Abstract:The demand for wound care is increasing globally and the traditional wound dressings offer limited support to the healing process, failing to meet the requirements of current healthcare system. Drug-controlled release nanotechnology has garnered interest recently in the field of biomedicine. Electrospun nanofibers are considered as an advanced wound dressing due to their unique structure and a biological function similar to the extracellular matrix. In this study, a polycaprolactone (PCL)-polyethylene glycol (PEG) loaded with nisin was developed by electrospinning technique. The fabricated nanofiber was characterized by scanning electron microscopy (SEM) and Fourier transform infrared spectroscopy (FTIR). The degradation index, in vitro release studies, antioxidant activity, and cell cytotoxicity analyses of the nanofibers were also performed. The antibacterial activity of the nanofiber was evaluated against wound causing skin pathogens like Staphylococcus aureus, Pseudomonas aeruginosa, Enterococcus faecalis , and Klebsiella pneumoniae . The FTIR data demonstrate the interactions between PCL, PEG, and nisin. The presence of nisin in the fiber was confirmed by the characteristic bands of nisin at 3398.63, 1643.23, and 1242.17 cm −1 . The nanofibers exhibited maximum controlled nisin release of 94.3% at 24 h. The nisin-loaded nanofibers exhibited antibacterial activity against all the indicator organisms used in the study. About 78% viability was observed on nisin-loaded nanofibers during the MTT assay. The results indicated that the fabricated antibacterial nanofiber has the potential for wound healing application and the treatment of bacterial infections in wounds.

Abstract:The dressing materials that provide surface protection, bacteriostatic activities, and tissue regeneration are important for the treatment and management of complex wounds. This study aimed to evaluate the wound-healing properties of electrospun nanofibers containing a blend of methylcellulose (MC) and polyvinyl alcohol (PVA). The nanofibers were tested in single-layered (S-NFs) and multilayered (M-NFs) forms (PCL/MC-PVA/PCL). In vitro scratch assay using L929 cells and in vivo experiments on Wistar rats were conducted. The results showed that both S-NFs and M-NFs significantly accelerated wound closure by promoting cell migration. M-NFs demonstrated superior wound healing activity compared to S-NFs. Additionally, M-NFs exhibited faster skin epithelization compared to S-NFs. Histopathological evaluation confirmed the absence of irritation or lesions on the healed wound surface. Overall, the study concluded that these polymeric nanofibers have the potential to be used as self-wound healing dressings. They are safe, non-toxic, biodegradable, and biocompatible.

High Glucose Concentration Stimulates NHE-1 Activity in Distal Nephron Cells: the Role of the Mek/Erk1/2/p90RSK and p38MAPK Signaling Pathways

Abstract:Aims: In models of diabetes, distal nephron cells contribute to glucose uptake and oxidation. How these cells contribute to the use of glucose for the regulation of H+ extrusion remains unknown. We used Madin-Darby Canine Kidney (MDCK) cells to investigate the effect of acute or chronic high glucose concentration on the abundance and activity of the Na+/H+ exchanger (NHE-1). Methods: Using RT-PCR, we also evaluated the mRNA expression for sodium glucose co-transporters SGLT1 and SGLT2. Protein abundance was analyzed using immunoblotting, and intracellular pH (pHi) recovery was evaluated using microscopy in conjunction with the fluorescent probe BCECF/AM. The Na+-dependent pHi recovery rate was monitored with HOE-694 (50 µM) and/or S3226 (10 µM), specific NHE-1 and NHE-3 inhibitors. Results: MDCK cells did not express the mRNA for SGLT1 or SGLT2 but did express the GLUT2, NHE-1 and NHE-3 proteins. Under control conditions, we observed a greater contribution of NHE-1 to pHi recovery relative to the other H+ transporters. Acute high glucose treatment increased the HOE-694-sensitive pHi recovery rate and p-Erk1/2 and p90RSK abundance. These parameters were reduced by PD-98059, a Mek inhibitor (1 µM). Chronic high glucose treatment also increased the HOE-694-sensitive pHi recovery rate and p-p38MAPK abundance. Both parameters were reduced by SB-203580, a p38MAPK inhibitor (10 µM). Conclusion: These results suggested that extracellular high glucose stimulated NHE-1 acutely and chronically through Mek/Erk1/2/p90RSK and p38MAPK pathways, respectively.

