Abstract:The isotopic composition of nitrous oxide (N2O) provides useful information for evaluating N2O sources and budgets. Due to the co-occurrence of multiple N2O transformation pathways, it is, however, challenging to use isotopic information to quantify the contribution of distinct processes across variable spatiotemporal scales. Here, we present an overview of recent progress in N2O isotopic studies and provide suggestions for future research, mainly focusing on: analytical techniques; production and consumption processes; and interpretation and modelling approaches. Comparing isotope-ratio mass spectrometry (IRMS) with laser absorption spectroscopy (LAS), we conclude that IRMS is a precise technique for laboratory analysis of N2O isotopes, while LAS is more suitable forin situ/inline studies and offers advantages for site-specific analyses. When reviewing the link between the N2O isotopic composition and underlying mechanisms/processes, we find that, at the molecular scale, the specific enzymes and mechanisms involved determine isotopic fractionation effects. In contrast, at plot-to-global scales, mixing of N2O derived from different processes and their isotopic variability must be considered. We also find that dual isotope plots are effective for semi-quantitative attribution of co-occurring N2O production and reduction processes. More recently, process-based N2O isotopic models have been developed for natural abundance and(15)N-tracing studies, and have been shown to be effective, particularly for data with adequate temporal resolution. Despite the significant progress made over the last decade, there is still great need and potential for future work, including development of analytical techniques, reference materials and inter-laboratory comparisons, further exploration of N2O formation and destruction mechanisms, more observations across scales, and design and validation of interpretation and modelling approaches. Synthesizing all these efforts, we are confident that the N2O isotope community will continue to advance our understanding of N2O transformation processes in all spheres of the Earth, and in turn to gain improved constraints on regional and global budgets.

Abstract:Measurements of the four most abundant stable isotopocules of N2O (14N14N16O, 15N14N16O, 14N15N16O, and 14N14N18O) can provide a valuable constraint on source attribution of atmospheric N2O. N2O isotopocules at natural abundance levels can be analyzed by isotope-ratio mass-spectrometry (IRMS) [1] and more recently optical isotope ratio spectroscopy (OIRS) [2]. OIRS instruments can analyze the N2O isotopic composition in gaseous mixtures in a continuous-flow mode, providing real-time data with minimal or no sample pretreatment, which is highly attractive to better resolve the temporal complexity of N2O production and consumption processes. Most importantly, OIRS laser spectroscopy is selective for position-specific 15N substitution due to the existence of characteristic rotational-vibrational spectra.By allowing both in-situ application and measurements in high temporal resolution, laser spectroscopy has established a new quality of data for research on N2O in particular and N cycling in general. However, applications remain challenging and are still scarce as a metrological characterization of OIRS analyzers, reporting factors limiting their performance is still missing. In addition, only since recently two pure N2O isotopocule reference materials have been made available through the United States Geological Survey (USGS), which however, only offer a small range of δ15N and δ18O values (< 1 ‰) and are therefore not suited for a two-point calibration approach [3].This presentation will highlight the recent progress achieved within the framework of the EMPIR project “Metrology for Stable Isotope Reference Standards (SIRS)”, namely:(1) The development of pure and diluted N2O reference materials (RMs), covering the range of isotope values required by the scientific community. These gaseous standards are available as pure N2O or N2O diluted in whole air. N2O RMs were analyzed by an international group of laboratories for δ15N, δ18O (MPI-BGC, Tokyo Institute of Technology, UEA), δ15Nα, δ15Nß (Empa, Tokyo Institute of Technology) and δ17O (UEA) traceable to the existing isotope ratio scales. (2) The metrological characterization of the three most common commercial N2O isotope OIRS analyzers (with/without precon QCLAS, OA-ICOS and CRDS) for gas matrix effects, spectral interferences of enhanced trace gas concentrations (CO2, CH4, CO, H2O), short-term and long-term repeatability, drift and dependence of isotope deltas on N2O concentrations [4]. In summary, the authors suggest to include appropriate RMs following the identical treatment (IT) principle during every OIRS measurement to retrieve compatible and accurate results. Abstract:Abstract. For the past two decades, the measurement of nitrous oxide (N2O) isotopocules – isotopically substituted molecules 14N15N16O, 15N14N16O and 14N14N18O of the main isotopic species 14N14N16O – has been a promising technique for understanding N2O production and consumption pathways. The coupling of non-cryogenic and tuneable light sources with different detection schemes, such as direct absorption quantum cascade laser absorption spectroscopy (QCLAS), cavity ring-down spectroscopy (CRDS) and off-axis integrated cavity output spectroscopy (OA-ICOS), has enabled the production of commercially available and field-deployable N2O isotopic analyzers. In contrast to traditional isotope-ratio mass spectrometry (IRMS), these instruments are inherently selective for position-specific 15N substitution and provide real-time data, with minimal or no sample pretreatment, which is highly attractive for process studies. Here, we compared the performance of N2O isotope laser spectrometers with the three most common detection schemes: OA-ICOS (N2OIA-30e-EP, ABB – Los Gatos Research Inc.), CRDS (G5131-i, Picarro Inc.) and QCLAS (dual QCLAS and preconcentration, trace gas extractor (TREX)-mini QCLAS, Aerodyne Research Inc.). For each instrument, the precision, drift and repeatability of N2O mole fraction [N2O] and isotope data were tested. The analyzers were then characterized for their dependence on [N2O], gas matrix composition (O2, Ar) and spectral interferences caused by H2O, CO2, CH4 and CO to develop analyzer-specific correction functions. Subsequently, a simulated two-end-member mixing experiment was used to compare the accuracy and repeatability of corrected and calibrated isotope measurements that could be acquired using the different laser spectrometers. Our results show that N2O isotope laser spectrometer performance is governed by an interplay between instrumental precision, drift, matrix effects and spectral interferences. To retrieve compatible and accurate results, it is necessary to include appropriate reference materials following the identical treatment (IT) principle during every measurement. Remaining differences between sample and reference gas compositions have to be corrected by applying analyzer-specific correction algorithms. These matrix and trace gas correction equations vary considerably according to N2O mole fraction, complicating the procedure further. Thus, researchers should strive to minimize differences in composition between sample and reference gases. In closing, we provide a calibration workflow to guide researchers in the operation of N2O isotope laser spectrometers in order to acquire accurate N2O isotope analyses. We anticipate that this workflow will assist in applications where matrix and trace gas compositions vary considerably (e.g., laboratory incubations, N2O liberated from wastewater or groundwater), as well as extend to future analyzer models and instruments focusing on isotopic species of other molecules.

