Abstract:N-(1,3-dimethylbutyl)-N '-phenyl-p-phenylenediamine (6PPD) as a rubber antioxidant has attracted global concern, since its ozone-oxidation product 6PPD-quinone (6PPDQ) was found to be the primary toxicant responsible for urban runoff mortality syndrome in coho salmon. However, the biotransformation fate and associated toxicological mechanism of 6PPD have not received much study yet. In this work, the in vitro assays showed 6PPD can be transformed into 6PPDQ by cytochromes P450 (CYP450) in human liver microsomes (HLMs) with 0.98 % production rate, and the adducts of 6PPDQ with calf thymus DNA and the N-N coupling product between 6PPD and 6PPDQ were further identified after 6PPD incubation in HLMs. Further evidence for the 6PPDQ formation can be obtained from the in vivo assays that the 6PPDQ-DNA adducts and 6PPD-N-N6PPDQ dimer were detected in mice by oral gavage with 6PPD, and the latter dimer species was detected as well in 6PPD exposure to zebrafish larvae. Especially, the bioaccumulation property and high reactivity of 6PPDQ result in the continuous formation of the significant DNA adducts and 6PPD-N-N-6PPDQ dimer even in case of low production rate of biotransformation of 6PPD to 6PPDQ, which may provide potentially effective biomarkers for such process. DFT computations revealed the formation mechanism of 6PPDQ is the (N)Habstraction of 6PPD by CYP450, followed by amino radical rebound at the nearby ortho-carbon, yielding a quinol intermediate due to spin delocalization, that might readily undergo further oxidation by CYP450 into 6PPDQ.
Computational Evidence for Α-Nitrosamino Radical As Initial Metabolite for Both the P450 Dealkylation and Denitrosation of Carcinogenic Nitrosamines B

Abstract:The mutagenic and carcinogenic potency of α-CHₙ-nitrosamines such as N-nitrosodimethylamine (NDMA) is caused by their P450-catalyzed α-hydroxylation and subsequent dealkylation, yielding alkyl diazonium ions (R−NN⁺) as potent electrophiles. Alternatively, P450s may also catalyze their denitrosation as metabolic detoxification. DFT calculations at the UB3LYP/LANL2DZ(Fe)/6-31G+**(H,C,N,O,S)//LANL2DZ(Fe)/6-31G(H,C,N,O,S) level of theory show that H-abstraction from the α-C of NDMA as initial metabolic step yields an α-nitrosamino radical (•CH₂N(CH₃)NO) as common first intermediate for both the oxidative dealkylation (toxification) and denitrosation (detoxification) pathways. In particular, the calculated kinetic isotope effect for the P450-mediated dealkylation of NDMA is in good agreement with experimental information. The results show further that the initial α-hydroxylation of NDMA may proceed in two spin states. Besides a stepwise high-spin (HS, quartet) route with a separate rebound barrier, there is a concerted low-spin (LS, doublet) pathway. Interestingly, the resultant two-state reactivity appears to discriminate between metabolic toxification and detoxification: Evaluation of calculated free energy barriers of the H-abstraction (ΔG‡) through the Eyring equation suggests that the dealkylation:denitrosation product ratio is governed by the LS:HS ratio of the overall metabolic process. Moreover, inclusion of three further α-CHₙ-nitrosamines in the computational analysis demonstrates that the initial H-abstraction barrier is proportional to the C–H bond dissociation enthalpy (BDE) of the substrates, which enables the estimation of spin-averaged reaction barriers through ground-state BDE calculations. The discussion includes also reductive denitrosation pathways that according to current computational evidence appear to be unlikely for aliphatic nitrosamines.
Cytochrome P450 IIIA may be responsible for carbon monoxide generation during microsomal metabolism of 2,4,5-trifluoroaniline

