Abstract:OBJECTIVE To quantify peripheral blood mononuclear cell JAK3 mRNA levels after allogeneic haematopoietic stem cell (allo-HSCT) transplantation, and to explore its relationship with graft vs host disease (GVHD). METHODS Serial monitoring of JAK3 mRNA levels by fluorescent quantitative reverse transcriptase polymerase chain reaction (FQ-RT-PCR) technique was performed on 26 cases (83 samples) after allo-HSCT. RESULTS The mean JAK3/ABL expression levels was 8.71 in 14 patients without GVHD; the JAK3/ABL expression level was 31.94 in one patient with acute GVHD (aGVHD); the mean JAK3/ABL expression levels was 19.23 in 11 patients with chronic GVHD (cGVHD), and the cGVHD was diagnosed at the mean time of six months (3 - 12 months). The JAK3/ABL expression levels did not change in 4 patients one month after allo-HSCT, however, the levels increased again when GVHD were diagnosed in these 4 patients, and the JAK3/ABL expression levels decreased again when GVHD was remitted. CONCLUSION The JAK3 expression level related to the remission and relapse in patients with GVHD.
Parental Investment Under Predation Threat in Incubating Common Eiders (Somateria mollissima): A Hormonal Perspective

Abstract:Predation risk affects the costs and benefits of prey life-history decisions. Predation threat is often higher during reproduction, especially in conspicuous colonial breeders. Therefore, predation risk may increase the survival cost of breeding, and reduce parental investment. The impact of predation risk on avian parental investment decisions may be hormonally mediated by prolactin and corticosterone, making them ideal tools for studying the trade-offs involved. Prolactin is thought to promote parental care and commitment in birds. Corticosterone is involved in allostasis and may either mediate reduced parental investment (corticosterone-fitness hypothesis), or promote parental investment through a reallocation of resources (corticosterone-adaptation hypothesis). Here, we used these hormonal proxies of incubation commitment to examine the impact of predation risk on reproduction in common eiders ( Somateria mollissima ) breeding in the Baltic Sea. This eider population is subject to high but spatially and temporally variable predation pressure on adults (mainly by the white-tailed eagle Haliaeetus albicilla and introduced mammalian predators) and nests (by the adult predators and exclusive egg predators such as hooded crows Corvus cornix ). We investigated baseline hormonal levels and hatching success as a function of individual quality attributes (breeding experience, female and duckling body condition), reproductive investment (clutch weight), and predation risk. We expected individuals nesting in riskier environments (i.e., on islands where predation on adults or nests is higher, or in less concealed nests) to reduce their parental investment in incubation, reflected in lower baseline prolactin levels and either higher (corticosterone-fitness hypothesis) or lower (corticosterone-adaptation hypothesis) baseline corticosterone levels. Contrary to our predictions, prolactin levels showed a positive correlation with nest predation risk. The unexpected positive relationship could result from the selective disappearance of low-quality females (presumably having low prolactin levels) from risky sites. Supporting this notion, female body condition and hatching success were positively correlated with predation risk on females, and baseline prolactin concentrations were positively correlated with duckling body condition, a proxy of maternal quality. In line with the corticosterone-adaptation hypothesis, baseline corticosterone levels increased with reproductive investment, and were negatively associated with nest predation risk. Hatching success was lower on islands where nest predation risk was higher, consistent with the idea of reduced reproductive investment under increased threat. Long-term individual-based studies are now needed to distinguish selection processes occurring at the population scale from individually plastic parental investment in relation to individual quality and variable predation risk.

Abstract:In birds, the link between parental care behaviour and prolactin release during incubation persists after hatching in altricial birds, but has never been precisely studied during the whole rearing period in precocial species, such as ducks. The present study aims to understand how changes in parental care after hatching are related to circulating prolactin levels in mallard hens rearing ducklings. Blood was sampled in hens over at least 13 post-hatching weeks and the behaviour of the hens and the ducklings was recorded daily until fledging. Contacts between hens and the ducklings, leadership of the ducklings and gathering of them steadily decreased over post-hatching time. Conversely, resting, preening and agonistic behaviour of hens towards ducklings increased. Plasma prolactin concentrations remained at high levels after hatching and then fell after week 6 when body mass and structural size of the young were close to those of the hen. Parental care behaviour declined linearly with brood age, showed a disruption of the hen-brood bond at week 6 post-hatching and was related to prolactin concentration according to a sigmoid function. Our results suggest that a definite threshold in circulating prolactin is necessary to promote and/or to maintain post-hatching parental care in ducks.

