Abstract:Red-skinned pear is a promising commercial fruit due to its attractive appearance and nutritious value. Anthocyanin is the determinant of the red coloration of the pear peel. However, differences in anthocyanin accumulation exist among red pear cultivars with different genetic backgrounds. In this study, we analyzed the anthocyanin content and gene expression patterns in the fruits and different tissues of the red pear 'Red Zaosu' at different developmental stages and found a difference in anthocyanin accumulation between 'Red Zaosu' pear and its green mutant. The data showed that the expression profiles of transcripts that encoded critical anthocyanin biosynthetic genes were basically consistent with a tendency to a decreased anthocyanin content during fruit development, indicating that a synergistic effect of these genes was responsible for anthocyanin biosynthesis and regulation. Tissue-specific expression analysis of anthocyanin biosynthetic genes showed that they could be expressed in all tissues but at different levels. PbF3H, PbDFR, and PbANS were mainly expressed during the early flowering period, which explained the reduced levels of anthocyanin content in petals. Additionally, the content of anthocyanins and the expression levels of PbDFR, PbANS, and PbMYB10 significantly decreased in the green mutant of 'Red Zaosu', suggesting that PbDFR, PbANS, and PbMYB10 probably play a decisive role in determining the skin coloration of 'Red Zaosu' and its green mutant.
PpMYB36 Encodes a MYB-Type Transcription Factor That Is Involved in Russet Skin Coloration in Pear (Pyrus pyrifolia)

Abstract:Fruit color is one of the most important external qualities of pear (Pyrus pyrifolia) fruits. However, the mechanisms that control russet skin coloration in pear have not been well characterized. Here, we explored the molecular mechanisms that determine the russet skin trait in pear using the F-1 population derived from a cross between russet skin ('Niitaka') and non-russet skin ('Dangshansu') cultivars. Pigment measurements indicated that the lignin content in the skin of the russet pear fruits was greater than that in the non-russet pear skin. Genetic analysis revealed that the phenotype of the russet skin pear is associated with an allele of the PpRus gene. Using bulked segregant analysis combined with the genome sequencing (BSA-seq), we identified two simple sequence repeat (SSR) marker loci linked with the russet-colored skin trait in pear. Linkage analysis showed that the PpRus locus maps to the scaffold NW_008988489.1: 53297-211921 on chromosome 8 in the pear genome. In the mapped region, the expression level of LOC103929640 was significantly increased in the russet skin pear and showed a correlation with the increase of lignin content during the ripening period. Genotyping results demonstrated that LOC103929640 encoding the transcription factor MYB36 is the causal gene for the russet skin trait in pear. Particularly, a W-box insertion at the PpMYB36 promoter of russet skin pears is essential for PpMYB36-mediated regulation of lignin accumulation and russet coloration in pear. Overall, these results show that PpMYB36 is involved in the regulation of russet skin trait in pear.
The Red Sport of 'zaosu' Pear and Its Red-Striped Pigmentation Pattern Are Associated with Demethylation of the PyMYB10 Promoter.

