Abstract:Aims: The purpose of this study was to investigate the possible mechanism(s) of saponins from Panax japonicus (SPJ) on alcohol-induced hepatic damage in mice. Methods: SPJ were identified by high performance liquid chromatography-evaporative light scattering detection-mass spectrometry (LCELSDMS). Non-toxic concentrations of SPJ were assayed on alcohol-induced hepatic injury in male ICR mice and human hepatic cells. The protective effects were evaluated by biochemical values, histopathological observations and the relative gene expression. Results. In vitro, SPJ showed significant hydroxyl radical scavenging capacity. In vivo, SPJ (50mg/kg) could rectify the pathological changes of aspartate transaminase, alanine transaminase, malondialdehyde, reduced glutathione (GSH), glutathione peroxidase (GPX), catalase (CAT) and superoxide dismutase (SOD) caused by alcohol metabolism to normal levels except for hepatic GSH and CAT. In hepatic cells, the results were in agreement with foregoing results determined in mice after pretreatment of SPJ (100g/ml). RTPCR results showed that CAT, GPX and SOD mRNA decreased by alcohol metabolism were reversed, in which GPX3, SOD1 and SOD3 could return to a normal level, but CAT, GPX1 and SOD2 mRNA were still evidently lower than the control. Histopathological observations provided supportive evidence for biochemical analyses. Conclusions: SPJ plays an important role in the protection of the structure and function of hepatic mitochondria and karyon by directly scavenging reactive oxygen species/free radicals and up-regulating the expression of antioxidant enzymes (SOD, GPX and CAT), especially to GPX3, SOD1 and SOD3.

Abstract:Polysaccharides are rich in Panax notoginseng residue after extraction. This study aims to explore the structural characteristics of PNP‐20, which is a homogeneous polysaccharide, separated from P. notoginseng residue by fractional precipitation and evaluate the anti‐enteritis effect of PNP‐20. The structure of PNP‐20 was determined by spectroscopic analyses. A mouse model with enteritis induced by restraint stress (RS) and lipopolysaccharide (LPS) was used to evaluate the pharmacological effect of PNP‐20. The results indicated that PNP‐20 consisted of glucose (Glc), galactose (Gal), Mannose (Man) and Rhamnose (Rha). PNP‐20 was composed of Glcp‐(1→, →4)‐α‐Glcp‐(1→, →4)‐α‐Galp‐(1→, →4,6)‐α‐Glcp‐(1→, →4)‐Manp‐(1→ and →3)‐Rhap‐(1→, and contained two backbone fragments of →4)‐α‐Glcp‐(1→4)‐ α‐Glcp‐(1→ and →4)‐α‐Galp‐(1→4)‐α‐Glcp‐(1→. PNP‐20 reduced intestinal injury and inflammatory cell infiltration in RS‐ and LPS‐induced enteritis in mice. PNP‐20 decreased the expression of intestinal tumor necrosis factor‐α, NOD‐like receptor family pyrin domain containing 3, and nuclear factor‐κB and increased the expression of intestinal superoxide dismutase 2. In conclusion, PNP‐20 may be a promising material basis of P. Notoginseng for the treatment of inflammatory bowel disease.

Abstract:Polygonum perfoliatum L. has a long history of medicinal and edible applications. Studies have shown that it can significantly protect liver injury, but the mechanism is unclear. The purpose of this study was to explore the protective mechanism of P. perfoliatum on chronic alcoholic liver injury from the perspective of lipid metabolism. After 8 weeks of alcohol exposure in male Wister mice, the levels of aspartate aminotransferase (AST), alanine aminotransferase (ALT) and alkaline phosphatase (ALP) in serum were significantly increased, and the activities of alcohol dehydrogenase (ADH) and acetaldehyde dehydrogenase (ALDH) in liver were significantly decreased. Meanwhile, pathological changes of liver tissue in mice were observed by histopathology. Then, Ultra-High Performance Liquid Chromatography (UHPLC) QExactive Plus Mass Spectrometer lipidomics and matrix-assisted laser desorption/ionization time-of-flight/time -of-flight (MALDI-TOF/TOF) mass spectrometry imaging methods were established to analyze lipid metabolism in mice. Ten different lipids were identified by statistical analysis, including Fatty Acyls, Glycerophospholipids, Prenol lipids and Sphingomyelins. After intervention with P. perfoliatum extracts at different doses (25 to 100 mg/kg), levels of AST, ALT, ALP in serum, and activities of ADH and ALDH in liver were significantly corrected. The hepatic cord structure was clear, and the liver cells were closely arranged without other obvious abnormalities. Non-target lipidomics analysis showed that P. perfoliatum extract could regulate the metabolic disorders of the 10 different lipids caused by continuous alcohol exposure. Pathway analysis suggested that the mechanism of P. perfoliatum extract on chronic alcoholic liver injury may be related to the regulation of linoleic acid and α-linolenic acid.