Abstract:Aerobic incubation of 2,4,5-trifluoroaniline (2,4,5-TFA) with NADPH fortified hepatic microsomes from dexamethasone (DEX) pretreated rats resulted in significant generation of carbon monoxide (CO). Microsomes prepared from naive rats or rats pretreated with ��-naphthoflavone failed to generate CO, and microsomes from phenobarbital (PB) pretreated rats catalyzed the reaction at 25% of the rate seen with DEX induced microsomes. The correlation of CO generation with other marker activities of P450s, i.e. erythromycin N-demethylase (ERY), pentoxyresorufin-O-dealkylase, ethoxyresorufin-O-deethylase, and metabolic intermediate complex formation from troleandomycin (TAO) or p-amino-tert-butylbenzoate were examined. Only the markers of cytochrome P450 IIIA, i.e. ERY and TAO were significantly correlated with the CO generation. Pretreatment of DEX induced rats with TAO resulted in similar diminution of microsomal P450 content, TAO, ERY and CO generation. These results revealed that P450 IIIA played a major role in the microsomal CO generation of 2,4,5-TFA, and the CO generation from 2,4,5-TFA might be a new probe to characterize cytochrome P450 IIIA.

Abstract:The interfacial width of polyolefins blends has been probed as a function of distance away from the critical point by using neutron reflectivity. For strongly immiscible polymer pairs, the width of the interface increases slowly when the degree of immiscibility is decreased and the interfacial width varies with the interaction parameter chi of the polymers. Closer to the critical point the dependence on the degree of miscibility becomes stronger and the way in which the interfacial width diverges, as criticality is approached, is related to both the chain length and chi. The self-consistent field theory numerical calculations, with the additional contribution due to capillary waves, provides a good description of the width of the interface between two polymer bulk phases in particular at intermediate values of the degree of immiscibility.
The influence of 4-alkyl substituents on the formation and reactivity of 2-methoxy-quinone methides: evidence that extended pi-conjugation dramatically stabilizes the quinone methide formed from eugenol

Abstract:The effects of para-alkyl substituents on both the cytochrome P450-catalyzed oxidation of phenols to quinone methides (QMs; 4-methylene-2,5-cyclohexadien-1-ones), and on the rates of nucleophilic additions to the QMs were investigated. The derivatives of 4-alkyl-2-methoxyphenol studied were 4-methyl (creosol), 4-ethyl, 4-propyl, 4-isopropyl, and 4-allyl (eugenol). The relative reactivities of QMs derived from these phenols with water were 4-methyl > 4-ethyl = 4-propyl > 4-isopropyl > 4-allyl. These variations in rate were rationalized by differences in stabilization of positive charge density at the site of nucleophilic attack. In particular, saturation of the vinyl substituent on eugenol-QM increases the solvolysis rate 100-fold. This effect is presumably due to the loss of the contribution of an additional aromatic resonance structure to the overall resonance hybrid of the QM from 2-methoxy-4-propylphenol. Finally, the kinetic results show that there is a 472-fold difference in reactivity within this series of QMs. The QM glutathione conjugates were synthesized and characterized by 1H-NMR and electrospray mass spectrometry and a HPLC assay was developed to quantify QM formation in rat liver microsomes. The general trend is increasing alkyl substitution at the para position results in more QM; however, in contrast to the large range of reactivities of the QMs observed in the kinetic experiments, the amounts of P450-derived QM GSH adducts varied only by a factor of 3. In particular, similar amounts of the QMs from eugenol and 2-methoxy-4-propylphenol were produced which suggests that the lack of reported hepatotoxicity for the latter phenol in mice depleted of GSH, may be due to the extreme reactivity of 4-propyl-QM that would be rapidly detoxified by hydrolysis. These data suggest that there may be a threshold cytotoxicity level for QMs related to their reactivity which may affect the relative toxicities of 4-alkyl-2-methoxyphenols.
Molecular Characterization of Carbapenem Resistant Isolates of Acinetobacter baumannii in An Intensive Care Unit of A Tertiary Care Centre at Central India.

