Abstract:Glutathione transferases (GSTs) constitute an ancient, ubiquitous, multi-functional antioxidant enzyme superfamily that has great importance on cellular detoxification against abiotic and biotic stresses as well as plant development and growth. The present study aimed to a comprehensive genome-wide identification and functional characterization of GST family in one of the economically important legume plants— Medicago truncatula . Here, we have identified a total of ninety-two putative MtGST genes that code for 120 proteins. All these members were classified into twelve classes based on their phylogenetic relationship and the presence of structural conserved domain/motif. Among them, 7 MtGST gene pairs were identified to have segmental duplication. Expression profiling of MtGST transcripts revealed their high level of organ/tissue-specific expression in most of the developmental stages and anatomical tissues. The transcripts of MtGSTU 5, MtGSTU 8, MtGSTU 17, MtGSTU 46, and MtGSTU 47 showed significant up-regulation in response to various abiotic and biotic stresses. Moreover, transcripts of MtGSTU 8, MtGSTU 14, MtGSTU 28, MtGSTU 30, MtGSTU 34, MtGSTU 46 and MtGSTF 8 were found to be highly upregulated in response to drought treatment for 24h and 48h. Among the highly stress-responsive MtGST members, MtGSTU17 showed strong affinity towards its conventional substrates reduced glutathione (GSH) and 1‐chloro‐2,4‐dinitrobenzene (CDNB) with the lowest binding energy of—5.7 kcal/mol and -6.5 kcal/mol, respectively. Furthermore, the substrate-binding site residues of MtGSTU17 were found to be highly conserved. These findings will facilitate the further functional and evolutionary characterization of GST genes in Medicago .

Abstract:Plant glutathione S-transferases (GSTs) are integral to normal plant metabolism and biotic and abiotic stress tolerance. The GST gene family has been characterized in diverse plant species using molecular biology and bioinformatics approaches. In the current study, in silico analysis identified 44 GSTs in Vigna radiata. Of the total 44 GSTs identified, chromosomal locations of 31 GSTs were confirmed. The pI value of GST proteins ranged from 5.10 to 9.40. The predicted molecular weights ranged from 13.12 to 50 kDa. Subcellular localization analysis revealed that all GSTs were predominantly localized in the cytoplasm. The active site amino acids were confirmed to be serine in tau, phi, theta, zeta, and TCHQD; cysteine in lambda, DHAR, and omega; and tyrosine in EF1G. The gene architecture conformed to the two-exon/one-intron and three-exon/two-intron organization in the case of tau and phi classes, respectively. MEME analysis identified 10 significantly conserved motifs with the width of 8-50 amino acids. The motifs identified were either specific to a specific GST class or were shared by multiple GST classes. The results of the current study will be of potential importance in the characterization of the GST gene family in V. radiata, an economically important leguminous crop.

Abstract:There is an acknowledged need for better information to guide resource allocation and service planning in the health services. Despite the recognition of the important role of socioeconomic factors, difficulties with the appropriate presentation of daya have so far proved insuperable. Social area analysis (SAA), which is a generic name for a number of methods employing census and other data to classify small areas into similar socioeconomic groups, is an approach which quantifies data in a useful fashion and has important applications in medical, epidemiological and health services research. Most previous British exercises in SAA have been in the field of town planning. The potentialities of the approach for community medicine are evaluated, by the use of information from two existing studies. This is shown to discriminate more effectively than does existing health information between hypotheses concerning geographical variations in mortality, and it provides adequate explanations for urban area differentials in infant mortality, the uptake of vaccination, and the incidence of infectious disease. Specific applications of SAA in health planning and research are discussed.

