Abstract:Summary Plant can recruit beneficial microbes to enhance their ability to resist disease. Selenium is well established as a beneficial element in plant growth, but its role in mediating microbial disease resistance remained poorly understood. Here, we investigated the correlation between selenium, oilseed rape rhizosphere microbes and Sclerotinia sclerotiorum. Soil application of 0.5 and 1.0 mg/kg selenium significantly increased the resistance of oilseed rape to Sclerotinia sclerotiorum compared with no selenium application, and the disease inhibition rate was higher than 20%. The disease resistance of oilseed rape was related to rhizosphere microorganisms, and beneficial bacteria isolated from the rhizosphere inhibited Sclerotinia stem rot. Burkholderia cepacia, and synthetic community enhanced plant disease resistance through transcriptional regulation and activated plant-induced systemic resistance to protect plants. Besides, inoculation of isolated bacteria optimized the bacterial community structure of leaves and enriched beneficial microorganisms such as Bacillus, Pseudomonas and Sphingomonas. Bacillus isolated from the leaves were sprayed on the detached leaves, and it also performed a significant inhibition effect on Sclerotinia sclerotiorum. Overall, our results suggested that selenium drive plant rhizosphere microorganisms to increase resistance to Sclerotinia sclerotiorum in oilseed rape.

Seed Film Coating with Uniconazole Improves Rape Seedling Growth in Relation to Physiological Changes under Waterlogging Stress

Abstract:Waterlogging is an important constraint of global production of rape. The effects of seed film coating with 0.0075% uniconazole on the growth and physiology was investigated using seedlings from three varieties of rape (Brassica napus L.) subjected to waterlogging. While seed coating with uniconazole had no significant effect on germination percentage during waterlogging stress, it enhanced root vigour, increased root length, root volume and root dry weight. It also significantly enhanced leaf dry weight and ratio of root to shoot, but induced a significant decrease in shoot height and stem dry weight. Seed film coating with uniconazole also significantly increased the activities of the antioxidant enzymes, POD, CAT and SOD, and soluble sugar concentration during waterlogging. Thus, it suggests that seed film coating with uniconazole at a suitable concentration can improve rape seedling growth and increase seedling establishment during waterlogging.

Abstract:Ascorbic acid (AsA) and selenium nanoparticles (SeNPs) were versatile plant growth regulators, playing multiple roles in promoting plant growth under heavy metal stresses. This study aimed to evaluate the beneficial role of individual and combined effects of AsA and SeNPs on morpho-physio-biochemical traits of rice with or without chromium (Cr) amendment. The results indicated that Cr negatively affected plant biomass, gas exchange parameters, total soluble sugar, proline, relative water contents, and antioxidant-related gene expression via increasing reactive oxygen species (MDA, H2O2, O2•−) formation, resulting in plant growth reduction. The application of AsA and SeNPs, individually or in combination, decreased the uptake and translocation of Cr in rice seedlings, increased seedlings with tolerance to Cr toxicity, and significantly improved the rice seedling growth. Most notably, AsA + SeNP treatment strengthened the antioxidative defense system through ROS quenching and Cr detoxification. The results collectively suggested that the application of AsA and SeNPs alone or in combination had the potential to alleviate Cr toxicity in rice and possibly other crop species.
Se Ameliorates Cd Toxicity in Oilseed rape (Brassica napus L.) Seedlings by Inhibiting Cd Transporter Genes and Maintaining root Plasma Membrane Integrity