Abstract:OBJECTIVE To detect genes encoding carbapenem resistance in Acinetobacter baumannii in an intensive care unit. METHODS A. baumannii isolates were recovered from various clinical specimens of hospitalized patients admitted to the Medical and Surgical intensive care units of a tertiary care centre in Pune. Bacterial identification was performed by routine conventional microbial culture and biochemical tests using standard recommended techniques. Antibiotic sensitivity test was performed by standard Kirby Bauer disc diffusion technique. PCR amplification and automated sequencing was carried out. RESULTS A total of 155 /368 (42.11%) isolates A. baumannii were found to have reduced susceptibility to imipenem (diameter of zones of inhibition ≤13mm) by disc diffusion method. Among these 155 isolates tested 130 (83.87%) isolates showed MIC values for imipenem and meropenem ranging from16-64 mg/L as per CLSI breakpoints. Among these 155 isolates, Carbapenemase production was confirmed by Modified Hodge test for 93 (60%) isolates. Out of 155 isolates, DDST was positive for 89 (57.41%), CDST was positive for 73(47.09%) and MBL (IP/IPI) E-test was positive for 105 (67.74%). blaOXA-51 gene was detected in 47/105 (44.76%), blaOXA-23 gene in 55/105 (52.38%) and blaOXA-58 like gene in 15/105 (14.28%). CONCLUSION MBL production along with co- production of OXA enzymes are considered to be the important reason for resistance to imipenem in Acinetobacter in our health care settings. Hence, early detection of these drug resistant genes by molecular methods is essential in limiting the spread of infection due to these organisms.
Metabolic activation and cytotoxicity of 4-Methylquinoline mediated by CYP3A4 and sulfotransferases in rats

Abstract:4-Methylquinoline (4-MQ) is a quinoline derivative widely present in groundwater and soil and has been reported to be genotoxic. The mechanisms of the toxic action remain unknown. This study aimed to elucidate the metabolic activation of 4-MQ and to determine the possible role of reactive metabolites in 4-MQ-induced liver injury in rats. In the present study, a hydroxylation metabolite (M1), a GSH conjugate (M2) and an NAC conjugate (M3) derived from 4-MQ were detected in vitro and in vivo . The structures of the two conjugates were verified by chemical synthesis, mass spectrometry, and nuclear magnetic resonance. CYP3A4 was found to dominate the hydroxylation of 4-MQ. Sulfotransferases also participated in the metabolic activation of 4-MQ. Pretreatment of primary hepatocytes with ketoconazole (KTC) or 2,6-dichloro-4-nitrophenol (DCNP) not only reduced the production of GSH conjugate M2 but also decreased the susceptibility of hepatocytes to the cytotoxicity of 4-MQ. Urinary NAC conjugate M3 was found in rats given 4-MQ, and M3 may be a potential biomarker for 4-MQ exposure.

Abstract:Existing multi-agent reinforcement learning (MARL) communication methods have relied on a trusted third party (TTP) to distribute reward to agents, leaving them inapplicable in peer-to-peer environments. This paper proposes reward distribution using Neuron as an Agent (NaaA) in MARL without a TTP with two key ideas: (i) inter-agent reward distribution and (ii) auction theory. Auction theory is introduced because inter-agent reward distribution is insufficient for optimization. Agents in NaaA maximize their profits (the difference between reward and cost) and, as a theoretical result, the auction mechanism is shown to have agents autonomously evaluate counterfactual returns as the values of other agents. NaaA enables representation trades in peer-to-peer environments, ultimately regarding unit in neural networks as agents. Finally, numerical experiments (a single-agent environment from OpenAI Gym and a multi-agent environment from ViZDoom) confirm that NaaA framework optimization leads to better performance in reinforcement learning.
Spectroscopic Characterization of Two Peroxyl Radicals During the O 2 -Oxidation of the Methylthio Radical

Abstract:The atmospheric oxidation of dimethyl sulfide (DMS) yields sulfuric acid and methane sulfonic acid (MSA), which are key precursors to new particles formed via homogeneous nucleation and further cluster growth in air masses. Comprehensive experimental and theoretical studies have suggested that the oxidation of DMS involves the formation of the methylthio radical (CH 3 S•), followed by its O 2 -oxidation reaction via the intermediacy of free radicals CH 3 SO x • ( x = 1–4). Therefore, capturing these transient radicals and disclosing their reactivity are of vital importance in understanding the complex mechanism. Here, we report an optimized method for efficient gas-phase generation of CH 3 S• through flash pyrolysis of S -nitrosothiol CH 3 SNO, enabling us to study the O 2 -oxidation of CH 3 S• by combining matrix-isolation spectroscopy (IR and UV–vis) with quantum chemical computations at the CCSD(T)/aug-cc-pV(X + d)Z (X = D and T) level of theory. As the key intermediate for the initial oxidation of CH 3 S•, the peroxyl radical CH 3 SOO• forms by reacting with O 2 . Upon irradiation at 830 nm, CH 3 SOO• undergoes isomerization to the sulfonyl radical CH 3 SO 2 • in cryogenic matrixes (Ar, Ne, and N 2 ), and the latter can further combine with O 2 to yield another peroxyl radical CH 3 S(O) 2 OO• upon further irradiation at 440 nm. Subsequent UV-light irradiation (266 nm) causes dissociation of CH 3 S(O) 2 OO• to CH 3 SO 2 •, CH 2 O, SO 2 , and SO 3 . The IR spectroscopic identification of the two peroxyl radicals CH 3 SOO• and CH 3 S(O) 2 OO• is also supported by 18 O- and 13 C-isotope labeling experiments.

