Quality Analysis of Five Bioactive Compounds in Mentha Haplocalyx Extracts by High Performance Liquid Chromatography

Abstract:Mentha haplocalyx is derived from the aerial part of Mentha haplocalyx Brip. Caffeic acid, hesperidin, rosmarinic acid, buddleoside and diosmetin are active constituent of Mentha haplocalyx used in many traditional Chinese medicines. This paper describes a sensitive and specific assay for the determination of five bioactive compounds in Mentha haplocalyx extracts. In this paper, the five components were separated on an Agilent Zorbax Eclipse XDB-C-18 column (250 x 4,6 mm, 5 mu m) and detected by diode array detector (DAD). Mobile phase was composed of (A) aqueous phosphoric acid (0.5 %, v/v) and (B) acetortitrile using a gradient elution. Analytes were performed at 30 degrees C with a flow rate of 1 mL/min and UV deteCtion at 340 nm. All calibration curves showed good linear regression (r(2) >= 0.9999) within tested ranges. The LOD and LOQ were 031-5.93. and 2.98-57 mu g/mL, respectively. Overall intra-day and inter-day variations were less than 2.19 % and the average recoveries were 98.23-100.04 % for the analytes. This newly established method is validated as simple, precise and accurate. It can be used as a valid analytical method for intrinsic quality control of Mentha haplocalyx. In conclusion, the proposed method would be-sensitive enough and reliable for comprehensive quality control for Clinical use and modernization of traditional Chinese medicines.
Phenolic Compounds and Biological Activity of Selected Mentha Species

Abstract:Mentha species are widely used as food, medicine, spices, and flavoring agents. Thus, chemical composition is an important parameter for assessing the quality of mints. In general, the contents of menthol, menthone, eucalyptol, and limonene comprise one of the major parameters for assessing the quality of commercially important mints. Building further on the phytochemical characterization of the quality of Mentha species, this work was focused on the composition of phenolic compounds in methanolic extracts. Thirteen Mentha species were grown under the same environmental conditions, and their methanolic extracts were subjected to the LC-MS/MS (liquid chromatography-tandem mass spectrometry) profiling of phenolics and the testing their biological activities, i.e., antioxidant and tyrosinase inhibition activities, which are important features for the cosmetic industry. The total phenolic content (TPC) ranged from 14.81 ± 1.09 mg GAE (gallic acid equivalents)/g for Mentha cervina to 58.93. ± 8.39 mg GAE/g for Mentha suaveolens. The antioxidant activity of examined Mentha related with the content of the phenolic compounds and ranged from 22.79 ± 1.85 to 106.04 ± 3.26 mg TE (Trolox equivalents)/g for M. cervina and Mentha x villosa, respectively. Additionally, Mentha pulegium (123.89 ± 5.64 mg KAE (kojic acid equivalents)/g) and Mentha x piperita (102.82 ± 15.16 mg KAE/g) showed a strong inhibition of the enzyme tyrosinase, which is related to skin hyperpigmentation. The most abundant compound in all samples was rosmarinic acid, ranging from 1363.38 ± 8323 to 2557.08 ± 64.21 μg/g. In general, the levels of phenolic acids in all examined mint extracts did not significantly differ. On the contrary, the levels of flavonoids varied within the species, especially in the case of hesperidin (from 0.73 ± 0.02 to 109. 39 ± 2.01 μg/g), luteolin (from 1.84 ± 0.11 to 31.03 ± 0.16 μg/g), and kaempferol (from 1.30 ± 0.17 to 33.68 ± 0.81 μg/g). Overall results indicated that all examined mints possess significant amounts of phenolic compounds that are responsible for antioxidant activity and, to some extent, for tyrosinase inhibition activity. Phenolics also proved to be adequate compounds, together with terpenoids, for the characterization of Mentha sp. Additionally, citrus-scented Mentha x villosa could be selected as a good candidate for the food and pharmaceutical industry, especially due its chemical composition and easy cultivation, even in winter continental conditions.

