Abstract:Eutrophication, mainly caused by the oversupply of nitrate (NO3−) and phosphate, has become a serious environmental problem in Xiangshan Bay (east China), but the sources of the nitrate are poorly understood. In this study, the estuarine distributions of dual stable isotopes (δ15N and δ18O) of NO3− measured in May 2012 were used in combination with concurrent measurements of salinity, pH, dissolved oxygen, and nutrients (nitrate, nitrite, ammonium, and phosphate) to investigate NO3− sources and the processes that control NO3− distributions within the bay. In the lower bay, high concentrations of NO3− (up to 60.6 μmol L−1), an N/P ratio of 52, and low values of nitrate δ15N (δ15NNO3 = 5.7‰–6.7‰) and δ18O (δ18ONO3 = 1.8‰–3.5‰) indicate that NO3− in this area was likely from the Changjiang and Qiantangjiang rivers as well as local nitrification-associated inputs. In the upper bay, constituent concentrations and high values of δ15NNO3 (up to 8.8‰) and δ18ONO3 (up to 6.8‰) suggest that NO3− distributions here were controlled mainly by the mixing of coastal and fresh waters, phytoplankton assimilation, and inputs of riverine NO3−, possibly derived from manure and sewage. Spatial variations of Δ(15,18), the nitrate δ15N-to-δ18O anomaly, is possibly resulted from the different water residence times across the bay, which could in turn lead to different degrees of nitrification or variable amounts of NO3− from atmospheric deposition.

Abstract:Eutrophication caused by inputs of excess nitrogen（N） has become a serious environmental problem in Hangzhou Bay（China）,but the sources of this nitrogen are not well understood.In this study,the August 2019 distributions of salinity,nutrients [nitrate（NO 3 - ）,nitrite,ammonium,and phosphate],and the stable isotopic composition of NO 3 - (δ 15 N and δ 18 O) were used to investigate sources of dissolved inorganic nitrogen（DIN） to Hangzhou B ay.Spatial distributions of nitrate,salinity,and nitrate δ 18 O indicate that the Qiantang River,the Changjiang River,and nearshore coastal waters may all contribute nitrate to the bay.Based on the isotopic compositions of nitrate in these potential source waters and conservative mixing of nitrate in our study area,we suggest that the NO 3 - in Hangzhou B ay was likely derived mainly from soils,synthetic N fertilizer,and manure and sewage.End-member modeling indicates that in the upper half of the bay,the Qiantang River was a very important DIN source,possibly contributing more than 50% of DIN in the bay head area.In the lower half of the bay,DIN was sourced mainly from strongly intruding coastal water.DIN coming directly from the Changjiang River made a relatively small contribution to Hangzhou Bay DIN in August 2019.

Abstract:The spatial variations in nitrate(NO-3),nitrite(NO-2) and ammonium(NH+4) concentrations in the Changjiang Estuary and the adjacent East China Sea were described on the basis of the summer cruise in 2006.NO-3 concentrations decreased along a salinity gradient.Owing to the input of the sewage from Shanghai City NO-2 and NH-4 concentrations were very high(up to 8.64,19.81 μmol/dm3 respectively) in the Changjiang Estuaxy area and decreased seaward.Generally NO-3 and NH+4 concentrations were significantly correlated with salinity,with the coefficient(r2) of 0.815 and 0.255 respectively,suggesting that these two nutrients behaved nearly conservatively,while NO-2 did not behave conservatively(r2=0.074).Theoretical dilution line(TDL) was obtained by assuming river and ocean end-members.NO-3,NO-2 and NH+4 concentrations were above values expected for a conservative mixing relationship with salinity at the freshwater end member due to high nitrogen input from the sewage effluence,and in deep water at the oceanic stations because of the decomposition of the organic matter.In contrast,at the salinity plume with high dissolved oxygen(DO) concentrations,the surface NO-3 concentrations were below values expected for the conservative mixing by 1~19 mol/dm3,as a result of high primary production in the euphotic zone,where NO-2 and NH+4 concentrations were also negatively deviated to various extents.In waters with the highest turbidity in the Changjiang Estuary,NH+4 was removed due to the adsorption of suspended matter.Overall,NO-3,NO-2 and NH+4 concentrations at the surface at the oceanic stations deviated from the TDL to less extent,while their concentrations at the bottom were generally beyond values expected for the TDL because the organic matter from the photic zone decomposed and nutrients regenerated.

