Abstract:The non-judicious use of pesticides in agro-food poses a severe threat to food safety and human health. As an emerging chromatographic fingerprint provider, surface-enhanced Raman spectroscopy analysis (SERS) sheds bright light on sensitive and nondestructive detection of pesticide residues. This research proposed a novel strategy to detect three-pesticide residues (thiabendazole, carbendazim, and chlorpyrifos) on tomato peel based on the flexible and sticky SERS substrate. After selecting the best commercial adhesive tape (3M9080), the SERS substrate was constructed by optimizing the parameters in the preparation process of AuNPs. Therefore, a new simple “tape-wrapped SERS” way for pesticide residue analysis was established with a simple procedure of “absorption, separation, and drop addition.” Based on chemometrics method, the limit of semiquantitative detection was 20, 36, and 80 ng/cm2 for thiabendazole, carbendazim, and chlorpyrifos, respectively, on tomato surface, which indicated that the proposed method could meet the requirement of actual application with a large prospect in agro-food safety detection.

Abstract:The overuse of mixed pesticides poses a threat to both the ecosystem and human health, yet general pesticide detection methods are complex in terms of steps, such as the need for pretreatment and cleaning. Surface-enhanced Raman scattering (SERS) has been used for real-life trace detection of residues due to its rapid detection and ultra-high sensitivity. Here, we prepared a novel Ti 3 C 2 Tx (MXene)-noble metal hybrid flexible substrate with a layer of MXene on top of flower-like Ag nanoparticles (AgNFs), and finally grew Au nanoparticles (AuNPs) by galvanic replacement and self-reduction reactions. The limit of detection (LOD) can be as low as 2×10 -12 M by detecting 4-mercaptobenzoic acid (4MBA). For efficient sampling, transparent and flexible platform made of polydimethylsiloxane (PDMS) were used for the extraction and detection of thiabendazole (TBZ), thiram, methyl parathion (MP) and their mixtures. Notably, the lowest concentrations detected by swabbing on the tomato surface (3.7 ng/mL, 2.8 ng/mL) were lower than the EU maximum permissible limits of pesticides on raw fruits and vegetables (4.0 mg/L, 6.33 mg/mL). This high performance Ti 3 C 2 Tx-noble metal hybrid flexible substrate offers a great advantage in the trace detection of molecules.

Abstract:Surface-enhanced Raman scattering (SERS) is a promising method for rapid and sensitive detection of pesticide residues. Herein, a three-dimensional MoS2/Au/Ag nanostructure was developed by employing composite systems containing semiconductors, noble metal nanoparticles, and internal standard. The MoS2 nanoflowers provide abundant anchors for the loading of plasmonic nanoparticles so as to generate intense electromagnetic “hot spots,” and the Au-MBN/Ag nanostructures can increase the accuracy of SERS detection with the calibration ability of the internal standard molecules (4-mercaptobenzonitrile, 4-MBN). Therefore, the MoS2/Au/Ag nanostructures integrate the advantages of high-enhancement SERS substrates and high-accuracy ratiometric Raman detection. Moreover, the nanostructures were fabricated via a facile procedure and showed high sensitivity, accuracy, and reproducibility in SERS detection. The nanostructures also showed good linear fitting between the ratios of Raman intensity and logarithmic concentrations of analytes in ratiometric SERS analysis. In addition, thiabendazole (TBZ) in apple juice was successfully detected by using the as-prepared SERS substrates with a limit of detection down to 0.1 ppm in practical applications. To conclude, the MoS2/Au/Ag nanostructures showed excellent SERS activity in ratiometric analysis and the substrate is expected to have great potential for the quantitative detection of low-concentration pesticide residues.

Abstract:The purpose of this study was to develop a flexible substrate methylcellulose-decorated silver nanoparticles (MC/Ag NPs) film and explore its application in fruits and vegetables by surface enhanced Raman spectroscopy (SERS) technology for rapid detection of pesticides. The performance of the MC/Ag NPs film substrate was characterized by Nile blue A (NBA), and the detection limit was as low as 10−8 M. The substrate also exhibited satisfactory Raman signal strength after two months of storage. The impressive sensitivity and stability were due to the excellent homogeneity of the silver nanoparticles that were grown in situ in the methylcellulose matrix, which generated “hot spots” between the silver nanoparticles without a large amount of aggregation, and resulted in the ultra-high sensitivity and excellent stability of the MC/Ag NPs film substrate. The MC/Ag NPs film substrate was used to detect thiram pesticides on tomato and cucumber peels, and the minimum detectable level of thiram was 2.4 ng/cm2, which was much lower than the maximum residue level. These results indicate that the MC/Ag NPs film is sensitive to rapid detection of multiple pesticides in food.

