Abstract:Reconstructing past responses of coastal wetlands to climate change contextualizes ongoing and future developments in these globally important ecosystems. The molecular distributions and stable isotope ratios (delta H-2 and delta C-13) of sedimentary plant wax n-alkanes are frequently used to infer past vegetation and hydroclimate changes in wetland systems. However, there is limited modern information available about these compounds in subtropical wetlands. Here we analyzed mature leaves from 30 typical plant species and roots from 6 plant species collected in the Florida Everglades, including tree island plants, freshwater wetland plants, mangroves, and seagrass. The n-alkane abundance (2 to 884 mu g/g dry weight), percent of aquatic plants ratio (Paq, 0 to 1), average chain length (ACL(23-33), 24.0-30.7), concentration weighted average (CWA) delta H-2 (-231 to -78 parts per thousand) and delta C-13 values (-38.9 to -14.4 parts per thousand) spanned wide ranges with plant growth habit. Significant differences in n-alkane abundances, Paq, ACL(23-33), CWA delta H-2 and delta C-13 values were found to exist between the leaves and roots of some emergent aquatic plants. Simple mass balance calculations of wetland aquatic plants suggest that long chain n-alkanes (e.g., C-29 n-alkanes) are predominantly derived from leaves rather than roots in wetland surface sediments/soils. However, the contribution from mid-chain n-alkanes (e.g., C-23 n-alkane) from roots may be equal to or greater than those from leaves. This implies that the differences in the isotopic compositions between root and leaf derived material need to be taken into account when interpreting down core changes in mid-chain n-alkane delta H-2 and delta C-13 values, which may be derived from variable contributions from leaves and roots rather than a change in hydroclimate or vegetation. Considering the large variation in both n-alkane distribution proxies and isotopic composition, no single molecular index or stable isotope ratio can capture multivariate changes of wetland ecosystems in the past. Nevertheless, principal component analysis shows promising potential to resolve different plant functional types. Paleo-reconstruction of subtropical aquatic ecosystems using n-alkanes will be most useful if the full molecular and isotopic distribution information of plant waxes are used. (C) 2020 Elsevier Ltd. Leaf wax n-alkane distribution and hydrogen isotopic fractionation in fen plant communities of two Mediterranean wetlands (Tenaghi Philippon, Nisí fen—Greece)

Abstract:Many continental paleoclimate archives originate from wetland sedimentary sequences. While several studies have investigated biomarkers derived from peat-generating vegetation typical of temperate/boreal bogs (e.g., Sphagnum ), only scant information is available on emergent plants predominant in temperate/subtropical coastal marshlands, peri-lacustrine and fen environments. Here, we address this gap, focusing on two wetlands in the Mediterranean (Nisí fen and Tenaghi Philippon, Greece). We examined the concentration, homologue distribution, and hydrogen stable isotopic composition (δ 2 H) of leaf wax n -alkanes in 13 fen plant species, their surrounding soil, and surface water during the wet growing season (spring) and the declining water table period (summer). Our findings indicate that local graminoid species primarily contribute to the soil n -alkane signal, with a lesser influence from forbs, likely owing to differences in morphology and vegetation structure. The δ 2 H values of surface and soil water align with local average annual precipitation δ 2 H, reflecting winter-spring precipitation. Consistently, the average δ 2 H of local surface, soil, and lower stem water showed negligible evaporative enrichment, confirming minimal 2 H-fractionation during water uptake. We find that δ 2 H values of source water for wax compound synthesis in local fen plants accurately mirror local annual precipitation. Furthermore, despite differences between leaves and lower stems in n -alkane production rates, their δ 2 H values exhibit remarkable similarity, indicating a shared metabolic substrate, likely originating in leaves. Our net 2 H-fractionation values (i.e., precipitation to leaf n -alkanes) align with those in Chinese highlands and other similar environments, suggesting consistency across diverse climatic zones. Notably, our data reveal a seasonal decrease in the carbon preference index (CPI) in plant samples, indicating wax lipid synthesis changes associated with increased aridity. Additionally, we introduce a new parity isotopic difference index (PID) based on the consistent δ 2 H difference between odd and even n -alkane homologues. The PID demonstrates a strong anticorrelation with plant CPI, suggesting a potential avenue to trace long-term aridity shifts through δ 2 H analysis of odd and even n -alkane homologues in sedimentary archives. While further development of the PID is necessary for broad application, these findings highlight the intricate interplay between plant physiology, environmental parameters, and sedimentary n -alkanes in unravelling past climatic conditions.

