Abstract:Exopolysaccharide (EPS) produced by Paenibacillus polymyxa PYQ1 may be a promising agent used in the cosmetic field for skin care because of its protective benefits against short-wave ultraviolet (UVC, 100-280 nm) radiation, which induces cytotoxicity in HaCaT cells. We attempted to use low-intensity ultrasound to enhance its fermentation, and the yield of EPS increased by 24 % under the optimum ultrasonic treatment (90 W, 45 s in 9 h). Results of further experiments show that low-intensity ultrasound will kill a few bacterial cells. Therefore, the remaining bacterial cells have more space and more nutrients, growing faster and producing EPS more effi-ciently. The recoverable physical damage caused by ultrasound increased the cell permeability, and for the first time, we found that low-intensity ultrasound up-regulated the expression of membrane transporters involved with the transport of sucrose and polysaccharides. Owing to these changes, the utilization of carbon sources was accelerated, and the transport of EPS was promoted to the outside of bacterial cells such that the production of EPS was enhanced.
A New Strategy to Improve Ganoderma Polysaccharides Production by Symbiotic Fungi Elicitors Through Activating the Biosynthetic Pathway

Abstract:Ganoderma lucidum polysaccharides (GLP) attract growing attention due to their remarkable bioactivities, but the low content in raw materials remains a bottleneck severely restricting their application. We previously found a higher polysaccharides accumulation in Ganoderma lucidum cultured in continuous cropping soil, and soil symbiotic fungi are presumed as the key among many factors. Herein, 33 symbiotic fungi were isolated from the soil, and fungal elicitors were prepared to investigate their biotic eliciting effect on GLP biosynthesis. Most elicitors were found to significantly improve GLP production, among which the NO.16 molecularly identified as Penicillium citrinum, exhibited the optimum eliciting effect with GLP yield increasing by 3.4 times. Differences in the biosynthetic pathway genes expressions and the monosaccharide components of GLP were further analyzed. The transcriptions of the main genes of GLP biosynthetic pathway were up-regulated under PCE treatments, suggesting it improves GLP production by activating transcriptions of the biosynthetic pathway genes. Moreover, PCE eliciting significantly altered the monosaccharide compositions of GLP with Gal, Man, GalA, GlcA, and Fuc increasing by 8.17 %, 5.68 %, 5.41 %, 2.66 %, and 1.51 % respectively, but Glc decreased by 23.43 %, which may result in the activity change. It can serve as a new strategy to improve GLP production.

Abstract:Agaricus bitorquis (Quel.) Sacc. Chaidam (ABSC) is a wild edible fungus uniquely found in the Tibet Plateau. ABSC is rich in polysaccharides that are considered biologically active. This study aimed to determine the feasibility of enhancing exopolysaccharide (EPS) production by ABSC in shake flask culture by supplementing the fermentation medium with anthocyanin extract. Different concentrations of Lycium ruthenicum Murr. (LRM) anthocyanin crude extract were tested on ABSC fermentation. The activity of phosphoglucose isomerase (PGI), phosphoglucose mutase (PGM), and phosphomannose isomerase (PMI), enzymes presumably involved in EPS synthesis by ABSC, was determined. ABSC transcriptomic profile in response to the presence of anthocyanins during fermentation was also investigated. LRM anthocyanin crude extract (0.06 mg/mL) was most effective in increasing EPS content and mycelial biomass (by 208.10% and 105.30%, respectively, P < 0.01). The activity of PGI, PGM, and PMI was increased in a medium where LRM anthocyanin extract and its main components (proanthocyanidins and petunia anthocyanin) were added. RNA-Seq analysis showed that 349 genes of ABSC were differentially expressed during fermentation in the medium containing anthocyanin extract of LRM; 93 genes were up-regulated and 256 genes down-regulated. From gene ontology enrichment analysis, differentially expressed genes were mostly assigned to carbohydrate metabolism and signal transduction categories. Collectively, LRM anthocyanins extract positively affected EPS production and mycelial biomass during ABSC fermentation. Our study provides a novel strategy for improving EPS production and mycelial growth during ABSC liquid submerged fermentation.

