GRO/KC, interleukin-1alpha (IL-1alpha), and IL-1beta. F344 rats without struvite stones at necropsy had milder bladder lesions and significantly lower urinary levels of proinflammatory cytokines but a more prominent inflammatory response than did other rat strains. Based on our results, struvite stone formation is linked to a robust inflammatory response that does not resolve UTI but instead promotes damage to surrounding tissues.
Urothelial hyperplasia with calculi (papillomatosis) in the urinary bladder of a male spontaneous diabetic Torii rat

Abstract:A 40-week-old male spontaneous diabetic Torii rat, an animal model of type 2 diabetes mellitus, was found to have marked urinary calculi with hematuria in the urinary bladder on necropsy. Histological findings in the urinary bladder included a papillary growth pattern with a fibrovascular stroma without atypia. Fine granular materials in the bladder lumen were positive for Von Kossa staining but negative for periodic acid-Schiff or Gram staining, indicating no apparent bacterial infection in the urinary bladder. Scanning electron microscopy revealed that the urinary calculi were magnesium ammonium phosphate crystals (struvite). On the basis of the results, the lesion was diagnosed as urothelial hyperplasia with calculi (papillomatosis). Chronic inciting stimuli by struvite crystals were considered the primary cause of the bladder findings.
Struvite urolithiasis in a B6C3F1 mouse

Abstract:In a 2 year carcinogenicity bioassay using B6C3F1 mice, one male mouse developed clinical signs near termination of the study, comprising skin sores around the prepuce, penile prolapse and urine scalding. The predominant finding at necropsy was a markedly distended urinary bladder filled with numerous crystallized particles. Microscopically, there was subacute cystitis with marked hyperplasia of the transitional epithelium. X-ray diffraction analysis of the crystals showed a diffraction pattern characteristic of struvite (ammonium magnesium phosphate). The implications of the spontaneous occurrence of bladder stones in rodents on long-term toxicology studies are discussed.
Morphological lesions of the rat urinary tract induced by inoculation of mycoplasmas and other urinary tract pathogens

Abstract:The effects on the urinary tract after inoculation of Ureaplasma urealyticum into the rat bladder were evaluated and compared to that seen after Mycoplasma hominis, Escherichia coli and Proteus mirabilis inoculation. The inoculation of the urease-producing organisms P. mirabilis and U. urealyticum were associated with the formation of struvite bladder stones and predominantly hyperplastic lesions of the bladder. The P. mirabilis inoculated rats also displayed marked pyelonephritis. A similar but much less pronounced reaction also occurred in the kidneys of some of the U. urealyticum inoculated rats. P. mirabilis could frequently be recultured. In contrast, this was not possible with U. urealyticum, but the organism was detected by scanning electron microscopy 2 weeks after the inoculation. Inoculation of M. hominis was associated with a few mild lesions of the bladder, but inflammatory lesions were not present in the kidneys. The study confirms the potential of Ureaplasma to form struvite stones in rat urinary tract. It also demonstrates that it can induce inflammatory changes in both bladder and kidney of rats without concomitant stone formation.
Reactions to vesical foreign bodies in two strains of rats

Abstract:Foreign bodies in the urinary tract induce uroliths. This study examined reactions to vesical foreign bodies in Brattleboro rats manifesting diabetes insipidus and Sprague-Dawley rats. A silk suture was placed in the bladder of these rats and the occurrence of vesical uroliths, stone composition, and mucosal morphology were examined. Sprague-Dawley rats readily formed bladder stones in addition to a urolith formed over the suture, but there was little evidence that Brattleboro rats developed similar stones. Stone composition was primarily ammonium magnesium phosphate. The mucosal reactive hyperplasia was pronounced in the Sprague-Dawley, but was negligible in the Brattleboro rats. In conclusion, vesical foreign bodies readily induced uroliths in Sprague-Dawley rats, but there was no similar evidence in Brattleboro rats. It is suggested that the excessive diuresis of the latter may play a major role for this resistance to form stones, but the precise mechanisms of it are complex and remain to be explored.
A Modified Rat Model for Infection-Induced Stone Formation

