Abstract:ConspectusTransition metal catalyzed asymmetric hydrofunctionalization of readily available unsaturated hydrocarbons presents one of the most straightforward and atom-economic protocols to access valuable optically active products. For decades, noble transition metal catalysts have laid the cornerstone in this field, on account of their superior reactivity and selectivity. In recent years, from an economical and sustainable standpoint, first-row, earth-abundant transition metals have received considerable attention, due to their high natural reserves, affordable costs, and low toxicity. Meanwhile, the earth-abundant metal catalyzed hydrofunctionalization reactions have also gained much interest and been investigated gradually. However, since chiral ligand libraries for earth-abundant transition-metal catalysis are limited to date, the development of highly enantioselective versions remains a significant challenge.This Account summarizes our recent efforts in developing suitable chiral ligands for iron and cobalt catalysts and their applications in the highly enantioselective hydrofunctionalization reactions (hydroboration and hydrosilylation) of alkenes and alkynes. In ligand design, we envisioned that chiral unsymmetric NNN-tridentate (UNT) ligand scaffolds could promote these enantioselective transformations with earth-abundant metals. Therefore, several types of chiral UNT ligands were designed and prepared in our laboratory, utilizing readily available natural amino acids as chiral sources. In the very beginning, chiral oxazoline iminopyridine (OIP) ligands were proposed and investigated through the rational combination of nitrogen-containing ligand scaffolds. After a systematic survey of the ligand effects, the imine moiety in the rigid OIP ligands was replaced by a conformationally more flexible amine unit, leading to the construction of reactive oxazoline aminoisopropylpyridine (OAP) ligands. Subsequently, imidazoline iminopyridine (IIP) and thiazoline iminopyridine (TIP) ligands were prepared by altering the oxygen atom of oxazoline with nitrogen and sulfur linkers, respectively. To further expand the chiral ligand library, other tridentate ligands containing a twisted pincer, anionic, and nonrigid backbone were also designed and synthesized, including iminophenyl oxazolinyl phenylamine (IPOPA) and imidazoline phenyl picolinamide (ImPPA). The efficacy of these chiral UNT ligands for asymmetric induction in iron and cobalt catalysis has been demonstrated through asymmetric hydrofunctionalization of alkenes and asymmetric sequential hydrofunctionalization of alkynes, which exhibit excellent reactivity as well as high chemo-, regio-, and stereoselectivity with broad functional group tolerance. Notably, highly regio- and enantioselective hydrofunctionalization of challenging substrates, such as 1,1-disubstituted aryl alkenes and terminal aliphatic alkenes, was also achieved. Furthermore, the development of asymmetric sequential isomerization/hydroboration of internal alkenes and sequential hydrofunctionalization of alkynes further demonstrates the synthetic power of these catalytic systems. The chiral enantioenriched products obtained by these methodologies could be potentially utilized in organic synthesis, medicinal chemistry, and materials science. We believe that our continuous efforts in this field would be beneficial to the development of asymmetric earth-abundant metal catalysis.

Abstract:Catalytic olefin hydroamination reactions are some of the most atom-economical transformations that bridge readily available starting materials–olefins and high-value-added amines. Despite significant advances in this field over the last two decades, the formal hydroamination of nonactivated aromatic compounds remains an unsolved challenge. Herein, we report the extension of olefin hydroamination to aromatic π-systems by using arenophile-mediated dearomatization and Cu-catalysis to perform 1,2-hydroamination on nonactivated arenes. This strategy was applied to a variety of substituted arenes and heteroarenes to provide general access to structurally complex amines. We conducted DFT calculations to inform mechanistic understanding and rationalize unexpected selectivity trends. Furthermore, we developed a practical, scalable desymmetrization to deliver enantioenriched dearomatized products and enable downstream synthetic applications. We ultimately used this dearomative strategy to efficiently synthesize a collection of densely functionalized small molecules.