Abstract:Xestoquinone (XQN) (3 x 10(-7) to 3 x 10(-3) M), isolated from the sea sponge Xestospongia sapra, induced a concentration-dependent Ca(2+) release from the heavy fraction of fragmented sarcoplasmic reticulum (HSR) of rabbit skeletal muscle with an EC(50) value of approximately 30 microM. On the basis of the EC(50), XQN is 10 times more potent than caffeine. Dithiothreitol completely blocked XQN-induced Ca(2+) release from HSR without affecting that induced by caffeine. Caffeine-induced Ca(2+) release was reduced markedly by Mg(2+), procaine, and ruthenium red, agents that are known to block release of Ca(2+) from sarcoplasmic reticulum, whereas that induced by XQN was not inhibited. The bell-shaped profile of Ca(2+) dependence for XQN was significantly shifted upward in a wider range of pCa (between 7 and 3), whereas that for caffeine was shifted to the left in a narrower range of pCa (between 8 and 7). The maximum response to caffeine in (45)Ca(2+) release was not affected by 9-methyl-7-bromoeudistomin D, whereas the response was further increased by XQN. XQN caused a concentration-dependent decrease in [(3)H]ryanodine binding to HSR. This effect of XQN also was abolished in the presence of dithiothreitol. Scatchard analysis revealed that the mode of inhibition by XQN was noncompetitive in [(3)H]ryanodine binding to HSR. These results indicate that sulfhydryl groups are involved in both the XQN effect on ryanodine binding and on Ca(2+) release.