Abstract:'Zaosu' pear, a hybrid of Pyrus pyrifolia and Pyrus communis, is a popular cultivar developed in China. 'Zaosu Red' is a bud sport of 'Zaosu' with red shoots, young leaves, and fruit. After grafting of 'Zaosu Red', reverse mutations in some branches lead to a loss of colour in leaves and stems. Also, the mature fruit of 'Zaosu Red' exhibits two phenotypes; fully red and striped. The aim of this study was to establish the mechanism of the red colour mutation in 'Zaosu' and the striped pigmentation pattern in fruit of 'Zaosu Red'. The accumulation of anthocyanins and transcript levels of the genes PpUFGT2 and PyMYB10 were highly correlated. The open reading frames (ORF) and promoter regions of these two key genes were cloned and compared between 'Zaosu' and its bud sports, but no sequence differences were found. The R2R3 MYB, PyMYB10, can activate expression of genes encoding enzymes of the anthocyanin biosynthetic pathway. A yeast one-hybrid assay showed that PyMYB10 was associated with the -658 to -172 bp fragment of the PpUFGT2 promoter, probably via a MYB binding site (MBS) located at -466 bp. The PyMYB10 promoter had lower methylation levels in anthocyanin-rich tissues, indicating that the red bud sport of 'Zaosu' pear and the striped pigmentation pattern of 'Zaosu Red' pear are associated with demethylation of the PyMYB10 promoter. Abstract:Anthocyanins are one of highly attractive characteristics contributing attractive color and antioxidant properties in fruits. During pear ripening, phenylalanine ammonialyase (PAL) was pivotal enzyme involved in anthocyanin biosynthesis, which could be served as a determinant attribute of fruit quality. Results showed that a lower level of anthocyanin accumulation limited to the peel tissue in Red Bartlett, whereas higher amounts of anthocyanins were accumulated in both peel and flesh of Red d'Anjou. The Pyrus communis PAL (PcPAL) gene was cloned and its expression in different pear fruit tissues was analyzed. The PcPAL expression was shown to be tissue dependent and in both cultivars its increase was at similar rates in the early ripening period. Nonetheless, the PcPAL expression diminished at the end of Red Bartlett ripening, while it increased markedly in Red d'Anjou. The PcPAL expression was higher and displayed no correlation with the anthocyanin accumulation in the flesh of Red Bartlett pear. And the higher PcPAL expression profile could be related to the increased anthocyanin accumulation observed in Red d'Anjou.
Transcriptome profiling reveals differential gene expression in proanthocyanidin biosynthesis associated with red/green skin color mutant of pear (Pyrus communis L.)

Abstract:Anthocyanin concentration is the key determinant for red skin color in pear fruit. However, the molecular basis for development of red skin is complicated and has not been well-understood thus far. "Starkrimson" (Pyrus communis L.), an introduced red pear cultivated in the north of China and its green mutant provides a desirable red/green pair for identification of candidate genes involved in color variation. Here, we sequenced and annotated the transcriptome for the red/green color mutant at three stages of development using Illumina RNA-seq technology. The total number of mapped reads ranged from 26 to 46 million in six libraries. About 70.11-71.95% of clean reads could be mapped to the reference genome. Compared with green colored fruit, a total of 2230 differentially expressed genes (DEGs) were identified in red fruit. Gene Ontology (GO) terms were defined for 4886 differential transcripts involved in 15 Kyoto Encyclopedia of Genes and Genomes (KEGG) pathways. Three DEGs were identified as candidate genes in the flavonoid pathway, LAR, ANR, and C3H. Tellingly, higher expression was found for genes encoding ANR and LAR in the green color mutant, promoting the proanthocyanidin (PA) pathway and leading to lower anthocyanin. MYB-binding cis-motifs were identified in the promoter region of LAR and ANR. Based on these findings, we speculate that the regulation of PA biosynthesis might be a key factor for this red/green color mutant. Besides the known MYB and MADS transcription families, two new families, AP2 and WRKY, were identified as having high correlation with anthocyanin biosynthesis in red skinned pear. In addition, qRT-PCR was used to confirm the transcriptome results for 17 DEGs, high correlation of gene expression, further proved that AP2 and WARK regulated the anthocyanin biosynthesis in red skinned "Starkrimson," and ANR and LAR promote PA biosynthesis and contribute to the green skinned variant. This study can serve as a valuable new resource laying a solid foundation for functional gene identification in the anthocyanin pathway of red-skinned pear and provide a good reference for relevant research on molecular mechanisms of color variation in other pear species.

Abstract:Red pear is a popular fruit that is appreciated for its attractive and distinctive appearance and mild flavor. In this study, we investigated the mechanism underlying the red coloration of pear skin using the ‘Xinqihong’ cultivar—which was selected as a spontaneous bud sport mutant of the ‘Xinli 7′(Pyrus betulifolia Rehd.) variety and has a stronger red color that is retained in the mature fruit—as an experimental model. We compared the phenotype and gene expression patterns of the two varieties and found no significant differences at the early stage of fruit development. However, although the red color of ‘Xinli 7′ fruits began to fade 107 days after full bloom, that of ‘Xinqihong’ fruits persisted until the time of harvest. Transcriptome sequencing identified 639 genes that were differentially expressed between the two varieties, including genes related to light, calcium, and hormone signaling (e.g., PbPIF3, PbGA2ox, and the calmodulin related genes). Moreover, anthocyanin biosynthesis genes were downregulated as the red color of ‘Xinli 7′ fruits faded. These results provide insight into the molecular basis of color differences in red pears that can be useful for improving its fruit quality and commercial value.

