Abstract:C-type lectins (CTLs) are important pattern recognition receptors (PRRs) that play vital roles in innate immunity. In teleosts, a number of CTLs have been reported, but their in vivo effects on host defense are still limited. In this study, a CTL homolog (SsLec1) was identified from black rockfish, Sebastes schlegelii, and its structure, expression and biological function was analyzed. The open reading frame of SsLec1 is 633 bp, with a 5'- untranslated region (UTR) of 36 bp and a 3'- UTR of 117 bp. The deduced amino acid sequence of SsLec1 shares the highest overall identity (73.20%) with the CTL of Oplegnathus fasciatus. SsLec1 possesses conserved CTL features, including a carbohydrate-recognition domain, four disulfide bond-forming cysteine residues, the mannose-type carbohydrate-binding motif, the conserved calcium binding sites and a putative signal peptide. The expression of SsLec1 was highest in liver and could be induced by experimental infection with Listonella anguillarum. Recombinant SsLec1 (rSsLec1) purified from E. coli was able to bind and agglutinate the Gram-negative fish pathogens Vibrio ichthyoenteri and Vibrio vulnificus. The agglutinating ability of rSsLec1 was abolished in the presence of mannose or ethylenediaminetetraacetic acid. Further analysis showed that rSsLec1 could enhance phagocytosis by macrophages. In vivo experiments indicated that rSsLec1 could inhibit bacterial infection and promote viral invasion. Taken together, these results suggest that SsLec1 is a novel CTL that possesses apparent immunoregulation property and plays a critical role in host defense against pathogens invasion.
Recombinant expression and functional characterization of a CXC chemokine in black seabream (Acanthopagrus schlegeli)

Abstract:With the method of DNA recombination in vitro,a black sea bream CXC (BSCXC) chemokine mature peptide cod- ing sequence was sub cloned into expression vector pQE30 and expressed in E.Coli M15.About 8kDa expressed protein was purified by Ni-NTA chromatographic technology and identified by Western blotting using 6xHis tag antibodies.The bioactivity assay showed that the expressed protein had significantly chemotactic activities towards both leucocytes and macrophages in a dose dependent manner (P<0.01,P<0.05),and the BSCXC had stronger chemotaxis effects to leucocytes than that to macrophages.Furthermore,the BSCXC can significantly enhance the phagocytic activity of macrophages.The results indicate that the BSCXC is an incomplete ELR~+ CXC chemokine for it is structurally and func- tionally incomplete,and possesses ELR~+ CXC and ELR~- CXC chemokine activities.

Abstract:CC chemokines are a superfamily of chemotactic cytokines involved in recruitment,activation and adhesion of a variety of leukocyte types to inflammatory foci,the important component of the innate immune system.In this paper the structure and function of CC chemokine genes from economically important fishes such as rainbow trout(Oncorhynchus mykiss),flounder(Paralichthys olivaceus)and catfish were analysed by comparative genomics and bioinformatics methods.Phylogenetic analysis of all the new CC chemokines and the previously published fish chemokines using the neighbor joining method allowed the establishment of orthologies of three distinct CC chemokine groups with non-fish CC chemokines.The first orthology was established with human CCL27 and CCL28.Included within this clade were cichlid(Melanochromis auratus and Paralabidochromis chilotes) SCYA101 and SCYA101a,salmon(Salmo salar) CB510320,zebrafish(Danio rerio) BI839410,rainbow trout CK11,channel catfish(Ictalurus punctatus) BM027974 and BM029630.The second orthology was established with human CCL20.This clade contained the trout CK8a,CK8b,catfish SCYA112,and chicken(Gallus gallus) AAK84434.The chicken sequence was much closer to the human CCL20 than to any of the fish CC chemokines included in the clade.The third orthology was established between chicken XP424980 and blue catfish(Ictalurus furcatus) SCYA109.All the other fish CC chemokines were not grouped into clades containing non-fish CC chemokines.Phylogenetic analysis using Clustal W by heuristic search without bootstrapping was also performed to explore the relationship of fish CC chemokines and mammalian CC chemokines.Altogether,orthologies were suggested for seven groups of fish CC chemokines. Potential orthologies were suggested for CCL20 including Fundulus CN976993,the trout CK8a and CK8b,and the catfish SCYA112.Similarly,orthologies were suggested for CCL24 with five fish CC chemokines,for CCL22 with eight fish CC chemokines,for CCL17 with at least four fish CC chemokines,for CCL25 with 15 fish CC chemokines,for CCL27/CCL28 with seven fish CC chemokines,and for CCL19/CCL21 with 17 fish CC chemokines.Chemokine data from fish is crucial not only for comparative immunology,but also in efforts to understand and prevent a number of fast-acting diseases prevalent among important aquaculture species.Ongoing work in fish CC chemokines will help better understand the important relationship between fish disease resistance and innate immunity.[Journal of Fishery Sciences of China,2006,13(1):119-127]
Cloning and bioactivity analysis of a CXC ligand in black seabream Acanthopagrus schlegeli : the evolutionary clues of ELR + CXC chemokines