Abstract:High resolution records for long chain n-alkane and n-alkanol biomarkers have been obtained by extraction of sediments from the profiles of Luochan (0–36ka) and Xunyi (0–170ka) on the Chinese Loess Plateau (CLP). The two records are similar where they overlap over the last 36ka. At Xunyi, distributions of the n-alkane and n-alkanol homologues remain relatively constant over the full 170kyr. The n-alkanes are characterized by high odd over even carbon number preference and predominance of the C31 n-alkane, whereas the n-alkanols are dominated by even over odd carbon numbers and by the C28 n-alkanol. An Alkane Index [A.I., C3l/(C29+C31)] and an Alkanol Index [Alc.I., C30/(C28+C30)] are both positively related with magnetic susceptibility of the sediment, with higher values during interglacials. A Lipid Index, L.I., averaging the normalized Alkane and Alkanol indices, gives a measure of changing vegetation for palaeo-environmental construction. A further useful index is provided by the δ13C value of the most abundant n-alkane, C31, after adjustment for the calculated effect of the estimated atmospheric pCO2 and precipitation. The record of this index over the last 170kyr, which varies from −36 to −32‰, indicates that the vegetation on the CLP has been mainly C3 plants and became even more so in the glacial periods (especially MIS 6 and 2) as the minor population of warm season grasses (C4) favored in interglacial times declined when temperatures fell. The lipid distributions and carbon isotope values are in general accord with the pollen and phytolith evidence recorded for the Yuan areas of the CLP in indicating steppe or meadow-steppe vegetation, with abundant cold season grasses (C3) and some herbs, bushes and trees, especially in glacial times.
Compositions and Isotopic Differences of Iso- and Anteisoalkanes in Black Mangroves (avicennia Germinans) Across A Salinity Gradient in A Subtropical Estuary

Abstract:Environmental contextMangroves dominate at the interface between land and sea, especially along tropical and subtropical coasts. To gain a better understanding of how mangroves respond to various environmental stress factors, we investigated the use of monomethylalkanes as potential chemical tracers for black mangroves. The application of these chemical tracers could elucidate how black mangroves respond to environmental stress such as sea level rise in mixed mangrove environments. AbstractA series of iso- and anteiso-monomethylalkanes (MMAs) with carbon numbers from C23 to C35 and C14 to C34 respectively were detected in Avicennia germinans. These compounds were present in varying amounts up to 54.1, 1.0 and 3.4µg g–1 dry weight in the leaves, bark and the crustose lichens attached to the bark of A. germinans respectively. These MMAs were not detected in the leaf waxes of Rhizophora mangle and Laguncularia racemosa, but were detected in significantly lower abundances (2–6% of that in A. germinans leaf wax) in the bark and lichen of R. mangle. Significant odd-carbon number distributions and even-carbon number distributions were observed for long chain (C ≥ 25) iso- (maximising at C31) and anteiso-MMAs (maximising at C32) respectively in A. germinans leaf wax. However, no obvious carbon number preferences were detected for bark and lichen. The long chain (LC) iso- and anteiso-MMAs in A. germinans leaf waxes were found to be enriched in 13C by 0.3–4.3 and 0.7–4.2 per mille (‰) compared to the n-alkanes with the same carbon numbers respectively across the salinity gradient of 19.7–32.0 practical salinity units (psu). In comparison, the LC iso- and anteiso-MMAs were found to be more depleted in D by 6.1–55.1 and 7.3–57.0 ‰ compared to the n-alkanes with same carbon numbers respectively. The results imply that A. germinans could be another important source of iso- and anteiso-alkanes in sediments and soils, and that these compounds could potentially be used as biomarkers for this species in mixed mangrove environments.
Characterization of Glossy Spike Mutants and Identification of Candidate Genes Regulating Cuticular Wax Synthesis in Barley (hordeum Vulgare L.)