Abstract:Rapid identification and analysis of pesticide residue on real‐world sample by a portable platform is of growing interest for monitoring food safety and safeguarding human health. Herein, we developed a robust surface‐enhanced Raman spectroscopy (SERS) sensor based on ZrO2@TiO2/Ag substrate for the analysis of pesticide residues. The synergetic effects of the MOFs‐derived porous ZrO2@TiO2 heterostructure induced charge transfer enhancement and the electromagnetic enhancement from the plasmonic AgNPs hotspot generate significant SERS enhancement signal. The SERS substrate exhibits superior long‐term stability and signal homogeneity for SERS quantitative analysis. The platform has the capability to detect not only model molecule of 4‐ATP but also pesticides of triazophos and fonofos at the extremely low concentration of 10‐8 M. Moreover, we simulate the quantitative detection of pesticide residue on real objects by spraying pesticides on dendrobium and chrysanthemum leaves, followed by transferring to the SERS platform to perform analysis. The nearly linear fitting curve with a wide concentration range suggests the superior qualitative detection capability. Remarkably, two‐component and three‐component of the pesticides are readily identified in a rapid way. The approach provides the rapid analysis, low‐cost device, portable and quantitative pesticide identification, and offers a clear path to multicomponent analysis at a trace level.

CD8+ T cell subsets of cytotoxic T lymphocytes induced by Epstein-Barr virus infection in infectious mononucleosis.

Abstract:Mononuclear peripheral blood lymphocytes (PBL) from patient with infectious mononucleosis (IM) were tested in a 51Cr-release assay for cytotoxicity against autologous and allogeneic lymphoblastoid cell line (LCL), or Epstein-Barr virus (EBV)-genome positive and negative cell line. In acute phase, PBL lyse an autologous LCL as well as allogeneic LCL (Wa cells). High levels of cytotoxicity were observed in the combinations between effector and target cells sharing HLA-Class 1 product. EBV-genome positive Daudi and Raji cells which lack HLA-Class 1 antigen and have mismactched HLA-Class 1 antigen, respectively showed resistance to killing. EBV-genome negative tumor cells except NK sensitive K562 cells were not killed by IM lymphocytes. However, the IM lymphocytes without atypical form in convalescent phase failed to show killing activity against autologous and allogeneic LCL. These findings suggest that cell surface membrane antigen structure on EBV-infected LCL may be able to explain the recognition and triggering of lysis of target cells by HLA-Class 1 restricted cytotoxic T cells (CTL) from acute IM. Phenotypic analysis of PBL with atypical form from IM was made by two-color flow cytometry. The data demonstrate that CD8+ T cells quantitatively represent the major population of lymphocytes expanded during acute IM. Furthermore, approximately 70% of these CD8+ T cells express HLA-DR on these surface, suggesting that they have undergone activation. However, IL 2R (CD25 antigen) expression was not significantly elevated on activated T cells. The salient profile on cytofluorographs of an acute IM was the increased number of CD3+CD19-, CD8+CD11b-, CD8+CD28+ and CD8+S6F1+ cells. However, CD3-CD19+, CD8+CD11b+, CD8+S6F1-, CD4+Leu8- and CD25+HLA-DR+ antigens were little expressed. Increased number of CD8+CD11b-, CD8+CD28+ and CD8+S6F1+ cells, which are regarded as CTL were reduced according to the improvement of the clinical symptoms and laboratory findings. These results together with HLA typing analysis suggested a possibility HLA-Class 1 restriction of the CTL with surface phenotype of CD8+CD11b-, CD8+CD28+, and CD8+S6F1+.
Rapid Detection of Malathion, Phoxim and Thiram on Orange Surfaces Using Ag Nanoparticle Modified PDMS as Surface-Enhanced Raman Spectroscopy Substrate

