Abstract:The structure of the oviduct of female Macrobrachium rosenbergii during spawning was examined with optical microscope and transmission electron microscope(TEM).In Macrobrachium rosenbergii,a pair of oviducts originates from the anterolateral part of the ovary ends at the female genital pores situated in the basipodite of the 3rd thoracic appendage.The oviduct wall,folded in some areas, consists of a basal lamina and a layer of columnar epithelial cells.There are two types of epithelial cells,one with light cytoplasm and the other with dark cytoplasm,found in the oviduct wall.The free surface of the epithelial cells possesses long dense microvilli.And beneath the microvilli,there is a thick layer of secreted substance.The nucleus is in oval shape with one or several nucleoli.The cytoplasm contains abundant mitochondria and rough endoplasmic reticulums.After shedding the eggs,the cell membrane and the microvilli break and the cytoplasm leaks out.The layer of secreted substance disappears,only a little residue of it remains.

Abstract:Ultrastructure of development of compound eyes before and after hatching in Macrobrachium nipponense was studied by electron microscopy.The results of this study showed that(1) the dioptre system of an ommatidium included cornea,corneagenous cell,and crystalline cone surrounded by iris pigmental cells.From 16th day after spawning to the zoeaⅠstage,the thickness of cornea,the diameter of crystalline cone,and the amounts of organelles in corneagenous cells increased gradually,in addition,the blocks of dense electron granules in crystalline cone changed from 4 to 4-8 retinular cell appeared in the end of crystalline cone.(2) The sensate system of an ommatidium composed of 8 retinular cells.Microvilli protruded from retinular cells maked up of the 'Decapoda type' rhabdom,which arounded a thin layer of cytoplasm where imbibe vesicles and perirhadomal vacuoles could be seen.With the development,the diameter of rhabdom,the amounts of organelles such as mitochondria,endoplasmic reticulum,increased gradually.Futhermore,lamellar bodies,multivesicular bodies,proximal pigment granule also appeared.(3) Retinular cells have a differential process: at 16th day after spawning,the space between each of seven retinular cells was wide and separated by the four lobes of the eighth retinular cell,and the distal pigment granules appeared;at zoeaⅠstage,location of the eighth retinular cell has ascended,which made the other seven retinular cells circled around the rhabdom tightly,a great deal of proximal pigment granules can be seen in retinular cells.

Abstract:The structural change of the oviduct of freshwater shrimp (Macrobrachium nipponense) during spawning was examined by electron microscopy. The oviduct wall structural characteristics seem to be influenced significantly by the spawning process. Before the parturition and ovulation, two types of epithelial cells (types I and II) are found in the epithelium. The free surfaces of type I and type II cells have very dense long microvilli. Under the type I and type II cells, are a relatively thick layer of secreting material and a layer of mostly dead cells. After ovulation, two other types of epithelial cells (types III and IV) are found in the oviduct wall epithelium. The free surface of type III cells only has short microvilli scattered on the surface. The thick layer with secreting material and the dead cell layer disappeared at this stage. In some type III cells, the leaking out of cytoplasm from broken cell membrane led to the death of these type III cells. The transformation of all four types of epithelial cells was in the order: IV→I→ II→ III.
On the Function and Evolution of Electric Organs in Fish

