Abstract:Over the last decade, various magnetic nanomaterials have been developed as magnetic resonance imaging (MRI) contrast agents; the greatest challenges encountered for clinical application have been insufficient stability. In this paper, a lyophilization method for 2, 3-dimercaptosuccinic acid-modified iron oxide (γ-Fe2 O3 @DMSA) nanoparticles was developed to simultaneously overcome two disadvantages; these include insufficient stability and low-magnetic response. After lyophilization, the clusters of γ-Fe2 O3 @DMSA with the size of 156.7 ± 15.3 nm were formed, and the stability of the lyophilized powder (γ-Fe2 O3 @DMSA-LP) increased up to over 3 years. It was also found that rehydrated γ-Fe2 O3 @DMSA-LP could be ingested by RAW264.7 cells in very large quantities. Results of pharmacokinetics and biodistribution studies in vivo indicated that γ-Fe2 O3 @DMSA-LP is a promising liver-targeted material. Furthermore, it also exhibited higher MRI efficiency and longer imaging time in the liver than the well-known product Feridex(®) . Moreover, results of vascular irritation and long-term toxicity experiments demonstrated γ-Fe2 O3 @DMSA-LP could be a nontoxic, biocompatible contrast agent in vivo. Therefore, the proposed γ-Fe2 O3 @DMSA-LP can be used as a potential MRI contrast agent in clinic for hepatic diseases.

Abstract:The development of new types of high-performance nanoparticulate MR contrast agents with either positive (T-1) or dual-contrast (both positive and negative, T-1 + T-2) ability is of great importance. Here we report a facile synthesis of ultrasmall PEGylated iron oxide nanoparticles for dual-contrast T-1- and T-2-weighted MRI. The produced superparamagnetic iron oxide nanoparticles (SPIONs) are of high crystallinity and size uniformity with an average diameter of 5.4 nm, and can be individually dispersed in the physiological buffer with high stability. The SPIONs reveal an impressive saturation magnetization of 94 emu g(-1) Fe3O4, the highest r(1) of 19.7 mM(-1) s(-1) and the lowest r(2)/r(1) ratio of 2.0 at 1.5 T reported so far for PEGylated iron oxide nanoparticles. T-1-and T-2-weighted MR images showed that the SPIONs could not only improve surrounding water proton signals in the T-1-weighted image, but induce significant signal reduction in the T-2-weighted image. The good contrast effect of the SPIONs as T-1 + T-2 dual-contrast agents might be due to its high magnetization, optimal nanoparticle size for T-1 + T-2 dual-contrast agents, high size monodispersity and excellent colloidal stability. In vitro cell experiments showed that the SPIONs have little effect on HeLa cell viability.

Abstract:Ultra-small-sized iron oxide nanoparticles with good biocompatibility are regarded as promising alternatives for the gadolinium-based contrast agents, which are widely used as a positive contrast agent in magnetic resonance imaging (MRI). However, the current preparation of the iron oxide magnetic nanoparticles with small sizes usually involves organic solvents, increasing the complexity of hydrophilic ligand replacement and reducing the synthesis efficiency. It remains a great challenge to explore new iron oxide nanoparticles with good biocompatibility and a high T1 contrast effect. Here, we reported a cage-like protein architecture self-assembled by approximately 6–7 BSA (bovine serum albumin) subunits. The BSA nanocage was then used as a biotemplate to synthesize uniformed and monodispersed Fe2O3@BSA nanoparticles with ultra-small sizes (∼3.5 nm). The Fe2O3@BSA nanoparticle showed a high r1 value of 6.8 mM–1 s–1 and a low r2/r1 ratio of 10.6 at a 3 T magnetic field. Compared to Gd-DTPA, the brighter signal and prolonged angiographic effect of Fe2O3@BSA nanoparticles could greatly benefit steady-state and high-resolution imaging. Abstract:Magnetic resonance imaging (MRI) has gained wide interest in early accurate diagnoses due to the high resolution and low toxicity of magnetic nanoparticles. In order to develop potential alternatives of toxic Gd- or Mn-based chelating agents, we report the synthesis of water soluble ultra-small Fe3O4 nanoparticles by a modified co-precipitation method as T1-weighted positive contrast agents. The magnetic iron oxide nanoparticles (MIONs) were functionalized by polymer ligand dodecanthiol-polymethacrylic acid (DDT-PMAA) to enhance their colloidal stability. These MIONs have high longitudinal relaxivity (r1 = 8.18 mM−1·S−1) and exhibited good results in the in vitro and in vivo MR imaging. No toxicity was observed in cytotoxicity assay and histology toxicity analysis. The MIONs@DDT-PMAA(magnetic iron oxide nanoparticles @ dodecanthiol-polymethacrylic acid) present great potential as positive contrast agents for tumor diagnosis.
Moderate Cooling Coprecipitation for Extremely Small Iron Oxide As a Ph Dependent T1-MRI Contrast Agent.