Abstract:The aim of the work was to study the composition of the alcohol extract, antioxidant activity and pharmacological properties of peppermint leaves of three forms - menthol, linalool and carvone. Materials and methods: Chromatography-mass-spectrometric analysis was performed on an Agilent Technologies device. For the model evaluation of antioxidant activity, the aerometric method for measuring the total antioxidant activity was used, implemented in the TsvetYauza-01-AA device. Tonic activity was studied in terms of the duration of swimming of laboratory animal (male mice) with a load of 7% of the animal's body weight until complete fatigue - "The mouse forced swim test". The discussion of the results. When studying the chemical composition of medicinal plant raw materials of peppermint leaves of menthol, linalool, carvone types, more than 50 compounds were identified. A comparative analysis showed that the content of menthol in peppermint leaves of the menthol type can exceed the content of menthol in other chemotypes by ten or more times. At the same time, the tonic effect of an aqueous extract from peppermint leaves is mostly associated with the menthol. It should be taken into account when using this type of raw material in the manufacturing. Findings. The raw material of peppermint leaves may have a different degree of pharmacological activity depending on the chemotype, which should be taken into consideration when using this type of raw material.

Abstract:Plant products, especially medicinal herbs and spices, have been used for centuries as a remedy to support human health and improve the flavor of food. Therefore, the purpose of this study was to identify plant species distinguished by their high content of phenolic compounds and high antioxidant activity using advanced multivariate statistical techniques such as Principal Component Analysis (PCA) and Hierarchical Cluster Analysis (HCA). To realize the purpose of the study, the total phenolic (TPC) and flavonoids (FC) content, antioxidant activity (TAC) and Fe(II) ion chelating capacity (FIC) of medicinal herbs and spices from plants belonging to three botanical families, Lamiaceae, Apiaceae and Asteraceae were determined. The interpretation of the obtained data revealed that the studied samples are localized in the PCA and HCA plots according to their TPC, FC, TAC and FIC values. Chemometric analysis confirmed that medicinal herbs and spices from plants belonging to the Lamiaceae family are richer sources of phenolic compounds and exhibit stronger antioxidant activity than those raw materials from plants in the Apiaceae family. In addition, no significant differences were found in terms of TPC, FC, TAC and FIC values between medicinal herbs and spices from the same plant species, i.e., oregano (Origanum vulgare), common thyme (Thymus vulgaris), rosemary (Rosmarinus officinalis), caraway (Carum carvi) and lovage (Levisticum officinale). A close relationship between antioxidant properties and contents of phenolic compounds was also confirmed.

Functional differences between vowel onsets and offsets in temporal perception of speech: local-change detection and speaking-rate discrimination.

Abstract:To provide a perceptual framework for the objective evaluation of durational rules in speech synthesis, two experiments were conducted to investigate the differences between vowel (V) onsets and V-offsets in their functions of marking the perceived temporal structure of speech. The first experiment measured the detectability of temporal modifications given in four-mora (CVCVCVCV) Japanese words. In the V-onset condition, the inter-onset intervals of vowels were uniformly changed (either expanded or reduced) while their inter-offset intervals were preserved. In the V-offset condition, this was reversed. These manipulations did not change the duration of the entire word. Each of the modified words was paired with its unmodified counterpart, and the pair was given to listeners, who were asked to rate the difference between the paired words. The results show that there were no significant differences in the listeners' abilities to detect the temporal modification between the V-onset and V-offset conditions. In the second experiment, the listeners were asked to estimate the differences they perceived in speaking rates for the same stimulus set as that of the first experiment. Interestingly, the results show a clear difference in the listeners' performance between the V-onset and V-offset conditions. Specifically, changing the V-onset intervals changed the perceived speaking rates, which showed a linear relation (r = -0.9) despite the fact that the duration of the entire word remained unchanged. In contrast, modifying the V-offset intervals produced no clear relation with the perceived speaking rates. The second experiment also showed that the listeners performed well in speaking rate discrimination (3.5%-5% in the change ratio). These results are discussed in relation to the differences in the listeners' temporal processing range (local or global) between the two experiments.