Abstract:Diquat (DQ) poisoning has garnered attention in recent years, primarily due to the rising incidence of cases worldwide, coupled with the absence of a viable antidote for its treatment. Despite the fact that diquat monopyridone (DQ-M) has been identified as a significant metabolite of DQ, the enzyme responsible for its formation remains unknown. In this study, we have identified aldehyde oxidase (AOX) as a vital enzyme involved in DQ oxidative metabolism. The metabolism of DQ to DQ-M was significantly inhibited by AOX inhibitors including raloxifene and hydralazine. The source of oxygen incorporated into DQ-M was proved to be from water through a H2<sup>18</sup>O incubation experiment which further corroborated DQ-M formation via AOX metabolism. The product of DQ-M in vitro generated by fresh rat tissues co-incubation was consistent with its AOX expression. The result of the molecular docking analysis of DQ and AOX protein showed that DQ is capable of binding to AOX. Furthermore, the cytotoxicity of DQ was significantly higher than DQ-M at the same concentration tested in six cell types. This work is the first to uncover the involvement of aldehyde oxidase, a non-cytochrome P450 enzyme, in the oxidative metabolic pathway of diquat, thus providing a potential target for the development of detoxification treatment.

Oxidation reactions of hydroxy naphthoquinones: Mechanistic investigation by LC-Q-TOF-MS analysis

Abstract:PURPOSE: The hydroxyl radical ((●)OH)-induced oxidation reactions of isomeric hydroxy naphthoquinones (generally having anti-tumor activities) namely, lawsone and juglone, were carried out and the reaction mechanism was elucidated.MATERIALS AND METHODS: The degradation products from the reaction of (●)OH (produced by H(2)O(2)/UV) with lawsone and juglone were analyzed using a liquid chromatography quadrupole-time-of-flight mass spectrometer (LC-Q-TOF-MS). The transient intermediate studies were investigated using picosecond pulse radiolysis technique.RESULTS: Mono hydroxylated and dihydroxylated adducts of both lawsone and juglone were identified from the product analysis. The isomeric mono-hydroxylated adducts of lawsone were confirmed using survival yield (SY) analysis. The hydroxylated adducts of lawsone also underwent dimerization reaction. The transient spectral analysis using pulse radiolysis studies revealed the formation of hydroxycyclohexadienyl type radical of both lawsone and juglone as the initially formed intermediate.CONCLUSIONS: The (●)OH-induced reactions of both lawsone and juglone result in the mono and di-hydoxylated derivatives. The demonstration of the various isomeric products using mass spectrometry is a clear proof of the addition probability of (●)OH at different positions of lawsone and juglone, which is generally a difficult task using other analytical techniques.

Abstract:Advanced oxidation processes (AOPs) offer effective means to degrade recalcitrant contaminants, yet concerns about the formation of toxic products may hinder their practical application. This study compared the degradation of meta -chloronitrobenzene ( m CNB) by UV-activated hydrogen peroxide (UV/H 2 O 2 ) and persulfate (UV/PDS) oxidation. Both methods demonstrated efficient m CNB removal, with 73 % and 64 % removal observed, respectively, in 30 min at an oxidant dose of 1.0 mM. However, the products formed in the UV/PDS process exhibited higher toxicity compared to those formed in the UV/H 2 O 2 process. This difference in toxicity arose from the distinct rection behavior of hydroxyl radical (HO • ) and sulfate radical (SO 4 •− ), the principal reactive species in UV/H 2 O 2 and UV/PDS processes, respectively. HO • mainly attacked m CNB via an addition mechanism, forming hydroxylated products in UV/H 2 O 2 . In contrast, SO 4 •− reacted with m CNB via single-electron transfer, forming a radical cation ( m CNB •+ ). This radical cation underwent rapid dissociation or hydrolysis to 3-chrolophenol (3CP) and 2-chloro-4-nitrophenol (2C4NP), respectively. Subsequent oxidation of 2C4NP by SO 4 •− generated a phenoxyl radical. The coupling of m CNB •+ or phenoxyl radical with a nitrogen dioxide radical (NO 2 • ) derived from m CNB denitration, leading to the generation of more toxic dinitro-products. We assume these findings can be extrapolated to the treatment of other nitroaromatic compounds. SO 4 •− -based oxidation technologies tend to generate more polynitrated byproducts than HO • -based oxidation technologies, thus posing a greater potential risk. This consideration should guide the selection of appropriate treatment technologies for these contaminates.

