Abstract:The filtration of air has attracted increasing attention during recent waves of viral infection. This review considers published literature regarding the usage of cellulose-based materials in air filtration devices, including face masks. Theoretical aspects are reviewed, leading to models that can be used to predict the relationship between structural features of air filter media and the collection efficiency for different particle size classes of airborne particulates. Collection of particles can be understood in terms of an interception mechanism, which is especially important for particles smaller than about 300 nm, and a set of deterministic mechanisms, which become important for larger particles. The effective usage of cellulosic material in air filtration requires the application of technologies including pulp refining and chemical treatments with such additives as wet-strength agents and hydrophobic sizing agents. By utilization of high levels of refining, in combination with freeze drying and related approaches, there are opportunities to achieve high levels of interception of fine particles. A bulky layer incorporating nanofibrillated cellulose can be used in combination with a coarser ply to achieve needed strength in a filter medium. Results of recent research show a wide range of development opportunities for diverse air filter devices containing cellulose.

Virus inactivation using an electrically conducting virus filter in biopharmaceutical manufacturing process

Abstract:Biopharmaceutical manufacturing processes using mammalian cells or plasma carry the risk of viral contamination. To mitigate these risks, it is essential to ensure viral clearance during the downstream process. Virus-retentive filters are used for size-based virus filtration, offering robust viral removal of more than 99.99%. However, virus breakthroughs have also been reported during virus filtration under certain conditions. In addition, these virus-retentive filters are disposable to ensure the safety of bioproducts, leading to significant costs and environmental concerns. In this study, innovative electrically conducting virus filters were fabricated using free-standing carbon veils (CV) and used to achieve additional virus inactivation after filtration. The viruses were captured in a CV-assisted virus filter, which was electrically heated using direct current to inactivate the viruses. This electrically conducting virus filter can inactivate viruses and can be reused up to five times. These results demonstrate that electrical conduction through electrical conducting damaged the phage capsid and eliminated the RNA genome, leading to bacteriophage inactivation. Moreover, it was confirmed that the electrically conducting virus filter could be reused up to five times without any changes to its physical or chemical structure. This study contributes to the reduction of process costs and environmental impacts by enabling the reuse of virus filters and enhancing the safety of the virus filtration process by preventing undesired virus breakthroughs.

Abstract:Several chromatographic approaches have been established over the last decades for the production of pharmaceutically relevant viruses. Due to the large size of these products compared to other biopharmaceuticals, e.g., proteins, convective flow media have proven to be superior to bead-based resins in terms of process productivity and column capacity. One representative of such convective flow materials is membranes, which can be modified to suit the particular operating principle and are also suitable for economical single-use applications. Among the different membrane variants, affinity surfaces allow for the most selective separation of the target molecule from other components in the feed solution, especially from host cell-derived DNA and proteins. A successful membrane affinity chromatography, however, requires the identification and implementation of ligands, which can be applied economically while at the same time being stable during the process and non-toxic in the case of any leaching. This review summarizes the current evaluation of membrane-based affinity purifications for viruses and virus-like particles, including traditional resin and monolith approaches and the advantages of membrane applications. An overview of potential affinity ligands is given, as well as considerations of suitable affinity platform technologies, e.g., for different virus serotypes, including a description of processes using pseudo-affinity matrices, such as sulfated cellulose membrane adsorbers.

Abstract:Viruses are the most abundant microorganisms on the earth, their existence in contaminated waters possesses a significant threat to humans. Waterborne viral infections could be fatal to sensitive population including young child, the elderly, and the immune-compromised. It is imperative to remove viruses during water treatment to better protect public health, especially in the light of evidence of detection of coronaviruses genetic fragments in raw sewage. We reported bench-scale experiments evaluating the extent and mechanisms of removal of a model virus (spring viremia of carp virus, SVCV) in water by adsorption. Microspheres made by boronic acid-modified bacterial cellulose with excellent mechanical strength were successfully fabricated as packing materials for the column to remove glycoproteins and enveloped viruses from water. The synthesized adsorbent was characterized by attenuated total reflectance Fourier transform infrared (ATR-FTIR) spectroscopy, Atomic Force Microscopy (AFM), Scanning Electron Microscopy (SEM), and Brunauer Emmett Teller (BET) measurement. The adsorption efficiency of glycoproteins was investigated by SDS-PAGE and the Broadford protein assay, while the binding capacity with the virus (spring viremia of carp virus) was monitored by cell culture to calculate the viral cytopathic effect and viral titer caused by the virus. The data obtained from the above experiments showed that ∼3-log removal of SVCV in 3 h, which significantly reduced the virus concentration from microspheres packed column. The present study provides substantial evidence to prove beyond doubt that material based on bacterial cellulose seems to have the potential for virus removal from water which can be extended to systems of significant importance.