Abstract:Lavender is a valued plant due to its cosmetic, perfumery, culinary, and health benefits. A wide range of applications is related to the composition of bioactive compounds, the quantity and quality of which is determined by various internal and external factors, i.e., variety, morphological part of the plant, and climatic and soil conditions during vegetation. In the presented work, the characterization of antimicrobial properties as well as the qualitative and quantitative assessment of bioactive compounds in the form of polyphenols in ethanol extracts from leaves and flowers of Lavandula angustifolia Mill. intended for border hedges, cultivated in the region of southern Poland, were determined. The composition of the fraction of volatile substances and antioxidant properties were also assessed. The conducted research shows that extracts from leaves and flowers significantly affected the viability of bacterial cells and the development of mold fungi. A clear decrease in the viability of bacteria and C. albicans cells was shown in the concentration of 0.32% of extracts. Leaf extracts were characterized by a much higher content of polyphenols and antioxidant properties than flower extracts. The composition of volatiles measured by GC-MS was significantly different between the extracts. Linalyl acetate and ocimene isomers mix dominated in flower extracts, whereas coumarin, γ-cadinene, and 7-methoxycoumarin were identified as dominant in leaf extracts.

Abstract:The present work describes a coke oven soil treatability study by landfarming, conducted on-site in a field scale facility covering 100 m2. The soil contamination was mainly due to high concentrations of polynuclear aromatic hydrocarbons (PAHs) up to 1140 mg/Kg dry weight (ΣEPA). Along the treatment process the soil was characterised at the chemical, microbiological and ecotoxicological levels. After 3 months a reduction of 63% in total PAHs concentration was observed, being detected a more pronounced reduction for PAHs with 2, 3 and 4 rings (79%). Concomitantly, a change in the composition of the microbial population was observed with a significant increase in the PAHs degrading and total heterotrophic colonies. Concerning the ecotoxicity and genotoxicity data no effect was detected in the treated soil samples eluates.

Abstract:Culinary spices and herbs have been used to impart a characteristic flavour and aroma in food due to their appealing fragrance. Recently, bioactive compounds from herbs, especially phenolics, have gained much attention due to their potential health outcomes. The aim of this study was to characterize and quantify the phenolic compounds from 10 widely used Australian-grown herbs (oregano, rosemary, bay, basil, sage, fenugreek, dill, parsley, mint and thyme). For this purpose, liquid chromatography mass spectrometry (LC-MS) was used for the complete profiling of polyphenolic compounds and quantification of abundant phenolic compounds was completed with high-performance liquid chromatography—photodiode array detection (HPLC-PDA). Polyphenols from Australian-grown herbs were estimated through total phenolic content (TP), total flavonoids (TF) and total tannins (TT) along with their in-vitro antioxidant activities. Oregano and mint were estimated with the highest value of TP (140.59 ± 9.52 and 103.28 ± 8.08 mg GAE/g, milligram gallic acid equivalent/gram) while rosemary and mint had the highest TF (8.19 ± 0.74 and 7.05 ± 0.43 mg QE (quercetin equivalent)/g). In this study, eighty-four (84) phenolic compounds were screened and confirmed through LC-MS/MS by comparing their masses and fragmentation pattern with published libraries. The results of this study validate the use of these herbs as bioactives and their positive impact on human health.

Monoterpene composition of essential oil from peppermint (Mentha x piperita L.) with regard to leaf position using solid-phase microextraction and gas chromatography/mass spectrometry analysis

Abstract:Monoterpene compounds of leaf pairs and flowers of Mentha x piperita have been studied by direct headspace sampling using solid-phase microextraction coupled with gas chromatography/mass spectrometry (SPME-GC/MS). The content of peppermint-characteristic compounds such as menthol, menthyl acetate, and neomenthol increased in a basipetal direction (older plant parts), whereas menthone and isomenthone showed higher levels in the acropetal direction (younger plant parts). Higher levels of menthofuran were found in peppermint flowers in contrast to the leaves. SPME sampling resulted in relatively higher amounts of high-volatile monoterpenes and lower detection of less volatile compounds such as menthol and menthone, compared to solvent-based samples from essential oil distillation.

