Abstract:Osteocytes are thought to be the primary mechanosensory cells within bone, regulating both osteoclasts and osteoblasts to control load induced changes in bone resorption and formation. Osteocytes initiate intracellular responses including activating the Wnt/β-catenin signaling pathway after experiencing mechanical forces. In response to changing mechanical loads (strain) the osteocytes signal to cells on the bone surface. However, this process of osteocyte activation appears heterogeneous since it occurs in sub-populations of osteocytes, even within regions predicted to be experiencing similar global strain magnitudes determined based on traditional finite element modeling approaches. Several studies have investigated the strain responses of osteocyte lacunae using finite element (FE) models, but many were limited by the use of idealized geometries (e.g., ellipsoids) and analysis of a single osteocyte. Finite element models by other groups included more details, such as canaliculi, but all were done on models consisting of a single osteocyte. We hypothesized that variation in size and orientation of the osteocyte lacunae within bone would give rise to micro heterogeneity in the strain fields that could better explain the observed patterns of osteocyte activation following load. The osteocytes in our microscale and nanoscale models have an idealized oval shape and some are based on confocal scans. However, all the FE models in this preliminary study consist of multiple osteocytes. The number of osteocytes in the 3D confocal scan models ranged from five to seventeen. In this study, a multi-scale computational approach was used to first create an osteocyte FE model at the microscale level to examine both the theoretical lacunar and perilacunar strain responses based on two parameters: 1) lacunar orientation and 2) lacunar size. A parametric analysis was performed by steadily increasing the perilacunar modulus (5, 10, 15, and 20 GPa). Secondly, a nanoscale FE model was built using known osteocyte dimensions to determine the predicted strains in the perilacunar matrix, fluid space, and cell body regions. Finally, 3-D lacunar models were created using confocal image stacks from mouse femurs to determine the theoretical strain in the lacunae represented by realistic geometries. Overall, lacunar strains decreased by 14% in the cell body, 15% in the fluid space region and 25% in the perilacunar space as the perilacunar modulus increased, indicating a stress shielding effect. Lacunar strains were lower for the osteocytes aligned along the loading axis compared to those aligned perpendicular to axis. Increases in lacuna size also led to increased lacunar strains. These finite element model findings suggest that orientation and lacunar size may contribute to the heterogeneous initial pattern of osteocyte strain response observed in bone following in vivo applied mechanical loads. A better understanding of how mechanical stimuli directly affect the lacunae and perilacunar tissue strains may ultimately lead to a better understanding of the process of osteocyte activation in response to mechanical loading.
Evaluating the influence on osteocyte mechanobiology within the lacunar-canalicular system for varying lacunar equancy and perilacunar elasticity: A multiscale fluid-structure interaction analysis

Abstract:The lacunar morphology and perilacunar tissue properties of osteocytes in bone can vary under different physiological and pathological conditions. How these alterations collectively change the overall micromechanics of osteocytes in the lacunar-canalicular system (LCS) of an osteon still requires special focus. Therefore, a Haversian canal and LCS-based osteon model was established to evaluate the changes in the hydrodynamic environment around osteocytes under physiological loading using fluid-structure interaction analysis, followed by a sub-modelled finite element analysis to assess the mechanical responses of osteocytes and their components. Osteocytes were modelled with detailed configurations, including cytoplasm, nucleus, and cytoskeleton, and parametric variations in lacunar equancy (L.Eq) and perilacunar elasticity (Pl.E) were considered within the osteon model. The study aimed to conduct a comparative study among osteon models with varying L. Eq and Pl. E to check the resulting differences in osteocyte mechanobiology. The results demonstrated that the average mechanical stimulation of each subcellular component of osteocytes increased with decreases in L. Eq and Pl. E, reflecting conditions typically seen in young, healthy bone as per previous literature. However, hydrodynamic responses, such as fluid flow and fluid shear stress on osteocytes, varied proportionally with the elasticity difference between the bone matrix and the perilacunar region during Pl. E variation. Additionally, the findings revealed that a minimal percentage of energy was used to transmit mechanical responses through microtubules from the cell membrane to the nucleus, and this energy percentage increased with higher L. Eq. The outcomes of the study could help to quantify how the osteocyte microenvironment and its mechanosensitivity within cortical bone changes with L. Eq and Pl. E alterations in different bone conditions, from young to aged and healthy to diseased.
Interactive effects of various loading parameters on the fluid dynamics within the lacunar-canalicular system for a single osteocyte