Abstract:The development of innovative multifunctional wound healing materials based on natural bioactives has attracted a great deal of attention in recent years due to their ability to potentiate the healing process and minimize the challenges linked with conventional wound dressings. The current study demonstrates the development and evaluation of a novel nanofibrous wound healing mat made of Gallic acid-chitosan conjugate (GAC) incorporated within a polycaprolactone (PCL) matrix using an electrospinning technique. The GAC conjugate was synthesized by green synthesis using a free radical polymerization reaction to improve the chitosan's free radical scavenging properties. FTIR, DSC, XRD, and UV spectral analysis characterized the formation of GAC conjugate. The red shift observed in the UV spectrum suggested alterations in electronic transitions resulting from the creation of covalent bonds between CA and GS. The appearance of broader and weaker peaks in DSC and the reduced crystallinity in XRD spectrum, signify structural modifications, thereby providing valuable insights into the interaction and structural changes occurring in the GAC conjugates.The SEM image revealed a porous flake-like structural network of GAC conjugate due to the reduced intermolecular hydrogen bonding between them. The functional layer of the nanofibrous mat was produced by electrospinning GAC conjugate with polycaprolactone (PCL) to provide mechanical integrity to the nanofibrous mat. The electrospun nanofibers were characterized using FTIR, DSC, and SEM and evaluated for cell cytotoxicity, angiogenesis, and in vivo wound healing potential in female Wistar rats. The FTIR spectra confirmed the presence of GAC conjugate in the nanofibers, validating the conjugation process. The results demonstrated the successful incorporation of GAC conjugate within the PCL nanofibrous mat, thereby confirming the improvement in the biological properties of the mats. The nanofibrous mat showed angiogenesis activity in the yolk sac membrane assay confirming the property of nanofibers to facilitate cellular adhesion, migration and promotion of angiogenesis with no cytotoxic effect on the viability of mouse stromal cells. On the sixteenth day of therapy, wounds dressed with GAC/PCL nanofibrous mat exhibited a significant improvement in the percent wound closure (94.43%) as compared to the control group (p < 0.05).These results supported the application of GAC/PCL as a novel biomaterial for wound dressing to accelerate the process of wound healing. Abstract:Wound dressings can be used to create a temporary healing environment and expedite the wound healing process. Ulvan (ULV) is a sulfated polysaccharide with potent antiviral and anti-inflammatory activities. Polycaprolactone (PCL) is a hydrophobic biodegradable polyester that exhibits slow degradation, strong mechanical strength, and excellent biocompatibility. Electrospun nanofiber matrices mimic the microstructure of the extracellular matrix, allowing them to promote cell proliferation and differentiation. Therefore, the primary objective of this study was to fabricate a polycaprolactone-ulvan fibrous composite mat (PCL-ULV) using the electrospinning technique and to investigate its physical and chemical properties. To assess the characteristics of PCL-ULV, scanning electron microscopy (SEM) was utilized to examine its morphology and diameter distribution. Fourier transform infrared (FTIR) spectroscopy, calcofluor white staining, and monosaccharide analysis were employed to analyze the components of PCL-ULV. Additionally, the water contact angle was measured to evaluate the hydrophilicity. Furthermore, the proliferation and morphology of and gene expression in NIH3T3 fibroblasts on PCL-ULV were assessed. The results showed that the average PCL-ULV fiber diameter was significantly smaller than that of the PCL fibers. The water contact angle measurements indicated that PCL-ULV exhibited better hydrophilicity than the PCL mat. FTIR, calcofluor white staining, and monosaccharide analyses demonstrated that ULV could be successfully coelectrospun with PCL. NIH3T3 fibroblasts cultured on PCL and PCL-ULV showed different cellular behaviors. On PCL-ULV, cell adhesion, proliferation, and stretching were greater than those on PCL. Moreover, the behavior of NIH3T3 fibroblasts on PCL and PCL-ULV differed, as the cells on PCL-ULV exhibited higher proliferation and more stretching. Furthermore, NIH3T3 fibroblasts cultured on ULV-PCL showed higher α-SMA and MMP-9 gene expression and a lower ratio of TIMP-1/MMP-9 than those cultured on PCL. Notably, scarless wounds display lower TIMP/MMP expression ratios than scarring wounds. Thus, the fibrous composite mat PCL-ULV shows potential as a wound dressing for scarless wound healing.

