Abstract:The cDNAs of two crustacean hyperglycemic hormones, designated as C1 and C2, were isolated from the eyestalk of mud crab Scylla paramamosain through RT-PCR and RACE. Sequence analysis indicated that C1 had an open reading frame of 420bp encoding 139aa of 15.326kDa and pI at 5.540; C2 had an open reading frame of 426bp encoding 141aa of 15.621kDa and pI at 7.618. The amino acid sequences of C1 and C2 possessed 92%, 80%, 78% identity with the CHHs of S. olivacea, Charybdis japonica, Portunus trituberculatus respectively. C1 and C2 protein firstly clustered with CHHs of S. olivacea and Charybdis japonica in the phylogenetic analysis. The expression of C1 and C2 in tissues were analyzed by Real-Time PCR, the result showed that C1 and C2 were expressed in eyestalk, hepatopancreas, intestinal, heart, gill and stomach, but not in muscle. After 24h＇s stress in salinity, the expression of C2 increased by a factor of thirty com- pared with C1. The expression of C2 in salinity 10 was not significant compared with salinity 5 （P〉0.05）, but significant with salinity 15 （P〈0.05） and great significant with salinity 20, 25, 30 （P〈0.01）. The more the salinity changed, the more the C2 expressed. These results served further studies on functions and regulation mechanism of CHHs.

Abstract:The most abundant neuropeptide,crustacean hyperglycemic hormone(CHH) has been first extracted from sinus glands of Eriocheir sinensis,and purified to homogeneity by reverse-phase high performance liquid chromatogram(RP-HPLC).The sinus glands of Eriocheir sinensis are the organs to store neuropeptides,which are quite perfect to do research for the neurosecretory system of Eriocheir sinensis.For the crab Eriocheir sinensis,CHH accounts for(153.63±32.23) ng(n=4) per SG,while the extracts of one sinus gland reach(1252.63±298.40) ng(n=4).In crustaceans,the CHH and the structurally related peptides MIH (molt-inhibiting hormone),VIH(vitellogenesis inhibiting hormone) and MOIH(mandibular organ inhibiting hormone),are classically considered as the major physiological regulators involved in the control of carbohydrate metabolism and in different functions such as molt,reproduction and hydromineral metabolism.The amino acid sequences of these neuropeptides hormones are similar to each other: they consist of 72-78 residues with six conserved cysteine residues arranged in three disulfide bridges.CHH shows obviously hyperglycemic activity,which can make the haemolymph of the crab Eriocheir sinensis reach the highest level(56.14±10.58) mg·100 mL~(-1)) only in 30 minutes,while the CHHs from any other species that have been examined show hyperglycemic activity in about 2 hours.The molecular weight of CHH was examined by SDS-PAGE under the indication of protein Marker(BBI,from 4.1 kD to 66 kD),the result was 5.1 kD with the use of GDS Microsoft to analyze,which is much lower than any other CHH(7-9 kD) that has already been known.In recent years,the amino acid sequences of the CHHs from many species of crustaceans have been determined,however,no sequence data for CHH from Eriocheir sinensis has been reported so far,and will be reported later.In this paper CHH from Eriocheir sinensis has been separated and its biological activity has been examined,the results of the experiment will be helpful for the further research in the synthesis and secretion of the neuropeptides,as well as have laid the foundation to determine the physiological function of other neuropeptides hormones in the sinus glands.

Abstract:Crustacean hyperglycemic hormone (CHH) is a polypeptide neurohormone involved in regulation of multiple physiological processes. We report here the cloning from thoracic ganglia of the blue crab (Callinectes sapidus) a cDNA (CsCHH-2) encoding a putative CHH isoform (CsCHH-2). CsCHH-2 is structurally similar to a putative preproCHH (CsCHH-1) previously cloned from eyestalk ganglia of C. sapidus. The two preprohormones possess an identical signal peptide and CHH precursor related peptide, but differ in the mature CHH polypeptide. An analysis by RT-PCR of the tissue distribution of CsCHH-1 and CsCHH-2 revealed the former is restricted to eyestalk neural ganglia, while the latter is widely distributed among tissues. The type of CHH transcript present in eyestalk and thoracic ganglia did not vary as a function of the molt cycle. An assessment of transcript abundance in tissues of intermolt crabs showed the abundance of the CsCHH-1 transcript in eyestalk ganglia far exceeds the abundance of the CsCHH-2 transcript in extra-eyestalk tissue. An assessment of transcript abundance during a molt cycle showed CsCHH-1 transcript abundance in eyestalk ganglia was low during intermolt, rose during premolt, reaching a peak in D(3), then fell prior to molting, and remained low during postmolt. By contrast, CsCHH-2 transcript abundance in thoracic ganglia was low during intermolt, rose sharply during D(2), then dropped in D(3) and remained low during postmolt. The results are consistent with the hypothesis that CsCHH-1 and CsCHH-2 differ with respect to physiological function.

