Abstract:This study focuses on the aggregation behavior of the biodegradable amphiphilic block copolymers based on methoxy poly(ethylene glycol) (mPEG) as a hydrophilic block and either crystalline poly(caprolactone-b-l-lactide) (P(CL-LLA)) or amorphous poly(caprolactone-b-D,L-lactide) (P(CL-DLLA)) as a hydrophobic block. These block copolymers have a strong tendency to form micelles in aqueous medium, with very low critical micelle concentrations (CMCs). The CMC of P(CL-LLA)-b-mPEG is higher than that of P(CL-DLLA)-b-mPEG when the mPEG block has the same molecular weight. Furthermore, the partition equilibrium coefficient (K(v)) of pyrene in the micellar solution of P(CL-LLA)-b-mPEG copolymer was lower than that of P(CL-DLLA)-b-mPEG copolymer when the mPEG block was the same length. These differences were believed to be related to the physical state of the core-forming blocks, i.e., the crystalline P(CL-LLA) block and the amorphous P(CL-DLLA) block. The TEM images showed that micelles formed by P(CL-LLA)-b-mPEG assembled in a cylindrical morphology, whereas those formed by P(CL-DLLA)-b-mPEG took a classical spherical shape. In addition, with differential scanning calorimetry (DSC) and wide-angle X-ray diffraction (WAXD) analyses, it is believed that the crystallization tendency of the core-forming blocks is the main factor governing the morphology of micelles in water. A possible mechanism for the cylindrical assembly morphology was discussed.

Abstract:Amphiphilic block copolymers consisting of methoxy poly(ethylene glycol) (mPEG) and either crystalline poly(caprolactone-b-L-lactide) [P(CL-LLA)]or amorphous poly(caprolactone-b-D,L-lactide) [P(CL-DLLA)], respectively, were prepared under mild conditions using dicyclohexylcarbodiimide as coupling agent. Micelles were formed from the resultant copolymers with predetermined hydrophobic and hydrophilic blocks in water by dialysis method. The composition and structure of the copolymers were characterized by H-1-NMR, C-13-NMR, and GPC measurements. Differential scanning calorimetry analysis revealed that the crystallization behavior of P(CL-LLA) block is affected significantly by the molecular weight of the conjugated mPEG block, and the glass transition temperature (T-g) of P(CL-DLLA) block is depressed by the existence of mPEG moiety in the block copolymer. The crystallization behavior was also characterized by the technique of polarized light microscopy. The hydrodynamic diameter and size distribution of micelles were determined by particle size measurements. Transmission electron microscopy images showed that P(CL-LLA)-b-mPEG block copolymer self-aggregated into thread-like shape in water, whereas P(CL-DLLA)-b-mPEG adopted a classical spherical shape. It is suggested that both the high enthalpy of crystallization and hydrophobicity of P(CL-LLA) core-forming blocks would be responsible for the thread-like morphology. Abstract:Block copolymer (BCP) self-assembly and crystallization determine their fundamental mechanical, optical, and transport properties. Multiple physical and chemical routes to control BCP self-assembly have been developed over the years. In contrast, modulating the crystallization behavior, particularly crystallite size and orientation, has received considerably less attention. This paper underpins that 1,2-dipalmitoyl-sn-glycero-3-phosphocholine (DPPC) phospholipid additives enable precise control of BCP crystallization via a combination of interfacial and confinement effects. The assembly and crystallization behavior of methoxy-poly(ethylene glycol)-b-poly(ε-caprolactone) (mPEG-b-PCL) hybridized with DPPC was elucidated in solid-supported dry-film forms. mPEG-b-PCL/DPPC hybrid films coassemble into multilamellar structures with BCP and lipid domains stacked in registry across the film thickness. With small amounts of DPPC (3 wt %), mPEG-b-PCL/DPPC hybrid films unexpectedly exhibit nonadditive thermal conductivity, reaching ∼0.1 W m–1 K–1, a value considerably lower than pure BCP or DPPC films. Hybridization alters DPPC chain packing and induces specific PCL crystallite orientation at the nanoscale. PCL chains on the folding plane of an orthorhombic lattice optimize molecular interactions with vertically aligned DPPC alkyl chains by orienting PCL crystallites at a 20° tilt. These results enable new applications of polymeric systems in which switchable crystallization routes are used to tune heat transport.