Abstract:The osteocyte lacunar-canalicular system (LCS) serves as a mechanotransductive core where external loading applied to the skeleton is transduced into mechanical signals (e.g., fluid shear) that can be sensed by mechanosensors (osteocytes). The fluid velocity and shear stress within the LCS are affected by various loading parameters. However, the interactive effect of distinct loading parameters on the velocity and shear stress in the LCS remains unclear. To address this issue, we developed a multiscale modeling approach, combining a poroelastic finite element (FE) model with a single osteocytic LCS unit model to calculate the flow velocity and shear stress within the LCS. Next, a sensitivity analysis was performed to investigate individual and interactive effects of strain magnitude, strain rate, number of cycles, and intervening short rests between loading cycles on the velocity and shear stress around the osteocyte. Lastly, we developed a relatively simple regression model to predict those outcomes. Our results demonstrated that the strain magnitude or rate alone were the main factors affecting the velocity and shear stress; however, the combination of these two was not directly additive, and addition of a short rest between cycles could enhance the combination of these two related factors. These results show highly interactive effects of distinct loading parameters on fluid velocity and shear stress in the LCS. Specifically, our results suggest that an enhanced fluid dynamics environment in the LCS can be achieved with a brief number of load cycles combined with short rest insertion and high strain magnitude and rate.

Finite Element Models of Osteocytes and Their Load-Induced Activation

Abstract:Purpose of review: Osteocytes are the conductors of bone adaptation and remodelling. Buried inside the calcified matrix, they sense mechanical cues and signal osteoclasts in case of low activity, and osteoblasts when stresses are high. How do osteocytes detect mechanical stress? What physical signal do they perceive? Finite element analysis is a useful tool to address these questions as it allows calculating stresses, strains and fluid flow where they cannot be measured. The purpose of this review is to evaluate the capabilities and challenges of finite element models of bone, in particular the osteocytes and load-induced activation mechanisms. Recent findings: High-resolution imaging and increased computational power allow ever more detailed modelling of osteocytes, either in isolation or embedded within the mineralised matrix. Over the years, homogeneous models of bone and osteocytes got replaced by heterogeneous and microstructural models, including, e.g. the lacuno-canalicular network and the cytoskeleton. The lacuno-canalicular network induces strain amplifications and the osteocyte protrusions seem to be stimulated much more than the cell body, both by strain and fluid flow. More realistic cell geometries, like minute constrictions of the canaliculi, increase this effect. Microstructural osteocyte models describe the transduction of external stimuli to the nucleus. Supracellular multiscale models (e.g. of a tunnelling osteon) allow to study differential loading of osteocytes and to distinguish between strain and fluid flow as the pivotal stimulatory cue. In the future, the finite element models may be enhanced by including chemical transport and intercellular communication between osteocytes, osteoclasts and osteoblasts.
Both network architecture and micro cracks effects on lacuno-canalicular liquid flow efficiency within the context of multiphysics approach for bone remodeling

Abstract:When physical forces are applied to bone, its mechanical adaptive behaviors change according to the microarchitecture configuration. This leads to changes in biological and physical thresholds in the remodeling cell population, involving sensor cells (osteocytes) interacting with each other and changes in osteocyte shape due to variation in lacunar shape. The resulting alterations in fluid flow leads to changes in the membrane electrical potential and shear stress. Eventual creation of microcracks, may lead in turn to modify cell activity. In contrast, the redundancy in the lacuno canalicular network (LCN) interconnectivity maintains partial flow. Our goal was to investigate the role of fluid flow in LCN by proposing a model of electro-mechanical energy spread through inhomogeneous microarchitectures. We focused on mechano-sensitivity to changes in load-induced flow impacted by neighboring micro cracks and quantifying its critical role in changing, velocity, shear stress and orientation of liquid mass transportation from one cell to another. To enhance the concept of intricacy LCN micro-structure to fluid flow, we provide a new combined effects factor considered as osteocytes sensor efficiency. We customized an influence function for each osteocyte, coupling: in one hand, the spatial distribution within remodeling influence areas, conducting a significant fluid spread, leading hydro-dynamic behavior and impacted further by presence of micro cracks and; in other hand, the fluid electro kinetic behavior. As an attempt to fill the limitations stated by many of the recent studies, we reveal in numerical simulation, some results which cannot be measured in vitro/in vivo studies. Numerical calculations were performed in order to evaluate, among many others, how liquid flow conditions changes between lacunas, how the orientation and the magnitude of the governing flow in LCN can regulate osteocytes efficiency. In addition to be regulated by osteocytes, a direct effects of fluid flow are also acting on osteoblast activity. In summary, this new approach considers mechano-sensitivity in relation to liquid flow dynamic and suggests additional pathway for Osseo integration via osteoblast regulation. However, this novel modeling approach may help improve the mapping and design bone scaffolds and/or selection of scaffold implantation regions.

