Abstract:It is of importance for bioimaging of fungal cells using biocompatible and low toxic carbon dots (CDs) as labels in plant protection field because a clearer understanding on the infection mechanism of fungi on plant can be achieved. Meanwhile, long wavelength, especially, red/near-infrared (MR) emissive CDs are more biocompatible than short wavelength emissive ones. In this work, CDs with red emission were synthesized by solvothermal pyrolysis of citric acid, aciylamide dissolved in formamide. Fungal cells stained by the CDs with red emission were brightly illuminated when imaged on a fluorescent microscope with excitation by a green laser pulse, suggesting the CDs are of an excellent label for bioimaging of fungal cell in red color region. Moreover, the CDs show a selective response to Hg2+ in the NaAc-HAc buffer solution, while ziram can form a more stable complex with Hg2+, leading to a recovery of the quenched fluorescence of the CDs. Therefore, methods for the detections of Hg2+ and ziram based on the "off-on" fluorescence of the CDs were established with limits of detection as low as 0.19 mu M and 0.55 mu g/ml. Fluorescent Nitrogen and Sulfur Co-Doped Carbon Dots from Casein and Their Applications for Sensitive Detection of Hg2+ and Biothiols and Cellular Imaging.

Abstract:Fluorescent nitrogen and sulfur co-doped carbon dots (NSCDs) were synthesized by a one-step pyrolysis strategy using casein as carbon, nitrogen and sulfur sources, and characterized by UVevis spectrum, fluorescent spectrum, X-ray photoelectron spectroscopy (XPS) and FT-IR, etc. The synthesized NSCDs displayed a blue emission under ultraviolet illumination with a quantum yield of 31.8%, and a good aqueous solubility, photostability and biocompatibility. It was found that the fluorescence intensity of NSCDs could be selectively quenched by Hg2+, so NSCDs was used as an effective probe for the detection of Hg2+. The linear range and the limit of detection (LOD) of the fluorescent sensor based on NSCDs for the detection of Hg2+ were 0.01-0.25 mu M and 6.5 mu M, respectively. Spiked water samples were detected by the sensor with the recovery of 95.4-106.3% and relative standard deviation (RSD) of 3.6-8.6%. It was also observed that the quenched NSCDs-Hg2+ system could be restored by the addition of biothiols such as L-cysteine (Lcy), homocysteine (Hcy) and glutathione (GSH), thus NSCDs-Hg2+ system was employed as a fluorescent sensor for the detection of biothiols. The linear range and LOD of the NSCDs-Hg2+ system were 1-10 mM and 23.6 nM for Lcy, 0.2-2.5 mM and 12.3 nM for Hcy, and 0.1-2.0 mM and 16.8 nM for GSH, respectively. The NSCDs-Hg2+ system was applied for the detection of biothiols in serum samples with satisfied results. In addition, the study in vitro imaging HeLa cells revealed that the synthesized NSCDs could be used as effective fluorescent probes in cellular imaging without noticeable cytotoxicity. An Improved Synthesis of Water-Soluble Dual Fluorescence Emission Carbon Dots from Holly Leaves for Accurate Detection of Mercury Ions in Living Cells.

Abstract:Background Carbon dots (CDs) emitting near-infrared fluorescence were recently synthesized from green leaves. However, the Hg2+ detection of CDs was limited because of the insufficient water solubility, low fluorescence and poor stability. Methods Dual fluorescence emission water-soluble CD (Dual-CD) was prepared through a solvothermal method from holly leaves and low toxic PEI1.8k. PEG was further grafted onto the surface to improve the water solubility and stability. Results The Dual-CD solution can emit 487 nm and 676 nm fluorescence under single excitation and exhibit high quantum yield of 16.8%. The fluorescence at 678 nm decreased remarkably while the emission at 470 nm was slightly affected by the addition of Hg2+. The ratiometric Hg2+ detection had a wide linear range of 0–100 μM and low detection limit of 14.0 nM. In A549 cells, there was a good linear relation between F487/F676 and the concentration of Hg2+ in the range of 0–60 μM; the detection limit was 477 nM. Furthermore, Dual-CD showed visual fluorescence change under Hg2+. Conclusion Dual-CD has ratiometric responsiveness to Hg2+ and can be applied for quantitative Hg2+ detection in living cells.