Structural characterization of lateral phase separation in polymer-lipid hybrid membranes

Abstract:Hierarchic self-assembly is the main mechanism used to create diverse structures using soft materials. This is a case for both synthetic materials and biomolecular systems, as exemplified by the non-covalent organization of lipids into membranes. In nature, lipids often assemble into single bilayers, but other nanostructures are encountered, such as bilayer stacks and tubular and vesicular aggregates. Synthetic block copolymers can be engineered to recapitulate many of the structures, forms, and functions of lipid systems. When block copolymers are amphiphilic, they can be inserted or co-assembled into hybrid membranes that exhibit synergistic structural, permeability, and mechanical properties. One example is the emergence of lateral phase separation akin to the raft formation in biomembranes. When higher-order structures, such as hybrid membranes, are formed, this lateral phase separation can be correlated across membranes in the stack. This chapter outlines a set of important methods, such as X-ray Scattering, Atomic Force Microscopy, and Cryo-Electron Microscopy, that are relevant to characterizing and evaluating lateral and correlated phase separation in hybrid membranes at the nano and mesoscales. Understanding the phase behavior of polymer-lipid hybrid materials could lead to innovative advancements in biomimetic membrane separation systems.

Abstract:Blue phase liquid crystals (BPLCs) self-assembling into soft cubic crystalline structures possess significant applications in the field of dynamic photoelectron spectroscopy. However, three scientific challenges limit their broader application, namely, the difficulty to form a free-standing structure due to fluidity, dependence on nanoparticles or polymer matrix to widen the BP temperature range, and high working voltages to reorientate the LCMs. Herein, we proposed a new concept of free-standing BPLC microcapsules (BPLCMs) prepared by microfluidic emulsification and interfacial polymerization. The temperature range of BPLCs could be widened by surface anchoring and physical confinement. The BPLCMs built a color library in the visible region in the thermal response process. Most importantly, the stable BPLCMs exhibited a reversible surface-switch mode driven by an electric field lower than 0.55 V mu m(-1), which was the lowest record for BPLCs. The BPLCMs demonstrate intriguing prospects in the fields of smart display and reversible sensors.

Abstract:Structural properties of plasmenyl-glycerophospholipids (plasmalogens) have been scarcely studied for plasmalogens with long polyunsaturated fatty acid (PUFA) chains, despite of their significance for the organization and functions of the cellular membranes. Elaboration of supramolecular assemblies involving PUFA-chain plasmalogens in nanostructured mixtures with lyotropic lipids may accelerate the development of nanomedicines for certain severe pathologies (e.g., peroxisomal disorders, cardiometabolic impairments, and neurodegenerative Alzheimer’s and Parkinson’s diseases). Here, we investigate the spontaneous self-assembly of bioinspired, custom-produced docosapentaenoyl (DPA) plasmenyl (ether) and ester phospholipids in aqueous environment (pH 7) by synchrotron small-angle X-ray scattering (SAXS) and cryogenic transmission electron microscopy (cryo-TEM). A coexistence of a liquid crystalline primitive cubic Im3m phase and an inverted hexagonal (H II ) phase is observed for the DPA-ethanolamine plasmalogen (C16:1p-22:5n6 PE) derivative. A double-diamond cubic Pn3m phase is formed in mixed assemblies of the phosphoethanolamine plasmalogen (C16:1p-22:5n6 PE) and monoolein (MO), whereas a coexistence of cubic and lamellar liquid crystalline phases is established for the DPA-plasmenyl phosphocholine (C16:1p-22:5n6 PC)/MO mixture at ambient temperature. The DPA-diacyl phosphoinositol (22:5n6-22:5n6 PI) ester lipid displays a propensity for a lamellar phase formation. Double membrane vesicles and multilamellar onion topologies with inhomogeneous distribution of interfacial curvature are formed upon incorporation of the phosphoethanolamine plasmalogen (C16:1p-22:5n6 PE) into dioleoylphosphocholine (DOPC) bilayers. Nanoparticulate formulations of plasmalogen-loaded cubosomes, hexosomes, and various multiphase cubosome- and hexosome-derived architectures and mixed type nano-objects (e.g., oil droplet-embedding vesicles or core–shell particles with soft corona) are produced with PUFA-chain phospholipids and lipophilic antioxidant-containing membrane compositions that are characterized by synchrotron SAXS and cryo-TEM imaging. The obtained multiphase nanostructures reflect the changes in the membrane curvature induced by the inclusion of DPA-based PE and PC plasmalogens, as well as DPA-PI ester derivative, and open new opportunities for exploration of these bioinspired nanoassemblies.