Abstract:Se (Selenium) has been reported to be an important protective agent to decreases Cd (Cadmium) induced toxic in plants. However, it remains unclear how Se mitigates the uptake of Cd and increased the resistance to Cd toxicity. Hydroponic experiments were arranged to investigate the changes of physiological properties, root cell membrane integrity and Cd-related transporter genes in rape seedlings. Comparison of the biomass between the addition of Se and the absence of Se under Cd exposure showed that the Cd-induced growth inhibition of rape seedlings was alleviated by Se. Cd decreased the photosynthetic rate ( Pn ), stomatal conductance ( Gs ) and photosynthetic pigment content including chlorophyll a, chlorophyll b and carotenoid. However, all these parameters were all significantly improved by Se addition. Moreover, exposure to Se resulted in a decrease in Cd concentration in both shoot and root, ranging from 4.28 to 27.2%. Notably, the application of Se at a concentration of 1 μmol L − 1 exhibited the best performance. Furthermore, Se enhanced cell membrane integrity and reduced superoxide anion levels, thereby contributing to the alleviation of cadmium toxicity in plants. More critically, Se decreased the expression levels of root Cd-related transporter genes BnIRT1 , BnHMA2 and BnHMA4 under Cd stress, which are responsible for Cd transport and translocation. These results are important to increase crop growth and reduce Cd load in the food chain from metal toxicity management and agronomical point of view.

Abstract:Selenium (Se) is an essential micro-element for human and animals. In higher plants, Se essentiality or phyto-toxicity is less explored. Therefore, we aimed to examine the effects of Se (0, 25, 50, and 100 µM) as sodium selenite on the physio-chemical, cell ultra-structural and genomic alterations in hydroponically grown seedlings of four cultivars of B. napus (cvs. Zheda 619, Zheda 622, ZS 758, and ZY 50). Results showed that excessive (100 µM) Se (IV) exhibited significant reduction in plant growth parameters, declined pigment contents, lower water-soluble protein levels, and overproduction of H2O2 and MDA contents. A significant increase in antioxidant enzyme activities and transcript levels of superoxide dismutase (SOD), peroxidase (POD), ascorbate peroxidase (APX), and glutathione reductase (GR), except catalase (CAT) were noticed in the leaves and roots. Non-enzymatic antioxidants including glutathione (GSH) and oxidized glutathione (GSSG), except GSSG in roots were enhanced under higher Se (IV) levels. Transmission electron microscopy analysis revealed the ultrastructural damages in leaf mesophyll and root tip cells induced by excessive Se (IV). Less-significant phytotoxic effects were observed in above-mentioned parameters at 50 µM Se (IV). Overall, Se (IV) supplementation at 25 µM displayed marginal beneficial effect by enhancing plant growth, pigment contents, protein levels and restrict H2O2 and MDA overproduction. A marginal increase/decrease in ROS-detoxifying enzymes (except CAT activity) and elevated GSH and GSSG levels were noticed. The accumulation of Se (IV) was much higher in roots as compared to leaves. This accumulation was maximum in Zheda 622 and minimum in ZS 758, followed by Zheda 619 and ZY 50. Overall findings showed that Zheda 622 was the most sensitive and ZS 758 as most tolerant to Se (IV) phyto-toxicity. In addition, Se (IV) was found beneficial until 25 µM Se (IV) but phytotoxic at higher Se levels especially at 100 µM Se (IV).

Abstract:As an effective neonicotinoid insecticide, imidacloprid (IMI) has been widely used in crop production, but its residue affects normal plant growth. Selenium (Se) is a non-essential mineral nutrient in higher plants, that acts as the active centre of glutathione peroxidase (GSH-Px), which removes harmful peroxides. In this study, we investigated the mechanism by which selenium improves the growth status of IMI-treated garlic plants through analyses of apparent morphology and antioxidant enzyme activity as well as the dynamic changes in nutrients and metabolites in the plants. The results showed that 80 μg/kg Na2SeO3 had a strong effect on alleviating the damage in garlic plants exposed to IMI (1.2 mg/kg) by increasing the absorption of mineral elements to enhance the synthesis of chlorophyll and antioxidant enzymes. A nontarget metabolomics analysis based on gas chromatography-mass spectrometry (GC-MS) indicated that the addition of Na2SeO3 to IMI-treated garlic could reconstruct the plant metabolic distribution by enhancing the nitrogen and indole metabolism, maintaining lower concentrations of secondary metabolites and maintaining the balance of the plant energy metabolism. Our study provides novel insights into the molecular mechanisms by which garlic plants responds to IMI exposure and suggests the use of selenium with IMI-contaminated plants as a solution for the advancement of sustainable agricultural pesticide use.
Selenium mitigates the chromium toxicity in Brassicca napus L. by ameliorating nutrients uptake, amino acids metabolism and antioxidant defense system

