Abstract:This study was designed to evaluate the effect of ractopamine on finishing pigs fed two levels of total lysine (0.81% vs. 1.11%) with and without ractopamine (0 vs. 10 ppm) in a two-way factorial arrangement. Three hundred and twenty crossbred barrows and gilts with body weight of 971.12 kg housed in an environmentally controlled facility were blocked and randomly allotted to the four dietary treatments. Pigs’ weight and feed intake were recorded weekly until the average of the pen reached 115-kg body weight. Ractopamine significantly promoted growth rate (965 vs. 1110 g/day, S.E. =23.3, P<0.01) and improved Gain: Feed ratio (0.28 vs. 0.33, S.E. =0.0067, P<0.01). The improvement in growth rate by ractopamine was 133% higher at high lysine level than low lysine level. In addition, the improvement by ractopamine on growth rate decreased with time (1158 vs. 1441 g/day in week 1, 946 vs 1050 g/day in week 2 and 791 vs 839 g/day in week 3, S.E. =40.3, P=0.014). Average backfat depth was reduced by ractopamine supplementation only when high lysine diet was fed (0.76 vs 0.79 cm at 0.81% lysine and 0.80 vs 0.70 at 1.11% lysine, S.E. =0.025, P<0.01). Average loin depth was significantly increased by the addition of ractopamine (2.79 vs 2.90 cm, S.E. =0.021, P<0.01). Increased lean muscle percentage by ractopamine was only observed in high lysine group (55.2% vs 55.4% at 0.81% lysine and 55.1% vs 56.3% at 1.11% lysine, s.e. =0.22, P=0.026), whereas dressing percentage was significantly increased by ractopamine only in low lysine group (73.7% vs 75.6% at 0.81% lysine and 74.2% vs 74.6% at 1.11% lysine, s.e. =0.31, P=0.021). In conclusion, ractopamine improved growth performance and carcass characteristics in pigs fed the high lysine diet and increased dressing percentage in finishing pigs fed low lysine diet.

Abstract:Abstract Heat stress (HS) compromises farm animal productivity and it is thus both an economic and food security issue. Reduced output (e.g., milk yield, muscle growth, egg production) during HS was traditionally thought to result from decreased nutrient intake (a classic biological response shared by all animals during environmental-induced hyperthermia). Utilizing pair-feeding experimental designs clearly indicate that reduced feed intake only accounts for a portion of the decreased productivity. The etiological epicenter of many (if not most) of the physiological consequences of HS is gastrointestinal tract hyperpermeability. For reasons not clearly understood, HS compromises the intestinal barrier allowing luminal antigens to infiltrate into local and systemic circulation. The invading antigens are recognized and stimulate the immune system. An activated immune system rearranges the hierarchy of nutrient partitioning, and the entire body participates in ensuring that leukocytes receive the fuel and building blocks necessary to mount an effective response. Central to this is increased insulin secretion coupled with simultaneous decreased adipose tissue and skeletal muscle insulin resistance. Activated leukocytes switch their energetic metabolism away from oxidative phosphorylation to aerobic glycolysis. Utilizing a LPS-Euglycemic clamp we have estimated that an intensely activated immune system can use more than 2 kg of glucose/d in lactating cows. We extended this in growing steers and pigs and the amount of glucose used is approximately 1 g/Kg BW0.75/h regardless of species and physiological state. If the heat load is severe enough, the consequences can become lethal. This is primarily explained by inflammation induced hypercoagulation, endothelial barrier dysfunction and disseminated intravascular coagulation. If the animal survives, these changes result in the reprioritization of fuel selection during HS which appears to be primarily responsible for reduced animal productivity during the warm summer months.