Abstract:Eutrophication in coastal waters has been increasing remarkably, severely impacting the water quality in mariculture bays. In this study, we conducted multiple isotopic measurements on suspended particulate nitrogen (δ 15 N-PN) and dissolved nitrate (δ 15 N-NO − 3 and δ 18 O-NO − 3 ) in Zhanjiang Bay, a typical mariculture bay with a high level of eutrophication in South China, to investigate the changes in nitrogen sources and their cycling between the rainy and dry seasons. During the rainy season, the study found no significant relation between δ 15 N-PN and δ 15 N-NO − 3 due to the impact of heavy rainfall and terrestrial erosion. In the upper bay, a slight nitrate loss and slightly higher δ 15 N-NO − 3 and δ 18 O-NO − 3 values were observed, attributed to intense physical sediment-water interactions. Despite some fluctuations, nitrate concentrations in the lower bay mainly aligned with the theoretical mixing line during the rainy season, suggesting that nitrate was primarily influenced by terrestrial erosion and that nitrate isotopes resembled the source. Consequently, the isotopic values of nitrate can be used for source apportionment in the rainy season. The results indicated that soil nitrogen (36%) and manure and sewage (33%) were the predominant nitrogen sources contributing to nitrogen loads during this period. In contrast, the dry season saw a deficient ammonium concentration (<0.2 μmol/L) in the bay, due to nearly complete consumption by phytoplankton during the red tide period. Additionally, the significant loss of nitrate and simultaneous increase in the stable isotopes of dissolved and particulate nitrogen suggest a strong coupling of assimilation and mineralization during the dry season. More active biogeochemical processes during the dry season may be related to decreased runoff and increased water retention time. Overall, our study illustrated the major seasonal nitrogen sources and their dynamics in Zhanjiang Bay, providing valuable insights for formulating effective policies to mitigate eutrophication in mariculture bays.

Abstract:Isotope measurements were performed on dissolved nitrate (NO3−) and ammonium (NH4+) in the coastal waters of the northeastern Beibu Gulf, China, to investigate the seasonal nitrate sources and their biogeochemical processes, which are due to the rapid development of local industrialisation and urbanisation. The high N/P ratio observed in the coastal bay during both fall and spring suggests that P is a limiting nutrient, which in turn indicates that increasing P causes conditions favourable for algal blooms. Higher nutrient concentrations and δ15N-NO3− and δ15N-NH4+ values were found in the nearshore area in the fall, suggesting that nutrients originated mainly from land-based pollution. A Bayesian isotope mixing model was used to calculate the contribution of potential NO3− sources and the results showed that in the nearshore area, NO3− originated mainly from manure and sewage (58%). In the spring, however, in addition to the impact of urban sewage effluents, the exchange of sediment and water was another important factor causing higher nutrient concentrations and positive NO3− isotopes in the nearshore area. There were lower concentrations of nutrients and an increase in δ15N-NO3− and δ15N-NH4+ values in the offshore area in the fall, and the NO3− loss in the surface water was mainly caused by the process of assimilation. However, the exchange of sediment and water was the dominant factor causing higher nutrient concentrations (except for NO3−) and positive dual nitrate isotopes but lower NO3− concentration in the offshore area during the spring. Overall, isotope analysis of NO3− and NH4+ helps to illustrate the major sources of the former and their biological transformation in the northeastern Beibu Gulf.