Abstract:1. The electric discharges of Gymnarchus niloticus and of representative species of seven genera of the Mormyridae have been examined in their natural habitat in Africa and in the laboratory. 2. Comparable investigations of the South American Gymnotidae have shown the existence of two discharge types in both these unrelated fish families. 3. The first type of electric discharge consists of very regular sequences of continuously emitted, monophasic pulses, varying from species to species in frequency, and within narrower limits from individual to individual. 4. Fish emitting this first type of pulses include Gymnarchus , Hypopomus and Eigenmannia . The frequency range for these fish lies between 60 and 400 discharges/sec. 5. The frequency does not alter with the state of excitation of the fish. The duration of individual pulses is relatively long, i.e. 2-10 msec. 6. The second type of discharge is less regular in frequency, the pulse duration much shorter and the pulse shape more complex. The individual discharge from the whole electric organ lasts about 0.2 msec, in Petrocephalus . 7. This type of discharge is found in all the examined species of the Mormyridae and in such forms as Gymnotus carapo and Staetogenes elegans . 8. The basic discharge rate of a resting mormyrid is somewhat variable and not strictly rhythmical. It usually lies between 1 and 6 pulses/sec. 9. Stimuli which excite the mormyrids cause an increase in the discharge frequency. The recorded maximum is about 130 pulses/sec. 10. Suitable stimuli can inhibit the discharges of the Mormyridae for prolonged periods. 11. In Gymnotus carapo and Staetogenes elegans the basic discharge rate is higher and of regular rhythmicity. Depending on temperature the frequencies lie between 30 and 87 pulses/sec. When these fish are excited the frequencies are increased up to 200 pulses/sec, for a short time. 12. The shape of the electric field, which is set up with each pulse around the fish, has been examined. 13. A theory has been proposed which suggests that these fish, by means of their electric pulses, can locate objects if their electrical conductivity differs from that of water. 14. These fish have shown themselves extremely sensitive to influences affecting the electric field. This has been studied by applying artificial electric stimuli, by studying the effects of conductors and non-conductors introduced into the field, and the reactions towards magnetic fields and electrostatic charges. 15. Conditioned reflex experiments with Gymnarchus niloticus and Gymnotus carapo have shown that these fish can detect the presence of a stationary magnet, and that they can discriminate between conductors and non-conductors. 16. The prey of these fish does not appear to be affected by the discharges. Inter alia , the electric pulses have a social significance. 17. This locating mechanism may be considered as an adaptation to life in turbid water. 18. Gymnotidae and Mormyridae (taken to include Gymnarchus ) show striking features of convergent evolution. 19. Unusual locomotory adaptations such as swimming by means of the dorsal fin ( Gymnarchus ), the anal fin (Gymnotidae) and ‘Gemminger9s bones’ (Mormyridae) may be considered as a means which tends to make the axis of symmetry of the fish and of its electric field coincide during active movements. 20. A new theory for the evolution of electric organs has been suggested. A major prerequisite appears to be a receptor sensitive to electrical stimulation. 21. It is suggested that special sensory and nervous differentiations of the lateralis system (‘mormyromasts’, valvulae cerebelli) are concerned with the perception and integration of electric stimuli. 22. Muscular action potentials have been recorded in the water at some distance from non-electric fish. 23. The easiest explanation for the evolution of strong electric organs would appear to start from such muscular action potentials, and proceed via weak electric organs used for orientation, to the powerful offensive and defensive electric organs.
A rare case of intra-ovarian oocyte maturation

Abstract:Summary The intra-ovarian presence of ootids, i.e. female gametes that have completed meiosis, is considered exceptional in the animal kingdom. The present study explores the first such case to be reported in a sea cucumber (Echinodermata: Holothuroidea). In the overwhelming majority of animals, including holothuroids, oocytes (i.e. immature female gametes) that are developing in the ovary undergo a primary arrest at the prophase stage of meiosis, which may last from days to decades. In free-spawning taxa, this arrest is normally lifted only during or shortly before transit in the gonoduct, when gamete release (spawning) is imminent. However, oocytes of the holothuroid Chiridota laevis were discovered to have resumed the second meiotic division including the completion of germinal vesicle breakdown and polar-body expulsion inside the ovary, effectively reaching the ootid stage concomitantly with ovulation (i.e. escape from follicle cells) prior to spawning. The potential drivers and significance of this exceptionally rare case of full intra-ovarian oogenic maturation are discussed.

