Abstract:Type 2 glycine transporter (GlyT2) mediates intracellular glycine transport and is expressed selectively in glycinergic neurons. Expression of GlyT2 gene promoter-driven enhanced green fluorescent protein (eGFP) in BAC transgenic mice allows selective visualization of glycinergic neurons by fluorescence microscopy. Here, we show that cerebellar interneuron precursors identified by the transcription factor Pax2, including gamma-aminobutyric acid (GABA)ergic interneurons of the molecular layer (ML; basket and stellate cells), transiently express GlyT2-eGFP during development. In contrast, expression of endogenous GlyT2 is restricted to glycinergic Golgi cells. Comparison with knock-in mice expressing eGFP in GABAergic neurons [glutamic acid decarboxylase (GAD)67-eGFP] revealed that GlyT2-eGFP expression often precedes GAD67-eGFP and is therefore a marker of immature GABAergic interneurons. In the internal granule cell layer, GABAergic Golgi cells differentiated shortly after birth, prior to glycinergic Golgi cells. In the ML, GlyT2-eGFP-positive precursor cells migrated until the boundary with the external granule cell layer, forming an inside-out maturation gradient that determined the final position of interneurons in the ML. After migration, GlyT2-eGFP gradually disappeared, while interneurons differentiated morphologically and became immunoreactive for parvalbumin, the GABA(A) receptor alpha1 subunit, and the K(+)Cl(-) exchanger KCC2 (K(+)Cl(-) cotransporter type 2). Numerous presumptive GABAergic synaptic terminals were seen on immature ML interneurons as early as P4, preceding the expression of these neurochemical markers. These results suggest that GABAergic synaptogenesis marks the onset of differentiation of basket and stellate cells in the mouse cerebellum, and that GABAergic synaptic function might contribute to the differentiation of interneurons in the cerebellar cortex.

Activity-dependent plasticity of presynaptic GABAB receptors at parallel fiber synapses

Abstract:Parallel fiber synapses in the cerebellum express a wide range of presynaptic receptors. However, presynaptic receptor expression at individual parallel fiber synapses is quite heterogeneous, suggesting physiological mechanisms regulate presynaptic receptor expression. We investigated changes in presynaptic GABAB receptors at parallel fiber-stellate cell synapses in acute cerebellar slices from juvenile mice. GABAB receptor-mediated inhibition of excitatory postsynaptic currents (EPSCs) is remarkably diverse at these synapses, with transmitter release at some synapses inhibited by >50% and little or no inhibition at others. GABAB receptor-mediated inhibition was significantly reduced following 4 Hz parallel fiber stimulation but not after stimulation at other frequencies. The reduction in GABAB receptor-mediated inhibition was replicated by bath application of forskolin and blocked by application of a PKA inhibitor, suggesting activation of adenylyl cyclase and PKA are required. Immunolabeling for an extracellular domain of the GABAB2 subunit revealed reduced surface expression in the molecular layer after exposure to forskolin. GABAB receptor-mediated inhibition of action potential evoked calcium transients in parallel fiber varicosities was also reduced following bath application of forskolin, confirming presynaptic receptors are responsible for the reduced EPSC inhibition. These data demonstrate that presynaptic GABAB receptor expression can be a plastic property of synapses, which may compliment other forms of synaptic plasticity. This opens the door to novel forms of receptor plasticity previously confined primarily to postsynaptic receptors.