Abstract:The phytotoxicity of chromium (Cr) makes it obligatory for the researchers to develop strategies that seek to hinder its accumulation in food chains. While, protective role of selenium (Se) has not been discussed in detail under adverse conditions in oilseed rape. Here, our aim was to investigate the potential use of Se (0, 5 and 10 μM) in alleviating the Cr toxicity (0, 100 and 200 μM) in Brassica napus L. Results delineated that Se-supplementation notably recovered the Cr-phytotoxicity by reducing the Cr accumulation in plant tissues and boosted the inhibition in plant growth and biomass. Under Cr stress, the exogenously applied Se significantly recovered the impairment in photosynthesis related parameters (chlorophyll a, chlorophyll b, carotenoids, net photosynthetic rate, stomatal conductance, and photochemical efficiency of photosystem II), and counteracted the reduction in nutrients uptake and improved the essential amino acids (EAAs) levels. In addition, Se activated the antioxidants enzymes included in AsA-GSH cycle (SOD, CAT, APX, GR, DHAR, MDHAR, GSH, and AsA) and glyoxalase (Gly) system (Gly I and Gly II) and minimized the excessive generation of reactive oxygen species (ROS) and methylglyoxal (MG) contents in response to Cr stress. In a nutshell, Se (more effective at 5 μM) alleviated the Cr and MG induced phytotoxicity and oxidative damages by minimizing their (Cr and MG) accumulation and enhanced the plant growth, nutrients element level, nutrition quality by improving EAAs, antioxidant and Gly system. By considering the above-mentioned biomarkers, the addition of exogenous Se in Cr polluted soils might be effective approach to decrease the Cr uptake and its linked phytotoxicity in B. napus.
Effects of Selenium Application on Reducing Cadmium Uptake and Ameliorates Cadmium Stress on Oilseed Rape (brassica Napus L.) in Cadmium-Contaminated Soil

Abstract:Cadmium is considered a highly toxic metallic element that does not have any beneficial biological functions for humans or plants. It has been reported that the antagonism of selenium to heavy metal stress has been observed in a variety of plants, and appropriate selenium could alleviate heavy metal-induced oxidative damage and reduce the accumulation of heavy metals in plants. The changes of physiological characteristics, root tip cells, cadmium concentration and accumulation of rape under cadmium stress were investigated in this study through pot experiment. Results showed that selenium could alleviate the inhibitory effect of cadmium on the growth of rape seedlings. The concentration and accumulation of cadmium were decreased after the selenium application in rape seeds, ranging from 19.93 to 22.97% and 27.96 to 43.88% respectively. And the decrease of photosynthetic pigment content induced by cadmium was significantly improved. The results of transmission electron microscopy showed that exogenous selenium and cadmium had metal complexation reaction and formed black precipitation, which may be related to the detoxification effect of selenium on cadmium. More critically, with the addition of selenium, the plasma membrane damage and free radical accumulation in root tips induced by cadmium stress were gradually alleviated in the histochemical staining experiment of rape root tips. These results may provide evidence for exploring effective measures to reduce cadmium accumulation in rape under cadmium-contaminated areas.
Components of a Rice-Oilseed Rape Production System Augmented with Trichoderma sp. Tri-1 Control Sclerotinia sclerotiorum on Oilseed Rape

