Abstract:Volatile profiles of unifloral honeys and meads prepared in different ways (boiled-saturated, not boiled-unsaturated) were investigated by headspace solid-phase micro extraction (HS-SPME) and dehydration homogeneous liquid-liquid extraction (DHLLE) followed by GC-FID/MS analyses. The obtained data were analyzed by principal component analysis (PCA) to evaluate the differences between the investigated products. The volatile profiles of honey as well as the boiled and the not boiled meads prepared from it showed significant discrepancies. The meads contained more aliphatic acids and esters but fewer monoterpenes and aliphatic hydrocarbons than the honey. Significant/substantial differences were found between the boiled (more aliphatic alcohols and acids) and the not boiled meads (more aliphatic hydrocarbons and esters). Some compounds related to yeast metabolism, such as tryptophol, may be considered markers of honey fermentation. This research allowed us to identify chemical markers of botanical origin, retained and detectable in the meads: 4-isopropenylcyclohexa-1,3-diene-1-carboxylic acid and 4-(1-hydroxy-2-propanyl)cyclohexa-1,3-diene-1-carboxylic acid for linden; valeric acid, γ-valerolactone, p-hydroxybenzoic acid for buckwheat; 4-hydroxybenzeneacetic acid, homovanillic acid and trans-coniferyl alcohol for honeydew; and methyl syringate for canola.

Abstract:The botanical origin of honey determines its composition and hence properties and product quality. As a highly valued food product worldwide, assurance of the authenticity of honey is required to prevent potential fraud. In this work, the characterisation of Spanish honeys from 11 different botanical origins was carried out by headspace gas chromatography coupled with mass spectrometry (HS-GC-MS). A total of 27 volatile compounds were monitored, including aldehydes, alcohols, ketones, carboxylic acids, esters and monoterpenes. Samples were grouped into five categories of botanical origins: rosemary, orange blossom, albaida, thousand flower and "others" (the remaining origins studied, due to the limitation of samples available). Method validation was performed based on linearity and limits of detection and quantification, allowing the quantification of 21 compounds in the different honeys studied. Furthermore, an orthogonal partial least squares-discriminant analysis (OPLS-DA) chemometric model allowed the classification of honey into the five established categories, achieving a 100% and 91.67% classification and validation success rate, respectively. The application of the proposed methodology was tested by analysing 16 honey samples of unknown floral origin, classifying 4 as orange blossom, 4 as thousand flower and 8 as belonging to other botanical origins.

Abstract:Honey is a very appreciated product for its nutritional characteristics and its benefits for human health, comprising antioxidant, anti-inflammatory, antifungal, and antibacterial activities. These attributes depend on the specific composition of each honey variety, with the botanical origin as one of the distinctive features. Indeed, honeydew and blossom honeys show different physicochemical properties, being the antioxidant capacity, mainly relying on the phenolic compound content, one of the most important. In this work, Folin–Ciocalteu (FC) index, total flavonoid content (TFC), and the antioxidant capacity based on the ferric reducing antioxidant power (FRAP) assay were determined for a total of 73 honeys (50 blossom honeys and 23 honeydew honeys). Mean content of oxidizable species (FC index) ranges from 0.17 to 0.7 mg eq. gallic acid g−1, with honeydew honeys being the ones with higher values. Regarding TFC, mean values above 1.5 mg eq. quercetin g−1 (method applied in the absence of NaNO2) were obtained for honeydew honeys and heather honey. Lower and not discriminatory values (below 0.3 mg eq. epicatechin g−1) were obtained in the presence of NaNO2. The maximum antioxidant capacity was observed for thyme honeys (2.2 mg eq. Trolox g−1) followed by honeydew and heather honeys. Individually, only the FC index was able to discriminate between honeydew and blossom honeys, while the other spectroscopic indexes tested allowed the differentiation of some honey types according to the botanical origin. Thus, a holistic treatment of the results was performed using partial least square discriminant analysis (PLS-DA) for classification purposes using FC, TFC, and FRAP results as data. Honeydew and blossom honey were satisfactorily discriminated (error 5%). In addition, blossom honeys can be perfectly classified according to their botanical origin based on two-class PLS-DA classification models.