Abstract:Peppermint essential oil (EO) has a multitude of applications, such as a fragrance in cosmetics, personal care and industrial products, or as a flavoring ingredient in food and beverages. Despite its popularity and economic significance, peppermint EO is often adulterated to reduce production costs and to increase profits. Although the ISO standard for peppermint EO exists, detecting sophisticated forms of adulteration remains challenging.The current study used conventional and chiral GC/MS analysis of volatiles compounds, and chemometric techniques to evaluate an extensive set of authentic peppermint EO (n = 22) and commercial products (n = 36) purported to contain peppermint EO. Specifically, thirty-six terpenoids were examined in each sample and compared with the ISO standard. Fifty-three percent of the selected commercial products did not meet the ISO specifications and the ratio between menthone/isomenthone was proven to be a good indicator for authentication and adulteration detection. Chiral GC/MS was further employed to measure eight terpenoids: α-pinene, β-pinene, limonene, menthol, menthone, isomenthone, pulegone, and menthyl acetate. The enantiomeric compositions of 27 commercial products were above or below the norm measured from authentic peppermint EOs. Of the 27 samples, eight met the ISO standard. A sample class prediction (SCP) model based on partial least squares-discriminant analysis (PLS-DA) of conventional GC/MS data was constructed using authentic peppermint EOs and cornmint EOs. The model can distinguish the most common types of peppermint EOs (US, India, and US/India blend) and cornmint EOs sold in the US market. After construction, the SCP model was then used to analyze commercial samples. One sample, which passed both ISO specification and chiral analysis, was identified as outlier by the SCP model. Overall, the applicability of combining both conventional and chiral GC/MS along with chemometric tools has been successfully demonstrated to address the overall quality of peppermint EOs in commerce and may help combat sophisticated adulteration.
LC-MS-based metabolite profiling of methanolic extracts from the medicinal and aromatic species Mentha pulegium and Origanum majorana

Abstract:Introduction: There has been increasing interest dedicated to the phenolic compounds with a view to their antioxidant and healthy properties. Recent studies have focused on plants from the Lamiaceae family with special interest in phenolic compounds antioxidant potential. Objective: The metabolite profile of methanolic extracts from two Lamiacea medicinal plants was investigated. Materials and methods: Mentha pulegium and Origanum majorana methanolic extracts were analysed using reversed-phase ultra-high-performance liquid chromatography (RP-UHPLC) coupled to electrospray ionisation quadrupole time-of-flight mass spectrometry (ESI-QTOF-MS) detection in the negative ion mode. Results: A total of 85 metabolites were characterised from different families, such as organic acids and derivatives, amino acids and derivatives, nucleosides, phenolic compounds as well as other polar metabolites, by using the MS and MS/MS information provided by the QTOF-MS. However, the total phenols and flavonoids were also quantified spectrophotometrically and they registered higher amounts in Mentha pulegium than in Origanum majorana extract. Gallocatechin was the major compound in M. pulegium extract whereas quercetin dimethyl ether, jaceidin and dihydrokaempferide were the major ones in O. majorana extract. Conclusion: The distribution of phenolic compounds in the methanolic extract showed a variation among studied plants. Mentha pulegium can be considered as a source of gallocatechin.

Abstract:Medicinal plants are a rich source of secondary metabolites, which have tremendous potential in the development and discovery of new drugs. Mint leaves are used to treat diarrhea and diarrhoea, hypertension, liver and spleen infections, asthma, rheumatic pain, joint irritation, sensitivity and jaundice. The menthol content in natural mint ointment is used in the medicine, perfumery and food businesses. This research aims to determine the yield and content of active secondary metabolite compounds in mint (Mentha arvensis) leaves with an emphasis on alkaloids, flavonoids, tannins, saponins, terpenoids and steroids. The examination strategy uses maceration (watering) techniques for 3 days. Then phytochemical screening was carried out on mint leaves using standard methods. The results showed that the yield of 96% ethanol concentrate from mint leaves was 5.75% and contained a mixture of secondary metabolites of alkaloids, tannins, saponins and steroids.