Abstract:Metabotropic GABAB receptors mediate slow inhibitory effects presynaptically and postsynaptically through the modulation of different effector signalling pathways. Here, we analysed the distribution of GABAB receptors using highly sensitive SDS-digested freeze-fracture replica labelling in mouse cerebellar Purkinje cells. Immunoreactivity for GABAB1 was observed on presynaptic and, more abundantly, on postsynaptic compartments, showing both scattered and clustered distribution patterns. Quantitative analysis of immunoparticles revealed a somato-dendritic gradient, with the density of immunoparticles increasing 26-fold from somata to dendritic spines. To understand the spatial relationship of GABAB receptors with two key effector ion channels, the G protein-gated inwardly rectifying K+ (GIRK/Kir3) channel and the voltage-dependent Ca2+ channel, biochemical and immunohistochemical approaches were performed. Co-immunoprecipitation analysis demonstrated that GABAB receptors co-assembled with GIRK and CaV2.1 channels in the cerebellum. Using double-labelling immunoelectron microscopic techniques, co-clustering between GABAB1 and GIRK2 was detected in dendritic spines, whereas they were mainly segregated in the dendritic shafts. In contrast, co-clustering of GABAB1 and CaV2.1 was detected in dendritic shafts but not spines. Presynaptically, although no significant co-clustering of GABAB1 and GIRK2 or CaV2.1 channels was detected, inter-cluster distance for GABAB1 and GIRK2 was significantly smaller in the active zone than in the dendritic shafts, and that for GABAB1 and CaV2.1 was significantly smaller in the active zone than in the dendritic shafts and spines. Thus, GABAB receptors are associated with GIRK and CaV2.1 channels in different subcellular compartments. These data provide a better framework for understanding the different roles played by GABAB receptors and their effector ion channels in the cerebellar network.

Abstract:Inhibitory synaptic plasticity is important for shaping both neuronal excitability and network activity. Here we investigate the input and GABAA receptor subunit specificity of inhibitory synaptic plasticity by studying cerebellar interneuron–Purkinje cell (PC) synapses. Depolarizing PCs initiated a long-lasting increase in GABA-mediated synaptic currents. By stimulating individual interneurons, this plasticity was observed at somatodendritic basket cell synapses, but not at distal dendritic stellate cell synapses. Basket cell synapses predominantly express β2-subunit-containing GABAA receptors; deletion of the β2-subunit ablates this plasticity, demonstrating its reliance on GABAA receptor subunit composition. The increase in synaptic currents is dependent upon an increase in newly synthesized cell surface synaptic GABAA receptors and is abolished by preventing CaMKII phosphorylation of GABAA receptors. Our results reveal a novel GABAA receptor subunit- and input-specific form of inhibitory synaptic plasticity that regulates the temporal firing pattern of the principal output cells of the cerebellum.

GABAA currents in immature dentate gyrus granule cells

Abstract:We used whole cell patch clamp and gramicidin perforated patch recordings in hippocampal slices to study gamma-aminobutyric acid (GABA) currents in granule cells (GCs) from juvenile rat dentate gyrus (DG). GCs are generated postnatally and asynchronously such that they can be detected at different stages of their maturation in DG within the first month. In contrast, inhibitory interneurons are generated embryonically, and their circuitry is well developed even as their target GCs and GC excitatory connections are still being formed. In this study, two GABA currents evoked in GCs by medial perforant path stimulation are compared. The first, pharmacologically isolated by glutamate receptor blockade, is the product of direct activation of GABA interneurons with monosynaptic input to the recorded GC (monosynaptic GABAA). Monosynaptic GABAA displays slight outward rectification of its current-voltage relation, is 97% eliminated by 10 microM bicuculline and coincides temporally with the excitatory components of GC postsynaptic currents as has been described for GABAA currents in other brain regions. The second is a novel GABA response that is detectable in 10 microM bicuculline and is present on GCs only at the earliest stages of their maturation. Unlike monosynaptic GABAA, this transient GABA is eliminated by glutamate receptor blockade and hence is likely to be generated by interneurons activated via an intervening glutamatergic synapse (polysynaptically). It is predominantly chloride mediated, has a relative bicarbonate/chloride permeability ratio of 26%, and is unchanged by bath-applied saclofen and strychnine or by intracellular calcium chelation. It is 97% antagonized by 100 microM picrotoxin and 99% antagonized by 100 microM bicuculline. This current is thus a relatively bicuculline (BMI)-resistant GABAA current (BMIR-GABAA). Compared with monosynaptic GABAA, BMIR-GABAA has a later peak, slower time course of decay, and marked outward rectification. Its reversal potential is 7-8 mV depolarized to that of monosynaptic GABAA whether recorded in whole cell or with gramicidin perforated patch to preserve native internal chloride concentration. Together these data may suggest that BMIR-GABAA is evoked by an anatomically segregated population of interneurons activating a unique, developmentally regulated GABAA receptor. Further, the transient nature of this current coupled with its temporal characteristics that preclude overlap with the excitatory components of the synaptic response are consistent with a role that is trophic or signaling rather than primarily inhibitory.