Nitrate Source Apportionment in a Subtropical Watershed Using Bayesian Model

Abstract:Nitrate (NO3-) pollution in aquatic system is a worldwide problem. The temporal distribution pattern and sources of nitrate are of great concern for water quality. The nitrogen (N) cycling processes in a subtropical watershed located in Changxing County, Zhejiang Province, China were greatly influenced by the temporal variations of precipitation and temperature during the study period (September 2011 to July 2012). The highest NO3- concentration in water was in May (wet season, mean±SD=17.45±9.50 mg L(-1)) and the lowest concentration occurred in December (dry season, mean±SD=10.54±6.28 mg L(-1)). Nevertheless, no water sample in the study area exceeds the WHO drinking water limit of 50 mg L(-1) NO3-. Four sources of NO3(-) (atmospheric deposition, AD; soil N, SN; synthetic fertilizer, SF; manure & sewage, M&S) were identified using both hydrochemical characteristics [Cl-, NO3-, HCO3-, SO42-, Ca2+, K+, Mg2+, Na+, dissolved oxygen (DO)] and dual isotope approach (δ15N-NO3- and δ(18)O-NO3-). Both chemical and isotopic characteristics indicated that denitrification was not the main N cycling process in the study area. Using a Bayesian model (stable isotope analysis in R, SIAR), the contribution of each source was apportioned. Source apportionment results showed that source contributions differed significantly between the dry and wet season, AD and M&S contributed more in December than in May. In contrast, SN and SF contributed more NO3- to water in May than that in December. M&S and SF were the major contributors in December and May, respectively. Moreover, the shortcomings and uncertainties of SIAR were discussed to provide implications for future works. With the assessment of temporal variation and sources of NO3-, better agricultural management practices and sewage disposal programs can be implemented to sustain water quality in subtropical watersheds.

Abstract:Coupled nitrogen and oxygen isotopes of nitrate have proven useful in identifying nitrate sources and transformation in rivers. However, isotopic fractionation and low-resolution monitoring limit the accurate estimation of nitrate dynamics. In the present study, the spatio-temporal variations of nitrate isotopes (N-15 and O-18) and hydrochemical compositions (NO3- and Cl-) of river water were examined to understand nitrate sources in the Xijiang River, China. High-frequency sampling campaigns and isotopic analysis were performed at the mouth of the Xijiang River to capture temporal nitrate variabilities. The overall values of delta N-15-NO3- and delta O-18-NO3- ranged from +4.4% to +14.1% and from -0.3% to +6.8%, respectively. The results of nitrate isotopes indicated that NO3- mainly originated from soil organic nitrogen (SON), chemical fertilizer (CF), and manure and sewage wastes (M&S). The negative correlation of nitrate isotopic valueswith NO3-/Cl- ratios suggested the importance of denitrification in NO3- loss. The results of Bayesianmodel with incorporation of isotopic fractionation during the denitrification showed that SON and CF contributed to the most (72-73%) nitrate in the wet season; whereas approximately 58% of nitrate was derived from anthropogenic inputs (M&S and CF) in the dry season. The nitrate flux was 2.08 x 10(5) tons N yr(-1) during one hydrologic year between 2013 and 2014, with 86% occurring in the wet season. Long-term fluctuations in nitrate flux indicated that nitrate export increased significantly over the past 35 years, and was significantly correlated with nitrate concentrations. The seasonal pattern of nitrate dynamics indicated the mixing of nitrified NO3- and denitrified NO3- between surface flow and groundwater flow under different hydrological conditions. Overall, the present study quantitatively evaluates the spatio-temporal variations in nitrate sources in a subtropical watershed, and the high-frequency monitoring gives a better estimate of nitrate exports and proportional contributions of nitrate sources. Abstract:Elevated nitrate (NO 3 − ) loadings in groundwater may cause health effects in drinking water and nutrient enrichment of aquatic ecosystems. To reveal the sources and seasonal variations of NO 3 − in the coastal groundwater of Beihai, southern China, we carried out hydrochemical and isotopic (δ 15 N-δ 18 O in NO 3 − ) investigations in the summer and winter, respectively, concerning multiple-aquifer groundwater, rainwater, seawater, and surface water. The sources of the main elements present in the waters were interpreted by ionic ratios. NO 3 − sources were identified by combined use of the δ 15 N values and δ 18 O values or NO 3 − /Na + molar ratios, with estimations of the proportional contribution by the Bayesian stable isotope mixing model. Denitrification was interpreted along the flow paths. The results show groundwater main elements are originated primarily from silicate weathering, and secondarily from anthropogenic inputs and carbonate dissolution. Its qualities are largely affected by seawater intrusion along the coastline. Because of difference in the predominant minerals within the aquifers and in scale and extent of seawater intrusion, the groundwater displays distinct ionic ratio characters. NO 3 − concentrations are up to 33.9 mg/L, with higher loadings in the plains relative to along the coastline. Soil N, domestic sewage, rainwater, chemical fertilizers, and algae are NO 3 − sources, with average proportional contributions of 0.255, 0.221, 0.207, 0.202, and 0.116, respectively. In relation to the winter, higher production of NO 3 − from nitrification of soil N- and algae-derived ammonium induced by higher temperatures in the summer accounts for increases in groundwater NO 3 − loadings. In the rural areas, elevated loadings of NO 3 − in the winter may be due to larger infiltration fractions of sewage. Seasonal variations of atmospheric NO 3 − deposition and farming may also cause the dynamics. Our results improve the understanding of sources and seasonal dynamics of NO 3 − in coastal groundwater.