We present three existence theorems showing that sp ecific behaviour of the distribution of Y in the neighbourhood of zero is essential. Simple consequences of these theorems give evidence to the existence of the moments for particular random variables; some of th ese results are well known from standard probability theory. However, we obtain them in a simple way.

Abstract:The objective of this study is to use the finite element (FE) method to predict the mechanical signals (interstitial fluid velocity, strain, pore pressure, and pore fluid velocity) produced by osteocyte during physiological activities. The model predicts that the amplitude and distribution of the mechanical signals are mainly affected by the loading rate. The magnitude of mechanical signals in the lacunar-canalicular system increases as the amplitude, frequency and amount of direction of load increase. Collagen hillocks can effectively amplify strain signals at the process. The established model can be used for studying the mechanism of bone mechanotransduction at the micro-level.
Blood pressure – lower targets, greater challenges

Abstract:The world health organization has estimated that high blood pressure causes one in eight deaths world wide making hypertension the third leading killer in the world (1). The seventh joint national committee (JNC-7) report was recently published in the Journal of the American Medical Association (2) and presents new guidelines for the prevention and management of high blood pressure, based on a review of all the latest scientific literature. The guidelines were developed by the National High Blood Pressure Education Program Coordinating Committee, administered by the National Heart, Lung, and Blood Institute. The report addresses areas including blood pressure measurement and classification, patient evaluation and testing, treatment approaches and monitoring, and certain special considerations (2). The new guidelines categorize blood pressure as normal (<120/80 mmHg), prehypertension (120/ 80 to 139/89), stage 1 hypertension (140/90 to 159/ 99), and stage 2 hypertension (‡160/100). The JNC7 report on hypertension takes a stronger view than its predecessors (3), reclassifying ‘high normal’ blood pressure as ‘prehypertension’ and calling for aggressive treatment, first by changing lifestyle and then with drugs. The guidelines also emphasize the importance of a systolic blood pressure of over 140 mmHg as an important cardiovascular risk factor for people over 50 years of age. Cardiovascular disease risk increases as the blood pressure rises above 115/75 mmHg, increasing 2-fold for each 20/10 mmHg increment. Lowering blood pressure from 140/90 towards the new goal level of 120/80 mmHg will decrease heart attacks (20– 25%), heart failure (50%), stroke (35–40%), kidney disease, and will save lives (2). The blood pressures at which most doctors consider treatment may be beneficial are in fact well above the level at which blood pressure starts to cause harm. JNC-7 emphasizes the importance of aging which is the single most important factor associated with the development of hypertension (4). Unless preventive steps are taken, stiffness and other damage to arteries worsen with age and make high blood pressure more and more difficult to treat. The new prehypertension category reflects this risk and will prompt people to take preventive action early. Data from the Framingham Heart Study suggests that individuals who are normotensive at 55 years of age have a 90% lifetime risk for developing hypertension. So the aim should be to slow the adverse effects of aging and delay the onset of hypertension. In practice lifestyle modification is the only way, at present, to counteract the effects of age and to try to prevent the development of hypertension. JNC-7 recommends that Governments and others should modify working conditions to reduce the excessive demands and time pressures on those living in the developed countries of the world. In addition they should also promote exercise, weight reduction, the consumption of less alcohol and the taking of less sodium in the diet. Specifically JNC-7 recommends change to the ‘DASH’ diet – high in fruits and vegetables, potassium, and calcium – (5). Most important smoking cessation should be encouraged and children and young people should be discouraged from starting to smoke. The JNC-7 guidelines include new data on US control, awareness, and treatment rates for high blood pressure. According to a national survey, 70% of Americans are aware of their high blood pressure, 59% are being treated for it, and 34% of those with hypertension have it under control (6). Those percentages represent a slight improvement over rates for 10 years ago, when 68% of Americans were aware of their high blood pressure, 54% were being treated for it, and 27% of those with hypertension had it under control (6). Analysis of Osteocyte Morphology in Terms of Sensation of In Vivo Stress Applied on Bone