Abstract:Cuticular waxes comprise the hydrophobic layer that protects crops against nonstomatal water loss and biotic and abiotic stresses. Expanding on our current knowledge of the genes that are involved in cuticular wax biosynthesis and regulation plays an important role in dissecting the processes of cuticular wax metabolism. In this study, we identified the Cer-GN1 barley (Hordeum vulgare L.) mutant that is generated by ethyl methanesulfonate mutagenesis with a glossy spike phenotype that is controlled by a single recessive nuclear gene. A physiological analysis showed that the total cuticular wax loads of Cer-GN1 were one-third that of the progenitor wild-type (WT), and its water loss rate was significantly accelerated (p < 0.05). In addition, Cer-GN1 was defective in the glume’s cuticle according to the toluidine blue dye test, and it was deficient in the tubule-shaped crystals which were observed on the glume surfaces by scanning electron microscopy. Using metabolomics and transcriptomics, we investigated the impacts of cuticular wax composition and waxy regulatory genes on the loss of the glaucous wax in the spikes of Cer-GN1. Among the differential metabolites, we found that 16-hydroxyhexadecanoic acid, which is one of the predominant C16 and C18 fatty acid-derived cutin monomers, was significantly downregulated in Cer-GN1 when it was compared to that of WT. We identified two novel genes that are located on chromosome 4H and are downregulated in Cer-GN1 (HvMSTRG.29184 and HvMSTRG.29185) that encode long-chain fatty acid omega-monooxygenase CYP704B1, which regulates the conversion of C16 palmitic acid to 16-hydroxyhexadecanoic acid. A quantitative real-time PCR revealed that the expression levels of HvMSTRG.29184 and HvMSTRG.29185 were downregulated at 1, 4, 8, 12, and 16 days after the heading stage in Cer-GN1 when it was compared to those of WT. These results suggested that HvMSTRG.29184 and HvMSTRG.29185 have CYP704B1 activity, which could regulate the conversion of C16 palmitic acid to 16-hydroxyhexadecanoic acid in barley. Their downregulation in Cer-GN1 reduced the synthesis of the cuticular wax components and ultimately caused the loss of the glaucous wax in the spikes. It is necessary to verify whether HvMSTRG.29184 and HvMSTRG.29185 truly encode a CYP704B1 that regulates the conversion of C16 palmitic acid to 16-hydroxyhexadecanoic acid in barley.
Compound Specific Δd and Δ13c Analyses As a Tool for the Assessment of Hydrological Change in a Subtropical Wetland

Abstract:Compound specific carbon and deuterium stable isotope values (δ13C and δD) and the relative abundance of mid-chain n-alkanes (Paq) were determined for a series of dominant wetland plants, a surface slough-to-ridge soil transect, and slough and ridge soil cores, to assess historical vegetation successions induced by hydrological modification in an anthropogenically impacted, subtropical wetland, the Florida Everglades, USA. A difference of as much as 3.6 and 130 ‰ in their δ13C and δD values was observed between the two most abundant emergent macrophyte species (Cladium and Eleocharis), respectively. A clear n-alkane δD value depletion (−130 to −167 ‰) and decreasing Paq was observed along the slough-to-ridge soil transect, likely the result of an eco-hydrological transition from slough-to-ridge dominated vegetation (Eleocharis to Cladium). In agreement with the relatively constant Paq values, the lack of significant changes in the δD depth profile for the slough core, suggest a consistent slough type of vegetation composition over time at that location. In contrast, changes of both n-alkane δ13C and δD values for the ridge core, especially after ~1960 AD, coincide with the expected plant successions from historically long hydroperiod (>8 months), slough type plants (Eleocharis, Utricularia, Nymphaea) to present day, shorter hydroperiod (<8 months), ridge type plants (Cladium). These δ13C and δD changes seem to be driven by vegetation shifts associated with hydrological change. The application of the compound-specific stable isotope determinations may strongly complement the biomarker approach for paleo-hydrological assessments in wetland ecosystems.
Mangrove Leaf Species-Specific Isotopic Signatures along a Salinity and Phosphorus Soil Fertility Gradients in a Subtropical Estuary