Abstract:Clearance of Deposited Silicon Carbide Whisker from Rat Lungs Inhaled during a 4‐Week Exposure: Izumi Akiyama, et al. Department of Environmental Health Engineering, Institute of Industrial Ecological Sciences, University of Occupational and Environmental Health— Pulmonary deposition and the clearance of deposited fiber particles from lungs are key determinant factors in assessing potential carcinogenicity and fibrogenicity. Forty‐two Wistar male rats (9 wk old) were exposed to silicon carbide whisker (SiCW) for 6 h/d, 5 d/wk for 4 wk. The mass median aerodynamic diameter was 2.5 µm as detected by an Andersen sampler, and the geometric mean fiber diameter and length determined by a scanning electron microscope were 0.4 and 2.2 µm, respectively. The daily average exposure concentrations were 10.4 ± 0.5 mg/m3 (214 ± 31 fibers/ ml) during the exposure period. The rats were sacrificed after 3 d, 2 wk, 1, 2, 3, 6 and 12 months after 4‐wk exposure. At the time of sacrifice, the wet lung weights and the deposited SiCW amounts in the lungs were measured by an X ray reference method. The amount of SiCW deposited was 0.60 ± 0.09 mg 3 d after a 4‐wk exposure. The apparent deposition fraction was 4.8 ± 0.7 (%). During the clearance period, the amount of SiCW deposited in the rat lungs decreased exponentially with the increasing duration of the clearance period. The biological half time in the onecompartment model was determined to be 4.0 months which is similar to the values for glass fiber, potassium titanate whisker and aluminium‐silicate ceramic fiber under similar exposure conditions.
Gap Junctions May Have A Computational Function In The Cerebellum: A Hypothesis

Abstract:In the cerebellum, granule cells make parallel fibre contact on (and excite) Golgi cells and Golgi cells inhibit granule cells, forming an open feedback loop. Parallel fibres excite Golgi cells synaptically, each making a single contact. Golgi cells inhibit granule cells in a structure called a glomerulus almost exclusively by GABA spillover acting through extrasynaptic GABA A receptors. Golgi cells are connected dendritically by gap junctions. It has long been suspected that feedback contributes to homeostatic regulation of parallel fibre signals activity, causing the fraction of the population that are active to be maintained at a low level. We present a detailed neurophysiological and computationally-rendered model of functionally grouped Golgi cells which can infer the density of parallel fibre signals activity and convert it into proportional modulation of inhibition of granule cells. The conversion is unlearned and not actively computed; rather, output is simply the computational effect of cell morphology and network architecture. Unexpectedly, the conversion becomes more precise at low density, suggesting that self-regulation is attracted to sparse code, because it is stable. A computational function of gap junctions may not be confined to the cerebellum.

[The Pharmacology of the GABA-induced Biphasic Response in Guinea-Pig Celiac Ganglion Neurons in Vitro].

Abstract:In guinea-pig celiac ganglion neurons, GABA induced a biphasic response, i.e. a depolarizing response associated with a resistance decrease followed by a hyperpolarizing response associated with a resistance increase. Such response persisted in low Ca2+ high Mg2+ medium. The GABA-induced biphasic response was mimicked by muscimol and inhibited by bicuculline methiodide (100 mumol/L) and picrotoxin (100-300 mumol/L). At doses producing almost equal depolarizing responses, muscimol was weaker in producing hyperpolarizing responses than GABA. Baclofen had no effect on membrane potentials of the ganglion cells. These results suggest that the depolarizing phase of the GABA-induced biphasic response is mediated by GABAA/Cl- ionophore receptor complex and the hyperpolarizing phase may be mediated by a novel type of GABA receptor possessing similar pharmacological properties as the classical GABAA receptor.