Abstract:Plant extracts are a source of valuable ingredients that can be used in many industries. This paper presents research on the content of selected bioactive compounds in extracts obtained from various plant materials. Raw materials have a documented use in traditional medicine not only in Poland. The tested plants were: bitter melon (fruit), elderberry (flowers, fruit, leaves), wild rose (fruit, flesh, seeds), mountain ash (fruit), guelder rose (fruit), and sea buckthorn (fruit, leaves, pomace). The main goal of these tests is to indicate the potential raw materials that may constitute an alternative source of bioactive compounds with antimicrobial activity. The plant material was tested for the content of bioactive antioxidant compounds and possible antimicrobial activity. The content of polyphenols (phenolic acids and flavonoids) was analyzed using UPLC/PDA, sterols, organic acids, and other bioactive compounds. The minimum inhibitory concentration (MIC) was determined. The total free phenolic acids (TPC) and total free flavonoids (TFC) of all plant raw materials was varied and ranged from 0.21 (mg RUTE/1 g of extract) to 38.30 mg RUTE/1 g of extract) for TFC. The concentration of sterols was, on average, about 10 mg/1 g of extract. The value of approx. 20 mg/1 g of the extract was recorded for bitter melon and beach rose. The content of organic acids was about 1.5 mg/1 g of the extract to even 13 mg/1 g of the extract for sea buckthorn berries. The most sensitive to the extracts' activity were the following bacteria: M. luteus, P. mirabilis, P. fragii, S. enteritidis, and E. coli. The tested plant materials can be used in various industries as a source of bioactive compounds of an antibacterial nature.

Abstract:Even though Melissa officinalis L. (lemon balm, fam. Lamiaceae) is a well-known medicinal, aromatic, and spicy plant, and its physicochemical profile and biological activity have been investigated, there is no detailed research regarding the influence of solvent nature (polarity) on the extraction of active compounds (total polyphenols and total flavonoids) and antioxidant activities of the obtained extracts. Therefore, this study aimed to evaluate the polyphenol and flavonoid contents, as well as the antioxidant potential of lemon balm extracts obtained by varying the polarity of the extraction solvents (methyl alcohol, acetone, ethyl acetate, and deionized water) and using three extraction techniques (maceration-MAC, ultrasound-UAE, and microwave-assisted extraction-MAE). Two in vitro tests (ABTS and FRAP assays) were used to determine the antioxidant activity of the extracts. Total tannin and total protein contents, extraction yield, and physical properties of the selected extracts were measured as well. The highest content of polyphenols was found for the methanolic and water extracts obtained by all three extraction techniques, while the highest flavonoid yield was detected only in the mentioned methanolic extracts. The microwave reactor provided methanolic, ethyl acetate, and water extracts with the highest ABTS radical scavenging activity, while in the case of UAE, it was methanolic and water extracts, and in the case of MAC only water extracts. On the other hand, among lemon balm extracts from MAC, water extract possessed the highest ferric reducing power, whereas in UAE and MAE, it was ethyl acetate extract. Total tannin content determined in selected water extracts was 2.55, 4.53, and 1.83 mg tannic acid equivalent (TAE)/mL, while total proteins amounted to 1.34, 1.50, and 1.31 mg/mL using MAC, UAE, and MAE, respectively. The content of total extractive substances in the form of the extraction yield was also determined for selected water extracts and amounted to 12.6 % for MAC, 17.2 % for UAE, and 36.8 % for MAE. Further, this research has included the investigation of some physical properties of lemon balm water extracts, such as conductivity (3.68-4.14 mS/cm), pH (5.99-6.43), density (0.854-0.901 g/mL), surface tension (26.0-31.7 mN/m), and viscosity (1.18-1.21 mPa·s). This research represents the base for the future encapsulation of lemon balm extracts, enriched in polyphenol content, in a novel type of biofunctional carrier that potentially can be applied in the pharmacy, chemical industry, and biotechnics.
The Profile and Content of Polyphenolic Compounds and Antioxidant and Anti-Glycation Properties of Root Extracts of Selected Medicinal Herbs

