Abstract:A novel composite material with an ultra-high strength and a low elastic modulus, comprising a carbon fibre reinforced liquid crystalline polymer (LCP/CF) has been developed. An experimental osteotomy of the distal femur in nineteen rabbits weighing on average 4.8 (3.7-6.3) kg was fixed with an intramedullary LCP/CF rod of 3.2 mm in diameter and 50 mm in length. The initial mechanical properties of the rods had a mean shear strength of 180 MPa, a flexural strength of 450 MPa, and a flexural modulus of 40 GPa. The rods were implanted for periods of 3, 6, 12, 24, and 52 weeks. The healing of the osteotomy was analyzed by radiographic, histological, microradiographic, and oxytetracycline fluorescence studies. The strength retention of the LCP/CF rods was measured by mechanical testing. Solid union of the osteotomy was seen within 6 weeks. The implants retained their original strength properties up to 52 weeks. In three rabbits, a non-union with granulation tissue and carbon fibre particles was noticed at the osteotomy site, where disruption of the implant surface was seen at the level of the osteotomy. Further studies with a special interest in the fabrication process of the LCP/CF-implants are needed.

Abstract:Several felid species have been shown to be susceptible to infection with highly pathogenic avian influenza (HPAI) viruses of the H5N1 subtype. Infection of felids by H5N1 HPAI virus is often fatal, and cat-to-cat transmission has been documented. Domestic cats may then be involved in the transmission of infection to other animals but also to humans. A particular concern is the hypothetical role of the cat in the adaptation of the virus to mammalian species, thus increasing the pandemic risk. Therefore, the development of a HPAI vaccine for domestic cats should be considered a veterinary and also a public health priority. Here we show that vaccination of cats with a recombinant canarypox (ALVAC)(1)) virus, expressing the hemagglutinin (HA) of influenza virus A/chicken/Indonesia/03 (H5N1) confers protection against challenge infection with two antigenically distinct H5N1 virus isolates from humans. Despite low hemagglutination inhibiting (HI) antibody titers at the time of challenge, all vaccinated cats were protected against mortality and had reduced histopathological changes in the lungs. Importantly, viral shedding was reduced in vaccinated cats as compared to controls, suggesting that vaccination of cats could reduce the risk of viral transmission. In conclusion this study showed that the recombinant canarypox virus protected cats against homologous and heterologous H5N1 HPAI virus challenges.

Abstract:Feline panleukopenia virus (FPV), feline calicivirus (FCV), and feline herpesvirus type-1 (FHV-1) are major infectious pathogens in cats. We evaluated the immunogenicity of a new vaccine containing inactivated FPV, two FCVs, and FHV-1 in animals. An FPV, two FCVs, and an FHV-1 isolate were continuously passaged 70, 50, 80, and 100 times in CRFK cells. FP70, FC50, FC80, and FH100 were propagated and used as vaccine antigens. Two inactivated feline virus vaccines, Rehydragel-adjuvanted vaccine (FRAV) and Cabopol-adjuvanted vaccine (FCAV) were prepared and inoculated into mice and guinea pigs. Humoral immune responses were measured using hemagglutination inhibition (HI) for FPV and virus-neutralizing antibody (VNA) for two FCVs and FHV-1 tests. Serial passages in CRFK cells resulted in increase in titers of FPV and two FCVs but not FHV-1 The FCAV induced higher mean HI and VNA titers than the FRAV in guinea pigs; therefore, the FCAV was selected. Cats inoculated with FCAV developed a mean HI titer of 259.9 against FPV, and VNA titers of 64, 256, and 3.2 against FCV17D03, FCV17D283, and FHV191071, respectively. Therefore, cats inoculated with the FCAV showed a considerable immune response after receiving a booster vaccination.