Abstract:OBJECTIVE:To employ a newly modified rat model for infection-induced bladder stone formation.METHODS:24 adult male Sprague Dawley rats were divided into 3 groups, model group (n=12), sham operation group (n=8) and control group (n=4). The surgical procedures were performed aseptically under anesthesia (25% Ultane 1.0 g/kg). The bladder in model group was exposed through a short lower midline abdominal incision, the puncture needle (G18) with guideline was inserted aseptically into bladder, a metal wire, which have been contaminated by the Proteus mirabilis, was put into the puncture canal, then implanted into the bladder lumen through the guideline. In the sham operated group the puncture needle (G18) with guideline was inserted into bladder without metal wire implanted into the bladder. There was no any operation in control group. The rats were sacrificed by excessive anesthesia at 21 days post challenge. The bladder were removed aseptically and inspected for evidence of urolithiasis.RESULTS:On Day 2 after surgery, two rats died in model group, no rats died in other groups. Twenty-one days after surgery, all of rats in model group developed various-sized bladder stones. There was no stone formation in sham operation group and control group. All stones were verified by infrared spectroscopy and optical crystallography. These stone were struvite stone.CONCLUSION:This model has a less trauma, faster recovery and excellent stone formation so that it may be used for the study of infection stone.
An ecological study of infected urinary stone genesis in an animal model

Abstract:Direct molecular and morphological techniques of modern microbiology were used to monitor the sequential development of bacterial microcolonies and biofilms in a rat model of urinary infection and to demonstrate that the urease activity of the infecting organisms sets in course a series of reactions in which struvite and apatite crystals develop within the matrix of the enlarging bacterial aggregate. This forms multiple stone nidi on the uroepithelial surface upon which succeeding bacterial biofilms develop and with the incorporation of other urine components, such as urinary mucroproteins, establishes a matrix skeleton that becomes mineralised, thus allowing for the growth of the stone in concentric layers. To arrive at this hypothesis, we studied infection stone genesis using a newly developed model for infection-induced bladder stone formation in the rat. We examined in detail the sequential events in the evolving microbial ecology of progressive struvite calculogenesis, using conventional microbiological techniques, direct ultrastructural observation, newly developed ultrastructural cytochemical localisation techniques and immunological procedures for stabilisation of the biofilm glycocalix and stone matrix in the rat model. It was concluded that the organic glycocalix material secreted by the associated bacteria comprises a substantial and aetiologically important part of the infection stone calculogenesis and matrix production.
Establishment of Rat Model of Ceftriaxone Associated Urolithiasis

Abstract:Objective To establish a rat model of ceftriaxone associated urolithiasis and investigate the possible mechanism of its formation.Methods Thirty-two male Wistar rats aged 8 weeks were randomly divided to four groups:group A served as control group;groups B,C and D received ceftriaxone sodium for 10 days with dosage of 100,150 and 200 mg/kg,respectively.The 24-h urine was gathered before intraperitoneal injection was conducted and before the end of experiment for the determination of oxalate and biochemical analysis.All rats were sacrificed with over-dose of carbon dioxide.The blood was collected for biochemical analysis.Kidneys were collected to observe the urinary stone formation and for Von Kossa staining.All the data were processed with the statistical software of SPSS 16.0.Results There were no visible calculi in these four groups,however,Von Kossa stains were positive in the three experimental groups,with the incidence of 50.0％ (4/8) in group B,62.5％ (5/8) in group C and 62.5％ (5/8) in group D.The incidence of massive calcium crystal deposits was 12.5％ (1/8) in group C and 12.5％ (1/8) in group D.No significant differences were observed in the incidences of calcium crystal deposits in groups with different dosages.Both the 24-h urinary excretion of calcium and uCa/Cr ratio in the 150 mg/kg group were increased markedly as compared with the control group.Conclusion Ceftriaxone could induce the formation of crystals in the kidney of rats.Ceftriaxone has the potential to significantly increase urinary excretion of calcium,which may be responsible for ceftriaxone associated urolithiasis.
Urolithiasis in dogs: Evaluation of trends in urolith composition and risk factors (2006-2018)