Abstract:Carbon dots (CDs) have garnered extensive interest in basic physical chemistry as well as in biomedical applications due to their low cost, good biocompatibility, and great aqueous solubility. However, the synthesis of multi-functional carbon dots has always been a challenge for researchers. Here, we synthesized novel CDs with a high quantum yield of 28.2% through the straightforward hydrothermal method using Diaminomaleonitrile and Boc-D-2, 3-diaminopropionic acid. The size, chemical functional group, and photophysical properties of the CDs were characterized by TEM, FTIR, XPS, UV, and fluorescence. It was demonstrated in this study that the prepared CDs have a high quantum yield, excellent photostability, and low cytotoxicity. Regarding the highly water-soluble property of CDs, they were proven to possess selective and sensitive behavior against Cu2+ ions (linear range = 0-9 μM and limit of detection = 1.34 μM). Moreover, the CDs were utilized in fluorescent ink in anti-counterfeiting measures. Because of their low cytotoxicity and good biocompatibility, the CDs were also successfully utilized in cell imaging. Therefore, the as-prepared CDs have great potential in fluorescence sensing, anti-counterfeiting, and bioimaging.

Abstract:Recently, widespread attention has been paid to red emissive carbon dots (CDs) which have desirable optical properties, low toxicity, and biocompatibility. Despite great efforts, the facile preparation of red dual-emissive CDs useful for ratiometric detection and bioimaging remains challenging. Here, we report a facile synthesis of red dual-emissive CDs and their potential for ratiometric fluorescence sensing and cellular imaging. Derived from the hydrothermal treatment of dicyandiamide and o-phenylenediamine in dilute sulfuric acid, the CDs are surface-tailored with nitrogen-, oxygen-, and sulfur-containing functional groups. The as-prepared CDs show various good features, including good water solubility, biocompatibility, excitation-independent dual-emission with two photoluminescence (PL) peaks centered at 630 and 680 nm, and an absolute quantum yield (QY) of 30.2% in water. The CDs exhibit a selective, sensitive, rapid, and stable ratiometric fluorescence response toward methyl blue, giving a linear relationship in the range of 0.5-300 mu M with a correlation coefficient (R-2) of 0.997. We also study ratiometric fluorescence sensing for the accurate detection of pH. Moreover, the CDs possess good cellular imaging ability, indicating their promising applicability for biomedical applications. These results pave a way toward the fabrication of red dual-emissive carbon-based nanomaterials useful for both ratiometric sensing and bioimaging.

Abstract:Carbon dots (CDs) are a new class of photoluminescent (PL), biocompatible, environment-friendly, and low-cost carbon nanomaterials. Synthesis of highly efficient red-emitting carbon dots (R-CDs) on a gram scale is a great challenge at present, which heavily restricts the wide applications of CDs in the bioimaging field. Herein, R-CDs with a high quantum yield (QY) of 53% are produced on a gram scale by heating a formamide solution of citric acid and ethylenediamine. The as-prepared R-CDs have an average size of 4.1 nm and a nitrogen content of about 30%, with an excitation-independent emission at 627 nm. After detailed characterizations, such strong red fluorescence is ascribed to the contribution from the nitrogen- and oxygen-related surface states and the nitrogen-derived structures in the R-CD cores. Our R-CDs show good photostability and low cytotoxicity, and thus they are excellent red fluorescence probes for bioimaging both in vitro and in vivo.
N,S-self-doped carbon quantum dots from fungus fibers for sensing tetracyclines and for bioimaging cancer cells

Abstract:In this work, nitrogen and sulfur dual-doped carbon quantum dots (N,S-CDs) from naturally renewable biomaterial fungus fibers were prepared by a biosynthesis and hydrothermal method. The N,S-CDs displayed good water solubility, excellent stability, high quantum yield (QY = 28.11%) as well as remarkable features for fluorescence quenching-based detection and cellular imaging of cancer cells. It was worth mentioning that the heteroatoms doped carbon quantum dots made from the fungus fibers had a satisfactory QY and could be used as a selective, efficient, and sensitive fluorescent probe to determine tetracyclines by the synergistic effects of static quenching and internal filtration effect. The probe demonstrated a wide linear range and low detection limit. For tetracycline, the linear range was 0.5 μM to 47.6 μM, and the corresponding detection limit was 15.6 nM. Significantly, the test papers prepared by using N,S-CDs could detect tetracyclines in aquiculture wastewater rapidly. The produced N,S-CDs did not affect the cell viability and showed great promises for cellular imaging.