Abstract:Free amino acids of 58 unifloral honeys of 17 different floras (cedar, eucalyptus, vitex, carob, clover, honeydew, sunflower, citrus, heather, thyme, flower, chestnut, sideritis, acacia, lavender, cotton, and mad honeys) from various regions across Turkey were analyzed using ultra performance liquid chromatography with electrospray ionization tandem mass spectrometry (UPLC-ESI-MS/MS). A method for the determination of free amino acids in honeys was developed and validated. The method comprises sample preparation involving ultrasonic extraction and separation, identification, confirmation, and quantitation. The obtained data have been inquired by principal components analysis (PCA), allowing differentiation of studied honeys by their floral origins. The main amino acids detected in Turkish honeys were phenylalanine (4024.53 mg/kg), proline (1138.18 mg/kg), tyrosine (693.42 mg/kg), and isoleucine (749.81 mg/kg). Thus, the honeys consist of rich free amino acids, and can be accordingly consumed as not only traditional sweeteners, but also supplementary materials for food products especially for foods for children. This method may be used for the determination of individual free amino acids in honeys.

Abstract:Chestnut, heather, honeydew, multifloral, orange blossom and thyme honey samples obtained either from beekeepers or supermarkets from Portugal, Greece, Oceania and Asia, were characterised by means of poly-parametric analysis: palynological, °Brix, moisture, pH, electrical conductivity, free acidity, total dissolved solids, salinity, vitamin C, specific weight, CIELAB colour parameters, Pfund and colour intensity determinations, along with bio-activity and volatile compounds analyses were carried out. Results showed that the botanical and geographical origin of honey samples had a similar impact on the determined parameters, using dimension reduction and classification techniques. Among them, moisture, °Brix, electrical conductivity, salinity, pH, colour parameters <i>L*</i> and <i>b*</i>, colour intensity, 1-hydroxy-2-propanone, benzaldehyde, 2-ethyl-1-hexanol, HMF and benzeneacetaldehyde were the potential markers related to the geographical and botanical origin of six honey varieties from different countries. Finally, the identification of methylglyoxal in Portuguese honeydew honey creates the basis for new research on the isolation and identification of this compound in European honey.

Abstract:This study aims to predict several quality traits in commercial honey samples simultaneously and to reveal possible honey adulteration using a field-deployable portable infrared spectrometer without any sample preparation. A total of one hundred and forty-seven commercial honey samples were purchased from local and online markets in Turkey and the United States of America (USA), and their soluble solids (°Brix), pH, free acidity, moisture, water activity (aw), glucose, fructose, sucrose, and hydroxymethyl furfural (HMF) contents were determined using reference methods. The HMF (n = 11 samples) and sucrose (n = 21) concentrations were higher than the regulatory limits in some tested samples. The exceeding HMF content may imply temperature abuse during storage and prolonged storing. On the other hand, high sucrose content may indicate possible adulteration with commercial sweeteners. Therefore, soft independent modeling of class analogies (SIMCA) analysis was conducted to reveal this potential sweetener adulteration in the samples, and the SIMCA model was able to identify all the flagged samples. The suggested FT-IR technique may be helpful in regulatory bodies in determining honey authenticity issues as well as assessing the quality characteristics of honey samples in a shorter period and at a lower cost.

Abstract:A delayed traumatic intracerebral hematoma was seen in a patient after a shotgun wound to the brain. The cause of the hemorrhage seemed to be a traumatic aneurysm. Both complications are reviewed, and it is suggested that clinical and intracranial pressure monitoring should lead to early diagnosis and may minimize the effects of this problem and that angiography may be appropriate early in the course of patients who have had fragments pass near intracranial vessels.
Bile lipid secretion in obese and non-obese individuals with and without gallstones.