Abstract:Background: Urolithiasis is a common and often recurrent problem in dogs. Objective: To evaluate trends in urolith composition in dogs and to assess risk factors for urolithiasis, including age, breed, sex, neuter status, urolith location, and bacterial urolith cultures. Sample population: A total of 10 444 uroliths and the dogs from which they were obtained. Methods: The laboratory database at the UC Davis Gerald V. Ling Urinary Stone Analysis Laboratory was searched for all urolith submissions from dogs between January 2006 and December 2018. Mineral type, age, breed, sex, neuter status, urolith location, and urolith culture were recorded. Trends were evaluated and variables compared to evaluate risk factors. Results: Calcium oxalate (CaOx) and struvite-containing uroliths comprised the majority of all submissions from dogs, representing 47.0% and 43.6%, respectively. The proportion of CaOx-containing uroliths significantly decreased from 49.5% in 2006 to 41.8% in 2018 (P = .006), with no change in the proportion of struvite-containing urolith submissions. Cystine-containing uroliths comprised 2.7% of all submissions between 2006 and 2018 and a significant nonlinear increase in this mineral type occurred over time (1.4% of all submissions in 2006 to 8.7% in 2018; P < .001). Of all cystine-containing uroliths, 70.3% were from intact male dogs. Age, breed, and sex predispositions for uroliths were similar to those previously identified. Conclusions and clinical importance: Although calcium oxalate- and struvite-containing uroliths continue to be the most common uroliths submitted from dogs, a decrease in the proportion of CaOx-containing uroliths and an increase in the proportion of cystine-containing uroliths occurred during the time period evaluated.
Concrement formation in the urinary bladder in rats inoculated with Ureaplasma urealyticum

Abstract:To study the concrement-forming ability of Ureaplasma urealyticum in the urinary tract, viable and heat-killed ureaplasmas as well as urease and non-urease-producing bacteria were inoculated into the bladder in rats. Viable ureaplasmas, in contrast to heat-killed, caused the formation of bladder stones with a frequency corresponding to urease-producing bacteria (Proteus mirabilis). It was not possible to reculture the inoculated ureaplasmas from the urinary tract. Non-urease producing microorganisms (Escherichia coli and Mycoplasma hominis) only occasionally induced stone formation. The results indicate that U. urealyticum can initiate stone formation, a property that appears to be associated with the urease activity of the organism.
Canine and feline urolithiasis: examination of over 50 000 urolith submissions to the Canadian veterinary urolith centre from 1998 to 2008

Abstract:This study reports on trends in canine and feline urolithiasis in Canada during the past 10 years. Age, sex, breed of animals and mineral composition from 40 637 canine and 11 353 feline bladder uroliths submitted to the Canadian Veterinary Urolith Centre between 1998 and 2008 were recorded. Struvite and calcium oxalate uroliths comprised > 85% of all uroliths submitted. In dogs, the number of struvite submissions has declined and the number of calcium oxalate submissions has increased. Struvite uroliths were most common in female dogs and calcium oxalate uroliths in male dogs. The shih tzu, miniature schnauzer, bichon frisé, lhasa apso, and Yorkshire terrier were the breeds most commonly affected for both struvite and calcium oxalate uroliths. Urate uroliths were most common in male dalmatians. In cats, struvite submissions declined and calcium oxalate submissions remained constant. Struvite and calcium oxalate uroliths were common in domestic, Himalayan, Persian, and Siamese cats. Urate uroliths were over-represented in Egyptian maus.

Abstract:This study aimed to investigate the clinical, laboratory, radiography and ultrasonography findings for diagnosis of urolithiasis in cats and dogs. This study was consisted of totally 17 animals, of those, 7 cats and 11 dogs were different ages, breeds and sex. Urolithiasis diagnosis was made through urine analysis, direct and indirect radiography and ultrasonographic findings. Radiopaque stones were detected in the bladder (n=13), only in the urethra (n=2), and both in the urethra and bladder (n=2). Urethrotomy was performed in two animals with urolith in the urethra, and cystotomy was performed in two cases by pushing the stones in the urethra into the urinary bladder with uroretropropulsion. In other cases, uroliths were removed from the bladder by cystotomy. The uroliths were analyzed by biochemical method. The results of the present study revealed that clinical, hematological, biochemical, urinalysis, radiographic, and ultrasonographic examinations are essential in dogs and cats for the accurate diagnosis of urolithiasis.
A novel technique for atraumatic transurethral catheterisation of male rats