Abstract:Cross-linked polyvinyl alcohol (PVA) and polyvinyl pyrrolidone (PVP) composite nanofibrous membranes have been prepared by electrospinning. Mechanical properties of the membranes improved significantly with PVP addition. PVP improved hydrophilicity and sustainable degradation of the membranes. Biocompatibility of the membranes was assessed by in vitro culture of native skin cells (L929 fibroblast and HaCaT keratinocytes). Tests showed sustained release of the antibiotic ciprofloxacin hydrochloride monohydrate by the membranes. Further, zone of inhibition study against Staphylococcus aureus growth demonstrated protective action against external pathogenic microbes. These studies show these simple PVA-PVP nanofibrous membranes are promising interactive antibiotic-eluting wound dressing materials.

Abstract:Chronic wounds have become a growing concern as they can have a profound impact on individuals, potentially resulting in mortality. It is crucial to prevent and manage bacterial infections, particularly drug-resistant ones. Antimicrobial peptides, such as LL-37, can firmly eliminate pathogens. Additionally, the process of angiogenesis, facilitated by growth factors like VEGF, is essential for tissue repair and wound healing. To enhance the stability and bioavailability of therapeutic agents, targeted delivery strategies utilizing Chitosan-based carriers have been employed. Electrospun biopolymers in advanced wound dressings have revolutionized wound care by providing a more effective and efficient solution for promoting tissue regeneration and speeding up the healing process. The present investigation utilized Chitosan nanoparticles to encapsulate the recombinant LL37 peptide and VEGF. An in-depth investigation was carried out to analyze the biophysical and morphological traits of the LL37-CSNPs and VEGF-CSNPs. The first support layer consisted of PCL electrospun nanofiber, followed by the electrospinning of PVA/CsLL37, PVA/CsVEGF, and PVA/CsLL37/CsVEGF onto the PCL layer. An in vitro examination assessed the fabricated nanofibers' morphological, mechanical, and biological characteristics. The antimicrobial effects were tested on methicillin-resistant Staphylococcus aureus (MRSA). The in vivo experiments assessed the antibacterial and wound-healing capabilities of the nanofibers. The findings validated the continuous release of LL37 and VEGF. The composite material PCL/PVA/CsLL37/CsVEGF demonstrated potent bactericidal and antioxidant characteristics. The cytotoxic assay demonstrated the biocompatibility of the fabricated nano mats and their potential to accelerate fibroblast cell proliferation. The efficacy of PVA/CsLL37/CsVEGF in promoting wound healing was confirmed through an in vivo wound healing assay. Furthermore, the histological analysis provided evidence of faster epidermal formation and improved antibacterial activity in wounds covered with PVA/CsLL37/CsVEGF. Adding LL37 and VEGF to the composite material improves the immune response and promotes blood vessel formation, accelerating wound healing and decreasing inflammation.

Abstract:Purpose: In the present study, the poly (ε-caprolactone)/cellulose nanofiber containing ZrO2 nanoparticles (PCL/CNF/ZrO2 ) nanocomposite was synthesized for wound dressing bandage with antimicrobial activity. Methods: PCL/CNF/ZrO2 nanocomposite was synthesized in three different zirconium dioxide amount (0.5, 1, 2%). Also the prepared nanocomposites were characterized by Infrared spectroscopy (FT-IR), X-ray diffraction (XRD), differential scanning calorimetry (DSC), and thermogravimetric analysis (TGA). In addition, the morphology of the samples was observed by scanning electron microscopy (SEM). Results: Analysis of the XRD spectra showed a preserved structure for PCL semi-crystalline in nanocomposites and an increase in the concentrations of ZrO2 nanoparticles, the structure of nanocomposite was amorphous as well. The results of TGA, DTA, DSC showed thermal stability and strength properties for the nanocomposites which were more thermal stable and thermal integrate compared to PCL. The contact angles of the nanocomposites narrowed as the amount of ZrO2 in the structure increased. The evaluation of biological activities showed that the PCL/CNF/ZrO2 nanocomposite with various concentrations of ZrO2 nanoparticles exhibited moderate to good antimicrobial activity against all tested bacterial and fungal strains. Furthermore, cytocompatibility of the scaffolds was assessed by MTT assay and cell viability studies proved the non-toxic nature of the nanocomposites. Conclusion: The results show that the biodegradability of nanocomposite has advantages that can be used as wound dressing.

