Abstract:The use of yeast antagonists to suppress postharvest fruit diseases and reduce economic losses has been attracting widespread attentions. In this study, the effectiveness of Meyerozyma caribbica isolated from Tibet soils against Aspergillus ochraceus and underlying mechanisms were investigated. M. caribbica exhibited great resistances under adverse environment and could obviously inhibit the growth and ochratoxin A (OTA) production of A. ochraceus via mycelium destruction and down-regulation of gene expressions involved in OTA synthesis. Besides, when grapes inoculated with A. ochraceus, disease incidence was reduced by both M. caribbica and its produced volatile organic compounds (VOCs). M. caribbica enhanced grape resistance to A. ochraceus by increasing resistance-related enzyme activities and gene expressions. VOCs were identified with the main compounds including 3-methyl-1-butanol, oxirane,2,2,3-trimethyl and 2-methyl-1-propanol. Accumulation of endogenous γ-aminobutyric acid (GABA) content was observed in grapes treated by yeast VOCs. Therefore, M. caribbica can be applied as a promising biocontrol agent for postharvest fruit loss control.

Abstract:The alteration of safety-secured yeast is a crucial step before scale applications. Based on previous studies, Pichia pastoris showed great potential and value to improve its biocontrol ability. The original sequences of antimicrobial peptide Ac-AMP2 and MiAMP1 were optimized according to the preference of Pichia pastoris and ligated into pPICZ alpha A plasmid which is emerged as a high-performance vector for transformation and expression. The results of RT-qPCR and Western blotting could imply that pPICZ alpha A/Ac-AMP2 and pPICZ alpha A/MiAMP1 were successfully overexpressed in Pichia pastoris GS115. The peptide concentration of GS115/Ac-AMP2 reached a maximum value of 210 mg L-1 at 60 h while GS115/MiAMP1 was 220 mg L-1 at 96 h. The biocontrol experiment indicated that the recombinant strain GS115/Ac-AMP2 and GS115/MiAMP1 could highly suppress the pathogen Penicillium expansum in vivo, which was respectively 42 % (GS115/Ac-AMP2) and 29.2 % (GS115/MiAMP1) of incidence disease lower than the sterile distilled water treatment. In the case of the experimental results considered, the modified GS115/Ac-AMP2 and GS115/MiAMP1 might be promising biological agent in postharvest applications.

Abstract:Aims This research sought to improve the ability of biocontrol yeast to suppress postharvest fungal disease and explore possible mechanisms of action. Methods and Results The addition of 2% sodium glutamate (SG), which is edible and recognized as safe, enhances the inhibitory effect of Rhodosporidium paludigenum Fell & Tallman on Penicillium expansum in vivo and in vitro. Rhodosporidium paludigenum cells grown in medium with a final concentration of 2% SG, displayed viability under a variety of stress conditions, including sodium chloride (NaCl), calcofluor white (CFW), Congo red (CR) and sodium dodecyl sulphate (SDS). Activity and relative gene expression levels of antioxidant-related enzymes in R. paludigenum, including peroxisomal catalase (CAT), thioredoxin reductase (TrxR), glutathione peroxidase (GSH-PX), glutathione reductase (GR) and superoxide dismutase (SOD) were altered in the presence of SG. Levels of reactive oxygen species (ROS) increased in cells grown in the presence of SG as well as the content of several amino acids. Conclusions In the presence of 2% SG R. paludigenum inhibited P. expansum and exhibited tolerance to a number of stressful conditions which may involve the upregulation of antioxidant enzymes and amino acids. Significance and Impact of the Study The ability of culture conditions to enhance the fungal suppressive abilities of yeast has the potential to enhance the management of postharvest disease in fruit.

