Abstract:(1) Background: This study was conducted to investigate the effects of dietary fluoride (F) on tissue retention, digestive enzymes activities, mucosal immunity, and cecum microbial community of laying hens. (2) Methods: Total of 288 37-week-old Hy-Line Gray laying hens with similar laying rate (85.16% ± 3.87%) were adapted to the basal diets for ten days, and then allocated into three groups at random (n = 9, 6, 6 replicates/group). The concentrations of F in the diets were 31.19 (the control group, CON), 431.38 (F400, low-F group) and 1237.16 mg/kg (F1200, high-F group), respectively. The trial lasted for 59 days. (3) Results: Results suggested that F residuals in duodenum responded to dietary F concentrations positively. The activities of amylase, maltase and lactase were decreased in high-F group, compared with those in the control group. The mRNA expression levels of jejunum and ileum secretory immunoglobulin A (sIgA) and Mucin 2, and sIgA concentrations were decreased inhigh-F group, than those in the control group. The observed operational taxonomic units (OTUs) of laying hens in high-F group were higher than the CON and low-F groups, and the bacterial structure was different from the other two groups. The Lactobacillus was higher in the control group, while Gammaproteobacteria, Escherichia-Shigella, Streptococcaceae, and Enterobacteriaceae were higher in the high-F group. (4) Conclusions: The actual results confirmed that dietary high F intake increased the F residuals in duodenum, and reduced the digestion and absorption of nutrients and immunity via decreasing the activities of digestive enzymes, impairing intestine mucosal immunity, and disturbing the cecum microbial homeostasis of laying hens.

Abstract:The objective of this study was to investigate the effects of excess dietary fluoride (F) on laying performance and antioxidant capacity of laying hens. A total of 576 laying hens, 51 wk old, was randomly divided into 6 groups, each of which included 6 replicates of 16 hens. Graded amounts of sodium fluoride (NaF) were added to the basal diet to achieve concentrations of 16 (control), 200, 400, 600, 800, and 1,000 mg/kg F, respectively. Dietary F at 1,000 mg/kg significantly decreased ADFI, laying rate, and average egg weight, and increased feed conversion ratio (FCR) (P < 0.05). No significant differences were observed in serum total antioxidant capacity (T-AOC) level or catalase (CAT) concentration among all the treatments, while hens fed F at 800 and 1,000 mg/kg had higher activity of serum glutathione peroxidase (GSH-PX) and concentration of malondialdehyde (MDA) (P < 0.05) as compared to the control group. Compared with the control group, dietary F at 400 mg/kg increased liver MDA concentration (P < 0.001), and decreased CAT concentration of liver (P < 0.001); 600 mg/kg F decreased liver T-AOC levels (P < 0.001); and 800 mg/kg of F decreased liver total superoxide dismutases (T-SOD) activity (P < 0.001). Compared with the control group, feeding F at 600 mg/kg decreased kidney T-AOC levels and T-SOD activity (P < 0.001), and increased MDA concentration of kidney (P < 0.001), while dietary 1,000 mg/kg of F decreased kidney GSH-PX activity (P < 0.05) and CAT concentration (P < 0.001). In conclusion, these results indicated that excessive F ingestion had an adverse effect on laying performance by inducing oxidative stress and impairing the antioxidant system of laying hens.

Abstract:This study was conducted to investigate the effects of excess dietary fluoride (F) on serum biochemical indices, egg quality, and concentrations of F in soft tissues, eggs, and serum of laying hens. Commercial laying hens (n = 576, 51 weeks of age) were randomly allotted to 6 treatments with 6 replicates of 16 birds. The basal diets contained fluorine inclusions at a level of 16 mg/kg, and graded sodium fluoride was added to the basal diet to achieve fluorine inclusions, respectively, at a level of 200, 400, 600, 800, and 1000 mg/kg in the experimental diets. Dietary F levels at 600, 800, and 1000 mg/kg decreased (P < 0.05) albumin height and yolk color, while eggshell strength and eggshell thickness significantly decreased at 800 and 1000 mg/kg, respectively, compared with the control group. Fluoride concentrations in eggshell, albumin, yolk, liver, kidney, ovary, and oviduct responded to dietary F levels positively, and F concentrations in eggshell were the highest. Fluorine concentrations in albumin and yolk increased with the feeding time at the same dietary F levels (P < 0.05). Dietary F level at 400 mg/kg increased serum calcium level and activity of glutamic oxalacetic transaminase (P < 0.05). In conclusion, dietary F levels at 600 mg/kg decreased albumin height and yolk color, while eggshell strength and eggshell thickness significantly decreased at 800 and 1000 mg/kg, respectively. F concentrations in soft tissues, albumin, yolk, and eggshell of layers had a positive correlation with dietary F levels. By disturbing Ca and phosphorus metabolism, dietary F levels affected the formation of eggshell, reducing eggshell strength and eggshell thickness.