Abstract:Dual-emissive carbon dots (CDs) were fabricated for dual-channel ratiometric fluorometric determination of pH and mercury ion (Hg2+) and intracellular imaging. Dual-emissive CDs were synthesized by one-pot solvothermal treatment of cabbage. The CDs exhibited two distinctive fluorescence emissions at 500 and 678 nm under single excitation at 410 nm. The green emission (500 nm) had reversible linear response to pH (7.0–12.0) due to deprotonation and protonation of surface functional groups and their non-covalent interactions. On the other hand, the red emission (678 nm) had efficient and selective fluorescence response to Hg2+ by formation of non-emission complex between CDs and Hg2+. The limit of detection (LOD) and limit of quantification (LOQ) for Hg2+ were 6.25 and 20.63 nM, respectively. The CDs have been successfully applied for label-free ratiometric fluorometric determination of pH and Hg2+ in fish and human serum samples with good recoveries (92.0–108.3%). In addition, the CDs had excellent photostability, low cytotoxicity, and good biocompatibility for intracellular imaging. All in all, the system was multi-functional in determination, high in sensitivity, and excellent in selectivity, which demonstrated wide and promising applicability for biosensing and bioimaging in the future.

Abstract:Red-emitting carbon dots (R-CDs) have triggered ever-increasing interest in biochemical sensing due to their deep tissue penetration and minimal biological auto-fluorescence interference. Herein, dual-emitting intrinsic RCDs are fabricated and well-characterized. R-CDs display ratiometric and colorimetric sensing properties for heavy metal ions (HMIs, Cu2+, Hg2++, and Fe3++), with the corresponding detection limits of 24.5, 74.2, and 170 nM, respectively. Additionally, smartphone imaging colorimetry based on red-green-blue (RGB) value variations is employed to achieve real-time and on-site detection of these HMIs. Crucially, discrimination of these HMIs is facilitated by principal component analysis. Furthermore, R-CDs demonstrate effective optical sensing of these HMIs in tap/lake water and human urine with satisfactory recoveries (95.82-104.8 %). The underlying mechanism of optical sensing, which involves metal ion-triggered aggregation-induced quenching, is elucidated by spectroscopic, morphological analysis, and quantum mechanical calculations. This work presents a feasible realtime approach for the fluorescent and smartphone-assisted dual-mode detection of HMIs.

Abstract:Fluorescent red-emissive carbon dots (RCDs) were synthesized using the solvothermal method with citric acid as a carbon source, N , N -dimethylformamide as a nitrogen source, and formamide as a solvent. The as-synthesized RCDs show red fluorescence in an aqueous solution and have an excellent stability towards photobleaching as well as extremely low cytotoxicity and are successfully used for cell and zebrafish imaging. The results indicate that RCDs have potential applications in both in vitro and in vivo bioimaging.

Abstract:The authors describe strongly red-emitting carbon dots (CDs) which were obtained via microwave synthesis using phenylenediamine as the carbon source. The structural and optical properties of the resultant CDs are studied in some detail. The CDs possess (a) longwave emission (peaking at 620 nm under 470 nm excitation), (b) a quantum yield of ~15%, (c) a size of typically 3.8 nm; and (d) good photostability. The CDs have a pH-dependet response that covers the pH 5 to 10 range, and their fluorescence is quenched by ferric ions. The CDs can detect ferric ions in aqueous samples in the 0 to 30 μM concentration range with a lower detection limit of 15 nM. The CDs were also used to image pH values and ferric ions in E. coli bacteria. Graphical abstract The red-emitting carbon dots with high stability are synthesized which show dual response to pH-values and ferric ions in aqueous solution and biological media simultaneously.

Abstract:A water-soluble probe composite materials (CDs/ZnO) was synthesized by microwave pyrolysis methods of citric acid, urea and zinc oxide (ZnO) with aggregation-induced emission activity. We creatively realize the composite of CDs and nano-ZnO in the form of foam, then used as bactericide for the first time and showed excellent antibacterial properties. Also, the composite materials overcame the problem of aggregation-induced quenching (ACQ) and realized solid-state emission. The materials had two different emission centers, and the two emission peaks were varied with the change of excitation wavelength. Furthermore, Methyl red (MR) showed fluorescence quenching for CDs/ZnO with obvious linear correlation, which demonstrated potential in the measurement of MR in dyeing wastewater. The CDs/ZnO we obtained perfectly inherited the advantages of CDs and nano ZnO, it has not just no cytotoxicity to cells, but also has a good killing effect on bacteria. Highly selective and sensitive ratiometric detection of Hg2+ ions with NFS co-doped carbon dots: Real sample analysis, antibacterial properties, and cellular imaging applications