Abstract:Highly regio- and enantioselective intermolecular hydroamination of alkenes is a challenging process potentially leading to valuable chiral amines. Hydroamination of alkenes via metal-catalyzed hydrogen atom transfer (HAT) with good regioselectivity and functional group tolerance has been reported, however, high enantioselectivity has not been achieved due to the lack of suitable ligands. Here we report a ligand-promoted cobalt-catalyzed Markovnikov-type selective radical hydroamination of alkenes with diazo compounds. This operationally simple protocol uses unsymmetric NNN-tridentate (UNT) ligand, readily available alkenes and hydrosilanes to construct hydrazones with good functional group tolerance. The hydrazones can undergo nitrogen-nitrogen bond cleavage smoothly to deliver valuable amine derivatives. Additionally, asymmetric intermolecular hydroamination of unactivated aliphatic terminal alkenes using chiral N-imidazolinylphenyl 8-aminoquinoline (IPAQ) ligands has also been achieved to afford chiral amine derivatives with good enantioselectivities.
NiH-Catalyzed Functionalization of Remote and Proximal Olefins: New Reactions and Innovative Strategies

Abstract:Transition metal hydride catalyzed functionalization of remote and proximal olefins has many advantages over conventional cross-coupling reactions. It avoids the separate, prior generation of stoichiometric amounts of organometallic reagents and the use of preformed organometallic reagents, which are sometimes hard to access and may compromise functional group compatibility. The migratory insertion of metal hydride complexes generated in situ into readily available alkene starting materials, the hydrometalation process, provides an attractive and straightforward route to alkyl metal intermediates, which can undergo a variety of sequential cross-coupling reactions. In particular, with the synergistic combination of chain-walking and cross-coupling chemistry of nickel, NiH-catalyzed functionalization of remote and proximal olefins has undergone particularly intense development in the past few years. This Account aims to chronicle the progress made in this arena in terms of activation modes, diverse functionalizations, and chemo-, regio-, and enantioselectivity.We first provide a brief introduction to the general reaction mechanisms. Taking remote hydroarylation as an example, the four oxidation states of Ni have allowed us to develop two different reaction strategies to form the final product: a Ni(I)-H/X-Ni(II)-H platform that relies on stoichiometric reductants and a Ni(I/II/III) cycle and a redox-neutral functional group or FG-Ni(II)-H platform that reacts with an alkene substrate and forms the migratory products via a Ni(0/II) pathway. We also demonstrate that diverse functionalization, including general C-C bond-forming reactions and the more challenging C-N/C-S bond-forming reactions could be realized. Moreover, the employment of appropriate chiral ligands has allowed us to successfully realize the corresponding asymmetric hydrofunctionalization reactions of olefins, including hydroalkylation, hydroarylation, hydroalkenylation, hydroalkynylation, and hydroamination. Interestingly, the enantio-determining step could be enantioselective hydronickelation, selective oxidative addition, or selective reductive elimination. To realize more challenging asymmetric migratory hydrofunctionalization, we have developed a general ligand relay catalytic strategy with a combination of two simple ligands, the first for chain-walking and the second for asymmetric coupling. This novel strategy avoids the design of a single, possibly structurally complex chiral ligand to promote both steps of chain-walking and asymmetric coupling. In addition, the success of multicomponent hydrofunctionalization provides a convenient approach to gain simple access to complex molecules. Finally, alkyl halides could be used as olefin precursors to undergo a variety of reductive migratory cross-electrophile coupling reactions. Applications of these remote hydrofunctionalization reactions are also discussed. We hope this Account will inspire future development in the field to overcome key challenges, including conceptually new catalytic strategies, development of high-performance systems with enhanced reactivity and selectivity, cutting-edge catalyst design, and further mechanistic studies.
Mimicking Hydrogen-Atom-transfer-like Reactivity in Copper-Catalysed Olefin Hydrofunctionalization