Abstract:This study aims to establish the isolation and purification method of polysaccharides from medicinal residue of Panax notoginseng (PPN). The structure and protective effect of PPN on myelosuppression mice were investigated. One neutral polysaccharide (NPPN) and five acidic polysaccharides (APPN I, APPN II-A, APPN II-B, APPN III-A, and APPN III-B) were obtained. The results confirmed that NPPN, APPN I and APPN II-A are glycan with 1, 4 main chains. APPN III-A is a glycan. APPN II-B and APPN III-B are homogalacturonan pectin with 1, 4 main chains. This study demonstrated that NPPN played a bone marrow protective role in myelosuppression mice induced by cyclophosphamide. NPPN could relieve cell cycle arrest, reduce the apoptosis rate of marrow cells, and improve granulocyte-macrophage colony-stimulating (GM-CSF), thermoplastic polyolefin (TPO) and erythropoietin (EPO) serum level, which contributes to promoting the proliferation of hematopoietic cells.

Abstract:Radix Pueraria lobata can be used as medicine and food, whose polysaccharide is one of the main bioactive ingredients. To explore the effect and mechanism of Pueraria lobata polysaccharide, a homogeneous and novel water-soluble polysaccharide (PLP1) was successfully isolated and purified from P. lobata by column chromatography in the current study. Structure analysis revealed that PLP1 (M<sub>w</sub> = 10.43 kDa) was constituted of the residues including (1 → 4)-α-d-glucose and (1 → 4, 6)-α-d-glucose, which were linked together at a ratio of 5:1 and represented the main glycosidic units. In vitro experiments indicated that PLP1 exhibited a better free radical-scavenging ability than amylose and amylopectin, meanwhile in vivo experiments indicated that PLP1 effectively protected against liver injury in mice with acute ALD through significantly inhibiting oxidative stress to regulate lipid metabolism, increasing short-chain fatty acid production, and maintaining intestinal homeostasis by regulating intestinal flora. Taken together, our results illustrate that PLP1 can regulate intestinal microecology as a feasible therapeutic agent for protecting against ALD on the ground of the gut-liver axis, thus laying a theoretical foundation for the rational exploitation and utilization of P. lobata resources in the clinic.

Abstract:[Objective]To study the protective effect of polysaccharides from Rhodomytus fomenfosa (Ait.) Hassk. on acute liver injury in rats. [Method]The effects of R. fomenfosa polysaccharides on the content of ALT,AST,MDA,SOD and GSH-PX in serum of acute liver injury rats induced by D-GalN were respectively investigated. [Result] Each dose of R. fomenfosa polysaccharides could significantly reduce the content of ALT,AST,MDA and increase the content of SOD,GSH-P. [Conclusion]R. fomenfosa polysaccharides have good protective effects on acute liver injury in rats induced by D-GalN.