Abstract:The present investigation was envisaged for large scale in-silico genome wide identification and characterization of glutathione S-transferases (GSTs) in Chenopodium quinoa. In this study, a total of 120 GST genes (CqGSTs) were identified and divided into 11 classes of which tau and phi were highest in numbers. The average protein length of protein was found to be 279.06 with their corresponding average molecular weight of 31,819.4 kDa. The subcellular localization analysis results showed that proteins were centrally localized in the cytoplasm followed by chloroplast, mitochondria and plastids. Structural analysis revealed the presence of 2 -14 exons in CqGST genes. Most of the proteins possessed two exon one intron organization. MEME analysis identified 15 significantly conserved motifs with a width of 6-50 amino acids. Motifs 1, 3, 2, 5, 6, 8, 9 and 13 were found specifically in tau class family; motifs 3, 4, 5, 6, 7 and 9 were found in phi class gene family, while motifs 3, 4, 13 and 14 were found in metaxin class. Multiple sequence alignment revealed highly conserved N-terminus with active site serine (Ser; S) or cysteine (Cys; C) residue for the activation of GSH binding and GST catalytic activity. The gene loci were found to be unevenly distributed across 18 different chromosomes with a maximum of 17 genes located on chromosome number 7. Dominance of alpha helix was followed by coil, extended strand and beta turns. Gene duplication analysis revealed that segmental duplication and purifying type selection were highest in number and found to be main source of expansion of GST gene family. Cis acting regulatory elements analysis showed the presence of 21 different elements involved in stress, hormone and light response and cellular development. The evolutionary relationship of CqGST proteins carried out using maximum likelihood method revealed that all the tau and phi class GSTs were closely associated with those of G. max, O. sativa and A. thaliana. Molecular docking of GST molecules with the fungicide metalaxyl showed that the CqGSTF1 had the lowest binding energy. The comprehensive study of CqGST gene family in quinoa provides groundwork for further functional analysis of CqGST genes in the species at molecular level and has potential applications in plant breeding.

Abstract:The glutathione-S-transferase (GST) family of proteins is ancient and versatile, and plays an important role in combating exogenous pathogens, endogenous toxicants, and various abiotic stresses. Although the GST family has been studied in many crops, few studies have been reported in Chinese cabbage (Brassica rapa ssp. Pekinensis). In the present work, genome-wide analysis of the GST family in Chinese cabbage was carried out, and the expression and functions of genes under different conditions were investigated. A total of 88 GST genes were identified and divided into seven subfamilies according to their evolutionary relationships. Tandem duplication of genes was revealed as the main mechanism of expansion in this family. Transcriptome analysis under high and low temperatures and abiotic stress conditions revealed that most GST genes respond to environmental changes to varying degrees, particularly under herbicide and cadmium stress conditions. Our findings provide a basis for analysing the functions of GST family members in Chinese cabbage, especially in response to various abiotic stresses.
Genome-wide Identification and Transcriptional Analyses of MATE Transporter Genes in Root Tips of Wild Cicer Spp. under Aluminium Stress

Abstract:Chickpea is an economically important legume crop with high nutritional value in human diets. Aluminium-toxicity poses a significant challenge for the yield improvement of this increasingly popular crop in acidic soils. The wild progenitors of chickpea may provide a more diverse gene pool for Al-tolerance in chickpea breeding. However, the genetic basis of Al-tolerance in chickpea and its wild relatives remains largely unknown. Here, we assessed the Al-tolerance of six selected wild Cicer accessions by measuring the root elongation in solution culture under control (0 µM Al3+) and Al-treatment (30 µM Al3+) conditions. Al-treatment significantly reduced the root elongation in all target lines compared to the control condition after 2-day’s growth. However, the relative reduction of root elongation in different lines varied greatly: 3 lines still retained significant root growth under Al-treatment, whilst another 2 lines displayed no root growth at all. We performed genome-wide identification of multidrug and toxic compound extrusion (MATE) encoding genes in the Cicer genome. A total of 56 annotated MATE genes were identified, which divided into 4 major phylogeny groups (G1-4). Four homologues to lupin LaMATE (> 50% aa identity; named CaMATE1-4 ) were clustered with previously characterised MATEs related to Al-tolerance in various other plants. qRT-PCR showed that CaMATE2 transcription in root tips was significantly up-regulated upon Al-treatment in all target lines, whilst CaMATE1 was up-regulated in all lines except Bari2\_074 and Deste\_064, which coincided with the lines displaying no root growth under Al-treatment. Transcriptional profiling in five Cicer tissues revealed that CaMATE1 is specifically transcribed in the root tissue, further supporting its role in Al-detoxification in roots. This first identification of MATE-encoding genes associated with Al-tolerance in Cicer paves the ways for future functional characterization of MATE genes in Cicer spp., and to facilitate future design of gene-specific markers for Al-tolerant line selection in chickpea breeding programs.### Competing Interest StatementThe authors have declared no competing interest.

