Abstract:Many drug and therapeutic modalities have emerged over the past few years. However, successful commercialization is dependent on their safety and efficacy evaluations. Several preclinical models are available for drug-screening and safety evaluations, including cellular- and molecular-level models, tissue and organoid models, and animal models. Organoids are three-dimensional cell cultures derived from primary tissues or stem cells that are structurally and functionally similar to the original organs and can self-renew, and they are used to establish various disease models. Human hepatobiliary organoids have been used to study the pathogenesis of diseases, such as hepatitis, liver fibrosis, hepatocellular carcinoma, primary sclerosing cholangitis and biliary tract cancer, as they retain the physiological and histological characteristics of the liver and bile ducts. Here, we review recent research progress in validating drug toxicity, drug screening and personalized therapy for hepatobiliary-related diseases using human hepatobiliary organoid models, discuss the challenges encountered in current research and evaluate the possible solutions.

Abstract:Cancer global burden accounts for 19.3 million new cases and 10 million deaths in 2020. Despite advances on the understanding of disease progression and efforts to find successful therapeutic strategies, most drugs fail in clinical stages. Lack of models with clinical mimicry recapitulating the in vivo pathophysiology and the organ microenvironment where the tumor forms is a key driver reducing the success rate of anti-cancer drugs. Animal models remain the gold standard for preclinical research but fail to recapitulate the complex human tumor microenvironment, a critical component determining tumor development, progression, and response to treatments. Organ-on-a-chip models allow to control the environment and organization of cells in an in vivo-like arrangement by culturing multiple cell types to better replicate human conditions, and by implementing different biochemical and physical stimuli, such as fluid shear stress, chemical/nutrient gradients, and perfusion/flow to recapitulate the human vascular system physiology. We leverage our 3D bioprinting technologies to develop complex vascularized organ-on-a-chip models aimed to recapitulate the tumor and liver niche. Both organ-systems are interconnected by our h-VIOSTM platform, allowing dynamic perfusion for 21 days and enabling the simultaneous assessment of anti-tumor therapeutic efficacy and drug-induced liver injury. Our hydrogels feature a complex perfusable vasculature that interfaces with an interstitial chamber where tissues of interest are grown. The vasculature allows for continued and controlled delivery of nutrients, oxygen, and small molecule drugs. Dextran diffusion studies revealed diffusion coefficients ranging from 43.2 μm2/s (4kDa) to 8 μm2/s (70kDa). Vascular architectures are lined with endothelial cells forming a functional barrier expressing the junctional markers ZO-1 and PECAM-1. Moreover, we can modulate the permeability of our vasculature architecture, allowing the extravasation of immune and cancer cells. PBMC-derived T cells and Jurkat cells undergo transendothelial migration in response to a CXCL12 gradient, highlighting the applicability of our model for immunotherapeutic efficacy studies. The tumor interstitial chamber incorporates co-culture of cancer spheroids or tumoroids, fibroblasts, endothelial and immune cells. Colon carcinoma spheroids effectively integrate into the tumor niche with no growth arrest. The liver model consists of an interstitial chamber with a lobular microstructure supporting the monoculture of primary human hepatocytes, and sustaining their viability and function for 9 days as demonstrated by high albumin production, urea synthesis and CYP activity. Our organ-on-a-chip models are fully customizable and anticipated to contribute to advance personalized medicine and improve clinical translation rates. Research Progress of Organoids-on-chips in Biomedical Application