Abstract:Malathion, phoxim, and thiram are organophosphates and organosulfur pesticides widely used in agricultural products. The residues of these pesticides present a direct threat to human health. Rapid and on-site detection is critical for minimizing such risks. In this work, a simple approach was introduced using a flexible surface-enhanced Raman spectroscopy (SERS) substrate. The prepared Ag nanoparticles-polydimethylsiloxane (AgNPs-PDMS) substrate showed high SERS activity, good precision (relative standard deviation = 5.33%), and stability (30 days) after optimization. For target pesticides, the linear relationship between characteristic SERS bands and concentrations were achieved in the range of 10~1000, 100~5000, and 50~5000 μg L−1 with LODs down to 3.62, 41.46, and 15.69 μg L−1 for thiram, malathion, and phoxim, respectively. Moreover, SERS spectra of mixed samples indicated that three pesticides can be identified simultaneously, with recovery rates between 96.5 ± 3.3% and 118.9 ± 2.4%, thus providing an ideal platform for detecting more than one target. Pesticide residues on orange surfaces can be simply determined through swabbing with the flexible substrate before acquiring the SERS signal. This study demonstrated that the prepared substrate can be used for the rapid detection of pesticides on real samples. Overall, this method greatly simplified the pre-treatment procedure, thus serving as a promising analytical tool for rapid and nondestructive screening of malathion, phoxim, and thiram on various agricultural products.

Abstract:Rapid and effective detection of pesticide residues from complex surfaces of fruits and vegetables has important significance. Herein, we report a novel three-dimensional (3D) hierarchical porous functional surface-enhanced Raman scattering (SERS) substrate, which is fabricated by successive two-step hydrothermal synthesis strategy of silver nanoparticles (Ag NPs) and cobalt oxide nanowires (Co3O4 NWs) on the 3D copper foam framework as Cu@Co3O4@Ag-H. The strategy offers a new avenue for localized plasmonic materials distribution and construction, which exhibits better morphology regulation ability and SERS activity (or hotspots engineering) than physical spurring obtained Cu@Co3O4@Ag-S. The developed Cu@Co3O4@Ag-H possesses large surface area and rich hotspots, which contributes to the excellent SERS performance, including homogeneity (RSD of 7.8%), sensitivity (enhancement factor, EF of 2.24 × 108) and stability. The Cu@Co3O4@Ag-H not only provides plenty of Electromagnetic enhancement (EM) hotspots but also the trace detection capability for droplet rapid sensing within 2 s. Cu@Co3O4@Ag-H substrate is further developed as an effective SERS sensing platform for pesticide residues detection on the surfaces of fruits and vegetables with excellent LOD of 0.1 ppm, which is lower than the most similar reported works. This work offers new potential for bioassay, disease POCT diagnosis, national security, wearable flexible devices, energy storage and other related fields.

Abstract:The shrinkage of 3D hot spots droplets on a hydrophobic array substrate for the quantitative and multiplex detection of pesticide residues by using a portable Raman spectrometer is designed and demonstrated. The synergetic effects between MoS2 and AgNPs enhance the SERS sensitivity. The platform achieves quantitative analysis and multi‐components detection of pesticide residues on honeysuckle surface. The practical application of surface‐enhanced Raman spectroscopy (SERS) technology for quantitative analysis still confronts the hinder of controlling the uniformity of signal and anchoring target molecules in a designed space. Herein, a facile method of devising the shrinkage of three‐dimension (3D) hot spots droplets on a hydrophobic array substrate is demonstrated for the quantitative and multiplex detection of pesticide residues by using a portable Raman spectrometer. The shrunk droplet which is consisted of the analyte solution and AgNPs sol can induce simultaneously increasing density of plasmonic hot spots and immobilizing target molecules in hot spots. Besides, the closely packed Ag nanoparticles (AgNPs) are anchored in the gaps of the MoS2 nanosheet array, generating uniform hotspots distribution with relative standard deviation (RSD) of about 8 %. In addition, the synergetic effects between MoS2 and AgNPs further enhance the SERS sensitivity, and thereby target molecules of 10−9 m are readily detected. Besides, the proposed strategy exhibits excellent SERS sensitivity for the analysis of pesticides down to 10−8 m. Importantly, quantitative analysis and multi‐components detection of pesticide residues on honeysuckle surface are achieved. Therefore, it is expected to be used for quantitative analysis of complex pesticide systems on real samples.