Abstract:Oocyte maturation is a process that occurs in the ovaries, where an immature oocyte resumes meiosis to attain competence for normal fertilization after ovulation/spawning. In starfish, the hormone 1-methyladenine binds to an unidentified receptor on the plasma membrane of oocytes, inducing a conformational change in the heterotrimeric GTP-binding protein α-subunit (Gα), so that the α-subunit binds GTP in exchange of GDP on the plasma membrane. The GTP-binding protein βγ-subunit (Gβγ) is released from Gα, and the released Gβγ activates phosphatidylinositol-3 kinase (PI3K), followed by the target of rapamycin kinase complex2 (TORC2) and 3-phosphoinositide-dependent protein kinase 1 (PDK1)-dependent phosphorylation of serum- and glucocorticoid-regulated kinase (SGK) of ovarian oocytes. Thereafter, SGK activates Na+/H+ exchanger (NHE) to increase the intracellular pH (pHi) from ~6.7 to ~6.9. Moreover, SGK phosphorylates Cdc25 and Myt1, thereby inducing the de-phosphorylation and activation of cyclin B–Cdk1, causing germinal vesicle breakdown (GVBD). Both pHi increase and GVBD are required for spindle assembly at metaphase I, followed by MI arrest at pHi 6.9 until spawning. Abstract:The ultrastructure of human oocytes has been described only qualitatively. To offer a precise organelle spatial distribution and organelle volume during the main maturation stages, we previously conducted stereological studies on prophase-I (GV) and metaphase-I (MI) oocytes, and here we present results on metaphase-II (MII) oocytes. Five donor oocytes from different donors were processed for transmission electron microscopy, and quantification of organelle distribution was performed using point-counting stereology. Statistical tests compared the means of the relative volumes occupied by organelles among oocyte regions. The most abundant organelles were elements of the smooth endoplasmic reticulum (SER), such as SER small vesicles, SER medium vesicles, SER large vesicles and SER isolated tubules, along with mitochondria, followed by SER tubular aggregates, cortical vesicles and lysosomes. Significant differences between oocyte regions were found for lysosomes, cortical vesicles and SER large vesicles. Comparisons of MII oocytes to previous findings in GV and MI oocytes evidenced specific patterns of organelle distribution and relative volumes. This final evaluation thus enables to track organelle spatial reorganization across oocyte stages, which, in addition to gathered knowledge, may be useful to assist in improvements of stimulation protocols, in-vitro maturation media and cryopreservation techniques.

Abstract:This contribution describes the growth of oocytes, addressing the formation of structures that compose the follicular complex, as well as the remodeling of the extracellular matrix, specifically laminin, fibronectin and type IV collagen during gonadal maturation. Thirty-seven females of the Acari zebra fish, Hypancistrus zebra were captured and the ovaries were submitted to histological processing for light and electron microscopy and immunohistochemistry techniques. Oogonia and four stages (I – IV) of oocytes were distinguished, and structures such as the postovulatory follicle and atretic oocytes (initial and advanced atresia) were observed. The follicular complex consists of the mature oocyte, zona radiata (Zr1, Zr2 and Zr3), follicular cells, basement membrane and theca. During oocyte growth, proteins of the extracellular matrix showed different intensities of staining. Based on these observations, a model of oocyte growth is proposed to define specific characteristics of the oocyte and the remodeling of the extracellular matrix in the ovary of H. zebra. This model of oocyte growth can be extended to other species of ornamental fishes. This study contributes data for induced fertilization and eventual conservation of this species.

Abstract:The post ovulated eels could be divided into two types:one is smoothly ovulated eels(SOE),another is partly ovulated eels(POE).Ratio of Ⅳ +++ phase oocytes and Ⅴ phase oocytes in ovaries of SOE was much higher than that of POE. POE Ⅳ ++ phase oocytes and Ⅵ phase oocytes were commonly observed in ovaries.Concentrations of mGnRH, plasma gonadotropin and plasma estradiol 17 β were higher in SOE than in POE.Testosterone and cGnRH Ⅱ levels in plasma of SOE and POE were foumd without significant difference.

Abstract:In viviparous fish, a considerable degree of variation in placental structures have been described. However, no distinct structures are reported in Scorpaenidae. In this study, we demonstrate a new type of folliculogenesis and follicular placentation in Sebastes schlegelii. Before copulation, the germinal epithelium gradually surrounds the oocytes and develops to individually follicles with a stalk-like structure hanging on the ovigerous lamella, which ensures each follicle have access to spermatozoa after copulation. From stage V to early gestation, the cyp17-I highly expressed accompanied by cyp19a1a signals disappearance, and 11-ketotestosterone level keeps rising and peaks at blastula stage, while 17β-estradiol declines to the bottom. Meanwhile, the theca cells rapidly proliferate and invade outwards forming a highly hypertrophied and folded microvillous placenta. This unbalance of hormone might be an important factor driving the theca cells proliferation and invasion. Additionally, some conserved genes related to mammalian placentation are significantly high expression in follicular placenta suggesting the high convergence in vertebrate placenta evolution.