Abstract:Sclerotinia sclerotiorum causes serious yield losses on many crops throughout the world. A multicomponent treatment that consisted of the residual rice straw remaining after rice harvest and Trichoderma sp. Tri-1 (Tri-1) formulated with the oilseed rape seedcake fertilizer was used in field soil infested with S. sclerotiorum. This treatment resulted in oilseed rape seed yield that was significantly greater than the nontreated control or when the fungicide carbendizem was used in the presence of this pathogen in field trials. Yield data suggested that the rice straw, oilseed rape seedcake, and Tri-1 components of this treatment all contributed incrementally. Similar treatment results were obtained regarding reduction in disease incidence. Slight improvements in yield and disease incidence were detected when this multicomponent treatment was combined with a fungicide spray. Inhibition of sclerotial germination by this multicomponent treatment trended greater than the nontreated control at 90, 120, and 150 days in field studies but was not significantly different from this control. This multicomponent treatment resulted in increased yield relative to the nontreated control in the absence of pathogen in a greenhouse pot study, while the straw alone and the straw plus oilseed rape seedcake treatments did not; suggesting that Tri-1 was capable of promoting growth. Experiments reported here indicate that a treatment containing components of a rice-oilseed rape production system augmented with Tri-1 can control S. sclerotiorum on oilseed rape, be used in integrated strategies containing fungicide sprays for control of this pathogen, and promote plant growth.
Nano-selenium promotes the product quality and plant defense of Salvia miltiorrhiza by inducing tanshinones and salvianolic acids accumulation

Abstract:Selenium (Se) is a non-essential element for plants, but it has dosage effects on plant's life in some environments. Although many studies have revealed the beneficial effects of Se on the plant growth and defense of grains, fruits and vegetables, the underlying mechanism is largely unknown. Here, the effects of nanoparticle Se (Nano-Se) on Salvia miltiorrhiza , a valuable traditional Chinese medicine, were described. Ten mg/L Nano-Se was the optimum concentration for foliar application to stimulate plant growth, antioxidant capacity, and the accumulation of tanshinones and salvianolic acids. With the increase in the duration of Nano-Se exposure, plant biomass, nutrients and tanshinones contents gradually increased, while the salvianolic acids contents first increased significantly and then decreased. Transcriptomic analysis showed that the promotion effects of Nano-Se were probably due to the activation of photosynthesis, plant hormone signal transduction and antioxidant response in S. miltiorrhiza . Nano-Se might increase the contents of tanshinones and salvianolic acids by activating the salicylic acid (SA) and jasmonic acid (JA) signaling pathways and the downstream APETALA2/ethylene-responsive element binding factors (AP2/ERF) and WRKY transcription factors. Insect bioassays revealed that Nano-Se could reduce the survival and fecundity of aphids on S. miltiorrhiza by inducing the contents of salvianolic acid B (SalB) and rosmarinic acid (RA). Thus, these results indicate that Nano-Se could promote the crop yield, product quality and pest resistance of S. miltiorrhiza by inducing plant biomass and secondary metabolites. Our study suggests potential applications of Nano-Se in the production of industry crops.
Isolation and evaluation of Bacillus subtilis RSS-1 as a potential biocontrol agent against Sclerotinia sclerotiorum on oilseed rape

Abstract:Sclerotinia stem rot caused by Sclerotinia sclerotiorum is one of the most important diseases of oilseed rape in the world. Because of the absence of resistant varieties and the disadvantages of chemical control, application of antifungal microbes has become an eco-friendly and effective measure to control this disease. In this study, Bacillus subtilis strain RSS-1, isolated from soil samples, was identified based on morphological, physiological and biochemical tests, and DNA gyrase subunit A ( gyrA ), gyrB , DNA-directed RNA polymerase subunit beta ( rpoB ) and rpoC gene sequence analysis. It significantly inhibited mycelial growth and sclerotial production of S. sclerotiorum in vitro . In greenhouse experiments, all three tested concentrations (10 6 , 10 7 , 10 8 cfu mL −1 ) of cell fermentation broth and culture filtrate significantly reduced the severity of sclerotinia stem rot on oilseed rape ( P < 0.05). RSS-1 was more effective at reducing disease severity when applied 24 h before inoculation with S. sclerotiorum than at 24 h post inoculation, suggesting that RSS-1 should be applied as a prophylactic rather than a curative biological agent. Colonization tests indicated that the population density of RSS-1 on rapeseed leaves significantly decreased ( P < 0.05) over 6 days. However, RSS-1 could stably colonize in rhizospheric soil of rapeseed over 30 days. Challenge inoculation tests showed that RSS-1 significantly inhibited the activities of polygalacturonase and cellulase and accumulation of oxalic acid during the S. sclerotiorum infection. These results suggest that RSS-1 was a potential biological agent for controlling sclerotinia stem rot caused by S. sclerotiorum on oilseed rape.