Abstract:The present study aimed to evaluate the effects of excess dietary fluoride (F) on laying performance, egg quality, tissue retention, serum biochemical indices, and serum reproductive hormones of laying hens. A total of 384 Hy-Line Gray hens, 37 wk old, were treated with sodium fluoride added to a corn-soybean meal basal diet at 0, 400, 800, and 1200 mg fluorine/kg feed. The results showed that dietary F levels at 800 and 1200 mg/kg markedly decreased ADFI, laying rate, average egg weight, and increased feed conversion ratio (FCR) (P < 0.05). Dietary F levels at 800 and 1200 mg/kg dramatically decreased the egg quality of albumen height, yolk color, eggshell strength, and eggshell thickness, and on the 49th D, 400 mg/kg F group significantly decreased the eggshell strength, compared to those of control group. Fluoride residues in tissues of hens were increased significantly with the increase of dietary F supplemental levels (P < 0.05). Fluoride concentrations were generally high in feces, eggshell, tibia, kidney, and ovary, and the highest in feces, following with eggshell and tibia, lower in kidney and ovary, and the lowest in serum. Serum uric acid levels and alanine aminotransferase activity increased significantly (P < 0.05), and glucose, triglycerides, and phosphorus decreased significantly (P < 0.05) in response to dietary F concentration, compared to those of the control group, respectively. Dietary F supplementation at 1200 mg/kg significantly decreased (P < 0.05) the estrogen concentrations in serum, compared to those of the control group. Concentrations of progesterone in the fluoride-treated groups were significantly (P < 0.05) decreased relative to those of the control group. In conclusion, these results indicated that the excessive ingestion of F has had a detrimental effect on egg laying rate and quality of eggs by damaging the function of the liver, kidney, and ovary of laying hens.

Abstract:The effects of feeding dehydrated Enterococcus faecium on growth performance, immune response, and cecal microflora in broiler chickens challenged with Escherichia coli K88 were investigated. Two hundred eighty-eight 1-d-old birds were randomly assigned to 4 treatments: negative control birds (N-con) fed a basal diet and not challenged with E. coli K88; positive control birds (P-con) fed a basal diet and challenged with E. coli K88; birds fed a basal diet including dehydrated E. faecium (Ef) at 1 × 10(9) cfu/kg of feed and challenged with E. coli K88; and birds fed a basal diet including the antibiotic colistine sulfate (Anti) at 10 mg/kg of feed and challenged with E. coli K88. Birds fed E. faecium had greater (P < 0.05) BW on d 14, 21, and 28 and greater (P < 0.05) jejunal villus height on d 21 and 28 compared with birds on the other treatments. Jejunal crypt depth was decreased (P < 0.05) in birds fed either E. faecium or antibiotic compared with P-con treatment birds on d 10, 21, and 28. Birds fed E. faecium had a greater (P < 0.05) concentration of IL-4 in their jejunal mucosa than did those in the N-con treatment group on d 10, 14, and 21. Infected birds, with or without E. faecium feeding, had a higher (P < 0.05) tumor necrosis factor-α and secreted IgA in their jejunal mucosa than did those in the N-con treatment group on d 10 and 14. Birds fed E. faecium had lower (P < 0.05) concentrations of E. coli on d 14 and 28, less (P < 0.05) Clostridium perfringens on d 28, greater Lactobacillus counts on d 14 and 21, and greater (P < 0.05) Bifidobacterium in their cecal contents on d 21 than did the P-con birds. These results suggest that E. faecium can promote growth performance, improve intestinal morphology, and beneficially manipulate the cecal microflora in broilers challenged with E. coli K88.