Abstract:RationaleInformation on the isotopic composition of nitrous oxide (N2O) at natural abundance supports the identification of its source and sink processes. In recent years, a number of mass spectrometric and laser spectroscopic techniques have been developed and are increasingly used by the research community. Advances in this active research area, however, critically depend on the availability of suitable N2O isotope Reference Materials (RMs).MethodsWithin the project Metrology for Stable Isotope Reference Standards (SIRS), seven pure N2O isotope RMs have been developed and their 15N/14N, 18O/16O, 17O/16O ratios and 15N site preference (SP) have been analysed by specialised laboratories against isotope reference materials. A particular focus was on the 15N site‐specific isotopic composition, as this measurand is both highly diagnostic for source appointment and challenging to analyse and link to existing scales.ResultsThe established N2O isotope RMs offer a wide spread in delta (δ) values: δ15N: 0 to +104‰, δ18O: +39 to +155‰, and δ15NSP: −4 to +20‰. Conversion and uncertainty propagation of δ15N and δ18O to the Air‐N2 and VSMOW scales, respectively, provides robust estimates for δ15N(N2O) and δ18O(N2O), with overall uncertainties of about 0.05‰ and 0.15‰, respectively. For δ15NSP, an offset of >1.5‰ compared with earlier calibration approaches was detected, which should be revisited in the future.ConclusionsA set of seven N2O isotope RMs anchored to the international isotope‐ratio scales was developed that will promote the implementation of the recommended two‐point calibration approach. Particularly, the availability of δ17O data for N2O RMs is expected to improve data quality/correction algorithms with respect to δ15NSP and δ15N analysis by mass spectrometry. We anticipate that the N2O isotope RMs will enhance compatibility between laboratories and accelerate research progress in this emerging field.
Isotopomer labeling and oxygen dependence of hybrid nitrous oxide production

Abstract:Nitrous oxide (N 2 O) is a potent greenhouse gas and ozone depletion agent, with a significant natural source from marine oxygen-deficient zones (ODZs). Open questions remain, however, about the microbial processes responsible for this N 2 O production, especially hybrid N 2 O production when ammonia-oxidizing archaea are present. Using 15 N-labeled tracer incubations, we measured the rates of N 2 O production from ammonium (NH 4+ ), nitrite (NO 2- ), and nitrate (NO 3- ) in the eastern tropical North Pacific ODZ and the isotopic labeling of the central ( α ) and terminal ( β ) nitrogen (N) atoms of the N 2 O molecule. We observed production of both doubly and singly labeled N 2 O from each tracer, with the highest rates of labeled N 2 O production at the same depths as the near-surface N 2 O concentration maximum. At most stations and depths, the production of 45 N 2 O α and 45 N 2 O β were statistically indistinguishable, but at a few depths there were significant differences in the labeling of the two nitrogen atoms in the N 2 O molecule. Implementing the rates of labeled N 2 O production in a time-dependent numerical model, we found that N 2 O production from NO 3- dominated at most stations and depths, with rates as high as 1600 ± 200 pM N 2 O d −1 . Hybrid N 2 O production, one of the mechanisms by which ammonia-oxidizing archaea produce N 2 O, had rates as high as 230 ± 80 pM N 2 O d −1 that peaked in both the near-surface and deep N 2 O concentration maxima. Based on the equal production of 45 N 2 O α and 45 N 2 O β in the majority of our experiments, we infer that hybrid N 2 O production likely has a consistent site preference, despite drawing from two distinct substrate pools. We also found that the rates and yields of hybrid N 2 O production were enhanced at low dissolved oxygen concentrations ([O 2 ]), with hybrid N 2 O yields as high as 20 % at depths where [O 2 ] was below detection (880 nM) but nitrification was still active. Finally, we identified a few incubations with [O 2 ] up to 20 μ M where N 2 O production from NO 3- was still active. A relatively high O 2 tolerance for N 2 O production via denitrification has implications for the feedbacks between marine deoxygenation and greenhouse gas cycling.