Abstract:A series of liquid crystalline/crystalline block copolymers (BCPs) containing poly(methacrylate) block with liquid crystalline (LC) azobenzene moieties in the side chains (PMMAzo) and crystalline block poly(l-lactide) (PLLA) were prepared. The interplay of microphase separation, crystallization and LC ordering in these BCPs was investigated. It is revealed that microphase separation between two blocks is favorable to the LC ordering, which is attributed to the enhanced local concentration of LC moieties in PMMAzo microdomains. For a similar reason, crystallization of PLLA can intensify microphase separation thus facilitate LC ordering of PMMAzo. PLLA crystallization may also stabilize the LC structure, leading to phase transition temperatures of the BCPs higher than that of PMMAzo homopolymer. On the other hand, the LC ordering can conversely affect crystallization of PLLA. The crystallizability of PLLA is weakened by the chemically linked PMMAzo block. The special PLLA ε-crystals, which are usually formed in the presence of organic solvents, are unexpectedly observed under suitable conditions.
Hybrid polymer/liquid vesicles as new particles for drug delivery and cell mimics

Abstract:Hybrid copolymer/lipid vesicle are recently developed self-assembled structures that could present biocompatibility and biofunctionality of liposomes, as well as robustness, low permeability and functionality variability conferred by the copolymer chains. However, to date, physical and molecular parameters governing copolymer/lipid phase separation in these hybrid membranes are not well understood. In this work, we studied in detail the formation and phase separation in the membranes of both Giant Unilamellar Hybrid Vesicles(GHUVs) and Large Unilamellar Hybrid Vesicles (LHUVs) obtained from the mixture of phospholipids in the fluid (liquid disordered) or gel state (solid ordered) with various copolymers based on poly(dimethylsiloxane) (PDMS) and poly(ethylene glycol) (PEO) with different architectures (grafted, triblock) and molar masses. For GHUVs, phase separation at the micron scale and nanoscale was evaluated through confocal microscopy, and Fluorescence lifetime imaging microscopy technique (FLIM) respectively, where as acombination of Small angle neutron scattering (SANS), Cryo-transmission electron microscopy (Cryo-TEM) and Time-resolved Forster resonance energy transfer (TR-FRET) techniques was used for LHUVs. We demonstrate that the lipid/polymer fraction, lipid physical state, and the line tension at lipid polymer/lipid boundaries which can be finely modulated by the molar mass and architecture of the copolymer are important factors that govern the formation and structuration of hybrid vesicles. We also evidence that elasticity ofthe hybrid membrane can be modulated via the lipid polymer composition, through the use of micropipettes techniques.

Self-assembled Lyotropic Liquid Crystalline Phase Behavior of Monoolein-Capric Acid-Phospholipid Nanoparticulate Systems