Abstract:To study the effects of fluorine supplement on digestive function in piglets,ninety-six piglets(Duroc×Landrace×Yorkshire) with body weight of about 16kg were allotted to four groups by weight and sex and every group included three replicates of eight pigs each.The groups fed the same basal diet supplemented with 0,100,250,400 mg/kg fluorine from sodium fluoride.Results show that:①The fluorine supplements all decrease the apparent digestibility of dry matter and crude protein(P<0.02),and 250 mg/kg and 400 mg/kg fluorine decrease additionally the apparent digestibility of crude fat(P<0.02).②Every dosage fluorine supplement decreases the activities of total proteinase,lipase,amylase in duodenal content and lipase activity in pancreas(P<0.05),and 400 mg/kg fluorine decreases additionally the total proteinase in pancreas(P<0.01).③ Every dosage fluorine supplement decreases duodenum villus height(P<0.01),while increases crypt depth(P<0.01).④Excessive fluorine supplement apparently injures the structure of pancreatic epithelium cell and its mitochondrion,endoplasmic reticulum,etc.
Effects of Fluorosis on Lipid Peroxidation and Antioxidant Systems in Young Pigs

Abstract:This study was conducted to investigate effects of fluoride on lipid peroxidation and antioxidant systems in young pigs. Three groups of crossbred barrows about 50 days old were exposed to 100, 250, and 400 mg F-/kg (from NaF) in their diets for 50 days. Serum malondialdehyde (MDA) and nitric oxide (NO) levels were significantly increased, and the activities of superoxide dismutase (SOD), glutathione peroxidase (GSH-Px), and catalase (CAT) were significantly decreased. In thyroid, liver, and kidney tissues, the MDA level was significantly increased, and SOD and GSH-Px activities were significantly decreased. These results suggest that fluoride induces excessive production of NO and reactive oxygen species (ROS), enhances lipid peroxidation, and disturbs the antioxidant system of pigs. Oxidative damage from oxidative stress could therefore be an important pathway for fluoride toxicity in soft tissues.

Abstract:To investigate the effect of estrogen deficiency on the small intestinal mucosal barrier induced by fluoride (F), F exposure models of ovariectomy (OVX) rats (surgically removed ovaries) and non-OVX rats (normal condition) were established by adding sodium fluoride (NaF) (0, 25, 50, and 100 mg/L, calculated by F ion) in drinking water for 90 days. The intestinal mucosal histomorphology, mucosal barrier function, and protein expression levels of tight junctions (TJs), adhesion junctions (AJs), and desmosomes were evaluated in the duodenum, jejunum, and ileum. Hematoxylin-eosin (HE) staining and 5-Bromo-2-deoxyUridine (BrdU) measurement showed that excessive F-induced damage to intestinal epithelial cells and inhibited the proliferation of intestinal epithelial cells, eventually decreasing the number of goblet cells and decreasing glycoprotein secretion, as indicated by Alcian blue and periodic acid-Schiff (AB-PAS) and periodic acid-Schiff (PAS) staining. Further immunofluorescence analysis demonstrated that excessive F decreased the protein expression levels of occludin, zonula occludens-1 (ZO-1), E-cadherin, and desmoplakin (P < 0.05, P < 0.01) and enhanced the expression of claudin-2 (P < 0.01), suggesting that cell-to-cell junctions were disrupted. Collectively, F exposure impaired the small intestinal mucosal barrier by inducing damage to intestinal epithelial cells and inhibiting intestinal epithelial cell proliferation. Disorders in the junctional complex protein expression blocked the synergy between intercellular communication and aggravated mucosal injury. In particular, estrogen deficiency exacerbated F-induced enterotoxicity, which provides new explanations for the development and severity of intestinal disease in postmenopausal women with high-F areas.