Abstract:Human viral contamination in drinking and recreational water may persist for extensive periods of time and cause a significant health risk concern. The aim of this study is to evaluate a viral recovery method using a new electropositive charged nanoalumina filter and to compare results with the widely used negatively charged HAWP filter by Millipore Inc. The recovery of infectious recombinant adenovirus type 5 (rAd5) was tested using the Fluorescence-Activated Cell Sorting (FACS) assay, in parallel with viral genomes recovery assay by quantitative PCR (qPCR). The mean infectivity recoveries were 82–91% by nanoalumina filters eluted with 3% beef extract (BE, pH6.0), and 78–90% by HAWP filters eluted with 3% BE (pH 9.0), respectively, from 1L of environmental samples seeded with 1pfu/mL rAd5. The mean genome recoveries were 16–35% by nanoalumina filters eluted with BE (pH 6.0), and 29–66% by HAWP filters eluted with NaOH (pH 10.8) from different types of water, respectively. Water quality, concentration of viruses, filters, and elution buffers are factors that determine the viral recovery efficiencies. The nanoalumina filters also had higher filtration rates than HAWP filters for large volumes of environmental water samples (up to10L), thus, have an advantage in concentrating infectious viruses from environments without pre-filtration, adjusting pH or adding multivalent cations.

Abstract:Retention of bacteriophages (phi 6, PR772, T1, and PP7) and mammalian viruses (poliovirus and influenza A virus) by a hydrophilic triple layer PVDF microporous membrane, the Ultipor VF grade DV50 membrane, was evaluated. Challenges of membrane discs or pleated filter cartridges were performed at concentrations of 10(6)-10(8) PFU/mL in one or more of the following carrier fluids: water, saline, gelatin (0.1%) in phosphate buffer, Dulbecco's Modified Eagle Medium (MEM), and MEM supplemented with 10% fetal bovine serum (MEM + 10). The data demonstrate a minimum log titer reduction (LTR) of 6 for viruses larger than 50 nm irrespective of the carrier fluid. Protein transmission levels of greater than 95% for IgG and albumin were achieved. For integral pleated filter cartridges, correlation between a nondestructive integrity test (using the forward flow integrity test method) and virus retention was demonstrated. The Ultipor VF grade DV50 filter can be applicable in the manufacture of biologicals and biopharmaceuticals, where high protein transmission and consistent viral titer reduction are desired.

Abstract:Virus removal by membrane filtration is challenging due to the small size of these microorganisms. For microfiltration (MF) membranes, which have pore sizes above 100 nm (i.e., larger than most viruses), surface interactions are of critical importance in determining virus removal. The present study aims to evaluate the impact of surface interactions on virus removal by MF membranes wherein predictions by extended DLVO (XDLVO) modeling are compared with experimental data on virus clearance. The study employed four viruses (MS2, Qβ, phiX174, phi6), two MF cartridge membranes (nylon and PTFE), and several solution chemistry conditions to cover a range of virus-membrane surface interaction scenarios. For the hydrophilic nylon membrane, the highest log removal values (LRVs) were observed at the pH when the filter carried a slightly positive charge. For the hydrophobic PTFE membrane, higher LRVs were attained for high ionic strength solutions, wherein the electric double layer was compressed, even at pH values where the filter was negatively charged. We conclude that the electrostatic interaction determines whether viruses can approach the membrane and hydrophobic interactions are a critical factor in retaining viruses near the membrane surface, preventing viral leakage into the filtrate. The surface interactions have to be considered in virus membrane filtration.