Simultaneously inhibit cadmium and arsenic uptake in rice (Oryza sativa L.) by Selenium enhanced iron plaque: Performance and mechanism

Abstract:Selenium (Se) fortification is witnessed to simultaneously inhibit absorbing Cadmium (Cd) and Arsenic (As) by rice plants, but the mechanism is unclear. Here, the effects of Se on the root morphology, iron plaque (IP) content, soil Fe2+ content, radial oxygen loss (ROL), and enzyme activities of the rice plants in the soil contaminated by Cd and As were intensively investigated through the hydroponic and soil experiments. Se effectively alleviated the toxic effects of Cd and As on the plants and the dry weight, root length, and root width were increased by 203.18%, 33.41%, and 52.81%, respectively. It also elucidated that ROL was one of the key factors to elevate IP formation by Se and the specific pathways of Se enhancing ROL were identified. ROL of the plants in the experiment group treated by Se was increased 36.76%, and correspondingly IP was magnified 50.37%, compared to the groups with Cd and As. It was owing to Se significantly increased the root porosity (62.11%), facilitating O2 transport to the roots. Additionally, Se enhanced the activities of catalase (CAT) and superoxide dismutase (SOD) to promote the catalytic degradation of ROS induced by Cd and As stress. It indirectly increased O2 release in the rhizosphere, which benefit to form more robust IP serve as stronger barrier to Cd and As. The results of our study provide a novel molecular level insight for Se promoting root IP to block Cd and As uptake by the rice plants.
The use of selenium for controlling plant fungal diseases and insect pests

Abstract:The selenium (Se) applications in biomedicine, agriculture, and environmental health have become great research interest in recent decades. As an essential nutrient for humans and animals, beneficial effects of Se on human health have been well documented. Although Se is not an essential element for plants, it does play important roles in improving plants' resistances to a broad of biotic and abiotic stresses. This review is focused on recent findings from studies on effects and mechanisms of Se on plant fungal diseases and insect pests. Se affects the plant resistance to fungal diseases by preventing the invasion of fungal pathogen through positively affecting plant defense to pathogens; and through negative effects on pathogen by destroying the cell membrane and cellular extensions of pathogen inside plant tissues after invasion; and changing the soil microbial community to safeguard plant cells against invading fungi. Plants, grown under Se enriched soils or treated with Se through foliar and soil applications, can metabolize Se into dimethyl selenide or dimethyl diselenide, which acts as an insect repellent compound to deter foraging and landing pests, thus providing plant mediated resistance to insect pests; moreover, Se can also lead to poisoning to some pests if toxic amounts of Se are fed, resulting in steady pest mortality, lower reproduction rate, negative effects on growth and development, thus shortening the life span of many insect pests. In present manuscript, reports are reviewed on Se-mediated plant resistance to fungal pathogens and insect pests. The future perspective of Se is also discussed on preventing the disease and pest control to protect plants from economic injuries and damages.

Abstract:Straw amendment significantly enhances mercury (Hg) methylation and subsequent methylmercury (MeHg) bioaccumulation in Hg-contaminated paddy fields by releasing dissolved organic matter (DOM). This study comprehensively investigates the regulatory mechanisms of DOM and its different molecular weights derived from sulfur-rich rape straw (RaDOM) and composted rape straw (CRaDOM) applied in the rice-filling stage on soil MeHg production and subsequent bioaccumulation in rice grains. The results indicated that the amendment of RaDOM and CRaDOM significantly reduced soil MeHg content by 42.40–62.42%. This reduction can be attributed to several factors, including the suppression of Hg-methylating bacteria in soil, the supply of sulfate from RaDOM and CRaDOM, and the increase in the humification, molecular weight, and humic-like fractions of soil DOM. Additionally, adding RaDOM increased the MeHg bioaccumulation factor in roots by 27.55% while inhibiting MeHg transportation by 12.24% and ultimately reducing MeHg content in grains by 21.24% compared to the control group. Similarly, CRaDOM enhanced MeHg accumulation by 25.19%, suppressed MeHg transportation by 39.65%, and reduced MeHg levels in the grains by 27.94%. The assimilation of sulfate derived from RaDOM and CRaDOM into glutathione may be responsible for the increased retention of MeHg in the roots. Over the three days, there was a significant decrease in soil MeHg content as the molecular weight of RaDOM increased; conversely, altering the molecular weight of CRaDOM demonstrated an inverse trend. However, this pattern was not observed after 12 days. Applying sulfur-rich rape DOM can help mitigate MeHg accumulation in paddy fields by regulating the quality of soil DOM, sulfur cycling, and Hg-methylating bacteria.