Targeting Gut Microbiota and Metabolism Profiles with Coated Sodium Butyrate to Ameliorate High-Energy and Low-Protein Diet-Induced Intestinal Barrier Dysfunction in Laying Hens

Abstract:High energy diet are a risk factor for intestinal barrier damage. Butyrate, a major energy source for intestinal epithelial cells, has been shown to improve barrier dysfunction and modulate the gut microbiota. In this trial, we examined the preventative effect of coated sodium butyrate (CSB) on high-energy low-protein (HELP)-induced intestinal barrier injury in laying hens, and also worked to determine the underlying mechanisms by an integrative analysis of gut microbiota and the metabolome. A total of 216 healthy 28-weeks-old Huafeng laying hens were randomly assigned to 3 groups with 6 replicates each: the CON group (normal diet), HELP group (high-energy and low-protein diet) and CH500 group (500 mg/kg CSB added to HELP diet). The duration of the trial encompassed a period of 10 weeks. The results revealed that CSB treatment improved the laying rate (P<0.05) and mitigated the detrimental effects on intestinal barrier function and the inflammatory response induced by the HELP diet in laying hens (P<0.05). Microbial profiling analysis revealed that the CSB treatment reshaped the HELP-perturbed gut microbiota, and promoted the growth of beneficial bacteria (P<0.05). Untargeted metabolomics analysis revealed that CSB reduced the metabolites associated with intestinal inflammation (P<0.05). In conclusion, CSB did not merely modulate alterations in the gut microbiota composition and microbial metabolites but also yielded increased egg production, while mitigating intestinal barrier dysfunction and inflammatory responses induced by HELP in laying hens.
Antagonistic Effects of Different Selenium Sources on Growth Inhibition, Oxidative Damage, and Apoptosis Induced by Fluorine in Broilers

Abstract:Fluorosis can induce oxidative stress through leading to reactive oxygen species (ROS) generation. Selenium (Se) can eliminate ROS by direct and indirect manners. In this study, therefore, we investigated the possible protective effects of sodium selenite (SS) and selenomethionine (Se-Met) on fluorine (F)-induced oxidative stress in broilers. A total of 720 1-day-old Lingnan Yellow broilers were allotted to 4 groups (6 replicates of 30 birds each group) and fed with basal diet (control group), 800 mg/kg F (high F group), 800 mg/kg F+0.15 mg Se/kg as SS (SS group), or Se-Met (Se-Met group), respectively. The experiment lasted 50 d. High F group significantly decreased (P < 0.05) the average daily gain (ADG) and feed efficiency (FE) in comparison with control group. The contents of ROS, malondialdehyde, 8-hydroxydeoxyguanosine, protein carbonyl, and cysteinyl aspartate specific proteinases 3 in serum, liver, and kidney were higher (P < 0.05) in high F group than those in control group. Compared with control group, the decreased (P < 0.05) activities of glutathione peroxidase (GSH-Px) and cytoplasmic thioredoxin reductase (TrxR1) as well as contents of selenoprotein P (SelP), total protein (TP), and B-cell lymphoma-2 in serum and tissues were observed in high F group. Moreover, the pathological lesions of liver and kidney in high F group were more than those in control group. However, supplementation with SS and Se-Met could improve ADG and FE, increase SelP and TP concentrations, elevate GSH-Px and TrxR1 activities, minimize the changes of oxidative stress and apoptosis parameters as well as ultrastructure of liver and kidney, whereas the effects of Se-Met were better than those of SS. The results indicated that excess F could result in growth inhibition of broilers through inducing oxidative stress and subsequently caused oxidative damage to biological macromolecules and soft tissues as well as apoptosis, whereas dietary SS and Se-Met supplementation could antagonize high F induced growth retardation by inhibiting oxidative stress and a mechanism of apoptosis regulation and the impact was more with Se-Met.