Abstract:Recent evidence indicates that the heat stress loss on the growth performance of calves is associated with the diversion of nutrients to control enteritis and systemic inflammation. In this study, we investigated the impact of heat stress on markers of inflammation, feed use-efficiency, and growth of dairy calves. We hypothesized that dexamethasone, which is known for its immunosuppressive and anti-inflammatory properties, would reduce inflammation and restore the growth of calves exposed to heat stress. Thirty-two Holstein bull calves (body weight (BW) 68.5 ± 1.37 kg; age 3.5 ± 0.5-week-old; mean ± SD) were housed in individual pens in climate-controlled rooms at constant ambient temperature and allowed to adjust to facilities for 5 d before the start of treatments. Calves were randomly assigned to one of 4 treatments (n = 8/treatment) in a 2 × 2 factorial arrangement of environment (ENV, thermoneutral or heat stress) and intervention (INT, saline or dexamethasone) imposed for 5 d as follow: 1) thermoneutral (constant ambient temperature of 20°C 24 h/d) and administration of saline, 2) thermoneutral (constant ambient temperature of 20°C 24 h/d) and administration of dexamethasone, 3) cyclic heat stress (40°C ambient temperature, from 0800 to 1900 h/d) and administration of saline, 4) cyclic heat stress (40°C ambient temperature, from 0800 to 1900 h/d) and administration of dexamethasone. Dexamethasone (0.05 mg/kg BW), or saline (1.2 mL) was administered intramuscularly on d 1 and 3. Upon completion of treatments, calves were euthanized on d 5 to obtain jejunum mucosa samples. Commercial milk replacer, starter grain, and water were offered, and intake was monitored daily. Rectal temperature and respiratory rate were monitored 3 times daily. Blood samples were collected on d 1, 3, and 5 to determine serum pro-inflammatory cytokine concentrations. A section of the jejunum was collected and snap-frozen to determine the concentration of pro-inflammatory markers. Statistical analyses included a mixed model, fixed effects of ENV, INT, consecutive measurements taken over time (d, h, or both), replica, and random effects of calf and error (SAS version 9.4, SAS Institute Inc., Cary, NC). The measurements collected immediately before treatment allocation were included as covariates in the model. An ENV effect showed that heat stress increased rectal temperature (38.72 vs. 39.21°C), respiratory rate (36 vs. 108 breaths/min), and water intake (3.2 vs. 6.6 L/d). The treatments did not affect dry matter intake. An ENV × INT interaction showed that heat stress with saline decreased average daily gain (ADG) by 35% and tended to decrease feed use-efficiency by 36%, but the use of dexamethasone to treat heat stress restored ADG and feed use-efficiency comparable to their basal levels. An ENV × INT interaction revealed that heat stress with saline increased jejunal interleukin (IL)-6 concentration 2-fold, but dexamethasone treatment of heat stress restored jejunal IL-6 concentration to basal levels. The bioenergetic cost of the heat stress-immune pro-inflammatory response ranged between 1.18 and 1.50 Mcal of ME. Overall, the administration of dexamethasone reduced the jejunal concentration of a pro-inflammatory marker and restored the heat stress-associated reduction in growth and feed use-efficiency. The immunomodulation and anti-inflammatory effects of dexamethasone could be part of a homeorhetic change that results in a shift from maintenance functions to support growth on calves exposed to heat stress.

Abstract:Alcohol and other substance abuse treatment programs are focusing more on the wellbeing of the total patient rather than just targeting functional sobriety. Various explanations are offered in this article for the inclusion of a fitness and health promotion component in a chemical abuse treatment program. The treatment program at Charter Hospital of Fort Worth, Texas, is discussed in detail, including a comprehensive medical examination, fitness and nutritional screenings, and appropriate exercise activities. Educational sessions are offered on health fitness, risk reduction, stress management, smoking cessation, proper nutrition and the positive psychological aspects of exercise. The implications of a health/fitness component in the recovery process are emphasized.

Abstract:Abstract The objective of this study was to determine impacts on immune parameters, anti-oxidant capacity, and growth of finishing steers fed a Saccharomyces cerevisiae fermentation product (SCFP; NaturSafe; Diamond V, Cedar Rapids, IA) and ractopamine hydrochloride (RAC; Optaflexx; Elanco Animal Health, Greenfield, IN). Angus-crossbred steers (N = 288) from two sources were utilized in this 90-d study. Steers were blocked by source, stratified by initial body weight to pens of six steers, and pens randomly assigned to treatments (16 pens per treatment). Three treatments compared feeding no supplemental SCFP (control; CON) and supplemental SCFP for 57 d (SCFP57), and 29 d (SCFP29) before harvest. Supplementation of SCFP was 12 g per steer per d, and all steers were fed RAC at 300 mg per steer per d for 29 d before harvest. Blood samples were collected from3 steers per pen, and muscle samples were collected from 1 steer per pen at 57, 29 (start of RAC), and 13 (midRAC) days before harvest. Blood was analyzed from 2 steers per pen for ferric reducing anti-oxidant power (FRAP). Muscle gene expression of myokines, markers of anti-oxidant and growth signaling were assessed. Individual animal BW were also collected on 57, 29, 13, and 1 d before being harvested at a commercial facility (National Beef, Tama, IA). Data were analyzed using the Mixed procedure of SAS 9.4 (Cary, NC) with pen as the experimental unit. The model included fixed effects of treatment and group. Increased BW compared to CON was observed days −29, −13, and −1 in SCFP57 steers (P ≤ 0.05), with SCFP29 being intermediate days −13 and −1. Overall G:F was improved in SCFP29 and SCFP57 (P = 0.01). On day −29, FRAP was greater in SCFP57 than CON (P = 0.02). The percent of gamma delta T cells and natural killer cells in both SCFP29 and SCFP57 was greater than CON on day −13 (P = 0.02). There were no treatment × day effects for muscle gene expression measured (P ≥ 0.25). Interleukin 6 tended to decrease in SCFP29 and SCFP57 on day −13 (P = 0.10). No other treatment effects were observed for muscle gene expression. Muscle gene expression of interleukin 15 was increased (P = 0.01), and expression of interleukin 8 was decreased (P = 0.03) due to RAC feeding. Increased growth in SCFP-fed cattle may be related to changes in anti-oxidant capacity and the immune system.