RNA-Pt adducts following cisplatin treatment of Saccharomyces cerevisiae.

Abstract:The numerous regulatory roles of cellular RNAs suggest novel potential drug targets, but establishing intracellular drug-RNA interactions is challenging. Cisplatin (cis-diamminedichloridoplatinum(II)) is a leading anticancer drug that forms exchange-inert complexes with nucleic acids, allowing its distribution on cellular RNAs to be followed ex vivo. Although Pt adduct formation on DNA is well-known, a complete characterization of cellular RNA-Pt adducts has not been performed. In this study, the action of cisplatin on S. cerevisiae in minimal media was established with growth curves, clonogenic assays, and tests for apoptotic markers. Despite high toxicity, cisplatin-induced apoptosis in S. cerevisiae was not observed under these conditions. In-cell Pt concentrations and Pt accumulation on poly(A)-mRNA, rRNA, total RNA, and DNA quantified via ICP-MS indicate ∼4- to 20-fold more Pt accumulation in total cellular RNA than in DNA. Interestingly, similar Pt accumulation is observed on rRNA and total RNA, corresponding to one Pt per (14,600 ± 1,500) and (5760 ± 580) nucleotides on total RNA following 100 and 200 μM cisplatin treatments, respectively. Specific Pt adducts mapped by primer extension analysis of a solvent-accessible 18S rRNA helix occur at terminal and internal loop regions and appear as soon as 1 h post-treatment. Pt per nucleotide accumulation on poly(A)-mRNA is 4- to 6-fold lower than on rRNA but could have consequences for low copy-number or highly regulated transcripts. Taken together, these data demonstrate significant accumulation of Pt adducts on cellular RNA species following in cellulo cisplatin treatment. These and other small molecule-RNA interactions could disrupt processes regulated by RNA.
Quantification of singlet oxygen production in the reaction of superoxide with hydrogen peroxide using a selective chemiluminescent probe.

Abstract:A sensitive chemiluminescent probe that selectively reacts with singlet oxygen in the presence of superoxide and hydrogen peroxide has been used to quantify the production of singlet oxygen in the reaction of superoxide with hydrogen peroxide. The yield of singlet oxygen from this reaction was found to be low (0.2% relative to the initial superoxide concentration). No evidence for the formation of hydroxyl radical was observed in this reaction, ruling out the Haber-Weiss mechanism as a major singlet oxygen formation pathway. No singlet oxygen production was observed in the reaction of superoxide with 2-nitrobenzoic acid, which has a pKa similar to that of hydrogen peroxide, rendering the protonation of superoxide, followed by its disproportionation, an unlikely explanation for the formation of singlet oxygen in this system. The low yields of singlet oxygen and hydroxyl radical suggest that their formation in this reaction should be relatively unimportant in biological systems.

Abstract:Carbon isotope fractionation of sulfamethoxazole (SMX) during biodegradation by Microbacterium sp. strain BR1 (ipso-hydroxylation) and upon direct photolysis was investigated. Carbon isotope signatures (δ(13)C) of SMX were measured by LC-IRMS (liquid chromatography coupled to isotope ratio mass spectrometry). A new LC-IRMS method for the SMX metabolite, 3-amino-5-methylisoxazole (3A5MI), was established. Carbon isotope enrichment factors for SMX (ε(C)) were -0.6 ± 0.1‰ for biodegradation and -2.0 ± 0.1‰ and -3.0 ± 0.2‰ for direct photolysis, at pH 7.4 and pH 5, respectively. The corresponding apparent kinetic isotope effects (AKIE) for ipso-hydroxylation were 1.006 ± 0.001; these fall in the same range as AKIE in previously studied hydroxylation reactions. The differences in SMX and 3A5MI fractionation upon biotic and abiotic degradation suggest that compound specific stable isotope analysis (CSIA) is a suitable method to distinguish SMX reaction pathways. In addition, the study revealed that the extent of isotope fractionation during SMX photolytic cleavage is pH-dependent.