Abstract:Schizophyllum commune (S. commune) polysaccharides are biomacromolecules with multiple biological activities and wide applications. In this study, polysaccharide production through submerged fermentation of S. commune using different surfactants was investigated. The addition of 1 g/L of polyoxyethylene sorbitan monooleate (Tween 80) at the beginning of the fermentation showed the best promotional effects on collective exopolysaccharide (EPS) production (which increased by 37.17%) while shortening the production cycle by 2 days. The monosaccharide composition of the EPS produced when the added Tween 80 was similar to that of the control; however, the molecular weight (Mw) was lower. Notably, the addition of Tween 80 significantly increased the ATP levels and the transcription levels of phosphoglucomutase and beta-glucan synthase genes in the polysaccharide synthesis pathway. The addition of Tween 80 reduced the pellet size of the mycelium compared to that of the control, but did not significantly change the microstructure of the mycelial cells. This study proposes an efficient strategy for the production of polysaccharides through submerged fermentation of S. commune, and elucidates the detailed mechanism of using Tween 80 as a fermentation stimulatory reagent.

Abstract:Armillaria luteo-virens Sacc. is a special medicinal and edible mushroom in China, and its wild type has been investigated that it contains some functional components. The optimization of submerged culture conditions for mycelium growth and exo-polysaccharide (EPS) production in an edible mushroom A. luteo-virens Sacc. was studied in shake flasks and bioreactors with response surface methodology (RSM). A fractional factorial design (FFD) and central composite design (CCD) were applied to optimize the main factors that affect the fungal polysaccharides production in a submerged culture. Glucose is very important for EPS production by A. luteo-virens Sacc., and yeast extract powder also significantly affects fungal EPS production through fractional factorial experiment. The optimized cultivation medium constituents were derived from the predicted model using central composite design as follows: 31.26g/l glucose, 1.06g/l yeast extract powder, 1.00g/l K2HPO4, 0.50g/l KH2PO4, and 0.45g/l MgSO4·7H2O, pH unadjusted. The predicted maximal EPS concentration was 5.40g/l using the optimized culture medium compositions at 23°C for 5 days. Additionally, the submerged cultivation process was explained using the optimized cultivation medium. The cultivation kinetics model constructed was suitably depicting the culture process in bioreactor cultivation scale.

Abstract:Ganoderma lucidum has been a well-known species of basidiomycetes for a long time, and has been widely applied in the fields of food and medicine. Based on the simulation results of model iZBM1060 in our previous research, the effect of L-phenylalanine on G. lucidum extracellular polysaccharides (EPSs) was investigated in this study. EPS production reached 0.91 g/L at 0.4 g/L L-phenylalanine after a 24 h culture, which was 62.5% higher than that of control (0.56 g/L). Transcriptome and genome analysis showed that L-phenylalanine deaminase and benzoate 4-hydroxylase (related to L-phenylalanine metabolism) were significantly up-regulated, while the cell wall mannoprotein gene was down-regulated. Transmission electronic microscopy (TEM) and atomic force microscopy results showed that the cell wall thickness decreased by 58.58%, and cell wall porosity increased in cells treated with L-phenylalanine, which probably contribute to the increasing EPS production. This study provides an efficient strategy for fungal polysaccharide production with high output and low cost.

Abstract:To promote Agaricus sinodeliciosus var. Chaidam ZJU-TP-08 growth and metabolites accumulation, a novel integrated strategy was developed by adopting high levels of metal ions coupled with light treatment. The results revealed that yellow and blue light could significantly promote biomass and exopolysaccharides production, respectively. Furthermore, the yellow–blue light shift strategy could stimulate exopolysaccharides formation. Ca2+ ions coupled with blue light mostly promoted exopolysaccharides production related to oxidative stress, which was 42.00% and 58.26% higher than that of Ca2+ ions coupled with the non-light and dark cultivation without Ca2+ ions in 5-L bioreactor. RNA-seq was performed to uncover the underlined molecular mechanism regulated by light-induced gene expressions in exopolysaccharides biosynthesis and oxidative stress. The findings of this work provide valuable insights into adopting metal ions coupled with the light-assisted method for the macrofungus submerged fermentation for exopolysaccharides production.

