Abstract:Photodynamic therapy (PDT) is a promising therapeutic strategy for the clinical treatment of laryngeal cancer, and nanocarriers could be easily combined PDT with conventional chemotherapy for enhanced therapeutic outcomes. However, the encapsulation of photosensitizers (PS) and chemotherapy drugs into conventional nanocarriers achieves only a simple combination without synergistic effects. Herein, we explored an oxidation-sensitive polyphosphoester (PPE)-based nanocarrier to co-encapsulate chlorin e6 (Ce6) and docetaxel (Dtxl) for synergistic laryngeal cancer therapy. During the PDT process, the produced ROS not only induced cell apoptosis but also triggered the release of Dtxl through the hydrophobic to hydrophilic transition of the PPE core, resulting in a cascade PDT/chemotherapy to elicit a synergistic anticancer effect for the complete suppression of laryngeal cancer growth. Moreover, this oxidation-sensitive nanocarrier-mediated cascade PDT/chemotherapy can also induce an effective antitumor immune response via the immunogenic cell death effect (ICD) and potentiate the effectiveness of immune checkpoint blockade (ICB) antibodies to inhibit distant tumor growth.

Abstract:AbstractNanomedicines with photodynamic therapy and reactive oxygen species (ROS)‐triggered drug release capabilities are promising for cancer therapy. However, most of the nanomedicines based on ROS‐responsive nanocarriers still suffer from serious ROS consumption during the triggered drug release process. Herein, a photodynamic‐chemodynamic cascade strategy for the design of drug delivery nanosystem is proposed. A doxorubicin hydrochloride‐loaded ROS‐responsive polymersome (DOX‐RPS) is prepared via the self‐assembly of amphiphilic poly(ethylene glycol)‐poly(linoleic acid) and poly(ethylene glycol)‐(2‐(1‐hexyloxyethyl)‐2‐devinyl pyropheophorbide‐α)‐iron chelate (PEG‐HPPH‐Fe). The RPS can effectively deliver a drug to tumor site through passive targeting effect. Upon laser irradiation, the photosensitizer HPPH can efficiently generate ROS, which further causes in situ oxidation of linoleic acid chain and subsequent RPS structural destruction, permitting triggered drug release. Intriguingly, catalyzed by HPPH‐Fe, ROS will be regenerated from linoleic acid peroxide through a chemodynamic process. Therefore, ROS‐triggered drug release can be achieved without ROS over‐consumption. The in vitro and in vivo results confirmed ROS generation, triggered drug release behavior, and potent antitumor effect of the DOX‐RPS. This photodynamic‐chemodynamic cascade strategy provides a promising approach for enhanced combination therapy.

Abstract:Thalictrum is a well recognised genus of medicinal plants. It is widely distributed in temperate and tropical habitats of India. Alkaloid isomers from Thalictrum are reported to exhibit various pharmacological activities like antiamebic, antimicrobial, antitumour and antiviral. Thalictrum reniforme Wallich and Thalictrum neurocarpum Royale are very less studied species of the genera. In present study, we have discussed about simultaneous quantification of seven bioactive alkaloids in aerial parts of T. reniforme and T. neurocarpum using ultra performance liquid chromatography attached with hybrid triple quadrupole linear ion trap mass spectrometer (UPLC-QqQLIT-MS) operated in the multiple reactions monitoring (MRM) acquisition. The chromatographic separation of all the standards were carried out by using ACQUITY UPLC CSHTM C18 column using a gradient programming at a flow rate of 0.3 mL/min. Method was validated by determining the precision and recovery values of the samples in terms of relative standard deviations (RSDs) which was found 0.40-3.1% and 0.59-3.17% (98-101%), respectively. Calibration curves for all the seven analytes was determined as optimum linear detector response having R2 0.9988 over the concentration range of 0.5-1000 ng/mL. The validated method was then used to quantify the isoquinoline alkaloids which revealed that T. reniforme had high level of total content as compared to T. neurocarpum. The quantity of analyte was subjected to principal component analysis.