Abstract:Ethnopharmacological relevance Polygala fallax Hemsl. is a traditional folk medicine commonly used by ethnic minorities in the Guangxi Zhuang Autonomous Region, and has a traditional application in the treatment of liver disease. Polygala fallax Hemsl . polysaccharides (PFPs) are of interest for their potential health benefits. Aim of this study This study explored the impact of PFPs on a mouse model of cholestatic liver injury (CLI) induced by alpha-naphthyl isothiocyanate (ANIT), as well as the potential mechanisms. Materials and methods A mouse CLI model was constructed using ANIT (80 mg/kg) and intervened with different doses of PFPs or ursodeoxycholic acid. Their serum biochemical indices, hepatic oxidative stress indices, and hepatic pathological characteristics were investigated. Then RNA sequencing was performed on liver tissues to identify differentially expressed genes and signaling pathways and to elucidate the mechanism of liver protection by PFPs. Finally, Quantitative real-time polymerase chain reaction (qRT-PCR) and Western blotting were used to verify the differentially expressed genes. Results Data analyses showed that PFPs reduced the levels of liver function-related biochemical indices, such as ALT, AST, AKP, TBA, DBIL, and TBIL. PFPs up-regulated the activities of SOD and GSH, down-regulated the contents of MDA, inhibited the release of IL-1β, IL-6, and TNF-α, or promoted IL-10. Pathologic characterization of the liver revealed that PFPs reduced hepatocyte apoptosis or necrosis. The RNA sequencing indicated that the genes with differential expression were primarily enriched for the biosynthesis of primary bile acids, secretion or transportation of bile, the reactive oxygen species in chemical carcinogenesis, and the NF-kappa B signaling pathway. In addition, the results of qRT-PCR and Western blotting analysis were consistent with those of RNA sequencing analysis. Conclusions In summary, this study showed that PFPs improved intrahepatic cholestasis and alleviated liver damage through the modulation of primary bile acid production, Control of protein expression related to bile secretion or transportation, decrease in inflammatory reactions, and inhibition of oxidative pressure. As a result, PFPs might offer a hopeful ethnic dietary approach for managing intrahepatic cholestasis.

Abstract:OBJECTIVE: To study the protective effect of saponins from Panax japonicus against ethanol-induced hepatic cells L-O2 injury and their possible mechanisms. METHODS: Saponins from P. japonicus was purified by HPLC-evaporation light scattering detection (ELSD). The survival rate of hepatic cells was determined by MTT assay. The level of malondialdehyde (MDA), the activities of superoxide dismutase (SOD) and glutathione peroxidase (GSH-Px) in L-O2 cells were measured by spectrophotometer. RESULTS: Six major saponins from P. japonicus were identified by HPLC-ESI-MS: ginsenosides Rg 1, Re, Rf, F 3, Rg 2 and Rd. Rg 1 0.16 g·L -1, Re 1.28 g·L -1 and Rf 0.64 g·L -1 could promote normal cells L-O2 proliferation (P < 0.01), at the rate of 22.7%, 34.8% and 28.5%, respectively. Rd 0.16 g·L -1 and F 3 1.28 g·L -1 showed evident inhibition to normal cells L-O2 (P < 0.01), at the rate of 49.7% and 43.3%. Rg 1 0.16 g·L -1, Re 1.28 g·L -1 and Rf 0.64 g·L -1 also showed significantly protection against ethanol-induced hepatic cells L-O2 injury (P < 0.01). The rate of inhibition decreased to 23.3%, 26.9% and 26.6% compared with the ethanol-induced model control, the inhibition rate of which was 50.4%. Rd 0.16 g·L -1 and F 3 1.28 g·L -1 showed more evident toxicity to ethanol-induced hepatic cells L-O2. The inhibition rate was 83.2% and 64.8% (P < 0.01), respectively. Meanwhile, the content of MDA and the activity of SOD and GSH-Px in ethanol-induced L-O2 cells were (1.35 ± 0.05) mmol·L -1, (26.4 ± 2.8) kU·L -1 and (67.0 ± 4.3) kU·L -1, respectively. Rg 1 0.16 g·L -1, Re 1.28 g·L -1 and Rf 0.64 g·L -1 could decrease the level of intracellular MDA to (1.17 ± 0.04), (1.21 ± 0.03) and (1.21 ± 0.05) mmol·L -1 (P < 0.01), increase the activity of intracellular SOD and GSH-Px activity to 38.3 ± 4.8, 35.6 ± 4.4 and (39.0 ± 3.3) kU·L -1 (P < 0.01), and 86.3 ± 5.7, 78.4 ± 3.5 and (84.2 ± 4.4) kU·L -1 (P < 0.01), respectively. CONCLUSION: Rg 1, Re and Rf have evidently protective effect against ethanol-induced hepatic cells L-O2 injury. The possible mechanism may be related to the decline of MDA content and increase in antioxidant enzymes such as SOD and GSH-Px activities.