Abstract:Colon microenvironment and microbiota dysbiosis are closely related to various human metabolic diseases. In this study, a total of 72 healthy female mice were exposed to fluoride (F) (0, 25, 50 and 100 mg/L F−) in drinking water for 70 days. The effect of F on intestinal barrier and the diversity and composition in colon microbiota have been evaluated. Meanwhile, the relationship among F-induced colon microbiota alterations and antimicrobial peptides (AMPs) expression and short-chain fatty acids (SCFAs) level also been assessed. The results suggested that F decreased the goblet cells number and glycoprotein expression in colon. And further high-throughput 16S rRNA gene sequencing result demonstrated that F exposure induced the diversity and community composition of colonic microbiota significantly changes. Linear Discriminant Analysis Effect Size (LEfSe) analysis identified 11 predominantly characteristic taxa which may be the biomarker in response to F exposure. F-induced intestinal microbiota perturbations lead to the significantly decreased SCFAs levels in colon. Immunofluorescence results showed that F increased the protein expression of interleukin-17A (IL-17A) and IL-22 (P < 0.01) and disturbed the expression of interleukin-17 receptor A (IL-17RA) and IL-22R (P < 0.05 or P < 0.01). In addition, the increased expression of IL-17A and IL-22 cooperatively enhanced the mRNA expression of AMPs which response to F-induced microbiota perturbations. Collectively, destroyed microenvironment and disturbed AMPs are the primary reason of microbiota dysbiosis in colon after F exposure. Colonic homoeostasis imbalance would be helpful for finding the source of F-induced chronic systemic diseases.

Effects of Excessive Feed Fluorine on Structure and Function of Liver in Piglets

Abstract:Ninety-six piglets(Duroc×Landrace×Yorkshire) with body weight of 17kg were allotted into four groups by weight and sex.Every group included three replicates of eight pigs each.The groups received the same basal diet(containing 26.18mg/kg fluorine) supplemented with 0,100,250,400mg/kg fluorine as sodium fluoride.Results show that 250mg/kg and 400mg/kg fluorine increases the percentage of liver weight to live weight by 16.28%(P<0.02) and 20.00%(P<0.01) respectively;meanwhile,excessive fluorine apparently injures the structure of hepatocytes,including mitochondrions and endoplasmic reticulum,resulting in liver swelling and necrosis.In addition,100,250 and 400mg/kg fluorine decreases the succinate dehydrogenase activity of hepatocytes mitochondrion by 19.50%(P<0.05),29.02%(P<0.01) and 23.17%(P<0.03) respectively.400mg/kg fluorine improves the glutamate pyruvate transminase activity of serum by 36.43%(P<0.05).It is implicated that excessive feed fluorine could impair the structure and function of liver in piglets.
Th17-Related Cytokines Involved in Fluoride-Induced Cecal and Rectal Barrier Damage of Ovariectomized Rats

Abstract:To investigate fluoride (F)-induced intestine barrier damage and the role of estrogen deficiency in this progress, a rat model of estrogen deficiency was established through bilateral surgical removal of ovaries. The F exposure model was then continued by adding sodium fluoride (0, 25, 50, and 100 mg/L, calculated on a fluorine ion basis) to drinking water for 90 days. Afterward, intestinal mucosal structure, barrier function, and inflammatory cytokines were evaluated. The results showed that excessive F decreased the developmental parameters (crypt depth) of the cecum and rectum and inhibited the proliferation capacity of the intestinal epithelia, which are more obvious in the state of estrogen deficiency. The distribution of goblet cells and glycoproteins in the intestinal mucosa decreased with the increase in F concentration, and estrogen deficiency led to a further decline, especially in the rectum. Using the immunofluorescence method, the study showed that excessive F caused interleukin-17A (IL-17A) significantly decrease in the cecum and increase in the rectum. Meanwhile, F treatment remarkably upregulated the expression of intestinal IL-1β, IL-23, and IL-22, while the level of IL-6 was downregulated. In addition, estrogen deficiency increased IL-1β, IL-6, IL-23, and IL-22, but decreased IL-17A expression in the cecum and rectum. Collectively, F exposure damaged intestinal morphological structure, inhibited epithelial cell proliferation and mucus barrier function, and resulted in the disturbance of T helper (Th) 17 cell-related cytokines expression. Estrogen deficiency may further aggravate F-induced damage to the cecum and rectum.