Abstract:The renaissance of catalytic metal hydride hydrogen atom transfer (MHAT) offers advanced tools for radical chemistry on simple olefins. While 3d transition metals like cobalt, iron and manganese have been extensively studied in catalytic MHAT, the potential of copper remains unexplored. This is due to the polar reactivity exhibited by classical nucleophilic Cu(I)-H. Here we report copper-catalysed MHAT-like oxidative hydrofunctionalization reactions. In contrast to conventional Cu(I)-H chemistry, the putative Cu-MHAT process produces alkyl radicals with high chemoselectivity and regioselectivity, which are subsequently captured by Cu(II) species to undergo coupling reactions with a broad scope of oxygen-, nitrogen-, halogen- and carbon-based nucleophiles. Preliminary results suggest viable extension to asymmetric catalysis and radical polymerization. This work offers a complementary oxidative MHAT platform. Cobalt, iron or manganese catalysts are the metals of choice in alkene functionalization reactions via catalytic metal hydride hydrogen atom transfer (MHAT). Now a complementary MHAT-like system based on copper is proposed to operate in a regioselective Markovnikov addition reaction of nucleophiles to olefins.

Abstract:Cu-catalyzed enantioselective Cu-B(pin) addition to 1,1-disubstituted alkenes generating tertiary alkylcopper complexes that contain a quaternary stereogenic center, followed by the protonation of the complexes are presented. The resulting formal hydroboration products are formed in up to 98% yield and 99:1 enantiomeric ratio. The C-B(pin) moiety can be converted to a variety of useful functional groups. The method is applied to the enantioselective formal synthesis of (R)-ketoprofen. Detailed stereochemical models for enantioselective induction were elucidated through DFT calculations.

Abstract:Transition metal-catalyzed direct C-H functionalization has drawn great attention in the past several decades owing to its advantages compared to conventional organic transformations, including higher atom-, step-and cost-economy and the avoidance of tedious prefunctionalization and waste emission. At the current stage, to make the C-H functionalization more applicable, chemists have devoted themselves to expanding the substrate and reaction scope. In the past decade, we exerted ourselves to develop new transformations based on direct C-H functionalization. In this minireview we report on our recent achievements on the addition of C-H bonds to carbonyls and imines. The addition of organometallic reagents, such as Grignard reagents, toward carbon-heteroatom double bonds is one of the most powerful reactions in organic synthesis to produce secondary and tertiary alcohols and amines. This chemistry is broadly used in both laboratory and industry. However, this powerful transformation suffers from some drawbacks: (1) the preparation of initial organohalides from easily available fossil feedstocks is tedious and sluggish; (2) substantial amounts of metal halide salts are emitted as waste; (3) last but not least, the manipulation of organometallic reagents is complicated due to their sensitivity to air and moisture. In contrast, direct insertion of polar double bonds to C-H bonds via transition-metal catalysis is ideal from the viewpoint of atom-, step-and cost-economy and the avoidance of the waste emission, as well as of the complex manipulation of sensitive reagents. Starting from this point, we made a commitment to this project years ago and have made credible achievements in this field. We first carried out Ir-catalyzed addition of pyridinyl C-H bonds to aldehydes promoted by silane, showing an unusual C-3 selectivity. Later on, we developed Rh-catalyzed addition of aryl C-H bonds with aldimines in the absence of any additives with directing strategy with highest atom-and step-economy. The mechanism was investigated in depth by the isolation of key intermediates and systematic thermodynamic and kinetic studies. Such a concept was expanded to the coupling of aryl/alkenyl C-H bonds with aldehydes and imines. Notably, a tandem process of relayed C-H activation/alkyne insertion/ cyclization between benzoates/benzimide and alkynes was developed, indicating the potential of the direct coupling of esters and amides with C-H bonds. Ideally, this strategy opens a new window to approach the ideal reactions to produce amines and alcohols from hydrocarbons.