Abstract:Fruit skin color is an important parameter of outer quality and plays an important role in attracting customers. In many plants, it is the result of coordinative regulation of anthocyanin pathway genes. In our study, the differential expression of cDNA library in a pair of pear mutant with red and green color was investigated to find candidate genes which might regulate the anthocyanin biosynthesis and control the coloration of pear. We constructed a cDNA library using the cDNA-amplified fragment length polymorphism approach to analyze the transcriptional differences between the original cultivar “Early red Doyenne du Comice” with high anthocyanin content in the peel and its green color mutant with comparatively low anthocyanin content. Altogether, 47 transcript-derived fragments, putatively involved in anthocyanin biosynthesis, primary metabolism, stress, and defense responses, were identified. The relationships of differentially expressed genes and coloration were investigated by quantitative real-time PCR with fruit skin samples at different developmental stages. A gene putatively involved in anthocyanin biosynthesis was found and named as PyMADS18. Its sequence is similar to genes reported in the literature as regulators of anthocyanin biosynthesis. The expression results indicate that PyMADS18 is likely to be involved in anthocyanin accumulation and regulation of anthocyanin synthesis in early fruit development of pear.

Abstract:Pears with red skin are attractive to consumers and provide additional health benefits. Identification of the gene(s) responsible for skin coloration can benefit cultivar selection and breeding. The use of QTL-seq, a bulked segregant analysis method, can be problematic when heterozygous parents are involved. The present study modified the QTL-seq method by introducing a |Δ(SNP-index)| parameter to improve the accuracy of mapping the red skin trait in a group of highly heterozygous Asian pears. The analyses were based on mixed DNA pools composed of 28 red-skinned and 27 green-skinned pear lines derived from a cross between the ‘Mantianhong’ and ‘Hongxiangsu’ red-skinned cultivars. The ‘Dangshansuli’ cultivar genome was used as reference for sequence alignment. An average single-nucleotide polymorphism (SNP) index was calculated using a sliding window approach (200-kb windows, 20-kb increments). Nine scaffolds within the candidate QTL interval were in the fifth linkage group from 111.9 to 177.1 cM. There was a significant linkage between the insertions/deletions and simple sequence repeat markers designed from the candidate intervals and the red/green skin (R/G) locus, which was in a 582.5-kb candidate interval that contained 81 predicted protein-coding gene models and was composed of two subintervals at the bottom of the fifth chromosome. The ZFRI 130-16, In2130-12 and In2130-16 markers located near the R/G locus could potentially be used to identify the red skin trait in Asian pear populations. This study provides new insights into the genetics controlling the red skin phenotype in this fruit.

Abstract:With the progenies produced from the cross Pyrus pyrifolia Nakai ‘Whangkeumbae’× Pyrus bretschneideri Rehd.‘Dangshansu’,the method of bulked segregant analysis(BSA) was used to identify molecular markers linked to the trait of fruit russet skin in pear.Two hundred and eighty one pairs of SSR primers derived from pear and apple genome were screened.Two SSR markers,CH01c06 and Hi20b03,which linked to russet skin of fruit at a distance of 4.8 cM and 12.0 cM,respectively,were obtained.This suggests that the gene which determines fruit russet skin be located on LG8 of the pear consensus genetic map.