Abstract:Most bones are anisotropically loaded and seem to be adapted to the anisotropic stress or strain field by changing the anisotropy in their microstructure. Osteocyte (OCY) is believed to play an important role as a mechanosensor and regulator of modeling and/or remodeling orchestrating osteoblast and osteoclast activity to make bone suitable to resist the mechanical environment. In general, osteocytes sense magnitude of stress (strain) applied upon the bone and then work as a trigger to change bone mass to adjust bone’s mechanical function to the stress field. This structural optimization is an important aspect of the bone functional adaptation; another inevitable optimization might be achieved through the change in intrinsic material anisotropy including the preferential c -axis orientation of biological apatite (BAp) crystal. To achieve this adaptation through material anisotropy, osteocyte needs to be a mechanosensor which can detect anisotropic stress field. In the present study, osteocyte lacunae and canaliculi in the mid-diaphysis and the distal part of the rat femur were stained by a fluorescein dye for visualization and analysis. The mid-diaphysis shows greater degree of the preferential c -axis orientation of BAp crystal than the distal part in relation to the magnitude of uni-axial stress field. It was found that the osteocytes in long bone preferentially align along the bone long axis and the degree of alignment is greater in the mid-diaphysis than in the distal region, which seems to be effective for the sensation of the site-dependent specific stress field applied on the long bone.
Computational modeling based on confocal imaging predicts changes in osteocyte and dendrite shear stress due to canalicular loss with aging

Abstract:Exercise and physical activity exert mechanical loading on the bones which induces bone formation. However, the relationship between the osteocyte lacunar-canalicular morphology and mechanical stress experienced locally by osteocytes transducing signals for bone formation is not fully understood. In this study, we used computational modeling to predict the effect of canalicular density, the number of fluid inlets, and load direction on fluid flow shear stress (FFSS) and bone strains and how these might change following the microstructural deterioration of the lacunar-canalicular network that occurs with aging. Four distinct computational models were initially generated of osteocytes with either ten or eighteen dendrites using a fluid-structure interaction method with idealized geometries. Next, a young and a simulated aged osteocyte were developed from confocal images after FITC staining of the femur of a 4-month-old C57BL/6 mouse to estimate FFSS using a computational fluid dynamics approach. The models predicted higher fluid velocities in the canaliculi versus the lacunae. Comparison of idealized models with five versus one fluid inlet indicated that with four more inlets, one-half of the dendrites experienced FFSS greater than 0.8 Pa, which has been associated with osteogenic responses. Confocal image-based models of real osteocytes indicated a six times higher ratio of canalicular to lacunar surface area in the young osteocyte model than the simulated aged model and the average FFSS in the young model (FFSS = 0.46 Pa) was three times greater than the aged model (FFSS = 0.15 Pa). Interestingly, the surface area with FFSS values above 0.8 Pa was 23 times greater in the young versus the simulated aged model. These findings may explain the impaired mechano-responsiveness of osteocytes with aging.