Abstract:Mangrove ecotypes are distinct monospecific or mix-species assemblages and used as classification criteria to evaluate coastal biogeochemical cycles at the local, regional, and global scales. However, it is not clear how plant nitrogen and carbon content, including bulk delta C-13 and delta N-1(5) and n-alkane delta C-13 values, vary across species and within species when plants are exposed to the interaction between nutrient (nitrogen-N, phosphorus-P) availability and stressors (i.e., salinity). Here we present significant differences in green leaves wax n-alkane delta(13)(delta C-13(n-alkane)) values and brown-senescent leaves C:N atomic ratios and total phosphorus (TP) concentrations of three mangrove species (Rhizophora mangle, Laguncularia racemosa, and Avicennia germinans) that reflect ecophysiological adaptations to nutrient availability and salinity along the Shark River estuary (SRE), South Florida, USA. Linear models between leaf wax delta(13)C(n-alkane )values and species location along TP fertility and salinity gradients showed distinct differences, particularly between the species A. germinans and R. mangle. Our analyses showed that leaf wax o delta C-13(n-alkane) properly represented major differences in ecophysiological responses by each mangrove species. We also found that both R. mangle and L. racemosa showed different isotopic footprints among the SRE upper, middle and lower estuarine salinity regions. Further, the green leaves bulk delta C-13 values in R. mangle (-32.3%o to -27.6%o) were positively correlated with distance from the mouth of the estuary. In contrast, L. racemosa showed a negative relationship with distance and a narrower bulk delta C-13 range (-29.8 parts per thousand to -28.1 parts per thousand) in comparison to the other two species. A. germinans, a species found only in the brackish (salinity: 18.8 +/- 1.2) and saline (30.3 +/- 0.53) estuarine regions, also showed a positive bulk delta C-13 relationship with distance. Because of the well-defined species-specific leaf wax n-alkane delta C-13 values along both water column/soil pore water salinity and TP gradients, we propose these values as a potential salinity proxy for paleoclimate reconstruction.

Abstract:Terrestrial leaf wax n-alkane biomarkers provide considerable insights into paleoenvironmental reconstruction. Over decades, a substantial number of field investigations were performed to constrain hydroclimatic factors that influence leaf wax n-alkane biomarkers to improve their utility for paleoenvironmental applications. However, a critical issue, the plant type effects, does exist which potentially affects the fidelity of leaf wax n-alkane biomarkers for paleohydroclimate calibration. Here we review the effects of plant types on terrestrial leaf wax n-alkane biomarkers from three aspects: leaf wax n-alkane distribution (wood vs. non-wood), hydrogen isotope (δ2Hwax; dicot vs. monocot) and carbon isotope (δ13Cwax; C3 vs. C4) biomarkers. Then we demonstrate the relationships between three forms of leaf wax n-alkane biomarkers, and provide examples of the cross-calibration among them in paleo-applications. The in-depth review of plant type effects on leaf wax n-alkane biomarkers will be helpful to interpret the hydroclimate and vegetation signals in the geologic past.

Abstract:Annually laminated sediments in Lake Xiaolongwan, northeast China, contain a suite of n-alkanes (C17–C33) with a strong odd over even carbon number predominance. Biomarker n-alkane proxies (average chain length, Paq ratio, grass/tree ratio, carbon preference index and compound-specific δ13C values) were used to reconstruct climate changes that occurred over the last millennium. Compound-specific δ13C values show large differences between the distinctive chain length groups of n-alkanes that originate from algae, aquatic macrophytes and terrestrial plants. Long-chain n-alkanes (C27–C33) are predominantly derived from leaf wax lipids in the forest. Variations in long-chain n-alkane δ13C values may mainly record water-use efficiency, inasmuch as the contribution from C4 plants is negligible in the Lake Xiaolongwan catchment. Short- and middle-chain n-alkanes are mainly from algae and aquatic plants. They are strongly depleted in 13C. This feature may be linked to a methane-derived, negative δ13C pool and lake overturn, which regulates dinoflagellate blooms. Parallel fluctuations are observed in δ13C27–31 values, Paq, and the grass/tree ratio throughout the record. Variations in δ13C27–31 values and Paq are in agreement with historical documents on summer and winter climate conditions. They support earlier suggestions that δ13C values in the long-chain n-alkanes and Paq may be useful indicators of effective precipitation or drought stress in this forested area. The δ13C27–31 index and Paq show distinct decadal variations. Periods with high δ13C27–31 values and a low Paq index correspond with the warm phases of the Pacific Decadal Oscillation (PDO). Values are reversed during PDO cool phases. At the decadal timescale, summer monsoon rainfall in northeast China over the last millennium may have been regulated mainly by the PDO.