Abstract:Viral infections in cats can cause serious diseases and even death. Vaccines protect animals from diseases by inducing antibody production and cellular immune response. Primary and booster vaccinations are performed in accordance with the recommendations for the use of vaccines prescribed by the manufactures depending on the minimum duration of immunity. In case of feline panleukopenia, antibody titers correlate with the protection against infection, as for feline calicivirus infection and feline rhinotracheitis, there is no such correlation or it is less clear. Vaccination of cats against these diseases has been performed in the Russian Federation for many years, nevertheless, the feline panleukopenia virus (FPV), feline calicivirus (FCV) and feline herpesvirus (FHV) are still the main common cause of morbidity and mortality among cats. Virus-carrying cats play an important role in the transmission of respiratory viruses such as FHV and FCV in the feline population, and the long-term persistence of FPV in the body, stability in the environmental conditions and resistance to disinfecting agents can be a potential cause of the infection in susceptible kittens. Due to variety of antigenically different FCV strains, the use of the vaccines containing two or more viral strains may induce a broader heterologous protection. The purpose of this work was to evaluate the effectiveness of the vaccine against feline panleukopenia, feline calicivirus infection and feline viral rhinotracheitis developed at the Federal Center for Animal Health (Vladimir) subordinate to the Rosselkhoznadzor, containing 2 heterologous FCV strains (Pers strain genotype I and Fauna strain genotype II), FPV Sheba strain and FHV Lavr strain. The product was developed and tested for its quality in accordance with the requirements of the Russian Federation law. Clinical studies were conducted using 8–12 week-old kittens from different litters born from seronegative, non-vaccinated cats and kept in the household, in a veterinary hospital and animal shelters. The product has successfully passed comprehensive quality control and is registered in the territory of the Russian Federation.
Vaccination with an inactivated canine influenza H3N2 virus vaccine is safe and elicits an immune response in cats

Abstract:Objectives: The aim of this study was to evaluate safety and seroconversion when an inactivated H3N2 canine influenza virus (CIV) vaccine was administered to cats. Methods: Twenty 7-8-week-old seronegative cats were randomly assigned to two groups of 10 animals each. Cats in treatment group T01 were subcutaneously administered two doses of an adjuvanted placebo 3 weeks apart to serve as non-immunized controls. Cats in treatment group T02 were subcutaneously administered with two doses of H3N2 CIV vaccine at 3 weeks apart. All animals were actively monitored for 5 days after each injection for local and systemic reactions. Tympanic temperatures were recorded the day before and 5 days after each vaccination. Blood samples for serology were collected prior to each vaccination (days -1 and 20), and 7 and 14 days post-second vaccination. Results: Minor vocalization was observed in both control and vaccinated animals after the first and second dose administration. The only injection site reaction observed was mild swelling in one control cat, which resolved within 24 h. Transient fevers (39.5-39.7°C) that resolved within 24 h post-injection were observed in both treatment groups (T01 = 3/10 and T02 = 5/10). All vaccinated, but no control, animals successfully seroconverted within 14 days of second vaccination, with H3N2 CIV-specific hemagglutination inhibition (HAI) titers ranging from 32 to 128. Conclusions and relevance: Cats vaccinated subcutaneously with an inactivated H3N2 CIV vaccine had similar rates of adverse events post-vaccination as the control group. Increased HAI titers provided evidence of post-vaccination seroconversion with the H3N2 CIV-vaccinated group.

Abstract:Today more and more children are surviving childhood diseases that had been fatal in a previous generation. Medical advances and technology allow most children with cancer to enjoy long-term survival and cure. Research studies now show that there are late effects of treatment that affect growth, development, and cognitive functioning. Parents and teachers of long-term survivors of childhood cancer share concerns about deficits in cognitive functioning as it affects learning in school. Childhood cancer and the literature on cognitive late effects of treatment are discussed. Recommendations are made for early psychological evaluation and appropriate educational intervention.
Attempted immunisation of cats against feline infectious peritonitis using canine coronavirus