Natural and synthetic polymers may be electrospun in the form of a blend. Besides, the antibiotics such as linezolid, enrofloxacin, and vancomycin are used in wound dressing due to their antibacterial properties. In this research, the blend nanofibrous structures made of PCL and gelatin (Gel) with a 25:75 ratio were produced for wound dressing applications. Clindamycin HCL as a drug was added to Gel and PCL polymeric solutions. Surface morphology, functional groups, and hydrophilicity of nanofibers were examined using SEM, FT-IR spectroscopy, and contact angle measurement, respectively. In addition, the antibacterial properties of nanofibers were evaluated quantitatively. The drug release mechanism of samples was investigated which the best-fitted model was recognized Korsmeyer-Peppas model. SEM images of scaffolds demonstrated uniform and bead-free morphology that, with incorporating the 6% of the drug, the diameters of mats were decreased from 398 nm to 303 nm. Moreover, the samples showed proper hydrophilicity and antibacterial properties against a gram-positive (89%) and a gram-negative (98%) bacterium. Finally, the nanofibers are capable of releasing the clindamycin gradually for 6 days.
Curcumin‐loaded Poly(ε‐caprolactone) Nanofibres: Diabetic Wound Dressing with Anti‐oxidant and Anti‐inflammatory Properties

Abstract:1. Curcumin is a naturally occurring poly-phenolic compound with a broad range of favourable biological functions, including anti-cancer, anti-oxidant and anti-inflammatory activities. The low bioavailability and in vivo stability of curcumin require the development of suitable carrier vehicles to deliver the molecule in a sustained manner at therapeutic levels. 2. In the present study, we investigated the feasibility and potential of poly(caprolactone) (PCL) nanofibres as a delivery vehicle for curcumin for wound healing applications. By optimizing the electrospinning parameters, bead-free curcumin-loaded PCL nanofibres were developed. 3. The fibres showed sustained release of curcumin for 72 h and could be made to deliver a dose much lower than the reported cytotoxic concentration while remaining bioactive. Human foreskin fibroblast cells (HFF-1) showed more than 70% viability on curcumin-loaded nanofibres. 4. The anti-oxidant activity of curcumin-loaded nanofibres was demonstrated using an oxygen radical absorbance capacity (ORAC) assay and by the ability of the fibres to maintain the viability of HFF-1 cells under conditions of oxidative stress. 5. The curcumin-loaded nanofibres also reduced inflammatory induction, as evidenced by low levels of interleukin-6 release from mouse monocyte-macrophages seeded onto the fibres following stimulation by Escherichia coli-derived lipopolysaccharide. 6. The in vivo wound healing capability of the curcumin loaded PCL nanofibres was demonstrated by an increased rate of wound closure in a streptozotocin-induced diabetic mice model. 7. These results demonstrate that the curcumin-loaded PCL nanofibre matrix is bioactive and has potential as a wound dressing with anti-oxidant and anti-inflammatory properties.
Nanofibrous Wound Dressing with a Smart Drug Delivery System: Poly(N-Isopropylacrylamide)-Conjugated Polycaprolactone Nanofibers Loaded with Curcumin

Abstract:In response to clinical demands for advanced wound dressings, a "smart bandage" was developed by combining polycaprolactone (PCL) nanofibers and the thermoresponsive polymer poly(N-isopropylacrylamide) (PNIPAAm). This bandage incorporates curcumin, an anti-inflammatory and antioxidant agent, to enhance wound healing. The LCST of PNIPAAm (32 C) enables an "on-off" drug delivery system, transitioning from a hydrophilic coil to a hydrophobic state. The nanofiber matrix was characterized using FTIR spectroscopy, SEM, water contact angle, and dynamic mechanical analysis. Curcumin release was assessed both above and below the LCST. The bandage is comfortable, easy to apply and remove, and promotes rapid wound healing. In vitro studies confirmed its non-toxicity to human dermal fibroblast cells. This "smart bandage" represents a significant advancement in wound care, with the potential to meet clinical requirements and enhance patient compliance, offering controlled drug delivery combined with the therapeutic benefits of curcumin.