Abstract:A selective copper(I)-catalyzed benzylic C(sp3)–H geminal difunctionalization reaction of a benzylic-type sp3 carbon was developed. This novel strategy allowed simultaneous introduction of amide and hydroxyl group in a highly selective and efficient way via successive oxidative intramolecular amidation and hydroxylation. This method was also applied to the synthesis of 3-hydroxy-2,3-dihydro-1H-pyrrolo[3,4-b]quinoxalinones from readily available 3-methyl-N-substituted quinoxaline-2-carboxamides in moderate to good yields. The five-membered cyclic hemiaminal moiety of the pyrrolo[3,4-b]quinoxalinones can serve as an intermediates for subsequent transformations into other useful functional groups.
Experimental–Computational Synergy for Selective Pd(II)-Catalyzed C–H Activation of Aryl and Alkyl Groups

Abstract:C-H activation and functionalization are on the forefront of modern synthetic chemistry. Imagine if any C-H bond of a molecule could be converted to a C-X bond, where X is the target functionality-this would alter the synthetic blueprints for complex target molecules since it would provide novel disconnections in retrosynthetic analysis. Collaborations between many experimental and computational groups have led to rapid developments of new C-H functionalization methods. Our groups represent an example of this; we were brought together as part of the NSF-supported Center for Selective C-H Functionalization. Many examples of experimental-computational synergy for selective Pd(II)-catalyzed C-H activation of aryl and alkyl groups are described in this Account. We describe computations by the Houk group made in response to experimental stimuli by the Yu group. The first section discusses the experimental and computational investigations of oxazoline-directed stereoselective Pd(II)-catalyzed C(sp3)-H bond activation that occurs through the concerted metalation-deprotonation (CMD) pathway involving a monomeric Pd(II) complex. The second section involves two types of bidentate ligands, mono-N-protected amino acid (MPAA) and acetyl-protected aminoethyl quinoline (APAQ) ligands that promote the C-H activation reactions with the ligand as the internal base. In the MPAA-assisted remote C-H bond activation, the basic dianionic amidate ligand participates in the deprotonation of a specific C-H bond. This mechanism accounts for the improved reactivity and selectivity in C-H activation reactions with MPAA ligands. The chiral APAQ ligands enable asymmetric palladium insertion into prochiral C-H bonds on a single methylene carbon center. The dianionic amidate of the APAQ ligand acts as an intramolecular base to deprotonate the methylene C-H asymmetrically and facilitate chiral Pd-C bond formation. The origins of the dramatic differences of five-membered (relatively inactive) and six-membered (highly active) chelation in β-methylene C(sp3)-H activation reactions by a Pd(II) catalyst were explained with density functional theory (DFT) calculations. This is mainly due to the steric repulsions between the ArF group of the substrate and the quinoline group of the ligand. The steric repulsion between the ArF group of the substrate and the quinoline group of the APAQ ligand destabilizes the five-membered chelate transition structure, increasing the energy of the transition state. The third section discusses a mechanism involving a Pd-Ag heterodimeric complex intermediate in the template-directed, Pd(II)-catalyzed remote meta functionalization of toluene derivatives and benzoic acid derivatives. The nitrile directing group of the template coordinates with Ag while the Pd is placed adjacent to the meta-C-H bond in the transition state, leading to the observed high meta selectivity. The selective activation of remote meta-C-H bonds at various distances can be achieved by tuning the template. The dual role of AgOAc as both an oxidant and part of the heteronuclear active species in the mechanism involving PdAg(OAc)3 was determined by DFT calculations and is in accord with literature information about complexes. For the systems discussed in these three sections, the similarity is that they all proceed via the CMD mechanism. The differences lie in the proton acceptors and the active Pd species. Common CMD involves a monomeric Pd mechanism with acetate as the proton acceptor. Both MPAA and APAQ ligands react via monomeric Pd mechanisms with a ligand moiety (the amidate oxygen) as the proton acceptor. Nitrile-containing template-mediated meta-C-H activations proceed via a Pd-Ag heterodimeric mechanism, still with acetate as the proton acceptor. The interaction between our two groups, experts in experiment and computation, and the discoveries made possible by that interplay are highlighted in this Account.
Copper-Catalyzed Carbonylative Coupling of Alkyl Halides