Abstract:Heteropteran ovaries are meroistic, telotrophic type and composed of several ovarioles that consist of a terminal filament, tropharium, vitellarium, and ovariole pedicel. The tropharium is made up of trophocytes, young oocytes and prefollicular cells, while the vitellarium consists of linearly arranged ovarian follicles (oocytes surrounded by follicular epithelium). Contact of the oocytes with the tropharium is maintained by nutritive cords. Despite the general similarity in the organization of heteropteran ovarioles, there are significant differences in the structure of the tropharium and vitellarium. In primitive groups of Heteroptera, the tropharium is composed of individual cells, while in more advanced taxa, the trophic chamber is built of syncytial lobes containing several nuclei (the exception is Miridae). The vitellarium consists of a different number of ovarian follicles, from one to 12. Oocytes develop either synchronously or asynchronously. The differentiation and diversification of the follicular epithelium differs even in closely related groups. The data so far comes from studies on representatives of 22 families (Heteroptera includes 87 families) and are usually only partial. To analyse the anagenesis of the heteropteran ovaries, research on representatives of the other heteropteran families from the studied infraorders should be carried out, and from Enicocephalomorpha whose ovaries have not been examined.

A Single‐Cell Atlas of Crab Ovary Provides New Insights Into Oogenesis in Crustaceans

Abstract:Crabs are a valuable resource for aquatic ecosystems worldwide. This study presents a high‐resolution single‐cell atlas to study the oogenesis process in crabs. The results indicate the conserved trajectory of oogenesis from oogonia to oocytes. Moreover, based on the distribution features and subsequent functional analysis, the study determined several molecules that play decisive roles in crab oogenesis. Oogenesis is crucial for sexual reproduction and provides the material basis for population continuation. Nonetheless, the identity of the cells involved, the nature of transformation, and underlying regulators of oogenesis in crustaceans remain elusive. Here, an atlas of the ovary is plotted via single‐nuclei RNA sequencing (snRNA‐seq) in the mud crab Scylla paramamosain, resulting in five cell types, including germ cells, somatic cells, and three follicle cell types identified, which in turn provides abundant candidate markers for them. Moreover, profiles of ligand‐receptor in different cells of the crab ovary indicate the roles of cell communication in oogenesis. Dozens of transcription factors in the trajectory from oogonia to oocytes as well as the key molecules/pathways in somatic cells and follicle cells relevant to oogenesis are screened, which is evolutionarily conserved and its underlying regulatory mechanism is subject to some modification across various phyla. The spatiotemporal expression patterns of seven markers are further verified and the RNAi confirms the essential roles of piwi and VgR in oogenesis. These data help to elucidate the mechanism underlying gametogenesis and the evolution of reproductive strategy in invertebrates.

Abstract:Asexual reproduction by means of splitting, also called fissiparity, is a common feature in some asterozoan groups, especially in ophiactid brittle stars. Most fissiparous brittle stars show six instead of the usual five rays, live as epibionts on host organisms, and use clonal fragmentation to rapidly colonize secluded habitats and effectively expand the margins of their distribution area. While the biology and ecology of clonal fragmentation are comparatively well understood, virtually nothing is known about the evolution and geological history of that phenomenon. Here, we describe an exceptional fossil of an articulated six-armed brittle star from the Late Jurassic of Germany, showing one body half in the process of regeneration, and assign it to the new species Ophiactis hex sp. nov. Phylogenetic inference shows that the fossil represents the oldest member of the extant family Ophiactidae. Because the Ophiactis hex specimen shows an original six-fold symmetry combined with a morphology typically found in epizoic ophiuroids, in line with recent fissiparous ophiactid relatives, we assume that the regenerating body half is an indication for fissiparity. Ophiactis hex thus shows that fissiparity was established as a means of asexual reproduction in asterozoan echinoderms by the Late Jurassic.