Abstract:This study aimed to investigate the effects of fermented blueberry pomace (FBP) on the intestinal barrier function and cecal microbiome of aged laying hens. A total of 320 Yukou Jingfen No. 8 laying hens (345-day-old) were randomly divided into a control group, 0.25% FBP group, 0.5% FBP group, or 1.0% FBP group. The results showed that the villus height (VH) in the jejunum of the 0.25-0.5% FBP groups and villus surface area in the jejunum of the 0.25% FBP group were higher (p < 0.05), while 0.25% FBP supplementation displayed a higher (p = 0.070) VH in the ileum compared to the control group. Mucin-2 expression was upregulated (p < 0.05) in the jejunum of the 0.5% FBP group and the ileum of the 0.25-0.5% FBP groups. Compared to the control group, interleukin (IL)-4 and IL-13 expressions were upregulated (p < 0.05) in the 1.0% FBP group. Microbiota analysis revealed that Prevotella abundance in the cecum of the 0.5-1.0% FBP groups was higher (p < 0.05) than in the 0.25% FBP group. In addition, microbial function prediction analysis showed that cecal microbiota in the 0.25% FBP group were mainly enriched by alanine/aspartate/glutamate metabolism and methane metabolism. Moreover, Spearman's correlation analysis revealed the potential correlations between the abundance of the cecal microbiota and intestinal-barrier-function-related gene expressions, as well as the short-chain fatty acid content, of laying hens. In summary, dietary FBP supplementation enhanced intestinal barrier function by improving intestinal morphology, upregulating gene expressions related to barrier function, and altering the cecal microbiota of aged laying hens.

Abstract:Fluoride is a common contaminant of groundwater and agricultural commodity, which poses challenges to animal and human health. A wealth of research has demonstrated its detrimental effects on intestinal mucosal integrity; however, the underlying mechanisms remain obscure. This study aimed to investigate the role of the cytoskeleton in fluoride-induced barrier dysfunction. After sodium fluoride (NaF) treatment of the cultured Caco-2 cells, both cytotoxicity and cytomorphological changes (internal vacuoles or massive ablation) were observed. NaF lowered transepithelial electrical resistance (TEER) and enhanced paracellular permeation of fluorescein isothiocyanate dextran 4 (FD-4), indicating Caco-2 monolayers hyperpermeability. In the meantime, NaF treatment altered both the expression and distribution of the tight junction protein ZO-1. Fluoride exposure increased myosin light chain II (MLC2) phosphorylation and triggered actin filament (F-actin) remodeling. While inhibition of myosin II by Blebbistatin blocked NaF-induced barrier failure and ZO-1 discontinuity, the corresponding agonist Ionomycin had effects comparable to those of fluoride, suggesting that MLC2 serves as an effector. Given the mechanisms upstream of p-MLC2 regulation, further studies demonstrated that NaF activated RhoA/ROCK signaling pathway and myosin light chain kinase (MLCK), strikingly increasing the expression of both. Pharmacological inhibitors (Rhosin, Y-27632 and ML-7) reversed NaF-induced barrier breakdown and stress fiber formation. The role of intracellular calcium ions ([Ca2+]i) in NaF effects on Rho/ROCK pathway and MLCK was investigated. We found that NaF elevated [Ca2+]i, whereas chelator BAPTA-AM attenuated increased RhoA and MLCK expression as well as ZO-1 rupture, thus, restoring barrier function. Collectively, abovementioned results suggest that NaF induces barrier impairment via Ca2+-dependent RhoA/ROCK pathway and MLCK, which in turn triggers MLC2 phosphorylation and rearrangement of ZO-1 and F-actin. These results provide potential therapeutic targets for fluoride-induced intestinal injury.

Abstract:This study was conducted to investigate the effects of fluoride on pancreatic digestive enzyme activities and ultrastructure in young pigs. Three groups of crossbred barrows were exposed to 100, 250, and 400 mg F-/kg (from NaF) in their diets for 60 days. Compared to a control group, the activities of pancreatic lipase and protease (but not amylase) were significantly decreased. Pancreatic acinar cells showed markedly swollen mitochondria and loss of mitochondrial cristae. Endoplasmic reticulum (ER) was markedly dilated and its folds were irregular. These results indicate that excessive fluoride in the diet can inhibit pancreatic digestive enzyme activities and cause ultrastructural changes, which may lead to a series of biochemical and pathological abnormalities.