Abstract:Purpose of ReviewConnecting organ-scale loads to cellular signals in their local in vivo environment is a current challenge in the field of bone (re)modelling. Understanding this critical missing link would greatly improve our ability to anticipate mechanotransduction during different modes of stimuli and the resultant cellular responses. This review characterises computational approaches that could enable coupling links across the multiple scales of bone.Recent FindingsCurrent approaches using strain and fluid shear stress concepts have begun to link organ-scale loads to cellular signals; however, these approaches fail to capture localised micro-structural heterogeneities. Furthermore, models that incorporate downstream communication from osteocytes to osteoclasts, bone-lining cells and osteoblasts, will help improve the understanding of (re)modelling activities. Incorporating this potentially key information in the local in vivo environment will aid in developing multiscale models of mechanotransduction that can predict or help describe resultant biological events related to bone (re)modelling.SummaryProgress towards multiscale determination of the cell mechanical environment from organ-scale loads remains elusive. Abstract:Abstract Osteocytes, the most abundant and long-lived cells in bone, are the master regulators of bone remodeling. In addition to their functions in endocrine regulation and calcium and phosphate metabolism, osteocytes are the major responsive cells in force adaptation due to mechanical stimulation. Mechanically induced bone formation and adaptation, disuse-induced bone loss and skeletal fragility are mediated by osteocytes, which sense local mechanical cues and respond to these cues in both direct and indirect ways. The mechanotransduction process in osteocytes is a complex but exquisite regulatory process between cells and their environment, between neighboring cells, and between different functional mechanosensors in individual cells. Over the past two decades, great efforts have focused on finding various mechanosensors in osteocytes that transmit extracellular mechanical signals into osteocytes and regulate responsive gene expression. The osteocyte cytoskeleton, dendritic processes, Integrin-based focal adhesions, connexin-based intercellular junctions, primary cilium, ion channels, and extracellular matrix are the major mechanosensors in osteocytes reported so far with evidence from both in vitro and in vitro studies. This review aims to give a systematic introduction to osteocyte mechanobiology, provide details of osteocyte mechanosensors, and discuss the roles of osteocyte mechanosensitive signaling pathways in the regulation of bone homeostasis.

A finite element model of an osteoblast to quantify the transduction of exogenous forces to cellular components

Abstract:Encouraged by recent advances of biophysical and biochemical assays we introduce a 3D finite element model of an osteoblast, seeking an analogue between exogenous forces and intracellularly activated sensory mechanisms. The cell was reverse engineered and the dimensions of the internal cellular structures were based on literature data. The model was verified and validated against atomic force microscopy experiments and four loading scenarios were considered. The stress distributions developing on the main cellular components were calculated along with their corresponding strain values. The nucleus and mitochondria exhibited similar loading trends, with the mitochondria being stressed by an order of magnitude higher than the nucleus (e.g. 1.4 vs. 0.16 MPa). Equivalent stiffness was determined to increase by almost 50%, from the apex to the cell's periphery, as was the cell's elasticity, which was lowest when the load was exerted directly above the nucleus. The assessment of how extrinsic loads are propagated to a cell's internal structures is inherently a problem of high complexity. The findings presented in this study can provide important insight into biophysical and biochemical responses elicited in cells through mechanical stimulus. This was evident in both the nuclear and mitochondrial loading and would stipulate the important contribution of even more accurate models in the interpretation of cellular events. One Sentence Summary: The results of this numerical biomechanical study demonstrated that even minor extrinsic loads irrespective of the application site, are transduced by a fraction of the cytoskeleton to its internal structure (primarily to its mitochondria and secondary to the cell's nucleus), indicating mechanical stimulus as the dominant pathway to cell expression.

Abstract:Brittle bone diseases are a global healthcare problem for orthopaedic clinicians, that reduces bone strength and promotes bone fracture risk. In vivo studies reported that loading-induced fluid flow through the lacunar–canalicular channel (LCS) of bone tissue inhibit such bone loss and encourages osteogenesis i.e. new bone formation. Canalicular fluid flow converts mechanical signals into biological signals and regulates bone reconstruction by releasing signalling molecules responsible for mechanotransduction. In-silico model mostly considers canalicular fluid is Newtonian, however, physiological canalicular fluid may be non-Newtonian in nature as it contains nutrients and supplements. Accordingly, this study attempts to develop a two-dimensional in-silico model to compute loading-induced non-Newtonian canalicular fluid flow in a complex LCS of bone tissue. Moreover, canalicular fluid is considered as a Jeffery fluid, that can easily be reduced to Newtonian fluid as a special case. The results show that physiological loading modulates the canalicular fluid flow, wall shear stress (WSS) and streamline in bone LCS. Fluid velocity and WSS increases with increase in non-dimensional frequency and non-Newtonian parameter (Jeffery fluid parameters) and reduce with change in permeability. The outcomes of this study may provide new insights in the role of mechanical loading-induced non-Newtonian canalicular fluid flow dynamics in bone LCS. The key findings of this study can be used to improve the understanding of osteocyte mechanobiology involved inside the bone tissue.