Abstract:The enzymes known as glutathione S-transferases (GST), which are present in many evolved organisms, are essential for the defense against reactive oxygen species. GSTs have a role in the development of defenses against biotic and abiotic challenges, especially defending plants from various stresses such as drought, salinity, and heavy metal. This study uses a genome-wide investigation of the GST gene family in Phaseolus vulgaris to pinpoint several distinctive traits. 55 Pv-GST proteins have been identified in P. vulgaris . The molecular weights of these proteins range from 15.02 kDa to 47.99 kDa; the range of amino acid numbers 132 to 420 and the range of theoretical isoelectric points 5.03 to 9.61 were identified. Pv-GST genes are estimated to have at least 2 and a maximum of 10 exons, with an average of 4 exons. Phylogenetic analysis was performed with GST proteins from Arabidopsis thaliana , Glycine max , and Phaseolus vulgaris species, and subfamilies of these GSTs were identified. Using RNAseq data, the expression profiles of Pv-GST genes in leaf tissue of common bean during drought and salinity stress were identified. Using the obtained sequence data, primers for qRT-PCR were designed. Changes in the expression profiles of GST genes caused by salt and drought stress and melatonin treatments in two different common bean cultivars were determined by qRT-PCR experiments. Under drought and salt stress, the expression levels of GSTs decreased, while melatonin treatment with few exceptions increased these expression levels. As a result of this study, it was observed that Pv-GST genes may play a role in the growth and development of bean and may be involved in the response to abiotic stresses. Moreover, the results of this study will provide a basis for functional gene research and the expression profiles and qRT-PCR results under different stress conditions in common bean will provide a valuable contribution to the understanding of the functions of the GST gene family.

Abstract:Glutathione S-transferases (GSTs) play versatile functions in multiple aspects of plant growth and development. A comprehensive genome-wide survey of this gene family in the genomes of G. raimondii and G. arboreum was carried out in this study. Based on phylogenetic analyses, the GST gene family of both two diploid cotton species could be divided into eight classes, and approximately all the GST genes within the same subfamily shared similar gene structure. Additionally, the gene structures between the orthologs were highly conserved. The chromosomal localization analyses revealed that GST genes were unevenly distributed across the genome in both G. raimondii and G. arboreum. Tandem duplication could be the major driver for the expansion of GST gene families. Meanwhile, the expression analysis for the selected 40 GST genes showed that they exhibited tissue-specific expression patterns and their expression were induced or repressed by salt stress. Those findings shed lights on the function and evolution of the GST gene family in Gossypium species.

Abstract:Glutathione S-transferases (GSTs) are ubiquitous enzymes which play versatile functions including cellular detoxification and stress tolerance. In this study, a comprehensive genome-wide identification of GST gene family was carried out in potato (Solanum tuberosum L.). The result demonstrated the presence of at least 90 GST genes in potato which is greater than any other reported species. According to the phylogenetic analyses of Arabidopsis, rice and potato GST members, GSTs could be subdivided into ten different classes and each class is found to be highly conserved. The largest class of potato GST family is tau with 66 members, followed by phi and lambda. The chromosomal localization analysis revealed the highly uneven distribution of StGST genes across the potato genome. Transcript profiling of 55 StGST genes showed the tissue-specific expression for most of the members. Moreover, expression of StGST genes were mainly repressed in response to abiotic stresses, while largely induced in response to biotic and hormonal elicitations. Further analysis of StGST gene's promoter identified the presence of various stress responsive cis-regulatory elements. Moreover, one of the highly stress responsive StGST members, StGSTU46, showed strong affinity towards flurazole with lowest binding energy of -7.6kcal/mol that could be used as antidote to protect crop against herbicides. These findings will facilitate the further functional and evolutionary characterization of GST genes in potato.
Probing the diversity of the Arabidopsis glutathione S-transferase gene family