Abstract:SUMMARY Plants synthesize natural products via lineage‐specific offshoots of their core metabolic pathways, including fatty acid synthesis. Recent studies have shed light on new fatty acid‐derived natural products and their biosynthetic pathways in disparate plant species. Inspired by this progress, we set out to develop tools for exploring the evolution of fatty‐acid derived products. We sampled multiple species from all major clades of euphyllophytes, including ferns, gymnosperms, and angiosperms (monocots and eudicots), and we show that the compositional profiles (though not necessarily the total amounts) of fatty‐acid derived surface waxes from preserved plant specimens are consistent with those obtained from freshly collected tissue in a semi‐quantitative and sometimes quantitative manner. We then sampled herbarium specimens representing 57 monocot species to assess the phylogenetic distribution and evolution, of two fatty acid‐derived natural products found in that clade: beta‐diketones and alkylresorcinols. These chemical data, combined with analyses of 26 monocot genomes, revealed a co‐occurrence (though not necessarily a causal relationship) between whole genome duplication and the evolution of diketone synthases from an ancestral alkylresorcinol synthase‐like polyketide synthase. Limitations of using herbarium specimen wax profiles as proxies for those of fresh tissue seem likely to include effects from loss of epicuticular wax crystals, effects from preservation techniques, and variation in wax chemical profiles due to genotype or environment. Nevertheless, this work reinforces the widespread utility of herbarium specimens for studying leaf surface waxes (and possibly other chemical classes) and reveals some of the evolutionary history of fatty acid‐derived natural products within monocots.

Leaf Wax N-Alkane Distributions in Arid Zone South African Flora: Environmental Controls, Chemotaxonomy and Palaeoecological Implications

Abstract:The environmental controls on leaf wax n-alkane distributions and associated interpretation of such distributions in geological archives have long remained rather enigmatic. Studies of contemporary vegetation often conflate changing environmental conditions and species differences between biomes, making it difficult to assess the extent to which variation is driven by plant adaptation to prevailing environmental conditions and/or more fixed chemotaxonomic patterns. We present a case study of arid and semi-arid regions of South Africa that considers these issues. We anticipate that such conditions may impart strong controls upon leaf wax synthesis.Leaf wax n-alkane data from 215 plants and 93 soils from the Succulent Karoo and Fynbos biomes of South Africa revealed yield and distributions to be highly variable. While many plants exhibited concentrations comparable with previous reports, several succulent plants produced n-alkane yield up to 100x the modal n-alkane concentration for their biome. The data demonstrate that, on average, leaf wax n-alkane distributions in the Succulent Karoo are different from those of the Fynbos biome, with the former associated with longer maximum chain length, less dispersed distributions and stronger odd numbered chain length preference. The patterns were closely mirrored in the soils. Average chain length was weakly correlated with climate and this is interpreted to be a function of multiple factors. We hypothesise that a key control is the shifting proportion of plant functional type between biomes, with a greater abundance of succulent growth forms in the more arid Succulent Karoo (which tended to produce longer and less dispersed n-alkane chain length distributions), differentiating this biome from the Fynbos, in which woody shrubs are more dominant. Inter-biome variability was also apparent, with some same taxonomic or functional groups common to both biomes tending to produce longer chain length n-alkane distributions under more arid conditions. There was, however, considerable individual plant variability and we observed both insensitivity to environmental conditions (i.e. consistent n-alkane distributions) and marked variability in chain length distributions.A key finding is the high yield of n-alkanes from multiple succulent plants, many of which use the CAM photosynthetic pathway. Compound specific delta C-13/delta D analyses (e.g. estimates of C-3 and C-4 biomass) for regions with potential succulent plant contributions should therefore be carried out cautiously. Heritability of the Structures and 13C Fractionation in Tomato Leaf Wax Alkanes: A Genetic Model System to Inform Paleoenvironmental Reconstructions