Abstract:Background The ELR + CXC chemokines are multifunctional mediators that are mainly responsible for the recruitment of leucocytes to sites of inflammation and infection. Because of their high sequence identity with mammalian IL-8, fish IL-8-like CXCs have been named as piscine 'IL-8' and included in the ELR + subgroup, even though there is no reliable functional or evolutionary evidence to support this classification. Results In this investigation, a homologue of piscine 'IL-8' from black seabream ( Acanthopagrus schlegeli) , called BS CXC, has been cloned and analyzed. The results revealed that BS CXC has a high gene similarity and tertiary structure similarity with piscine and mammalian CXC chemokines, both ELR - CXC and ELR + CXC, although it has a lower identity with ELR - CXC, compared with ELR + CXC chemokines. Like other piscine IL-8, BS CXC has only an incomplete ELR motif, which is essential for the mammalian ELR + CXC ability to attract granulocytes. Bioactivity assay demonstrated that the BS rCXC produced in E. coli significantly stimulated migration of fish neutrophils and macrophages, but had no effect on rat neutrophils and macrophages, whereas hrIL-8 induced strong chemotaxis of fish neutrophils but did not affect fish macrophages. BS CXC seems show some structural and functional properties of the intermediate between ELR - CXC and ELR + CXC. Conclusion As an incomplete ELR + CXC chemokine from a modern fish, BS CXC provides some clues on the evolution from ancient ELR - CXC to ELR + CXC by retaining some properties of the intermediate stage in evolution, and it may be more appropriate to call this molecule 'piscine CXC with an incomplete ELR', instead of terming it fish 'IL-8'.
Modelling of FGM piezoelectric transducers using graded finite element concept

Abstract:Piezoelectric materials generate displacements when an electric potential is applied and, electric potential when they are subjected to forces or pressure. Functionally Graded Materials (FGM) are composite advanced materials, which are made by changing gradually the properties with position inside material domain. The application of FGM concept to piezoelectric transducer design allows designing composite transducers without interface between materials (e.g. PZT and Aluminum), due to the continuous change of property values. Thus, large improvements can be achieved, as reduction of stress concentration, increasing bonding strength and fatigue-lifetime. Recent works about piezoelectric FGM show lack of computational methods to model these transducers and evaluate their performance considering property gradation function, futhermore, FE comercial softwares have no “tools” to simulate graded continuous materials for while. Thus, this work proposes the development of Finite Element (FE) algorithms to model FGM piezoelectric ceramics and to explore the FGM potential on piezoelectricity field. The continuous change of piezoelectric, dielectric, and elastic properties is achieved by using the graded finite element concept, where these material properties are interpolated inside the finite element using the FE shape functions. A software based on 4-node graded finite element (Q4) is implemented. Dynamic and static analyses are performed. In the examples, the material properties are graded along thickness direction to illustrate the influence of gradation on the output displacements, vibration modes, and resonance frequencies. These examples are compared with results of a homogeneous piezoceramic to show FGM advantages.
Comparative transcriptomic analysis of the liver and spleen in marbled rockfish (Sebastiscus marmoratus) challenged with polyriboinosinic polyribocytidylic acid (poly(I:C))