Abstract:Osteocytes perceive and process mechanical stimuli in the lacuno-canalicular network in bone. As a result, they secrete signaling molecules that mediate bone formation and resorption. To date, few three-dimensional (3D) models exist to study the response of mature osteocytes to biophysical stimuli that mimic fluid shear stress and substrate strain in a mineralized, biomimetic bone-like environment. Here we established a biomimetic 3D bone model by utilizing a state-of-art perfusion bioreactor platform where immortomouse/Dmp1-GFP-derived osteoblastic IDG-SW3 cells were differentiated into mature osteocytes. We evaluated proliferation and differentiation properties of the cells on 3D microporous scaffolds of decellularized bone (dBone), poly(L-lactide-co-trimethylene carbonate) lactide (LTMC), and beta-tricalcium phosphate (β-TCP) under physiological fluid flow conditions over 21 days. Osteocyte viability and proliferation were similar on the scaffolds with equal distribution of IDG-SW3 cells on dBone and LTMC scaffolds. After seven days, the differentiation marker alkaline phosphatase (Alpl), dentin matrix acidic phosphoprotein 1 (Dmp1), and sclerostin (Sost) were significantly upregulated in IDG-SW3 cells (p = 0.05) on LTMC scaffolds under fluid flow conditions at 1.7 ml/min, indicating rapid and efficient maturation into osteocytes. Osteocytes responded by inducing the mechanoresponsive genes FBJ osteosarcoma oncogene (Fos) and prostaglandin-endoperoxide synthase 2 (Ptgs2) under perfusion and dynamic compressive loading at 1 Hz with 5 % strain. Together, we successfully created a 3D biomimetic platform as a robust tool to evaluate osteocyte differentiation and mechanobiology in vitro while recapitulating in vivo mechanical cues such as fluid flow within the lacuno-canalicular network. STATEMENT OF SIGNIFICANCE: This study highlights the importance of creating a three-dimensional (3D) in vitro model to study osteocyte differentiation and mechanobiology, as cellular functions are limited in two-dimensional (2D) models lacking in vivo tissue organization. By using a perfusion bioreactor platform, physiological conditions of fluid flow and compressive loading were mimicked to which osteocytes are exposed in vivo. Microporous poly(L-lactide-co-trimethylene carbonate) lactide (LTMC) scaffolds in 3D are identified as a valuable tool to create a favorable environment for osteocyte differentiation and to enable mechanical stimulation of osteocytes by perfusion and compressive loading. The LTMC platform imitates the mechanical bone environment of osteocytes, allowing the analysis of the interaction with other cell types in bone under in vivo biophysical stimuli.

Abstract:Background and Objective Detailed finite element models based on medical images ( μ -CT) are commonly used to analyze the mechanical behavior of bone at microscale. In order to simulate the tissue failure onset, isotropic failure criteria of lamellar tissue are often used, despite its non-isotropic and heterogeneous nature. The main goal of the present work is to estimate the in-plane ultimate stress of lamellar bone, considering the influence of mineral content and the porosity due to the osteocyte lacunae concentration. Methods To this aim, a representative volume cell of lamellar tissue is modeled numerically, including: (1) non-isotropic elastic properties of tissue as a function of the bone mineral density and (2) explicit modeling of the osteocyte lacunae, considering the range of porosity content, size and orientation of ellipsoid-shaped lacunae. Firstly, the element size for the finite element models have been defined from a preliminary convergence analysis. Bounds on the ultimate stress of non-porous lamellar tissue are estimated for two values of bone mineral density, considering the results of tensile and compressive tests of wet osteons from the literature. Subsequently, the ultimate stress of lamellar tissue considering several values of micro-porosity is addressed. Results Results obtained in this work show that the strength of lamellar bone decreases exponentially with the increase of lacunae porosity concentration. Ultimate stress of non-porous tissue ( p=0% ) increases with high mineral content, reaching a value of S ̄transc=355.40±39.80 MPa for compression in the transversal direction of the fiber bundles, being BMD=1.246g/cm3 . The mean value for the longitudinal to transverse strength ratio evaluated for porosity p=0%,1% and 5% and a mineral content BMD=1.2g/cm3 , is 2.47:1 for tension and 1.55:1 for compression. These values are in agreement with literature. Conclusions Osteocyte lacunae act as stress concentrators, acting as potential stimulus for the bone regeneration process. A novel micromechanical model for the in-plane ultimate stress of lamellar tissue as a function of mineral content and lacunae concentration is presented. Additional considerations about the intralamelar shear stress evolution are also given.