Abstract:Collected in a finely produced volume, the main pieces of the long and famous debate between Robert Platt and George Pickering about the nature of essential hypertension are particularly welcome and timely. Indeed, the debate has not only a historical interest as if today hypertension research delights in looking back at its roots, in revisiting the battlefields or, more properly, the tourney arenas in which it has developed. The fact is that, consciously or unconsciously, the debate is still going on, and this volume is a good opportunity and a healthy stimulus to discover how much of one side of the debate is yet concealed under the other, though the latter side is now considered as common wisdom and orthodox teaching. Professor John Swales, the editor of the publication, has made an excellent selection from the most important and pertinent writings of the two combatants and has admirably summarized the terms of the debate and its evolution in time. The technical controversy, as is well known, started about the unimodal or bimodal appearance of blood pressure frequency distribution curves, went on to the interpretation of hypertension as a single-gene-inherited disease or a multiple-gene-inherited condition, enlarged over the more philosophical issue of qualitative versus quantitative diseases. The essence of the debate was that in Platt's view hypertension was an inherited disease with dominant type transmission, with a specific (though unknown) defect, with a well-known natural history and with discrete lesions; whereas, according to George Pickering, hypertension was only the upper portion of a continuous distribution curve of blood pressure values, and, as blood pressure, depended on multiple-gene inheritance. In reading again all the arguments of the controversy together, one can see why the debate dragged on so long and was so long undecided. The first reason is that the data were rather scanty and the conclusions somewhat inferential and indirect. The number of Platt's observations was surprisingly small (350 sibs of 178 patients in his last paper), and George Pickering had an easy task in pointing out that any number of peaks could appear by mere chance in the distribution curves of such a small number of observations. Pickering's data were by far more numerous, but their manipulation to obtain age-and sex-adjusted scores, the different number of peaks described when the initial data were recalculated by Platt, the confusing argument of digit preference in measuring blood pressure values, all these aspects …
Modulation of presynaptic Ca2+ entry by AMPA receptors at individual GABAergic synapses in the cerebellum

Abstract:Cerebellar Purkinje cells (PCs) receive GABAergic input that undergoes powerful retrograde modulation by presynaptic cannabinoid and glutamate receptors. Here we examine a distinct modulatory mechanism at these synapses, which does not require postsynaptic depolarization and acts via presynaptic AMPA receptors. We find that this mechanism operates mainly in the somatic vicinity of PCs in which large boutons of basket cell axons form synapses on the PC soma. We use fast confocal microscopy and detailed kinetic modeling to estimate that, in these boutons, an action potential opens 100-200 Ca2+ channels, eliciting a brief 3-5 microM transient, followed by a longer-term, 15-30 nM rise of free Ca2+ (above the resting level of approximately 100 nM). Brief activation of local AMPA receptors suppresses Ca2+ entry (probably by silencing 20-40 P/Q-type channels) in a subgroup of terminals that tend to show a higher dynamic range of Ca2+ signaling. The results provide the first quantitative description of presynaptic Ca2+ kinetics and its modulation by AMPA receptor activation (most likely via a glutamate spillover-mediated mechanism) at identified GABAergic synapses.