Abstract:Recently, the potential dangers of viral infection transmission through water and air have become the focus of worldwide attention, via the spread of COVID-19 pandemic. The occurrence of large-scale outbreaks of dangerous infections caused by unknown pathogens and the isolation of new pandemic strains require the development of improved methods of viruses' inactivation. Viruses are not stable self-sustaining living organisms and are rapidly inactivated on isolated surfaces. However, water resources and air can participate in the pathogens' diffusion, stabilization, and transmission. Viruses inactivation and elimination by adsorption are relevant since they can represent an effective and low-cost method to treat fluids, and hence limit the spread of pathogen agents. This review analyzed the interaction between viruses and carbon-based, oxide-based, porous materials and biological materials (e.g., sulfated polysaccharides and cyclodextrins). It will be shown that these adsorbents can play a relevant role in the viruses removal where water and air purification mostly occurring via electrostatic interactions. However, a clear systematic vision of the correlation between the surface potential and the adsorption capacity of the different filters is still lacking and should be provided to achieve a better comprehension of the global phenomenon. The rationalization of the adsorption capacity may be achieved through a proper physico-chemical characterization of new adsorbents, including molecular modeling and simulations, also considering the adsorption of virus-like particles on their surface. As a most timely perspective, the results on this review present potential solutions to investigate coronaviruses and specifically SARS-CoV-2, responsible of the COVID-19 pandemic, whose spread can be limited by the efficient disinfection and purification of closed-spaces air and urban waters.
Nanofibrous filters: A promising solution for the efficient capture of polydisperse viral aerosols

Abstract:This study investigates the effectiveness of electrospun nanofibrous filters in capturing polydisperse virus-containing aerosols and the subsequent release of viruses, in comparison with standard commercial filters used in respirators, military gas masks, and devices for airborne virus sampling. We assessed the performance of these filters in capturing and releasing polydisperse aerosols containing MS2 bacteriophage, as well as in their ability to filter monodisperse dioctyl phthalate aerosols measuring 0.185 and 0.3 μm in diameter. Our findings indicate that nanofibrous filters provide superior filtration efficiency for monodisperse aerosols, achieving a reduction in the concentration of penetrating aerosols by 2–3 orders of magnitude compared to their commercial counterparts. However, this enhanced efficiency is accompanied by a higher pressure drop and a lower quality factor, underscoring the need for further improvements. Additionally, our research confirms the feasibility of producing aligned nanofibers via multiple-jet needleless electrospinning, though alignment did not significantly impact filtration efficiency. Nanofibrous filters demonstrated filtration efficiency for aerosolized virus-containing particles that was comparable to or better than that of commercial filters. Notably, certain nanofibrous filters exhibited exceptionally low rates of viral aerosol capture and release, indicating a potential for virus neutralization. Moreover, filters made from water-soluble electrospun polyvinylpyrrolidone significantly outperformed those made from gelatin in terms of viral particle release, underscoring the potential of water-soluble electrospun materials in improving viral particle collection. Overall, our study highlights the significant promise of electrospun nanofibers in public health, especially in enhancing defenses against the transmission of viral aerosols. Recombinant products such as monoclonal antibodies are expressed by mammalian, bacteria or yeast cells in fermentors, and within ascites fluid, or fluids from transgenic mammals. Mammalian cell lines may contain endogenous viruses that are generated in the bioreactor. Endogenous retroviruses are expressed because the retroviral genome is integrated into the cell line and cannot be screened-out during the creation of the Master Cell Bank. This causes retrovirus like particles, or RVLP’s, to be produced within the bioreactor. Products can also become contaminated by adventitious viruses which enter the process streams.

Abstract:The control of airborne contaminants is of great interest in improving air quality, which has deteriorated more and more in recent years due to strong industrial growth. In the last decades, cellulose has been largely proposed as suitable feedstock to build up eco-friendly materials for a wide range of applications. Herein, the issue regarding the use of cellulose to develop air-filtering systems is addressed. The review covers different cellulose-based solutions, ranging from aerogels and foams to membranes and films, and to composites, considering either particulate filtration (PM10, PM2.5, and PM0.3) or gas and water permeation. The proposed solutions were evaluated on the bases of their quality factor (QF), whose high value (at least of 0.01 Pa−1 referred to commercial HEPA (high-efficiency particulate air) filters) guarantees the best compromise between high filtration efficiency (>99%) and low pressure drop (<1 kPa/g). To face this aspect, we first analyzed the different morphological aspects which can improve the final filtration performance, outlining the importance on using nanofibers not only to increase surface area and to modulate porosity in final solutions, but also as reinforcement of filters made of different materials. Besides the description of technological approaches to improve the mechanical filtration, selected examples show the importance of the chemical interaction, promoted by the introduction of active functional groups on cellulose (nano)fibers backbone, to improve filtration efficiency without reducing filter porosity.