Abstract:Specific pathogen free kittens were vaccinated with an unattenuated field isolate of canine coronavirus (CCV) either by aerosol or subcutaneously, and received boosting vaccinations four weeks later. Aerosolisation elicited a homologous virus-neutralising (VN) antibody response that increased steadily over a four-week period and levelled off one to two weeks after revaccination. The initial aerosolised dose produced an asymptomatic infection with excretion of CCV from the oropharynx up to eight days after vaccination; virus shedding was not detected, however, after the second inoculation. Cats vaccinated subcutaneously developed low VN antibody titres after the first CCV dose and experienced a strong anamnestic response after the second dose. Neutralising antibody titres then levelled off one to two weeks after revaccination at mean values somewhat lower than in cats vaccinated by aerosol. CCV was not isolated from the oropharynx after either subcutaneous dose. Four weeks after CCV boosting inoculations, vaccinated cats and sham-vaccinated control cats were divided into three subgroups and challenged by aerosol with the virulent UCD1 strain of feline infectious peritonitis virus (FIPV UCD1) at three different dosage levels. Five of six cats (including sham-vaccinated controls) given the lowest challenge dose showed no signs of disease, while all other cats developed lesions typical of feline infectious peritonitis (FIP). The five surviving cats developed FIP after subsequent challenge with a fivefold higher dose of FIPV. Thus heterotypic vaccination of cats with CCV did not provide effective protection against FIPV challenge.
Anti-rabies humoral immune response in cats after concurrent vs separate vaccination against rabies and feline leukaemia virus using canarypox-vectored vaccines

Abstract:Objectives: Some expert groups recommend that cats should be vaccinated with non-adjuvanted feline leukaemia virus (FeLV) and rabies vector vaccines, which, in the European Union, are currently not licensed for concurrent use and have to be administered at least 14 days apart (different from the USA) and thus at separate visits, which is associated with more stress for cats and owners. The aim of this study was to assess the anti-rabies antibody response in cats after vaccination against rabies and FeLV at concurrent vs separate (4 weeks apart) visits using two canarypox-vectored vaccines (Purevax Rabies and Purevax FeLV; Boehringer Ingelheim) and to evaluate the occurrence of vaccine-associated adverse events (VAAEs). Methods: Healthy FeLV antigen-negative client-owned kittens (n = 106) were prospectively included in this randomised study. All kittens received primary vaccinations against rabies (week 0) and FeLV (weeks 4 and 8). After 1 year, the study group (n = 52) received booster vaccinations against rabies and FeLV concurrently at the same visit (weeks 50-52). The control group (n = 54) received booster vaccinations against rabies (weeks 50-52) and FeLV (weeks 54-56) separately. Anti-rabies virus antibodies (anti-RAV Ab) were determined by fluorescent antibody virus neutralisation assay at weeks 4, 50-52 and 54-56, and compared between both groups using a Mann-Whitney U-test. Results: Four weeks after the first rabies vaccination, 87/106 (82.1%) kittens had a titre ⩾0.5 IU/ml and 19/106 (17.9%) had a titre <0.5 IU/ml. Four weeks after the 1-year rabies booster, all cats had adequate anti-RAV Ab according to the World Organisation for Animal Health (⩾0.5 IU/ml), and the titres of the study group (median = 14.30 IU/ml) and the control group (median = 21.39 IU/ml) did not differ significantly (P = 0.141). VAAEs were observed in 7/106 (6.6%) cats. Conclusions and relevance: Concurrent administration of Purevax FeLV and Purevax Rabies vector vaccines at the 1-year booster does not interfere with the development of anti-RAV Ab or cause more adverse effects and thus represents a better option than separate vaccination visits for cats and owners.
Molecular Detection of Parvovirus in Captive Siberian Tigers and Lions in Northeastern China from 2019 to 2021

Abstract:The fact that wild felines are carriers of pernicious infectious viruses should be a major concern due to the potential cross-species transmission between the felines and human or domestic animals. However, studies on the virus in the captive wild felines, especially in tigers, are thin on the ground. In this study, we screened four infectious viruses, namely, feline parvovirus (FPV), feline coronavirus (FCoV), canine distemper virus (CDV), and influenza A virus (IAV), in the blood samples of 285 captive Siberian tigers (Panthera tigris altaica) and in the spleen samples of two deceased lions (Panthera leo), which were collected from 2019 to 2021 in three Siberian Tiger Parks from the northeast of China. Nucleic acids isolated from the blood samples collected from tigers and the spleen samples collected from two deceased lions were positive for FPV by PCR, and the positive rate was 4.6% (13/285) in tigers. Furthermore, the VP2 gene of FPV was amplified by nested PCR, and the sequences of the VP2 gene from these six FPV positive strains shared 98.3–99.9% homology with the reference. The key amino acid sites of VP2 protein were consistent with that of FPV reference strains. Phylogenetic analysis based on the VP2 gene showed that in this study, FPV-positive strains were grouped within the FPV clade and closely related to the Asian strains clade. The results of this study showed that FPV circulated in the captive Siberian tigers and lions in northeastern China and provided valuable information for the study of FPV epidemiology in wild felines. Therefore, we suggest that regular antibody monitoring and booster immunization for tigers should be performed.
Outbreak and genotyping of canine distemper virus in captive Siberian tigers and red pandas