Abstract:We report here the lyotropic liquid crystalline phase behavior of two lipid nanoparticulate systems containing mixtures of monoolein, capric acid, and saturated diacyl phosphatidylcholines dispersed by the Pluronic F127 block copolymer. Synchrotron small-angle X-ray scattering (SAXS) was used to screen the phase behavior of a library of lipid nanoparticles in a high-throughput manner. It was found that adding capric acid and phosphatidylcholines had opposing effects on the spontaneous membrane curvature of the monoolein lipid layer and hence the internal mesophase of the final nanoparticles. By varying the relative concentration of the three lipid components, we were able to establish a library of nanoparticles with a wide range of mesophases including at least the inverse bicontinuous primitive and double diamond cubic phases, the inverse hexagonal phase, the fluid lamellar phase, and possibly other phases. Furthermore, the in vitro cytotoxicity assay showed that the endogenous phospholipid-containing nanoparticles were less toxic to cultured cell lines compared to monoolein-based counterparts, improving the potential of the nonlamellar lipid nanoparticles for biomedical applications.
Facile Mixing of Phospholipids Promotes Self-Assembly of Low-Molecular-Weight Biodegradable Block Co-Polymers into Functional Vesicular Architectures

Abstract:In this work, we have used low-molecular-weight (PEG12-b-PCL6, PEG12-b-PCL9 or PEG16-b-PLA38; MW, 1.25–3.45 kDa) biodegradable block co-polymers to construct nano- and micron-scaled hybrid (polymer/lipid) vesicles, by solvent dispersion and electroformation methods, respectively. The hybrid vesicles exhibit physical properties (size, bilayer thickness and small molecule encapsulation) of a vesicular boundary, confirmed by cryogenic transmission electron microscopy, calcein leakage assay and dynamic light scattering. Importantly, we find that these low MW polymers, on their own, do not self-assemble into polymersomes at nano and micron scales. Using giant unilamellar vesicles (GUVs) model, their surface topographies are homogeneous, independent of cholesterol, suggesting more energetically favorable mixing of lipid and polymer. Despite this mixed topography with a bilayer thickness similar to that of a lipid bilayer, variation in surface topology is demonstrated using the interfacial sensitive phospholipase A2 (sPLA2). The biodegradable hybrid vesicles are less sensitive to the phospholipase digestion, reminiscent of PEGylated vesicles, and the degree of sensitivity is polymer-dependent, implying that the nano-scale surface topology can further be tuned by its chemical composition. Our results reveal and emphasize the role of phospholipids in promoting low MW polymers for spontaneous vesicular self-assembly, generating a functional hybrid lipid-polymer interface.

Abstract:Cell membranes are vital barriers that regulate the composition of the intracellular environment and facilitate communication processes essential for cellular function and survival. In comparison to lipid membranes, artificial polymeric membranes generally offer enhanced stability due to their higher molecular weight and greater variability in the nature of the macromolecular building blocks, which provides access to a broad chemistry toolbox to regulate important features such as fluidity and permeability. We recently developed an artificial cell platform based on a complex coacervate, in which a terpolymer, composed of a hydrophilic poly(ethylene glycol) segment, a hydrophobic poly(caprolactone-g-trimethylene carbonate) domain and a polyglutamate anchor (PEG-PCLgTMC-PGA) was used for stabilization. These membranized structures showed excellent permeability, due to the high fluidity of the membrane. However, the polymer membrane proved to be unselective with regard to the molecular weight of guest molecules that were exchanged with the environment. To advance this platform, a series of terpolymers with distinctive features were synthesized to further refine their regulatory features of the polymer membrane. Through investigation of structural terpolymer variants, including those in which the hydrophobic domain was based on PCLgTMC, poly(D,L-lactic acid) or polystyrene, their influence on membrane permeability, fluidity, and sequestration of hydrophobic molecules, such as cholesterol, was determined. With this extended range of membrane-forming building blocks, this coacervate platform is equipped with tailored properties and facilitates sequestration of hydrophobic molecules into the membrane and controlled fluidity.