Abstract:Drinking water reservoirs are threatened globally by anthropogenic nitrogen pollution. Hydrochemistry and isotopes were analyzed to identify spatial and temporal varieties of main nitrate sources in a large drinking water reservoir in East China. The results showed that NO3− was the main nitrogen form in both the dry and wet seasons, but dissolved organic nitrogen (DON) was increased in the wet season. The δ15N-NO3− values (+ 1.3‰ to + 11.8‰) and δ18O-NO3− values (+ 2.5‰ to + 13.5‰), combined with principal component analysis (PCA), indicated that chemical fertilizer was the main nitrate source during the dry season, while chemical fertilizer, soil N, and sewage/manure were the main nitrate sources during the wet season in the Qiandao Lake area. And, the nitrate isotopes showed the significant nitrification and assimilation in the Qiandao Lake area. A Bayesian isotopic mixing model (Stable Isotope Analysis in R) was applied to the spatial and seasonal trends in the proportional contribution of four NO3− sources (chemical fertilizer (CF), soil nitrogen (SN), sewage and manure (SM), and atmospheric deposition (AD)) in the Qiandao Lake area. It was revealed that CF was the most important nitrate source in the dry season, accounting for 53.4% with 19.2% of SM and 18.9% of SN, while the contribution of SN increased in the wet season, accounting for 31.6%, followed by CF (30.8%) and then SM (24.2%). The main nitrate sources in the urban area, rural area, and central lake area were CF and SN, accounting for 66.1% in the urban area, 71.7% in the rural area, and 68.2% in the central lake area. Measures should be made to improve chemical fertilizer use efficiency and to reduce nitrogen loss in the Qiandao Lake area..

Identification of the nitrate pollution in surface water of Dongshan Bay based on stable isotope technology

Abstract:Identifying and quantifying water nitrate pollution is crucial for managing aquatic environment of a bay. Dongshan Bay, a significant semi-enclosed bay in the southeastern coastal area of Fujian Province, features mangrove and coral reef ecosystems at its estuary and bay mouth, respectively. Dongshan Bay is impacted by human activities such as mariculture. We quantified and analyzed nitrate pollution status in the surface waters of Dongshan Bay by measuring physicochemical parameters, stable isotopes (δ15N-NO3-, δ18O-NO3- and δ15N-NH4+) of the surface waters, and using statistical methods including the MixSIAR isotope mixing model. The results showed that the concentrations of chlorophyll a and dissolved inorganic nitrogen in the surface waters exhibited a noticeable gradient change, decreasing from the estuary of the Zhangjiang River to the mouth of Dongshan Bay. The maximum concentrations of chlorophyll a, NH4+, NO3- and NO2- were 45.2 μg·L-1, 52.67 μmol·L-1, 379.2 μmol·L-1 and 3.93 μmol·L-1, respectively. The nitrogen and oxygen isotope values of NH4+ and NO3- in the surface waters showed significant spatial variations. According to the MixSIAR model results, nitrogen sources in the surface waters of Dongshan Bay were mainly freshwater inputs of the Zhangjiang River estuary, aquaculture wastewater, and groundwater. The freshwater input from the Zhangjiang River estuary contributed the most (25.2%), while aquaculture wastewater, groundwater and urban sewage accounted for 24.6%, 19.0%, and 15.1%, respectively. It is evident that freshwater input from the Zhangjiang River estuary is the primary source of nitrate in the surface waters of Dongshan Bay.
Identifying Diffused Nitrate Sources in a Stream in an Agricultural Field Using a Dual Isotopic Approach.