Abstract:A simple, low-cost hydrothermal method was employed to synthesize highly fluorescent nitrogen-, fluorine-, and sulfur-co-doped carbon dots (NFS-CDs) using flufenamic acid and L-cysteine as precursors. The synthesized NFS-CDs exhibited dual emission peaks at 490 and 580 nm with a quantum yield of 24.7 %. They exhibit excellent stability, excitation-dependent fluorescent, and particle sizes ranging from 2 to 8 nm. The fluorescent chemosensor probe, NFS-CDs, showed strong selectivity and sensitivity for Hg2+ over other metal ions investigated in aqueous solutions (pH ∼ 7.4). Strong fluorescent enhancement at 490 nm and considerable quenching at 580 nm was observed in the presence of Hg2+ ions. The stoichiometric ratio of the NFS-CDs/Hg2+ complex was optimized to 1:1 according to the Benesi-Hildebrand and Stern-Volmer plot methods. The NFS-CDs exhibited a linear dynamic detection range from 0 to 10 × 10-6 M for Hg2+ ions with a lower detection limit of 18.0 and 67.5 × 10-9 M, respectively, at 490 and 580 nm. Practical applications of NFS-CDs in detecting Hg2+ ions in natural water samples showed high recovery rates (98.9-104.6 %) and low relative standard deviation (RSD ≤ 2.47 %). The NFS-CDs/Hg2+ achieved 78.7 ± 2.6 % and 83.4 ± 2.3 % antibacterial activity against E. coli and S. aureus as NFS-CDs/Hg2+ could damage the bacterial walls when they entered the bacteria. Furthermore, the NFS-CDs were used to detect Hg2+ ions intracellularly in HCT116 cells with low toxicity using live cell imaging.

Abstract:Introduction: Iron is one of the most important needed elements for the growth and reproduction of living organisms. The detection of iron levels is important and developing fluorescent probes with excellent sensitivity for Fe 3+ ions is of great significance. Carbon dot (CDs) is a new type of fluorescent nanomaterial based on abundant and low-cost carbon elements. The use of widely distributed renewable agricultural waste straw as a carbon precursor to prepare CDs sensor can not only reduce the pollution caused by burning straw to the atmospheric environment, but also achieve the transformation of resources from waste to treasure. Methods: In this study, CDs were obtained from corn stalk powder by pyrolysis and microwave process. The sensitivity and linear response range of CDs sensor was studied through analyzing the effect of different Fe 3+ ions concentrations on the fluorescence quenching. The application of CDs in biological cell imaging was investigated using HGC-27 cells. Results: The fluorescence quenching showed a good linear relationship with the Fe3+ concentration in the range from 0 to 128 μM, and a low detection limit of 63 nM. In addition, the CDs have high recognition for Fe3+ ions. Meanwhile, the CDs have a low cytotoxicity and desirable biocompatibility, allowing the multicolor living cell imaging. Conclusion: The prepared CDs can be used as fluorescent sensors for the selective detection of Fe 3+ ions and biological cell imaging. Our results supported that the conversion of agricultural waste into carbon nanomaterials has great potential to be developed.

Abstract:Carbon dots (CDs) also nitrogen-doped CDs (N-CDs) were produced by green hydrothermal synthesis using Pea and ethanediamine as the carbon and nitrogen source, separately. Transmission electron microscopy (TEM) images displayed that the prepared CDs and N-CDs were well dispersed, had a spherical morphology. X-ray diffraction (XRD) figures of CDs and N-CDs presented a graphitic amorphous structure. Fourier transform infrared spectroscopy (FT-IR) verified that CDs and N-CDs carried many different hydrophilic groups (for example hydroxyl, carboxyl/carbonyl, amide, amino groups) on the surface, X-ray photoelectron spectroscopy (XPS) together verified this result. However, the optical properties and fluorescence quantum yield for N-CDs were obviously superior to those of CDs. Furthermore, the prepared N-CDs displayed outstanding advantages including low toxicity, satisfactory biocompatibility, and excellent chemical stability. More prominently, the prepared N-CDs could detect Hg2+ ions with high sensitivity and selectivity in both water samples and HeLa cells.

Abstract:A novel type of red-emitting carbon dots was synthesized using urea and neutral red via the solvothermal method at a temperature of 200 °C for 4 hours. The resulting CDs, named NR-CDs, display a distinct red emission at 620 nm when excited at a wavelength of 470 nm, achieving a quantum yield of 61.11% in acetone. Interestingly, NR-CDs exhibit strict solvent polarity dependence, i.e. the emission wavelength of NR-CDs red-shifts with an increase in solvent polarity. Besides, when incubated with NR-CDs and excited by an excitation wavelength of 470 nm, 4-T1 cells emit a clear red fluorescence. Meanwhile, high-quality in-vivo fluorescent images of zebrafish embryos and larvae were obtained by incubating with NR-CDs. Moreover, by modifying with polyetherimide (PEI), the peak value of PEI-NR-CDs shifts from 620 nm to 596 nm, and the emission is increased by 264%. Co-localization experiment with commercial dye (Lyso-Tracker Green) shows that PEI-NR-CDs are localized in lysosomes and could be used as a lysosomal tracer for cells. Furthermore, PEI-NR-CDs were prepared into fluorescent ink, which exhibited orange fluorescence under UV light. All results demonstrate that as-obtained NR-CDs and PEI-NR-CDs can be used as reagent for fluorescent-imaging application.