Abstract:Type-A receptors for the neurotransmitter GABA (γ-aminobutyric acid) are ligand-gated chloride channels that mediate inhibitory neurotransmission. Each subunit of the pentameric receptor protein has ligand-binding sites in the amino-terminal extracellular domain and four membrane-spanning regions, one of which forms a wall of the ion channel1. Each subunit also has a large intracellular loop that may be a target for protein kinases and be required for subcellular targeting and membrane clustering of the receptor, perhaps by anchoring the receptor to the cytoskeleton2,3,4. Neurotransmitter receptors need to be positioned in high density in the cell membrane at sites postsynaptic to nerve terminals releasing that neurotransmitter. Other members of the superfamily of ligand-gated ion-channel receptors associate in postsynaptic-membrane clusters by binding to the proteins rapsyn or gephyrin5,6,7. Here we identify a new cellular protein, GABAA-receptor-associated protein (GABARAP), which can interact with the γ2 subunit of GABAA receptors. GABARAP binds to GABAA receptors both in vitro and in vivo, and co-localizes with the punctate staining of GABAA receptors on cultured cortical neurons. Sequence analysis shows similarity between GABARAP and light chain-3 of microtubule-associated proteins 1A and 1B. Moreover, the N terminus of GABARAP is highly positively charged and features a putative tubulin-binding motif. The interactions among GABAA receptors, GABARAP and tubulin suggest a mechanism for the targeting and clustering of GABAA receptors.

Abstract:Long-term modification of transmission efficacy at synapses is the cellular basis of memory and learning1–4. A special type of synaptic plasticity in the cerebellum was postulated theoretically5,6, and has since been verified4,7–9. Each cerebellar Purkinje cell (PC) receives two distinct excitatory inputs, one from parallel fibres (PFs) and the other from a climbing fibre (CF). When these two types of inputs are conjunctively activated, PF–PC transmission undergoes long-term depression (LTD)4,7–9. Accumulated evidence suggests that LTD plays a role in the motor learning processes of the cerebellum10–12. At the molecular level, LTD appears to be caused by desensitization of receptor molecules in PC dendrites towards the PF neurotransmitter4, presumably L-glutamate (Glu) (ref. 12). Glu receptors are heterogeneous and can be divided into several subtypes13–15. In this study, we compared the potency of several Glu agonists in inducing LTD and found a highly selective dependency of LTD on the quisqualate(QA)-selective subtype of Glu receptors.

Serotonin drives a novel GABAergic synaptic current recorded in rat cerebellar purkinje cells: a Lugaro cell to Purkinje cell synapse

Abstract:We recorded a novel fast GABAergic synaptic current in cerebellar Purkinje cells in rat brain slices using patch-clamp techniques. Because of a relatively low sensitivity to bicuculline, these currents can be recorded under conditions in which basket and stellate cell inputs are blocked. The observations that the novel synaptic currents occur spontaneously only in the presence of serotonin, and the specific limited positions in the slice from which they can be electrically evoked, suggest that the presynaptic cell is the Lugaro cell. Cell-attached recordings confirm that the Lugaro cell is the only interneuron in the cerebellar cortex with firing behavior consistent with the spontaneous activity recorded in Purkinje cells. The input shows a strong presynaptic modulation mediated by GABA(A) receptors, resulting in a dynamic range from almost 0 to >90% release probability. Modeling GABA(A) receptor responses to different GABA transients suggests that the relatively low sensitivity of the synaptic currents to bicuculline, compared with the higher affinity GABA(A) receptor antagonist SR-95531 (2-(3-carboxypropyl)-3-amino-6-(4-methoxyphenyl) pyridazinium), is attributable to an unusually long GABA dwell time and/or high GABA concentration in the synaptic cleft. The significance of this novel input is discussed in relation to other GABAergic synapses impinging on Purkinje cells. We suggest that the release of GABA onto Purkinje cells from Lugaro cells would primarily occur during motor activity under conditions in which the activity of basket and stellate cells might be inhibited.

In this Classic Paper Revisited, the author recounts the scientific journey leading to a report published in the Proceedings of the National Academy of Sciences (PNAS) and shares several personal stories from her formative years and “research truths” that she has learned along the way. Briefly, the principal inhibitory neurotransmitter in the brain, γ-aminobutyric acid (GABA), was conventionally thought to regulate cognitive processes by activating synaptic GABA type A (GABAA) receptors and generating transient inhibitory synaptic currents. However, the author’s laboratory team discovered a novel nonsynaptic form of tonic inhibition in hippocampal pyramidal neurons, mediated by extrasynaptic GABAA receptors that are pharmacologically distinct from synaptic GABAA receptors. This tonic current is highly sensitive to most general anesthetics, including sevoflurane and propofol, and likely contributes to the memory-blocking properties of these drugs. Before the publication in PNAS, the subunit composition of GABAA receptors that generate the tonic current was unknown. The team’s research showed that GABAA receptors containing the α5 subunit (α5GABAARs) generated the tonic inhibitory current in hippocampal neurons. α5GABAARs are highly sensitive to GABA, desensitize slowly, and are thus well suited for detecting low, persistent, ambient concentrations of GABA in the extracellular space. Interest in α5GABAARs has surged since the PNAS report, driven by their pivotal roles in cognitive processes and their potential as therapeutic targets for treating various neurologic disorders.