Abstract:Objective Previous investigation demonstrated that PNS could protect many kinds of tissue against oxidative and inflammatory damage.In this study we observed the protection of PNS on liver damage,and discuss its potential mechanisms on this effect.Methods HepG2 cell already treated with 10-7-102 mg/ml of PNS was incubated with 0.6 mmol/L H2O2,and its livingness was detected by MTT method.The rats were treated with PNS,then were injected with 10% CCl4 abdominally,and after the injection of CCl4,their ALT,AST and TBil within serum were observed in time point of 24,48,72 h.Finally we studied the biopsy of the rat liver.The mRNA expression of HepG2 cell incubated with 10-3 mg/ml PNS was examined by human DNA Chips analysis system.Results By MTT method the absorbency of HepG2 cell with PNS treated was higher than its control cell which treated only with PBS.PNS could restrain the rise of ALT,AST and total bilirubin within serum when the rat liver was damaged by CCl4,and biopsy examination showed that PNS could decrease the denaturation,necrosis and inflammation.The mRNA microarray showed that PNS could affect expression of about 105 kinds of genes.Conclusions PNS can protect hepatocyte from damage induced by H2O2,can help rat against hepatic damage by CCl4,and it can probably exert protective effect by controlling the expression of some genes.

Abstract:Introduction: Ganshu Nuodan is a liver-protecting dietary supplement composed of Ganoderma lucidum ( G. lucidum ) spore powder, Pueraria montana (Lour.) Merr. ( P. montana ), Salvia miltiorrhiza Bunge ( S. miltiorrhiza ) and Astragalus membranaceus (Fisch.) Bunge. ( A. membranaceus ). However, its pharmacodynamic material basis and mechanism of action remain unknown. Methods: A mouse model of acute alcohol liver disease (ALD) induced by intragastric administration of 50% alcohol was used to evaluate the hepatoprotective effect of Ganshu Nuodan. The chemical constituents of Ganshu Nuodan were comprehensively identified by UPLC-QTOF/MS, and then its pharmacodynamic material basis and potential mechanism of action were explored by proteomics and network pharmacology. Results: Ganshu Nuodan could ameliorate acute ALD, which is mainly manifested in the significant reduction of alanine aminotransferase (ALT) and aspartate aminotransferase (AST) in serum and malondialdehyde (MDA) content in liver and the remarkably increase of glutathione (GSH) content and superoxide dismutase (SOD) activity in liver. Totally 76 chemical constituents were identified from Ganshu Nuodan by UPLC-QTOF/MS, including 21 quinones, 18 flavonoids, 11 organic acids, 7 terpenoids, 5 ketones, 4 sterols, 3 coumarins and 7 others. Three key signaling pathways were identified via proteomics studies, namely Arachidonic acid metabolism, Retinol metabolism, and HIF-1 signaling pathway respectively. Combined with network pharmacology and molecular docking, six key targets were subsequently obtained, including Ephx2, Lta4h, Map2k1, Stat3, Mtor and Dgat1. Finally, these six key targets and their related components were verified by molecular docking, which could explain the material basis of the hepatoprotective effect of Ganshu Nuodan. Conclusion: Ganshu Nuodan can protect acute alcohol-induced liver injury in mice by inhibiting oxidative stress, lipid accumulation and apoptosis. Our study provides a scientific basis for the hepatoprotective effect of Ganshu Nuodan in acute ALD mice and supports its traditional application.

Abstract:Penthorum chinense Pursh (P. chinense), a functional food, has been applied to protect the liver against alcohol-related fatty liver disease (ALD) for a long history in China. This study was designed to evaluate the ameliorative activity of the polyphenolic fraction in P. chinense (PGF) depending on the relief of ALD. The ALD mouse model was established by exposing the mice to a Lieber-DeCarli alcohol liquid diet. We found that PGF administration significantly ameliorated alcohol-induced liver injury, steatosis, oxidative stress, and inflammation in mice. Furthermore, alcohol-increased levels of the critical hepatic lipid synthesis proteins sterol regulatory element binding transcription factor (SREBP-1) and diacylglycerol o-acyltransferase 2 (DGAT2) were attenuated by PGF. Similarly, PGF inhibited the expression of the lipid transport protein very low-density lipoprotein receptor (VLDLR). Interestingly, PGF restored alcohol-inhibited expression of carnitine palmitoyltransferase 1 (CPT1) and peroxisome proliferator-activated receptor alpha (PPARα), essential fatty acid β-oxidation proteins. Mechanistic studies revealed that PGF protects against alcohol-induced hepatocyte injury and lipid deposition via the SIRT1/AMPK signaling pathway. In sum, this research clearly demonstrated the protective effects of PGF against ALD, which was mediated by activating SIRT1/AMPK pathways in hepatocytes. We provide a new theoretical basis for using P. chinense as a functional food in ALD.