Abstract:Genetic variation contributes to the phenotypic diversity of plants. Most red pear cultivars that accumulate anthocyanins originated from somatic variation and are termed bud sports. ‘Hongzaosu’ is a bud sport derived from the green pear ‘Zaosu’. A new genetic map was constructed from a population derived from the cross of ‘Yuluxiang’ and ‘Hongzaosu’, from which PpBBX24 was identified as a crucial gene controlling anthocyanin accumulation. Genetic and phylogenetic evidence revealed that PpBBX24 is a repressor of anthocyanin biosynthesis in pear. A 14 bp deletion was detected in the third exon of PpBBX24 in ‘Hongzaosu’, resulting in premature termination of protein translation. The truncated PpBBX24 protein was a positive regulator of anthocyanin biosynthesis by activating the transcription of PpCHS and PpUFGT. Three independent variations in the BBX24 coding region that led to premature termination of translation were detected in other pear bud sports and the progeny of a bud sport that all accumulate anthocyanins. The genome of ‘Hongzaosu’ was assembled using both long-read and short-read sequences. By combining genomic and transcriptomic data, allele-specific expression of PpMYB110a was observed in the fruit skin of ‘Hongzaosu’, which was likely caused by large variation in the promoter. This work enhances understanding of coloration in red pears and provides a novel genomic resource for further studies on ‘Hongzaosu’ pear.
PbGA2ox8 induces vascular-related anthocyanin accumulation and contributes to red stripe formation on pear fruit

Abstract:Abstract Fruit with stripes, which are generally longitudinal, can occur naturally, but the bioprocesses underlying this phenomenon are unclear. Previously, we observed an atypical anthocyanin distribution that caused red-striped fruit on the spontaneous pear bud sport “Red Zaosu” ( Pyrus bretschneideri Rehd.). In this study, comparative transcriptome analysis of the sport and wild-type “Zaosu” revealed that this atypical anthocyanin accumulation was tightly correlated with abnormal overexpression of the gene-encoding gibberellin (GA) 2-beta-dioxygenase 8, PbGA2ox8 . Consistently, decreased methylation was also observed in the promoter region of PbGA2ox8 from “Red Zaosu” compared with “Zaosu”. Moreover, the GA levels in “Red Zaosu” seedlings were lower than those in “Zaosu” seedlings, and the application of exogenous GA 4 reduced abnormal anthocyanin accumulation in “Red Zaosu”. Transient overexpression of PbGA2ox8 reduced the GA 4 level and caused anthocyanin accumulation in pear fruit skin. Moreover, the presence of red stripes indicated anthocyanin accumulation in the hypanthial epidermal layer near vascular branches (VBs) in “Red Zaosu”. Transient overexpression of PbGA2ox8 resulting from vacuum infiltration induced anthocyanin accumulation preferentially in calcium-enriched areas near the vascular bundles in pear leaves. We propose a fruit-striping mechanism, in which the abnormal overexpression of PbGA2ox8 in “Red Zaosu” induces the formation of a longitudinal array of anthocyanin stripes near vascular bundles in fruit.

Abstract:Skin color is an important factor in pear breeding programs. The degree of red coloration is determined by the content and composition of anthocyanins. In plants, many MYB transcriptional factors are involved in regulating anthocyanin biosynthesis. In this study, a R2R3-MYB transcription factor gene, PyMYB10, was isolated from Asian pear (Pyrus pyrifolia) cv. ‘Aoguan’. Sequence analysis suggested that the PyMYB10 gene was an ortholog of MdMYB10 gene, which regulates anthocyanin biosynthesis in red fleshed apple (Malus × domestica) cv. ‘Red Field’. PyMYB10 was identified at the genomic level and had three exons, with its upstream sequence containing core sequences of cis-acting regulatory elements involved in light responsiveness. Fruit bagging showed that light could induce expression of PyMYB10 and anthocyanin biosynthesis. Quantitative real-time PCR revealed that PyMYB10 was predominantly expressed in pear skins, buds, and young leaves, and the level of transcription in buds was higher than in skin and young leaves. In ripening fruits, the transcription of PyMYB10 in the skin was positively correlated with genes in the anthocyanin pathway and with anthocyanin biosynthesis. In addition, the transcription of PyMYB10 and genes of anthocyanin biosynthesis were more abundant in red-skinned pear cultivars compared to blushed cultivars. Transgenic Arabidopsis plants overexpressing PyMYB10 exhibited ectopic pigmentation in immature seeds. The study suggested that PyMYB10 plays a role in regulating anthocyanin biosynthesis and the overexpression of PyMYB10 was sufficient to induce anthocyanin accumulation.