Abstract:Glutathione S-transferases (GSTs) appear to be ubiquitous in plants and have defined roles in herbicide detoxification. In contrast, little is known about their roles in normal plant physiology and during responses to biotic and abiotic stress. Forty-seven members of the GST super-family were identified in the Arabidopsis genome, grouped into four classes, with amino acid sequence identity between classes being below 25%. The two small zeta (GSTZ) and theta (GSTT) classes have related GSTs in animals while the large phi (GSTF) and tau (GSTU) classes are plant specific. As a first step to functionally characterize this diverse super-family, 10 cDNAs representing all GST classes were cloned by RT-PCR and used to study AtGST expression in response to treatment with phytohormones, herbicides, oxidative stress and inoculation with virulent and avirulent strains of the downy mildew pathogen Peronospora parasitica. The abundance of transcripts encoding AtGSTF9, AtGSTF10, AtGSTU5, AtGSTU13 and AtGSTT1 were unaffected by any of the treatments. In contrast, AtGSTF6 was upregulated by all treatments while AtGSTF2, AtGSTF8, AtGSTU19 and AtGSTZ1 each showed a selective spectrum of inducibility to the different stresses indicating that regulation of gene expression in this super-family is controlled by multiple mechanisms. The respective cDNAs were over expressed in E. coli. All GSTs except AtGSTF10 formed soluble proteins which catalysed a specific range of glutathione conjugation or glutathione peroxidase activities. Our results give further insights into the complex regulation and enzymic functions of this plant gene super-family.

Abstract:The knowledge of moisture content in surface concrete structure is a major subject of interest frequently studied by ground-penetrating radar (GPR) techniques. This paper proposes a parametric study of an extraction technique of the direct wave dispersion-technique originally applied for seismological data and adapted to GPR waves. The herein procedure includes the picking of the direct wave in the material and the calculation of the dispersion curve of the phase velocity in function of frequency, using the f - β transform. Unfortunately, for GPR signals, many factors can introduce inaccuracies into the determination of the dispersion curves. In this case, we need to know the influence of different parameters used in this processing methodology.

Abstract:Glutathione S-transferases (GSTs; EC 2.5.1.18) are members of an isozyme family and catalyze the conjugation of the reduced tripeptide glutathione to a variety of hydrophobic and electrophilic substrates. Though members of different classes of the GST superfamily have been identified and characterized from many plant species including rice, no detailed information is available for the Lambda class gene family in rice. In this study, a genome-wide analysis was carried out to investigate expression patterns of three Lambda class GST members of rice including OsGSTL1, OsGSTL2 and OsGSTL3 in seedlings, at different growth and developmental stages as well as in response to various biotic and abiotic stresses. Expression analysis using microarray datasets and quantitative real-time reverse transcriptase polymerase chain reaction suggests that this gene family express differentially in various tissues, in response to hormones and during different biotic and abiotic stresses including heavy metals, cold, drought and salt stress. Massively Parallel Signature Sequencing (MPSS) analysis also showed differential expression of OsGSTLs during plant growth and development and under different stresses. Out of three members, maximum expression of OsGSTL2 was observed for the MPSS libraries in comparison to other members. We conclude that members of rice Lambda class GST family play an important role in plant growth and development and in combating different biotic and abiotic stresses.