Abstract:Transurethral catheterisation of male rats is technically difficult owing to anatomical peculiarities. In the male rat, the urethral striated sphincter consists of two lateral fascicles separated by an anterior and a posterior strip of connective tissue, which impedes the smooth insertion of a urinary catheter. For rat studies requiring continuous collection of urine, bladder irrigation, or measurement of bladder pressure, investigators either have to exclude the male population (be limited to the female population) or perform percutaneous (suprapubic) bladder puncture in male rats, which is more traumatic and invasive than transurethral catheterisation. This paper describes a novel, atraumatic method of transurethral catheterisation in the male rat, with the aid of a microscope and microsurgical instruments. Six Wistar rats were used for this experiment, all of which were catheterised successfully, with no evidence of bladder or urethral injury. The study shows that male rats can be safely catheterised via the urethra with the aid of a microscope and microsurgical instruments for both visual and tactile feedback. This is a relatively straightforward technique to learn and can allow for inclusion of male rats in future studies requiring urinary analysis or bladder irrigation, without the need for traumatic percutaneous (suprapubic) bladder puncture.

Abstract:In this study, the presence and types of urine crystals in cats and dogs with urolithiasis/crystaluria and the effects of some demographic factors (such as type, breed, age, sex, sterilization status) on them were evaluated. In the study, physical, chemical and sediment examinations of urine samples of 67 cats and dogs were performed. It was determined that 83.58% of the animals with crystals in their urine were male. It was determined that 63.64% of the female animals and 44.64% of the male animals with crystals detected in the urinary tract were sterilized. It was determined that urinary crystals were more common in cats between 1-3 years of age (AO) and in dogs older than 3 AO. It was determined that calcium oxalate crystal (64.41%) in cats and struvite crystal (50%) in dogs were the most common crystals. It was determined that urine crystals were found mostly in cross breeds in cats and dogs. It was determined that most of the cats and dogs with urine crystals were fed with home meals and non-branded foods. It was determined that urinary tract infections had an effect on the formation of urine crystals. As a result, it was concluded that it is important in terms of Veterinary Medicine that such studies should be routinely performed, especially in the United States of North America (such as the Minnesota Urolith Center), since urine crystals cause significant lower urinary system diseases.

Marked struvite crystalluria and its association with lower urinary tract signs in a cat with feline idiopathic cystitis

Abstract:Case report: We describe a case of a large amount of mineralised material, presumed to be struvite crystals, within the urinary bladder of a cat with feline idiopathic cystitis. The presence of this material coincided with episodes of lower urinary tract signs in this cat over a 2-year period. Clinical significance: Although struvite crystalluria is widely considered to be clinically insignificant, this generalisation may not be true for all cats with lower urinary tract disease. Imaging of the urinary tract is recommended in all cats with lower urinary tract signs.
Lower urinary tract lithiasis of cats in Algeria: Clinical and epidemiologic features

Abstract:Aim: This study aims to describe the clinical symptoms, diagnosis, and treatment of urolithiasis of the lower urinary tract and to determine the main risk factors involved in the occurrence of urinary lithiasis in cats in Algeria from 2016 to 2018. Materials and methods: During the study period, 465 cats were examined and 32 cases of urolithiases were selected and investigated by conducting physical examinations, blood analyses, urinalysis, X-ray radiography, and ultrasonography. Parameters such as breed, age, or sex were studied and reported in a farm return to analyze risk factors involved in the formation of lower urinary urolithiasis. Results: The most clinically relevant symptoms of urolithiasis observed in cats were dysuria, pollakiuria, hematuria, and stranguria. Urinalysis and blood analysis revealed a significant presence of urinary crystals and acute kidney failure in nine cats. The ultrasonography and radiography confirmed the diagnosis of urolithiasis with the incidence of 43.75% and 31.25%, respectively. The lower urinary tract urolithiasis appeared to be more frequent in European and Siamese cats. In addition, cats aged between 4 and 8 years old were the most affected. Male cats (87.50%) were more affected than female cats. Finally, the lower urinary tract urolithiasis was more frequent in cats consuming the commercial pet food, previously castrated, and confined inside the house. Conclusion: Complete clinical assessments, in addition to complementary examinations, are necessary and beneficial in treating the animal and preventing possible complications. Whether the choice of therapy is surgical or treatment with drugs, it is crucial to understand that the elimination of the stone is not an end, but the beginning of a series of investigations. Because of their impact on both the formation and elimination of metabolites, it has been found that factors, such as race, gender, age, diet, and lifestyle, should be considered as potential risk factors for urolithiasis.
Staphylococcus xylosus Cystitis and Struvite Urolithiasis in Nude Mice Implanted with Sustained-release Estrogen Pellets