Abstract:The mature fruit skin of ‘Dangshansuli’ pear (Pyrus bretchnederi Rehd.) is yellow-green, but the fruit of the mutant, ‘Xiusu’, is russetted. To understand the mechanism of russet formation, the suppression subtractive hybridization libraries were used to screen for differentially expressed genes between the wild type pear and the mutant. Three hundred and twenty nine and 366 unigenes were obtained from 934 and 929 colonies in the forward and reverse libraries, respectively. The results indicated that phenylpropanoid metabolism, ethylene metabolism and secondary metabolism might be involved in the russet formation process, and fruit skin is likely to be regulated by lignin synthesis, polyamine and H2O2 signalling. Furthermore, the accumulation of lignin and H2O2, together with up-regulated POD enzyme genes expression in the exocarp of ‘Xiusu’ compare with ‘Dangshansuli’ pear control, provided a comprehensive transcriptomics view on the coordination of the russet formation of mutant pear.

Abstract:Background Red coloration of fruit skin is one of the most important traits in peach ( Prunus persica ), and it is mainly due to the accumulation of anthocyanins. Three MYB10 genes, PpMYB10.1 , PpMYB10.2 , and PpMYB10.3 , have been reported as important regulators of red coloration and anthocyanin biosynthesis in peach fruit. In this study, contribution of PpMYB10.1 / 2 / 3 to anthocyanin accumulation in the fruit skin was investigated in the Japanese peach cultivars, white-skinned ‘Mochizuki’ and red-skinned ‘Akatsuki’. We then investigated the relationships between allelic type of PpMYB10.1 and skin color phenotype in 23 Japanese peach cultivars for future establishment of DNA-marker. Results During the fruit development of ‘Mochizuki’ and ‘Akatsuki’, anthocyanin accumulation was observed only in the skin of red ‘Akatsuki’ fruit in the late ripening stages concomitant with high mRNA levels of the last step gene leading to anthocyanin accumulation , UDP-glucose:flavonoid-3-O-glucosyltransferase ( UFGT ). This was also correlated with the expression level of PpMYB10.1 . Unlike PpMYB10.1 , expression levels of PpMYB10.2 / 3 were low in the skin of both ‘Mochizuki’ and ‘Akatsuki’ throughout fruit development. Moreover, only PpMYB10.1 revealed expression levels associated with total anthocyanin accumulation in the leaves and flowers of ‘Mochizuki’ and ‘Akatsuki’. Introduction of PpMYB10.1 into tobacco increased the expression of tobacco UFGT , resulting in higher anthocyanin accumulation and deeper red transgenic tobacco flowers; however, overexpression of PpMYB10.2 / 3 did not alter anthocyanin content and color of transgenic tobacco flowers when compared with wild-type flowers. Dual-luciferase assay showed that the co-infiltration of PpMYB10.1 with PpbHLH3 significantly increased the activity of PpUFGT promoter. We also found close relationships of two PpMYB10.1 allelic types, MYB10.1-1/MYB10.1-2, with the intensity of red skin coloration. Conclusion We showed that PpMYB10.1 is a major regulator of anthocyanin accumulation in red-skinned peach and that it activates PpUFGT transcription. PpMYB10.2 / 3 may be involved in functions other than anthocyanin accumulation in peach. The peach cultivars having two MYB10.1-2 types resulted in the white skin color. By contrast, those with two MYB10.1-1 or MYB10.1-1/MYB10.1-2 types showed respective red or pale red skin color. Proteomic analysis of ‘Zaosu’ pear (Pyrus bretschneideri Rehd.) and its red skin bud mutation