Abstract:Drug screening is traditionally based on the pharmacodynamic models from 2D cell culture or animal experiments. However, these models suffer from poor drug efficacy prediction due to their difference from human in vivo cell microenvironments. In recent years, advances in biotechnology and tissue engineering have enabled rapid growth of in vitro organoid culturing. These organoids cultured in matrigel can mimic human in vivo cell microenvironment and physiology more accurately than traditional 2D cell culture and animal models. Among them, tumor organoids, especially patient-derived tumour organoids, can be applied as effective cancer models for drug screening and personalized medicine. By integrating organoid culturing and microfluidics, organoid-on-a-chip platform has been developed to simulate the structure and function of human tissues or organs. In 2010, Ingber et al. reported a biomimetic microfluidic platform called lung-on-a-chip. Microscale engineering technologies were firstly utilized to create biomimetic microchips. Henceforth, increasing kinds of organ-on-a-chips were reported all over the world to mimic different organs, such as liver, kidney and brain. 2D planar cells were cultured in PDMS chambers in majority organ-on-a-chips. More and more studies focused on 3D cells or organoids instead recently due to their better biomimetic capability for drug screening and disease models. In 2018, Qin et al. reported a liver organoid from human iPSCs in a 3D perfusable chip system, which is the first study that uses human organoid in a microfluidic chip and executed drug testing successfully. The established liver organoid-on-a-chip system may provide a promising platform for engineering stem cell-based organoids with applications in regenerative medicine, disease modeling and drug testing. Conventionally, drug efficacy evaluation in organoid-on-a-chip was performed by cell staining. However, this approach only presents static results of live/dead cells, but cannot monitor the cells dynamically for long-term recording. Sensor integration with organoid-on-a-chips paves a new way for dynamic recording. Currently, Wang et al. developed a 3D electric cell/matrigel-substrate impedance sensing (3D-ECMIS) platform for real-time and non-invasive monitoring of 3D cell viability and drug susceptibility. This platform integrated impedance sensors to monitor the impedance changes during 3D cell culturing. Additionally, Wang et al. reported the 3D cardiomyocytes detection by combining microelectrode arrays (MEAs). Heart cells of neonatal rat were cultured on a tissue engineering scaffold, which was fabricated by 3D printing and electro-spinning. The experimental results demonstrated that extracellular field potential (spike amplitude and firing rate) of 3D cardiomyocytes can be recorded in real time. Moreover, non-invasive measurement of cellular metabolism is important to study the metabolism mechanism and drug efficacy. Thus, light-addressable potentiometric sensor (LAPS) was integrated with organoid-on-a-chip for chemical imaging of ions (H+, Na+, K+, Ca2+) induced by cellular metabolism, which enables accurate detection of different chemical parameters in organoid-on-a-chip. Multiple organoid chip linked by microfluidics, called human-on-a-chip is a newly emerging and frontier technology to mimic the interaction of multiple human organs in recent years. It can recapitulate the physiologically relevant structures and functions of the organs, as well as the interaction between multiple organs as in vivo, thereby offering alternative models for predicting human responses to various drugs and environmental stimulus. Organoid-on-a-chip demonstrates outstanding potential in drug screening, disease modeling and personalized precision medicine.

Abstract:Organ-on-a-chip systems are miniaturized microfluidic 3D human tissue and organ models designed to recapitulate the important biological and physiological parameters of their in vivo counterparts. They have recently emerged as a viable platform for personalized medicine and drug screening. These in vitro models, featuring biomimetic compositions, architectures, and functions, are expected to replace the conventional planar, static cell cultures and bridge the gap between the currently used preclinical animal models and the human body. Multiple organoid models may be further connected together through the microfluidics in a similar manner in which they are arranged in vivo, providing the capability to analyze multiorgan interactions. Although a wide variety of human organ-on-a-chip models have been created, there are limited efforts on the integration of multisensor systems. However, in situ continual measuring is critical in precise assessment of the microenvironment parameters and the dynamic responses of the organs to pharmaceutical compounds over extended periods of time. In addition, automated and noninvasive capability is strongly desired for long-term monitoring. Here, we report a fully integrated modular physical, biochemical, and optical sensing platform through a fluidics-routing breadboard, which operates organ-on-a-chip units in a continual, dynamic, and automated manner. We believe that this platform technology has paved a potential avenue to promote the performance of current organ-on-a-chip models in drug screening by integrating a multitude of real-time sensors to achieve automated in situ monitoring of biophysical and biochemical parameters.
A Novel 3d Liver Organoid System for Elucidation of Hepatic Glucose Metabolism

Abstract:Hepatic glucose metabolism is a key player in diseases such as obesity and diabetes as well as in antihyperglycemic drugs screening. Hepatocytes culture in two-dimensional configurations is limited in vitro model for hepatocytes to function properly, while truly practical platforms to perform three-dimensional (3D) culture are unavailable. In this work, we present a practical organoid culture method of hepatocytes for elucidation of glucose metabolism under nominal and stress conditions. Employing this new method of culturing cells within a hollow fiber reactor, hepatocytes were observed to self-assemble into 3D spherical organoids with preservation of tight junctions and display increased liver-specific functions. Compared to both monolayer culture and sandwich culture, the hepatocyte organoids displayed higher intracellular glycogen content, glucose consumption, and gluconeogenesis and approached the in vivo values, as also confirmed by gene expression of key enzymes. Moreover, hepatocyte organoids demonstrated more realistic sensitivity to hormonal challenges with insulin, glucagon, and dexamethasone. Finally, the exposure to high glucose demonstrated toxicities including alteration of mitochondrial membrane potential, lipid accumulation, and reactive oxygen species formation, similar to the in vivo responses, which was not captured by monolayer cultures. Collectively, hepatocyte organoids mimicked the in vivo functions better than hepatocyte monolayer and sandwich cultures, suggesting suitability for applications such as antihyperglycemic drugs screening.