Abstract:Marbled rockfish (Sebastiscus marmoratus) is a common coastal fish in China with considerable market value. The artificial culture of this species has not been widely popularized, and there have been few research on its vaccine or immune adjuvant. In this study, we performed transcriptome analysis of marbled rockfish spleen and liver tissue samples at 12 and 48 h after the polyinosinic-polycytidylic acid (poly(I:C)) challenge. Compared with the control group, 1067 and 598 differentially expressed genes (DEGs) were identified in the spleen and liver, respectively. GO, and KEGG enrichment analyses of DEGs revealed that some immune-related biological processes and pathways were significantly changed after poly(I:C) treatment, including primary immunodeficiency, the NF-kappa B signaling pathway, the Jak-STAT signaling pathway, and the PPAR signaling pathway. In conclusion, the findings of this research provide new insights into the immune response of marbled rockfish against poly(I:C) at the transcriptome level, and enrich the theoretical basis for the development of its immune adjuvant.
A novel calreticulin-related molecule that interacts with bacteria and enhances host resistance against bacterial infection in black rockfish, Sebastes schlegeli

Abstract:Calreticulin (CRT) is a highly conserved and multi-functional protein with diverse localizations. CRT has lectin-like properties and possesses important immunological activities in mammalian. In teleost, very limited studies on CRT immunologic function have been documented. In the present study, a CRT homologue (SsCRT) was cloned, identified and characterized from black rockfish, Sebastes schlegeli, an important aquaculture species in East Asia. The full length of SsCRT cDNA is 2180 bp and encoded a polypeptide of 425 amino acids. SsCRT contains a signal peptide, three distinct structural and functional domains (N-, P- and C-domains), and an endoplasmic reticulum (ER) retrieval signal sequence (KDEL). The deduced amino acid sequence of SsCRT shares 89-92% overall sequence identities with the CRT proteins of several fish species. SsCRT was distributed ubiquitously in all the detected tissues and was highly expressed in the spleen, muscle and liver. After the infection of fish extracellular bacterial pathogen Vibrio anguillarum and intracellular bacterial pathogen Edwardsiella tarda, the mRNA transcripts of SsCRT in spleen, liver, and head kidney were significantly up-regulated. The expression patterns were time-dependent and tissue-dependent. Recombinant SsCRT (rSsCRT) exhibited apparent binding activities against different bacteria and PAMPs. In vivo studies showed that the expressions of multiple immune-related genes such as TNF13B, IL-1β, IL-8, SAA, Hsp70, and ISG15 in head kidney were significantly enhanced when black rockfish were treated with rSsCRT. Furthermore, rSsCRT reduced pathogen dissemination and replication in fish kidney and spleen. These results indicated that SsCRT served as an immune receptor to recognize and eliminate the invading pathogens, which played a vital role in the immune response of Sebastes schlegeli. These findings provide new insights into understanding the roles of CRT proteins in immune response and pathogen infection in teleost.

Abstract:A C‐type lectin‐like protein (Ec‐ CTLP ) was cloned from the grouper Epinephelus coioides . The full‐length cDNA of Ec‐ CTLP was composed of 905 bp with a 522 bp open reading frame that encodes a 174‐residue protein. The putative amino acid sequence of Ec‐ CTLP contains a signal peptide of 19 residues at the N‐terminus and a CLECT domain from Cys43 to Arg169 and a conserved imperfect WND (Trp‐Asn‐Asp) motif. The homologous identity of deduced amino acid sequences is from 32 to 42% with other fishes. The expression of Ec‐ CTLP was differently upregulated in E. coioides spleen (germline stem) cells after being challenged at 16 and 4° C. Intracellular localization revealed that Ec‐ CTLP was distributed only in the cytoplasm. Recombinant Ec‐ CTLP ( rEc‐CTLP ) was expressed in Escherichia coli BL21 ( DE3 ) and purified for mouse Mus musculus anti‐Ec‐ CTLP serum preparation. The rEc‐CTLP fusion protein does not possess haemagglutinating activity, but improves survival from frozen bacteria. The survival of bacteria (including gram‐negative E. coli and gram‐positive Staphylococcus aureus ) was positively correlated with the concentration of the rEc‐CTLP . These findings can provide clues to help understand the probable C‐type lectin in marine fish innate immunity.