Effects of Rumen-Protected L-Tryptophan Supplementation on Productivity, Physiological Indicators, Blood Profiles, and Heat Shock Protein Gene Expression in Lactating Holstein Cows under Heat Stress Conditions

Abstract:In this study, we examined the effects of rumen-protected L-tryptophan supplementation on the productivity and physiological metabolic indicators in lactating Holstein cows under heat stress conditions. The study involved eight early lactating Holstein cows (days in milk = 40 ± 9 days; milk yield 30 ± 1.5 kg/day; parity 1.09 ± 0.05, p < 0.05), four cows per experiment, with environmentally controlled chambers. In each experiment, two distinct heat stress conditions were created: a low-temperature and low-humidity (LTLH) condition at 25 °C with 35–50% humidity and a high-temperature and high-humidity (HTHH) condition at 31 °C with 80–95% humidity. During the adaptation phase, the cows were subjected to LTLH and HTHH conditions for 3 days. This was followed by a 4-day heat stress phase and then by a 7-day phase of heat stress, which were complemented by supplementation with rumen-protected L-tryptophan (ACT). The findings revealed that supplementation with ACT increased dry matter intake as well as milk yield and protein and decreased water intake, heart rate, and rectal temperature in the HTHH group (p < 0.05). For plateletcrit (PCT, p = 0.0600), the eosinophil percentage (EOS, p = 0.0880) showed a tendency to be lower, while the monocyte (MONO) and large unstained cells (LUC) amounts were increased in both groups (p < 0.05). Albumin and glucose levels were lower in the HTHH group (p < 0.05). The gene expressions of heat shock proteins 70 and 90 in the peripheral blood mononuclear cells were higher in the ACT group (HTHH, p < 0.05). These results suggest that ACT supplementation improved productivity, physiological indicators, blood characteristics, and gene expression in the peripheral blood mononuclear cells of early lactating Holstein cows under heat-stress conditions. In particular, ACT supplementation objectively relieved stress in these animals, suggesting that L-tryptophan has potential as a viable solution for combating heat-stress-induced effects on the cattle in dairy farming.

Effect of Heat Stress on Bovine Mammary Cellular Metabolites and Gene Transcription Related to Amino Acid Metabolism, Amino Acid Transportation and Mammalian Target of Rapamycin (mTOR) Signaling

Abstract:Heat stress (HS) is one of the most serious factors to negatively affect the lactation performance of dairy cows. Bovine mammary epithelial cells are important for lactation. It was demonstrated that HS decreases the lactation performance of dairy cows, partly through altering gene expression within bovine mammary epithelial tissue. However, the cellular metabolism mechanisms under HS remains largely unknown. The objective of this study was to determine whether HS induced changes in intracellular metabolites and gene transcription related to amino acid metabolism, amino acid transportation and the mTOR signaling pathway. Immortalized bovine mammary epithelial cell lines (MAC-T cells, n = 5 replicates/treatment) were incubated for 12 h at 37 °C (Control group) and 42 °C (HS group). Relative to the control group, HS led to a greater mRNA expression of heat shock protein genes HSF1, HSPB8, HSPA5, HSP90AB1 and HSPA1A. Compared with the control group, metabolomics using liquid chromatography tandem–mass spectrometry identified 417 differential metabolites with p 1.0 in the HS group. HS resulted in significant changes to the intracellular amino acid metabolism of glutathione, phenylalanine, tyrosine, tryptophan, valine, leucine, isoleucine, arginine, proline, cysteine, methionine, alanine, aspartate and glutamate. HS led to a greater mRNA expression of the amino acid transporter genes SLC43A1, SLC38A9, SLC36A1, and SLC3A2 but a lower mRNA expression of SLC7A5 and SLC38A2. Additionally, HS influenced the expression of genes associated with the mTOR signaling pathway and significantly upregulated the mRNA expression of mTOR, AKT, RHEB, eIF4E and eEF2K but decreased the mRNA expression of TSC1, TSC2 and eEF2 relative to the control group. Compared with the control group, HS also led to greater mRNA expression of the CSN1S2 gene. Overall, our study indicates that bovine mammary epithelial cells may have the ability to resist HS damage and continue milk protein synthesis partly through enhanced intracellular amino acid absorption and metabolism and by activating the mTOR signaling pathway during HS.