Abstract:This work aimed to develop the submerged cultivation conditions for improved exopolysaccharides (EPS) production by Armillaria luteo-virens Sacc. The effects of culture temperature, aeration rate, inoculum level, initial pH, and additives on EPS formation and mycelial growth are investigated. The aeration rate, initial pH, and inoculum level significantly affected EPS production under the submerged cultivation. The developed conditions were as follows: cultivation temperature 23 °C, initial pH 5.0, aeration rate 0.5 vvm, 0.5% Tween 80, inoculum level 5% (v/v), and shaking speed 120 r/min. Under the developed conditions, the highest EPS production was 13.01 g/L at 5 days culture time. EPS production was examined in a 5 L bioreactor, and an unstructured kinetic model for EPS formation was well developed. The verified investigations in the large-scale cultivation system showed that the developed models are able to predict the submerged cultivation process of EPS formation. Current results revealed that the submerged cultivation conditions can be utilized to control EPS production, and the unstructured models developed are suitable for explaining EPS production by A. luteo-virens Sacc QH in a large-scale cultivation bioreactor.
Efficient Delignification of Sugarcane Bagasse by Fenton Oxidation Coupled with Ultrasound-Assisted NaOH for Biotransformation from Agaricus Sinodeliciosus Var. Chaidam

Abstract:Sugarcane bagasse (SCB), a widely available lignocellulosic by-product, could be potentially used in macrofungal bioconversion for high-value bioactive compounds. For macrofungus with poor lignin degradation capacity, its recalcitrant lignin structure blocked the release of fermentable sugars for cell growth. Herein, the main objective was to construct an effective delignification pretreatment strategy for SCB biotransformation and synthesis of bioactive substances by Agaricus sinodeliciosus var. Chaidam. Different pretreatments including chemical (alkali and Fenton oxidation) and ultrasound methods were used to intensify the delignification process. Physico-chemical characteristics of native and pretreated SCB were examined via XRD, XPS, FT-IR and C-13 ssNMR to characterize crystallinity index, surface lignin and chemical structure. Lignocellulose-degrading enzymes were also used to assess the ability of the macrofungus to degrade SCB. By comparing morphological structure, chemical composition and fermentation characteristics, a suitable multiple sequential pretreatment method was finally identified. Our findings revealed that Fenton-ultrasound-alkali pre-treatment has an optimum delignification effect on SCB with never-uncovered excellent ability to detoxify harmful substances produced by Fenton oxidation. Under this pretreatment condition, the cellulose content was increased by 106.45%, whereas hemicelluloses and lignin were decreased by 48.34% and 74.15%, respectively. The macrofungal growth, intracellular polysaccharides, total phenols and terpenoids formation were 2.4, 1.46, 1.36 and 1.66 times higher than those of the untreated, respectively. Fermentation kinetic results showed outstanding fit and prediction of macrofungal growth and enzymatic activity secretion on above pretreated SCB. This study shed light on developing a novel pretreatment strategy for low-cost SCB lignocellulose transformation into high-value products.

Abstract:Three kinds of water-soluble polysaccharides (PS, PFB and PM) were isolated from the sporocarp, fermentation broth and cultured mycelium of wild mushroom Agaricus bitorquis (Quél.) Sacc. Chaidam (ABSC). The Mw of PS, PFB and PM ranged from 893 Da to 38,340 Da. FTIR and NMR spectroscopy indicated that the skeleton of PS-I in PS contained α-D-mannopyranose with a backbone of (1 → 4)-α-D-glycosidic. The isolated fraction PFB-I in PFB was connected by (1 → 6) or (1 → 4)-α-D-glycosidic linkage. PM-I in PM was connected by (1 → 4)-α-D-glucopyranosyl residues with branched chain connected by (1 → 6)-α-glycosidic linkage. The anti-hypoxia activity results in vivo revealed that the supplementation with PS, PFS and PM could extend the survival time of mice under hypoxia stress, and increase GSH-Px, CAT and SOD activities and protein levels in liver tissues of mice, decrease MDA and NO levels. The findings suggested that such polysaccharides from ABSC showed anti-hypoxia activity.