Abstract:Postharvest spoilage fungi, such as Botrytis cinerea, are considered the main cause of losses of fresh fruit quality and vegetables during storage, distribution, and consumption. The current control strategy is the use of SO2 generator pads whose application is now largely under observation. A high quantity of SO2 can be deleterious for fresh fruits and vegetables and it is not allowed in organic agriculture. For this reason, great attention has been recently focused on identifying Biological Control Agents (BCA) to implement biological approaches devoid of chemicals. In this direction, we carried out our study in isolating five different non-Saccharomyces yeast strains from local vineyards in the South of Italy as possible BCA. We performed both in vitro and in vivo assays in semi-commercial conditions on detached grape berries stored at 0 °C, simulating the temperature normally used during cold storage, and obtained relevant results. We isolated three M. pulcherrima strains and one L. thermotolerans strain able to largely antagonize the development of the B. cinerea, at both in vitro and in vivo conditions. In particular, we detected the ability of the three isolates of M. pulcherrima strains Ale4, N20/006, and Pr7 and the L. thermotolerans strain N10 to completely inhibit (100% in reduction) the mycelial growth of B. cinerea by producing fungistatic compounds. We found, using an extracellular lytic enzymes activity assay, that such activity could be related to lipid hydrolyzation, β-1,3-glucanase and pectinase activity, and pectinase and protease activity, depending on the yeasts used. Results from our in vitro assays allowed us to hypothesize for M. pulcherrima strains Ale4 and N20/006 a possible combination of both the production of soluble metabolites and volatile organic compounds to antagonize against B. cinerea growth. Moreover, in semi-commercial conditions, the M. pulcherrima strain N20/006 and L. thermotolerans strain N10 showed relevant antagonistic effect also at low concentrations (with a significantly reduction of 'slip skin' incidence of 86.4% and 72.7%, respectively), thus highlighting a peculiar property to use in commercial development for organic agriculture and the handling process.

Abstract:Worldwide, microbial growth destroys large amounts of various products, causing yield losses in the agronomical and biotechnological industries. Traditionally, biocides have been used to manage these problems, but different disadvantages such as the establishment of resistant strains and the suppression of natural competitors have made alternatives such as biological control necessary. After harvest, many fruits are kept in cold storage to prolong their availability and shelf life. Often, this requires the application of a chemical fungicide to prevent postharvest decay from decay fungi. An alternative approach for preventing postharvest fungal decay during storage could be based on the treatment of the commodity with antagonistic yeasts. In this regard, the use of cold-adapted yeasts may offer a distinct advantage. Numerous cold-adapted yeasts species have been isolated from artificial cold environments, as well as cold-stored fruits. Recently, we isolated and identified epiphytic yeasts during the cold postharvest storage of pears and fine fruits from packinghouses in Argentinean Patagonia, and we tested their efficacy in controlling the postharvest diseases of different fruits caused by several pathogens. Additionally, killer yeasts as producers of mycocins or killer toxins that can neutralize the activities of spoilage yeasts in wines represent an interesting biocontrol strategy. Several screening studies focused to determine the occurrence of killer yeasts in winemaking environments have been carried out, and they have demonstrated the presence of killer phenotypes in yeasts from wines, cellar surfaces, and winery equipment. In previous studies carried out in our laboratory, most yeasts isolated from spontaneously fermenting grape musts evidenced killer character. These studies provide an exceptional source of potential antagonist yeasts to be used in biocontrol of undesired microorganisms in wine. Nonetheless, it is necessary to continue to identify new potential microorganisms and to develop a better understanding of the biology of yeast biocontrol systems to increase the potential of postharvest biocontrol as a viable alternative to synthetic postharvest fungicides and chemical preservatives against wine spoilage yeast.

Abstract:Changes in consumer expectations have led to increasing demand for novel plant protection strategies, in order to reduce the application of chemical products, reduce the occurrence of new pests and the impact that all these actions generate in the environment. In recent years there have been numerous investigations related to biological control and the use of microorganisms as new control strategies. As part of integrated disease management, antagonistic microorganisms have been investigated lately and presented great interest. Such microorganisms can be applied in conventional and in organic farming as biological control agents (BCA). Many of these microorganisms are present in the microbial ecology generating interactive associations between surrounding microorganisms. For these reasons, it has become necessary to search new natural antimicrobial agents as alternatives to synthetic and chemical products. It has been discovered that there are microorganisms, particularly yeasts, that have antagonistic activity and different mechanisms of action, indicating that they could be interesting candidates for the development of BCA. Here, we evaluate the antagonist effect of four endophytic yeast, Cryptococcus antarcticus, Aureobasidium pullulans, Cryptococcus terrestris and Cryptococcus oeirensis over the growth of Botrytis cinerea, Monilinia laxa, Penicillium expansum and Geotrichum candidum in in vitro assays (inhibition zone diameter assay and confrontation assay).The results revealed that the four yeast strains evaluated showed antagonistic activity against the phytopathogens tested, suggesting that these yeasts produce compounds capable of inhibiting the growth of fungi and, depending on the assay, the evaluated antagonist-yeasts have differential biocontrolling-effect against the postharvest pathogens tested.