Abstract:Leaf wax n-alkanes are broadly used to reconstruct paleoenvironmental information. However, the utility of n-alkanes as a paleoenvironmental proxy may be modulated by the extent to which biological as well as environmental factors influence the structural and isotopic variability of leaf waxes. In paleoclimate applications, there is usually an implicit assumption that most variation of leaf wax traits through a time series can be attributed to environmental change and that biological sources of variability within plant communities are small. For example, changes in hydrology affect the δ2H of waxes via rainwater and the δ13C of leaf waxes by changing plant communities. We measured the degree of genetic control over δ13C variation in leaf waxes within closely related species with an experimental greenhouse growth study. We measured the proportion of variability in structural and isotopic leaf wax traits that is attributable to genetic variation using a set of 76 introgression lines (ILs) between two interfertile Solanum (tomato) species: S. lycopersicum cv M82 (hereafter cv M82) and S. pennellii. Leaves of S. pennellii, a wild desert tomato relative, produced significantly more iso-alkanes than cv M82, a domesticated tomato cultivar adapted to water-replete conditions. We report a methylation index to summarize the ratio of branched (iso- and anteiso-) to total alkanes. Between Solanum pennellii and cv M82, the iso-alkanes were found to be enriched in 13C by 1.2–1.4‰ over n-alkanes. The broad-sense heritability values (H2) of leaf wax traits describe the degree to which genetic variation contributes to variation of these traits. Variation of individual carbon isotopic compositions of alkanes were of low heritability (H2 = 0.13–0.19), suggesting that most variation in δ13C of leaf waxes in this study can be attributed to environmental variance. This supports the interpretation that variation in the δ13C of wax compounds recorded in sediments reflects paleoenvironmental and vegetation changes. Average chain length (ACL) values of n-alkanes were of intermediate heritability (H2 = 0.30), suggesting that ACL values are more strongly influenced by genetic cues.

Abstract:Summary The plant cuticle is a hydrophobic barrier, which seals the epidermal surface of most aboveground organs. While the cuticle biosynthesis of angiosperms has been intensively studied, knowledge about its existence and composition in nonvascular plants is scarce. Here, we identified and characterized homologs of Arabidopsis thaliana fatty acyl‐CoA reductase (FAR) ECERIFERUM 4 (AtCER4) and bifunctional wax ester synthase/acyl‐CoA:diacylglycerol acyltransferase 1 (AtWSD1) in the liverwort Marchantia polymorpha (MpFAR2 and MpWSD1) and the moss Physcomitrium patens (PpFAR2A, PpFAR2B, and PpWSD1). Although bryophyte harbor similar compound classes as described for angiosperm cuticles, their biosynthesis may not be fully conserved between the bryophytes M. polymorpha and P. patens or between these bryophytes and angiosperms. While PpFAR2A and PpFAR2B contribute to the production of primary alcohols in P. patens, loss of MpFAR2 function does not affect the wax profile of M. polymorpha. By contrast, MpWSD1 acts as the major wax ester‐producing enzyme in M. polymorpha, whereas mutations of PpWSD1 do not affect the wax ester levels of P. patens. Our results suggest that the biosynthetic enzymes involved in primary alcohol and wax ester formation in land plants have either evolved multiple times independently or undergone pronounced radiation followed by the formation of lineage‐specific toolkits.

Disentangling Temperature Effects on Leaf Wax N-Alkane Traits and Carbon Isotopic Composition from Phylogeny and Precipitation