Abstract:The aim of this study was to evaluate the effect of seasonal heat stress (HS) on productive performance, physiological responses, metabolism, and hematological profile of hair breed male lambs finished in feedlot. Twenty Dorper × Katahdin male lambs (body weight = 34.6 ± 1.4 kg and age = 4.5 months) were housed in individual pens and exposed to environmental conditions of summer HS (n = 10, temperature = 28.3 ± 4.0 °C and THI = 77.2 ± 5.4 units) or winter thermoneutrality (n = 10, temperature = 19.2 ± 2.6 °C and THI = 64.0 ± 3.0 units). Each season, a 30-day feeding test was conducted to measure study variables. Compared to thermoneutral lambs, heat-stressed lambs had lower (P < 0.01) growth rate and feed efficiency without changing dry matter intake. Heat-stressed lambs presented higher (P < 0.01) rectal temperature and respiratory rate through the daytime than termoneutral lambs. On most sampling days, summer HS caused lower (P < 0.01) serum concentrations of glucose, cholesterol, total protein, urea, potassium, and thyroid hormones, but higher (P < 0.01) serum triglyceride and chlorine values. Overall serum concentrations of cortisol and insulin were unaffected by summer HS. The blood of heat-stressed lambs showed lower (P ≤ 0.03) erythrocyte and platelet counts, hemoglobin, and hematocrit, but higher (P ≤ 0.03) erythrocyte size and leucocyte count than the blood of thermoneutral lambs. In conclusion, hair breed male lambs in response to chronic conditions of summer HS had slow growth but avoided hyperthermia due to the activation of physiological, metabolic and endocrine adjustments.

Abstract:Dairy cows experiencing heat stress (HS) during the pre-calving portion of the transition period give birth to smaller calves and produce less milk and milk protein. Supplementation of rumen-protected methionine (RPM) has been shown to modulate protein, energy, and placenta metabolism, making it a potential candidate to ameliorate HS effects. We investigated the effects of supplementing RPM to transition cows under HS induced by electric heat blanket (EHB) on cow-calf performance. Six weeks before expected calving, 53 Holstein cows were housed in a tie-stall barn and fed a control diet (CON, 2.2% Met of MP) or a CON diet supplemented with Smartamine®M (MET, 2.6% Met of MP, Adisseo Inc., France). Four weeks pre-calving, all MET and half CON cows were fitted with an EHB. The other half of the CON cows were considered thermoneutral (TN), resulting in 3 treatments: CONTN (n = 19), CONHS (n = 17), and METHS (n = 17). Respiratory rate (RR), skin temperature (ST), and rectal temperature (RT) were measured thrice weekly and core body temperatures recorded bi-weekly. Post-calving body weights (BW) and BCS were recorded weekly, and DMI was calculated and averaged weekly. Milk yield was recorded daily and milk components were analyzed every third DIM. Biweekly AA and weekly nonesterified fatty acids (NEFA), β-hydroxybutyrate (BHB), insulin, and glucose were measured from plasma. Calf birth weight and 24 h growth, thermoregulation, and hematology profile were measured and apparent efficiency of absorption (AEA) of immunoglobulins was calculated. Data were analyzed using the MIXED procedure of SAS with 2 preplanned orthogonal contrasts: CONTN vs. the average of CONHS and METHS (C1) and CONHS vs. METHS (C2). Relative to TN, EHB cows had increased RT during the post-calving weeks and increased RR and ST during the entire transition period. Body weight, BCS, DMI, and milk yield were not impacted by the EHB or RPM. However, protein % and SNF were lower in CONHS, relative to METHS cows. At calving, METHS dams had higher glucose concentrations, relative to CONHS, and during the post-calving weeks, the EHB cows had lower NEFA concentrations than TN cows. Calf birthweight and AEA were reduced by HS, while RR was increased by HS. Calf withers height tended to be shorter and RT were lower in CONHS, compared with MTHS heifers. Overall, RPM supplementation to transition cows reverts the negative impact of HS on blood glucose concentration at calving and milk protein % in the dams and increases wither height while decreasing RT in the calf.
Altered rumen microbiome and correlations of the metabolome in heat-stressed dairy cows at different growth stages