Abstract:The wide availability and diversity of dangerous microbes poses a considerable problem for health professionals and in the development of new healthcare products. Numerous studies have been conducted to develop membrane filters that have antibacterial properties to solve this problem. Without proper protective filter equipment, healthcare providers, essential workers, and the general public are exposed to the risk of infection. A combination of nanotechnology and biosorption is expected to offer a new and greener approach to improve the usefulness of polysaccharides as an advanced membrane filtration material. Nanocellulose is among the emerging materials of this century and several studies have proven its use in filtering microbes. Its high specific surface area enables the adsorption of various microbial species, and its innate porosity can separate various molecules and retain microbial objects. Besides this, the presence of an abundant OH groups in nanocellulose grants its unique surface modification, which can increase its filtration efficiency through the formation of affinity interactions toward microbes. In this review, an update of the most relevant uses of nanocellulose as a new class of membrane filters against microbes is outlined. Key advancements in surface modifications of nanocellulose to enhance its rejection mechanism are also critically discussed. To the best of our knowledge, this is the first review focusing on the development of nanocellulose as a membrane filter against microbes.

Abstract:Air pollution is an increasing concern all over the world due to its adverse effects on human health. It claims thousands of lives every year. Hence, the demand for the ventilator, respirator, facemask, body protection, and hospital air filtration has been surging dramatically during the COVID-19 pandemic. Nanofiber membranes with optimal characteristics, such as a high specific surface area and porous microstructure with interconnected pores, can efficiently capture the fine particles (such as bacteria, fungi, virus, etc.). Recently, various types of polymers have been fabricated as electrospun fibrous membranes to be used as an anti-bacterial or anti-viral air filtration media. This review presents a brief overview of air filtration history and its main mechanisms and then the latest research about air filtration with antibacterial and antiviral properties will be reviewed.

Serial virus filtration: A case study evaluating the product-dependent impact of control strategies on process efficiency

Abstract:The production of biopharmaceutical products carries an inherent risk of contamination by adventitious viruses. Historically, these manufacturing processes have incorporated a dedicated virus filtration step to ensure product safety. However, challenging process conditions can lead to passage of small viruses to the permeate pool and an overall decrease in the desired virus logarithmic reduction value (LRV) for the process. The implementation of serial virus filtration has improved the robustness of such processes, albeit concerns about increased operating times and process complexity have limited its implementation. This work focused on optimizing a serial filtration process and identifying process control strategies to provide maximum efficiency while ensuring proper controls for process complexity. Constant TMP was identified as the optimal control strategy, which combined with the optimal filter ratio, resulted in a robust and faster virus filtration process. To demonstrate this hypothesis, data with two filters connected in series (1:1 filter ratio) are presented for a representative non-fouling molecule. Similarly, for a fouling product, the optimal setup was a combination of a filter connected in series to two filters operated in parallel (2:1 filter ratio). The optimized filter ratios bring cost- and time-savings benefits to the virus filtration step, thereby offering improved productivity. The results of risk and cost analyses performed as part of this study combined with the control strategy, offer companies a toolbox of strategies to accommodate products with different filterability profiles in their downstream processes. This work demonstrates that the safety advantages of performing filters in series can be achieved with minimal increases in time, cost, and risk.
Coliphages as viral indicators of sanitary significance for drinking water

Abstract:Coliphages are virus that infect coliform bacteria and are used in aquatic systems for risk assessment for human enteric viruses. This mini-review appraises the types and sources of coliphage and their fate and behavior in source waters and engineered drinking water treatment systems. Somatic (cell wall infection) and F + (male specific) coliphages are abundant in drinking water sources and are used as indicators of fecal contamination. Coliphage abundances do not consistently correlate to human enteric virus abundance, but they suitably reflect the risks of exposure to human enteric viruses. Coliphages have highly variable surface characteristics with respect to morphology, size, charge, isoelectric point, and hydrophobicity which together interact to govern partitioning and removal characteristics during water treatment. The groups somatic and F + coliphages are valuable for investigating the virus elimination during water treatment steps and as indicators for viral water quality assessment. Strain level analyses (e.g., Qβ or GA-like) provide more information about specific sources of viral pollution but are impractical for routine monitoring. Consistent links between rapid online monitoring tools (e.g., turbidity, particle counters, and flow cytometry) and phages in drinking water have yet to be established but are recommended as a future area of research activity. This could enable the real-time monitoring of virus and improve the process understanding during transient operational events. Exciting future prospects for the use of coliphages in aquatic microbiology are also discussed based on current scientific evidence and practical needs.