Abstract:Many studies have reported maternal transfer of organic contaminants in oviparous species, with inconsonant results. Egg-laying sequence and selected maternal tissues may impact on assessment of potential maternal transfer of contaminants. Here, this hypothesis was verified by exposing chickens (Gallus domesticus) to polychlorinated biphenyls (PCBs), polybrominated diphenyl ethers (PBDEs), and dechlorane plus (DPs). Concentrations in eggs laid during exposure exhibited periodic fluctuations (conforming to egg-laying cycles) and a decreasing trend during depuration. Fluctuation patterns of DPs and BDE209 differed from those of other compounds. The PBDE congener profiles in eggs were dominated by BDE209 during exposure and by BDE100 and 153 during depuration. The abundance of PCB congener (CB138) which is recalcitrant to metabolism increased with laying sequence. Maternal transfer potential was negatively correlated (P = 0.0014, R2 = 0.7874) to the log KOW of chemicals (log KOW >7) when the muscle, heart, lung, or stomach was used. No correlations were found when the liver, fat, kidneys, or intestine was used (log KOW >7), although DPs and BDE209 showed the highest maternal transfer potential. Different fluctuation patterns of DPs and BDE209 in eggs and increased abundance of BDE209 in eggs laid in the initial egg-laying period imply that the liver, fat, kidney, or intestinal tissues could be more appropriate in assessing maternal transfer of the target analytes.

Abstract:Due to the limited number of field studies investigating associations between environmentally relevant per- and polyfluoroalkyl substances (PFAS) mixtures and reproductive impairment, there is uncertainty as to whether birds are affected by PFAS pollution, whether species differ in sensitivity to PFAS, and whether the observed reproductive impairment is caused by PFAS or rather due to other potential confounding variables. Therefore, we investigated PFAS concentrations in eggs and blood plasma of great tit (Parus major) and blue tit (Cyanistes caeruleus) nestlings near a PFAS hotspot in Belgium, reproductive impairment, and associations between the accumulated levels and nestling body condition. In total, 29 eggs and 22 blood plasma samples of great tit clutches, and 10 egg and 10 blood plasma samples of blue tit clutches, were collected. Despite more types of PFAS being detected in eggs compared to plasma, only minor differences in profiles were observed between species. On the other hand, tissue-specific differences were more pronounced and likely reflect a combination of maternal transfer and dietary exposure post-hatching. Despite the high concentrations detected in both species, limited reproductive impairment was observed. Our results support previous findings that great tits and blue tits may not be very susceptible to PFAS pollution and provide evidence that other factors, including ecological stoichiometry, may be more important in explaining inter-species variation in PFAS accumulation and reproductive impairment.

Abstract:The peptide hormone prolactin plays an important role in the expression of parental care behaviours across bird and mammal taxa. While a great deal is known about how plasma prolactin concentrations vary across the reproductive cycle, the few studies that investigate how prolactin relates to individual-level variation in parental care have reported mixed results. We argue that, since parental care is also affected by social interactions and environmental constraints, prolactin may better reflect behaviours that are indirectly related to parenting than the absolute level of care that is eventually expressed. In this study, we tested for associations between plasma prolactin and the expression of both parental care and proximity to the partner in incubating black-headed gulls, Chroicocephalus ridibundus. Baseline prolactin levels increased with calendar date but were unrelated to incubation behaviours. However, parents who showed a weaker decrease in prolactin to an acute stressor spent more time in close proximity to their incubating partner while not on the nest themselves, suggesting that individual variation in stress-induced prolactin changes reflect differences in parents' tendency to be closely associated with their partner and the joint nesting attempt. Baseline and stress-induced levels of the stress hormone corticosterone were unrelated to both prolactin levels and parental behaviours, suggesting that this hormone is not a strong moderator of parental care in black-headed gulls. One potential explanation for the link between prolactin dynamics and partner proximity is that prolactin reflects parental motivation to provide parental care or retain contact with the breeding partner, but further work is needed to directly test this hypothesis.

Exploring solvent effects upon the Menshutkin reaction using a polarizable force field.