Abstract:In this study, four canine distemper virus (CDV) strains were isolated from captive Siberian tigers (Panthera tigris altaica) and red pandas (Ailurus fulgens) during two separate CDV outbreaks in a zoo in Guangdong province, China. Sequence alignment and phylogenetic analyses based on the full-length hemagglutinin (H) and fusion (F) genes showed that they were closely identical to genotype Asia-1. Prior to confirmation of CDV in Siberian tigers, to control spread of the disease, a live attenuated combination CDV vaccine was used among almost all carnivore animals except for red pandas in which another recombinant combination CDV vaccine was used. However, about two months later, CDV re-emerged and caused the death among red pandas. Based on the vaccination records, the live combination vaccine could be considered an ideal weapon against CDV in zoo carnivore animals. Although the recombinant combination CDV vaccine was safe for red pandas, its protection effectiveness remains to be further investigated. Moreover, according to the outbreak interval time and sequence characterization, we suspected that stray cats circulating in the zoo were the intermediate host, which contributed to CDV spread from stray dogs to zoo animals. This study revealed the importance of vaccination and biosecurity for zoo animals.

Abstract:SARS-CoV-2 has exhibited varying pathogenesis in a variety of Mammalia family's including Canidae, Mustelidae, Hominidae, Cervidae, Hyaenidae, and Felidae. Novel SARS-CoV-2 variants characterized by spike protein mutations have recently resulted in clinical and epidemiological concerns, as they potentially have increased infectious rates, increased transmission, or reduced neutralization by antibodies produced via vaccination. Many variants have been identified at this time, but the variant of continuing concern has been the Delta variant (B.1.617.2), due to its increased transmissibility and infectious rate. Felines vaccinated using an experimental SARS-CoV-2 spike protein-based veterinary vaccine mounted a robust immune response to the SARS-CoV-2 spike protein. Using a reporter virus particle system and feline serum, we have verified that vaccinated felines produce antibodies that neutralize the SARS-CoV-2 Wuhan strain and variant B.1.617.2 at comparable levels.

Abstract:A trivalent (feline panleucopenia, feline herpesvirus, feline calicivirus), modified live, commercially available cat vaccine was used at either 6, 9 and 12 weeks of age (early schedule) or 9 and 12 weeks of age (conventional schedule), and the serological response to vaccination was assessed. The level of maternally derived antibody present at 6 weeks of age was also established. The use of early vaccination at 6 weeks of age induced an antibody response to each virus by 9 weeks of age in a significant proportion of kittens compared with unvaccinated littermates. There was no difference between the conventionally and early-vaccinated groups in terms of antibody response to any antigen by 12 and 15 weeks of age.

Abstract:Cytauxzoonosis is caused by Cytauxzoon felis (C. felis), a tick-borne parasite that causes severe disease in domestic cats in the United States. Currently, there is no vaccine to prevent this fatal disease, as traditional vaccine development strategies have been limited by the inability to culture this parasite in vitro. Here, we used a replication-defective human adenoviral vector (AdHu5) to deliver C. felis-specific immunogenic antigens and induce a cell-mediated and humoral immune response in cats. Cats (n = 6 per group) received either the vaccine or placebo in two doses, 4 weeks apart, followed by experimental challenge with C. felis at 5 weeks post-second dose. While the vaccine induced significant cell-mediated and humoral immune responses in immunized cats, it did not ultimately prevent infection with C. felis. However, immunization significantly delayed the onset of clinical signs and reduced febrility during C. felis infection. This AdHu5 vaccine platform shows promising results as a vaccination strategy against cytauxzoonosis.
Onset of Protection of FeLV Vaccine Containing Purified P45 FeLV-Envelope Antigen after One Single Injection of Primary Vaccination in Cats