Abstract:The objective of our study was to analyse the extracts from six medicinal herb roots (marshmallow, dandelion, liquorice, angelica, burdock, and comfrey) in terms of antioxidant capacity (ABTS, DPPH) and inhibition of advanced glycation end product (AGEs) formation. The quantification of phenolic acids and flavonoids was analysed using the UHPLC-DAD-MS method. Fifteen polyphenolic compounds were detected in the studied herbs. The higher number of polyphenols were found in marshmallows (ten polyphenols), while the lowest was in comfrey (five compounds). Liquorice root revealed the highest individual phenolic concentration (382 µg/g dm) with the higher contribution of kaempferol-3-O-rutinoside. Comfrey root extract was characterised by the most abundant TPC (Total Phenolic Content) value (29.79 mg GAE/ g dm). Burdock and comfrey showed the strongest anti-AGE activity studies with the BDA-GLU model. Burdock root was also characterised by the highest anti-AGE activity in the BSA-MGO model. The highest antioxidant capacity was determined by ABTS (72.12 µmol TE/g dw) and DPPH (143.01 µmol TE/g dw) assays for comfrey extract. The p-coumaric acid content was significantly correlated with anti-AGE activity determined by the BSA-MGO model. This research sheds new light on the bioactivity of root herbs, explaining the role of p-coumaric acid in preventing diabetes.

Abstract:Plants of the genus Monarda receive growing interest as the sources of herbal raw materials with wide range of potential applications in food, cosmetics, and phytopharmaceutical industry. This study aimed to evaluate the differences in chemical characteristics and biological activity among different organs of plants representing three underinvestigated species of this genus: Monarda bradburiana L. C. Beck, Monarda × media Willd., and Monarda punctata L. The content of phenolic compounds and the antioxidant activity of leaves, stems, and inflorescences were determined. Essential oil (EO) content, composition, and antimicrobial activity were also examined. M. punctata leaves and inflorescences had the highest EO content (4.43% and 5.59%, respectively), with carvacrol as a dominant constituent. Leaf EO was also rich in thymoquinone (17.48%). In EOs of M. bradburiana and M. × media, thymol dominated. EOs inhibited the growth of all tested strains of microorganisms at a concentration of 0.625 μL × mL‐1. The studied plant organs were rich in phenolic compounds, especially rosmarinic acid. M. bradburiana inflorescences were distinguished by high linarin content. Differences in flavonoid distribution seem to have special chemotaxonomic importance. Further research is needed to facilitate standardisation of the investigated plant organs as potential new herbal raw materials.

Biological Activity of Horehound (Marrubium vulgare L.) Herb Grown in Poland and Its Phytochemical Composition

Abstract:Marrubium vulgare (Lamiaceae) is a plant which has long been known and used in traditional medicine for various purposes. However, few recent studies have documented its chemical composition and biological properties. The present study investigated the phytochemical composition of horehound, as well as its protective, antioxidant, and antimicrobial potential. GC-MS analysis revealed that the major components of horehound essential oil are E-caryophyllene (35.7%), germacrene D (25.2%), and bicyclogermacrene (10.6%). The biological activity of horehound hydroethanolic herb extract derives from multiple chemical compounds, including polyphenols (55.72 mg/mL), flavonoids (11.01 mg/mL), phenolic acids (4.33 mg/mL), and tannins (4.46 mg/mL). Chromatographic analyses of the extract identified 12 phenolic compounds, of which ferulic acid, catechin, quercetin, protocatechuic acid, rutin, and syringic acid (35.42, 24.69, 20.65, 18.70, 14.46, and 12.69 mg/mL, respectively) were the main constituents. Its DPPH radical scavenging ability was 68.29%, while its antioxidant properties, determined by the FRAP method, were at the level of 1.22 mmol/L. Moreover, M. vulgare extract decreased the level of intracellular reactive oxygen species in the fibroblasts and keratinocytes in vitro, achieving the strongest antioxidant effect at a concentration of 2.5% in the case of both types of skin cells. Extracts from the horehound herb showed significant antimicrobial and anti-biofilm activity, confirming the plant's potential in therapeutic applications against various microbial pathogens (gram-positive and gram-negative bacteria and fungi). The research results demonstrate the protective effect of horehound extract on the viability of both fibroblasts and keratinocytes in vitro. To sum up, M. vulgare, as a valuable natural material with high preventive and therapeutic effectiveness, is a potential candidate for new applications in the pharmaceutical and cosmetics industries.