Abstract:The energetics of the Menshutkin reaction between triethylamine and ethyl iodide have been computed using B3LYP and MP2 with the LANL2DZ, LANL2DZd, SVP, MIDI!, 6-311G(d,p), and aug-cc-PVTZ basis sets. Small- and large-core energy-consistent relativistic pseudopotentials were employed. Solvent effect corrections were computed from QM/MM Monte Carlo simulations utilizing free-energy perturbation theory, PDDG/PM3, and both a nonpolarizable OPLS and polarizable OPLS-AAP force field. The B3LYP/MIDI! theory level provided the best DeltaG(++) values with a mean absolute error (MAE) of 4.9 kcal/mol from experiment in cyclohexane, CCl(4), THF, DMSO, acetonitrile, water, and methanol. However, the relative rates in cyclohexane, and to a certain extent CCl(4), were determined to be greatly underestimated when using the nonpolarizable OPLS force field. An overall reduction in the MAE to 3.1 kcal/mol using B3LYP/MIDI!/OPLS-AAP demonstrated the need for a fully polarizable force field when computing solvent effects for highly dipolar transition structures in low-dielectric media. The MAEs obtained with PDDG/PM3/OPLS and OPLS-AAP of 5.3 and 3.8 kcal/mol, respectively, provided comparable results to B3LYP at a fraction of the computational resources. The large rate accelerations observed in the reaction were correlated to an increased stabilization of the emerging charge separation at the transition state via favorable solute-solvent interactions.
Decreasing prolactin levels leads to a lower diving effort but does not affect breeding success in Adélie penguins

Abstract:Current research on seabirds suggests a key role of hormones in the trade-off between self-maintenance and parental investment through their influence on foraging decisions during the breeding period. Although prolactin is known to have major effects on parental care, its role in foraging behavior has rarely been investigated in seabirds to date. The aim of this study was to assess the influence of an experimental decrease in prolactin levels on foraging decisions and its consequences on breeding success in free-living seabirds. To achieve this, we implanted bromocriptine (an inhibitor of prolactin secretion) in male Adélie penguins (Pygoscelis adeliae), monitored their foraging behavior using time-depth recorders over several trips, and recorded their reproductive output. On average 8±0.5days after implantation, we showed that bromocriptine administration led to an efficient decrease in prolactin levels. However, no differences were seen in foraging trip durations between bromocriptine-implanted birds and controls. Moreover, the time spent diving and the number of dives performed per trip were similar in both groups. By contrast, all diving parameters (including diving efficiency) were negatively affected by the treatment during the first at-sea trip following the treatment. Finally, the treatment did not affect adult body condition or chick growth and survival. Our study highlights the short-term negative effect of low prolactin levels on diving effort, but indicates that a short-term and/or low-magnitude decrease in prolactin levels alone is not sufficient to modify consistently the body maintenance or the parental investment of Adélie penguins.

Abstract:A case of fibromuscular dysplasia (FMD) with involvement of the aorta is described in a 44-year-old woman. So far only 3 cases of aortal FMD have been mentioned, all in female patients. Our observation is characterized by a solitary stenosis of the lower abdominal aorta and it resembles histologically the "intimal fibroplasia" type of renal FMD. The review of the literature about FMD in general reveals that mostly the renal arteries are affected, followed by cervical artery FMD with only rare occurrence in other vessels. Though the etiology is basically unknown several factors have been made responsible, namely hereditary disposition, congenital rubella infection, female hormones, paucity of the vasa vasorum, and stretch-traction-stress. The disease has been classified into several histological subgroups and also according to the roentgenologic form which typically shows a "string of beads" appearance. There is a definitive female sex preponderance of 5:1, which is the same for all affected vessels; whereas in respect to age there is a marked distinction in so far that renal FMD patients on average are 11 years younger than patients with the cervicocephalic form of the disease.
Per- and polyfluoroalkyl substances (PFAS) in little penguins and associations with urbanisation and health parameters