Abstract:Abstract. Nitrous oxide (N 2 O) is the primary atmospheric constituent involved in stratospheric ozone depletion and contributes strongly to changes in the climate system through a positive radiative forcing mechanism. The atmospheric abundance of N 2 O has increased from 270 ppb during the pre-industrial era to approx. 330 ppb in 2018. Even though it is well known that microbial processes in agricultural and natural soils are the major N 2 O source, the contribution of specific soil processes is still uncertain. The relative abundance of N 2 O isotopocules ( 14 N 14 N 16 N, 14 N 15 N 16 O, 15 N 14 N 16 O and 14 N 14 N 18 O) carries process-specific in-formation and thus can be used to trace production and consumption pathways. While isotope ratio mass spectroscopy (IRMS) was traditionally used for high-precision measurement of the isotopic composition of N 2 O, quantum cascade laser absorption spectroscopy (QCLAS) has been put forward as a complementary technique with the potential for on-site analysis. In recent years, preconcentration combined with QCLAS has been presented as a technique to resolve subtle changes in ambient N 2 O isotopic composition. From the end of May until the beginning of August 2016, we investigated N 2 O emissions from an intensively managed grassland at the study site Fendt in Southern Germany. In total, 612 measurements of ambient N 2 O were taken by combining preconcentration with QCLAS analyses, yielding δ 15 N α , δ 15 N β , δ 18 O and N 2 O concentration with a temporal resolution of approximately one hour and precisions of 0.46 ‰, 0.36 ‰, 0.59 ‰ and 1.24 ppb, respectively. Soil δ 15 N-NO 3 − values and concentrations of NO 3 − and NH 4 + were measured to further constrain possible N 2 O-emitting source processes. Furthermore, the concentration footprint area of measured N 2 O was determined with a Lagrangian particle dispersion model (FLEXPART-COSMO) using local wind and turbulence observations. These simulations indicated that night-time concentration observations were largely sensitive to local fluxes. While bacterial denitrification and nitrifier denitrification were identified as the primary N 2 O-emitting processes, N 2 O reduction to N 2 largely dictated the isotopic composition of measured N 2 O. Fungal denitrification and nitrification-derived N 2 O accounted for 34–42 % of total N 2 O emissions and had a clear effect on the measured isotopic source signatures. This study presents the suitability of on-site N 2 O isotopocule analysis for disentangling source and sink processes in-situ and found that at the Fendt site bacterial denitrification/nitrifier denitrification is the major source for N 2 O, while N 2 O reduction acted as a major sink.
Tracing N2O formation in full-scale wastewater treatment with natural abundance isotopes indicates control by organic substrate and process settings

Abstract:Nitrous oxide (N2O) dominates greenhouse gas emissions in wastewater treatment plants (WWTPs). Formation of N2O occurs during biological nitrogen removal, involves multiple microbial pathways, and is typically very dynamic. Consequently, N2O mitigation strategies require an improved understanding of nitrogen transformation pathways and their modulating controls. Analyses of the nitrogen (N) and oxygen (O) isotopic composition of N2O and its substrates at natural abundance have been shown to provide valuable information on formation and reduction pathways in laboratory settings, but have rarely been applied to full-scale WWTPs. Here we show that N-species isotope ratio measurements at natural abundance level, combined with long-term N2O monitoring, allow identification of the N2O production pathways in a full-scale plug-flow WWTP (Hofen, Switzerland). Heterotrophic denitrification appears as the main N2O production pathway under all tested process conditions (0-2 mgO2/l, high and low loading conditions), while nitrifier denitrification was less important, and more variable. N2O production by hydroxylamine oxidation was not observed. Fractional N2O elimination by reduction to dinitrogen (N2) during anoxic conditions was clearly indicated by a concomitant increase in site preference, δ18O(N2O) and δ15N(N2O). N2O reduction increased with decreasing availability of dissolved inorganic N and organic substrates, which represents the link between diurnal N2O emission dynamics and organic substrate fluctuations. Consequently, dosing ammonium-rich reject water under low-organic-substrate conditions is unfavorable, as it is very likely to cause high net N2O emissions. Our results demonstrate that monitoring of the N2O isotopic composition holds a high potential to disentangle N2O formation mechanisms in engineered systems, such as full-scale WWTP. Our study serves as a starting point for advanced campaigns in the future combining isotopic technologies in WWTP with complementary approaches, such as mathematical modeling of N2O formation or microbial assays to develop efficient N2O mitigation strategies.
Marine N2O cycling from high spatial resolution concentration, stable isotopic and isotopomer measurements along a meridional transect in the eastern Pacific Ocean

Abstract:Nitrous oxide (N 2 O) is a potent greenhouse gas and ozone depleting substance, with the ocean accounting for about one third of global emissions. In marine environments, a significant amount of N 2 O is produced by biological processes in Oxygen Deficient Zones (ODZs). While recent technological advances are making surface N 2 O concentration more available, high temporal and spatial resolution water-column N 2 O concentration data are relatively scarce, limiting global N 2 O ocean models' predictive capability. We present a N 2 O concentration, stable isotopic composition and isotopomer dataset of unprecedently large spatial coverage and depth resolution in the broader Pacific, crossing both the eastern tropical South and North Pacific Ocean ODZs collected as part of the GO-SHIP P18 repeat hydrography program in 2016/2017. We complement these data with dissolved gases (nitrogen, oxygen, argon) and nitrate isotope data to investigate the pathways controlling N 2 O production in relation to apparent oxygen utilization and fixed nitrogen loss. N 2 O yield significantly increased under low oxygen conditions near the ODZs. Keeling plot analysis revealed different N 2 O sources above the ODZs under different oxygen regimes. Our stable isotopic data and relationships between the N 2 O added by microbial processes (ΔN 2 O) and dissolved inorganic nitrogen (DIN) deficit confirm increased N 2 O production by denitrification under low oxygen conditions near the oxycline where the largest N 2 O accumulations were observed. The slope for δ 18 O-N 2 O versus site preference (SP, the difference between the central (α) and outer (β) N atoms in the linear N 2 O molecule) in the eastern tropical North Pacific ODZ was lower than expected for pure N 2 O reduction, likely because of the observed decrease in δ 15 N β . This trend is consistent with prior ODZ studies and attributed to concurrent production of N 2 O from nitrite with a low δ 15 N or denitrification with a SP >0‰. We estimated apparent isotope effects for N 2 O consumption in the ETNP ODZ of 3.6‰ for 15 N bulk , 9.4‰ for 15 N α , -2.3‰ for 15 N β , 12.0‰ for 18 O, and 11.7‰ for SP. These values were generally within ranges previously reported for previous laboratory and field experiments.