Abstract:Photodynamic therapy (PDT) induces aggravation of tumor hypoxia, which leads to worsening tumor invasiveness and metastasis. Combining hypoxia-responsive prodrugs with photosensitizers could transform "enemy" (tumor hypoxia) into "friend" (positive amplifier) to facilitate efficient activation of prodrugs. However, it remains challenging to develop a highly effective co-delivery nanoplatform due to the poor compatibility between vehicles and cargoes. Herein, we selected podophyllotoxin (PPT) as a model drug to synthesize hypoxia-responsive polyphenol prodrug (PAG). By introducing a chemical modification strategy, PAG and Chlorin e6 (Ce6) could be remotely co-encapsulated into liposomes (PAG/Ce6 LPs) at a synergistic ratio utilizing metal ions gradient. In vitro experiments demonstrated that PAG/Ce6 LPs could kill tumor cells through multiple pathways; i) generating reactive oxygen species (ROS) for direct cell killing; ii) consuming intracellular glutathione (GSH) to disturb intracellular redox homeostasis to induce tumor cell apoptosis. iii) aggravating hypoxia micro-environment to promote PPT release for chemotherapy. Furthermore, the excellent stability of PAG/Ce6 LPs guaranteed extended drug blood circulation time, enhanced tumor accumulation and improved anticancer efficacy. Additionally, PAG/Ce6 LPs kept fluorescence quench in circulation and specifically recovered at the tumor site, avoiding the obstacle of phototoxicity. Such a co-delivery nanoplatform, integrating hypoxia-responsive polyphenol prodrugs and photosensitizer into one nanocarrier, provides new insight into solving the challenges of chemo-photodynamic therapy.

Abstract:Tumor hypoxia, which is indispensable to tumor propagation and therapy resistance, has been one of the most important factors influencing clinical outcomes. To modulate the hypoxia microenvironment, we herein developed reactive oxygen species (ROS)-sensitive arylboronic ester-based biomimetic nanocarriers co-encapsulated with a photosensitizer chlorin e6 (Ce6) and a hypoxia-activated prodrug tirapazamine (TPZp) for tumor-specific release and synergistic photodynamic chemotherapy. In order to bypass macrophage uptake and improve tumor penetration, the nanocarriers were further modified with the red blood cell membrane and iRGD peptide (denoted as NPs@i-RBMCe6+TPZp). After administration, NPs@i-RBMCe6+TPZp exhibited prolonged blood circulation, selective tumor accumulation and excellent penetration into the tumor interior. Upon light irradiation, ROS were generated by Ce6 for photodynamic therapy (PDT), which subsequently caused dissociation of the ROS-responsive nanocarriers. An enhanced therapeutic effect was further achieved through the activation of TPZp in the aggravated local hypoxia microenvironment. The synergistic cancer therapy based on NPs@i-RBMCe6+TPZp significantly suppressed tumor growth with negligible side effects. The biomimetic nanocarriers have great potential to overcome hypoxia-limited PDT, and significantly improve the anticancer efficacy by synergistic tumor-targeted PDT and hypoxia-activated chemotherapy.