Abstract:Abstract Background Breeding for strong red skin color is an important objective of the pear breeding program. There are few reports of proteome research in green skin pear and its red skin bud mutation. The manuscript at hand is one of the first studies dealing with 2D-PAGE-based analysis of pear fruits and leaves, establishing a suitable sample preparation and testing different 2D-PAGE protocols. Therefore, it may grant a basis for further studies on the pear proteome being the studies main goal. A proteomic analysis was conducted on leaves and fruits of ‘Zaosu’ pear ( Pyrus bretschneideri Rehd.) and its red skin bud mutation in order to reveal their genetic differences in the protein level. Results In the present study, the optimized two-dimensional (2-D) gel electrophoresis system of pear leaf and fruit was set up, and applied to analyze the leaves and fruit protein. The interesting peptide fragments were determined using 4800 Plus MALDI TOF/TOFTM Analyzer mass spectrometer, and the sequence obtained was blasted in NCBInr to identify the differentially-expressed protein. In the 1.5-fold differently-expressed proteins between ‘Zaosu’ pear and its mutant, 10 out of 35 proteins in fruit and 12 out of 24 ones in leaves were identified successfully. Among the 22 identified proteins, 7 protein spots were related to photosynthesis and energy metabolism; 4 were associated with environmental stress; 4 with disease defense; 2 with amino acid metabolism; 2 with cytoskeleton; 1 with antioxidant function; 1 with calcium metabolism; and 1 with unknown function. Moreover, related physiological index, such as chlorophyll content, Rubisco content and polyphone oxidase activity, were different between ‘Zaosu’ pear and its mutant. Conclusion A 2-D gel electrophoresis system of pear leaves and fruits was established, which was suitable for the analysis of proteome comparison. To the best of our knowledge, we have performed the first analysis of the proteomic changes in leaves and fruits of ‘Zaosu’ pear and its red skin bud mutation. Our study provides important information on the use of proteomic methods for studying protein regulation of ‘Zaosu’ pear and its red skin bud mutation.

PbRVE6 Promotes Anthocyanins Accumulation in Pear Fruit Peel by Regulating Key Biosynthetic Genes

Abstract:Red peel color, a desirable trait in pears, is determined by anthocyanin accumulation. While REVEILLE (RVEs) transcription factors regulate anthocyanin biosynthesis in some plant species, their role in pear peel has not been well‐studied. This study investigates the function of RVEs in anthocyanin biosynthesis in 'Zaosu' (low anthocyanin content) and its red bud mutant, 'Red Zaosu' (high anthocyanin content) fruit peel. Consistent with higher anthocyanin content, 'Red Zaosu' pears exhibited increased PbRVE6 expression compared to 'Zaosu', while PbRVE3a and PbRVE3b levels remained unchanged. Additionally, PbRVE6 was localized to the nucleus. Overexpression of PbRVE6 in 'Zaosu' pear pericarp significantly increased anthocyanin content and upregulated key anthocyanin pathway genes PbANS and PbUFGT. Conversely, VIGS silencing of PbRVE6 in 'Red Palacer' pears led to decreased expression of PbANS and PbUFGT and a slight reduction in anthocyanin content. Yeast one‐hybrid and dual‐luciferase assays confirmed that PbRVE6 can bind and activate the promoters of PbANS and PbUFGT. These findings demonstrate that PbRVE6 promotes anthocyanin accumulation in pear peel by directly regulating PbANS and PbUFGT expression. This study provides a valuable foundation for understanding the regulatory network of anthocyanins in pear peels, and offers potential genetic resources for the production of new pear germplasm.

Abstract:The color of red-skinned pear (Pyrus spp.) is primarily attributed to accumulation of anthocyanins, which provide nutritional benefits for human health and are closely associated with the commercial value of fruits. Here, we reported the functional characterization of a R2R3-MYB repressor PyMYB107, which forms an 'activator-repressor' loop to control anthocyanin accumulation in the red-skinned pear. PyMYB107 overexpression inhibited anthocyanin biosynthesis in both pear calli and fruits, while virus-induced gene silencing of PyMYB107 increased anthocyanin accumulation in pear fruits. Furthermore, ectopic expression of PyMYB107 decreased anthocyanin accumulation in tomato, strawberry and tobacco. PyMYB107 can competitively bind to PybHLH3 with PyMYB10/MYB114, thereby suppressing the transcriptional activation of key anthocyanin biosynthesis genes, PyANS and PyUFGT. Site-directed mutagenesis showed that mutations within the R3 domain and EAR motif of PyMYB107 eliminated its repressive activity. Additionally, PyMYB107 exhibited a comparable expression pattern to PyMYB10/MYB114 and was transcriptionally activated by them. Our finding advanced comprehension of the repression mechanism underlying anthocyanin accumulation, providing valuable molecular insights into improving quality of pear fruits.