Abstract:Transition metal-catalyzed carbonylation reactions represent a direct and atom-economical approach to introduce oxygen functionality into organic compounds, with CO acting as an inexpensive and readily available C1 feedstock. Despite the long history of carbonylation catalysis, including many processes that have been industrialized at bulk scale, there remain several challenges to tackle. For example, noble metals such as Pd, Rh, and Ir are typically used as catalysts for carbonylation reactions, rather than earth-abundant alternatives. Additionally, while carbonylation of C(sp<sup>2</sup>)-hybridized substrates (e.g., aryl halides) is well-known, carbonylation of unactivated alkyl electrophiles, especially where β-hydride elimination can compete with desired CO migratory insertion at the catalyst site, remains challenging for many systems. Recently, base metal catalysis based on Mn, Co, and other metals has enabled advances in carbonylative coupling of alkyl electrophiles, though the nucleophiles are often limited to alcohols or amines to generate esters or amides as products. Thus, we have targeted base metal-catalyzed carbonylative C-C and C-E (E = N, H, Si, B) coupling reactions as a method for approaching diverse carbonyl compounds of synthetic importance.Initially, we designed a heterobimetallic catalyst platform for carbonylative C-C coupling of alkyl halides with arylboronic esters (i.e., carbonylative Suzuki-Miyaura coupling) to generate aryl alkyl ketones. Subsequently, we developed multicomponent carbonylation reactions of alkyl halides using NHC-Cu catalysts (NHC = N-heterocyclic carbene). These reactions operate by radical mechanisms, converting alkyl halides into either acyl radical or acyl halide intermediates that undergo subsequent C-C or C-E coupling at the Cu site. This mechanistic paradigm is relatively novel in the metal-catalyzed carbonylation area, allowing us to discover a previously unexplored chemical space in carbonylative coupling catalysis. We have successfully developed the following reactions: (a) hydrocarbonylative coupling of alkynes with alkyl halides; (b) borocarbonylative coupling of alkynes with alkyl halides; (c) reductive aminocarbonylation of alkyl halides with nitroarenes; (d) reductive carbonylation of alkyl halides; (e) carbonylative silylation of alkyl halides; (f) carbonylative borylation of alkyl halides. These reactions provide a broad range of valuable products including ketones, allylic alcohols, β-borylenones, amides, alcohols, acylsilanes, and acylborons in an efficient manner. Notably, the preparation of some of these products has previously required multistep syntheses, harsh conditions, or specialized reagents. By contrast, the multicomponent coupling platform that we have developed requires only readily available building blocks and rapidly increases molecular complexity in a single synthetic manipulation.

The distinct reactivity and selectivity observed in Cu-MHAT system is attributed to the involvement of Cu(II)–H species, a hydrogen atom donor that possesses substantially altered charge distribution and weakened bonding in contrast to conventional Cu(I) hydrides. Preliminary results suggest potential extension to asymmetric catalysis and radical polymerization are viable. This work opens up new opportunities for MHAT chemistry by going beyond the common oxidation states.

Abstract:There is ongoing interest in catalysis with the earth-abundant metal copper (Cu). This density functional theory (DFT) study explores the detailed workings of the first CuH-catalyzed hydroarylation of alkenes with polyfluoroarenes involving C–F bond functionalization. The CuH complex undergoes Markovnikov hydrocupration with the aryl alkene substrate to give a Cu(I) benzyl intermediate, and the regioselectivity is due to a combination of orbital and dispersion interactions induced by the aryl alkene substrate. The benzyl ligand allows a 1,3-cupratropic shift for the Cu(I) complex to have an exposed carbonanionic atom facilitating nucleophilic aromatic substitution to the polyfluoroarene substrate. The distinct regioselectivity of para-C–F substitution is controlled by the noncovalent C(sp3)–H/π and C(sp2)–H●●●O interactions. These mechanistic insights can have implications for the development of new transitional metal-catalyzed hydroarylation and C–F functionalization reactions.