Abstract:Per- and Polyfluoroalkyl substances (PFAS) are increasingly detected in wildlife and present concerning and unknown health risks. While there is a growing body of literature describing PFAS in seabird species, knowledge from temperate Southern Hemisphere regions is lacking. Little penguins (Eudyptula minor) can nest and forage within heavily urbanised coastal environments and hence may be at risk of exposure to pollutants. We analysed scat contaminated nesting soils (n = 50) from 17 colonies in lutruwita/Tasmania for 16 PFAS, plasma samples (n = 45) from nine colonies, and three eggs for 49 PFAS. We detected 14 PFAS across the sample types, with perfluorooctanesulfonic acid (PFOS) and perfluorohexanesulfonic acid (PFHxS) most commonly detected. Mean concentration of PFOS in plasma was 2.56 ± 4.3 ng/mL (<LOD-22.3), soil was 2.92 ± 5.39 μg/kg (<LOD-29.8) and eggs were 11.89 ± 11.30 μg/kg (0.27-22.84). Mean concentration of PFHxS in plasma was 0.9 ± 0.84 ng/mL (<LOD-5.6), soil was 11.4 ± 39.18 μg/kg (<LOD-213.3) and eggs were 0.27 ± 0.11 μg/kg (<LOD-0.38). Urbanisation of breeding sites was positively associated with PFOS and PFHxS concentrations in soils and blood. Phenology appeared to be an important predictor of PFHxS load, with moulting birds having lower levels compared to non-moulting birds. We investigated PFAS associations with health parameters and found that haematocrit and total plasma proteins in females were negatively associated with PFOS and positively associated in males. The number of erythrocytic nuclear abnormalities in males was positively associated with PFOS. Findings are the first to document the presence of PFAS in this resident seabird nesting and foraging close to anthropogenically modified habitats. We demonstrate that even in low concentrations, PFAS may have detectable relationships with penguin health. Ongoing research is needed to investigate foraging ecology and trophic bioaccumulation pathways, as well as understand sublethal consequences from exposure.
Exposure of an endangered seabird species to persistent organic pollutants: Assessing levels in blood and link with reproductive parameters

Abstract:Ocean contamination, particularly from persistent organic pollutants (POPs), remains a significant threat to marine predators that occupy high trophic positions. Long-lived procellariform seabirds are apex predators in marine ecosystems and tend to accumulate contaminants. Prolonged exposure to pollutants negatively affects their fitness including reproductive success. Low breeding success may represent a hurdle for the restoration of small and endangered seabird populations, including several highly threatened gadfly petrels. Here we investigated the annual variation (2019 and 2022) in organochlorine pesticide (OCP), polychlorinated biphenyl ether (PCB), polybrominated diphenyl ether (PBDE), and polycyclic aromatic hydrocarbon (PAH) exposure in the endangered Bermuda petrel ( Pterodroma cahow ), and the relationship between female contaminant burden and breeding parameters. We found that petrels were exposed to a wide range of pollutants (33 out of 55 showed measurable levels) with PCBs dominating the blood contaminant profiles in both years. Only 9 compounds were detected in >50 % of the birds. Specifically, among OCPs, p, p'- DDE and hexaclorobenzene were the most frequently detected while fluorene and acenaphthene were the most common PAH. The concentrations of ∑ 5 PCBs and ∑ 7 POPs were higher in older birds. Furthermore, females with greater contaminant burdens laid eggs with a lower probability of hatching. However, female investment in egg production (size and volume) was unrelated to their blood contaminant load. Overall, this study highlights the presence of a wide range of contaminants in the petrel's food web, and it sheds light on the potential impact of chronic exposure to sub-lethal levels of PCBs on the breeding success of seabirds. We claim that toxicological testing should be a practice integrated in the management of seabirds, particularly of endangered species to monitor how past and present anthropogenic activities impact their conservation status.

Influence of perfluoroalkyl acids and other parameters on circulating thyroid hormones and immune-related microRNA expression in free-ranging nestling peregrine falcons

Abstract:Exposure to certain perfluoroalkyl acids (PFAAs) can have considerable effects on the endocrine and immune systems, although such effects remain largely uncharacterized in wildlife. Using an apex avian predator, we investigated possible relationships of thyroid hormones (THs), specifically free (F) and total (T) thyroxine (FT4; TT4) and triiodothyronine (FT3; TT3), and the expression of an immune-related microRNA biomarker (i.e., miR-155), with the concentrations of 11 PFAAs in nestling peregrine falcons (Falco peregrinus). Nestling peregrines (n = 56; usually two chicks of each sex per nest) were blood sampled when 23 ± 4 days old in urban and rural regions of the Laurentian Great Lakes Basin (Ontario, Canada) in 2016 and 2018. The circulating concentrations of several PFAAs were significantly associated with THs and estimated thyroid gland activity (TT3:TT4; FT3:FT4), including PFHxS (FT3; FT3:FT4), PFDS (TT3; TT3:TT4), PFOA (TT4; FT3:FT4), PFTeDA (TT4; FT3:FT4), PFHxDA (TT4; TT3:TT4) and ΣPFCAs (TT4). Our novel evaluation of miR-155 in peregrine nestlings identified significantly negative relationships of plasma miR-155 counts with PFHxS and PFOA concentrations, indicating potential down-regulation of miR-155 expression and impaired immunity. Several PFAA homologues significantly predicted the variation in THs and miR-155 in conjunction with year (e.g., inter-annual differences in weather, ambient temperature, rainfall), region (urban/rural), nestling age, and/or diet (trophic position; δ15N), which suggests that multiple environmental and biological stressors, including PFAA exposure, influenced thyroid activity and immune function in these nestlings. Further research is warranted to identify the mechanisms and additional impacts of PFAA-related thyroid and immune disruption on the growth, development, and health risks in developing birds.
Impacts of a warming climate on concentrations of organochlorines in a fasting high arctic marine bird: Direct vs. indirect effects?