Structure Studies of Micelle-Enhanced Polyelectrolyte Microcapsules

Abstract:Hybrid calcium carbonate particles, synthesized III the presence of block copolymer polystyrene-b-poly(acrylic acid) (PS-b-PAA), were used to fabricate polyelectrolyte (PE) such as polylallyamine hydrochloride)/ poly(strene sulfonate sodium) (PAH/PSS) and poly (diallyldimethyl ammonium chloride)/poly (styrene sulforate sodium) (PADH/PSS) Capsules, by layer-by-layer (LbL) technique. Novel micelle-enhanced polyelectrolyte (PE capsules were thus obtained. The structure of the micelle-enhanced PE microcapsules was studied in this article. Due to the presence of a large amount of negatively charged micelles inside PE capsule interior, which were liberated from templates, the capsule wall was tightly re-constructed. Different from the wall properties of the conventional PE capsules, the wall of the micelle-enhanced PE capsules showed stability under an extensive pH range from I to 14, but the micelled enhanced capsules showed dented morphology in solutions with high NaCl concentration such as 0.5 mol/L and I mol/L. The state of the block copolymer in the capsules was investigated in dialysis bag by simulating the Solution condition of the capsule interior after the template being removed. It was found that the micelle size of the block copolymer In solution of templates dissolution was tens of nanometers after the solution was dialyzed for 24 h. With the dialysis time increasing up to at least 72 It, the block copolymer began to precipitate onto the dialysis bag from the solution The interaction between the 2-D polyelectrolyte multilayer film and micelles formed by block copolymer in templates solution was studied by UV-Vis measurements as well. It was found that PE multilayer could interact with micelles via electrostatic force. At the same time I the block copolymer remained in the capsules interior could be precipitated onto the PE multilayer via dialyzing in deionized water for a long time (pH similar to 6). A micelle-related layer was formed which was stabilized by the "bridge effect" of calcium ions. That is, the wall of the micelle-enhanced PE capsules had a bilayer structure and was formed by the polyelectrolyte multilayer and micelle-related layer. It seemed that the two layers with different properties clung to each other via electrostatic force. The formation mechanism of the bilayer structure was well illuminated oil the aforementioned studies. This is a key property of the micelle-enhanced PE capsules with which all the properties of the capsules were linked. Based on the bilayer structure of the capsules, the morphology changes of the capsules in solutions with different conditions such as high NaCl concentrations could be elucidated well. However, more direct proofs and further studies were needed to completely resolve the fine structure of the micelle-enhanced PE capsules.

Abstract:Block copolymers (BCPs) have the capacity to self-assemble into a myriad of well-defined aggregate structures, offering great promise for the construction of drug delivery, photolithographic templates, and complex nanoscale assemblies. A uniqueness of these materials is their propensity to become kinetically frozen in non-equilibrium states, implying that the process of self-assembly can be utilized to remodel the resulting structures. Here, a new semiconfined system for processing the BCP self-assembly is constructed, in which an unusual dual-phase separation occurs, including nonsolvent-induced microphase separation and osmotically driven macrophase separation, ultimately yielding heterogeneous BCP membranes. These membranes with cellular dimensions show unique anisotropy that can be used for cell encoding and patterning, which are highly relevant to biology and medicine. This processing method not only provides new levels of tailorability to the structures and encapsulated contents of BCP assemblies, but can also be generalized to other block polymers, particularly those with attractive electronic and/or optical properties.