Abstract:Iron based nanomedicine (IBNM) has been one powerful diagnostic tool as a magnetic resonance imaging (MRI) contrast agent (CA) in the clinic for years. Conventional IBNMs are generally employed as T2-MRI CAs, but most of them are constrained in clinical indication expansion by magnetic susceptibility artifacts. In comparison, extremely small iron oxide (ESIO) with a core size less than 5 nm has demonstrated the T1-MRI effect, which provides prospects for a Gd-based agent alternative. Nevertheless, currently developed ESIOs for T1-MRI CAs always require harsh conditions such as a high temperature and high boiling point reagent. Moreover, very few of the currently developed ESIOs meet the stringent pharmaceutical standard. Herein, on the basis of a crystal nuclear precipitation-dissolution equilibrium mechanism and outer/inner sphere T1-MRI theory, monodisperse ESIOs with an average size of 3.43 nm (polydispersity index of 0.104) are fabricated using a moderate cooling procedure with mild coprecipitation reaction conditions. The as-synthesized ESIOs display around 3-fold higher T1 MRI signal intensity than that of commercial Ferumoxytol (FMT), comparable to that of Gd-based CAs in vitro. Additionally, the T1-MRI performance of the ESIOs is pH dependent and delivers bright signal augmentation. Eventually, the internalization into mesenchymal stem cells of the ESIO is realized in the absence of a transferring agent. Considering the identical structure and composition of the ESIOs as compared to that of FMT, they could meet the pharmaceutical criteria, thus providing great potential as T1-MRI Cas, for instance as stem cell tracers.

Abstract:alpha-Amino acid N-thiocarboxyanhydrides (NTAs) are promising cyclic monomers to synthesize polypeptides and polypeptoids via controlled ring-opening polymerizations. Superior to N-carboxyanhydrides requiring protection on hydroxyl groups, NTAs are able to tolerate such nucleophiles. In this work, we report the synthesis of NTA monomers containing unprotected phenolic hydroxyl groups of 3,4-dihydroxy-L-phenylalanine (DOPA) and L-tyrosine (Tyr). Their controlled ROPs and sequential copolymerizations with polysarcosine (PSar) yield PDOPA, PTyr, and PDOPA-b-polysarcosine (PDOPA-b-PSar) products quantitatively with designable degrees of polymerization. Micellar nanoparticles of Fe3+@PDOPA-b-PSar have been prepared thanks to the strong chelation of iron(III) cation by catechol ligands that act as T1-weighted magnetic resonance imaging (MRI) contrast agents. For instance, Fe3+@PDOPA(10)b-PSar(50) exhibits higher longitudinal relaxivity (r(1) = 5.6 mM(-1) s(-1)) than commercial Gd3+-based compounds. Effective MRI contrast enhancement in vivo of nude mice with a moderate duration (150 min) and 3D magnetic resonance angiography in rabbit illustrated by using volume rendering and maximal intensity projection techniques ignite the clinical application of Fe3+-based polypept(o)ides in diagnostic radiology as Gd-free MRI contrast agents.