Abstract:A highly enantio- and regioselective copper-catalyzed hydroamination reaction of alkenes has been developed using diethoxymethylsilane and esters of hydroxylamines. The process tolerates a wide variety of substituted styrenes, including trans-, cis-, and β,β-disubstituted styrenes, to yield α-branched amines. In addition, aliphatic alkenes coupled to generate exclusively the anti-Markovnikov hydroamination products.

Abstract:Carbonylation reactions have been widely used to construct carbonyl-containing molecules or carbon enhancement reactions, which are mostly catalyzed by noble metals (Pd, Rh, Ru, Ir). In this review, we introduce the copper-catalyzed carbonylation reactions that have been developed in our group. Abstract:We herein report the copper-catalyzed asymmetric conjugate addition of β-substituted alkenyl heteroarenes, the one of most challenging Michael acceptors, with alkenes as the latent nucleophiles. Diverse chiral heteroarenes bearing two vicinal stereocenters were obtained in good to excellent yields, generally excellent enantioselectivity, and a good level of diastereoselectivity. The products of the Michael addition can be readily transformed into other valuable acyclic enantioenriched structures bearing three contiguous stereocenters via chiral amine catalysis. Mechanistic studies, including an isotope labeling experiment, a nonlinear effect study, kinetic isotope effect experiments, and initial-rate kinetics studies, were implemented, and the experimental results indicated that the hydrocupration step might be the turnover-limiting step. Moreover, the origin of preferential alkene hydrocupration in the presence of β-substituted alkenyl heteroarenes and the Ph-BPE-ligated CuH catalyst was also elucidated via some control experiments.

Abstract:Density functional theory calculations have been performed to provide the unified mechanism of Cu(II)-catalyzed and amide-oxazoline (Oxa)-directed C(sp2)–H functionalization reactions. The common steps of the studied seven reactions (such as C–H bond vinylation, phenylation, trifluoromethylation, amination, alkynylation, and hydroxylation) are complexation, N–H and C–H bond deprotonation, and Cu(II)/Cu(II) → Cu(I)/Cu(III) disproportionation, leading to the Cu(III) intermediate. The mechanism of the studied C–H functionalization reactions, initiated from the Cu(III) intermediate, depends on the nature of coupling partners. With vinyl- or phenyl-Bpin, which bear no acidic proton (called as a Type-I reaction), the coupling partners are the in situ generated (by addition of anions) anionic borates, which coordinate to the Cu(III) intermediate and undergo concerted transmetalation and reductive elimination to form a new C–C bond. In contrast, with imidazole, aromatic amines, terminal alkyne, and water (called as a Type-II reaction), which bear an acidic proton, the real coupling partners are their in situ generated deprotonated derivatives, which coordinate to copper and lead to a final product with the C–Y bond (Y = C, N, and O) via the reductive elimination pathway. The C(sp2)–H bond trifluoromethylation with TMSCF3 is identified as a special case, positioned between the Type-I and Type-II reaction types. The real coupling partner of this reaction is the in situ generated (via the CF3–-to-OH– ligand exchange) CF3– anion that binds to the Cu(III) intermediate and undergoes the C–CF3 reductive elimination. Abstract:Transition metal catalyzed C-H bond activation have become one of the important tools for constructing new chemical bonds. Introducing directing group to the substrates are the key to successful reaction, these directing groups can also be further transformed in the reaction. As amidines with unique structure and reactivity, makes them ideal substrates for transition metal-catalyzed C-H transformations. This review describes the major progress and mechanistic investigations on C-H activation/annulation tandem reaction of amidines until early 2024, focusing on metal-catalyzed C-H activation of amidines with unsaturated compounds, such as alkynes, olefins vinylene carbonate, cyclopropanols and their derivatives. Meanwhile this manuscript also with a view to the reaction of amidines with diffierent carbene precursor, for example diazo compounds, azide, triazoles, pyriodotriazoles, sulfoxonium ylides. And its own C-H bond activation/cyclization reaction. A bright outlook was provided at the end of the manuscript.