Abstract:Currently, single chemotherapeutics or photosensitizers implemented in clinic present huge challenges in terms of unfavorable therapeutic efficacy, physiological instability and non-selective toxicity. In order to remedy the faultiness of insufficient efficiency of monotherapy, we herein synthesized an "all-in-one" targeting drug delivery system with multimodal therapy based on chemo-photodynamic synergistic therapy. Specifically, mesoporous silica nanorods were first synthesized through doping chlorin e6 (Ce6) in the sol–gel process, and then were used to load chemotherapy drug. After the amino groups and boronic acid groups were sequentially modified on the surface of nanoparticles, lactobionic acid (LA) was chosen as an intermediary to achieve simultaneous crosslinking of targeting agent LA and sealing agent sericin through pH responsive boronic ester linkage. The obtained ultimate nanocomposites possessed good biocompatibility, colloidal stability and hemocompatibility. Besides, in vitro endocytosis experiment performed with confocal laser scanning microscopy and flow cytometry testified the LA-mediated accurate tumor-specific targeting ability of synthesized nanocarriers. Moreover, a series of in vitro assays demonstrated that fabricated therapeutic nanoplatform could not only release loaded drugs in response to acidic environment to achieve chemotherapy, but also could generate singlet oxygen from photosensitizer Ce6 under external light stimulation to realize photodynamic therapy. Most interestingly, the combination therapy including photodynamic therapy and chemotherapy exhibited a synergistic effect against tumor cells in both in vivo and ex vivo experiments. Briefly, the drug delivery system reported here could attain effective chemo-photodynamic synergistic therapy to dispose of the insufficient lethality of monotherapy while guaranteeing the safety during in vivo application.

Abstract:Cyclodextrin (CD) has special spatial structure and well biological safety, so it has been widely used for constructing CD-based nanoplatforms. Through functionalization, cyclodextrin can form various stimulus-response nanoplatforms, such as pH, temperature, redox, light and magnetic fields. In this study, we designed a highly sensitive reactive oxygen species (ROS)-responsive polymer PCP which encapsulated doxorubicin (DOX) and purpurin 18 (P18) to achieve the synergy of photodynamic and chemotherapy. The high content of reactive oxygen species (ROS) in the tumor microenvironment (TME) triggers the cleavage of the borate bond of MPEG-CD-PHB (PCP), thereby promoting the release of drugs. When irradiated with near-infrared laser, the photosensitizer P18 released by polymer micelles can produce reactive oxygen species to promote cell apoptosis. Compared with monotherapy, a series of experiments confirmed that our micelles had enhanced anti-cancer activity. This work was beneficial to the design of ROS-responsive materials and provides an effective strategy for the application of collaborative anti-tumor therapy.

Abstract:Cancer has been one of the major healthcare burdens, which demands innovative therapeutic strategies to improve the treatment outcomes. Combination therapy hold great potential to leverage multiple synergistic pathways to improve cancer treatment. Cancer cells often exhibit an increased generation of reactive oxygen species (ROS) and antioxidant species compared with normal cells, and the levels of these species can be further elevated by common therapeutic modalities such as photodynamic therapy (PDT) or chemotherapy. Taking advantage that cancer cells are vulnerable to further oxidative stress, we aim to design a drug delivery system by simultaneously increasing the cellular ROS level, reducing antioxidative capacity, and inducing anticancer chemotherapy in cancer cells. Here, we designed a star-shape polymer, PEG(-b-PCL-Ce6)-b-PBEMA, based on the Passerini three-component reaction, which can both enhance ROS generation during PDT and decrease the GSH level in cancer cells. The polycaprolactone conjugated with photosensitizer Ce6 served as hydrophobic segments to promote micelle formation, and Ce6 was used for PDT. The H2O2-labile group of arylboronic esters pendent on the third segment was designed for H2O2-induced quinone methide (QM) release for GSH depletion. We thoroughly investigated the spectral properties of blank micelle during its assembling process, ROS generation, and H2O2-induced QM release in vitro. Moreover, this polymeric micelle could successfully load hydrophobic anti-cancer drug Doxorubicin (DOX) and efficiently deliver DOX into cancer cells. The triple combination of ROS generation, GSH elimination, and chemotherapy dramatically improved antitumor efficiency relative to each of them alone in vitro and in vivo.