Abstract:Abstract Background Polysaccharides are important active ingredients in Ophiocordyceps gracilis with many physiological functions. It can be obtained from the submerged fermentation by the anamorph ( Paraisaria dubia ) of Ophiocordyceps gracilis . However, it was found that the mycelial pellets of Paraisaria dubia were dense and increased in volume in the process of fermentation, and the center of the pellets was autolysis due to the lack of nutrient delivery, which extremely reduced the yield of polysaccharides. Therefore, it is necessary to excavate a fermentation strategy based on morphological regulation for Paraisaria dubia to promote polysaccharides accumulation. Results In this study, we developed a method for enhancing polysaccharides production by Paraisaria dubia using microparticle enhanced technology, talc microparticle as morphological inducer, and investigated the enhancement mechanisms by transcriptomics. The optimal size and dose of talc were found to be 2000 mesh and 15 g/L, which resulted in a high polysaccharides yield. It was found that the efficient synthesis of polysaccharides requires an appropriate mycelial morphology through morphological analysis of mycelial pellets. And, the polysaccharides synthesis was found to mainly rely on the ABC transporter-dependent pathway revealed by transcriptomics. This method was also showed excellent robustness in 5-L bioreactor, the maximum yields of intracellular polysaccharide and exopolysaccharides were 83.23 ± 1.4 and 518.50 ± 4.1 mg/L, respectively. And, the fermented polysaccharides were stable and showed excellent biological activity. Conclusions This study provides a feasible strategy for the efficient preparation of cordyceps polysaccharides via submerged fermentation with talc microparticles, which may also be applicable to similar macrofungi. Optimization of Submerged Culture for Exopolysaccharides Production by Morcella esculenta and its Antioxidant Activities In Vitro

Abstract:Morcella esculenta is an excellently edible and delicious morel mushroom found growing in China, Japan, and other Asian countries. Polysaccharides extracted from fungi have shown a variety of medical activities; however, the industrial large-scale production of Morcella esculenta can not be achieved now due to the natural conditions. So this paper is concerned with optimization of submerged culture conditions for mycelial growth and exopolysaccharides (EPS) production of Morcella esculenta. by airlift fermentation. And the antioxidant activities of EPS were revealed by various free radical scavenging assays in vitro. The optimal medium constituents were determined as follows: (g/L): glucose 20 g/L, peptone 4.5 g/L, KH2PO4 0.8 g/L, and MgSO4·7H2O 1.0 g/L. The optimum parameters of liquid culture were temperature 25 °C, cultivation time 5 d, rotary speed 140 rpm, and initial pH 6.0. This optimization strategy in shake flask culture leads to a mycelia yield of 4.93 ± 0.28 g/L, and EPS production of 1.65 ± 0.28 g/L, respectively. Under optimal culture conditions, the maximum mycelia and EPS concentration in a 7 L airlift tower loop reactor were 6.13 g/L and 2.12 g/L for the first report, which were considerably higher than those obtained in preliminary studies that indicated the EPS production was closely correlated to the mycelia growth by Morcella esculenta. Futuremore, the EPS demonstrated positively antioxidant potential on 1,1-diphenyl-2-picrylhydrazyl radical scavenging, reducing power, and hydroxyl radical scavenging. These findings may provide the basis for the popular use of EPS in functional food or medicine.
Study on the Microbial Mechanisms of Enhancing Agaricus Bisporus Growth Through Inoculation with Pseudomonas Putida