Abstract:Ethnopharmacological relevance According to the theory of traditional Chinese medicine, the main factors related to alcoholic liver disease (ALD) are qi stagnation and blood stasis of the five viscera. Previously, we showed that the bioactive components of Alhagi honey have various pharmacological effects in treating liver diseases, but the influence of Alhagi honey on ALD (and its mechanism of action) is not known. Aim of the study To determine the efficacy of the main active component of Alhagi honey, the polysaccharide AHPN80, in ALD and to explore the potential mechanism of action. Materials and methods AHPN80 was isolated from dried Alhagi honey and identified by transmission electron microscopy , Fourier-transform infrared spectroscopy, and gas chromatography. Venous blood, liver tissue, and colon tissue were collected in a mouse model of alcohol-induced acute liver injury. Histology, staining (Oil Red O, Alcian Blue–Periodic Acid Schiff) and measurement of reactive oxygen species (ROS) levels were used to detect histopathologic and lipid-accumulation changes in the liver and colon. Lipopolysaccharide (LPS) levels and the content of proinflammatory cytokines in serum were measured by enzyme-linked immunosorbent assays. Commercial kits were employed to detect biochemistry parameters in serum and the liver. A terminal deoxynucleotidyl transferase dUTP nick end labeling (TUNEL) staining kit was used to identify hepatocyte apoptosis. Expression of tight junction-associated proteins in colon tissues and nuclear factor erythroid 2-related factor 2/heme oxygenase-1/toll-like receptor-4/mitogen-activated protein kinase (Nrf2/HO-1/TLR4/MAPK) pathway-related proteins in liver tissues and HepG2 cells were analyzed by immunofluorescence or western blotting. Results In a mouse model of alcohol-induced acute liver injury, AHPN80 therapy: significantly improved liver parameters (cytochrome P450 2E1, alcohol dehydrogenase, aldehyde dehydrogenase , superoxide dismutase, malondialdehyde, glutathione peroxidase , catalase, total cholesterol, triglycerides, alanine transaminase , aspartate transaminase); reduced serum levels of LPS, interleukin (IL)-1β, IL-6, and tumor necrosis faction-α; increased levels of IL-10 and interferon-gamma. AHPN80 reduced ALD-induced lipid accumulation and ROS production, improved alcohol-induced inflammatory damage to hepatocytes, and inhibited hepatocyte apoptosis. Immunofluorescence staining and western blotting suggested that AHPN80 might eliminate hepatic oxidative stress by activating the Nrf2/HO-1 signaling pathway, repair the intestinal barrier, inhibit the LPS/TLR4/MAPK signaling pathway, and reduce liver inflammation. Conclusions AHPN80 may activate the Nrf2/HO-1 pathway to eliminate oxidative stress, protect the intestinal barrier, and regulate the TLR4/MAPK pathway to treat ALD in mice. AHPN80 could be a functional food and natural medicine to prevent ALD and its complications.

Abstract:Introduction: Polysaccharides are important components of Panax notoginseng that contribute to its immunomodulatory ability. This study aimed to isolate polysaccharides from notoginseng and investigate the structural feature and potential immunomodulatory activity. Methods: The polysaccharide was isolated from notoginseng by anion exchange and gel permeation chromatography. Its preliminary structure was characterized by Fourier transform infrared (FT-IR) spectroscopy, gas chromatography-mass spectrometry (GC-MS) and nuclear magnetic resonance (NMR) spectroscopy. The immunoregulatory function was further investigated in cyclophosphamide induced immunosuppressive mice, murine splenocytes and macrophages. Results: A novel homogeneous polysaccharide (PNPB1) was isolated from notoginseng with the molecular weight of 9.3 × 10 5 Da. Monosaccharide composition analysis indicated that PNPB1 consisted of Glc (88.2%), Gal (9.0%), Ara (2.4%) and trace GlcA, with the major backbone of (1→4)-linked α-Glcp, (1→6)-linked β-Glcp, and (1, 4→6)-linked β-Glcp. The polysaccharide was found to significantly enhance murine body weight, improve their thymus and spleen indices and increase the white blood cells (WBC). PNPB1 significantly enhanced splenic lymphocyte proliferation, NO and cytokine (TNF-α, IL-2, IL-10 and IFN-γ) production, as well as the phagocytosis and TLR2 expression of peritoneal macrophages, indicating potent immunoenhancement effect. Discussion: These findings provide a theoretical basis for elucidating the structure and immune activity of notoginseng polysaccharides.