Abstract:Differences in coloration exist among red pear cultivars. Here, we selected six red pear cultivars with different genetic backgrounds to elucidate the characteristics of fruit pigmentation. We detected anthocyanin contents and the expression levels of anthocyanin synthesis-related genes in these cultivars at different stages of fruit development. The anthocyanin contents of all six cultivars showed a rise–drop tendency. Principal component and hierarchical cluster analyses were used to distinguish the types of cultivars and the genes crucial to each anthocyanin accumulation pattern. The six cultivars were divided into three groups. Red Zaosu were clustered into one group, Red Sichou and Starkrimson into another group, and Palacer, Red Bartlett, and 5 Hao clustered into a third group. The expression levels of F3H, UFGT2, MYB10, and bHLH3 were similar among the differential coloration patterns of the six cultivars, suggesting a critical and coordinated mechanism for anthocyanin synthesis. Anthocyanin transporters (GST) and light-responsive genes, such as COP1, PIF3.1, and PIF3.2 played limited roles in the regulation of anthocyanin accumulation. This study provides novel insights into the regulation of anthocyanins synthesis and accumulation in red pears.

Abstract:Red fruits are popular and widely accepted by consumers because of an enhanced appearance and enriched anthocyanins. The molecular mechanism of anthocyanin regulation in red-skinned pear (Pyrus) has been studied, and the genes encoding the biosynthetic steps and several transcription factors (TFs) have been characterized. In this study, a candidate R2R3 MYB TF, PyMYB114, was identified by linkage to the quantitative trait loci (QTL) for red skin color on linkage group 5 in a population of Chinese pear (Pyrus bretschneideri). The function of PyMYB114 was verified by transient transformation in tobacco (Nicotinana tabacum) leaves and strawberry (Fragaria) and pear fruits, resulting in the biosynthesis of anthocyanin. Suppression of PyMYB114 could inhibit anthocyanin biosynthesis in red-skinned pears. The ERF/AP2 TF PyERF3 was found to interact with PyMYB114 and its partner PybHLH3 to co-regulate anthocyanin biosynthesis, as shown by a dual luciferase reporter system and a yeast two-hybrid assay. In addition, the transcript abundance of PyMYB114 and PyMYB10 were correlated, and co-transformation of these two genes into tobacco and strawberry led to enhanced anthocyanin biosynthesis. This interaction network provides insight into the coloration of fruits and the interaction of different TFs to regulate anthocyanin biosynthesis.
PyWRKY26 and PybHLH3 cotargeted the PyMYB114 promoter to regulate anthocyanin biosynthesis and transport in red-skinned pears

Abstract:Abstract Red pear is favored because of its bright appearance and abundant anthocyanins. Anthocyanin biosynthesis is controlled by transcription factors (TFs) forming regulatory complexes. In red-skinned pears, the WRKY TFs have a significant relationship with anthocyanin biosynthesis, but the molecular mechanism of the WRKY TFs involved in regulating color formation in red-skinned pear is unclear. In this study, the TFs PyWRKY31 and PyWRKY26 were screened as candidate genes for controlling anthocyanin biosynthesis by transcriptome data and bioinformatics analysis. The effect of anthocyanin accumulations after cotransformation of PyWRKY31 or PyWRKY26 with its partners PyMYB10 , PyMYB114 , and PybHLH3 was verified in tobacco leaves and strawberry receptacles by a transient expression system. RT-qPCR analysis and a dual-luciferase reporter system further confirmed that this cotransformation activated the expression of PyDFR , PyANS , and PyUFGT in anthocyanin biosynthesis and PyGST in anthocyanin transport instead of the PyABC transporter and PyAVP . Furthermore, the cotransformed PyWRKY26 and PybHLH3 could bind to the PyMYB114 promoter, and PyWRKY26 directly activated the transcription of PyMYB114 . In addition, the TF PyWRKY26 could interact with PybHLH3, as confirmed by firefly luciferase complementation and yeast two-hybrid (Y2H) assays. These results showed that the interaction of PyWRKY26 and PybHLH3 could cotarget the PyMYB114 promoter, which resulted in anthocyanin accumulation in red-skinned pear. This study further strengthened the understanding of the regulatory mechanism of anthocyanin accumulation and contributed to improving the appearance of red-skinned pears.