Abstract:The suppressors of cytokine signaling (SOCS) family are important soluble mediators to inhibit signal transduction via the Janus kinase/signal transducer and activator of transcription (JAK-STAT) pathway in the innate and adaptive immune responses. SOCS1 is the primary regulator of a number of cytokines. In this study, two spliced transcripts of SOCS1 were identified and characterized from black rockfish (Sebastes schlegeli), named SsSOCS1a and SsSOCS1b. SsSOCS1a and SsSOCS1b contained conserved structural and functional domains including KIR region, ESS region, SH2 domain and SOCS box. SsSOCS1a and SsSOCS1b were distributed ubiquitously in all the detected tissues with the higher expression level in liver and spleen. After stimulation in vivo with Vibrio anguillarum and Edwardsiella tarda, the mRNA expression of SsSOCS1a and SsSOCS1b were induced in most of the immune-related tissues, including head kidney, spleen and liver. Meanwhile, poly I:C and IFNγ up-regulated the expression of SsSOCS1a and SsSOCS1b that reached the highest level at 24 h in macrophages in vitro. Luciferase assays in HEK293 cells showed SsSOCS1a and SsSOCS1b had the similar function in inhibiting ISRE activity after poly I:C and IFNγ treatment. Furthermore, KIR domain in black rockfish was determined to have a negative regulatory role in IFN signaling. SsSOCS1a and SsSOCS1b were found to interact strongly with each other by Co-immunoprecipitation analyses. These results indicated that the function of SOCS1 in the negative regulation of IFN signaling is conserved from teleost to mammals which will be helpful to further understanding of the biological functions of teleosts SOCS1 in innate immunity.

Abstract:A CXCL13-like chemokine cDNA was isolated from large yellow croaker (Pseudosciaena crocea) by expressed sequence tag (EST) analysis (LycCXCL13) The full-length cDNA of LycCXCL13 is 796 nucleotides (nt) encoding a protein of 97 amino acids (aa), with a putative molecular weight of 10 7 kDa. The deduced LycCXCL13 contains a 24-aa signal peptide and a 73-aa mature polypeptide, which possesses the typical arrangement of four cysteines as found in other known CXC chemokines (C-25, C-27 C-52 and C-68) It shares 35, 36 and 39% aa sequence identities to green puffer CXCL13-like, Atlantic salmon CXCL13 and Japanese flounder CXCL13 chemokines, and 24-29% identities to CXCL13 chemokines in mammals, respectively Phylogenetic analysis showed that LycCXCL13 is more closely related to the CXCL13 subgroup than to any other CXC chemokine Subgroups. LycCXCL13 gene was constitutively expressed in all tissues examined, except for intestine. Genome-wide identification and characterization of Toll-like receptor genes in black rockfish (Sebastes schlegelii) and their response mechanisms following poly (I:C) injection

Abstract:Toll-like receptors (TLRs) are canonical transmembrane receptors that play an important role in defending against invading pathogens. In this study, we identified a total of 12 TLR genes in black rockfish (Sebastes schlegelii) with an analysis of their sequence characterizations. The phylogenetic analysis suggested that 12 distinct TLRs were grouped into five subfamilies (i.e., TLR1, TLR3, TLR5, TLR7, and TLR11 subfamilies), and each SsTLR gene respectively corresponded to the orthologs genes of other species. The protein domain analysis indicated that TLRs are type I transmembrane proteins, including an extracellular leucine-rich repeat (LRR), a transmembrane region (TM) domain and an intracellular Toll/IL-1 receptor (TIR) domain. The evolutionary ratios indicted that 12 SsTLRs were under purifying selection. qRT-PCR assays exhibited diverse TLRs molecular expression patterns in the heart, brain, head kidney, kidney, liver, intestine, and spleen of 3 black rockfish, and the expression levels were high in some immune tissues (e.g., head kidney, kidney, and spleen). Subsequently, 30 fish were equally divided into 2 groups i.e., poly (I:C)-treated and PBS-Control groups. After poly (I:C) injection, eight SsTLRs, i.e., SsTLR2, SsTLR2-1, SsTLR2-2, SsTLR3, SsTLR5S, SsTLR7, SsTLR8 and SsTLR22, were dramatically increased. Altogether these results contribute to understanding how SsTLRs respond to immune defense after poly (I:C) injection and provide researchers with comprehensive TLR gene family data of black rockfish.