Abstract:Introducing Pseudomonas putida optimizes the microbial community within the casing layer, thereby enhancing Agaricus bisporus growth. Nevertheless, the underlying mechanisms governing microbial community succession within the casing layer following microbial agent introduction remained elusive. This study delineated the interplay among microbial populations in the casing layer across the growth cycle of A. bisporus, comparing cohorts treated with P. putida (AP) against untreated controls (CK). The inoculation with Pseudomonas promoted the growth of A. bisporus, shortened its growth cycle, and linearly enhanced the relative abundance of key bacterial genera integral to its growth, including Flavobacterium, Rhodoferax, Pseudarthrobacter, and Vogesella (adjusted R-2 = 0.238-0.866). Importantly, a positive correlation was identified between the proliferation of these beneficial bacteria and positive community cohesion (adjusted R-2 = 0.353-0.838). Additionally, analysis of the symbiotic network topology within the AP group indicated a more stable community following inoculation. The niche results (AP = 11.7672 +/- 1.8938, CK = 11.0481 +/- 3.1273, P < 0.05) underscored the positive impact of Pseudomonas inoculation on ecosystem dynamics. Concentrations of essential nutrients for A. bisporus growth, notably readily available phosphorus (On day 17, the AP group was 78.8333 +/- 1.26623 mg/kg, while the CK group was 30.0333 +/- 0.98658 mg/kg) and readily available potassium (On day 17, the AP group was 2693.9 +/- 14.79561 mg/kg, while the CK group was 1646.9333 +/- 10.71276 mg/kg), were significantly elevated in the AP group. These enhancements strongly correlated with the increased abundance of pivotal microbial populations. Our research provided insights into the application of microbial inoculants to enhance the production of edible fungi.

Abstract:This study aims to develop a fermentation method instead of fruiting body extraction for Cordyceps polysaccharide (CP). The fermentation characteristics of P. dubia at different pH were inspected. The mechanism of microcycle conidiation of P. dubia was activated under pH 9.0 and produced yeast-like spores. A pH-shift strategy was raised based on the analysis of fermentation kinetics parameters. And, yeast-like spores were used as fermentation seeds to increase culture density. To further improve efficiency, a fed-batch fermentation with a pH-shift strategy was executed, CP production reached 13.54 g/L, with the productivity of 0.42 g/(L d), and the specific rate of CP to biomass was increased to 0.27, which improved by 47.6 %, 64.3 % and 97.1 % respectively, compared with fermentation without process controls. And, CP from fermented and extracted had the same monosaccharide composition except for differences in molecular weight. This study provides a feasible strategy for the efficient preparation of CP through spores fermentation.

Abstract:A medicinal mushroom Fomes fomentarius, was isolated from the fruiting body of a wild F. fomentarius and identified by ITS-5.8S rDNA sequencing analysis. Then, the optimization of submerged culture conditions and nutritional requirements of mycelial biomass and exopolysaccharide (EPS) production from F. fomentarius was studied using orthogonal matrix method. Under the optimal culture condition, the maximum EPS concentration reached 3.64 g l(-1), which is about four times higher than that at the basal medium. Furthermore, the EPS from F. fomentarius has a direct antiproliferative effect in vitro on SGC-7901 huaman gastric cancer cells in a dose- and time-dependent manner. Moreover, it was about three times that EPS at noncytocxity concentration of 0.25 mg ml(-1) could sensitize doxorubicin(Dox)-induced growth inhibition of SGC-7901 cells after 24h treatment.
Optimization of Effect Factors for Mycelial Growth and Exopolysaccharide Production by Schizophyllum Commune

Abstract:This paper is concerned with the optimization of effect factors for mycelial growth and exopolysaccharide production by Schizophyllum commune by one-factor-at-a-time and orthogonal methods. The one-factor-at-a-time method was adopted to investigate the effects of six different compounds (sodium carboxymethylcellulose, l -glutamic acid, V B1 , naphthalene acetic acid, oleic acid, and Tween 80) on mycelial growth and exopolysaccharide production. Among these factors, oleic acid, V B1 and Tween 80 were identified to be the most important factors. Subsequently, the concentration of oleic acid, V B1 and Tween 80 were optimized using the orthogonal matrix method. The effects of the factors on the mycelial growth of S. commune were in the order of oleic acid > V B1 > Tween 80, and those on exopolysaccharide production were in the same order. The optimal concentration for mycelia and exopolysaccharide were determined as oleic acid 0.1% ( v / v ), V B1 0.5 mg/L, and Tween 80 6 mg/L. The subsequent verification experiments confirmed the validity of the models. Under this optimized conditions in shake flask culture, the mycelial yield and exo-biopolymer production were 25.93 and 2.79 g/L, respectively, which were considerably higher than those obtained in the preliminary studies. The result was further confirmed in a 7-L fermentor experiments.