Abstract:Post-harvest decay of fresh table grapes causes considerable annual production losses. The main fungal agents of decay both in pre- and post-harvest are B. cinerea, Penicillium spp., Aspergillus spp., Alternaria spp., and Cladosporium spp. To date, the use of agrochemicals and SO2 are the main methods to control grape molds in pre- and postharvest, respectively. Significant improvements, however, have already been made in to apply innovative and more environmentally sustainable control strategies, such as Biological Control Agents (BCAs), which can reduce disease severity in both pre- and post-harvest. In this study, 31 new non-Saccharomyces yeast strains, isolated from berries of native Apulian table grape genotypes, were tested for their in vivo effectiveness against grey mold of table grapes, resulting in two St. bacillaris (‘N22_I1’ and ‘S13_I3’), one S. diversa (‘N22_I3’), one A. pullulans (‘OLB_9.1_VL’) and one H. uvarum (‘OLB_9.1_BR’) yeast strains that were marked as efficient and good BCAs. Their mechanisms of action were characterized through in vitro assays, and additional characteristics were evaluated to assess the economic feasibility and viability for future technological employment. Their effectiveness was tested by reducing the working concentration, their antagonistic effect on a wide range of fungal pathogens, their ability to survive in formulations with long shelf life, and their safety to human health.

Abstract:Fungal pathogens in fruits and vegetables cause significant losses during handling, transportation, and storage. Biological control with microbial antagonists replacing the use of chemical fungicides is a major approach in postharvest disease control, and several products based on single antagonists have been developed but have limitations related to reduced and inconsistent performance under commercial conditions. One possible approach to enhance the biocontrol efficacy is to broaden the spectrum of the antagonistic action by employing compatible microbial consortia. Here, we explore commercial kefir grains, a natural probiotic microbial consortium, by culture-dependent and metagenomic approaches and observed a rich diversity of co-existing yeasts and bacterial population. We report effective inhibition of the postharvest pathogen Penicillium expansum on apple by using the grains in its fresh commercial and milk-activated forms. We observed few candidate bacteria and yeasts from the kefir grains that grew together over successive enrichment cycles, and these mixed fermentation cultures showed enhanced biocontrol activities as compared to the fresh commercial or milk-activated grains. We also report several individual species of bacteria and yeasts with biocontrol activities against Penicillium rots on apple and grapefruit. These species with antagonistic properties could be further exploited to develop a synthetic consortium to achieve enhanced antagonistic effects against a wide range of postharvest pathogens.

Abstract:Epiphytic yeasts were isolated from different cultivars of apples and lemons and identified by a combination of PCR-RFLP of 5.8S rRNA region and sequencing of D1/D2 domain of the 26S rRNA gene. Among 69 isolates, Aureobasidium pullulans GE17 and Meyerozyma guilliermondii KL3 strains showed the greatest antagonistic activity against two significant apple and lemon postharvest pathogens, Penicillium expansum DSM62841 (blue mold) and Penicillium digitatum DSM2750 (green mold), after preliminary screening. Yeasts were applied as single and mixed cultures with two different cell concentrations of 106 and 108 cells/ml in the present study. It was determined that antagonistic activity of two yeast strains studied emerged with a combination of several mechanisms of action including competition for space and nutrients, production of volatile organic compounds (VOCs), secretion of extracellular lytic enzymes and inhibition of fungal spore germination. The highest inhibition of mycelial growth on P. expansum DSM62841 and P. digitatum DSM2750 (83.4% and 74.7%, respectively) was achieved by utilization of single culture of A. pullulans GE17. Otherwise, the application of mixed culture at the ratio of 108 cells/ml inhibited spore germination of both pathogens from 86% to 95%. Results of this study suggest that an increase in yeast cell concentrations positively affected their biocontrol activity against blue and green molds. According to the results, employing single culture of M. guilliermondii KL3 did not exhibit effective antagonistic activity against blue and green molds. However, utilization of A. pullulans GE17 alone and mixed culture showed succesfull controlling against both P. expansum DSM62841 and P. digitatum DSM2750.
Biological and physical approaches to improve induced resistance against green mold of stored citrus fruit