Abstract:Although iron oxide (Fe3O4) nanoparticles have broad application prospects as magnetic resonance imaging (MRI) contrast agent, their biocompatibility and biotoxicity still need to be improved. In this study, we prepared Fe3O4@Angelica sinensis polysaccharide nanoparticles (Fe3O4@ASP NPs) with a 9 nm Fe3O4 core and ASP as the coating material. The Fe3O4@ASP NPs are superparamagnetic, can be taken up by liver and spleen macrophages in the circulatory system in vivo, and are a good-biocompatibility and low-toxicity transverse relaxation time (T2) and T2-star (T2*) magnetic resonance imaging (MRI) contrast agent for the liver. The cytotoxicity assessment using HeLa cells and the pathological tests in mice validate that Fe3O4@ASP NPs have low toxicity and good biocompatibility in vivo, which can be attributed to the ASP as a natural polysaccharide with good biocompatibility and its function of protecting the liver. Fe3O4@ASP NPs are a potential new MRI contrast agent with high signal intensity in vivo.<h3 class="a-plus-plus">Graphic abstract</h3>

Abstract:Spherical superparamagnetic iron oxide nanoparticles have been developed as T 2 -negative contrast agents for magnetic resonance imaging in clinical use because of their biocompatibility and ease of synthesis; however, they exhibit relatively low transverse relaxivity. Here we report a new strategy to achieve high transverse relaxivity by controlling the morphology of iron oxide nanoparticles. We successfully fabricate size-controllable octapod iron oxide nanoparticles by introducing chloride anions. The octapod iron oxide nanoparticles (edge length of 30 nm) exhibit an ultrahigh transverse relaxivity value (679.3±30 mM −1 s −1 ), indicating that these octapod iron oxide nanoparticles are much more effective T 2 contrast agents for in vivo imaging and small tumour detection in comparison with conventional iron oxide nanoparticles, which holds great promise for highly sensitive, early stage and accurate detection of cancer in the clinic.

Abstract:Early diagnosis of disease and follow-up of therapy is of vital importance for appropriate patient management since it allows rapid treatment, thereby reducing mortality and improving health and quality of life with lower expenditure for health care systems. New approaches include nanomedicine-based diagnosis combined with therapy. Nanoparticles (NPs), as contrast agents for in vivo diagnosis, have the advantage of combining several imaging agents that are visible using different modalities, thereby achieving high spatial resolution, high sensitivity, high specificity, morphological, and functional information. In this work, we present the development of aluminum hydroxide nanostructures embedded with polyacrylic acid (PAA) coated iron oxide superparamagnetic nanoparticles, Fe3O4@Al(OH)3, synthesized by a two-step co-precipitation and forced hydrolysis method, their physicochemical characterization and first biomedical studies as dual magnetic resonance imaging (MRI)/positron emission tomography (PET) contrast agents for cell imaging. The so-prepared NPs are size-controlled, with diameters below 250 nm, completely and homogeneously coated with an Al(OH)3 phase over the magnetite cores, superparamagnetic with high saturation magnetization value (Ms = 63 emu/g-Fe3O4), and porous at the surface with a chemical affinity for fluoride ion adsorption. The suitability as MRI and PET contrast agents was tested showing high transversal relaxivity (r2) (83.6 mM−1 s−1) and rapid uptake of 18F-labeled fluoride ions as a PET tracer. The loading stability with 18F-fluoride was tested in longitudinal experiments using water, buffer, and cell culture media. Even though the stability of the 18F-label varied, it remained stable under all conditions. A first in vivo experiment indicates the suitability of Fe3O4@Al(OH)3 nanoparticles as a dual contrast agent for sensitive short-term (PET) and high-resolution long-term imaging (MRI).