Abstract:Ionic plasticity, a form of synaptic plasticity unique to inhibitory neurotransmission, can be induced in cerebellar Purkinje neurons by brain-derived neurotrophic factor (BDNF). It is expressed as a decrease in synaptic strength of GABA-A transmission onto Purkinje neurons due to reduced transmembrane chloride gradient. By making whole-cell recordings, we found that the effect of BDNF is mediated by neuronal potassium and chloride transporter KCC2 because it is blocked by inhibitors of KCC2 or by raising the intracellular chloride concentration. Under these conditions in which KCC2 activity is reduced, BDNF augments evoked GABA-A currents suggesting a direct facilitatory effect of BDNF on GABA-A receptor. We also found that the effect of BDNF is highly localized at the GABA-A synapse and is secured by physical coupling between GABA-A receptor and KCC2, as revealed by coimmunoprecipitation studies. Based on these results, we hypothesize that the interaction between KCC2 and specific subunit of GABA-A receptor represents a fundamental mechanism rendering the rapid induction of ionic plasticity in individual or input-specific GABA synapses possible. Such a mechanism may be important for the function of Purkinje neurons that are known to express GABA-A receptors with different subunit compositions.
Dominant role of GABAB2 and Gbetagamma for GABAB receptor-mediated-ERK1/2/CREB pathway in cerebellar neurons

Abstract:gamma-aminobutyric acid type B (GABA(B)) receptor is an allosteric complex made of two subunits, GABA(B1) and GABA(B2). GABA(B2) plays a major role in the coupling to G protein whereas GABA(B1) binds GABA. It has been shown that GABA(B) receptor activates ERK(1/2) in neurons of the central nervous system, but the molecular mechanisms underlying this event are poorly characterized. Here, we demonstrate that activation of GABA(B) receptor by either GABA or the selective agonist baclofen induces ERK(1/2) phosphorylation in cultured cerebellar granule neurons. We also show that CGP7930, a positive allosteric regulator specific of GABA(B2), alone can induce the phosphorylation of ERK(1/2). PTX, a G(i/o) inhibitor, abolishes both baclofen and CGP7930-mediated-ERK(1/2) phosphorylation. Moreover, both baclofen and CGP7930 induce ERK-dependent CREB phosphorylation. Furthermore, by using LY294002, a PI-3 kinase inhibitor, and a C-term of GRK-2 that has been reported to sequester Gbetagamma subunits, we demonstrate the role of Gbetagamma in GABA(B) receptor-mediated-ERK(1/2) phosphorylation. In conclusion, the activation of GABA(B) receptor leads to ERK(1/2) phosphorylation via the coupling of GABA(B2) to G(i/o) and by releasing Gbetagamma subunits which in turn induce the activation of CREB. These findings suggest a role of GABA(B) receptor in long-term change in the central nervous system.