Abstract:We present measurements of site preference (SP) and bulk (15)N/(14)N ratios (δ(15)N(bulk)(N2O)) of nitrous oxide (N(2)O) by quantum cascade laser absorption spectroscopy (QCLAS) as a powerful tool to investigate N(2)O production pathways in biological wastewater treatment. QCLAS enables high-precision N(2)O isotopomer analysis in real time. This allowed us to trace short-term fluctuations in SP and δ(15)N(bulk)(N2O) and, hence, microbial transformation pathways during individual batch experiments with activated sludge from a pilot-scale facility treating municipal wastewater. On the basis of previous work with microbial pure cultures, we demonstrate that N(2)O emitted during ammonia (NH(4)(+)) oxidation with a SP of -5.8 to 5.6 ‰ derives mostly from nitrite (NO(2)(-)) reduction (e.g., nitrifier denitrification), with a minor contribution from hydroxylamine (NH(2)OH) oxidation at the beginning of the experiments. SP of N(2)O produced under anoxic conditions was always positive (1.2 to 26.1 ‰), and SP values at the high end of this spectrum (24.9 to 26.1 ‰) are indicative of N(2)O reductase activity. The measured δ(15)N(bulk)(N2O) at the initiation of the NH(4)(+) oxidation experiments ranged between -42.3 and -57.6 ‰ (corresponding to a nitrogen isotope effect Δδ(15)N = δ(15)N(substrate) - δ(15)N(bulk)(N2O) of 43.5 to 58.8 ‰), which is considerably higher than under denitrifying conditions (δ(15)N(bulk)(N2O) 2.4 to -17 ‰; Δδ(15)N = 0.1 to 19.5 ‰). During the course of all NH(4)(+) oxidation and nitrate (NO(3)(-)) reduction experiments, δ(15)N(bulk)(N2O) increased significantly, indicating net (15)N enrichment in the dissolved inorganic nitrogen substrates (NH(4)(+), NO(3)(-)) and transfer into the N(2)O pool. The decrease in δ(15)N(bulk)(N2O) during NO(2)(-) and NH(2)OH oxidation experiments is best explained by inverse fractionation during the oxidation of NO(2)(-) to NO(3)(-).

While isotope ratio mass spectroscopy (IRMS) was traditionally used for high-precision measurement of the isotopic composition of N2O, quantum cascade laser absorption spectroscopy (QCLAS) has been put forward as a complementary technique with the potential for on-site analysis. In recent years, preconcentration combined with QCLAS has been presented as a technique to resolve subtle changes in ambient N2O isotopic composition. From the end of May until the beginning of August 2016, we investigated N2O emissions from an intensively managed grassland at the study site Fendt in Southern Germany. In total, 612 measurements of ambient N2O were taken by combining preconcentration with QCLAS analyses, yielding δ15Nα, δ15Nβ, δ18O and N2O concentration with a temporal resolution of approximately one hour and precisions of 0.46 ‰, 0.36 ‰, 0.59 ‰ and 1.24 ppb, respectively. Soil δ15N-NO3− values and concentrations of NO3− and NH4+ were measured to further constrain possible N2O-emitting source processes. Furthermore, the concentration footprint area of measured N2O was determined with a Lagrangian particle dispersion model (FLEXPART-COSMO) using local wind and turbulence observations. These simulations indicated that night-time concentration observations were largely sensitive to local fluxes. While bacterial denitrification and nitrifier denitrification were identified as the primary N2O-emitting processes, N2O reduction to N2 largely dictated the isotopic composition of measured N2O. Fungal denitrification and nitrification-derived N2O accounted for 34–42 % of total N2O emissions and had a clear effect on the measured isotopic source signatures. This study presents the suitability of on-site N2O isotopocule analysis for disentangling source and sink processes in-situ and found that at the Fendt site bacterial denitrification/nitrifier denitrification is the major source for N2O, while N2O reduction acted as a major sink.