Abstract:Amphiphilic block copolymers with a sitosteryl methacrylate‐based hydrophobic part and poly(carboxyethyl acrylate) for the hydrophilic segment are synthesized. These block copolymers, combined with soybean L‐α‐phosphatidylcholine, form small and giant hybrid vesicles with different properties compared to the corresponding liposomes with molecular sitosterol, advancing the understanding of integrating natural and synthetic compounds in hybrid membranes. Amphiphilic block copolymer and lipids can be assembled into hybrid vesicles (HVs), which are an alternative to liposomes and polymersomes. Block copolymers that have either poly(sitostryl methacrylate) or statistical copolymers of sitosteryl methacrylate and butyl methacrylate as the hydrophobic part and a poly(carboxyethyl acrylate) hydrophilic segment are synthesized and characterized. These block copolymers assemble into small HVs with soybean L‐α‐phosphatidylcholine (soyPC), confirmed by electron microscopy and small‐angle X‐ray scattering. The membrane's hybrid nature is illustrated by fluorescence resonance energy transfer between labeled building blocks. The membrane packing, derived from spectra when using Laurdan as an environmentally sensitive fluorescent probe, is comparable between small HVs and the corresponding liposomes with molecular sitosterol, although the former show indications of transmembrane asymmetry. Giant HVs with homogenous distribution of the block copolymers and soyPC in their membranes are assembled using the electroformation method. Abstract:Polymer-supported phospholipid bilayers (PLBs) are popular model systems for the study of transmembrane proteins under conditions close to cellular membrane environments. In this work, by combining the techniques of dissipative quartz crystal microbalance and atomic force microscopy, we investigate the deposition of vesicles on a hydrated cationic poly(diallyldimethylammonium chloride) (PDDA) layer as a function of phospholipid composition and sodium chloride concentration. The vesicles used consist of phospholipid mixtures with varying amounts of net negative charge. Uniform PLBs are formed by either increasing the negative charge density of the vesicles or decreasing sodium chloride concentration, suggesting that the electrostatic attraction between the vesicle and PDDA layer is the driving force for the formation of the PLBs. Our results indicate that the PLB formation is a fast adsorption-rupture process of the vesicles, without passing through a critical vesicle density. We further contend that the fluctuating PDDA support plays a central role for this process. This work provides a framework for understanding the key factors that influence the formation of PLBs.
Design, fabrication, and characterization of archaeal tetraether free-standing planar membranes in a PDMS- and PCB-based fluidic platform

Abstract:The polar lipid fraction E (PLFE) isolated from the thermoacidophilic archaeon Sulfolobus acidocaldarius contains exclusively bipolar tetraether lipids, which are able to form extraordinarily stable vesicular membranes against a number of chemical, physical, and mechanical stressors. PLFE liposomes have thus been considered appealing biomaterials holding great promise for biotechnology applications such as drug delivery and biosensing. Here we demonstrated that PLFE can also form free-standing "planar" membranes on micropores (∼100 μm) of polydimethylsiloxane (PDMS) thin films embedded in printed circuit board (PCB)-based fluidics. To build this device, two novel approaches were employed: (i) an S1813 sacrificial layer was used to facilitate the fabrication of the PDMS thin film, and (ii) oxygen plasma treatment was utilized to conveniently bond the PDMS thin film to the PCB board and the PDMS fluidic chamber. Using electrochemical impedance spectroscopy, we found that the dielectric properties of PLFE planar membranes suspended on the PDMS films are distinctly different from those obtained from diester lipid and triblock copolymer membranes. In addition to resistance (R) and capacitance (C) that were commonly seen in all the membranes examined, PLFE planar membranes showed an inductance (L) component. Furthermore, PLFE planar membranes displayed a relatively large membrane resistance, suggesting that, among the membranes examined, PLFE planar membrane would be a better matrix for studying channel proteins and transmembrane events. PLFE planar membranes also exhibited a sharp decrease in phase angle with the frequency of the input AC signal at ∼1 MHz, which could be utilized to develop sensors for monitoring PLFE membrane integrity in fluidics. Since the stability of free-standing planar lipid membranes increases with increasing membrane packing tightness and PLFE lipid membranes are more tightly packed than those made of diester lipids, PLFE free-standing planar membranes are expected to be considerably stable. All these salient features make PLFE planar membranes particularly attractive for model studies of channel proteins and transmembrane events and for high-throughput drug screening and artificial photosynthesis. This work can be extended to nanopores of PDMS thin films in microfluidics and eventually aid in membrane-based new lab-on-a-chip applications.
Adsorption and fusion of hybrid lipid/polymer vesicles onto 2D and 3D surfaces