Abstract:ABSTRACT Heat stress is one of the major stressors affecting dairy cow production, which causes great losses in animal husbandry production and economic development. This study combined 16S rDNA sequencing, metagenomic sequencing, and metabolomic analysis to investigate the effects of heat stress on the rumen microbiome and metabolism of cows. To achieve this, 10 cows each of growing heifers, heifers, and lactating cows were selected for sample collection in April and August (a total of 60 cows). Ruminal fluid was collected, filtered through gauze, and immediately transferred to liquid nitrogen prior to metagenomic, 16S rDNA sequencing, and metabolomic analyses. Heat stress increased the abundance of pathogenic bacteria, such as Treponema_2 , Sphingobacterium , and Streptococcus , in cattle at all growth stages, with the structure and composition of rumen microbial communities being the most affected by heat stress in growing heifers and the least affected by heat stress in lactating cows. Heat stress led to a significant decrease in carbohydrate metabolic function in growing heifers and heifers, whereas microbial function was the most stable in lactating cows, suggesting that changes in microbial function are not only more conserved than microbial composition but are also affected by age. Heat stress led to significant enrichment of fatty acyl metabolites in growing heifers, heifers, and lactating cows, and the heat stress indicator species Bacillus , Lactococcus , Streptococcus , and Enterococcus were significantly associated with fatty acyl compounds in growing heifers and heifers. Therefore, heat stress alters the metabolic status of cows by modulating fatty acid biosynthesis, the PPAR signaling pathway, and arachidonic acid metabolism through the regulation of long-chain fatty acids and other biomarkers in cows at three growth stages. IMPORTANCE Heat stress is one of the main causes of economic losses in the dairy industry worldwide; however, the mechanisms associated with the metabolic and microbial changes in heat stress remain unclear. Here, we characterized both the changes in metabolites, rumen microbial communities, and their functional potential indices derived from rumen fluid and serum samples from cows at different growth stages and under different climates. This study highlights that the rumen microbe may be involved in the regulation of lipid metabolism by modulating the fatty acyl metabolites. Under heat stress, the changes in the metabolic status of growing heifers, heifers, and lactating cows were closely related to arachidonic acid metabolism, fatty acid biosynthesis, and energy metabolism. Moreover, this study provides new markers for further research to understand the effects of heat stress on the physiological metabolism of Holstein cows and the time-dependent changes associated with growth stages.

Abstract:Abstract The 70 % of Earthʼs surface covered by oceans supports significant biological diversity and immense untapped potential for marine bioproducts. The recently completed international Census of Marine Life (2000–2010) invested heavily in evaluating the diversity, abundance, and distribution of life in the ocean but concluded that at least 50 % and potentially > 90 % of marine species remain undescribed by science. Despite this potential, and numerous successes spanning pharmaceuticals, nutraceuticals, anti-foulants and adhesives, biofuels, biocatalysts (enzymes), and cosmetics, several impediments have slowed marine bioproduct development. First, the sheer size of the ocean constrains comprehensive exploration. Second, marine taxonomists and ecologists generally do not focus on the most promising groups for bioproduct development. Third, the geographic mismatch between (often remote) biodiversity hotspots and science capacity limit discovery. Despite these challenges, new ocean sampling tools (digital imaging, remote vehicles, genetic approaches, in situ samplers), many developed or improved during the Census of Marine Life, should enhance future marine biodiversity and thus marine bioproduct discovery.
Folic Acid and Taurine Alleviate the Impairment of Redox Status, Immunity, Rumen Microbial Composition and Fermentation of Lambs under Heat Stress

Abstract:The aim of this study was to investigate if the supplementation of folic acid and taurine can relieve the adverse effects of different levels of heat stress (HS) on growth performance, physiological indices, antioxidative capacity, immunity, rumen fermentation and microbiota. A total of 24 Dorper × Hu crossbred lambs (27.51 ± 0.96 kg) were divided into four groups: control group (C, 25 °C), moderate HS group (MHS, 35 °C), severe HS group (SHS, 40 °C), and the treatment group, under severe HS (RHS, 40 °C, 4 and 40 mg/kg BW/d coated folic acid and taurine, respectively). Results showed that, compared with Group C, HS significantly decreased the ADG of lambs (p < 0.05), and the ADG in the RHS group was markedly higher than in the MHS and SHS group (p < 0.05). HS had significant detrimental effects on physiological indices, antioxidative indices and immune status on the 4th day (p < 0.05). The physiological indices, such as RR and ST, increased significantly (p < 0.05) with the HS level and were significantly decreased in the RHS group, compared to the SHS group (p < 0.05). HS induced the significant increase of MDA, TNF-α, and IL-β, and the decrease of T-AOC, SOD, GPx, IL-10, IL-13, IgA, IgG, and IgM (p < 0.05). However, there was a significant improvement in these indices after the supplementation of folic acid and taurine under HS. Moreover, there were a significant increase in Quinella and Succinivibrio, and an evident decrease of the genera Rikenellaceae_RC9_gut_group and Asteroleplasma under HS (p < 0.05). The LEfSe analysis showed that the genera Butyrivibrio, Eubacterium_ventriosum_group, and f_Bifidobacteriaceae were enriched in the MHS, SHS and RHS groups, respectively. Correlated analysis indicated that the genus Rikenellaceae_RC9_gut_group was positively associated with MDA, while it was negatively involved in IL-10, IgA, IgM, and SOD (p < 0.05); The genus Anaeroplasma was positively associated with the propionate and valerate, while the genus Succinivibrio was negatively involved in TNF-α (p < 0.05). In conclusion, folic acid and taurine may alleviate the adverse effects of HS on antioxidant capacity, immunomodulation, and rumen fermentation of lambs by inducing changes in the microbiome that improve animal growth performance.