Abstract:Health and environmental concerns have point out the need to improve or change several manufacturing steps in the food chain. In this context particular attention should be given to the technologies involved in fruits and vegetables production. Nearly all fresh fruit and vegetables are subjected to different periods of storage and/or shelf-life before of their consumption. This implies the need to protect the commodities from microbial spoilage. Some Citrus species (e.g. lemon and grapefruit) may be stored for several months before consumption and then post-harvest treatments are essential to contain green (Penicillium digitatum) and blue (P. italicum) moulds. Alternative approaches to chemicals usually have a lower efficacy in containing rots but fulfill the consumer's expectation. Among the alternative strategies, the improvement of host natural resistance is promising. In this regard, we report some results concerning the use of biotic (yeast) and abiotic agents as inducers of phytoalexin (i.e. scoparone and/or scopoletin) accumulation in Citrus rind and its importance in the control of fungal decay. In all experiments the inducers were applied on fruits before or 24 h after inoculation with P. digitatum and the rot severity was monitored 7 days later. The accumulation of phytoalexins was monitored according to a standard methodology by HPLC. In all experiments a positive correlation was found between increase of the phytoalexin scoparone in host tissue and reduction of decay.
Characterization of new yeast isolates collected from different fruits in Tunisia and biocontrol activity against Penicillium expansum on apples

Abstract:Biological control of fungal pathogens by antagonistic yeasts is increasingly becoming a valid strategy for a sustainable crop production and bioformulates are available on the market in some countries. However, in tropical and subtropical areas little research has focused on selection and development of local biocontrol yeasts adapted to such environments. To this aim, several yeasts were collected from the carposphere of different fruits in various locations in Tunisia. Representative isolates, selected for their fruit source and morphological characteristics, were identified by analysing their ITS regions and assayed for their biocontrol activity against the postharvest pathogen Penicillium expansum on stored apples, in comparison with three Italian yeast isolates already studied in depth as biocontrol agents. The selected yeasts were further characterized considering some important phenotypic features such as tolerance to abiotic stresses (oxidative, osmotic and UV-light stress), growth at different temperatures, compatibility with common agrochemicals and capacity to produce biofilm. Among the new investigated yeasts, seven isolates displayed a high biocontrol protection of apples from P. expansum (from 50 to 96% rot reduction). The most effective isolates were Cystobasidium minutum L3aid and Rhodotorula sp. Or1, reducing Penicillium rot of apples by 96% and 88%, respectively. These isolates and others, however, with good biocontrol activity, also showed remarkable resistance to environmental stresses and compatibility with common agrochemicals. Therefore, the selected isolates are worthy of further studies aimed at setting up new formulates to be used in biological or integrated control of fungal diseases of fruit and vegetables.

Abstract:The interaction of a number of potentially complementary monomers with poly(A) has been investigated by equilibrium dialysis, optical rotatory dispersion and ultraviolet absorption measurements. Experiments were conducted at pH 7.0, 0.15 M Na+, where poly(A) exists as a random coil with some degree of base-stacking, and at pH 6.0, 0.15 M Na+, where poly(A) adopts the protonated double-helical acid form structure below 15 degrees C. Binding isotherms show that, at 3.5 degrees C, poly(A) forms a 1 : 1 complex with xanthine at pH 6, and a 2:1 complex at pH 7, while oxoformycin forms a 1:1 complex with poly(A) at both pH 6 and pH 7. Poly(A) forms a complex, tentatively assigned 1:1 stoichiometry, with 8-azaxanthine at pH 6, but no complexing occurs at pH 7. The complexes have been characterized by their optical rotatory dispersion and ultraviolet spectra, their thermal stabilities, and their rates of formation at low temperature. All the complexes are laevorotatory at long wavelengths (greater than 300 nm) and unplex formation at low temperature is a slow process requiring many hours for completion. The complexes of poly(A) with 3-methylxanthine have been reinvestigated and shown to undergo normal helix-coil transitions; the anomalous melting behaviour noted previously [Biopolymers, 10, 21 -- 33 (1971)] has been explained. From a comparison of optical rotatory dispersion spectra, it is concluded that the poly(A) with 3-methylxanthine have similar structures, which are quite different from the structures of the corresponding complexes with 7-methylxanthine. The structures and properties of the poly(A) - monomer complexes are discussed, and compared with those of other polynucleotide - monomer complexes. No significant interaction was observed between poly(A) and hypoxanthine, allopurinol, 6,8-dihydroxypurine, 1-methylxanthine, 9-methylxanthine, theophylline, theobromine or 3,9-dimethylxanthine.
Isolation and Identification of Wild Yeast from Malaysian Grapevine and Evaluation of Their Potential Antimicrobial Activity against Grapevine Fungal Pathogens