Abstract:Feline Leukaemia Virus (FeLV) is a viral pathogen causing fatal disease in cat populations worldwide, mainly in young kittens with outdoor access. This study aimed to assess the protection against feline leukaemia virus (FeLV), provided by a FeLV vaccine containing purified p45 FeLV-envelope antigen (Leucogen®) after one single injection of primary vaccination in kittens. Twenty-five 9-week-old cats, negative for FeLV antibodies and antigens the day of vaccination, were randomized into two groups. One group of 10 kittens was not vaccinated and kept as control, and one group of 15 animals received one dose of Leucogen® vaccine. The serological response was measured 14 and 21 days post vaccination. Three weeks after the vaccination, all kittens were inoculated by oronasal route with a virulent FeLV-A strain. The development of persistent viraemia was monitored for 15 weeks following inoculation. The animals were clinically monitored closely during the experimental phase. All cats remained in good health, presented physiological growth along the study and did not show any unexpected reaction after vaccination. In the control group, 90% of cats (9/10 cats) developed a persistent infection, confirming that this experimental infection model was valid, since the minimum of 80% required by the European monograph was achieved. On the other side, 73% of the vaccinated cats (11/15 cats) did not develop a persistent infection following the challenge strain inoculation. In the FeLV vaccinated group, 73% of cats were protected from the first vaccine injection. The first injection of primary vaccination with Leucogen® vaccine provided complete protection against FeLV persistent viraemia in 73% of the kittens. The second injection of the primary vaccination is necessary to ensure a complete protection of the whole population and a long lasting immunity. The early protection against FeLV infection conferred after one single injection of primary vaccination with FeLV vaccine containing purified p45 FeLV-envelope antigen could be a contributing factor in the reduction of prevalence of FeLV infection.

Abstract:Feline calicivirus (FCV) is a worldwide pathogen that affects the general health status of cats. The aim of this study was to assess the efficacy of the FCV component of two multivalent commercial vaccines after one injection. The cats assigned to the vaccinated groups received one subcutaneous injection of either the Leucofeligen™ FeLV/RCP (Virbac, France) vaccine or the Purevax™ RCPFeLV (Boehringer Ingelheim, Germany) vaccine. In the control group, the cats did not receive any vaccine. Three weeks after the vaccine injection, all the cats were inoculated with a heterologous calicivirus strain, isolated in the field. The clinical signs of calicivirosis and the viral shedding were assessed. The results demonstrated that Leucofeligen™ FeLV/RCP could protect kittens against FCV infection by reducing clinical signs and viral excretion from 3 weeks after the first injection of primary vaccination, whereas Purevax™ RCPFeLV couldn't offer the same level of protection in similar experimental conditions.

Abstract:Protective antibody titers against core vaccines have not been standardized for cheetahs (Acinonyx jubatus) under human care. Vaccine-induced disease has been suspected after administration of modified live virus vaccine (MLVV), but it has not been confirmed as the causative agent. MLVV and killed virus vaccines (KVV) elicit humoral response in cheetahs; however, the use of both vaccines for initial immunization in cheetah cubs <6 months old within the same population has not been reported. The current case series describes viral disease presentation in two cheetah litters after using both vaccines and presents results for serum neutralization titers against feline calicivirus (FCV) and feline herpesvirus-1 (FHV-1) and hemagglutination inhibition titers against feline panleukopenia virus (FPV). For Litter 1, MLVV was administered at 6 and 9 wk old. On week 11, one male developed ocular, oral, and dermal lesions. Viral isolation recovered FCV. Because of suspected vaccine-induced FCV, KVV was administered on weeks 13 and 16. Litter 2 was vaccinated with KVV via the same vaccination schedule. Fifty-three days after the last booster, two cubs presented with ocular, respiratory, and oral clinical signs; both were PCR positive for FHV-1. Serology reported a better anamnestic response and protective titers against FCV and FPV with the protocol used with Litter 1. In Litter 2, FCV and FHV-1 titer measurement failed in three of four cubs, limiting comparison of titers between litters. In spite of limited measurements, absence of a statistical evaluation, and presence of infection, serology showed a better humoral response when MLVV was used.