Abstract:The recent COVID-19 pandemic has once again drawn attention to the risks of pathogenic viruses, as viruses can be transmitted through water environments, posing a threat to human health. Membrane technology, as a novel, green, and effective water treatment process, has been widely applied in the removal of pathogenic viruses from wastewater and drinking water. However, compared to refined centralized water treatment processes in urban areas, rural areas still lack effective water treatment strategies for decentralized water supply and drainage systems. Therefore, this review provides a comprehensive analysis of the hazards and transmission pathways of viruses, the mechanisms and influencing factors of virus removal by membrane processes. Based on the characteristics of centralized and decentralized water treatment systems, it further summarizes the application scenarios and effectiveness of the most widely used membrane processes. We emphasize the need for context-specific development of membrane-based water treatment processes tailored to urban or rural areas, addressing both water supply and wastewater treatment processes, to ensure comprehensive public health safety in water environments. In addition, this review also discusses several challenges that membrane technology faces in virus removal, providing new insights for further research in membrane processes.

Abstract:In this study, a hollow polyvinylidene fluoride (PVDF) fiber was developed and modified into a Janus membrane (Janus-VM) and an entirely hydrophobic membrane (Hydrophobic-VM) for removing 20-nm-scale viruses during biopharmaceutical processing. The Janus-VM has anionic properties on the outer and feed hydrophilic surfaces, and the inner pore surface (I.P.S) maintains the anionic-hydrophobic properties of the PVDF. Theoretically, an electrostatic repulsion force might be formed between the surface of the strong anionic membrane and anionic bacteriophages, preventing bacteriophages from adsorbing to the membrane surface. As a result, water flux can be increased through the reduction of membrane fouling. The PVDF membrane was modified through UV-irradiation, which causes –OH radicals to form on the membrane surface, and chemical grafting with anionic-hydrophobic/anionic-hydrophilic modifiers, which enhances the membrane's anion characteristics (−27, −48 mV). Additionally, the average pore size was reduced from 34 nm to less than 20 nm through chemical surface modification, after which the Janus membrane showed an excellent initial water flux, with the protein recovery ratio (PRR) approaching 98.9 %. UV absorbance analysis revealed 100 % removal efficiency for 20- and 40-nm gold particles on the membranes. Furthermore, the initial log reduction value (LRV) was observed through flux performance using MS2 and PP7 bacterial phages, for which the Janus membrane showed an excellent virus removal performance (LRV > 6.8 and 6.5).

Abstract:Human enteric viruses are important etiological agents of waterborne diseases. Environmental waters are usually contaminated with low virus concentration requiring large concentration factors for effective detection by (RT)-qPCR. Low-pressure reverse osmosis is often used to remove water contaminants, but very few studies focused on the effective virus removal of reverse osmosis treatment with feed concentrations as close as possible to environmental concentrations and principally relied on theoretical virus removal. The very low viral concentrations usually reported in the permeates (i.e. at least 5 log of removal rate) mean that very large volumes of water need to be analysed to have sufficient sensitivity and assess the process efficiency. This study evaluates two methods for the concentration of adenoviruses, enteroviruses and MS2 bacteriophages at different viral concentrations in large (< 200 L) and very large (> 200 L) volumes. The first method is composed of two ultrafiltration membranes with low-molecular weight cut-offs while the second method primarily relies on adsorption and elution phases using electropositive-charged filters. The recovery rates were assessed for both methods. For the ultrafiltration-based protocol, recovery rates were similar for each virus studied: 80% on average at high virus concentrations (106-107 viruses L-1) and 50% at low virus concentrations (103-104 viruses L-1). For the electropositive-charged filter-based method, the average recoveries obtained were about 36% for ADV 41, 57% for CV-B5 and 1.6% for MS2. The ultrafiltration-based method was then used to evaluate the performance of a low-pressure reverse osmosis lab-scale pilot plant. The retentions by reverse osmosis were similar for all studied viruses and the validated recovery rates applied to the system confirmed the reliability of the concentration method. This method was effective in concentrating all three viruses over a wide range of viral concentrations. Moreover, the second concentration method using electropositive-charged filters was studied, allowing the filtration of larger volumes of permeate from a semi-industrial low-pressure reverse osmosis pilot plant. This reference method was used because of the inability of the UF method to filter volumes on the order of one cubic metre.