Abstract:We investigated the formation of hybrid lipid/polymer (1,2-dioleoyl-sn-glycero-3-phosphocholine and poly(ethylene oxide-b-butadiene); DOPC/EO22Bd37) films onto planar silica surfaces. Using laser scanning confocal microscopy, atomic force microscopy, and quartz crystal microbalance analysis, we monitored the adsorption and fusion of hybrid lipid/polymer vesicles onto planar borosilicate glass cleaned via chemical etching or RF/air plasma treatment. In addition we used cryo-electron microscopy to characterize film formation on mesoporous silica nanoparticles. As the polymer content in the vesicles increased, the resulting hybrid lipid/polymer films on borosilicate glass - cleaned by chemical etching or plasma treatment - were more heterogeneous, indicating a large number of adsorbed vesicles rather than continuous bilayer films at higher polymer loadings. The observed lateral fluidity of both DOPC and hybrid lipid/polymer films also decreased substantially with increasing polymer fraction and was found to be relatively insensitive to changes in pH. Films prepared from vesicles with higher polymer loadings were completely immobile. We also found that polymer vesicles did not interact with clean plasma-treated glass surfaces, which may be due to elevated OH and Si-OH on plasma-treated surfaces. Conformal hybrid lipid/polymer coatings consistent with bilayers could be formed on mesoporous silica nanoparticles and imaged via cryo-electron microscopy. These results expand the library of biocompatible materials that can be used for coating silica-based materials and nanoparticles.

Abstract:Hybrid phospholipid/block copolymer membranes where polymers and lipids are molecularly mixed or phase-separated into polymer-rich and lipid-rich domains are promising drug delivery materials. Harnessing the chemical diversity of polymers and the biocompatability of lipids is a compelling approach to design the next generation of drug carriers. Here, we report on the development of a microfluidics-based strategy analogous to produce lipid nanoparticles (LNPs) for the nanomanufacturing of multilayered hybrid nanoparticles (HNPs). Using X-ray scattering, Cryo-electron, and polarized microscopy we show that phosphatidylcholine (PC) and PBD-b-PEO (poly(butadiene-block-ethylene oxide)) hybrid membranes can be nanomanufactured by microfluidics into HNPs with dense and multilayered cores which are ideal carriers of low-solubility drugs of the Biopharmaceutical Classification System (BCS) II and IV such as antimalarial DSM265 and Paclitaxel, respectively.
Experimental validation of a phase-field model to predict coarsening dynamics of lipid domains in multicomponent membranes

Abstract:<p>Membrane phase-separation is a mechanism that biological membranes often use to locally concentrate specific lipid species in order to organize diverse membrane processes. Phase separation has also been explored as a tool for the design of liposomes with heterogeneous and spatially organized surfaces. These "patchy" liposomes are promising platforms for delivery purposes, however their design and optimization through experimentation can be expensive and time-consuming. We developed a computationally efficient method based on the surface Cahn–Hilliard phase-field model to complement experimental investigations in the design of patchy liposomes. The method relies on thermodynamic considerations to set the initial state for numerical simulations. We show that our computational approach delivers not only qualitative pictures, but also accurate <em>quantitative</em> information about the dynamics of the membrane organization. In particular, the computational and experimental results are in excellent agreement in terms of lipid domain area fraction, total lipid domain perimeter over time and total number of lipid domains over time for two different membrane compositions (DOPC:DPPC with a 2:1 M ratio with 20% Chol and DOPC:DPPC with a 3:1 M ratio with 20% Chol). Thus, the computational phase-field model informed by experiments has a considerable potential to assist in the design of liposomes with spatially organized surfaces, thereby containing the cost and time required by the design process.</p>
Lipid domain coarsening and fluidity in multicomponent lipid vesicles: A continuum based model and its experimental validation

Abstract:Liposomes that achieve a heterogeneous and spatially organized surface through phase separation have been recognized to be a promising platform for delivery purposes. However, their design and optimization through experimentation can be expensive and time-consuming. To assist with the design and reduce the associated cost, we propose a computational platform for modeling membrane coarsening dynamics based on the principles of continuum mechanics and thermodynamics. This model couples phase separation to lateral flow and accounts for different membrane fluidity within the different phases, which is known to affect the coarsening dynamics on lipid membranes. The simulation results are in agreement with the experimental data in terms of liquid ordered domains area fraction, total domains perimeter over time and total number of domains over time for two different membrane compositions (DOPC:DPPC with a 1:1 molar ratio with 15% Chol and DOPC:DPPC with a 1:2 molar ratio with 25% Chol) that yield opposite and nearly inverse phase behavior. This quantitative validation shows that the developed platform can be a valuable tool in complementing experimental practice.