Prevalence of Four Viruses in Captive Siberian Tigers from Northeastern China.

Abstract:Our study investigated the prevalence of feline herpesvirus-1 (FHV-1), feline calicivirus (FCV), feline immunodeficiency virus (FIV), and feline leukemia virus (FeLV) in captive Siberian tigers in Northeastern China. A total of 324 blood samples and 33 nasopharyngeal swab samples of Siberian tigers collected from 2019 to 2021 in three cities were investigated by nested PCR. The results showed that 28.1% (91/324) tigers were infected with at least one virus; the positive rates of FHV-1, FCV, and FIV were 17.3%, 13.6%, and 0.9%, respectively; and the coinfection prevalence was 13.2%. No FeLV-positive sample was detected. And we found that the blood is the best for FCV, FIV, and FeLV detection, but nasopharyngeal swabs for FHV-1. By comparing the gD genes, TK gene, and gI gene of FHV-1, the homology of the three FHV-1 positive strains in this study was found to be 91.5%-99.9% shared with tigers and domestic cats. Based on a comparison of the nucleic acid sequences of 13 FCV strains, we found that the homology of strain HB-1926 with the other strains in this study was only about 77.7%, but shared 99.3% and 98.6% homology with Urnaba strain in American cat and TG1 strain in Chinese tiger, respectively. However, the other 12 FCV strains shared 87.1%-87.5% homology compared with the Chinese domestic cats. Phylogenetic tree analysis showed that the HB-1926 strain was not in the same clade as other strains. The fragments gag-p26, pol-RT, and pol-RNAse of Siberian tiger FIV shared more than 99% homology than domestic cats FIV subtype A. This study demonstrated that captive Siberian tigers in Northeastern China were exposed to FHV-1, FCV, and FIV, and it is necessary to develop more effective vaccines and improve daily management measures.

Abstract:Scott and colleagues are to be commended on a very readable review of the pitfalls in diagnostic test research. Their catalogue of possible biases, however, misses the problem of post-hoc determination of cut points, a very common bias that has only recently been described. Scott mentions that the requirement to select the optimal cut point or test threshold, however, does not comment on the problem of cut points being set before or after examination of the data. If the cut point is selected to optimize test performance in that particular set of research patients, it will probably benefit from random error, which will not necessarily be present in a subsequent set of patients. This bias was estimated in a simulation study to be at least five percentage points of sensitivity or specificity in a quarter of all datasets with less than 50 cases of disease. Although a good scientific method stipulates stating the analysis before seeing the data, the standard method of choosing the best cut point from the receiver operating curve goes against this edict. In a review of B type natriuretic peptide (BNP) studies 47 papers presented 54 analyses ofwhich 7used prespecified cut points and 47 used data derived post-hoc cut points. Meta regression failed to show an increase in test performance for the post-hoc studies, but there was sufficient diversity of results to swamp this effect.
Serologic Survey of Selected Pathogens in Free-Ranging Bengal Tigers (Panthera tigris tigris) in Nepal

Abstract:Serum samples of 11 Bengal tigers (Panthera tigris tigris) from Chitwan National Park in Nepal, collected between 2011-17, were evaluated for the presence of antibodies to eight diseases commonly investigated in large felids. This initial serologic survey was done to establish baseline information to understand the exposure of Nepal's free-ranging tiger population to these diseases. Tiger serum samples collected opportunistically during encounters such as translocation, human conflict, and injury were placed in cold storage for later use. Frozen serum samples were assessed for feline coronavirus (FCoV), feline immunodeficiency virus, feline leukemia virus, feline herpesvirus (FHV), canine distemper virus, canine parvovirus-2 (CPV-2), leptospirosis (LEP; seven serovars), and toxoplasmosis (TOX). Six tigers were found to be positive for LEP, eight for CPV-2, five for FHV, one for FCoV, and 10 for TOX. Tigers, like other wild felids, have been exposed to these common pathogens, but further research is needed to determine the significance of these pathogens to the Nepali population.