Abstract:The present study examined how climate changes may impact the concentrations of lipophilic organochlorines (OCs) in the blood of fasting High Arctic common eiders ( Somateria mollissima ) during incubation. Polychlorinated biphenyls (PCBs), 1-dichloro-2,2-bis ( p -chlorophenyl) ethylene ( p,p′ -DDE), hexachlorobenzene (HCB) and four chlordane compounds (oxychlordane, trans -chlordane and trans - and cis -nonachlor) were measured in females at chick hatching ( n = 223) over 11 years (2007–2017). Firstly, median HCB and p,p′ -DDE concentrations increased ~75 % over the study period, whereas median chlordane concentrations doubled (except for oxychlordane). PCB concentrations, in contrast, remained stable over the study period. Secondly, both body mass and clutch size were negatively associated with OC levels, suggesting that females with high lipid metabolism redistributed more OCs from adipose tissue, and that egg production is an important elimination route for OCs. Thirdly, the direct climate effects were assessed using the mean effective temperature (ET: air temperature and wind speed) during incubation, and we hypothesized that a low ET would increase redistribution of OCs. Contrary to expectation, the ET was positively correlated to most OCs, suggesting that a warmer climate may lead to higher OCs levels, and that the impact of ET may not be direct. Finally, potential indirect impacts were examined using the Arctic Oscillation (AO) in the three preceding winters (AO winter 1 – 3 ) as a proxy for potential long-range transport of OCs, and for local spring climate conditions. In addition, we used chlorophyll a (Chl a ) as a measure of spring primary production. There were negative associations between AO winter 1 and HCB, trans -chlordane and trans -nonachlor, whereas oxychlordane and cis -chlordane were negatively associated with Chl a. This suggests that potential indirect climate effects on eiders were manifested through the food chain and not through increased long-range transport, although these relationships were relatively weak.

Abstract:Assessing the physiological stress responses of wild animals opens a window for understanding how organisms cope with environmental challenges. Since stress response is associated with changes in body temperature, the use of body surface temperature through thermal imaging could help to measure acute and chronic stress responses non-invasively. We used thermal imaging, acute handling-stress protocol and an experimental manipulation of corticosterone (the main glucocorticoid hormone in birds) levels in breeding king penguins ( ), to assess: 1. the potential contribution of the Hypothalamo-Pituitary-Adrenal (HPA) axis in mediating chronic and acute stress-induced changes in adult surface temperature, 2. the influence of HPA axis manipulation on parental investment through thermal imaging of eggs and brooded chicks, and 3. the impact of parental treatment on offspring thermal’s response to acute handling. Maximum eye temperature ( ) increased and minimum beak temperature ( ) decreased in response to handling stress in adults, but neither basal nor stress-induced surface temperatures were significantly affected by corticosterone implant. While egg temperature was not significantly influenced by parental treatment, we found a surprising pattern for chicks: chicks brooded by the (non-implanted) partner of corticosterone-implanted individuals exhibited higher surface temperature (both and ) than those brooded by glucocorticoid-implanted or control parents. Chick’s response to handling in terms of surface temperature was characterized by a drop in both and independently of parental treatment. We conclude that the HPA axis seems unlikely to play a major role in determining chronic or acute changes in surface temperature in king penguins. Changes in surface temperature may primarily be mediated by the Sympathetic-Adrenal-Medullary (SAM) axis in response to stressful situations. Our experiment did not reveal a direct impact of parental HPA axis manipulation on parental investment (egg or chick temperature), but a potential influence on the partner’s brooding behaviour.