Two CcCCL19bs Orchestrate an Antibacterial Immune Response in Yellow River Carp (cyprinus Carpio Haematopterus)

Abstract:Chemokines are a group of chemotactic cytokines with an essential role in homeostasis as well as immunity via specific G protein-coupled receptors and atypical receptors. In our study, two Yellow River carp (Cyprinus carpio haematopterus) CCL19b genes (CcCCL19bs), tentatively named CcCCL19b_a and CcCCL19b_b, were cloned. The open reading frames (ORFs) of CcCCL19b_a and CcCCL19b_b were both 333 bp that encoded a 12 kDa protein with 110 amino acid residues. CcCCL19bs contained a signal peptide and a SCY domain with four typical conserved cysteine residues. The two CcCCL19b proteins shared high similarities with each other in both secondary and three-dimensional structure. Phylogenetic analysis showed that CcCCL19bs and other CCL19bs from tetraploid cyprinid fish were clustered into one clade. CcCCL19bs were highly expressed in gill and intestine in healthy fish, and a significant up-regulation of gene expression after Aeromonas hydrophila infection and poly(I:C) stimulation was observed in gill, liver, and head kidney. Furthermore, chemotaxis and antibacterial activity of CcCCL19bs were studied. The results indicated that recombinant CcCCL19b_a and CcCCL19b_b protein (rCcCCL19b_a and rCcCCL19b_b) exhibited significant attraction to primary head kidney leukocytes (HKLs). Meanwhile, both of rCcCCL19bs could promote the proliferation of HKLs, and significantly up-regulate the expressions of IL-1β, CCR7, and IL-6, and down-regulate the expression of IL-10 in primary HKLs. In vitro, rCcCCL19bs could bind and aggregate A. hydrophila and Staphylococcus aureus. The rCcCCL19bs exhibited significant antibacterial activity against A. hydrophila, but not S. aureus. Moreover, they inhibited the growth of A. hydrophila and S. aureus. In vivo, overexpression of CcCCL19bs contributed to the bacterial clearance. These studies suggested that CcCCL19bs orchestrate an antibacterial immune response.
Molecular characterization, structural and expression analysis of twelve CXC chemokines and eight CXC chemokine receptors in largemouth bass (Micropterus salmoides)

Abstract:The chemokine-receptor system plays important roles in the leukocyte trafficking, inflammation, immune cell differentiation, cancer and other biological processes. In the present study, the sequence features, structures and expression patterns of twelve CXC chemokine ligands (CXCL8a.1, CXCL8a.2, CXCL8b.1, CXCL8b.2, CXCL12a, CXCL12b, CXCL13.1, CXCL13.2, CXCL14, CXCL18a, CXCL18b and CXCL19) and eight CXC chemokine receptors (CXCR1, CXCR2, CXCR3.1, CXCR3.2, CXCR3.3, CXCR4a, CXCR4b and CXCR5) of largemouth bass (Micropterus salmoides) were analyzed. All the CXCLs and CXCRs of largemouth bass shared high sequence identities with their teleost counterparts and possessed conserved motifs and structures of CXCLs and CXCRs family. Realtime qPCR revealed that these CXCLs and CXCRs were ubiquitously expressed in all examined tissues, with high expression levels in the immune-related tissues (spleen, head kidney, and gill). Following lipopolysaccharide (LPS) and polyinosinic-polycytidylic acid (polyI:C) stimulations, most of these CXCLs and CXCRs were significantly up-regulated in spleen. In addition, the potential interacted molecules of these CXCLs and CXCRs were analyzed by protein-protein interaction network analysis. To the best of our knowledge, this is the first study that in detail analyzes the CXCLs and CXCRs of largemouth bass. Our results provide valuable basis for study the function and mechanism of chemokine-receptor system in largemouth bass.
CCL19 variants mediate chemotactic response via CCR7 in grass carp Ctenopharyngodon idella

Abstract:CC chemokine ligand 19 (CCL19) plays a key role in the regulation of immune responses including homeostasis, inflammation, and immune tolerance. In this study, two variants of CCL19 homologues (CCL19a2 and CCL19b) and CCR7 were investigated in grass carp Ctenopharyngodon idella. The three genes were widely expressed in immune tissues and could be modulated by stimulation with LPS, PHA and poly(I:C), and infection with Flavobacterium columnare and grass carp reovirus. In an in vitro chemotaxis assay, the recombinant CCL19a2 and CCL19b were active to promote the migration of HEK293 T cells expressing CCR7 and leucocytes isolated from the gills, head kidney and spleen. Moreover, their chemotactive effects were validated in vivo. We found that the cells recruited by CCL19a2 and CCl19b are mainly monocytes/macrophages expressing high levels of IL-1β, IFN-γ, colony stimulating factor 1 receptor (CSF1R) and MHC II. Our work suggests that CCL19a2 and CCl19b are involved in recruitment of antigen presenting cells in fish.
A CXCL ortholog from Hippocampus abdominalis: Molecular features and functional delineation as a pro-inflammatory chemokine