Abstract:Pathogenic fungi belonging to the genera Botrytis, Phaeomoniella, Fusarium, Alternaria and Aspergillus are responsible for vines diseases that affect the growth, grapevine yield and organoleptic quality. Among innovative strategies for in-field plant disease control, one of the most promising is represented by biocontrol agents, including wild epiphytic yeast strains of grapevine berries. Twenty wild yeast, isolated and molecularly identified from three different Malaysian regions (Perlis, Perak and Pahang), were evaluated in a preliminary screening test on agar to select isolates with inhibition against Botrytis cinerea. On the basis of the results, nine yeasts belonging to genera Hanseniaspora, Starmerella, Metschnikowia, Candida were selected and then tested against five grape berry pathogens: Aspergillus carbonarius, Aspergillus ochraceus, Fusarium oxysporum, Alternaria alternata and Phaeomoniella chlamydospora.Starmerella bacillaris FE08.05 and Metschnikowia pulcherrima GP8 and Hanseniaspora uvarum GM19 showed the highest effect on inhibiting mycelial growth, which ranged between 15.1 and 4.3 mm for the inhibition ring. The quantitative analysis of the volatile organic compound profiles highlighted the presence of isoamyl and phenylethyl alcohols and an overall higher presence of low-chain fatty acids and volatile ethyl esters. The results of this study suggest that antagonist yeasts, potentially effective for the biological control of pathogenic moulds, can be found among the epiphytic microbiota associated with grape berries.

Abstract:<section class="article-section__content"><h3 class="article-section__sub-title section1"> Aims</h3><p><i>Alternaria alternata</i> is a major contaminant of wine grapes, meaning a health risk for wine consumers due to the accumulation of toxic metabolites. To develop a successful biofungicide, the effectiveness of epiphytic wine grape yeasts against <i>A. alternata</i> growth and toxin production was assessed <i>in vitro</i> under temperature and a<sub>W</sub> conditions that simulate those present in the field. </p></section><section class="article-section__content"><h3 class="article-section__sub-title section1"> Methods and results</h3><p>The effect of 14 antagonistic yeasts was evaluated on growth and alternariol (AOH), alternariol monomethyl ether (AME) and tenuazonic acid (TA) production by three <i>A. alternata</i> strains in a synthetic medium with composition similar to grape (SN) at three temperatures (15, 25 and 30 °C). All <i>Metschnikowia</i> spp. yeast strains evaluated completely prevented <i>A. alternata</i> growth and mycotoxin production at all temperatures in SN medium. Meanwhile, the growth inhibition exerted by <i>Starmerella bacillaris</i> yeast strains was higher at 30 °C, followed by 25 and 15 °C, being able to show a stimulating or inhibiting effect. <i>Hanseniaspora uvarum</i> yeast strains showed a growth promoting activity higher at 15° C, followed by 25 and 30 °C. Even at conditions where <i>A. alternata</i> growth was stimulated by the <i>S. bacillaris</i> and <i>H. uvarum</i> yeasts, high inhibitions of mycotoxin production (AOH, AME and TA) were observed, indicating a complex interaction between growth and mycotoxin production. </p></section><section class="article-section__content"><h3 class="article-section__sub-title section1"> Conclusion</h3><p>There is a significant influence of temperature on the effectiveness of biocontrol against <i>A. alternata</i> growth and mycotoxin production. <i>Metschnikowia</i> spp. strains are good candidates to compose a biofungicide against <i>A. alternata</i>. </p></section><section class="article-section__content"><h3 class="article-section__sub-title section1"> Significance and Impact of the study</h3><p>Among the different antagonistic yeasts evaluated, only <i>Metschnikowia</i> spp. strains were equally effective reducing <i>A. alternata</i> growth and mycotoxin at different temperatures underlining the importance of considering environmental factors in the selection of the antagonists. </p></section>