Abstract:Ethnopharmacological relevance Penthorum chinense Pursh (PCP) has acknowledged as an edible herbal medicinal plant for the prevention and treatment of alcoholic liver injury (ALI). However, only few of researches focus on the chemical material basis and potential mechanisms of PCP against ALI. Aim of the study Herein, we explored the therapeutic effects of PCP extract against ALI based on the integration of network pharmacology, molecular docking, and experiment validation. Methods Based on the standard quality control of PCP herbs by UPLC fingerprint and quantitative determination, 80% ethanol extract fraction of PCP containing more polyphenols, compared to aqueous extract fraction of PCP, were chosen for further experiments. After oral administration of PCP ethanol extract, serum pharmacochemistry based on UPLC-Q-Exactive-MS analysis was implemented to evaluate the potential effective compounds. These absorbed prototypes in PCP were used to construct network pharmacology and predict the potential mechanisms of PCP extract against ALI. Then, the predicted targets and biological mechanisms of PCP extract were validated using animal experiments and molecular docking analysis. Results Although totally 19 polyphenol compounds were identified in PCP ethanol extract by UPLC-MS analysis, only 18 absorbed prototypes were found in the serum collected from mice at 1 h post-administration with PCP extract. These candidate active compounds were further screened into 13 compounds to construct network pharmacology and 433 targets were identified as PCP targets. GO and KEGG pathway enrichment analyses indicated that the effects of PCP extract would involve in Ras signaling pathway. The animal experiments on chronic ALI model mice shown that the oral administration of PCP can alleviate ALI by attenuating hepatic oxidative stress, inflammation and down-regulating the target proteins in Ras/Raf/MEK/ERK pathway. Molecular docking analysis revealed the good binding ability between the three polyphenols (i.e. quercetin, qpigenin, thonningianin B) in PCP with the top contribution in network pharmacology, and these target proteins (Ras, Raf, MEK1/2, and ERK1/2). Conclusion Our results clarified that PCP ethanol extract could effectively alleviate ALI by down-regulating Ras/Raf/MEK/ERK signaling pathway promisingly. Quercetin, apigenin, and thonningianin B may be the active compounds of PCP, attributing to the intervention benefits of PCP against ALI.

Abstract:The aim of this work was designed to investigate the antioxidant, antiaging, and hepatoprotective effects of alkali-extractable polysaccharides (AlAPS) and their three purified fractions (AlAPS-1, AlAPS-2, and AlAPS-3) from Agaricus bisporus in D-galactose induced aging mice. For in vitro antioxidant analysis, both AlAPS and its fractions exhibited moderate reducing power, Fe2+-chelating activities, and potent scavenging activities on hydroxyl and 1,1-diphenyl-2-picrylhydrazyl (DPPH) radicals. The in vivo results demonstrated that the polysaccharides, especially AlAPS-2, showed potential antiaging and hepatoprotective effects by enhancing the antioxidant status, decreasing serum hepatic enzyme activities, and improving the lipid metabolism. This study suggested that the polysaccharides extracted and purified from A. bisporus could be exploited as a potent dietary supplement to attenuate aging and prevent age-related diseases.