Abstract:Exopolysaccharides (EPS) of fungal origin have attracted special attention from researchers due to their multifarious applications in the food and pharmaceutical industries. In the present study, optimization of the process parameters for the production of exopolysaccharide by Schizophyllum commune AGMJ-1 was studied using one factor at a time (OFAT) method, Plackett–Burman design (PBD) and response surface methodology (RSM). OFAT method revealed xylose and yeast extract to be the most effective carbon and nitrogen sources and pH 5.3 as an optimum for maximum EPS production. Xylose, yeast extract and KCl were screened as statistically significant variables for EPS production using PBD. RSM based on the central composite design estimated that maximum EPS (4.26 g L−1), mycelial biomass (14 g L−1) and specific yield (0.45 g g−1) were obtained when concentration of xylose, yeast extract and KCl were set at 2.5 g % (w/v), 0.83 g % (w/v) and 6.53 mg % (w/v), respectively, in the production medium.

Identification of Growth-Promoting Bacterial Resources by Investigating the Microbial Community Composition of Polyporus umbellatus Sclerotia

Abstract:The sclerotium of the edible mushroom Polyporus umbellatus (Zhuling) exhibits various medicinal properties. However, given its long growth cycle and overexploitation, wild resources are facing depletion. Macrofungal growth depends on diverse microbial communities; however, the impact of soil bacteria on P. umbellatus development is unknown. Here, we combined high-throughput sequencing and pure culturing to characterize the diversity and potential function of bacteria and fungi inhabiting the P. umbellatus sclerotium and tested the bioactivities of their isolates. Fungal operational taxonomic units (OTUs) were clustered and classified, revealing 1275 genera. Bacterial OTUs yielded 891 genera. Additionally, 81 bacterial and 15 fungal strains were isolated from P. umbellatus sclerotia. Antagonism assays revealed three bacterial strains (FN2, FL19, and CL15) promoting mycelial growth by producing indole-3-acetic acid, solubilizing phosphate, and producing siderophores, suggesting their role in regulating growth, development, and production of active compounds in P. umbellatus. FN2-CL15 combined with bacterial liquid promoted growth and increased the polysaccharide content of P. umbellatus mycelia. This study reports new bioactive microbial resources for fertilizers or pesticides to enhance the growth and polysaccharide accumulation of P. umbellatus mycelia and offers guidance for exploring the correlation between medicinal macrofungi and associated microbial communities.
Identification of Anamorph Yeast of Tremella aurantialba and Optimization of Medium Composition for Production of Exopolysaccharides

Abstract:A yeast-like fungus strain B1 isolated from wild fungus Tremella aurantialba was identified and initially characterized. Two phylogenetic trees were generated based on the sequences of large subunit ribosomal RNA gene D1/D2 regions and internal transcribed spacer (ITS) regions of related fungi, respectively. The analysis of D1/D2 regions and ITS sequences showed that fungus B1 was clustered together with T. aurantialba, T. aurantia and T. microspore in the phylogenetic trees. Both the morphological characteristic and phylogenetic analysis established that fungus B1 was one of the anamorph strains of T. aurantialba and belongs to Tremella genus. A fermentation medium for exopolysaccharides (EPS) production by T. aurantialba B1 . Plackett–Burmen design was used to evaluate the effects of different components in the culture medium. Glucose and yeast extract have significant influence on the EPS production. The concentrations of two factors were optimized subsequently using central composite design and response surface analysis. The results showed that 49.2 g/L glucose and 10.4 g/L yeast extract could lead to the maximum production of EPS (4.99 g/L). The optimized medium led to a 1.5-fold enhancement of the production of EPS by T. aurantialba B1 , as compared with that without optimization.