Hydrogen Sulfide Protects against Paraquat-Induced Acute Liver Injury in Rats by Regulating Oxidative Stress, Mitochondrial Function, and Inflammation

Abstract:In addition to the lung, the liver is considered another major target for paraquat (PQ) poisoning. Hydrogen sulfide (H 2 S) has been demonstrated to be effective in the inhibition of oxidative stress and inflammation. The aim of this study was to investigate the protective effect of exogenous H 2 S against PQ-induced acute liver injury. The acute liver injury model was established by a single intraperitoneal injection of PQ, evidenced by histological alteration and elevated serum aminotransferase levels. Different doses of NaHS were administered intraperitoneally one hour before exposure to PQ. Analysis of the data shows that exogenous H 2 S attenuated the PQ-induced liver injury and oxidative stress in a dose-dependent manner. H 2 S significantly suppressed reactive oxygen species (ROS) generation and the elevation of malondialdehyde content while it increased the ratio of GSH/GSSG and levels of antioxidant enzymes including SOD, GSH-Px, HO-1, and NQO-1. When hepatocytes were subjected to PQ-induced oxidative stress, H 2 S markedly enhanced nuclear translocation of Nrf2 via S-sulfhydration of Keap1 and resulted in the increase in IDH2 activity by regulating S-sulfhydration of SIRT3. In addition, H 2 S significantly suppressed NLRP3 inflammasome activation and subsequent IL-1 β excretion in PQ-induced acute liver injury. Moreover, H 2 S cannot reverse the decrease in SIRT3 and activation of the NLRP3 inflammasome caused by PQ in Nrf2-knockdown hepatocytes. In summary, H 2 S attenuated the PQ-induced acute liver injury by enhancing antioxidative capability, regulating mitochondrial function, and suppressing ROS-induced NLRP3 inflammasome activation. The antioxidative effect of H 2 S in PQ-induced liver injury can at least partly be attributed to the promotion of Nrf2-driven antioxidant enzymes via Keap1 S-sulfhydration and regulation of SIRT3/IDH2 signaling via Nrf2-dependent SIRT3 gene transcription as well as SIRT3 S-sulfhydration. Thus, H 2 S supplementation can form the basis for a promising novel therapeutic strategy for PQ-induced acute liver injury.

Abstract:In this study, we examined the metabolic and gut microbiome responses to paraquat (PQ) in male Wistar rats, focusing on oxidative stress effects. Rats received a single intraperitoneal injection of PQ at 15 and 30 mg/kg, and various oxidative stress parameters (i.e., MDA, SOD, ROS, 8-isoprostanes) were assessed after three days. To explore the omic profile, GC-qTOF and UHPLC-qTOF were performed to assess the plasma metabolome; 1H-NMR was used to assess the urine metabolome; and shotgun metagenomics sequencing was performed to study the gut microbiome. Our results revealed reductions in body weight and tissue changes, particularly in the liver, were observed, suggesting a systemic effect of PQ. Elevated lipid peroxidation and reactive oxygen species levels in the liver and plasma indicated the induction of oxidative stress. Metabolic profiling revealed changes in the tricarboxylic acid cycle, accumulation of ketone body, and altered levels of key metabolites, such as 3-hydroxybutyric acid and serine, suggesting intricate links between energy metabolism and redox reactions. Plasma metabolomic analysis revealed alterations in mitochondrial metabolism, nicotinamide metabolism, and tryptophan degradation. The gut microbiome showed shifts, with higher PQ doses influencing microbial populations (e.g., Escherichia coli and Akkermansia muciniphila) and metagenomic functions (pyruvate metabolism, fermentation, nucleotide and amino acid biosynthesis). Overall, this study provides comprehensive insights into the complex interplay between PQ exposure, metabolic responses, and gut microbiome dynamics. These findings enhance our understanding of the mechanisms behind oxidative stress-induced metabolic alterations and underscore the connections between xenobiotic exposure, gut microbiota, and host metabolism.