Abstract:<h3 class="u-h4 u-margin-m-top u-margin-xs-bottom">Background</h3><p>Osteocytes, composing over 90% of bone cells, are well known for their mechanosensing abilities. Aged osteocytes with impaired morphology and function are less efficient in mechanotransduction which will disrupt bone turnover leading to osteoporosis. The aim of this systematic review is to delineate the mechanotransduction mechanism at different stages in order to explore potential target for therapeutic drugs.</p><h3 class="u-h4 u-margin-m-top u-margin-xs-bottom">Methods</h3><p>A systematic literature search was performed in PubMed and Web of Science. Original animal, cell and clinical studies with available English full-text were included. Information was extracted from the included studies for review.</p><h3 class="u-h4 u-margin-m-top u-margin-xs-bottom">Results</h3><p>The 26 studies included in this review provided evidence that mechanical loading are sensed by osteocytes via various sensing proteins and transduced to different signaling molecules which later initiate various biochemical responses. Studies have shown that osteocyte plasma membrane and cytoskeletons are emerging key players in initiating mechanotransduction. Bone regulating genes expressions are altered in response to load sensed by osteocytes, but the genes involved different signaling pathways and the spatiotemporal expression pattern had made mechanotransduction mechanism complicated. Most of the included studies described the important role of osteocytes in pathways that regulate mechanosensing and bone remodeling.</p><h3 class="u-h4 u-margin-m-top u-margin-xs-bottom">Conclusions</h3><p>This systematic review provides an up-to-date insight to different steps of mechanotransduction. A better understanding of the mechanotransduction mechanism is beneficial in search of new potential treatment for osteoporotic patients. By delineating the unique morphology of osteocytes and their interconnected signaling network new targets can be discovered for drug development.</p><h3 class="u-h4 u-margin-m-top u-margin-xs-bottom">Translational potential of this article</h3><p>This systematic review provides an up-to-date sequential overview and highlights the different osteocyte-related pathways and signaling molecules during mechanotransduction. This allows a better understanding of mechanotransduction for future development of new therapeutic interventions to treat patients with impaired mechanosensitivity.</p>

Abstract:Osteocytes and their cell processes reside in a large, interconnected network of voids pervading the mineralized bone matrix of most vertebrates. This osteocyte lacuno-canalicular network (OLCN) is believed to play important roles in mechanosensing, mineral homeostasis, and for the mechanical properties of bone. While the extracellular matrix structure of bone is extensively studied on ultrastructural and macroscopic scales, there is a lack of quantitative knowledge on how the cellular network is organized. Using a recently introduced imaging and quantification approach, we analyze the OLCN in different bone types from mouse and sheep that exhibit different degrees of structural organization not only of the cell network but also of the fibrous matrix deposited by the cells. We define a number of robust, quantitative measures that are derived from the theory of complex networks. These measures enable us to gain insights into how efficient the network is organized with regard to intercellular transport and communication. Our analysis shows that the cell network in regularly organized, slow-growing bone tissue from sheep is less connected, but more efficiently organized compared to irregular and fast-growing bone tissue from mice. On the level of statistical topological properties (edges per node, edge length and degree distribution), both network types are indistinguishable, highlighting that despite pronounced differences at the tissue level, the topological architecture of the osteocyte canalicular network at the subcellular level may be independent of species and bone type. Our results suggest a universal mechanism underlying the self-organization of individual cells into a large, interconnected network during bone formation and mineralization.