Abstract:Heat stress is a physiological condition when animals can no longer regulate their internal euthermic temperature. When livestock such as pigs are subjected to this environmental stress, it can be detrimental to performance, health and well-being, and if severe enough even death. Growing pigs are particularly susceptible to heat stress and one of the major organs first affected by heat stress is the gastrointestinal tract. As a result, reductions in appetite, intestinal function and integrity and increased risk of endotoxemia can modify post-absorptive metabolism and tissue accretion. These changes in intestinal integrity may be a result of altered expression of tight junction proteins, increased circulating endotoxin concentrations and markers of cellular stress (heat shock and hypoxia response), which is evident as early on as 2 h after heat-stress onset. Due to restricted blood flow, the ileum is more severely affected compared with the colon. Interestingly, many of the negative effects of heat stress on intestinal integrity appear to be similar to those observed with pigs reared under reduced nutrient and caloric intakes. Altogether, these depress pig performance and health, and extend days to market. Despite this impact on the gastrointestinal tract, under heat-stress conditions, intestinal glucose transport pathways are upregulated. This review discussed how heat stress (directly and indirectly via reduced feed intake) affects intestinal integrity and how heat stress contributes to decreased growth performance in growing pigs.

Impact of heat stress and a feed supplement on hormonal and inflammatory responses of dairy cows

Abstract:The aim of this trial was to evaluate the effects of an immunomodulatory supplement (OmniGen AF, OG; Phibro Animal Health Corp.) and heat stress on hormonal, inflammatory, and immunological responses of lactating dairy cows. Sixty multiparous Holstein cows were randomly assigned to 4 treatments in a 2 × 2 factorial arrangement using 2 environments: cooled using fans and misters, or noncooled, and 2 top-dressed feed supplements (56 g/d): OG or a placebo (CTL). Temperature-humidity index averaged 78 during the 8-wk trial. Blood was drawn to analyze cortisol, prolactin, and circulating tumor necrosis factor (TNF)-α and IL-10. Peripheral blood mononuclear cells (PBMC) were isolated and stimulated with hydrocortisone, prolactin, or lipopolysaccharide (LPS), individually or in several combinations, to assess induced proliferation and cytokine production. At d 52, 6 cows per treatment were injected i.v. with an LPS bolus (ivLPS) to assess hormone and cytokine responses. For cooled cows, feeding OG increased plasma cortisol concentration relative to CTL. Noncooled cows fed CTL had lower circulating TNF-α concentrations than noncooled-OG and cooled-CTL cows, with cooled-OG intermediate. Hydrocortisone+LPS-stimulated PBMC from OG cows tended to proliferate more than CTL. Relative to cooled cows, PBMC from noncooled cows produced more TNF-α and IL-10 when stimulated with LPS. Following ivLPS, cooled-OG cows had a greater cortisol response than the other treatments. In conclusion, OG supplementation enhanced cortisol release under basal condition and induced inflammation with cooling compared with CTL. This suggests that heat stress inhibits OG-mediated cortisol release. Heat stress seemed to enhance the inflammatory responses of PBMC from lactating cows. However, OG supplementation promoted PBMC proliferation under stress, or in the presence of hydrocortisone.

Abstract:Objective: This study evaluated the effects of increased levels of dietary total digestible nutrient (TDN) and crude protein (CP) using protected fat (PF), soybean meal (SB), and heat-treated soybean meal (HSB) on performance, physiological parameters, and behavioral measurements of early-fattening Hanwoo steers under heat stress conditions. Methods: Thirty-six steers (480.9 ± 58.6 kg, 15.9 ± 1.4 months) were assigned to 4 treatments: Control (TDN 75%, CP 15%, rumen degradable protein [RDP]:rumen undegradable protein [RUP]=62:48), PF (TDN 82.5%, CP 15%, RDP:RUP=62:48), PF+SB (TDN 82.5%, CP 16.5%, RDP:RUP=62:48), and PF+SB+HSB (TDN 82.5%, CP 16.5%, RDP:RUP=48:52) for a total of 16 weeks with division of 4 weeks. The average temperature-humidity index (THI) was 82.9 (1st; Moderate), 76.9 (2nd; Mild), 70.9 (3rd; Comfort), and 65.8 (4th period; Comfort). Results: Dry matter intake during whole period did not differ among treatments but decreased by 34% and 19%, respectively during 1st and 2nd compared to 4th. Average daily gain (ADG) of PF+SB+HSB was higher (p<0.05) than that of Control during the 1st period, and those of both PF+SB and PF+SB+HSB were higher (p<0.05) than Control during the 2nd. The ADG during whole period was in the order of PF+SB+HSB (1.23), PF+SB (1.18), PF (1.11), Control (0.98 kg/d) (p<0.05). As THI increased, rectal temperature and all blood parameters increased, while blood glucose levels decreased (p<0.05). Behavioral changes during 1st period compared to 3rd included decreases in Lying (24%), Walking (48%), and Eating (40%), and increases in Total standing (50%) and Drinking (43%) (p<0.05). Rumination during standing was 38% higher, and Rumination during lying was 32% lower (p<0.05). Conclusion: This study demonstrates 10% of increased levels of dietary TDN using PF and CP considering RUP can prevent performance reduction in early-fattening of heat-stressed Hanwoo steers and have positive effects on performance recovery from post-heat stress.