Abstract:The current study focuses on the development of gelatin-coated polycaprolactone (PCL) nanofibers co-loaded with luliconazole and naringenin for accelerated healing of infected diabetic wounds. Inherently, PCL nanofibers have excellent biocompatibility and biodegradation profiles but lack bioadhesion characteristics, which limits their use as dressing materials. So, coating them with a biocompatible and hydrophilic material like gelatin can improve bioadhesion. The preparation of nanofibers was done with the electrospinning technique. The solid state characterization and in-vitro performance assessment of nanofibers indicate the formation of uniformly interconnected nanofibers of 200–400 nm in diameter with smooth surface topography, excellent drug entrapment, and a surface pH of 5.6–6.8. The antifungal study showed that the nanofiber matrix exhibits excellent biofilm inhibition activity against several strains of Candida . Further, in-vivo assessment of nanofiber performance on C. albicans infected wounds in diabetic rats indicated accelerated wound healing efficacy in comparison to gauge-treated groups. Additionally, a higher blood flow and rapid re-epithelialization of wound tissue in the treatment group corroborated with the results obtained in the wound closure study. Overall, the developed dual-drug-loaded electrospun nanofiber mats have good compatibility, surface properties, and excellent wound healing potential, which can provide an extra edge in the management of complex diabetic wounds.

Abstract:Rhamnus prinoides leaf contains carbohydrates, saccharides, phenolic acids, and diterpenes with antibacterial, wound-healing, and anti-inflammatory properties. In this study, Rhamnus prinoides leaf extract was successfully incorporated into polycaprolactone-cellulose acetate (PCL-CA) nanofibers through electrospinning technique for the first time. The mats' morphology, diameter, chemical, and crystalline structure were characterized. The study investigated the mats' antibacterial activity, wound healing, cytotoxicity, drug release behaviour, hydrophilicity, and water absorbency properties. The results revealed that the mats exhibited continuous, smooth, without-beads, and interconnected structures, with average fiber diameters ranging from 385 ± 21 nm to 332 ± 74 nm. The antibacterial effeciency was remarkable against S. aureus and E. coli, achieving bacterial reduction percentages exceeding 99 % at concentrations of 3 % and above against S. aureus and 5 % and above against E. coli. Cytotoxic tests showed low-cytotoxicity up to an extract concentration of 7 %. The extract release increases with an increase in concentration. In vitro wound healing assay, the mats enhanced cell migration to the wound area. Additionally, the incorporation of Rhamnus prinoides significantly improved the hydrophilicity and water absorbency of the nanofibers. Overall, the study highlights the mats' broad antimicrobial and wound healing properties with less cytotoxicity, hydrophilicity, and water absorbency, making them promising for use as wound dressings.

Abstract:This study presents the development of tunable scaffolds with bimodal porosity comprising poly(ε-caprolactone) (PCL) micro-meshes and PCL/gelatin/ε-polylysine (ε-PL) fibrous layers. Pure PCL scaffolds were prepared using the fused deposition modeling technique featuring grid geometry and interconnected micro-pores, followed by electrospinning to produce PCL/gelatin/ε-PL nanofibrous layers. Field emission scanning electron microscopy was employed to investigate the morphological features of the scaffolds, while the physicomechanical properties were studied using tensile and contact angle tests. Antibacterial performance and skin cell toxicity of the scaffolds were determined by bacterial disc diffusion and viability assays, respectively. Morphological analysis showed the presence of micro-to nano-sized pores in the developed scaffolds. The mechanical test results revealed that the prepared scaffolds exhibited Young's modulus values similar to the human skin with higher strain. The nanocomposite scaffolds were cytocompatible and effectively eradicated common bacteria associated with cutaneous wounds. In light of the aforementioned results along with facile fabrication, the tunable PCL/gelatin/ε-PL porous scaffolds hold great promise for applications in skin wound repair.