Abstract:BACKGROUND Multiple primary malignant neoplasms in a single patient have been well documented in the literature. The lesions can be limited to a single organ or involve multiple organ systems and can be synchronous or metachronous. AIM Report of a new case. CASE REPORT We report the case of a 63 year old women with a rectocolonic carcinoma; during surgery an ileal nodule was discovered also as liver metastasis. The final pathologic report on the specimen revealed an adenocarcinoma of the colon and a carcinoid tumor of the small bowel with liver metastasis. Tumoral association is relatively common for patients with small intetinal carcinoids. CONCLUSION The association carcinoide of hail with primitive tumour not carcinoide is reported primarily with carcinomes of the digestive tract in particulary recto colics carcinoma.

Abstract:: Six peptides belonging to the crustacean hyperglycemic hormone (CHH) family were isolated from extracts of the sinus glands of the giant tiger prawn Penaeus monodon by reverse-phase high-performance liquid chromatography. These were designated Pem-SGP-A to Pem-SGA-F (Pem, Penaeus monodon; SGP, sinus gland peptide) and their amino-terminal amino acid sequences were analyzed. Five of the 6 peptides (Pem-SGP-A, -B, -D, -E, and -F) exhibited similar amino acid sequences to those of CHH peptides that had been characterized previously from the same prawn species by the other research groups, while 1 peptide (Pem-SGP-C) exhibited a novel sequence. Pem-SGP-C showed sequence similarity to known putative molt-inhibiting hormones (MIHs), particularly to an MIH from the kuruma prawn Penaeus japonicus, and less similarity to the CHHs determined thus far. Two similar complementary DNAs encoding Pem-SGP-C were cloned and found to encode very similar but distinct peptides, which were named Pem-SGP-C1 and Pem-SGA-C2. The open reading frame of each cDNA consisted only of a signal peptide and an MIH-like peptide. We also cloned 2 corresponding genes, both of which consisted of 3 exons and 2 introns. Analyses by reverse transcriptase polymerase chain reaction demonstrated that both Pem-SGP-C1 and Pem-SGP-C2 transcripts were detected only in cDNA synthesized using total RNA from the eyestalk but not in that from brain, thoracic ganglia, abdominal ganglia, abdominal muscle, hepatopancreas, or heart tissue of P. monodon.

Abstract:Synthesis of ecdysteroids (molting hormones) by crustacean Y-organs is regulated by a neuropeptide, molt-inhibiting hormone (MIH), produced in eyestalk neural ganglia. We report here the molecular cloning of a cDNA encoding MIH of the edible crab, Cancer pagurus. Full-length MIH cDNA was obtained by using reverse transcription-polymerase chain reaction (RT-PCR) with degenerate oligonucleotides based upon the amino acid sequence of MIH, in conjunction with 5'- and 3'-RACE. Full-length clones of MIH cDNA were obtained that encoded a 35 amino acid putative signal peptide and the mature 78 amino acid peptide. Of various tissues examined by Northern blot analysis, the X-organ was the sole major site of expression of the MIH gene. However, a nested-PCR approach using non-degenerate MIH-specific primers indicated the presence of MIH transcripts in other tissues. Southern blot analysis indicated a simple gene arrangement with at least two copies of the MIH gene in the genome of C. pagurus. Additional Southern blotting experiments detected MIH-hybridizing bands in another Cancer species, Cancer antennarius and another crab species, Carcinus maenas.