Abstract:Biliary lipid secretion rates were measured in non-obese and obese individuals with and without cholesterol gallstones, using a steady-state, amino acid duodenal perfusion method. In addition, biliary lipid secretion rates were measured in five obese gallstone patients receiving high-dose chenodeoxycholic acid therapy (16-22 mg day-1 kg-1). Bile acid secretion rates in the non-obese patients with cholesterol gallstones (563 +/- SEM 70 mumol/h, n = 6) were significantly lower than in the non-obese controls (1078 +/- 210 mumol/h, n = 10, P less than 0.05), whereas cholesterol secretion rates were similar in the non-obese individuals with and without gallstones (51 +/- 7 and 42 +/- 4 mumol/h respectively). In the obese, both with and without gallstones, the major abnormality was hypersecretion of cholesterol (107 +/- 7 mumol/h, n = 7, and 81 +/- 15 mumol/h, n = 7, respectively). Both these values were significantly greater than those in the non-obese controls (P less than 0.01-0.02). Biliary cholesterol secretion rates correlated significantly with bile acid secretion rates but, for every mole of bile acid secreted, the obese secreted more cholesterol than the non-obese. Chenodeoxycholic acid treatment lowered biliary cholesterol saturation in obese gallstone patients by reducing biliary cholesterol secretion. These results suggest that there are two major types of defect in biliary lipid secretion in gallstone patients: reduced biliary bile acid secretion in non-obese gallstone patients and excessive biliary cholesterol secretion in the obese.

Abstract:Pulmonary renal syndrome (PRS) is characterized by both diffuse alveolar haemorrhage and glomerulonephritis as pathological features. Several immunologic and non-immunologic mechanisms including anti-neutrophil cytoplasmic antibody (ANCA)-positive vasculitis, anti-glomerular basement membrane disease, and systemic lupus erythematosus are commonly involved in the pathogenesis of the syndrome. We herein present a 60-year-old woman, non-smoker, who presented with fever, polyarthralgia, and the spreading of generalized purpuric rashes. The patient displayed rapid deterioration over the following two weeks marked by progressive declining renal function followed by haemoptysis. The patient was subsequently diagnosed with PRS, which was confirmed by the radiological evidence of alveolar haemorrhage and the histopathological evidence of pauci-immune glomerulonephritis. All immune markers including ANCA were negative. The patient was successfully treated with hemodialysis and immunosuppressive therapy. ANCA-negative vasculitis is a rare entity and even more rare as an etiology of PRS. An early diagnosis of this disease and its timely intervention is crucial.
Botanical traceability of unifloral honeys by chemometrics based on head-space gas chromatography

Abstract:The botanical origin of honey is subjected to severe controls by Food Control Institutions, both for health protection and for frauds prevention. The complexity of honey makes it very difficult to verify the botanical origin. Among the available validated methods, sensory analysis and melissopalynology are the most widely employed. These methods require a long time and deep consolidated expertise. To shorten analysis time while simplifying the analytical procedure, head-space flash gas chromatography was applied in the present study. Chromatographic peak areas were processed by chemometrics (in particular principal components analysis and linear discriminant analysis). Three hundred and thirty-nine honey samples from twelve categories of unifloral honey were analyzed: acacia, citrus, chestnut, thistle, tree heath, eucalyptus, sunflower, rhododendron, lime, French honeysuckle, fir honeydew, and wood honeydew. Each sample was a priori classified by sensory analysis. The multivariate models were validated by cross validation and test-set validation, with predictive abilities always higher than 80%: good results were obtained both in calibration and in prediction mode, showing a good agreement between this new approach and the traditional one for the determination of the botanical origin of honey.