Abstract:Herein, we developed a PDMS-based flexible SERS substrate modified with gold nanostars (AuNSs) for insitu detection of pesticide residues on fruit surfaces. AuNSs with sharp branches were assembled on aminated PDMS membrane by chemical bonding effect. The AuNSs/PDMS substrate showed good signal uniformity, stability and sensitivity using the Raman signal molecule 4-MBA and the detection concentration of 4-MBA was as low as 10(-8) mol/L. Then the AuNSs/PDMS substrate was used to detect methyl parathion (MP) standard solution. A good linear relationship between SERS intensity at 1342 cm(-1) and MP concentration was established in the range of 4 mu g/mL-100 mu g/mL, and the limit of detection was down to 1.946 mu g/mL. Moreover, the AuNSs/PDMS substrate was directly covered on the surface of MP contaminated apple, and the Raman laser was incident from the back of substrate to achieve in-situ detection of pesticide residues. The recovery ratio indicated that the fabricated SERS substrate was realized successfully in real sample detection. AuNSs/PDMS substrate eliminates the requirement for sample preprocessing steps before testing, which can be used as a method for rapid inspection of pesticide in agricultural products on site. Abstract:We report a surface magnetic solid-phase extraction–surface-enhanced Raman scattering (SMSPE-SERS) method based on silver-nanoparticle-coated Fe 3 O 4 /chitosan (Fe 3 O 4 /CS@Ag) microspheres as the substrate, and this method integrates all steps from sample pretreatment to detection. Fe 3 O 4 /CS was synthesized by a one-step solvothermal method in which chitosan (CS) was used as a surface modifier and adsorbent. Fe 3 O 4 /CS@Ag microspheres exhibit both adsorption ability and SERS activity. Therefore, we used the SMSPE-SERS method to detect pesticide residues on fruit peel. The procedures of capturing, separating and enriching pesticides, as well as detection, are all integrated. In addition, the SERS substrate allows label-free detection of thiram pesticide in both fruit peel and apple juice. Owing to the uniform distribution of Ag NPs and the adsorption ability of CS, the thiram-detection sensitivity was sufficiently high to detect the lowest concentration of 1.2 ng/cm 2 , which was significantly lower than the maximum thiram residue limit (7 μg/cm 2 ) in fruits. The method was comparable to high-performance liquid chromatography with recovery ranging from 86.60 to 109.69 %.

Abstract:Surface-enhanced Raman spectroscopy (SERS) is becoming an attractive technique for applications in food safety detection, environmental monitoring, disease diagnosis, and molecular identification. However, SERS substrates often suffer from low sensitivity, poor signal homogeneity, and stability. Herein, solution-processable monodisperse Ag nanoparticles (M-AgNPs) with an average diameter of 66 nm are synthesized by a size-controllable seeded-growth method. Furthermore, the M-AgNPs were coated on the surface of bacterial nanocellulose (BNC) to form flexible monodisperse Ag nanoparticles@bacterial nanocellulose (M-Ag@BNC) SERS sensors by a simple vacuum-assisted filtration. The SERS performance of flexible M-Ag@BNC substrates with various M-Ag loadings is systematically investigated. Due to the highly homogeneous distribution of M-AgNPs, the optimal M-Ag-12@BNC SERS sensor exhibits high sensitivity with a detection sensitivity of 10–13 M for methylene blue, excellent reproducibility (relative standard deviation (RSD) = 4.48%), and prominent storage stability (over 20 days). Besides, the hydrophilic BNC with good permeability and adsorption properties can absorb target molecules in the high-density hot-spot regions to further enhance SERS performance. Finite-difference time domain (FDTD) is also used to simulate and verify the strong electric field distribution of a M-AgNP-based SERS sensor. Finally, the M-Ag-12@BNC SERS sensor is successfully applied to detect pesticide residues on irregular fruit surfaces by a simple and feasible paste-and-read method. The sensitivity of a flexible M-Ag@BNC SERS sensor for thiram is up to 10–8 M. The flexible, ultrasensitive, and stable M-Ag@BNC SERS sensor exhibits potential applications in trace identification of hazardous organic molecules.