Abstract:This study was undertaken to elucidate the mechanism of organ specificity and differential efficacy of garlic organosulfides (OSCs) [diallyl sulfide (DAS), diallyl disulfide (DADS), diallyl trisulfide (DATS), dipropyl sulfide (DPS) and dipropyl disulfide (DPDS)] in preventing benzo(a)pyrene (BP)-induced tumorigenesis in mice. The results of the present study reveal a good correlation between chemopreventive efficacies of garlic OSCs and their inductive effects on the expression of NAD(P)H:quinone oxidoreductase (NQO), an enzyme implicated in the detoxification of activated quinone metabolites of BP. Treatment of mice with DADS and DATS, which are potent inhibitors of BP-induced forestomach tumorigenesis, resulted in a statistically significant increase (2.4- and 1.5-fold, respectively) in forestomach NQO activity. In addition, DADS and DATS were much more potent inducers of forestomach NQO activity than DAS, which is a weak inhibitor of BP-induced forestomach tumorigenesis than the former compounds. Propyl-group containing OSCs (DPS and DPDS), which do not inhibit BP-induced tumorigenesis, did not affect forestomach NQO activity. Similar to forestomach, a good correlation was also observed between effects of these OSCs against BP-induced pulmonary tumorigenesis and their effects on NQO expression in the lung. For example, treatment of mice with DAS, which is a potent inhibitor of BP-induced pulmonary tumorigenesis, resulted in about 3.2-fold increase in pulmonary NQO activity. On the other hand, this activity was increased by about 1.5-fold upon DATS administration, which does not inhibit BP-induced cancer of the lung. In conclusion, our results suggest that induction of NQO may be important in anti-cancer effects of garlic OSCs.
Trapping DNA-radicals with DMPO reduced hypochlorous acid-induced 8-oxo-7,8-dihydro-2'-deoxyguanosine and mutagenesis in lung epithelial cells

Abstract:Irritation causes the recruitment and activation of neutrophils in the stressed airways. This process is known as neutrophilic inflammation. This process results in myeloperoxidase (MPO), an enzyme contained inside neutrophil azurophilic granules, being released as neutrophil extracellular traps (NETs), which also contain genomic DNA, modified histones, and other proteins. In the airways, released MPO can be taken up by bystander tissue epithelial cells. MPO is the only mammalian peroxidase enzyme that under physiological conditions produces hypochlorite (HOCl). Intracellularly produced HOCl may damage the cell genome, with the intermediacy of DNA-centered free radicals, which upon reaction with molecular oxygen decay to mutagenic end-oxidation products, such as 8-oxo-7,8-dihydro-2'-deoxyguanosine (8-oxo-dGuo). Herein, we aimed to test whether HOCl-induced DNA-centered radicals precede the oxidation of DNA and mutagenesis in A549 human lung epithelial cells as an in vitro model that resembles neutrophilic inflammation in irritated airways. Interestingly, by trapping HOCl-induced DNA-centered radicals, the nitrone spin trap 5,5-dimethyl-1-pyrroline N-oxide (DMPO) blocks the formation of 8-oxo-dGuo and possibly other end-oxidation products, forming DNA-DMPO nitrone adducts, thus reducing mutagenesis in the hypoxanthine phosphoribosyl transferase (hrpt) gene, one of the most sensitive genes to oxidative damage. P53 is a transcription factor known as the master regulator of the cell response to genomic damage. By trapping DNA-centered radicals, DMPO also blocks the translocation of p53 to the cell nucleus, suggesting that by trapping DNA-centered radicals with DMPO, end-oxidation products are prevented, and the cell response to genomic damage is not sensed. DMPO traps DNA-centered radicals, reduces 8-oxo-dGuo accumulation, and blocks hrpt gene mutation. Trapping DNA-centered radicals to reduce the accumulation of HOCl-induced mutagenic end-oxidation products in the genome of bystander cells, which have taken MPO from the inflammatory milieu, will provide new therapeutic avenues to reduce genotoxic damage at sites of neutrophilic inflammation, such as in the irritated airways.