Abstract:Synthesis of magnetic particles with superparamagnetism, high magnetization, excellent biocompatibility and small particle size is highly desirable for biomedical applications. Herein, we report that sub-100 nm biocompatible Fe3O4 nanocrystal clusters can be prepared via a solvothermal method by using water as a size-control agent and sodium citrate as a stabilizer. The resultant Fe3O4 particles have uniform spherical shape, superparamagnetic properties, high magnetization values, excellent colloidal stability and good biocompatibility. Notably, the particle size can be tuned over a range of 70-180 nm by changing the content of H2O in the system. Moreover, the T-2-weighted magnetic resonance imaging (MRI) experiments show that 85 nm Fe3O4 particles have an ultra-high r(2) relaxivity of 175.4 mM(-1) s(-1), suggesting a great potential as MRI probes. In addition, the labeling efficiency of mesenchymal stem cells with as-obtained Fe3O4 particles is much higher than that of commercial superparamagnetic iron oxide nanoparticles, which further indicates their promising in biomedicine.

Abstract:Conjunctival synthetic fiber ("teddy bear") granulomas result from a granulomatous response to synthetic fabric fibers introduced into the conjunctival sac. There is a striking propensity for these lesions to develop in the pediatric age group, to be unilateral, and to be situated in the inferior conjunctival fornix. It is believed that proximity of the eye to comforting blankets, fabric toys, or forcibly applied pullover sweaters may provide the opportunity for ocular inoculation. Described below is the case of a 6-year-old girl who had a conjunctival synthetic fiber granuloma that exhibited the characteristic clinical and pathologic features of these lesions.

Abstract:In this work, ultra-small iron oxide nanoparticles were synthesized using high temperature pyrolysis. For further functionalization, dopamine was anchored onto the surface of ultra-small iron oxide nanoparticles (Fe3O4@DOPA NPs). Gd(III) ions were chelated with diethylenetriaminepentaacetic acid (DTPA) which was linked with dopamine. The resulting Gd(III) complex conjugated ultra-small Fe3O4 NPs [Fe3O4@DOPA(Gd-DTPA) NPs] were characterized by XRD pattern and TEM. The longitudinal relaxivity (r(1)) of Fe3O4@DOPA(Gd-DTPA) NPs was 9.97 mM(-1) S-1. Compared to ultra-small Fe3O4 NPs and Gd(III) complexes, Fe3O4@DOPA(Gd-DTPA) NPs showed a significant improvement of the T-1-weighted MR image in aqueous solution, in vitro and in vivo.

Abstract:AIM:Investigated the self-assembly and characterization of novel antifouling polyethylene glycol (PEG)-coated iron oxide nanoparticles as nanoprobes for magnetic resonance imaging (MRI) contrast agent. METHOD:Monodisperse oleic acid-coated superparamagnetic iron oxide cores are synthesized by thermal decomposition of iron oleate. The self-assembly behavior between iron oxide cores and PEG-lipid conjugates in water and their characteristics are confirmed by transmission electron microscope, X-ray diffraction, thermogravimetric analysis, Fourier transform infrared spectroscopy, and vibrating sample magnetometer. RESULT:Dynamic light scattering shows superparamagnetic iron oxide nanoparticles coated with PEG are stable in water for pH of 3-10 and ionic strengths up to 0.3 M NaCl, and are protein resistant in physiological conditions. Additionally, in vitro MRI study demonstrates the efficient magnetic resonance imaging contrast characteristics of the iron oxide nanoparticles. CONCLUSION:The result indicates that the novel antifouling PEG-coated superparamagnetic iron oxide nanoparticles could potentially be used in a wide range of applications such as biotechnology, MRI, and magnetic fluid hyperthermia.