Selection and application of antifungal VOCs-producing yeasts as biocontrol agents of grey mould in fruits

Abstract:Rotting caused by grey mould (Botrytis cinerea) is a concerning disease for numerous crops both pre- and postharvest stages. Application of antagonistic yeasts is a promising strategy for controlling grey mould incidence which could mitigate undesirable consequences of using synthetic fungicides. In this work, a screening for detection of yeasts isolated from figs producers of antifungal volatile organic compounds (VOCs) were performed by confrontation in double dishes systems. Eleven out of 34 yeasts confronted reduced B. cinerea growth parameter in vitro. This reduction was correlated (p ≤ 0.050) with the production of 10 volatile compounds: two acids (acetic acid and octanoic acid), 7 esters (Ethyl propionate, n-Propyl acetate, Isobutyl acetate, 2-methylbutyl acetate, furfuryl acetate, phenylmethyl acetate, 2-phenylethyl acetate) and one ketone (Heptan-2-one). In bases on in vitro assay, Hanseniaspora uvarum 793 was applied to in vivo assays with strawberries and cherries. The reduction of incidence of B. cinerea in strawberries at 7 °C and 25 °C was 54.9 and 72.1% after 6 and 3 days, respectively. The reduction of incidence of B. cinerea in cherries at 7 °C and 25 °C was 48.9 and 45.6% after 5 and 4 days, respectively. These results showed that VOCs produced by Hanseniaspora uvarum 793 are effective in the control of incidence of Botrytis cinerea in fruits, being a potential alternative to chemical fungicide.
Screening Methods for Isolation of Biocontrol Epiphytic Yeasts against Penicillium digitatum in Lemons

Abstract:Worldwide, the green rot caused by Penicillium digitatum is one of the most aggressive postharvest diseases of lemons. Searching for sustainable alternatives to chemical fungicides, epiphytic yeasts as potential biocontrol agents were isolated from citrus fruits using a tailor-made selective medium. For disclosing their antagonistic potential against P. digitatum, obtained isolates were subjected to direct screening methods, both in vitro and in vivo. In the course of the primary in vitro screening that comprised dual culture assays, 43 yeast strains displaying antagonistic activities against the pathogen were selected. Subsequently, such strains were subjected to an in vivo screening that consisted of a microscale test, allowing the selection of six yeast strains for further analysis. In the final screening using macroscale in vivo tests, three strains (AcL2, AgL21, and AgL2) displaying the highest efficiencies to control P. digitatum were identified. The protection efficiencies in lemons were 80 (AcL2), 76.7 (AgL21), and 75% (AgL2). Based on sequence analysis of the PCR amplified D1/D2 domains of the 26S rRNA genes, they were identified as representatives of the species Clavispora lusitaniae. Interestingly, the strains exhibited a broad action spectrum among citrus fruits as they were also able to combat the green mold disease in grapefruit and two orange varieties. The direct screening methods applied in this study favored the recovery of efficient candidates for application as biological control agents to combat fungal infestations of citrus fruits.

Abstract:Significant losses in harvested fruit can be directly attributable to decay fungi. Some of these pathogenic fungi are also the source of mycotoxins that are harmful to humans. Biological control of postharvest decay of fruits, vegetables and grains using antagonistic yeasts has been explored as one of several promising alternatives to chemical fungicides, the use of which is facing increasingly more stringent regulation. Yeast species have been isolated over the past two decades from a variety of sources, including fruit surfaces, the phyllosphere, soil and sea water, and their potential as postharvest biocontrol agents has been investigated. Several mechanisms have been proposed as responsible for their antagonistic activity, including competition for nutrients and space, parasitism of the pathogen, secretion of antifungal compounds, induction of host resistance, biofilm formation, and most recently, the involvement of reactive oxygen species (ROS) in defense response. It has been recognized that a biocontrol system is composed of a three-way interaction between the host (commodity), the pathogen and the yeast, all of which are affected by environmental factors. Efficacy and consistent performance in controlling postharvest diseases are the hurdles that must be overcome if the use of yeast biocontrol agents and other alternatives are to be widely used commercially. Therefore, attempts have been made to combine alternative treatments in order improve their overall performance. The current review provides a brief overview of the topic of the use of yeasts as postharvest biocontrol agents and includes information on the sources from which yeast antagonists have been isolated, their mode of action, and abiotic stress resistance in yeast as it relates to biocontrol performance. Areas in need of future research are also highlighted.