Abstract:This experiment was conducted to evaluate the effects of sodium sulfate supplementation in laying hens diet by determining performance,antioxidant capacity,intestinal morphology and cecal microflora population. Four hundred and fifty 20-week-old Hyline W hite laying hens with similar body weight and laying rate were randomly allocated to 5 groups with 6 replicates per group and 15 hens per replicate. Hens in the control group was fed a basal diet,and hens in experimental groups were fed the basal diets supplemented with 0.3%,0.6%,1.5%,3.0% sodium sulfate,respectively. The trial lasted for 8 weeks. The results showed as follows:1) diets supplemented with sodium sulfate tended to improve the laying performance of laying hens with no significant effect( P 0.05),and the effect of lowdose groups( 0.3% and 0.6%) was better. 2) Compared with control group,diets supplemented with sodium sulfate had no significant influences on antioxidant parameters( P0.05). Compared with 0.6% group,the activities of glutathione peroxidase( GSH-Px) in plasma and total superoxide dismutase( T-SOD) in liver of high dose( 1. 5% and 3. 0%) groups were significantly decreased( P0.05 or P0.01),group added by 3.0% had significantly reduced the total antioxidant capacity( T-AOC)in plasma and the activity of GSH-Px in liver( P 0.05). 3) Compared with control group,the villus height in duodenum and ileum of 0.6% group had significantly raised by 30. 51% and 42. 63%( P 0. 01); the crypt depth in duodenum was significantly reduced( P 0.05) and the ratio of villus height to crypt depth( V / C) in duodenum was significantly raised( P 0.05). Compared with 0.6% group,the villus height in duodenum and ileum and V / C in duodenum of high dose( 1.5% and 3.0%) groups were significantly decreased( P 0.01),and the crypt depth in duodenum and ileum of 3. 0% group was significantly increased( P 0. 01). 4) Compared with control group,0. 3% and 0. 6% groups had remarkably reduced the number of Escherichia coli in cecum( P 0.01),and the group supplemented with 0. 6% was the best; compared with 0. 6% group,high dose( 1.5% and 3.0%) groups had significantly increased the number of Enterobacteria and Escherichia coli in cecum( P 0.05 or P 0.01). It is concluded that dietary sodium sulfate supplemental level at 0.3% to 0.6%tend to improve the antioxidant capacity and intestinal morphology,and regulate the cecal microflora population in laying hens. The optimum supplemental level of sodium sulfate is 0.6% in laying hens diet.
Alterations in Intestinal Antioxidant and Immune Function and Cecal Microbiota of Laying Hens Fed on Coated Sodium Butyrate Supplemented Diets

Abstract:This study was designed to evaluate the effects of dietary coated sodium butyrate (CSB) on the intestinal antioxidant, immune function, and cecal microbiota of laying hens. A total of 720 52-week-old Huafeng laying hens were randomly allocated into five groups and fed a basal diet supplemented with CSB at levels of 0 (control), 250 (S250), 500 (S500), 750 (S750), and 1000 (S1000) mg/kg for eight weeks. The results revealed that CSB supplementation quadratically decreased the malondialdehyde content and increased the superoxide dismutase activity of the jejunum as well as the total antioxidative capacity activity of the ileum (p < 0.05). Dietary CSB supplementation linearly decreased the diamine oxidase and D-lactic acid content of the serum (p < 0.05). Compared with the control group, the addition of CSB resulted in linear and/or quadratic effects on the mRNA expression of inflammatory cytokines TNF-α, IL-6, and IL-10 in the jejunum and ileum (p < 0.05). The short-chain fatty acid concentrations increased quadratically as supplemental CSB improved (p < 0.05). Additionally, dietary CSB levels had no effect on microbial richness estimators, but ameliorated cecal microbiota by raising the abundance of probiotics and lowering pathogenic bacteria enrichment. In conclusion, our results suggest that dietary supplementation with CSB could improve the intestinal health of laying hens via positively influencing the antioxidant capacity, inflammatory cytokines, short-chain fatty acids, and gut microbiota. In this study, 500 mg/kg CSB is the optimal supplement concentration in the hens’ diet.