Abstract:In this paper, we proposed a simple Surface-Enhanced Raman Scattering (SERS) detection method for the possible pesticide residues on fruit surfaces. 3-aminopropyl triethoxysilane (APTES) is used to modify Ag nanoparticles (AgNPs) on polydimethylsiloxane (PDMS) membranes. The PDMS was attached to the fruit epidermis to adsorb the substance, and then the SERS test was performed to analyze whether the epidermis had pesticide residues. In the present study, the AgNPs were modified using the Tollens synthesis method while glucose was set to be the reducing agent, and the AgNPs were synthesized into a uniform size of 20-50nm. In this study, we fabricated a simple flexible SERS substructure, which achieved the detection limits of R6G and Thiram at 10(-8) M. In the future, our work can be extended to biomedical applications, or focusing on the characteristic substances that may threaten the national defense and security, which has a broad application scenario.
Detection of Pesticides in Fruits by Surface-Enhanced Raman Spectroscopy Coupled with Gold Nanostructures

Abstract:There is growing concern in recent years for consumers about contamination of pesticides in fruits due to increasing use of pesticides in fruits. The objective of this study was to use surface-enhanced Raman spectroscopy (SERS) to detect and characterize pesticides extracted from fruit surfaces. Gold-coated SERS-active nanosubstrates were used for SERS measurement. Three types of pesticides (carbaryl, phosmet, and azinphos-methyl) widely used in apples and tomatoes were selected. Significantly enhanced Raman signals of pesticides were acquired by SERS from the extract of fruit samples and exhibited characteristic patterns of the analytes. Multivariate statistical methods such as partial least squares and principal component analysis were used to develop quantitative and qualitative models. SERS was able to detect all three types of pesticides extracted from fruit samples at the parts per million level. The study of detection limit demonstrated that at 99.86% confidence interval, SERS can detect carbaryl at 4.51 ppm, phosmet at 6.51 ppm, and azinphos-methyl at 6.66 ppm spiked on apples; and carbaryl at 5.35 ppm, phosmet at 2.91 ppm, and azinphos-methyl at 2.94 ppm on tomatoes. Most of these detection limits meet the maximum residue limits established by Food and Agriculture Organization of the United Nations and World Health Organization. Satisfactory recoveries (78–124%) were achieved for samples with concentrations at and larger than the detection limit. These results demonstrate that SERS coupled with novel SERS-active nanosubstrates is a rapid, sensitive, and reliable method for detection and characterization of chemical contaminants in foods.

Large-Scale Fabrication of Ultrasensitive and Uniform Surface-Enhanced Raman Scattering Substrates for the Trace Detection of Pesticides

Abstract:Technology transfer from laboratory into practical application needs to meet the demands of economic viability and operational simplicity. This paper reports a simple and convenient strategy to fabricate large-scale and ultrasensitive surface-enhanced Raman scattering (SERS) substrates. In this strategy, no toxic chemicals or sophisticated instruments are required to fabricate the SERS substrates. On one hand, Ag nanoparticles (NPs) with relatively uniform size were synthesized using the modified Tollens method, which employs an ultra-low concentration of Ag+ and excessive amounts of glucose as a reducing agent. On the other hand, when a drop of the colloidal Ag NPs dries on a horizontal solid surface, the droplet becomes ropy, turns into a layered structure under gravity, and hardens. During evaporation, capillary flow was burdened by viscidity resistance from the ropy glucose solution. Thus, the coffee-ring effect is eliminated, leading to a uniform deposition of Ag NPs. With this method, flat Ag NPs-based SERS active films were formed in array-well plates defined by hole-shaped polydimethylsiloxane (PDMS) structures bonded on glass substrates, which were made for convenient detection. The strong SERS activity of these substrates allowed us to reach detection limits down to 10−14 M of Rhodamine 6 G and 10−10 M of thiram (pesticide).

Abstract:It is of great significance to develop a simple and effective method for constructing large-scale high-quality surface-enhanced Raman scattering (SERS) substrate. Here Ag nanoparticle-assembled micro-bowl array was prepared by a close-packed polystyrene (PS) sphere monolayer templated electrodeposition approach. The fabricated Ag nanoparticle-assembled micro-bowl array shows high SERS sensitivity to rhodamine 6G (R6G) under an ultralow concentration of 1 fM, and exhibits excellent SERS spectral uniformity with a small relative standard deviation (RSD) of 7.6% and good reproducibility (a RSD ~8.2% for the average peak intensities from different batches of SERS substrates). The fabricated micro-bowl array SERS substrate was employed to detect pesticide residue (thiram and methyl parathion) on vegetables. The limit of detections (LODs) for the two pesticides are lower than the maximum residue limits (MRLs) set by European Union respectively, showing promising application in rapid inspection of food safety.