Abstract:Abstract Insult to the gastrointestinal tract (GIT) during heat stress has profound effects on epithelium integrity and subsequent regulation of inflammatory immune responses. The objective of this study was to evaluate the effects of different dietary buffering feeding strategies, on serum amyloid A (SAA), lipopolysaccharide-binding protein (LBP), aspartate aminotransferase (AST), glutamate dehydrogenase (GLDH), gamma-glutamyl transpeptidase (GGT), alkaline phosphatase (ALP), partial pressure of carbon dioxide (pCO2), base excess (BE), blood pH and bicarbonate (HCO3) of grain-fed steers exposed to a simulated heat wave event. Angus steers [n = 12; body weight (BW) = 582 ± 8.92 kg) were housed within climate control rooms for 21 d and randomly allocated into one of three dietary treatments (4 steers/treatment): 1) Treatment 1 (T1), finisher diet; 2) Treatment 2 (T2), transitioned from finisher to heat load diet (HLD) on d 4, fed HLD until d 16; 3) Treatment 3 (T3), fed finisher and supplemented with a calcareous marine algae (CMA; Acid Buffer, Celtic Sea Minerals) on d 4 until d 16. On d 17, T2 and T3 transitioned back to finisher. Days were split into three phases consisting of i) d 0 to 6 [PI, Temperature Humidity Index (THI), 65-78); ii) d 7 to 11 (PII, THI 83-87); iii) d 12 to 20 (PIII, THI 65-78). Individual steer jugular vein blood samples (40 mL) were obtained on d 0, 4 (PI), 7 to 11 (PII), 12 to 17, 18 and 20 (PIII), with plasma extracted. Data were analyzed using a repeated measures model, with a first order auto-regressive error structure, including fixed effects for treatment, phase and day, animal ID as a random effect. During PII, all steers had decreased pCO2 (P < 0.0001), HCO3 (P < 0.04) and BE (P < 0.01), indicating compensated respiratory alkalosis associated with hyperventilation, as blood pH remained unchanged (P = 0.15; Table 1). Both AST (P ≤ 0.009) and GLDH (P ≤ 0.004) were at a maximum during PIII, with T1 having greater AST (P ≤ 0.01) and GLDH (P ≤ 0.04) compared with T2 and T3. Greater AST and GLDH are suggestive of liver damage with reduced detoxifying capabilities becoming overloaded during PII. Interestingly, GGT remained similar (P ≤ 0.94) for TRT × phase, it remains unclear why, warranting further exploration. Alkaline phosphatase was decreased (P < 0.0001) for all treatments during PII and PIII compared with PI, potentially ALP was diverted to the GIT to aide in protection from endotoxemia. During PII, SAA tended to increase (P = 0.08), LBP remained at similar (P = 0.12) levels to PI, before both inflammatory markers decreased (P ≤ 0.05) during PIII. Observationally, steers had gut lining epithelium (mucin casts) within their feces from d 10 to 13. In conclusion, the heat wave initiated a respiratory alkalosis, upregulation of inflammatory marker SAA and mucin casts within feces, suggestive of acute heat stress. Subsequent liver damage, as indicated by greater AST and GLDH post-heat wave, were potentially mediated by inclusion of CMA in the diet.

Abstract:Abstract The objective of this study was to evaluate the impact of vitamin E (vitE) or polyphenols supplemented in feed or drinking water as a heat abatement strategy in growing pigs. Individually housed pigs (n = 128, 47.3 ± 5.0 kg BW) were assigned within weight blocks and sex to a 2x4 factorial arrangement consisting of 2 environments (thermo-neutral [21.2°C] or heat-stressed [30.9°C]) and 4 supplementation treatments (control diet [25 IU/kg dl-α-tocopherol acetate]; control+100 IU/L vitE [d-α-tocopherol] in water; control+200 IU/kg vitE [dl-α-tocopherol acetate] in feed; or control+400 mg/kg polyphenols in feed). Supplementation was started 7 d prior to temperature treatments applied for 28 d. Heat stress reduced (P ≤ 0.001) final BW, ADG, and ADFI (-7.4 kg, -26.7%, and -25.4%, respectively) and increased (P < 0.001) respiration rate and rectal temperature, but no effects of supplementation were detected. Serum vitamin E concentration increased (P < 0.001) with vitE supplementation (1.64, 3.59, 3.24, and 1.67 mg/kg for control, vitE in water, vitE in feed, and polyphenols, respectively) and was greater when supplemented in water vs. feed (P = 0.002), especially when measured on d 28 (chronic) vs. d 2 (acute) of heat stress. Liver vitamin E increased (P < 0.001) with vitE supplementation, especially in water, but not polyphenols (3.9, 31.8, 18.0, 4.9 ppm for control, vitE in water, vitE in feed, and polyphenols, respectively). Serum malondialdehyde (MDA) was greater (P < 0.05) for supplemented pigs compared to control, and heat stress reduced (P = 0.014) serum MDA on d 2, but not d 28. No differences were detected for intestinal morphology or MDA in mucosa of jejunum or ileum. Heat stress decreased (P < 0.03) TNF-α in mucosa of ileum and jejunum, and supplementation reduced (P < 0.05) TNF-α in mucosa of the ileum, but not jejunum. Heat stress markedly reduced performance of growing pigs, and supplementing antioxidants in feed or water was not effective in alleviating the impact of heat stress.