The Use of Pseudomonas Fluorescens P13 to Control Sclerotinia Stem Rot (sclerotinia Sclerotiorum) of Oilseed Rape

Abstract:Sclerotinia stem rot (SSR) caused by the fungus Sclerotinia sclerotiorum has been an increasing threat to oilseed rape ( Brassica napus L.) cultivation. Efficient and environment—friendly treatments are much needed. Here we focus on microbial control. The Pseudomonas fluorescens P13 that was isolated from oilseed rape cultivation soil, proved to be a useful biocontrol strain for application. Morphology, physiological and biochemical tests and 16S rDNA analysis demonstrated that it was P. fluorescens P13 and that it had a broad antagonistic spectrum, significantly lessening the mycelial growth of S. sclerotiorum by 84.4% and suppressing sclerotial formation by 95–100%. Scanning electron microscopy studies attested that P13 deformed S. sclerotiorum mycelia when they were cultured together. P13 did not produce chitinase but did produce hydrogen cyanide (HCN) which was likely one of the antagonistic mechanisms. The density of P13 remained at a high level (≥10 6 CFU/ml) during 5 weeks in the rhizosphere soil and roots. P13 reduced SSR severity at least by 59% in field studies and also promoted seedling growth ( p <0.05) at the seedling stage. From these data, our work provided evidence that P13 could be a good alternative biological resource for biocontrol of S. sclerotiorum .
Effect of selenium on macro- and microelement distribution and physiological parameters of rape and wheat seedlings exposed to cadmium stress

Abstract:Cadmium (Cd) exerts a detrimental effect on the metabolism of plants, whereas selenium (Se) may protect them against various stressors through its antioxidative activity. In this in vitro study we investigated the impact of Se (2 µM Na2SeO4) on the growth, nutrient (P, S, K, Ca, Mg, B, Mn, Fe and Zn) concentrations and cell integrity of rape (Brassica napus oleifera) and two wheat (Triticum aestivum) genotypes subjected to Cd stress (600 µM CdCl2). Rape accumulated both Cd and Se more than did wheat. In all plants, Cd markedly reduced the biomass, enhanced lipid peroxidation and diminished plasmalemma fluidity. A drop in the K uptake and the reduced plasmalemma permeability diminished the K efflux from the leaf cells. In contrast, Cd elevated S concomitantly with Zn, indicating an activity of detoxifying SH groups and SOD isoenzymes. When added alone, Se promoted the growth of all plants, it enhanced the accumulation of S, but the impact on other nutrients remained minor. In Cd-stressed plants, Se tended to counterbalance the Cd-induced changes in nutrients, it also reduced the lipid peroxidation and exerted positive effects on the cell membrane stability. The Cd stress and the protective role of Se were most evident in rape. The Finnish wheat genotype was less tolerant to Cd than the Polish one.