Abstract:Amalgamation of near-infrared laser phototherapies with chemotherapy in multi-modal synergistic therapy holds great promise for future precision cancer nanomedicine due to its minimal invasiveness, reduced adverse reactions, and high anticancer efficacy. Herein, CuO nanoparticles were functionalized with photosensitizer molecule, chlorin e6 (Ce6) and coated with polydopamine (PDA) to achieve a drug delivery system (CuO@Ce6-PDA) with photothermal/photodynamic therapy (PTT/PDT). Subsequently, chemical drug PTX was loaded for chemotherapy, and folic acid (FA) serving as cancer-targeting exterior material. Prepared FA@CuO@Ce6-PDA/PTX nanoparticles were nano-sized with favorable biocompatibility, colloidal stability, optimal surface charge, effective PTX loading, and controllable PTX release. In vitro studies on 4T1 cells showed that FA@CuO@Ce6-PDA/PTX had noteworthy synergistic therapeutic antitumour effects featuring chemo/PTT/PDT with IC 50 of 50 μg/mL lower than that FA@CuO@Ce6-PDA/PTX without NIR laser irradiation (225 μg/mL). Additionally, FA@CuO@Ce6-PDA/PTX produced intracellular high reactive oxygen species (ROS) in presence of 660 nm laser, altering mitochondrial membrane potential and promoting tumour cell death. In vivo results indicate nanoplatform could accumulate in tumour spots enabling thermal imaging capabilities and exhibit synergistic therapeutic effect if irradiated with NIR laser (808 and 660 nm), evident from in vitro antitumour assay. Therefore, in vitro finding postulates FA@CuO@Ce6-PDA/PTX could be an intriguing nanoplatform for Chemo/PTT/PDT-based combination therapy.

Abstract:Although combined chemotherapy (Chemo), photothermal (PTT) and photodynamic (PDT) in cancer therapy has drawn significant attention due to its superior anticancer ability, the required high intensity of irradiation results in serious photo-toxicity to healthy neighboring cells, and thus limits its biomedical applications. Herein, we developed an ultralow-intensity near infrared (NIR) light synchronously activated collaborative Chemo/PTT/PDT nanoplatform. The nanoplatform is composed of a highly emissive upconversion (UC) core, chlorin e6 (Ce6) photosensitizer and the anticancer drug doxorubicin hydrochloride (DOX) co-loaded in a mesoporous silica (MS) shell, and polyethylene glycol-modified graphene (PGO) acts as both the photothermal reagent and smart switch for promoted drug release. Upon 808 nm NIR light exposure with ultralow intensity (0.25 W cm-2), which is below the maximum permissible exposure (MPE, 0.33 W cm-2) for skin, the mild hyperpyrexia of PGO induced both cancer cell irreversible death for PTT and greatly promoted drug release for enhanced Chemo. On the other hand, the upconverted 660 nm light from UC activated Ce6 to generate reactive oxygen species for PDT, while the upconverted 540 nm light from UC could be employed for visualizing the treatment process. The in vitro and in vivo anticancer experiments demonstrate that the ultralow-intensity NIR light synchronously activated Chemo/PTT/PDT nanoplatform exhibits remarkable therapeutic efficacy with minimal photodamage.

Abstract:Comprehensive SummaryNanoscale drug delivery systems (NDDSs) have emerged as promising carriers for combinational therapy by co‐delivery of multiple drugs and modalities. However, most co‐delivery systems require the use of complicated materials and formulations. Herein, we report the single use of a polymeric material, namely mPEG‐block‐poly(phosphotyrosine) (mPEG‐b‐PpY), as a multi‐functional carrier for the facile fabrication of NDDS Pt/Ce6@mPEG‐b‐PpY (PtCeNP) for the co‐delivery of cisplatin and photosensitizer chlorin e6 (Ce6) via phosphato‐platinum coordination and π‐π stacking, respectively. PtCeNP improves the solubility, cellular uptake, and bioavailability of both parental drugs, and showed strong synergistic antitumor efficacy both in vitro and in vivo through combined chemo‐photodynamic therapy. Our results indicate that PpY is a biocompatible, multifunctional, and promising carrier material suitable for a variety of drugs and may simplify the design for co‐delivery systems.