Abstract:Two cDNA clones (634 and 1366 bp) encoding MIH/VIH (molt-inhibiting hormone/vitellogenesis-inhibiting hormone)-related peptides were isolated and sequenced from a Macrobrachium rosenbergii eyestalk ganglia cDNA library. The clones contain a 360 and 339 bp open-reading frame, and their conceptually translated peptides consist of a 41 and 34 amino acid signal peptide, respectively, and a 78 amino acid residue mature peptide hormone. The amino acid sequences of the peptides exhibit higher identities with other known MIHs and VIH (44–69%) than with CHHs (28–33%). This is the first report describing the cloning and sequencing of two MIH/VIH-related peptides in a single crustacean species. Transcription of these mRNAs was detected in the eyestalk ganglia, but not in the thoracic ganglia, hepatopancreas, gut, gill, heart, or muscle.

Abstract:In crustaceans, chitinases (Chts) play crucial roles in physiological functions such as chitinous food digestion, moulting and stress responses. Chitinases have been identified in both prokaryotes and eukaryotes, but the composition and number of Cht family members vary across species, especially in crustaceans. Seven Cht transcripts, all of which contained the conserved enzyme activity motif, were cloned from Scylla paramamosain for the first time, and SpCht1-SpCht6 were clearly classified into groups 1-6, respectively, through phylogenetic analysis. Notably, SpCht7, which had two catalytic domains, was identified for the first time in crabs and crustaceans. Although SpCht7 could not be grouped into any known Cht subclasses, it clustered together with insect Cht7 and could be classified into a novel subclass that we named group 7. The tissue distributions of SpChts, as well as their expression profiles during embryonic and larval development, growth-related moulting, salinity and cadmium stresses, were also investigated. The results suggested that SpChts might have different and varied biological functions. These results would be helpful for understanding and clarifying the roles of chitinases in crab development, moulting and responses to environmental stress. These results also provide new insight into the origin and evolution of the chitinase gene family in crabs and other crustaceans.

Abstract:Molt-inhibiting hormone (MIH), a member of the crustacean hyperglycemic hormone (CHH) family, inhibits the synthesis of ecdysteroid in Y-organ (YO) and plays a significant role in the regulation of molting and growth of crustaceans. A complete cDNA sequence encoding MIH (Ers-MIH, GenBank Accession No.: DQ341280) was cloned from eyestalk of Chinese mitten crab (Eriocheir sinensis) by 5' and 3' RACEs and PCR cloning. The full-length cDNA consists of 1457 bp with a 330 bp open reading frame, encoding 110 amino acids, containing a 75 amino acid mature peptide. The deduced amino acid sequence contains a typical CHH domain. Transcripts of Ers-MIH mRNA were detected in eyestalk by Northern blotting. The production of purified recombinant Ers-MIH (rErs-MIH) expressed in Escherichia coli was 0.3g/L. The LC-ESI-MS analysis showed that two peptide fragments of the recombinant protein were identical to the deduced amino acid sequence of Ers-MIH. By in vitro assay on E. sinensis YOs, a cGMP mediated suppression of rErs-MIH on ecdysteroidogenesis could be observed. Accumulation of cGMP in YOs showed a concentration-dependent manner within 0.01-1 nmol/mL of rErs-MIH; ecdysteroid secretion was inhibited significantly at the range of 0.01-100 nmol/mL rErs-MIH; furthermore, a significant inhibition effect on ecdysteroid releasing was shown when cGMP analog (8-Br-cGMP) concentration rose up to 100 nmol/mL. This study would facilitate to investigate the roles of MIH in molt cycle regulation.

Abstract:Molt-inhibiting hormone (MIH) activity was tested in terms of the inhibitory activity of ecdysteroid secretion by cultured Y-organs of the crayfish, Procambarus clarkii. Several neuropeptides were separated from a crude extract of sinus glands of P. clarkii by means of reverse-phase high-performance liquid chromatography (HPLC) and tested for MIH activity. An extremely high level of activity was found in one fraction in reverse-phase HPLC, suggesting that the novel neuropeptide in this fraction may be a P. clarkii MIH.
Identification and characterization of a hyperglycemic hormone from freshwater giant prawn, Macrobrachium rosenbergii

Abstract:Crustacean hyperglycemic hormone (CHH), a physiologically important neurohormone stored in the sinus gland of eyestalks, primarily regulates carbohydrate metabolism and also plays significant roles in reproduction, molting and other physiological processes. In the freshwater giant prawn, Macrobrachium rosenbergii, an injection of X-organ sinus gland (XOSG) extract evoked a hyperglycemic response, peaked in 1 h. The hyperglycemic effect of the eyestalk extract was maximal at the dose of 0.5 eyestalk equivalent. CHH fractionated by RP-HPLC, in M. rosenbergii was identified by its hyperglycemic activity and partial amino acid sequence, and the molecular weight of 8534 was determined by matrix-assisted laser desorption ionization mass spectrometry--time of flight analysis (MALDI-TOF). The amino acid sequence of the first 25 residues of CHH showed 72% homology with the first 25 residues of CHH A and CHH B of the American lobster Homarus americanus.