Abstract:Per- and polyfluoroalkyl substances (PFAS) are synthetic chemicals widely used in consumer products. PFAS can accumulate in animal tissues, resulting in biomagnification and adverse effects on wildlife, such as reproductive impairment. In bird species, PFAS are transferred from mothers to eggs along with essential nutrients and may affect embryo development. However, the extent of maternal PFAS transfer across different species and compounds remains poorly understood. Here, we conducted a systematic review and meta-analysis to quantify maternal PFAS transfer in wild birds and investigate potential sources of variation. We tested the moderating effects of compounds' physicochemical properties and biological traits of studied birds. The dataset included 505 measurements of PFAS concentration and 371 effect sizes derived from 13 studies on 16 bird species and 25 compounds. Overall, across all studies and species, we found a 41% higher concentration of PFAS in offspring than in mothers. Specifically, contaminants were concentrated in the yolk, longer and heavier compounds showed preferential transfer, larger clutch size was associated with decreased PFAS transfer and a higher transfer rate was shown in species with piscivorous and opportunistic/diverse diets. A validation assessment showed good robustness of the overall meta-analytic result. Given the crucial role of birds in maintaining ecological balance, this research article has relevant implications for modelling the impacts of PFAS on wildlife, ecosystems, and human health.

Abstract:Time trends and regional differences of polychlorinated dibenzo-p-dioxins and -furans (PCDD/Fs), polychlorinated biphenyls (PCBs), polychlorinated naphthalenes (PCNs), DDTs, polybrominated biphenyls (PBBs) and polybrominated diphenylethers (BDEs) were studied in unhatched osprey eggs collected by bird ringers in 1972-2017 from four areas in Finland. Two study areas were from Baltic Sea, Northern Quark and Finnish Archipelago Sea, while the two others were inland lake areas, eutrophicated Lake Vanajanselkä affected by industrial emissions, and Pristine SW Lake Area. The highest concentrations of most compound groups were in Lake Vanajanselkä consistent with high emissions, the predominance of bream as a prey, and higher concentrations in bream compared to other prey fish. Concentrations of all chlorinated compounds decreased significantly in all study areas. Average annual decreases were ∑PCDD/F 2.3-4.9%, ∑PCB 2.2-4.2%, ∑PCN 2.6-7.0% and ∑DDT 7.1-9.5%, primarily in line with decreased levels in prey fish. From 1972 PBBs and BDEs increased significantly until 1990s declining rapidly thereafter. PCDD/F congener profile was dominated by 2,3,4,7,8-PeCDF, except in Lake Vanajanselkä by 1,2,3,6,7,8-HxCDD. PCB congener profile was dominated by PCB 153 in all study areas, followed by PCB 180 and PCB 138. Among dioxin-like compounds PCBs contributed 82%, PCDDs 14% and PCDFs 4% to toxic equivalent quantity (∑TEQ). PCB 126 contributed most to ∑TEQ, followed by 1,2,3,7,8-PeCDD. BDE 47 being the dominant BDE congener, followed by BDE 100. ∑DDT concentrations were relatively similar across all study areas, with DDE contributing about 90%. Productivity of chicks per active nest was significantly decreased in Lake Vanajanselkä, and the likely explanation is embryotoxicity of dioxin-like compounds. It is plausible that dioxin-like compounds influenced embryonic survival among highly exposed ospreys prior to 2010, especially in Lake Vanajanselkä and Northern Quark. However, decreased survival due to DDE-induced eggshell thinning seems unlikely after 1985, and BDE levels were below those potentially causing adverse effects.

Abstract:Laying hens were exposed to feeds spiked with a series of perfluoroalkyl carboxylates (PFCAs) ranging from perfluorobutanoic acid (C4) to perfluorooctadecanoic acid (C18) to investigate their bioaccumulation, tissue distribution, and maternal transfer. We found that PFCAs with longer carbon chains (>8) were more efficiently absorbed in the gastrointestinal tract than those with shorter chains (≤8), and that the rate of depuration varied inversely with the carbon chain length in a U-shaped pattern. Moreover, bioaccumulation potential increased with increasing carbon-chain length, except for C4. Distinct affinities were observed for specific carbon-chain PFCAs across various tissues, evident from their differential accumulation during both uptake and depuration phases. Specifically, C9 showed a higher affinity for serum and liver, C12 was more prevalent in yolk, C14 was notably abundant in the brain, and C18 was predominant in other tissues. Furthermore, the egg-maternal ratio (EMR) increased with increasing carbon-chain length from C7 to C11 and reached a plateau phase for C12 to C18. Our study also confirmed the key role of phospholipids in the tissue distribution and maternal transfer of long-chain PFCAs. This study sheds light on the interaction between PFCAs and biological tissues and reveals the toxicokinetic factors that influence the bioaccumulation of PFCAs. Further research is needed to identify the specific proteins or components that mediate the tissue-specific affinity for different carbon-chain lengths of PFCAs.