Abstract:Leaf wax n-alkanes are terrestrial plant biomarkers that have characteristics that are widely employed as proxies for climatic conditions. Understanding the relationship between different environmental factors to the amounts and types of leaf wax n-alkanes in modern plants is crucial for the application of these proxies to paleoenvironmental reconstructions. However, the effects of climate conditions on plant wax characteristics remain complicated due to the interactions among temperature, precipitation and phylogeny. To evaluate the effect of temperature with minimized interfering factors, we collected 106 Artemisia plants across a 15 degrees C mean annual temperature gradient along the 400 mm isohyet in China. Both total n-alkane concentration (Sigma alk) and carbon preference index (CPI) varied greatly but did not correlate with temperature. Average chain length (ACL) increased with temperature, especially summer temperature (T-JJA , June-August), indicating that ACL could be used as proxy for temperature. The stable carbon isotope compositions of n-C-27 , n-C-29 and n-C-31 were very similar in each plant (-38.2 parts per thousand, to -30.0 parts per thousand, -39.0 parts per thousand, to -30.2 parts per thousand, -38.7 parts per thousand, to -30.5 parts per thousand, respectively), which reflects a similar biosynthetic process for all three n-alkane homologues of Artemisia. There was a positive relationship between delta(13) C of bulk leaf tissues (delta C-13(b)ulk ) and of n-alkanes (delta C-13(alk)), and the average offset of delta C-13(29) relative to delta C-13(bulk) (epsilon c29/bulk) was -7.1 parts per thousand. Increasing trends in both delta C-13(bulk) and delta C-13(29) were found with temperature. However, correlation of delta C-13(b)ulk with temperature (R-2 = 18%) was much weaker than that of delta C-13(29) with temperature (R-2 = 60%). Abstract:-The plant cuticle, multifunctional hydrophobic air-to-plant boundary, vary dramatically among plant species and responds dynamically to environmental and biotic factors. These dynamics, certainly driven by cost-benefit selection, are poorly understood up to now. -13C labelling and compound-specific isotope analysis open a new avenue to study the cuticle dynamics. We studied side specific leaf cuticular wax regeneration, in different compounds, and separately for intra-(IW) and epicuticular wax (EW), as well as the effect of epicuticular wax removal on the regeneration rate. -Mature leaves that reached the final area before the start of the experiment deposited new IW at first, but quickly equilibrated with EW. Removal of EW accelerated this equilibration but not the net rate of new wax deposition. The n-alkanes had the fastest turnover. Pentacyclic triterpenoids (ursolic acid) had a surprisingly slow turnover due to the large pool but also slow deposition rate. Adaxial and abaxial leaf surfaces exhibited small but consistent differences. -We demonstrated for the first time that EW removal does not affect wax deposition rate in evergreen leaf cuticle. We also confirmed that collodion discriminates between EW and IW well and does not inhibit future leaf metabolism and wax deposition in our species.

Abstract:Long chain n-alkanes are plant wax biomarkers that are widely used in paleoenvironmental reconstructions. Temperature is an important influence on the composition of leaf wax n-alkanes. However, to date, understanding of changes in the abundance and distribution of leaf wax n-alkanes with temperature remains unclear. The main reason is that the interference of precipitation on leaf wax n-alkanes has not been effectively removed in most investigations of natural ecosystems. We sampled 357 plants across a temperature gradient along an isohyet in China to minimize the influence of precipitation and evaluate the effect of temperature on the abundance and distribution of leaf wax n-alkanes. Most plant functional groups showed similar distributions in total n-alkane concentration (Sigma alk), carbon preference index (CPI) and average chain length (ACL). CPI values were not sensitive to temperature change. Sigma alk values were negatively correlated with summer mean temperature (T-JJA, June to August) for most plant functional groups. ACL values were always positively related to temperature, especially T-JJA. However, large variability in Sigma alk and ACL occurred within plants from the same site. Therefore, this study suggests that Sigma alk and ACL can indicate relative environmental temperature, but they should not be used as quantitative proxies of paleo-temperature. (C) 2018 Elsevier Ltd. Lipid biomarkers and stable isotopes uncover paleovegetation changes in extremely species-rich forest-steppe ecosystems, Central Europe