Abstract:The activation or interruption of the responses induced by regulatory peptides are ensured by ectoenzymes, the most important of them belonging to the group of zinc metallopeptidases. Thus angiotensin converting enzyme (ACE) forms the hypertensive peptide angiotensin II from its inactive precursor AI. This also the case for aminopeptidase N (APN) and neutral endopeptidase 24.11 (NEP, CALLA) which together inactivate the endogenous opioid peptides, enkephalins, whereas only NEP is involved in the metabolism of the atrial natriuretic factor (ANP) at the kidney and vascular levels. The pharmacological effects resulting from the inhibition of these enzymatic processes will appear only in tissues where the peptide substrate is tonically or phasically released. This promising approach is expected to avoid, or at least to minimize, the side effects resulting from excessive and ubiquitous stimulation of peptide receptors by exogenously administered agonists or antagonists. The essential amino acids known to be present in the active site of the bacterial endopeptidase thermolysin from crystallographic studies, have also been found in NEP by using a new program of sequence comparison associated with mutagenesis experiments. Several classes of selective inhibitors of NEP, APN and ACE have been rationally designed by taking into account the structural differences in the active site of these peptidases. Thus, the retro-inversion of the amide bond of the NEP inhibitor thiorphan resulted in the elimination of a residual interaction with ACE. Moreover, we have proposed to associate inhibitory potencies towards two peptidases in the same compound. Thus kelatorphan HONH-CO-CH2-CH(CH2 phi)-CONH-CH(CH3)-COOH and other systemically-active mixed NEP/APN inhibitors were shown capable of completely blocking enkephalin metabolism in vivo. This concept has been extended to mixed NEP/ACE inhibitors with compounds such as HS-CH2-CH(CH2 phi)-CONH-CH(CH2R)-COOH where R = CH-(CH3)2 (ES 34) or -OCH2 phi (ES 37). Only mixed inhibitors of NEP and APN are able to produce potent analgesia after intracerebroventricular or systemic administration without the major side effects of morphine (tolerance and dependence). Thiorphan or its prodrugs acetorphan or sinorphan lead to a increase in natriuresis and diuresis by protection of ANP degradation, but without any significant antihypertensive effect. Contrastingly mixed NEP/ACE inhibitors such as ES34 induce decreases in blood pressure higher than those that produced by the association of selective NEP and ACE inhibitors.

Abstract:Abstract Adverse weather conditions are a large constraint to maximizing farm animal productivity. Heat stress, in particular, compromises almost every metric of animal agriculture profitability. Suboptimal production during HS was traditionally thought to result from hypophagia. However, independent of inadequate nutrient consumption, HS affects a plethora of endocrine, physiological, metabolic, circulatory, and immunological variables. Mounting evidence suggest that direct effects of HS originating at the gastrointestinal tract precede the observed effects on the aforementioned systems. Heat stress compromises intestinal barrier integrity causing the appearance of luminal contents, e.g. endotoxin, in circulation. Endotoxin stimulates both a classic immune response with local and systemic inflammatory reactions as well as directly acting on numerous organs and tissues. Once activated, leukocytes switch from oxidative phosphorylation to aerobic glycolysis where the glucose requirement of an intensely triggered immune system can exceed 2 kg/d in a lactating dairy cow. Whole body metabolic adjustments are primarily characterized by increased basal and stimulated circulating insulin, increased hepatic glucose output, decreased adipose tissue mobilization, and decreased skeletal muscle flexibility characterized by a reliance on glucose as a fuel substrate rather than lipid. Ultimately, the metabolic and physiological consequences of heat stress share a similar phenotype with immune challenges. Describing the physiology and mechanisms that underpin how HS jeopardizes animal performance is critical for developing approaches to ameliorate current production issues and requisite for generating future strategies (genetic, managerial, nutritional, and pharmaceutical) aimed at optimizing animal well-being, and improving the sustainable production of high-quality protein for human consumption.