Abstract:Background: Purple sweet potato Ipomoea batatas (L.) has long been used as a medicine and a food. It contains various bioactive substances such as polysaccharides, anthocyanins, and flavonoids. Purple sweet potato polysaccharides are known to have anti-oxidant, anti-tumor, and immunomodulatory functions. Nevertheless, studies on the structural characterization of purple sweet potato polysaccharides and their ability to prevent non-alcoholic fatty liver disease (NAFLD) have rarely been reported. Results: A novel polysaccharide (PSPP-A) was extracted and isolated from purple sweet potato, and its structural characteristics and preventive effects on NAFLD were investigated. The results indicated that PSPP-A was composed of l-rhamnose, d-arabinose, d-galactose, d-glucose, and d-glucuronic acid with molar ratios of 1.89:8.45:1.95:1.13:1. Its molecular weight was 2.63 × 103 kDa. Methylation and nuclear magnetic resonance (NMR) analysis indicated that the glycosidic linkages were →3)-α-L-Araf-(1→, α-L-Araf-(1→, →2,4)-α-L-Rhap-(1→, 4-O-Me-β-D-GlcAp-(1→, →4)-α-D-Glcp-(1→, →4)-β-D-Galp-(1→, and →6)-β-D-Galp-(1→. Scanning electron microscopy (SEM) indicated that the structure of PSPP-A was irregular. Subsequently, the protective effect of PSPP-A on NAFLD was investigated. The results indicated that bodyweight, liver index, and triglyceride (TG), total cholesterol (TC), aspartate transaminase (AST), and alanine transaminase (ALT) content were significantly reduced by intervention of purple sweet potato polysaccharide-A (PSPP-A) compared with the - high-fat diet group. Liver histopathological analysis indicated that PSPP-A attenuated irregular hepatocyte patterns and excessive lipid vacuoles. Conclusions: The novel polysaccharide, PSPP-A, mainly contains arabinose, which has certain preventive effects on NAFLD. This study provides a theoretical basis for further elucidating the hepatoprotective effect of purple sweet potatoes as a functional food. © 2022 Society of Chemical Industry.
Hepato-Protective Effect Of Ginsenosides From The Fruits Of Panax Ginseng Against Acetaminophen-Induced Liver Damage In Mice

Abstract:Background and Objective: Acetaminophen (APAP)-induced hepatotoxicity is a severe public health problem in western countries. Current treatment methods for poisoning are limited and novel therapeutic strategies are needed. The aim of the present study was to investigate the protective effect of ginsenosides from the fruits of Panax ginseng (GFG)against APAP-induced liver injury in mice and its potential molecular mechanisms of action. Materials and Methods: In this study, mice were orally administered with 150 or 300 mg kg(-1) of GFG for 7 consecutive days, followed by a single injection of APAP (250 mg kg(-1)). Severe liver injury was observed after 24 h APAP injection and the protective effect of GFG was assessed. Results: The results showed that pre-treatment with GFG reduced the levels of serum aspartate aminotransferase (AST) and alanine aminotransferase (ALT). Moreover,GFG showed anti-oxidant activities characterized by reducing hepatic MDA contents and increasing hepatic SOD and GSH levels, accompanied by inhibiting expression level of 4-HNE. Likewise, GFG decreased APAP-induced the expression of cytochrome P450 El (CYP2E1). Pre-treatment with GFG significantly inhibited pro-inflammatory factors tumor necrosis factor alpha (TNF-alpha), interleukin-1 beta(lL-1 beta), Bax, Bcl-2 and cyclooxygenase-2 (COX-2) levels expression of which contributed to ameliorating APAP caused hepatotoxicity. Furthermore, liver histopathological observation provided further evidence that GFG pretreatment significantly inhibited APAP-induced hepatocyte necrosis, inflammatory cell infiltration. Conclusion: The present study clearly showed that GFG exerted a protective effect against APAP-induced hepatotoxicity due to its anti-oxidant, anti-apoptotic and anti-inflammatory effects.
[A case of retroperitoneal mucinous cystadenocarcinoma].

Abstract:We reported is the histogenesis of a case of a primary retroperitoneal cystadenocarcinoma that was resected surgically in a 42-year-old woman who was admitted to hospital for SLE. On echographic examination, she was found to have a right abdominal tumor and underwent an operation for the complete removal of an abdominal tumor located in the right retroperitoneal cavity. A histological examination of the tumor revealed it to be compatible with a mucinous cystadenocarcinoma of the ovary. Only four cases of a primary retroperitoneal mucinous cystadenocarcinoma have been reported in the literature. This rare lesion was studied histopathologically and its histogenesis is discussed.