Abstract:PURPOSE To describe a typical day for nurses who work with patients with cancer. DESIGN Multi-institutional, descriptive, qualitative. SETTING Six sites in different regions of the United States; rural and urban cancer and noncancer centers. SAMPLE 38 oncology nurses (mean age = 35 years; average time in nursing = 10 years; and in oncology = 7 years; 47% bachelor of science in nursing, 29% diploma, 13% associate degree in nursing, and 11% master's prepared. METHODS Phenomenological; content analysis of interviews. FINDINGS There is no typical day for oncology nurses. As such, the atypical becomes typical. This dialectic influenced how the nurses organized and managed their work. Management became a paradox or a juggling act--an attempt to control the uncontrollable, to manage the unmanageable. Time was a central metaphor used in the descriptions. The amount of time available influenced nurses' abilities to perform a certain quality and quantity of work. Demands on time, whether legitimate or illegitimate, were the context for the day-to-day work. IMPLICATIONS FOR NURSING PRACTICE It is necessary to recognize the value that nurses continually place on participation in patients' daily care, the unpredictability inherent in cancer and patient care, and the organizational routines that nurses use to accomplish their work.
An Enzyme-responsive Porphyrin Metal-organic Framework Nanosystem for Targeted and Enhanced Synergistic Cancer Photo-chemo Therapy

Abstract:Background: The clinical efficiency of photodynamic therapy (PDT) in combination with chemotherapy has proven to be a promising strategy for tumor treatment, yet is restricted by the high glutathione (GSH) concentration at the tumor site and nonspecific drug targeting. Objective: The goal of the current research was to create a biocompatible GSH-depleting and tumor- targeting nanoparticle (denoted as DOX/CA@PCN-224@HA) for the combined photodynamic and chemo photo-chemo) therapy. Methods: The nanoparticles were characterized by transmission electron microscopy (TEM). A UV-vis spectrophotometer was used to measure the drug loading efficiency (DE) and encapsulation efficiency (EE). The GSH-depleting ability was measured using Ellman's test. Confocal laser scan microscopy (CLSM) was used to assess the cellular uptake. MTT was adopted to evaluate the cytotoxicity of DOX/CA@PCN-224@HA against 4T1 cells. Results: The altered PCN-224 showed excellent monodispersing with a dimension of approximately 193 nm ± 2 nm in length and 79 nm ± 3 nm in width. The larger and spindle grid-like structure of PCN-224 obtains better dual-drug loading ability (DOX: 20.58% ± 2.60%, CA: 21.81% ± 1.98%) compared with other spherical PCN-224 nanoparticles. The ultimate cumulative drug release rates with hyaluronidase (HAase) were 74% ± 1% (DOX) and 45% ± 2% (CA) after 72 h. DOX/CA@PCN-224@HA showed GSH-consuming capability, which could improve the PDT effect. The drug-loaded nanoparticles could accurately target 4T1 cells through biological evaluations. Moreover, the released DOX and CA display cooperative effects on 4T1 cells in vitro. DOX/CA@PCN-224@HA nanoparticles showed inhibition against 4T1 cells with an IC50 value of 2.71 μg mL-1. Conclusion: This nanosystem displays great potential for tumor-targeted enhanced (photo-chemo) therapy.