Abstract:Drug toxicity assays using conventional 2D static cultures and animal studies have limitations preventing the translation of potential drugs to the clinic. The recent development of organs-on-a-chip platforms provides promising alternatives for drug toxicity/screening assays. However, most studies conducted with these platforms only utilize single endpoint results, which do not provide real-time/ near real-time information. Here, a versatile technology is presented that integrates a 3D liver-on-a-chip with a label-free photonic crystal-total internal reflection (PC-TIR) biosensor for rapid and continuous monitoring of the status of cells. This technology can detect drug-induced liver toxicity by continuously monitoring the secretion rates and levels of albumin and glutathione S-transferase α (GST-α) of a 3D liver on-a-chip model treated with Doxorubicin. The PC-TIR biosensor is based on a one-step antibody functionalization with high specificity and a detection range of 21.7 ng mL-1 to 7.83 x 103 ng mL-1 for albumin and 2.20 ng mL-1 to 7.94 x 102 ng mL-1 for GST-α. This approach provides critical advantages for the early detection of drug toxicity and improved temporal resolution to capture transient drug effects. The proposed proof-of-concept study introduces a scalable and efficient plug-in solution for organ-on-a-chip technologies, advancing drug development and in vitro testing methods by enabling timely and accurate toxicity assessments.
Development of an automated 3D high content cell screening platform for organoid phenotyping

Abstract:The use of organoid models in biomedical research has grown substantially since their inception. As they gain popularity among scientists seeking more complex and biologically relevant systems, there is a direct need to expand and clarify potential uses of such systems in diverse experimental contexts. Herein we outline a high-content screening (HCS) platform that allows researchers to screen drugs or other compounds against three-dimensional (3D) cell culture systems in a multi-well format (384-well). Furthermore, we compare the quality of robotic liquid handling with manual pipetting and characterize and contrast the phenotypic effects detected by confocal imaging and biochemical assays in response to drug treatment. We show that robotic liquid handling is more consistent and amendable to high throughput experimental designs when compared to manual pipetting due to improved precision and automated randomization capabilities. We also show that image-based techniques are more sensitive to detecting phenotypic changes within organoid cultures than traditional biochemical assays that evaluate cell viability, supporting their integration into organoid screening workflows. Finally, we highlight the enhanced capabilities of confocal imaging in this organoid screening platform as they relate to discerning organoid drug responses in single-well co-cultures of organoids derived from primary human biopsies and patient-derived xenograft (PDX) models. Altogether, this platform enables automated, imaging-based HCS of 3D cellular models in a non-destructive manner, opening the path to complementary analysis through integrated downstream methods.
Organotypic and Microphysiological Human Tissue Models for Drug Discovery and Development-Current State-of-the-Art and Future Perspectives

Abstract:The number of successful drug development projects has been stagnant for decades despite major breakthroughs in chemistry, molecular biology, and genetics. Unreliable target identification and poor translatability of preclinical models have been identified as major causes of failure. To improve predictions of clinical efficacy and safety, interest has shifted to three-dimensional culture methods in which human cells can retain many physiologically and functionally relevant phenotypes for extended periods of time. Here, we review the state of the art of available organotypic culture techniques and critically review emerging models of human tissues with key importance for pharmacokinetics, pharmacodynamics, and toxicity. In addition, developments in bioprinting and microfluidic multiorgan cultures to emulate systemic drug disposition are summarized. We close by highlighting important trends regarding the fabrication of organotypic culture platforms and the choice of platform material to limit drug absorption and polymer leaching while supporting the phenotypic maintenance of cultured cells and allowing for scalable device fabrication. We conclude that organotypic and microphysiological human tissue models constitute promising systems to promote drug discovery and development by facilitating drug target identification and improving the preclinical evaluation of drug toxicity and pharmacokinetics. There is, however, a critical need for further validation, benchmarking, and consolidation efforts ideally conducted in intersectoral multicenter settings to accelerate acceptance of these novel models as reliable tools for translational pharmacology and toxicology. SIGNIFICANCE STATEMENT: Organotypic and microphysiological culture of human cells has emerged as a promising tool for preclinical drug discovery and development that might be able to narrow the translation gap. This review discusses recent technological and methodological advancements and the use of these systems for hit discovery and the evaluation of toxicity, clearance, and absorption of lead compounds.