Abstract:Synaptic currents in immature Purkinje cells from rats on postnatal days 0–14 (P0–P14) were studied using whole-cell patch-electrodes applied to cerebellar slices (200 μm in thickness). Purkinje cells (held at −40 mV) showed excitatory postsynaptic currents (EPSCs) and inhibitory postsynaptic currents (IPSCs) spontaneously. From P2 to P12 the frequencies of miniature EPSCs and miniature IPSCs in the Purkinje cells increased by 10-fold or more, suggesting progressive formation of functional synapses during this period. Application of glycine (100 μM) to an immature Purkinje cell at P3–10 immediately increased the frequencies of both EPSCs and IPSCs. The effects of glycine showed maximum at P5–6 for EPSCs and at P9–10 for IPSCs and decreased thereafter. Facilitatory effects of glycine were suppressed by strychnine (1 μM), a specific blocker of the ionotropic glycine receptor, while the effects were also induced by other glycinergic agonists, including α-l-alanine (1 mM), l-serine (1 mM) and taurine (500 μM). The site of glycinergic effects was studied by removing the action potential generation in cerebellar slices. Following the addition of tetrodotoxin (TTX, 1 μM), the glycine-induced facilitation of EPSC almost disappeared, while that of IPSC remained (i.e. miniature IPSCs) and reached more than half of the value without TTX. These findings suggest that the ionotropic glycinergic receptors are expressed transiently but profoundly in the developing cerebellum, and that the distributions of these receptors causing excitation are different at excitatory and inhibitory presynaptic neurons. The glycine receptors may play distinct roles in the maturation and organization of cerebellar neural circuits.

Abstract:Glycine and γ-aminobutyric acid (GABA) are the major determinants of inhibition in the central nervous system (CNS). These neurotransmitters target glycine and GABAA receptors, respectively, which both belong to the Cys-loop superfamily of pentameric ligand-gated ion channels (pLGICs). Interactions of the neurotransmitters with the cognate receptors result in receptor opening and a subsequent influx of chloride ions, which, in turn, leads to hyperpolarization of the membrane potential, thus counteracting excitatory stimuli. The majority of glycine receptors and a significant fraction of GABAA receptors (GABAARs) are recruited and anchored to the post-synaptic membrane by the central scaffolding protein gephyrin. This ∼93 kDa moonlighting protein is structurally organized into an N-terminal G-domain (GephG) connected to a C-terminal E-domain (GephE) via a long unstructured linker. Both inhibitory neurotransmitter receptors interact via a short peptide motif located in the large cytoplasmic loop located in between transmembrane helices 3 and 4 (TM3-TM4) of the receptors with a universal receptor-binding epitope residing in GephE. Gephyrin engages in nearly identical interactions with the receptors at the N-terminal end of the peptide motif, and receptor-specific interaction toward the C-terminal region of the peptide. In addition to its receptor-anchoring function, gephyrin also interacts with a rather large collection of macromolecules including different cytoskeletal elements, thus acting as central scaffold at inhibitory post-synaptic specializations. Dysfunctions in receptor-mediated or gephyrin-mediated neurotransmission have been identified in various severe neurodevelopmental disorders. Although biochemical, cellular and electrophysiological studies have helped to understand the physiological and pharmacological roles of the receptors, recent high resolution structures of the receptors have strengthened our understanding of the receptors and their gating mechanisms. Besides that, multiple crystal structures of GephE in complex with receptor-derived peptides have shed light into receptor clustering by gephyrin at inhibitory post-synapses. This review will highlight recent biochemical and structural insights into gephyrin and the GlyRs as well as GABAA receptors, which provide a deeper understanding of the molecular machinery mediating inhibitory neurotransmission.

Abstract:Abstract GABA A receptors are ligand-gated chloride channels and ionotropic receptors of GABA, the main inhibitory neurotransmitter in vertebrates. In this review, we discuss the major and diverse roles GABA A receptors play in the regulation of neuronal communication and the functioning of the brain. GABA A receptors have complex electrophysiological properties that enable them to mediate different types of currents such as phasic and tonic inhibitory currents. Their activity is finely regulated by membrane voltage, phosphorylation and several ions. GABA A receptors are pentameric and are assembled from a diverse set of subunits. They are subdivided into numerous subtypes, which differ widely in expression patterns, distribution and electrical activity. Substantial variations in macroscopic neural behavior can emerge from minor differences in structure and molecular activity between subtypes. Therefore, the diversity of GABA A receptors widens the neuronal repertoire of responses to external signals and contributes to shaping the electrical activity of neurons and other cell types.