Abstract:Nitrous oxide (N2O), a potent greenhouse gas that contributes significantly to climate change, is emitted mostly from soils by a suite of microbial metabolic pathways that are nontrivial to identify, and subsequently, to manage. Using either natural abundance or enriched stable isotope methods has aided in identifying microbial sources of N2O, but each approach has limitations. Here, we conducted a novel pairing of natural abundance and enriched assays on two dissimilar soils, hypothesizing this pairing would better constrain microbial sources of N2O. We incubated paired natural abundance and enriched soils from a corn agroecosystem and a subalpine forest in the laboratory at 10%–95% soil saturation for 28 hr. The natural abundance method measured intramolecular site preference (SP) from emitted N2O, whereas the enriched method measured emitted 15N2O from soils amended with 15N‐labeled substrate. The isotopic composition of emitted N2O was measured using a laser‐based N2O isotopic analyzer, yielding two key findings. First, both methods revealed that denitrification was the primary source of N2O in all soils: isotopic enrichment revealed clear NO3− $\mathrm{N}{{\mathrm{O}}_{3}}^{-}$ reduction to N2O, while SP indicated a likely combination of fungal and bacterial denitrification. Second, we quantified, to our knowledge for the first time, persistent (>55%) β‐position‐specific enrichment in N2O emitted from 15 NO3− $\mathrm{N}{{\mathrm{O}}_{3}}^{-}$ ‐amended soils. This counter‐intuitive enrichment pattern could be indicative of codenitrification, an understudied but potentially important contributor to N2O emissions. Our work revealed the ubiquity of denitrification among the soils tested. Future pairings of natural abundance and enriched methods could better characterize diverse denitrification pathways.

Abstract:We have previously indicated that the ideal animal tumor model should mimic the human disease. This means that the investigator should be able to ascertain the influence of host factors on the initiation of tumorigenesis, mimic the susceptibility of tumor response based on age and reproductive history, and determine the response of the tumors induced to chemotherapy. The utilization of experimental models of mammary carcinogenesis in risk assessment requires that the influence of ovarian, pituitary, and placental hormones, among others, as well as overall reproductive events are taken into consideration, since they are important modifiers of the susceptibility of the organ to neoplastic development. Several species, such as rodents, dogs, cats, and monkeys, have been evaluated for these purposes; however, none of them fulfills all the criteria specified previously. Rodents, however, are the most widely used models; therefore, this work will concentrate on discussing the rat rodent model of mammary carcinogenesis.
Oxygen exchange between (de)nitrification intermediates and H2O and its implications for source determination of NO3- and N2O: a review

Abstract:Stable isotope analysis of oxygen (O) is increasingly used to determine the origin of nitrate (NO(3)-) and nitrous oxide (N(2)O) in the environment. The assumption underlying these studies is that the (18)O signature of NO(3)- and N(2)O provides information on the different O sources (O(2) and H(2)O) during the production of these compounds by various biochemical pathways. However, exchange of O atoms between H(2)O and intermediates of the (de)nitrification pathways may change the isotopic signal and thereby bias its interpretation for source determination. Chemical exchange of O between H(2)O and various nitrogenous oxides has been reported, but the probability and extent of its occurrence in terrestrial ecosystems remain unclear. Biochemical O exchange between H(2)O and nitrogenous oxides, NO(2)- in particular, has been reported for monocultures of many nitrifiers and denitrifiers that are abundant in nature, with exchange rates of up to 100%. Therefore, biochemical O exchange is likely to be important in most soil ecosystems, and should be taken into account in source determination studies. Failing to do so might lead to (i) an overestimation of nitrification as NO(3)- source, and (ii) an overestimation of nitrifier denitrification and nitrification-coupled denitrification as N(2)O production pathways. A method to quantify the rate and controls of biochemical O exchange in ecosystems is needed, and we argue this can only be done reliably with artificially enriched (18)O compounds. We conclude that in N source determination studies, the O isotopic signature of especially N(2)O should only be used with extreme caution.
Isotopologue ratios of N2O emitted from microcosms with NH4+ fertilized arable soils under conditions favoring nitrification