Abstract:The historical development of the vegetation of semi-dry grasslands in Central Europe is not satisfactorily understood. Long-term continuity of open vegetation or, conversely, deep-past forest phases are considered possible sources of the current extreme species diversity of these ecosystems. We aimed to reveal the trajectory of paleovegetation development in these ecosystems through detailed analysis of terrestrial in-situ soil geoarchives. We measured the bulk soil carbon and nitrogen contents, lipid molecular distribution, and compound-specific stable carbon and hydrogen isotopic signatures of mid- and long-chain n-alkanes extracted from soil and modern plant material tissues (i.e., deciduous and Pinus leaves and grass/herbaceous species). The C23-C33 n-alkane homologues were identified in soils with different abundances. Normally, C27 and C29 n-alkanes were the most abundant homologues in tree-leaf samples, while grass-derived n-alkanes were mostly C31 and C33 homologues. Soils were largely dominated by C29 and C31 n-alkanes. Odd-numbered C27-C33 soil n-alkane δ13C values ranged from -36.2‰ to -23.2‰, whereas their δ2H values showed a wider range of variability that fluctuated from -224‰ to -172‰. Molecular distribution in combination with radiocarbon analysis of soil organic matter (SOM) and δ13C and δ2H values of n-alkanes revealed a large contribution of C3 trees (both deciduous and coniferous trees/pine trees) as the main source of n-alkanes between the late Pleistocene and early Holocene (ca 15,000-8200 calibrated year before present/cal year BP). A clear shift toward more grassy/herbaceous vegetation was observed from the early Holocene (ca 11,700-8200 cal year BP) onwards. Distribution patterns of lipids and soil geochemical parameters showed that plants are the main source of SOM and that biodegradation and kinetic isotope fractionation are not the main reasons for 13C enrichment in soil profiles. Past C3 vegetation shifts as well as paleoclimate changes (i.e., aridity) can have played a role in the observed 13C depth profiles.

Abstract:Fallen leaves and their decomposition directly deposit leaf wax n-alkanes into sediments, which can be used to identify local flora. These n-alkanes are important for studying past vegetation and climate, but their distribution in sediments must be known. Aeolian sand n-alkanes are particularly important for understanding paleoclimates in arid regions, despite the challenges of extraction due to their extremely low abundance. To investigate the preservation of plant leaf wax n-alkanes in deserts, we analyzed n-alkanes in aeolian sands from the Northern Ulan Buh Desert (UBD), China, and compared them to the surrounding vegetation. We calculated the total n-alkane concentration (ΣALK), average chain length (ACL21–35), and carbon preference index (CPI21–35). In the Northern UBD, aeolian sand n-alkanes have lower ΣALK, indicating microbial degradation. The eastern aeolian sand has lower CPI21–35 and ACL21–35 than the adjacent vegetation, whereas the western sand values are consistent with the plants, likely due to the transport of plant-derived materials by wind and water from the nearby mountains. Our study shows that sedimentary n-alkane signatures are not only determined by local vegetation but also influenced by environmental factors like temperature and precipitation. Additionally, local deposition processes play a significant role in determining the properties of these n-alkanes.

Abstract:We present an 8000-year biomarker and stable carbon isotope record from the Verlorenvlei Estuary, South Africa. We assessed how leaf wax lipids, insoluble macromolecular organic matter, bulk C/N data and compound-specific stable carbon isotopes were linked to the site’s palynological record and to evidence for regional sea level and environmental change. Down-core trends in bulk δ 13 C are closely coupled to trends in pollen types from saline-tolerant taxa. These trends are mirrored by variations in the incorporation of reduced sulphur into macromolecular organic matter. This process, quantified with the thiophene ratio, is closely associated with periods of higher sea level 8,000–4,300 cal yr BP. We propose the thiophene ratio is a proxy for relative marine influence within (peri) estuarine sediments. All measured variables indicate differences between early-middle Holocene (8,000–4,300 cal BP) and late Holocene conditions at Verlorenvlei. The former period was more saline and preserves more labile macromolecular organic matter. Marine influence declined after 4,300 cal yr BP, and although the abundance of short-chain-length n -alkanes suggests continued presence of wetland flora until 2,500 cal yr BP, organic matter preservation became poorer and a drying trend was inferred, most notably for the interval 2,500–900 cal BP. Increasing freshwater inundation is apparent during the last 700 cal yr, consistent with several records from this region. Leaf wax n -alkane distributions are largely uncorrelated with bulk organic matter variables, with the exception of the abundance of C 31 and C 33 n -alkanes, which are negatively correlated with δ 13 C TOC . Furthermore, C 31 –C 33 n -alkane δ 13 C values are uncorrelated with C 23 –C 29 δ 13 C and δ 13 C TOC . They are also higher than our newly measured terrestrial (C 3 ) vegetation C 29 and C 31 end-member values of −35 ± 2 and −34 ± 1 ‰, respectively. These patterns are best explained by a dominant contribution of local riparian vegetation to the C 23 –C 29 n -alkanes, but time-varying contributions of non-local leaf waxes to the C 31 –C 33 signals. This renders inferences concerning regional environmental change from long-chain leaf waxes potentially challenging in this setting.