Abstract:Extremely small-sized iron oxide nanoparticles (ESIONPs) with sizes less than 5 nm have shown great promise as T1 contrast agents for magnetic resonance imaging (MRI). However, their facile and scalable production with simultaneously endowed biocompatible surface chemistry remains difficult to be realized. In this study, by using the coprecipitation method implemented in a specially designed gas/liquid mixed phase fluidic reactor, polyglucose sorbitol carboxymethyether (PSC) coated ESIONPs were continuously synthesized with controllable particle sizes ranging from 1.8 to 4 nm. Among the differently sized ESIONPs, the 3.7-nm ESIONPs exhibit the best performance as T1 MRI contrast agent, featuring a high r1 value of 4.11 (mmol L−1)−1 s−1 and low r2/r1 ratio of 7.90 under a clinical 3 T MR scanning, as well as the excellent T1 MRI contrast effect in not only water but also the cellular environment and blood vessel. Furthermore, the ESIONPs possess long-term stability and good dispersity in aqueous dispersions, making them ideal candidates as safe and effective T1-weighted MRI contrast agent for real clinical use.

Abstract:Traditional T2 magnetic resonance imaging (MRI) contrast agents have defects inherent to negative contrast agents, while chemical exchange saturation transfer (CEST) contrast agents can quantify substances at trace concentrations. After reaching a certain concentration, iron-based contrast agents can "shut down" CEST signals. The application range of T2 contrast agents can be widened through a combination of CEST and T2 contrast agents, which has promising application prospects. The purpose of this study is to develop a T2 MRI negative contrast agent with a controllable size and to explore the feasibility of dual contrast enhancement by combining T2 with CEST contrast agents. The study was carried out in vitro with HCT-116 human colon cancer cells. A GE SIGNA Pioneer 3.0 T medical MRI scanner was used to acquire CEST images with different saturation radio-frequency powers (1.25/2.5/3.75/5 μT) by 2D spin echo-echo planar imaging (SE-EPI). Magnetic resonance image compilation (MAGiC) was acquired by a multidynamic multiecho 2D fast spin-echo sequence. The feasibility of this dual-contrast enhancement method was assessed by scanning electron microscopy, transmission electron microscopy, Fourier transform infrared spectroscopy, dynamic light scattering, ζ potential analysis, inductively coupled plasma, X-ray photoelectron spectroscopy, X-ray powder diffraction, vibrating-sample magnetometry, MRI, and a Cell Counting Kit-8 assay. The association between the transverse relaxation rate r2 and the pH of the iron-based contrast agents was analyzed by linear fitting, and the linear relationship between the CEST effect in different B1 fields and pH was analyzed by the ratio method. Fe3O4 nanoparticles (NPs) with a mean particle size of 82.6 ± 22.4 nm were prepared by a classical process, and their surface was successfully modified with -OH active functional groups. They exhibited self-aggregation in an acidic environment. The CEST effect was enhanced as the B1 field increased, and an in vitro pH map was successfully plotted using the ratio method. Fe3O4 NPs could stably serve as reference agents at different pH values. At a concentration of 30 μg/mL, Fe3O4 NPs "shut down" the CEST signals, but when the concentration of Fe3O4 NPs was less than 10 μg/mL, the two contrast agents coexisted. The prepared Fe3O4 NPs had almost no toxicity, and when their concentration rose to 200 μg/mL at pH 6.5 or 7.4, they did not reach the half-maximum inhibitory concentration (IC50). Fe3O4 magnetic NPs with a controllable size and no toxicity were successfully synthesized. By combining Fe3O4 NPs with a CEST contrast agent, the two contrast agents could be imaged simultaneously; at higher concentrations, the iron-based contrast agent "shut down" the CEST signal. An in vitro pH map was successfully plotted by the ratio method. CEST signal inhibition can be used to realize the pH mapping of solid tumors and the identification of tumor active components, thus providing a new imaging method for tumor efficacy evaluation.