Abstract:The current research work focuses on preparing the polycaprolactone (PCL) based nanocomposite films embedded with surface modified Halloysite Nanotube (HNT). The avenue of the study is to unravel the applicability of polymer nanocomposites for wound healing. The flexible property of HNT was taken as the major force to accomplish the addition of biopolymer pectin onto its surface. Functionalization of HNT with pectin has certainly enhanced its binding nature with the polymer. The PCL nanocomposite films were characterized by several promising techniques such as FTIR, XRD, DSC-TGA, FESEM, TEM, AFM, and mechanical properties were too examined along. When compared to the plane PCL film, the nanocomposite films manifested favorable results in terms of mechanical and chemical properties. Additionally, biometric studies such as in-vitro swelling, enzymatic degradation, and hemolysis performed on the films gave extremely good results. The haemolytic percentage recorded for the films exhibited a steady decrease with increasing amount of nanofillers. The MTT assay showed cell proliferation and its increase as the embedded HNT is more in the matrix. Wound closure study performed on NIH3T3 cell line with 1, 3 and 5wt% of films has given a strong proof for the involvement of polymer and HNT in the healing procedure.

Abstract:Bipolar disorder is a lifelong illness with a course that is usually chronic or recurrent. Severity of complications is generally proportionate to the number of episodes, especially depression. In addition to potentially preventing episodes, effective treatment reduces mortality. This article reviews long-term treatment strategies for bipolar disorder, focusing on depressive episodes, and discusses treatment studies, including problems in design. Treatment effectiveness, including reduction of suicide risk, is enhanced if patients and physicians collaboratively recognize and treat prodromal symptoms, preventing the emergence of episodes. Strategies for treatment differ as one progresses from obtaining syndromal recovery in the acute episode, to functional recovery during continuation treatment, to stability during maintenance treatment. Successful long-term treatment of bipolar disorder requires integrated pharmacologic and nonpharmacologic treatments combined with a therapeutic alliance that facilitates a proactive, preventive approach to the illness.

Abstract:Chronic wounds often result in multiple infections with various kinds of bacteria and uncontrolled wound exudate, resulting in several healthcare issues. Advanced medicated nanofibres prepared by electrospinning have gained much attention for their topical application on infected chronic wounds. The objective of this work is to enhance the critical variables of ciprofloxacin-loaded polycaprolactone-silk sericin (PCL/SS-PVA-CIP) nanofibre production via the process of electrospinning. To examine the antibacterial effectiveness of PCL/SS-PVA-CIP nanocomposites, the material was tested against P. aeruginosa and S. aureus. The combination of PCL/SS-PVA-CIP exhibited potent inhibitory properties, with the most effective concentrations of ciprofloxacin (CIP) being 3 μg/g and 7.0 μg/g for each bacterium, respectively. The biocompatibility was evaluated by conducting cell reduction and proliferation studies using the human epidermal keratinocyte (HaCaT) cells and human gingival fibroblasts (HGFs) in vitro cell lines. The PCL/SS-PVA-CIP showed good cell compatibility with HaCaT and HGF cells, with effective proliferation even at antibiotic doses of up to 7.0 μg/g. The drug release effectiveness of the nanocomposites was assessed at various concentrations of CIP, resulting in a maximum cumulative release of 76.5% and 74.4% after 72 h for CIP concentrations of 3 μg/g and 7 μg/g, respectively. In summary, our study emphasizes the possibility of combining silk sericin (SS) and polycaprolactone (PCL) loading with CIP nanocomposite for wound management.

Abstract:A 23-year-old man presented for cosmetic consultation for symmetrically distributed, red-to-purple, hyperkeratotic papules that had been present since early childhood. Histopathologic features included ectasia of upper dermal vessels with overlying hyperkeratosis. Serum alpha-galactosidase A level was diminished. Fabry disease is an x-linked recessive disorder in which deficiency of the lysosomal enzyme alpha-galactosidase A leads to progressive accumulation of globotriaosylceramide in vital organs. The complexity and rarity of this disease mandates a multidisciplinary approach that includes initiation of enzyme replacement therapy.