Abstract:Chemokines are a family of chemotactic cytokines that regulate leukocyte migration. They are classified into four groups namely, CXC, CC, C and CX3C, based on the formation of a disulfide bridge. Among these, CXC chemokines have been identified as the largest group of chemokines in humans. In this study, we identified and functionally characterized a homolog of CXC chemokine from the big-belly seahorse, Hippocampus abdominalis, and designated it as ShCXCL. The cDNA of ShCXCL composed of a 342-bp open reading frame encoding 113 amino acids (aa). The CXC family-specific small cytokine domain (SCY) was identified from the mature peptide region, which comprised of a conserved CXC motif. As ShCXCL lacks an ELR (Glutamic acid-Leucine-Arginine) motif, it belongs to ELR- subfamily. The recombinant ShCXCL protein strongly induced the nitric oxide (NO) production in macrophage cells (RAW 264.7 cell line) and showed the chemotactic effect on flounder peripheral blood leukocytes. Tissue profiling showed a ubiquitous expression pattern in all examined tissues, with a high abundance in spleen. The up-regulated mRNA expression pattern of ShCXCL was observed in blood and kidney tissues after immune stimulation by live bacteria, such as Streptococcus iniae and Edwardsiella tarda, and mitogens, such as lipopolysaccharides (LPS) and polyinosinic:polycytidylic acid (poly I:C), suggesting its important role in host immune defense against microbial infection.
Molecular characterization and expression analysis of Japanese flounder (Paralichthys olivaceus) chemokine receptor CXCR2 in comparison with CXCR1

Abstract:Chemokines are categorized into five families; one of the families is the CXC chemokines, which are critical in the pro-inflammatory process. CXC chemokines transmit signals and mediate a cell's biological activities by binding to cell surface receptors known as chemokine receptors (CXCRs). In this study, the CXCR2 from Japanese flounder (Paralichthys olivaceus) (JfCXCR2) was identified and characterized at the molecular level. The JfCXCR2 gene has a 1077 bp open reading frame that encodes a protein of 359 amino acid residues with seven transmembrane domains. Phylogenetic analysis of JfCXCR2 revealed that it belonged to the fish CXCR2 subfamily. Furthermore, JfCXCR2 was compared with the previously identified Japanese flounder CXCR1 (JfCXCR1). The expression analysis of uninfected Japanese flounder showed that JfCXCR1 and JfCXCR2 were expressed in all the tissues and organs tested but mainly in immune-related organs, including the kidney and spleen. Infection by Streptococcus iniae significantly increased the level of JfCXCR1 and JfCXCR2 mRNA in the kidney at days 1 and 3 post-infection On the other hand, VHSV (viral hemorrhagic septicemia virus) and Edwardsiella tarda infection significantly increased JfCXCR2 mRNA levels in the kidney at days 3 and 6 post-infection, respectively. Conversely, JfCXCR1 expression was not significantly changed by either E. tarda or VHSV infection. Additionally, the peripheral blood leukocytes (PBLs) stimulated by recombinant proteins rCXCL8_L1a and rCXCL8_L1b were found to have significantly increased levels of JfCXCR1 and JfCXCR2 mRNA. Interestingly, even higher levels of JfCXCR1 and JfCXCR2 expression were observed in PBLs stimulated with rCXCL8_L1a and rCXCL8_L1b than in PBLs stimulated with either recombinant protein. These data suggest that bacterial infections may activate JfCXCR1. By contrast, JfCXCR2 may be activated by both bacterial and viral infection to mediate the immune response. These data can contribute to further understanding the functions of CXCR1 and CXCR2 in the fish immune system.