Abstract:Ecdysteroid molting hormone synthesis is directed by a pair of molting glands or Y-organs (YOs), and this synthesis is inhibited by molt-inhibiting hormone (MIH). MIH is a member of the crustacean hyperglycemic hormone (CHH) neuropeptide superfamily, which includes CHH and insect ion transport peptide (ITP). It is hypothesized that the MIH receptor is a Class A (Rhodopsin-like) G protein-coupled receptor (GPCR). The YO of the blackback land crab, Gecarcinus lateralis , expresses 49 Class A GPCRs, three of which (Gl-CHHR-A9, -A10, and -A12) were provisionally assigned as CHH-like receptors. CrusTome, a transcriptome database assembled from 189 crustaceans and 12 ecdysozoan outgroups, was used to deorphanize candidate MIH/CHH GPCRs, relying on sequence homology to three functionally characterized ITP receptors (BNGR-A2, BNGR-A24, and BNGR-A34) in the silk moth, Bombyx mori . Phylogenetic analysis and multiple sequence alignments across major taxonomic groups revealed extensive expansion and diversification of crustacean A2, A24, and A34 receptors, designated C HH F amily R eceptor C andidates (CFRCs). The A2 clade was divided into three subclades; A24 clade was divided into five subclades; and A34 was divided into six subclades. The subclades were distinguished by conserved motifs in extracellular loop (ECL) 2 and ECL3 in the ligand-binding region. Eleven of the 14 subclades occurred in decapod crustaceans. In G. lateralis , seven CFRC sequences, designated Gl-CFRC-A2α1, -A24α, -A24β1, -A24β2, -A34α2, -A34β1, and -A34β2, were identified; the three A34 sequences corresponded to Gl-GPCR-A12, -A9, and A10, respectively. ECL2 in all the CFRC sequences had a two-stranded β-sheet structure similar to human Class A GPCRs, whereas the ECL2 of decapod CFRC-A34β1/β2 had an additional two-stranded β-sheet. We hypothesize that this second β-sheet on ECL2 plays a role in MIH/CHH binding and activation, which will be investigated further with functional assays.

Abstract:Mandibular organs (MO) produce a crustacean juvenile hormone, methyl farnesoate (MF). MO activity is negatively regulated by factors, called mandibular organ inhibiting hormones (MOIHs), from the crustacean sinus gland X-organ complex in the eyestalks. Three MOIHs have been isolated previously from the spider crab Libinia emarginata and are characterized as members of the crustacean hyperglycemic hormone (CHH) neuropeptide family. In the research reported here, a full length cDNA sequence of 972 bp of a MOIH was isolated by screening a cDNA library constructed from the eyestalks of Libinia emarginata . This cDNA sequence encodes a preprohormone peptide with 137 amino acid residues, including a 26-amino acid long signal peptide, a 34-amino acid long precursor peptide, a dibasic peptide, the full length of 72-amino acid long MOIH, and a tri-peptide Gly-Lys-Lys which designates the potential amidation site at the C-terminus of the mature peptide.

Abstract:Using two-step reverse phase HPLC, we isolated and purified three peptides with mandibular organ inhibiting hormone (MO-IH) activity from the spider crab Libinia emarginata. One of the peptides, P22, gave a yield of 355 ng/SG. The others gave lower yields: P21, 9 ng/SG; P25, 67.5 ng/SG. The molecular weight was determined to be 8,439 for P25, 8,474 for P22, and 8,398 for P21 by mass spectrometry. All three peptides have similar amino acid compositions and contain 72-76 residues. We believe these peptides to be different isoforms of one family. The MO is more sensitive to the two minor isoforms, P21 and P25. All three isoforms can inhibit MO activity to a maximum inhibition of 70%. All three isoforms gave a significant hyperglycemic effect when injected into de-eyestalked fiddler crabs Uca pugilator. We believe the MO-IHs to be members of the crustacean hyperglycemic hormone (CHH) family, having similar amino acid compositions and both biological activities. (C) 1996 Wiley-Liss, Inc.