Abstract:Nitrate (NO3(-)) pollution is a severe problem in aquatic systems in Taihu Lake Basin in China. A dual isotope approach (δ(15)NNO3(-) and δ(18)ONO3(-)) was applied to identify diffused NO3(-) inputs in a stream in an agricultural field at the basin in 2013. The site-specific isotopic characteristics of five NO3(-) sources (atmospheric deposition, AD; NO3(-) derived from soil organic matter nitrification, NS; NO3(-) derived from chemical fertilizer nitrification, NF; groundwater, GW; and manure and sewage, M&S) were identified. NO3(-) concentrations in the stream during the rainy season [mean±standard deviation (SD)=2.5±0.4mg/L] were lower than those during the dry season (mean±SD=4.0±0.5mg/L), whereas the δ(18)ONO3(-) values during the rainy season (mean±SD=+12.3±3.6‰) were higher than those during the dry season (mean±SD=+0.9±1.9‰). Both chemical and isotopic characteristics indicated that mixing with atmospheric NO3(-) resulted in the high δ(18)O values during the rainy season, whereas NS and M&S were the dominant NO3(-) sources during the dry season. A Bayesian model was used to determine the contribution of each NO3(-) source to total stream NO3(-). Results showed that reduced N nitrification in soil zones (including soil organic matter and fertilizer) was the main NO3(-) source throughout the year. M&S contributed more NO3(-) during the dry season (22.4%) than during the rainy season (17.8%). AD generated substantial amounts of NO3(-) in May (18.4%), June (29.8%), and July (24.5%). With the assessment of temporal variation of diffused NO3(-) sources in agricultural field, improved agricultural management practices can be implemented to protect the water resource and avoid further water quality deterioration in Taihu Lake Basin.

Abstract:Anthropogenic nitrate (NO 3 − ) production has been increasing and is exported to the ocean via river networks, causing eutrophication and ecological damage. While studies have focused on river NO 3 − pollution, what has been lacking is the quantification of the sources of NO 3 − in coastal rivers. This study applied the dual isotopes (δ 15 N/δ 18 O-NO 3 − ) to quantify the sources and their fluxes of NO 3 − in two inflow rivers of the Qinzhou Bay. By adding our results to the NO 3 − source apportionment in Qinzhou Bay, we, for the first time, established the NO 3 − budgets of the terrestrial-to-marine continuum in both high- and low-flow seasons. We quantitatively showed the direct and indirect roles (e.g., the stimulation of nitrification by sewage ammonium-NH 4 + ) of terrestrial sources in driving the high NO 3 − loading in the estuary. The results highlighted the necessity to consider coastal rivers and estuary as a whole, which could shed light on the effective reduction of NO 3 − pollution in coastal environments.

Abstract:Identification and quantification of sources of nitrate (NO3–) in freshwater lakes provide useful information for management of eutrophication and improving water quality in lakes. Dual δ15N- and δ18O-NO3– isotopes and a Bayesian isotope mixing model were applied to identify sources of NO3– and estimate their proportional contributions to concentrations of NO3– in Tai Lake, China. In waters of Tai Lake, values for δ15N-NO3– ranged from 3.8 to 10.1‰, while values of δ18O ranged from 2.2 to 12.0‰. These results indicated that NO3– was derived primarily from agricultural and industrial sources. Stable isotope analysis in R called SIAR model was used to estimate proportional contributions from four potential NO3– sources (agricultural, industrial effluents, domestic sewage, and rainwater). SIAR output revealed that agricultural runoff provided the greatest proportion (50.8%) of NO3– to the lake, followed by industrial effluents (33.9%), rainwater (8.4%), and domestic sewage (6.8%). Contributions of those primary sources of NO3– to sub-regions of Tai Lake varied significantly (p < 0.05). For the northern region of the lake, industrial source (35.4%) contributed the greatest proportion of NO3–, followed by agricultural runoff (27.4%), domestic sewage (21.3%), and rainwater (15.9%). Whereas for the southern region, the proportion of NO3– contributed from agriculture (38.6%) was slightly greater than that contributed by industry (30.8%), which was similar to results for nearby inflow tributaries. Thus, to improve water quality by addressing eutrophication and reduce primary production of phytoplankton, NO3– from both nonpoint agricultural sources and industrial point sources should be mitigated.Graphical abstractᅟ