Development and validation of a high-throughput headspace solid-phase microextraction gas chromatography-mass spectrometry methodology for target and suspect determination of honey volatiles

Abstract:The determination of volatile compounds is essential for the chemical characterisation of honey's aroma and its correlation to its sensory profile and botanical origin. The present study describes the development, optimization and validation of a new, simple and reliable method for the determination of volatile compounds in honey using headspace solid-phase microextraction combined with gas chromatography/mass spectrometry (HS-SPME-GC-MS). The optimization of the SPME conditions showed that the ratio of honey: water (2:1) and the incubation temperature (60 °C) are the most critical parameters. Gas chromatography was performed with medium polar Varian CP-Select 624 column and the experimental Retention Index for a number of compounds was determined as an additional identification feature for suspect analysis. The simultaneous use of four internal standards chlorobenzene, benzophenone, 2-pentanol and 4-methyl-2-pentanone and matrix matched calibration enhanced method accuracy achieving recoveries 73-114 % and repeatability ranging between 3.9 and 19 % relative standard deviations. Furthermore, the superiority of the HS-SPME to static head space technique was verified exhibiting four-to nine-fold higher sensitivity. Target and suspect screening were applied to 30 Greek honey samples and 53 volatile compounds belonging to different chemical classes, such as alkanes, aldehydes, ketones, alcohols, and esters were identified with quantified concentrations ranging between 3.1 μg kg-1 (Limonene) up to 20 mg kg-1 (Benzeneacetaldehyde). Among the new findings is the detection of Myrtenol in Greek pine honey and 2,3-butanediol in Greek oak honey. The developed analytical protocol can be a valuable tool in order to chemically characterize honey based on the volatile content.

Abstract:Honey consumption is attributed to potentially advantageous effects on human health due to its antioxidant capacity as well as anti-inflammatory and antimicrobial activity, which are mainly related to phenolic compound content. Phenolic compounds are secondary metabolites of plants, and their content in honey is primarily affected by the botanical and geographical origin. In this study, a high-resolution mass spectrometry (HRMS) method was applied to determine the phenolic profile of various honey matrices and investigate authenticity markers. A fruitful sample set was collected, including honey from 10 different botanical sources (n = 51) originating from Greece and Poland. Generic liquid–liquid extraction using ethyl acetate as the extractant was used to apply targeted and non-targeted workflows simultaneously. The method was fully validated according to the Eurachem guidelines, and it demonstrated high accuracy, precision, and sensitivity resulting in the detection of 11 target analytes in the samples. Suspect screening identified 16 bioactive compounds in at least one sample, with abscisic acid isomers being the most abundant in arbutus honey. Importantly, 10 markers related to honey geographical origin were revealed through non-targeted screening and the application of advanced chemometric tools. In conclusion, authenticity markers and discrimination patterns were emerged using targeted and non-targeted workflows, indicating the impact of this study on food authenticity and metabolomic fields.

Abstract:The present study aimed to differentiate Mediterranean honey samples with regards to their botanical and geographical origin, using UV-vis spectroscopy, combined with multivariate statistical analysis and chemometrics. A total of 151 commercial honey samples were collected from different countries (Greece, Malta, Spain, Tunisia, and Turkey), which were characterized as thyme, pine, or polyfloral. Discrimination of geographical and botanical origin was achieved successfully with Principal Component Analysis (PCA) together with Random Forest (RF), Partial Least Squares – Discriminant Analysis (PLS – DA), and Data Driven – Soft Independent Modelling of Class Analogies (DD-SIMCA) using the spectral range of 220–550 nm. DD-SIMCA achieved very high accuracy, sensitivity, and specificity on the discrimination of both the geographical and botanical origin of honey samples. The results support the good predictive capability of UV-vis spectroscopy combined with chemometrics for the determination of honey origin. This methodology could be utilized for the rapid and non-destructive assessment of Mediterranean honey origin.

Abstract:The objective of the present work was the geographical discrimination of the most common honeydew honeys produced in Greece, namely pine and fir, on the basis of sugar, free amino acid and organic acid content, determined by nuclear magnetic resonance spectroscopy (1H NMR) and high-performance liquid chromatography (HPLC), along with moisture content, sugar ratios, or sugars to moisture ratio, using chemometrics. For this purpose, 39 pine and 31 fir honey samples were collected from professional beekeepers from eight different regions in Greece. Data were subjected to multivariate analysis and modeled using supervised statistical methods. The combination of 1H NMR and HPLC based on metabolites along with the aforementioned physicochemical data resulted in the geographical discrimination of pine and fir honeys. Respective prediction rates were 76.9 and 80.6%, using a model validation technique: the cross-validation method. Present results support the combined use of instrumental and conventional methods for honey geographical origin differentiation.