Postharvest biological control of Botrytis cinerea and the mechanisms underlying the induction of disease resistance in grapes by Lactobacillus plantarum CM-3

Abstract:Biological control by microbial antagonists has been proposed as a viable strategy for the control of postharvest diseases in fruit. In the present study, biocontrol efficacy of the bacterial antagonist Lactobacillus plantarum CM-3 against gray mold infection on harvested grapes was documented along with the underlying physiological and molecular mechanisms responsible for biocontrol activity. In this regard, a transcriptomic analysis of the response of grapes to L. plantarum CM3 was conducted. Results indicated that L. plantarum CM-3 significantly decreased the level of natural postharvest decay in grapes without causing any adverse effects on quality traits. Treatment of grapes with L. plantarum CM-3 (1 × 109 CFU mL−1) resulted in a 55 % reduction in disease incidence caused by Botrytis cinerea. Meanwhile, L. plantarum CM-3 enhanced the activity of several enzymes in grapes that are closely associated with defense, including POD, PPO, CAT, APX, PAL, and GLU. RNA-seq analysis indicated extensive changes in the gene expression profile of grape berries in response to being treated with L. plantarum CM-3. In total, 1,327 differentially expressed genes (DEGs) were identified, among which 78.1 % (1,037) were upregulated and 21.9 % (290) were downregulated. Numerous pathways were strongly influenced by the L. plantarum CM-3 treatment, including signal perception and transduction, maintenance of reactive oxygen species (ROS) homeostasis, transcriptional and post-transcriptional regulation, primary metabolic pathways, and secondary metabolism. Additionally, the expression of 22 DEGs linked to enhancing disease resistance was analyzed by RT-qPCR and their expression levels were found to well correspond with the results obtained in the analysis of the RNA-seq data. Our data indicate that L. plantarum CM-3 can enhance host resistance by altering the expression of defense genes and their corresponding proteins, including PR1, WRKY33, WRKY22, PTI6 and peroxidase isoenzyme. Collectively, results of our study indicate that L. plantarum CM-3 treatment enhances the disease resistance of harvested grapes through a comprehensive and complex regulatory network. These findings provide new information on the molecular mechanism responsible for the enhancement of host disease resistance in grapes by L. plantarum CM-3. It also demonstrates a biocontrol strategy that can be used to elicit defense responses in fruit against fungal pathogens.

Abstract:Pathogenic fungi can cause significant annual losses in the quality and quantity of fruits belonging to Rosaceae family during pre‐ and postharvest. Synthetic fungicides are considered the main method for controlling diseases in food crops; however, they pose a great risk to human health and to the environment. As a result, biological control methods, which are based on living microorganisms to reduce the population or to inhibit the growth of pathogens, have arisen as a safe alternative. In this review, we highlight the principal mechanisms (e.g., nutrition and space competition, mycoparasitism, induced resistance and growth enhancement, production of volatile organic compounds, and antibiosis) utilized by the most frequently reported non‐Saccharomyces biocontrol agents (BCAs) yeasts: Meyerozyma guilliermondii, Pichia membranifaciens, Wickerhamomyces anomalus, Metschnikowia pulcherrima, Hanseniaspora uvarum, and Aureobasidium pullulans, against the common pathogenic fungi found within Rosaceae family: Penicillium expansum, Botrytis cinerea, and Monilinia spp. Understanding BCAs' importance and mode of action is a necessary step in order to reduce the detrimental effect of spoilage fungi in the agriculture and food industry, by achieving a more sustainable and safer control of them.