Abstract:The eutrophication degree in the coastal bay has been increasing significantly for the past years, due to the increasing nutrient discharge. However, the factors controlling sources and nitrogen (N) cycling in the different types of bays are still poorly understood. In this comparative study, nitrate dual isotopes (δ15N−NO3− and δ18O−NO3−) and ammonium nitrogen isotope (δ15N−NH4+) , were used to determine the origin of nitrate in Qinzhou Bay (with riverine input) and Tieshangang Bay (without riverine input) in Beibu Gulf and to study biogeochemical processes associated with nitrogen cycling. The nutrient concentrations generally showed a decreased trend from the upper bay to the outer bay. The isotopic values in Tieshangang Bay were generally higher than that in the Qinzhou Bay, suggesting that there are differences in nitrate sources and transformation processes between the two bays. The dominant sources from manure and sewage (41%) and soil N (30%) from runoff input were responsible for the high nitrate observed in the upper Qinzhou Bay. Though manure and sewage (59%) were also dominant nitrate sources in the upper Tieshangang Bay, a decrease in source from soil N (20%) occurred due to less runoff input. Nutrients were retained in the upper Tieshangang Bay due to weak hydrodynamic conditions, which caused higher NH4+ concentrations in the upper bay. Significant nitrate loss occurred in the outer Qinzhou Bay, which was related to the intense hydrodynamic condition. Moreover, phytoplankton assimilation mainly utilized NH4+ due to sufficient NH4+ in the outer Qinzhou Bay. In contrast, nitrate loss was also found in the outer Tieshangang Bay, which is mainly related to the phytoplankton assimilation due to the weak hydrodynamic condition. In addition, the greater enrichment of δ18O−NO3− than δ15N−NO3− during both bays suggests that atmospheric deposition also contributes to the nitrate pool in the water and the impact of atmospheric deposition on the whole Beibu Gulf is relatively consistent. By this comparative study, we found that different nitrate biogeochemical processes occurred in these two types of bays, which were mainly determined by the difference of human activities and hydrodynamic conditions.

Abstract:A high concentration of nitrate (NO3−) in surface water threatens aquatic systems and human health. Revealing nitrate characteristics and identifying its sources are fundamental to making effective water management strategies. However, nitrate sources in multi-tributaries and mix land use watersheds remain unclear. In this study, based on 20 surface water sampling sites for more than two years’ monitoring from April 2012 to December 2014, water chemical and dual isotopic approaches (δ15N-NO3− and δ18O-NO3−) were integrated for the first time to evaluate nitrate characteristics and sources in the Huashan watershed, Jianghuai hilly region, China. Nitrate-nitrogen concentrations (ranging from 0.02 to 8.57 mg/L) were spatially heterogeneous that were influenced by hydrogeological and land use conditions. Proportional contributions of five potential nitrate sources (i.e., precipitation; manure and sewage, M & S; soil nitrogen, NS; nitrate fertilizer; nitrate derived from ammonia fertilizer and rainfall) were estimated by using a Bayesian isotope mixing model. The results showed that nitrate sources contributions varied significantly among different rainfall conditions and land use types. As for the whole watershed, M & S (manure and sewage) and NS (soil nitrogen) were major nitrate sources in both wet and dry seasons (from 28% to 36% for manure and sewage and from 24% to 27% for soil nitrogen, respectively). Overall, combining a dual isotopes method with a Bayesian isotope mixing model offered a useful and practical way to qualitatively analyze nitrate sources and transformations as well as quantitatively estimate the contributions of potential nitrate sources in drinking water source watersheds, Jianghuai hilly region, eastern China.