Abstract:A potentiometric electronic tongue (PE-tongue) and a voltammetric electronic tongue (VE-tongue) were used as rapid techniques to classify and predict the honey samples from different floral and geographical origins. The PE-tongue, which was named alpha-ASTREE, was developed by Alpha M.O.S. (Toulouse, France), and it comprises seven potentiometric chemical sensors. The VE-tongue was self-developed at Zhejiang University and comprises six metallic working sensors. Four types of honey of different floral origins (acacia, buckwheat, data, and motherwort) and four types of acacia honey of different geographical origins were classified by both multisensor systems. Multivariate statistical data analysis techniques such as principal component analysis (PCA) and discriminant function analysis (DFA) were used to classify the honey samples. Both types of electronic tongue have good potential to classify the honey samples, and the positions of the data point for the samples in the PCA score plots based on the VE-tongue were much more closely grouped. Three regression modes, principal component regression (PCR), partial least squares regression (PLSR), and least squared-support vector machines (LS-SVM), were applied for category forecasting. These regression models exhibited a clear indication of the prediction ability of the two types of electronic tongue, and a positive trend in the prediction of the floral and geographical origin of honey was found. Moreover, the performance of these regression models for predicting the four types of honey of different geographical origins by the VE-tongue is very stable. (C) 2014 Elsevier B.V. Abstract:Honey is a natural sweetener constituted by numerous macro- and micronutrients. Carbohydrates are the most representative, with glucose and fructose being the most abundant. Minor honey components like volatile organic compounds (VOCs), minerals, vitamins, amino acids are able to confer honey-specific properties and are useful to characterize and differentiate between honey varieties according to the botanical origin. The present work describes the chemical characterization of honeys of different botanical origin (multifloral, acacia, apple–dandelion, rhododendron, honeydew, and chestnut) produced and collected by beekeepers in the Trentino Alto-Adige region (Italy). Melissopalynological analysis was conducted to verify the botanical origin of samples and determine the frequency of different pollen families. The carbohydrate composition (fourteen sugars) and the profile of VOCs were evaluated permitting to investigate the relationship between pollen composition and the chemical profile of honey. Statistical analysis, particularly partial least squares discriminant analysis (PLS-DA), demonstrates the importance of classifying honey botanical origin on the basis of effective pollen composition, which directly influences honey's biochemistry, in order to correctly define properties and value of honeys.

Abstract:The volatile profiles of 51 samples from 12 monofloral-labelled Portuguese honey types were assessed. Honeys of bell heather, carob tree, chestnut, eucalyptus, incense, lavender, orange, rape, raspberry, rosemary, sunflower and strawberry tree were collected from several regions from mainland Portugal and from the Azores Islands. When available, the corresponding flower volatiles were comparatively evaluated. Honey volatiles were isolated using two different extraction methods, solid-phase microextraction (SPME) and hydrodistillation (HD), with HD proving to be more effective in the number of volatiles extracted. Agglomerative cluster analysis of honey HD volatiles evidenced two main clusters, one of which had nine sub-clusters. Components grouped by biosynthetic pathway defined alkanes and fatty acids as dominant, namely n-nonadecane, n-heneicosane, n-tricosane and n-pentacosane and palmitic, linoleic and oleic acids. Oxygen-containing monoterpenes, such as cis- and trans-linalool oxide (furanoid), hotrienol and the apocarotenoid α-isophorone, were also present in lower amounts. Aromatic amino acid derivatives were also identified, namely benzene acetaldehyde and 3,4,5-trimethylphenol. Fully grown classification tree analysis allowed the identification of the most relevant volatiles for discriminating the different honey types. Twelve volatile compounds were enough to fully discriminate eleven honey types (92%) according to the botanical origin.