Abstract:Chemo‐photodynamic therapy is an attractive strategy in tumor treatment. However, the combination of chemotherapeutic drug and photosensitizer in nanocarriers always elevates the risk of composition uncertainty. Here, a dual‐targeting photosensitizer‐peptide amphiphile conjugate (PpIX‐GGGK(TPP)GG‐GFLG‐R7‐RGD or pPAC) is designed to encapsulate doxorubicin (DOX) for enhanced chemo‐photodynamic tumor therapy. The amphiphilic nature of pPAC leads to the formation of core‐shell structured nanomicelles in aqueous media, where DOX is loaded in the inner core. The DOX@pPAC nanomicelle displays efficient tumor cellular uptake via integrin receptor‐mediated endocytosis using RGD peptide. After successful cell internalization with the aid of R8 peptide, DOX@pPAC exhibits rapid release of DOX and protoporphyrin IX‐peptide conjugate (PpIX‐peptide) due to the Cathepsin B‐triggered hydrolysis of GFLG linker. Simultaneously, triphenylphosphonium TPP cation will accumulate photosensitizer PpIX in subcellular mitochondria, followed by in situ generation of phototoxic reactive oxygen species (ROS) under light irradiation. In vitro investigations demonstrate that the synergistic chemotherapy and photodynamic therapy (PDT) of fabricated nanomicelles can significantly maximize the therapeutic effect against tumor cells with the minimal off‐target cytotoxicity. This work may provide an all‐in‐one nanosystem toward enhanced chemo‐photodynamic tumor therapy.

ROS-Responsive Polymeric Nanocarriers with Photoinduced Exposure of Cell-Penetrating Moieties for Specific Intracellular Drug Delivery

Abstract:In situ modulation of the surface properties on the micellar drug delivery nanocarriers offers an efficient method to improve the drug delivery efficiency into cells while maintaining stealth and stability during blood circulation. Light has been demonstrated to be a temporally and spatially controllable tool to improve cellular internalization of nanoparticles. Herein, we develop reactive oxygen species (ROS)-responsive mixed polymeric micelles with photoinduced exposure of cell-penetrating moieties via photodynamic ROS production, which can facilitate cellular internalization of paclitaxel (PTX) and chlorin e6 (Ce6)-coloaded micelles for the synergistic effect of photodynamic and chemotherapy. The thioketal-bond-linked block polymers poly(ε-caprolactone)-TL-poly(N,N-dimethylacrylamide) (PCL-TL-PDMA) with a long PDMA block are used to self-assemble into mixed micelles with PCL-b-poly(2-guanidinoethyl methacrylate) (PCL-PGEMA) consisting of a short PGEMA block, which are further used to coencapsulate PTX and Ce6. After intravenous injection, prolonged blood circulation of the micelles guarantees high tumor accumulation. Upon irradiation by 660 nm light, ROS production of the micelles by Ce6 induces cleavage of PDMA to expose PGEMA shells for significantly improved cellular internalization. The combination of photodynamic therapy and chemotherapy inside the tumor cells achieves improved antitumor efficacy. The design of ROS-responsive mixed polymeric nanocarriers represents a novel and efficient approach to realize both long blood circulation and high-efficiency cellular internalization for combined photodynamic and chemotherapy under light irradiation.
Red-light-triggered self-destructive mesoporous silica nanoparticles for cascade-amplifying chemo-photodynamic therapy favoring antitumor immune responses

Abstract:Although chemo-photodynamic therapy demonstrates promising synergetic therapeutic effects in malignant tumors, the light-controlled drug release, synergism and biocompatibility of current nanocarriers are limited. Herein, we report a red light-responsive, self-destructive carrier constructed using polyethylene glycol-modified, diselenide-bridged mesoporous silica nanoparticles. The carrier is co-encapsulated with the chemo-drug doxorubicin and the photosensitizer methylene blue for chemo-photodynamic therapy. Upon low-dose red light irradiation during photodynamic therapy (PDT), the reactive oxygen species (ROS) mediates a diselenide bond cleavage resulting in the degradation of the organosilica matrix and a dual drug release. This, in turn, results in a synergistic chemo-photodynamic performance in vitro and in vivo. More importantly, such cascade chemo-PDT boosts immunogenic cell death and robust anti-tumor immunity responses. Combination with a PD-1 checkpoint blockade further evokes a series of systemic immunity responses that suppress distant tumor growth and the pulmonary metastasis of breast cancer, as well as offer long-term protection against recurrent tumors. The presented work offers a controllable self-destruction nanoplatform for cascade-amplifying chemo-photodynamic therapy in response to external red light radiation.