Abstract:It is crucial to quantitatively track riverine nitrate (NO3-) sources and transformations in drinking water source watersheds for preventing current and future NO3- pollution, and ensuring a safe drinking water supply. This study identified the significant contributors to riverine NO3- in Zhaoshandu reservoir watershed of Zhejiang province, southeast China. To achieve this goal, we used hydrochemistry parameters and stable isotopes of NO3- (delta N-15-NO3- and delta O-18-NO3-) accompanied with a Markov Chain Monte Carlo mixing model to estimate the proportional contributions of riverine NO3- inputs from atmospheric deposition (AD), chemical nitrogen fertilizer (NF), soil nitrogen (SN), and manure and sewage (M&S). Results indicated that the main form of riverine nitrogen in this region was NO3-, constituting similar to 60% of the total nitrogen mass on average (total organic nitrogen similar to 37% & ammonium similar to 3%). Variations in the isotopic signatures of NO3- demonstrated that microbial nitrification of NF, SN and M&S was the primary nitrogen transformation process within the Zhaoshandu reservoir watershed, whereas denitrification was minimal. A classical dual isotope bi-plot incorporating chloride concentrations suggested NF, SN and M&S were the major contributors of NO3- to the river. Riverine NO3- source apportionment results were further refined using the Markov Chain Monte Carlo mixing model, which revealed that AD, NF, SN and M&S contributed 7.6 +/- 4.1%, 22.5 +/- 12.8%, 27.4 +/- 14.5% and 42.5 +/- 11.3% of riverine NO3- at the watershed outlet, respectively. Finally, uncertainties associated with NO3- source apportionment were quantitatively characterized as: SN > NF > M&S > AD. This work provides a comprehensive approach to distinguish riverine NO3- sources in drinking water source watersheds, which helps guide implementation of management strategies to effectively control NO3- contamination and protect drinking water quality. Summary of the main finding from this works (Capsule) We utilized NO3- stable isotope analysis and a Markov Chain Monte Carlo mixing model to quantify riverine nitrate pollution sources in a drinking water source watershed in Zhejiang province, southeast China. Markov Chain Monte Carlo mixing model output showed that NF, SN and M&S were the dominant sources of riverine NO3- during the sampling period in Zhaoshandu watershed. Uncertainty analysis characterized the variation strength associated with contributions of individual nitrate sources and indicated the greatest uncertainty for SN, followed by NF, M&S and AD.

Abstract:Eutrophication caused by inputs of excess nitrogen (N) has become a serious environmental problem in Hangzhou Bay (China), but the sources of this nitrogen are not well understood. In this study, the August 2019 distributions of salinity, nutrients [nitrate (NO − 3 ), nitrite, ammonium, and phosphate], and the stable isotopic composition of NO − 3 (δ 15 N and δ 18 O) were used to investigate sources of dissolved inorganic nitrogen (DIN) to Hangzhou Bay. Spatial distributions of nitrate, salinity, and nitrate δ 18 O indicate that the Qiantang River, the Changjiang River, and nearshore coastal waters may all contribute nitrate to the bay. Based on the isotopic compositions of nitrate in these potential source waters and conservative mixing of nitrate in our study area, we suggest that the NO − 3 in Hangzhou Bay was likely derived mainly from soils, synthetic N fertilizer, and manure and sewage. End-member modeling indicates that in the upper half of the bay, the Qiantang River was a very important DIN source, possibly contributing more than 50% of DIN in the bay head area. In the lower half of the bay, DIN was sourced mainly from strongly intruding coastal water. DIN coming directly from the Changjiang River made a relatively small contribution to Hangzhou Bay DIN in August 2019.

Abstract:Quantitative identification of non-point sources of nitrate in urban channels plays a critical role in effective nutrient management in urban regions. This is an emerging issue due to fast urbanization and the resultant complicated hydrological and hydraulic conditions in urban areas. In this study, we examine spatial-temporal characteristics of nitrogen concentration in urban channels based on dense in-situ samplings during a one-year period over a small urban catchment in China. We quantitatively identify nitrate sources into urban channels based on dual-isotope analyses and Bayesian isotope mixing model. Results show that nitrogen concentration peaks in winter as well as in urban channels and land surfaces in the urban core region. Sewage (47%) is the dominate contributor to NO3−-N in urban channels, followed by NH4+ in fertilizer (30%) as the second contributor. Sewage (NH4+ in fertilizer) contributes more NO3−-N to channels in winter (summer) with the proportion of 65% (44%), and more NO3−-N to urban core (suburban) channels with the proportion of 59% (42%). The rainfall and distribution of rainwater drains explain the monthly and spatial variations of contribution of NO3−-N sources well, respectively. In addition, less NO3−-N in the urban channels derives from nitrification, which is consistent with the results of high properties of NH4+-N/TN in this region. Our results highlight the key roles of land use types and rainfall in NO3−-N source apportionment, and provide support for the nitrogen management practices in urbanized regions.
Nitrogen pollution and sources in an aquatic system at an agricultural coastal area of Eastern China based on a dual-isotope approach