Abstract:(1) Background: Citrus honey constitutes a unique monofloral honey characterized by a distinctive aroma and unique taste. The non-targeted chemical analysis can provide pivotal information on chemical markers that differentiate honey based on its geographical and botanical origin. (2) Methods: Within the PRIMA project “PLANT-B”, a metabolomics workflow was established to unveil potential chemical markers of orange blossom honey produced in case study areas of Egypt, Italy, and Greece. In some of these areas, aromatic medicinal plants were cultivated to enhance biodiversity and attract pollinators. The non-targeted chemical analysis and metabolomics were conducted using ultra-high-performance liquid chromatography high-resolution mass spectrometry (UHPLC-HRMS). (3) Results: Forty compounds were disclosed as potential chemical markers, enabling the differentiation of the three orange blossom honeys according to geographical origin. Italian honey showed a preponderance of flavonoids, while in Greek honey, terpenoids and iridoids were more abundant than flavonoids, except for hesperidin. In Egyptian honey, suberic acid and a fatty acid ester derivative emerged as chemical markers. New, for honey, furan derivatives were identified using GC-MS in Greek samples. (4) Conclusions: The application of UHPLC-HRMS metabolomics combined with an elaborate melissopalynological analysis managed to unveil several potential markers of Mediterranean citrus honey potentially associated with citrus crop varieties and the local indigenous flora.

Abstract:The chemical composition and quality of honey are influenced by its botanical, geographic, and entomological origins, as well as climatic conditions. In this study, the physicochemical characteristics, microbial communities, and hydrocarbon compounds of honey produced by Apis mellifera, Apis cerana, Apis laboriosa, Apis dorsata, and Apis florea were elucidated. The physicochemical profile of the honey exhibited significant differences across species, including moisture content (18.27%–23.66%), fructose (33.79%–38.70%), maltose (1.10%–1.93%), electrical conductivity (0.37–0.74 mS/cm), pH (3.36–3.72), diastase activity (4.50–29.97 diastase number), and color (37.90–102.47 mm). Microbial analysis revealed a significant abundance of lactic acid bacteria, particularly the Apilactobacillus genus in A. laboriosa honey and the Lactobacillus in A. florea honey, indicating significant probiotic potential. Chemometric methods, principal component analysis, hierarchical cluster analysis, and orthogonal partial least squares discriminant analysis (OPLS-DA) were used to classify the honey samples based on the 12 beeswax-derived hydrocarbons. The OPLS-DA model demonstrated 100% accuracy in predicting the entomological origin of honey, indicating that specific hydrocarbons are reliable markers for honey classification.

Abstract:Honey is a highly consumed natural product produced by bees which is susceptible to fraudulent practices, some of them regarding its botanical origin. Two HPLC-UV non-targeted fingerprinting approaches were evaluated in this work to address honey characterization, classification, and authentication based on honey botanical variety. The first method used no sample treatment and a universal reversed-phase chromatographic separation. On the contrary, the second method was based on an off-line SPE preconcentration method, optimized for the isolation and extraction of polyphenolic compounds, and a reversed-phase chromatographic separation optimized for polyphenols as well. For the off-line SPE method, the use of HLB (3 mL, 60 mg) cartridges, and 6 mL of methanol as eluent, allowed to achieve acceptable recoveries for the selected polyphenols. The obtained HPLC-UV fingerprints were subjected to an exploratory principal component analysis (PCA) and a classificatory partial least squares-discriminant analysis (PLS-DA) to evaluate their viability as sample chemical descriptors for authentication purposes. Both HPLC-UV fingerprints resulted to be appropriate to discriminate between blossom honeys and honeydew honeys. However, a superior performance was accomplished with off-line SPE HPLC-UV polyphenolic fingerprints, being able to differentiate among the different blossom honey samples under the study (orange/lemon blossom, rosemary, thyme, eucalyptus, and heather). In general, this work demonstrated the feasibility of HPLC-UV fingerprints, especially those obtained after off-line SPE polyphenolic isolation and extraction, to be employed as honey chemical descriptors to address the characterization and classification of honey samples according to their botanical origin.