Abstract:Liver is the foremost organ of human being for drug metabolism, and it played a significant role in toxicity evaluation of drugs. Establishing a liver model in vitro can accelerate the process of the drug screening and new drug research and development. We provide a 3D printing based hepatic sinusoid-on-a-chip microdevice that reconstitutes organ-level liver functions to create a drug screening model of toxicity evaluation on chip. The microfluidic device, which recapitulates the hepatic sinusoid microenvironment, consists of PET polyporous membranes which mimic the perisinusoidal space, and experience fluid flow to mimic the hepatic arterial capillaries. The PET membrane was used to separate the hepatocyte and endotheliocyte. The endotheliocyte was cultured on the downside of the membrane and the hepatocyte were 3D seeded on the membrane via the 3D printer. This device was used to reproduce the in vitro liver model for drug toxicity assays. The expression of several biomarkers of liver was compared with the monoculture and 2D cultured conditions, and the results reveal that this organ-on-a-chip microdevice mimics the drug hepatoxicity that has not been possible by 2D cell-based and animal models, providing a useful platform for screening the drugs and developing an effective therapy in hepatopathy.

Abstract:<p>The past decade has seen many advancements in the development of three-dimensional (3D) <em>in vitro</em> models in pharmaceutical sciences and industry. Specifically, organoids present a self-organising, self-renewing and more physiologically relevant model than conventional two-dimensional (2D) cell cultures. Liver organoids have been developed from a variety of cell sources, including stem cells, cell lines and primary cells. They have potential for modelling patient-specific disease and establishing personalised therapeutic approaches. Additionally, liver organoids have been used to test drug efficacy and toxicity. Herein we summarise cell sources for generating liver organoids, the advantages and limitations of each cell type, as well as the application of the organoids in modelling liver diseases. We focus on the use of liver organoids as tools for drug validation toxicity and assessment.</p>

Pumpless, unidirectional microphysiological system for testing metabolism‐dependent chemotherapeutic toxicity

Abstract:Drug development is often hindered by the failure of preclinical models to accurately assess and predict the efficacy and safety of drug candidates. Body‐on‐a‐chip (BOC) microfluidic devices, a subset of microphysiological systems (MPS), are being created to better predict human responses to drugs. Each BOC is designed with separate organ chambers interconnected with microfluidic channels mimicking blood recirculation. Here, we describe the design of the first pumpless, unidirectional, multiorgan system and apply this design concept for testing anti‐cancer drug treatments. HCT‐116 colon cancer spheroids, HepG2/C3A hepatocytes, and HL‐60 promyeloblasts were embedded in collagen hydrogels and cultured within compartments representing "colon tumor", "liver", and "bone marrow" tissue, respectively. Operating on a pumpless platform, the microfluidic channel design provides unidirectional perfusion at physiologically realistic ratios to multiple channels simultaneously. The metabolism‐dependent toxic effect of Tegafur, an oral prodrug of 5‐fluorouracil, combined with uracil was examined in each cell type. Tegafur‐uracil treatment induced substantial cell death in HCT‐116 cells and this cytotoxic response was reduced for multicellular spheroids compared to single cells, likely due to diffusion‐limited drug penetration. Additionally, off‐target toxicity was detected by HL‐60 cells, which demonstrate that such systems can provide useful information on dose‐limiting side effects. Collectively, this microscale cell culture analog is a valuable physiologically‐based pharmacokinetic drug screening platform that may be used to support cancer drug development.