Abstract:Extremely small iron oxide nanoparticles (ESIONPs), which have a switchable contrast signal and a favorable biocompatibility, are gaining considerable attention as smart magnetic resonance imaging (MRI) contrast agents (CAs). Adaptive MRI CAs could improve the accuracy of tumor diagnosis because they realize rapid imaging signal conversion under an endogenous stimulus. Herein, based on the hydrophilic to hydrophobic transition of polymers, we propose a pH-responsive T1/T2 switchable ESIONP contrast agent. ESIONPs are function-alized at the surface with adamantane groups through a polyethylene glycol (PEG) chain, and subsequently, a stimuli-responsive polymer (PSDM) with pH-dependent solubility is introduced through the host-guest interaction between adaman-tane and g-cyclodextrin (g-CD). The obtained ESIONPs-PEG-PSDM is equipped with remarkable pH responsiveness, which triggers the aggregation of ESIONPs under the tumor acid condition, leading to the switch from a T1 to a T2 contrast agent. In vivo MRI experiments show that the T2 contrast signal is selectively enhanced at the tumor location after injection of ESIONPs-PEG-PSDM. Therefore, ESIONPs-PEG-PSDM with pH-induced hydrophilic to hydrophobic transition is a potential smart MRI contrast agent for precise tumor diagnosis.

Abstract:Oleic acid stabilized superparamagnetic iron oxide nanoparticles (SPION) were selected as the cores for fabrication of sub-50-nm monodisperse single-loaded SPION@SiO2 core-shell nanostructures. Parameters that influence the formation of SPION@SiO2 in the water-in-oil reverse microemulsion system have been systematically investigated. The sufficiently high concentration of well-dispersed SPION, together with an appropriately low injection rate of tetraethoxysilane, were found to be the keys to efficiently prevent the homogeneous nucleation of silica and obtain a high-quality single-loaded core-shell nanocomposite. A more detailed mechanism for incorporating oleic acid capped inorganic functional nanoparticles into silica is proposed on the basis of previous reports and our new experimental results. Finally, the as-synthesized SPION@SiO2 nanospheres are exploited as an MRI-enhanced contrast agent, and their contrast effect in solution is tested by using a clinical MRI instrument.

Abstract:Objective: To synthesize stable and water-soluble PMAT-Fe3O4 nanoparticles(NPs) as MRI contrast agent and characterize it.Methods: Poly maleic anhydride alt-1-tetradecene(PMAT) was utilized to modify the surface of oil-soluble NPs,and the obtained PMAT-Fe3O4 NPs were characterized by TEM,DLS,FT-IR,VSM and MRI.Results:(1) TEM and DLS studies showed that the PMAT-Fe3O4 NPs have a magnetic core size of about 10 nm and a hydrodynamic diameter of about 80 nm.(2) PMAT-Fe3O4 could keep stable in water and familiar buffers,such as MES,PBS and Tris without aggregation.(3) VSM measurements showed that the saturation magnetization(Ms) was about 14.0 emu·g-1,the relaxivity value(r2) of PMAT-Fe3O4 was 367.79 mM-1s-1.Conclusion: The obtained PMAT-Fe3O4 NPs possess outstanding water-solubility,good magnetic properties,and high r2 value,which are therefore expected to become an excellent MRI contrast agent.

Abstract:Superparamagnetic iron oxide nanoparticles (SPIONs) have been the subject of extensive research due to their potential biomedical applications. In the present investigation, superparamagnetic FA-PEI-Fe3O4 were successfully prepared and evaluated as a targeted MRI contrast agent. FTIR characteristics, TGA, VSM, and MR imaging confirmed the composition and magnetic properties of the synthesized nanoparticles. TEM showed that FA-PEI-Fe3O4 were spherical in shape and well dispersed. The nanoparticles were superparamagnetic at room temperature with a saturation magnetization value of 67.1 emu/g. The nanoparticles showed higher uptake efficiency due to receptor-mediated endocytosis. Moreover, specificity of FA-PEI-Fe3O4 to target tumor cells was demonstrated by the increased nanoparticle uptake and significant contrast enhancement of KB cells over MCF7 cells. The competitive inhibition of FA-PEI-Fe3O4 by free FA further confirmed the specific interaction of this conjugate with FA receptors. In vivo MR imaging studies showed a decreased signal intensity and enhanced tumor contrast post-injection of FA-PEI-Fe3O4. These results indicate that FA-PEI-Fe3O4 can be used as a promising tumor-targeting agent as well as a T-2 negative-contrast agent in MR imaging applications.