Abstract:Nitrogen (N) pollution of water courses is a major concern in most coastal watersheds in eastern China with intensive agricultural production. We use hydrogeological and dual-isotopic approaches to analyze the N concentrations, pollution, transformations, and sources of surface water and groundwater in an agricultural watershed of the Jiaozhou Bay (JZB) area. Results showed that dissolved total N (DTN) concentrations in sub-rivers (SRs) ranged from 6.0 to 25.3 mg N L−1 in the dry season and 9.1–26.7 mg N L−1 in the wet season, which indicated a positive relationship with the percentages of agricultural land. Meanwhile, the dominant dissolved N species in SRs changed from nitrate (NO3−, 64–100%) to dissolved organic N (DON, 52–77%) from the dry season to the wet season and the increased DON concentrations showed a positive relationship with the planted proportions of vegetable production systems. The NO3− concentrations of groundwaters ranged from 10.6 to 121.4 mg N L−1, which were over the limit for drinking water by the World Health Organization. Isotopic analysis indicated that most NO3− originated from the microbiological conversion via nitrification, whereas the deletion of denitrification was insignificant in this area. The results of the stable isotope analysis in R mixing model showed the contributions of potential NO3− sources which were in order of manure fertilizers (20.6–69.0%) > soil organic matter (19.5–53.2%) > chemical fertilizers (5.5–34.3%) > atmospheric deposition (1.3–18.8%). This study suggests that the management of crop productions and reasonable manure fertilizer application should be implemented to protect the quality of aquatic systems in the JZB area.

Abstract:The current developments with the production and utility of new generation conjugated typhoid vaccines have been meticulously reviewed by Szu [1]. Nevertheless, the efficacy of different vaccines conjugated to different carrier proteins like rEPA, diphtheria or tetanus toxoid, recombinant diphtheria toxin (CRM197), might be far from ideal during infection of vaccine recipients by Vi-negative Salmonella typhi strains. The existence of naturally occurring Vi-negative strains has been well established now during molecular studies though Vi-negative isolates would not be agglutinated by anti-Vi sera have been known for several decades. During the 1960s, they were identified during phage typing in various geographical locations globally [2]. Genetic control of Vi-expression has been determined, and it is possible to confirm Vi-negative S. typhi. The synthesis and transportation proteins of the Vi capsular polysaccharide of Salmonella enterica serovar Typhi (serovar Typhi) are encoded by the viaB operon, which resides on a 134-kb pathogenicity island known as SPI-7. During analysis of 60 stored strains of serovar Typhi from the Faisalabad region of Pakistan by PCR for the presence of SPI-7 and two genes essential for Vi production: tviA and tviB, 9 (15%) were tested negative for both tviA and tviB, while only two of such strains lacked SPI-7 [3]. Furthermore, the 42 multiplex PCR-positive isolates from the Faisalabad region of Pakistan included 9 Vi negatives [4]. The expanded usage of conjugated Vi vaccines [1] in areas with frequent circulation of Vi-negative isolates [3,4] could aid in selection and dissemination of the Vi-negative strains in the community. The role of circulating Vi-negative strains could be tremendous following extended usage of Vi vaccines. After mass vaccinations, they could be selected with extensive dissemination and disease morbidity and mortality. Such events could be avoided by the use of live attenuated vaccines. The clinical picture associated with such Vi-negative isolates would be indistinguishable from the one caused by isolates carrying Vi antigen [5]. The role of circulating Vi-negative isolates in the community for failures associated with Vi vaccines has not been examined. The reported failures encountered during different randomized trials with typhoid fever vaccines [6] might well have been associated with circulating Vi-negative strains.