Abstract:Allatostatins are important regulatory neuropeptides that inhibit juvenile hormone (JH) biosynthesis by the corpora allata (CA) in insects. However, to date, the structure and expression of the gene encoding allatostatins have not been reported in any species other than insects. In this study, we used a combination of a semi-nested polymerase chain reaction (PCR) and screening of a central nervous system cDNA library of Macrobrachium rosenbergii to isolate and sequence a cDNA clone (2885bp) encoding a 701 amino acid FGLamide allatostatin precursor polypeptide. This is the first reported allatostatin gene in crustacean. The deduced precursor was conceptually split into at least 35 FGLamide allatostatins at dibasic cleavage sites (Lys and Lys/Arg), far more than reported for any other known FGLamide allatostatin precursors from insects (13–14 allatostatins). Reverse transcription-polymerase chain reaction (RT-PCR) analysis demonstrated that the gene was expressed in the brain, gut, thoracic and abdominal ganglia, but not in the heart, muscle, ovary, gill, or hepatopancreas. Furthermore, developmentally-dependent expression of the gene was observed in the brain and thoracic ganglia of the prawn by using semi-quantitative RT-PCR analysis.
Identification and developmental expression of mRNAs encoding crustacean cardioactive peptide (CCAP) in decapod crustaceans

Abstract:Full-length cDNAs encoding crustacean cardioactive peptide (CCAP) were isolated from several decapod (brachyuran and astacuran) crustaceans: the blue crab Callinectes sapidus, green shore crab Carcinus maenas, European lobster Homarus gamarus and calico crayfish Orconectes immunis. The cDNAs encode open reading frames of 143 (brachyurans) and 139-140 (astacurans) amino acids. Apart from the predicted signal peptides (30-32 amino acids), the conceptually translated precursor codes for a single copy of CCAP and four other peptides that are extremely similar in terms of amino acid sequence within these species, but which clearly show divergence into brachyuran and astacuran groups. Expression patterns of CCAP mRNA and peptide were determined during embryonic development in Carcinus using quantitative RT-PCR and immunohistochemistry with whole-mount confocal microscopy, and showed that significant mRNA expression (at 50% embryonic development) preceded detectable levels of CCAP in the developing central nervous system (CNS; at 70% development). Subsequent CCAP gene expression dramatically increased during the late stages of embryogenesis (80-100%), coincident with developing immunopositive structures. In adult crabs, CCAP gene expression was detected exclusively in the eyestalk, brain and in particular the thoracic ganglia, in accord with the predominance of CCAP-containing cells in this tissue. Measurement of expression patterns of CCAP mRNA in Carcinus and Callinectes thoracic ganglia throughout the moult cycle revealed only modest changes, indicating that previously observed increases in CCAP peptide levels during premoult were not transcriptionally coupled. Severe hypoxic conditions resulted in rapid downregulation of CCAP transcription in the eyestalk, but not the thoracic ganglia in Callinectes, and thermal challenge did not change CCAP mRNA levels. These results offer the first tantalising glimpses of involvement of CCAP in environmental adaptation to extreme, yet biologically relevant stressors, and perhaps suggest that the CCAP-containing neurones in the eyestalk might be involved in adaptation to environmental stressors.
Chh Family Peptides From An 'Eyeless' Deep-Sea Hydrothermal Vent Shrimp, Rimicaris Kairei: Characterization And Sequence Analysis