Abstract:Nowadays, trace detection of thiram is in urgent demand due to its widespread application in agriculture and significant harmful effects on public health. In this work, a three-dimensional (3D) sea cucumber-like flexible porous surface-enhanced Raman scattering (SERS) substrate composed of a poly(vinylidene fluoride) (PVDF) membrane, ZnO nanorods, gold films, and Ag nanoparticles (Ag/Au/ZnO/P) has been established for the highly sensitive detection of thiram. The substrate takes advantage of the 3D morphology of the Ag/Au/ZnO system on a flexible porous PVDF membrane to produce abundant plasmonic hot spots. Meanwhile, the employment of an AgNPs/Au shell system combined the benefits of both gold and silver metals, thus guaranteeing stable and sensitive detection. With 4-mercaptobenzoic acid (4-MBA) as a probe molecule, the Ag/Au/ZnO/P substrate exhibited excellent linear detection in the range of 10-11-10-5 M, with a correlation coefficient (R2) of 0.99 and an enhancement cofactor of 7.09 × 107. The substrate exhibited excellent uniformity with a related standard deviation (RSD) value of 3.82% and demonstrated high stability during a 15 d-storage test. In addition, the substrate could detect thiram in an aqueous solution at concentrations as low as 10-10 M with excellent selectivity. Meanwhile, thiram on the surface of apple peel could be easily detected by the Ag/Au/ZnO/P substrate with the "paste-and-peel" method in less than 10 s, and the detection limit could be as low as 0.48 ng cm-2. Overall, the remarkable performance of the Ag/Au/ZnO/P SERS substrate demonstrated its great potential for the environmental monitoring of thiram.

Abstract:A new approach is presented to fabricate flexible surface-enhanced Raman scattering (SERS) substrate of Ag nanocubes monolayer-modified polydimethylsiloxane (Ag NCs/PDMS) through a powerful three-phase interface self-assemble method. The morphologies and crystal structures were characterized by scanning electron microscopy and X-ray diffraction. The self-assembled Ag NCs/PDMS substrate exhibited high SERS activity and good signal homogeneity, which was successfully used for quantitative detection of thiram; the detection limit reached 10 ng/mL, and the linear range is 10–1000 ng/mL. Furthermore, the flexible SERS substrates were successfully employed to detect thiram residues on factual apple samples, and trace amount (1 ng/cm 2 ) of thiram residues was detected on apple peels. The excellent SERS detection ability of self-assembled Ag NCs/PDMS substrate indicated that it will play an important role in pesticide detection in the future.

Abstract:Pesticides are still abused and overused in agricultural production. Food safety monitoring in terms of reliable detection and quantification of pesticide residues is an ever increasing need. Surface-Enhanced Raman Scattering (SERS)-based chemical sensors and adsorption materials crafted for specific pesticide should provide high sensitivity and selectivity. Herein, zeolitic imidazolate framework (ZIF-8) derived TiO2/ZnO three-dimensional (3D) heterostructures were constructed on FTO substrate and were utilized to assemble silver nanoparticles (AgNPs) to create electromagnetic hot spots. The formation of TiO2/ZnO heterostructure greatly improve the SERS performance of the substrate. The calculated enhancement factor (EF) of the TiO2/ZnO/Ag is 39-fold higher than the TiO2/Ag substrates. Furthermore, the substrate shows high signal sensitivity and reproducibility for rapid detection of multiple pesticides. Even with a portable Raman device, the substrate was found to be highly efficient for model molecules of crystal violet (CV) and 4-mercaptobenzoic acid (4-MBA), and the limit of detection (LOD) for both of 10−9 M was demonstrated. Importantly, real life detection of multiple pesticides residues can be achieved with the TiO2/ZnO/Ag sensor, which was demonstrated by the detection of three common pesticides, thiram, acephate and phoxim at trace concentrations.