Abstract:Abstract Chemicals in the systemic circulation can undergo hepatic xenobiotic metabolism, generate metabolites, and exhibit altered toxicity compared with their parent compounds. This article describes a 2-chamber liver-organ coculture model in a higher-throughput 96-well format for the determination of toxicity on target tissues in the presence of physiologically relevant human liver metabolism. This 2-chamber system is a hydrogel formed within each well consisting of a central well (target tissue) and an outer ring-shaped trough (human liver tissue). The target tissue chamber can be configured to accommodate a three-dimensional (3D) spheroid-shaped microtissue, or a 2-dimensional (2D) cell monolayer. Culture medium and compounds freely diffuse between the 2 chambers. Human-differentiated HepaRG liver cells are used to form the 3D human liver microtissues, which displayed robust protein expression of liver biomarkers (albumin, asialoglycoprotein receptor, Phase I cytochrome P450 [CYP3A4] enzyme, multidrug resistance-associated protein 2 transporter, and glycogen), and exhibited Phase I/II enzyme activities over the course of 17 days. Histological and ultrastructural analyses confirmed that the HepaRG microtissues presented a differentiated hepatocyte phenotype, including abundant mitochondria, endoplasmic reticulum, and bile canaliculi. Liver microtissue zonation characteristics could be easily modulated by maturation in different media supplements. Furthermore, our proof-of-concept study demonstrated the efficacy of this coculture model in evaluating testosterone-mediated androgen receptor responses in the presence of human liver metabolism. This liver-organ coculture system provides a practical, higher-throughput testing platform for metabolism-dependent bioactivity assessment of drugs/chemicals to better recapitulate the biological effects and potential toxicity of human exposures.

The hepatic enzyme assay showed that the 3D biomimetic microtissue maintained high basal CYP-1A1/2 and UGT activities, responded dynamically to enzyme induction/inhibition, and preserved great hepatic capacity of drug metabolism. Using the established biomimetic microtissue, the potential adverse drug reactions that induced liver injury were successfully analyzed via drug-drug interactions of clinical pharmaceuticals. The results showed that predosed pharmaceuticals which agitated CYP-1A1/2 and/or UGT activities would alter the toxic effect of the subsequently administrated drug. All the results validated the utility of the established biomimetic microtissue in toxicological studies in vitro. Also, we anticipate the microfluidics-based bioengineering strategy would benefit liver tissue engineering and liver physiology/pathophysiology studies, as well as in vitro assessment of drug-induced hepatotoxicity.

Abstract:The merging of three-dimensional in vitro models with multi-organ-on-a-chip (MOC) technology has taken in vitro assessment of chemicals to an unprecedented level. By connecting multiple organotypic models, MOC allows for the crosstalk between different organs to be studied to evaluate a compound's safety and efficacy better than with single cultures. The technology could also improve the toxicological assessment of aerosols that have been implicated in the development of chronic obstructive pulmonary disease, asthma, or lung cancer. Here we report the development of a lung/liver-on-a-chip, connecting in a single circuit, normal human bronchial epithelial (NHBE) cells cultured at the air-liquid interface (ALI), and HepaRG™ liver spheroids. Maintenance of the individual tissues in the chip increased NHBE ALI tissue transepithelial electrical resistance and decreased HepaRG™ spheroid adenosine triphosphate content as well as cytochrome P450 (CYP) 1A1/1B1 inducibility. CYP inducibility was partly restored when HepaRG™ spheroids were cocultured with NHBE ALI tissues. Both tissues remained viable and functional for 28 days when cocultured in the chip. The capacity of the HepaRG™ spheroids to metabolize compounds present in the medium and to modulate their toxicity was proven using aflatoxin B1 (AFB1). AFB1 toxicity in NHBE ALI tissues decreased when HepaRG™ spheroids were present in the same chip circuit, proving that the HepaRG™-mediated detoxification is protecting/decreasing from AFB1-mediated cytotoxicity. The lung/liver-on-a-chip platform presented here offers new opportunities to study the toxicity of inhaled aerosols or to demonstrate the safety and efficacy of new drug candidates targeting the human lung.

Abstract:As a full-fidelity simulation of human cells, tissues, organs, and even systems at the microscopic scale, Organ-on-a-Chip (OOC) has significant ethical advantages and development potential compared to animal experiments. The need for the design of new drug high-throughput screening platforms and the mechanistic study of human tissues/organs under pathological conditions, the evolving advances in 3D cell biology and engineering, etc., have promoted the updating of technologies in this field, such as the iteration of chip materials and 3D printing, which in turn facilitate the connection of complex multi-organs-on-chips for simulation and the further development of technology-composite new drug high-throughput screening platforms. As the most critical part of organ-on-a-chip design and practical application, verifying the success of organ model modeling, i.e., evaluating various biochemical and physical parameters in OOC devices, is crucial. Therefore, this paper provides a logical and comprehensive review and discussion of the advances in organ-on-a-chip detection and evaluation technologies from a broad perspective, covering the directions of tissue engineering scaffolds, microenvironment, single/multi-organ function, and stimulus-based evaluation, and provides a more comprehensive review of the progress in the significant organ-on-a-chip research areas in the physiological state.