Abstract:Reactive oxygen species (ROS)-responsive nanocarriers have aroused widespread interest in recent years. On the one hand, a high ROS level has been detected in many types of tumor cells. On the other hand, ROS generation is also induced during photodynamic, sonodynamic, or chemodynamic therapy. In addition, multiple types of polymers are sensitive to ROS. Therefore, numerous ROS-responsive polymeric nanocarriers with unique ROS-responsive characteristics have been developed. This review discusses ROS-sensitive polymeric nanocarriers to improve drug delivery efficacy. In particular, ROS-responsive nanocarriers for synergistic cancer therapy are highlighted. The development of novel ROS-sensitive nanocarriers holds great potential for combining ROS-mediated therapy, such as photodynamic therapy, and other therapies to achieve synergistic anticancer efficacy.

Abstract:The combination of multiple anti-tumor methods has shown significant application potential in overcoming the limitations of monotherapy. Photodynamic therapy (PDT) and chemotherapy combination is a promising strategy for reducing drug resistance and side effects. Here, inspired by the acidic environment of tumors, carboxymethyl chitosan-based pH-responsive nanovesicles were developed to co-deliver the chemotherapeutic drug doxorubicin (DOX) and photosensitizer 5-aminolevulinic acid (5-ALA). The in vitro drug release studies revealed that drugs could be responsively released when nanoparticles were triggered by the acidic environment. The controlled-release behavior improved drug retention and reduced the administration time. Our nanoparticles could significantly enhance the killing effect of drugs on tumor cells and increase intracellular levels of reactive oxygen species (ROS) compared to monotherapy, effectively achieving the effects of combined chemotherapy and PDT. The loaded DOX could kill tumor cells and the loaded 5-ALA could enhance the content of protoporphyrin IX (PpIX), resulting in excess ROS production to improve the effects of PDT. In summary, our nanoparticles could co-deliver the drugs and exert synergistical anti-tumor of PDT and chemotherapy by suppressing tumor cell proliferation and facilitating cell apoptosis.
Polymeric Nanoparticles with ROS‐Responsive Prodrug and Platinum Nanozyme for Enhanced Chemophotodynamic Therapy of Colon Cancer

Abstract:The combination of chemotherapy and photodynamic therapy (PDT) has promising potential in the synergistic treatment of cancer. However, chemotherapy and photodynamic synergistic therapy are impeded by uncontrolled chemotherapeutics release behavior, targeting deficiencies, and hypoxia‐associated poor PDT efficacy in solid tumors. Here, a platinum nanozyme (PtNP) loaded reactive oxygen species (ROS)‐responsive prodrug nanoparticle (CPT‐TK‐HPPH/Pt NP) is created to overcome these limitations. The ROS‐responsive prodrug consists of a thioketal bond linked with camptothecin (CPT) and photosensitizer‐2‐(1‐hexyloxyethyl)‐2‐devinyl pyropheophorbide‐a (HPPH). The PtNP in CPT‐TK‐HPPH/Pt NP can efficiently catalyze the decomposition of hydrogen peroxide (H2O2) into oxygen to relieve hypoxia. The production of oxygen can satisfy the consumption of HPPH under 660 nm laser irradiation to attain the on‐demand release of CPT and ensure enhanced photodynamic therapy. As a tumor diagnosis agent, the results of photoacoustic imaging and fluorescence imaging for CPT‐TK‐HPPH/Pt NP exhibit desirable long circulation and enhanced in vivo targeting. CPT‐TK‐HPPH/Pt NPs effectively inhibit tumor proliferation and growth in vitro and in vivo. CPT‐TK‐HPPH/Pt NP, with its excellent ROS‐responsive drug release behavior and enhanced PDT efficiency can serve as a new cancer theranostic agent, and will further promote the research of chemophotodynamic synergistic cancer therapy.