Abstract:Plant glutathione S -transferases (GSTs) are a large group of multifunctional proteins that are induced by diverse stimuli. Using proteomic approaches we identified 20 GSTs at the protein level in Arabidopsis cell culture with a combination of GST antibody detection, LC-MS/MS analysis of 23-30 kDa proteins and glutathione-affinity chromatography. GSTs identified were from phi, tau, theta, zeta and DHAR sub-sections of the GST superfamily of 53 members. We have uncovered preliminary evidence for post-translational modifications of plant GSTs and show that phosphorylation is unlikely to be responsible. Detailed analysis of GST expression in response to treatment with 0.01-1 mM of the plant defence signal salicylic acid (SA) uncovered some interesting features. Firstly, GSTs appear to display class-specific concentration-dependent SA induction profiles highlighting differences between the large, plant specific phi and tau classes. Secondly, different members of the same class, while sharing similar SA dose responses, may display differences in terms of magnitude and timing of induction, further highlighting the breadth of GST gene regulation. Thirdly, closely related members of the same class ( GSTF6 and GSTF7 ), arising via tandem duplication, may be regulated differently in terms of basal expression levels and also magnitude of induction raising questions about the role of subfunctionalisation within this family. Our results reveal that GSTs exhibit class specific responses to SA treatment suggesting that several mechanisms are acting to induce GSTs upon SA treatment and hinting at class-specific functions for this large and important, yet still relatively elusive gene family.

Abstract:The GATA transcription factors are important transcriptional regulators of plant growth and responses to environmental stimuli. Here, a total of 25 CaGATA genes was identified in chickpea (Cicer arietinum), and their basic characteristics, including gene structure, duplication patterns, conserved domains and various physical and chemical parameters were subsequently determined. Of our interest, the enrichment of the hormone- and stress-responsive cis-regulatory elements in the promoters of CaGATA genes has been analyzed to predict the CaGATA members with potential hormone-mediated functions in stress tolerance. Furthermore, the tissue-specific expression patterns of the CaGATA genes were assessed using the available transcriptome data. More importantly, transcript levels of the identified CaGATA genes were quantified in roots and leaves of chickpea seedlings exposed to ABA (abscisic acid) or dehydration treatment using real-time quantitative PCR. Expression levels of a total of 12 CaGATA genes were significantly altered in roots and/or leaves by both ABA and dehydration treatments, suggesting that these genes might play roles in regulation of chickpea response to water stress in an ABA-dependent manner. Out of these genes, only CaGATA04 was induced in both roots and leaves by ABA and dehydration treatments. Furthermore, CaGATA05 and 21 were the most highly induced in roots (8.55-fold) and leaves (4.90-fold), respectively, by dehydration. Findings of this study have provided important insights into the CaGATA family of chickpea, as well as useful information for selection of CaGATA genes of interest for in-depth functional characterizations that might lead to development of chickpea cultivars with improved performance under water-deficit conditions.

Comparative Analysis of the Glutathione S-Transferase Gene Family of Four Triticeae Species and Transcriptome Analysis of GST Genes in Common Wheat Responding to Salt Stress

Abstract:Glutathione S-transferases (GSTs) are ancient proteins encoded by a large gene family in plants, which play multiple roles in plant growth and development. However, there has been little study on the GST genes of common wheat (Triticum aestivum) and its relatives (Triticum durum, Triticum urartu, and Aegilops tauschii), which are four important species of Triticeae. Here, a genome-wide comprehensive analysis of this gene family was performed on the genomes of common wheat and its relatives. A total of 346 GST genes in T. aestivum, 226 in T. durum, 104 in T. urartu, and 105 in Ae. tauschii were identified, and all members were divided into ten classes. Transcriptome analysis was used to identify GST genes that respond to salt stress in common wheat, which revealed that the reaction of GST genes is not sensitive to low and moderate salt concentrations but is sensitive to severe concentrations of the stressor, and the GST genes related to salt stress mainly come from the Tau and Phi classes. Six GST genes which respond to different salt concentrations were selected and validated by a qRT-PCR assay. These findings will not only provide helpful information about the function of GST genes in Triticeae species but also offer insights for the future application of salt stress resistance breeding in common wheat.