Abstract:Abstract Selenium nanoparticles (SeNPs) have attracted considerable attention globally due to their significant potential for alleviating abiotic stresses in plants. Accordingly, further research has been conducted to develop nanoparticles using chemical ways. However, our knowledge about the potential benefit or phytotoxicity of bioSeNPs in rapeseed is still unclear. Herein, we investigated the effect of bioSeNPs on growth and physiochemical attributes, and selenium detoxification pathways compared to sodium selenite (Se (IV)) during the early seedling stage under normal and salt stress conditions. Our findings showed that the range between optimal and toxic levels of bioSeNPs was wider than Se (IV), which increased the plant’s ability to reduce salinity-induced oxidative stress. BioSeNPs improved the phenotypic characteristics of rapeseed seedlings without the sign of toxicity, markedly elevated germination, growth, photosynthetic efficiency and osmolyte accumulation versus Se (IV) under normal and salt stress conditions. In addition to modulation of Na + and K + uptake, bioSeNPs minimized the ROS level and MDA content by activating the antioxidant enzymes engaged in ROS detoxification by regulating these enzyme-related genes expression patterns. Importantly, the main effect of bioSeNPs and Se (IV) on plant growth appeared to be correlated with the change in the expression levels of Se-related genes. Our qRT-PCR results revealed that the genes involved in Se detoxification in root tissue were upregulated upon Se (IV) treated seedlings compared to NPs, indicating that bioSeNPs have a slightly toxic effect under higher concentrations. Furthermore, bioSeNPs might improve lateral root production by increasing the expression level of LBD16 . Taken together, transamination and selenation were more functional methods of Se detoxification and proposed different degradation pathways that synthesized malformed or deformed selenoproteins, which provided essential mechanisms to increase Se tolerance at higher concentrations in rapeseed seedlings. Current findings could add more knowledge regarding the mechanisms underlying bioSeNPs induced plant growth. Graphical Abstract

Abstract:1. We have investigated the differences between the nitric oxide synthase inhibitor L-NMMA, the guanylate cyclase inhibitor methylene blue and the potassium channel blockers apamin and charybdotoxin or apamin and iberiotoxin, in their abilities to increase vasoconstrictor responses in rat small mesenteric arterial rings. 2. When administered during the maintained contraction to PGF2alpha (10 microM), L-NMMA (100 microM) or the combination of apamin (0.7 microM) and charybdotoxin (0.1 microM) significantly increased the contractile response. Methylene blue (10 microM) increased the contraction, but this did not reach significance. However, apamin (0.7 microM) and iberiotoxin (0.1 microM) also significantly increased the contractile response. 3. The combination of L-NMMA or methylene blue with apamin/charybdotoxin produced significantly greater increases in the contractile response to PGF2alpha than achieved individually. 4. Relaxations to acetylcholine (10 microM) were significantly reduced by L-NMMA or methylene blue but not by apamin in combination with charybdotoxin or iberiotoxin. 5. Since apamin/iberiotoxin had similar effects to apamin/charybdotoxin, it is likely that the actions of these agents involve direct actions on smooth muscle potassium channels rather than inhibition of endothelium-derived hyperpolarising factor (EDHF). These results suggest that endothelium-derived nitric oxide but not EDHF has a major role in modulating vascular tone under these conditions.

Abstract:Selenium (Se) is an essential microelement for humans and a beneficial element for plants. Recently, biofortification with Se has emerged as a key strategy to increase crop Se content. Nonetheless, Se species matters a lot as inorganic Se species is mostly toxic to human health. In this study, we investigated the effects of different forms and mode of Se application on Se accumulation and speciation in rice. The results showed that root application of Se remarkably increased Se accumulation, photosynthetic rate, biomass accumulation and tolerance to cadmium stress in rice as compared to foliar application. However, the stimulatory effects of Se varied depending on the Se species used for root feeding. At vegetative stage, root application of Se-(Methyl) selenocysteine caused the highest water extractable Se content in leaves with major contribution from organic Se species such as Se-amino acid and non-amino acid organic Se. Further investigation at reproductive stage revealed that foliar application of sodium selenite (Na2SeO3) resulted in the highest total Se content in rice seeds which was largely attributed to inorganic Se. In contrast, the root application of Na2SeO3 led to the maximum accumulation of organic Se compounds which are advantageous to human health. Moreover, the root application of Se increased antioxidant capacity and selectively enhanced amino acids and essential element content in rice grain. This study deepens our understanding of the Se species in Se-enriched rice and suggests that root application of Se may ensure the safe intake of Se through rice.