Abstract:Chemokines are cytokines that mediate leukocyte traffic between the lymphoid organs, the bloodstream, and the site of tissue damage, which is essential for an efficient immune response. In particular, the gamma interferon (IFN- γ) inducible chemokines CXCL9, CXCL10, and CXCL11, and their receptor CXCR3, are involved in T cell and macrophage recruitment to the site of infection. The nature and function of these chemokines and their receptor are well-known in mammals, but further research is needed to achieve a similar level of understanding in fish immunity. Thus, in this study, we seek to identify the genes encoding the components of the Atlantic salmon (Salmo salar) CXCL9, CXCL10, CXCL11/CXCR3 axis (CXCL9-11/CXCR3), predict the protein structure from the amino acid sequence, and explore the regulation of gene expression as well as the response of these chemokines and their receptor to viral infections. The cxcl9, cxcl10, cxcl11, and cxcr3 gene sequences were retrieved from the databases, and the phylogenetic analysis was conducted to determine the evolutionary relationships. The study revealed an interesting pattern of clustering and conservation among fish and mammalian species. The salmon chemokine sequences clustered with orthologs from other fish species, while the mammalian sequences formed separate clades. This indicates a divergent evolution of chemokines between mammals and fish, possibly due to different evolutionary pressures. While the structural analysis of the chemokines and the CXCR3 receptor showed the conservation of critical motifs and domains, suggesting preserved functions and stability throughout evolution. Regarding the regulation of gene expression, some components of the CXCL9-11/CXCR3 axis are induced by recombinant gamma interferon (rIFN-γ) and by Infectious pancreatic necrosis virus (IPNV) infection in Atlantic salmon cells. Further studies are needed to explore the role of Atlantic salmon CXCL9-11 chemokines in regulating immune cell migration and endothelial activation, as seen in mammals. To the best of our knowledge, there have been no functional studies of chemokines to understand these effects in Atlantic salmon.
Functional Characterization of the ELR Motif in Piscine ELR + CXC-Like Chemokine

Abstract:To elucidate the functional role of piscine incomplete ELR motif, the recombinant CXC and its mutants (mELR and mLoop) were produced in Escherichia coli M15 based on the predicted mature peptide coding sequence of the black sea bream CXC (BS CXC) chemokine. Assays showed that the BS rCXC proteins displayed strong ability to induce fish blood neutrophils and head kidney (HK) macrophage migration in a dose-independent manner (10 to 200 ng), both in black sea bream and common carp. Although the ELR motif and the N-terminal loop of ELR + CXC chemokines are essential for chemotactic activity and receptor binding in mammals, the mELR and mLoop mutants showed no significant difference in their induction of chemotaxis of fish blood neutrophils compared with the full-length rCXC at the same dose. Human recombinant IL-8 (hrIL-8) can clearly induce piscine blood neutrophil migration and has no effect on macrophages, whereas the BS rCXC cannot induce chemotaxis in higher vertebrates, such as rat blood neutrophils or macrophages, even if the incomplete ELR motif in rCXC was mutated to ELR. The BS CXC and its mutants can promote the phagocytosis ability of piscine blood neutrophils and HK macrophages both in black sea bream and common carp, but have no effect on rat neutrophils or macrophages. Results showed that the piscine ELR + CXC-like chemokine represents an ancient version of a CXC chemokine; the ELR motif still does not show the higher specific polarization of function as found in mammalian.

Abstract:In 1997, the Royal College of Paediatrics and Child Health (RCPCH) published a framework for practice on the withholding or withdrawing of life sustaining treatment. Based on sound ethical and legal principles, the purpose of the document was to oVer guidance for those faced with diYcult treatment decisions that could not be resolved by appeal to scientific fact alone. 1 Unsurprisingly, the RCPCH document has attracted both acclaim and some criticism for its general objectives and specific conclusions. 23 Any attempt to provide such guidance might be interpreted as applying unnecessary constraints to clinical practice. Furthermore,such documents can be criticised as being too general to be useful, stigmatising to some individuals or groups, and striking the wrong balance between law and morality. Although these criticisms have some validity, questions such as those dealt with by the RCPCH document and others cannot simply be left to the moral values of individual clinicians or their (possibly mistaken) interpretation of the law. 4 Some considered advice—informed by appropriate collective multidisciplinary deliberation—is imperative. It seems likely, therefore, that other groups will wish to provide guidance or advice on ethicolegal matters.