Abstract:Oogenesis has universal characteristics among fishes, in which structures of vital importance for embryos are assembled. However, these universal structures may have additional functions depending on the evolutionary ecology of each species. The aim of this research was to describe histologically the structure of the ovary and the oogenesis of the pupfishes Cyprinodon artifrons and Garmanella pulchra , with emphasis on the follicular structures linked to the resistance of embryos to desiccation. The embryos of pupfishes experience periodic deleterious environmental changes caused by estuarine tides. They can survive periods out of water protected by accessory egg layers assembled in the ovaries during oogenesis. This research found that both species have a single ovary with asynchronous follicle development, associated with a frequent breeding activity. Also, multiple generations of cortical alveoli were described, larger in G. pulchra , related with the formation of perivitelline space, a structure that can buffer the effects of desiccation when embryos remain out of water. In both species, the accumulation of oil droplets in the oocytes was simultaneous with the synthesis of cortical alveoli. Oil droplets may be related to energy reserves during extended incubation periods. The zona pellucida of both species presented an intermediate thickness with respect to other species, and hair-like filaments arranged on the zona pellucida of G. pulchra were clearly detected. These structures as a whole, could be related to the survival of the embryos of the pupfishes of Yucatan in the tidal fluctuations in the estuarine environments in which they inhabit.

Abstract:Fish ovaries exhibit a remarkable diversity in shape, size, and organization, reflecting the myriad reproductive strategies employed by different species. This review delves into the intricate biology of fish ovaries, highlighting their structural diversity and the hormonal regulation that governs ovarian development and oocyte maturation. Key hormones include pituitary gonadotropins (GTHs) and maturation-inducing hormones (MIHs), which initiate oocyte growth and maturation. GTHs stimulate ovarian production of estradiol-17β and 17α,20β-DP, which induce oocyte maturation via MPF formation. Sex steroids like estrogens and progestogens, synthesized from cholesterol, play crucial roles. Other hormones, including growth hormone, prolactin, thyroid hormones, IGFs, ACTH, and melatonin, influence ovarian activity. The review also explores the varied reproductive strategies among fish, including oviparity and viviparity, and discusses how environmental factors like water temperature and photoperiod influence ovarian histology. Deciphering dynamic interactions between spermatozoa and the ovarian microenvironment through integrated multi-omics approaches in viviparous black rockfish ( Sebastes schlegelii )

Abstract:The ovarian microenvironment plays a critical role in ensuring the reproductive success of viviparous teleosts. However, the molecular mechanism underlying the interaction between spermatozoa and the ovarian microenvironment has remained elusive. This study aimed to contribute to a better understanding to this process in black rockfish (Sebastes schlegelii) utilizing integrated multi-omics approaches. The results demonstrated significant upregulation of ovarian complement-related proteins and pattern recognition receptors, along with remodeling of glycans on the surface of spermatozoa at early spermatozoa-storage stage (one month after mating). As spermatozoa were stored over time, ovarian complement proteins were progressively repressed by tryptophan and hippurate, indicating a remarkable adaptation of spermatozoa to the ovarian microenvironment. Near fertilization, a notable upregulation of cellular junction proteins was observed. The study revealed that spermatozoa bind to ZPB2a protein through GSTM3 and that ZPB2a promoted spermatozoa survival and movement in a GSTM3-dependent manner. These findings shed light on a key mechanism influencing the dynamics of spermatozoa in the female reproductive tract, providing valuable insights into the molecular networks regulating spermatozoa adaptation and survival in species with internal fertilization.

Abstract:Neither the overall differences in ovariole number nor the caste-specifically modulated expression of vitellogenin can fully explain the striking caste differences in honey bee reproduction, in particular the mechanisms that block oogenesis in virgin queens and in workers kept in the presence of a queen. For this reason we investigated the initial stages of oogenesis in queens in relation to mating status and in workers exposed to different social conditions. A striking feature in ovarioles of both castes was a considerably elongated terminal filament which consisted not only of normal terminal filament cells but also contained apparently undifferentiated cells that were tentatively considered as stem cells. BrdU incorporation was detected in the upper germarium, as well as in the terminal filament. Cytoskeleton analysis by TRITC-phalloidin labeling for F-actin, and immunofluorescence detection for beta-tubulin did not reveal structural differences in the early oogenesis steps between queens and queenless workers. In contrast, queenright workers showed signs of a disorganized microtubule and microfilament system that could explain the histological evidence for progressive cell death observed in the germaria. In addition to cytoplasmic tubulin we also detected marked intranuclear foci indicating the presence of nuclear beta(II)-tubulin.