Abstract:Female nude mice (J:NU-Foxn1nu; age, 6 wk) were injected with 1 million MCF7 human breast cancer cells in the fourth mammary fat pads and received a 21-d sustained-release estrogen pellet (0.25 mg) subcutaneously in the dorsum of the neck. All mice were maintained in sterile housing and provided sterile water and irradiated rodent chow. Approximately 6 wk after implantation, 4 of the 30 mice showed clinical signs of depression and dehydration. The 2 animals most severely affected were euthanized and presented for necropsy. The urinary bladders of these animals were distended with variable sized white, opaque uroliths. Urinalysis revealed coccal bacteria, erythrocytes, neutrophils and struvite crystals. Urine cultures from both necropsied animals grew heavy, pure growths of Staphylococcus xylosus. The organism was sensitive to all antibiotics tested except erythromycin (intermediate). Analysis of the uroliths revealed 100% struvite composition. Remaining mice in the study were evaluated clinically for hydration status, the ability to urinate, and the presence of palpable stones in the urinary bladder; one additional mouse had a firm, nonpainful bladder (urolithiasis suspected). Given the sensitivity of the organisms cultured from urine samples, the remaining mice were placed on enrofloxacin in the drinking water (0.5 mg/mL). All remaining mice completed the study without further morbidity or mortality. Previous studies have reported the association of estrogen supplementation with urinary bladder pathology, including infection and urolithiasis. Here we present a case of urolithiasis and cystitis in nude mice receiving estrogen supplementation that was associated with Staphylococcus xylosus, which previously was unreported in this context. When assessing these nude mice for urolithiasis, we found that visualizing the stones through the body wall, bladder palpation, and bladder expression were helpful in identifying affected mice.
Ectopic Bone as a Nidus for Calcium Oxalate Urocystolithiasis in a Cat

Abstract:A 7-year-old female spayed domestic shorthair cat was referred to the urology service for nonsurgical urocystolith removal. A urolith was attached to the urothelium by ectopic bone. Interventional removal without surgery was successful. Follow-up evaluation 3 years after urolith removal revealed recurrent uroliths, bladder wall mineralization, and marked renal mineralization. This case illustrates the metaplastic potential of the urothelium and that ectopic bone should be included among the pathological factors that promote lithogenesis.
[Establishment Nephrolithiasis Rat Model Induced by Nanobacteria and Analysis of Stone Formation].

Abstract:OBJECTIVE:To establish nephrolithiasis rat model induced by nanobacteria and to determine the relationship of nanobacteria with nephrolithiasis.METHODS:Thirty adult male SD rats were randomized into 3 groups (n=10 each): group A, given an intravenous injection of nanobacteria; group B, given an intravenous injection of nanobacteria and taken tetracycline; group C, as normal control. Eight weeks later, all rats were sacrificed and the kidneys were examined for pathology and the 24-h urine N-acetyl-beta-D-aminoglycoside enzyme (NAG) and lactate dehydrogenase (LDH) were detected.RESULTS:Histopathological studies revealed nanobacteria induced renal tubular crystallization, which was significantly different among the 3 groups (P=0.033). The crystallization was higher in group A than that in group B. The urinary NAG and LDH excretion were significantly different among the 3 groups (P<0.001).CONCLUSION:Nanobacteria may be involved in the pathogenesis of nephrolithiasis. Tetracycline may inhibit stone formation.