Abstract:Soils represent the major source of the atmospheric greenhouse gas nitrous oxide (N2O) and there is a need to better constrain the total global flux and the relative contribution of the microbial source processes. The aim of our study was to determine variability and control of the isotopic fingerprint of N2O fluxes following NH4+-fertilization and dominated by nitrification. We conducted a microcosm study with three arable soils fertilized with 0–140mgNH4+–Nkg−1. Fractions of N2O derived from nitrification and denitrification were determined in parallel experiments using the 15N tracer and acetylene inhibition techniques or by comparison with unfertilized treatments. Soils were incubated for 3–10 days at low moisture (30–55% water-filled pore space) in order to establish conditions favoring nitrification. Dual isotope and isotopomer ratios of emitted N2O were determined by mass spectrometric analysis of δ18O, average δ15N (δ15Nbulk) and 15N site preference (SP=difference in δ15N between the central and peripheral N positions of the asymmetric N2O molecule). N2O originated mainly from nitrification (>80%) in all treatments and the proportion of NH4+ nitrified that was lost as N2O ranged between 0.07 and 0.45%. δ18O and SP of N2O fluxes ranged from 15 to 28.4‰ and from 13.9 to 29.8‰, respectively. These ranges overlapped with isotopic signatures of N2O from denitrification reported previously. There was a negative correlation between SP and δ18O which is opposite to reported trends in N2O from denitrification. Variation of average 15N signatures of N2O (δ15Nbulk) did not supply process information, apparently because a strong shift in precursor signatures masked process-specific effects on δ15Nbulk. Maximum SP of total N2O fluxes and of nitrification fluxes was close to reported SP of N2O from NH4+ or NH2OH conversion by autotrophic nitrifiers, suggesting that SP close to 30‰ is typical for autotrophic nitrification in soils following NH4+-fertilization. The results suggest that the δ18O/SP fingerprint of N2O might be used as a new indicator of the dominant source process of N2O fluxes in soils.

Abstract:Abstract. Nitrous oxide (N2O) is a potent greenhouse gas and produced in denitrification and nitrification by various microorganisms. Site preference (SP) of 15N in N2O, which is defined as the difference in the natural abundance of isotopomers 14N15NO and 15N14NO relative to 14N14NO, has been reported to be a useful tool to quantitatively distinguish N2O production pathways. To determine representative SP values for each microbial process, we firstly measured SP of N2O produced in the enzyme reaction of hydroxylamine oxidoreductase (HAO) purified from two species of ammonia oxidizing bacteria (AOB), Nitrosomonas europaea and Nitrosococcus oceani, and that of nitric oxide reductase (NOR) from Paracoccus denitrificans. The SP value for NOR reaction (−5.9 ± 2.1‰) showed nearly the same value as that reported for N2O produced by P. denitrificans in pure culture. In contrast, SP value for HAO reaction (36.3 ± 2.3‰) was a little higher than the values reported for N2O produced by AOB in aerobic pure culture. Using the SP values obtained by HAO and NOR reactions, we calculated relative contribution of the nitrite (NO2–) reduction (which is followed by NO reduction) to N2O production by N. oceani incubated under different O2 availability. Our calculations revealed that previous in vivo studies might have underestimated the SP value for the NH2OH oxidation pathway possibly due to a small contribution of NO2– reduction pathway. Further evaluation of isotopomer signatures of N2O using common enzymes of other processes related to N2O would improve the isotopomer analysis of N2O in various environments.