Abstract:The crustacean eyestalk synthesizes and secretes several structurally-related peptides belonging to the crustacean hyperglycemic hormone (CHH) family, which are considered major physiological regulators during the crustacean life cycle. However, it is intriguing that eyestalks of many hydrothermal vent crustaceans prove to have varying degrees of reduction. In the present study, we characterized full-length cDNAs encoding two important eyestalk hormones of the CHH family, CHH and VIH (vitellogenesis-inhibiting hormone), from the 'eyeless' hydrothermal vent shrimp Rimicaris kairei. The two isoforms of Chh cDNA were 1027 and 1877 bp in length, respectively, and the deduced preprohormones contained 137 and 138 aa, respectively. The Vih cDNA was 907 bp in length, encoding a putative preprohormone of 113 aa. When compared with other known protein sequences of CHHs and VIHs, these polypeptides from hydrothermal vents show high similarity with their non-vent counterparts. These results may provide evidence for the mechanisms of eyestalk reduction and vent-adapting evolution of crustaceans. The hydrothermal vent shrimp with reduced eyestalks may take a different evolutionary pathway than eyestalk-holding crustaceans, and the reduced eyestalks can be considered a good example for the investigation of the diversity of crustacean evolution in different environments. Expression and release patterns of neuropeptides during embryonic development and hatching of the green shore crab, Carcinus maenas

Abstract:Crustacean ecdysis is controlled by at least three neuropeptides: moult-inhibiting hormone (MIH), which represses ecdysteroid synthesis; crustacean hyperglycaemic hormone (CHH), which not only influences ecdysteroid synthesis but also water uptake during moulting; and crustacean cardioactive peptide (CCAP), which is involved in stereotyped ecdysis behaviour. During embryonic development, moulting takes place in the egg, but there is little information regarding developmental expression of these neuropeptides during this period or during hatching--an event that is analogous to eclosion in insects. To address this problem, we determined expression profiles of MIH and CHH mRNA by quantitative RT-PCR, together with developmental peptide expression studies [confocal immunocytochemistry (ICC) and radioimmunoassay (RIA)]. Likely homologous events relating to neuropeptide surges of both CHH and CCAP were seen during larval hatching, when compared to the adult moult, and cell-specific copy concentration of both MIH and CHH mRNAs was identical to that of the adult during late embryonic development. We measured parallel mRNA and peptide expression of two neuropeptides (red pigment-concentrating hormone RPCH) and pigment-dispersing hormone (PDH) during development, as these have roles as neuromodulators and as classical neurohormonal roles. For MIH and CHH, gene expression was in accordance with peptide expression, but novel sites of CHH expression were found (abdominal peripheral neurones), the expression and release patterns of which may be related to larval eclosion and water uptake necessary for eggshell rupture and hatching. For RPCH and PDH, gene transcription and peptide expression were not in accordance. A significant contribution of maternally derived (non-translated) PDH mRNA to the embryo was seen, and for RPCH, high-level mRNA and peptide expression during late embryogenesis is related to a long ignored, but potentially important release site--the enigmatic post-commissural organs--which are the most prominent structures expressing RPCH during late embryogenesis.
Hyperglycaemic Hormones Inhibit Protein and Mrna Synthesis in in Vitro-Incubated Ovarian Fragments of the Marine Shrimp Penaeus Semisulcatus.

Abstract:The present work shows for the first time that peptides belonging to the Crustacean hyperglycaemic hormone family (CHH-family hormones) from Penaeus japonicus affect protein and mRNA synthesis in in vitro-incubated ovarian explant fragments removed from vitellogenic females of Penaeus semisulcatus. Reduced levels of protein synthesis, determined by TCA-precipitable 35S-labeled proteins, were found in the presence of crude sinus gland extracts from both P. semisulcatus and P. japonicus. A similar inhibitory effect compared to controls was found with each of the seven CHH-family peptides. Non-CHH-family peptides did not reduce protein synthesis. Crude sinus gland extracts prepared from P. semisulcatus were at least 20-fold more effective than sinus gland extracts of P. japonicus. The inhibition level was directly related to the concentration of the peptide in the incubation media, but its degree varied among the different tested peptides. The profile of proteins synthesized during in vitro incubation was analyzed using polyacrylamide gel electrophoresis under denatured and reduced conditions (SDS-PAGE), followed by autoradiography. Synthesis of several proteins was reduced, including proteins with electrophoretic mobility similar to that of vitellin. Immunoprecipitation with antiserum prepared against native ovarian vitellin confirmed the inhibitory effect of CHH-family peptides on vitellin synthesis. The crude sinus gland extract and CHH-family peptides also inhibited RNA synthesis, as determined by [3H]uridine incorporation into mRNA of ovarian fragments. It is concluded that in addition to their role in carbohydrate metabolism, CHH-family peptides may also influence ovarian physiology in crustaceans.