Abstract:Recent studies suggest that mild hyperhomocysteinemia may be a risk factor for venous thromboembolic disease (VTED). In this work we evaluated the prevalence of moderate hyperhomocysteinemia in patients with VTED in our area. We found hyperhomocysteinemia in 23.4% of 64 patients studied compared with 7.35% of 68 healthy controls (p = 0.014). Our results suggest that moderate hyperhomocysteinemia is one of the most prevalent abnormalities associated with VTED.

Abstract:Red-emitting carbon dots have attracted much attention because of their excellent fluorescence properties. It is of great significance to synthesize red-emitting carbon dots with high fluorescence quantum yield. Herein, by using citric acid and 1,8-diaminonaphthalene as precursors in N, N-dimethylformamide, novel carbon dots (NC-CDs) exhibiting red emission at 607 nm upon excitation at 514 nm were synthesized via a solvent-thermal approach. NC-CDs aggregated in lipid droplets of cells, and hemin quenched their fluorescence. Subsequently, cationic surfactants cetyltrimethylammonium bromide, anionic surfactants sodium dodecyl sulfate and nonionic surfactants were used to functionalize the NC-CDs. The results indicated that both ionic surfactants increased the fluorescence quantum yield and solubility of NC-CDs with extended fluorescence lifetime. This approach is applicable to a class of carbon dots that possess both carboxyl and amino groups. Additionally, these functionalized NC-CDs exhibited high recognition sensitivity towards ClO− and Cu2+ respectively except for hemin. This study provides a novel approach for the synthesis of highly red-emitting carbon dots, expanding their applications in the field of biological imaging and sensing. The findings hold significant implications for the real-time monitoring of hemin in lipid droplets. Additionally, this work offers a simple method to dramatic enhance their fluorescence quantum yield.

Abstract:Biomass-based carbon dots (Bio-CDs) were prepared from dehydroabietic acid using a one-step hydrothermal process. Characterization by TEM, XPS and FTIR spectroscopy showed that the Bio-CDs are spherical nanoparticles containing mainly C, N and O elements, with functional groups such as amino and carbonyl groups on their surface. The optical properties of the Bio-CDs were studied in detail. A solution of Bio-CDs exhibited excitation-dependent blue fluorescence emission. The solution showed excellent photostability under ultraviolet light and the fluorescence intensity could be enhanced by decreasing the temperature. The intensity of fluorescence emission of the solution was essentially unchanged over the pH range 3.91–8.69, and in the presence of different anions and cations, other than Fe 3+ and Pb 2+ . Fe 3+ and Pb 2+ ions, respectively, quenched and enhanced the intensity of the fluorescence emission of the solution, allowing sensitive and selective detection of Fe 3+ (LOD = 2.33 μM, Em = 437 nm) and of Pb 2+ (LOD = 0.27 μM, Em = 437 nm and LOD = 0.33 μM, Em = 500 nm). As a further demonstration of potential applications, the Bio-CDs were shown to have low cytotoxicity and to stain cell nuclei as effectively as the commonly used nuclear stain 4′,6-diamino-2-phenylindole (DAPI), demonstrating their promise in the field of cell imaging.

Abstract:In this article, carbon dots (CDs) were synthesized using methyl red (MR) as precursor in the absence and presence of ethanediamine (EDA) by facile hydrothermal pyrolysis, referring to as MR-CDs and MR-EDA-CDs, respectively. MR-CDs synthesized at different temperatures exhibited molecular state controlled photoluminescence (PL) property with the same characteristics (emission position and shape, no contribution from carbon core) as MR, except that their quantum yields (QYs) decreased with the reaction temperature increasing because of the consumption of MR. On the other hand, MR-EDA-CDs were fractionated into four fractions, which individually displayed unique surface state controlled PL property. Moreover, four fractions isolated from MR-EDA-CDs have specific surface groups that can selectively recognize pH, Fe3+, Ag+ and Hg2+, respectively, in aqueous solution, then were developed as the corresponding sensors. In addition, MR-EDA-CDs are potential fluorescent agents with excellent biocompatibility for bioimaging of fungal cells.