Abstract:The naturally occurring nitrogen (N) isotopes, 15N and 14N, exhibit different reaction rates during many microbial N transformation processes, which results in N isotope fractionation. Such isotope effects are critical parameters for interpreting natural stable isotope abundances as proxies for biological process rates in the environment across scales. The kinetic isotope effect of ammonia oxidation (AO) to nitrite (NO2–), performed by ammonia-oxidizing archaea (AOA) and ammonia-oxidizing bacteria (AOB), is generally ascribed to the enzyme ammonia monooxygenase (AMO), which catalyzes the first step in this process. However, the kinetic isotope effect of AMO, or εAMO, has been typically determined based on isotope kinetics during product formation (cumulative product, NO2–) alone, which may have overestimated εAMO due to possible accumulation of chemical intermediates and alternative sinks of ammonia/ammonium (NH3/NH4+). Here, we analyzed 15N isotope fractionation during archaeal ammonia oxidation based on both isotopic changes in residual substrate (RS, NH4+) and cumulative product (CP, NO2–) pools in pure cultures of the soil strain Nitrososphaera viennensis EN76 and in highly enriched cultures of the marine strain Nitrosopumilus adriaticus NF5, under non-limiting substrate conditions. We obtained εAMO values of 31.9–33.1‰ for both strains based on RS (δ15NH4+) and showed that estimates based on CP (δ15NO2–) give larger isotope fractionation factors by 6–8‰. Complementary analyses showed that, at the end of the growth period, microbial biomass was 15N-enriched (10.1‰), whereas nitrous oxide (N2O) was highly 15N depleted (−38.1‰) relative to the initial substrate. Although we did not determine the isotope effect of NH4+ assimilation (biomass formation) and N2O production by AOA, our results nevertheless show that the discrepancy between εAMO estimates based on RS and CP might have derived from the incorporation of 15N-enriched residual NH4+ after AMO reaction into microbial biomass and that N2O production did not affect isotope fractionation estimates significantly.

Abstract:Nitrous oxide (N2O) has been for long a major focus of all greenhouse gas accounting agreements. Understanding the mechanisms of its formation and clarifying its sources and sinks are highly important for mitigating N2O emissions. In this context, measuring the doubly substituted isotopocules of N2O can add new and unique opportunities to fingerprint and constrain the biogeochemical N2O cycle, similar to other atmospheric species such as CO2, CH4, and O2.We address this challenging research field by developing and validating a laser spectroscopic technique for selective analysis of the eight most abundant N2O isotopic species including the doubly substituted isotopocules 14N15N18O, 15N14N18O, and 15N15N16O. This method is based on quantum cascade laser absorption spectroscopy (QCLAS) and reaches a precision of 0.01 – 0.20 ‰ with 1 – 2 min spectral averaging on samples of 4 μmol of N2O in N2 at 4 hPa.Furthermore, we have established a new reference frame combining two independent approaches: (1) clumped N2O isotopocule abundances were linked to stochastic distribution by equilibrating the N–O bond in the N2O molecule over activated Al2O3 at 100 and 200 °C, and (2) individual isotopocule concentrations were calibrated using a set of high-accuracy gravimetric N2O-in-N2 gas mixtures. The latter approach, applied for the first time to clumped isotope measurements, has a particular potential in realizing regular multi-point calibration for species like 15N15N16O, because no procedure for equilibration of the N–N bond has been successful yet.Results of the validation measurements, using the QCLAS method and calibration approach, are presented for a large range of δ values (approx. 100 ‰ for d15N and d18O). Inter-comparison measurements with high-resolution mass spectrometry show compatible results for bulk isotopic composition (d15N, d(458+548)), but superior performance of QCLAS for determining site-selectivity (SP, SP18). In summary, this work provides new methodological basis for the measurements of clumped N2O isotopes and has a high potential to stimulate future research in the N2O community by establishing a new class of reservoir-insensitive tracers and molecular-scale insights.

Abstract:RATIONALE: Fungal denitrifiers can contribute substantially to N2 O emissions from arable soil and show a distinct site preference for N2 O (SP(N2 O)). This study sought to identify another process-specific isotopic tool to improve precise identification of N2 O of fungal origin by mass spectrometric analysis of the N2 O produced.METHODS: Three pure bacterial and three fungal species were incubated under denitrifying conditions in treatments with natural abundance and stable isotope labelling to analyse the N2 O produced. Combining different applications of isotope ratio mass spectrometry enabled us to estimate the oxygen (O) exchange accelerated by denitrifying enzymes and the ongoing microbial pathway in parallel. This experimental set-up allowed the determination of δ18 O(N2 O) values and isotopic fractionation of O, as well as SP(N2 O) values, as a perspective to differentiate between microbial denitrifiers.RESULTS: Oxygen exchange during N2 O production was lower for bacteria than for fungi, differed between species, and depended also on incubation time. Apparent O isotopic fractionation during denitrification was in a similar range for bacteria and fungi, but application of the fractionation model indicated that different enzymes in bacteria and fungi were responsible for O exchange. This difference was associated with different isotopic fractionation for bacteria and fungi.CONCLUSIONS: δ18 O(N2 O) values depend on isotopic fractionation and isotopic fractionation may differ between processes and organism groups. By comparing SP(N2 O) values, O exchange and the isotopic signature of precursors, we propose here a novel tool for differentiating between different sources of N2 O.