Abstract:Background: Organoids are three-dimensional structures that closely recapitulate tissue architecture and cellular composition, thereby holding great promise for organoid-based drug screening. Although growing in three-dimensional provides the possibility for organoids to recapitulate main features of corresponding tissues, it makes it incommodious for imaging organoids in two-dimensional and identifying surviving organoids from surrounding dead cells after organoids being treated by irradiation or chemotherapy. Therefore, significant work remains to establish high-quality controls to standardize organoid analyses and make organoid models more reproducible. Methods: In this study, the Z-stack imaging technique was used for the imaging of three-dimensional organoids to gather all the organoids' maximum cross sections in one imaging. The combination of live cell staining fluorescent dye Calcein-AM and ImageJ assessment was used to analyze the survival of organoids treated by irradiation or chemotherapy. Results: We have established a novel quantitative high-throughput imaging assay that harnesses the scalability of organoid cultures. Using this assay, we can capture organoid growth over time, measure multiple whole-well organoid readouts, and show the different responses to drug treatments. Conclusions: In summary, combining the Z-stack imaging technique and fluorescent labeling methods, we established an assay for the imaging and analysis of three-dimensional organoids. Our data demonstrated the feasibility of using organoid-based platforms for high-throughput drug screening assays.

Abstract:The effective Hamiltonian formalism is extended to vectorial electromagnetic waves in order to describe statistical properties of the field in reverberation chambers. The latter are commonly used in electromagnetic compatibility tests. As a first step, the distribution of wave intensities in chaotic systems with varying opening in the weak coupling limit for scalar quantum waves is derived by means of random matrix theory. In this limit the only parameters are the modal overlap and the number of open channels. Using the extended effective Hamiltonian, we describe the intensity statistics of the vectorial electromagnetic eigenmodes of lossy reverberation chambers. Finally, the typical quantity of interest in such chambers, namely, the distribution of the electromagnetic response, is discussed. By determining the distribution of the phase rigidity, describing the coupling to the environment, using random matrix numerical data, we find good agreement between the theoretical prediction and numerical calculations of the response.

Abstract:The understanding that systemic context and tissue crosstalk are essential keys for bridging the gap between in vitro models and in vivo conditions led to a growing effort in the last decade to develop advanced multi-organ-on-a-chip devices. However, many of the proposed devices have failed to implement the means to allow for conditions tailored to each organ individually, a crucial aspect in cell functionality. Here, we present two 3D-print-based fabrication methods for a generic multi-organ-on-a-chip device: One with a PDMS microfluidic core unit and one based on 3D-printed units. The device was designed for culturing different tissues in separate compartments by integrating individual pairs of inlets and outlets, thus enabling tissue-specific perfusion rates that facilitate the generation of individual tissue-adapted perfusion profiles. The device allowed tissue crosstalk using microchannel configuration and permeable membranes used as barriers between individual cell culture compartments. Computational fluid dynamics (CFD) simulation confirmed the capability to generate significant differences in shear stress between the two individual culture compartments, each with a selective shear force. In addition, we provide preliminary findings that indicate the feasibility for biological compatibility for cell culture and long-term incubation in 3D-printed wells. Finally, we offer a cost-effective, accessible protocol enabling the design and fabrication of advanced multi-organ-on-a-chip devices.

Abstract:3D organoid models have been widely used in drug evaluation because they effectively mimic physiological conditions and cell-cell interactions. However, current organoid-based drug evaluation heavily relies on invasive methods like qRT-PCR and immunofluorescence quantification, which disrupts the organoid's structure and physiological function during the evaluation process. Therefore, it is urgent to develop a non-destructive drug evaluation platform to continuously monitor organoid fate. Here, a non-destructive and real-time in situ platform based on field effect transistor sensor (NDRS-FET) was developed to monitor organoid fate and obtained accurate drug evaluation by conducting the dynamic models. Using liver organoid as the model, the transition kinetics of liver organoid fate in response to candidate drug compounds were recorded by the NDRS-FET platform. OSM, FH1 and BG45 were successfully identified as effective regulators of liver organoid fate. Compared to traditional methods, the NDRS-FET platform could not only accurately capture the drug effect on organoid in real time, but also reduce the intergroup deviation among samples, providing more accurate and reliable tool for drug development and screening.