Development of high-performance thin layer chromatography method for identification of phenolic compounds and quantification of rosmarinic acid content in some species of the Lamiaceae family

Abstract:Aim: Representatives of Nepetoideae Burnett subfamily are promising species of the Lamiaceae Martinov family because of accumulating such valuable groups of secondary metabolites as terpenoids and polyphenols. A high-performance thin layer chromatography (HPTLC) fingerprint method for the qualitative determination of phenolic compounds and for the quantification of rosmarinic acid (RA) content in methanol extracts of five species of this subfamily was developed for the first time. Materials and methods: Dried aerial parts of Dracocephalum moldavica L., Ocimum americanum L., Lophanthus anisatus (Nutt.) Benth., Monarda fistulosa L., and Satureja hortensis L. collected in flowering period were macerated with methanol. The HPTLC analysis was conducted using the CAMAG analytical system (Muttenz, Switzerland). The comparative analysis of RA contents was performed by HPTLC densitometric detection at λ = 366nm. Results: Identification of polyphenols in the investigated herbs was performed by comparison of a color and R f of the chromatographic zones with six reference standards: rutin, apigenin, luteolin, caffeic acid, chlorogenic acid, and RA. HPTLC method was also validated for the quantification of RA in the extracts of investigated herbs. RA contents decreased in such a sequence: D. moldavica (24.83 ± 0.78mg/g) > M. fistulosa (20.32 ± 0.64mg/g) > O. americanum (19.59 ± 0.61mg/g) > S. hortensis (18.77 ± 0.52mg/g) > L. anisatus (12.61 ± 0.43mg/g). Conclusion: Obtained data can facilitate the differentiation of investigated species using the chromatographic fingerprints of their phenolic compounds. Developed and validated HPTLC method provides an approach to estimate RA content as a common marker of investigated herbs.
The Influence of Copper and Zinc on Photosynthesis and Phenolic Levels in Basil (Ocimum basilicum L.), Borage (Borago officinalis L.), Common Nettle (Urtica dioica L.) and Peppermint (Mentha piperita L.)

Abstract:This work is aimed at relationships which govern zinc and copper uptake by four popular medicinal herbs: basil (Ocimum basilicum L.), borage (Borago officinalis L.), common nettle (Urtica dioica L.) and peppermint (Mentha piperita L.). They are often grown in soils with significant copper or zinc levels. Herbs were cultivated by a pot method in controlled conditions. Manganese, iron, copper and zinc concentrations were determined by High-Resolution Continuum Source Flame Atomic Absorption Spectrometry. The efficiency of photosynthesis was estimated by measuring the chlorophyll content, water use efficiency, net photosynthesis, intercellular CO2, stomatal conductance, and transpiration rate. Phenolic compounds were determined by the Folin–Ciocalteu method. Analysis of variance showed that herbs grown in soil treated with copper exhibited a lower iron content in roots, while manganese behaved in the opposite way. The only exception was borage, where a decrease in the manganese content in roots was observed. Both copper and zinc supplementations increased the total content of phenolics, while the highest increases were observed for common nettle and basil. Peppermint and borage responded less to supplementation. In the majority of samples, zinc and copper did not significantly affect the photosynthesis. Herbal extracts from common nettle and basil had unique antioxidant properties and may be good free radical scavengers.
