Abstract:Two aerobic, Gram-negative, non-motile, non-sporulating, yellow-pigmented bacteria, strains TH92(T) and TH91, were isolated from a hot spring located in Laibin, Guangxi, in the south-eastern geothermal area of China. The isolates grew at 40-77 degrees C (optimally at 70 degrees C) and at pH 6.0-9.5 (optimally at pH 7.5-8.0). Phylogenetic analysis of 16S rRNA gene sequences and levels of DNA-DNA relatedness together indicated that the new isolates represented a novel species of the genus Thermus with closest affinity to Thermus aquaticus, Thermus igniterrae and Thermus thermophilus. Compared with their closest relatives, strains TH92( T) and TH91 were able to assimilate a wider range of carbohydrates, amino acids and organic acids as sole carbon sources for growth, such as lactose and melibiose. The new isolates had lower combined levels of C(16 : 0 ) and iso-C(16 : 0) compared with their closest relatives. On the basis of polyphasic taxonomic characterization, strains TH92(T) and TH91 are considered to represent a single novel species of the genus Thermus, for which the name Thermus arciformis sp. nov. is proposed. The type strain is TH92(T) (=CGMCC 1.6992(T) =JCM 15153(T)).

Abstract:A strictly aerobic, Gram-stain-positive, motile and spore-forming bacterium, strain 3nP4(T), was isolated from the Puge hot spring located in the south-western geothermal area of China. Strain 3nP4(T) grew at 38-66 °C (optimum 57-60 °C), at pH 6.0-9.3 (optimum 7.0-7.5) and with 0-4 % (w/v) NaCl (optimum 0-0.5 %). Phylogenetic analysis of 16S rRNA gene sequences, as well as DNA-DNA relatedness values, indicated that the isolate represents a novel species of the genus Anoxybacillus, related most closely to Anoxybacillus voinovskiensis DSM 12111(T). Strain 3nP4(T) had diphosphatidylglycerol, phosphatidylglycerol, phosphatidylethanolamine and one unidentified phospholipid as major polar lipids and iso-C15 : 0 and iso-C17 : 0 as major fatty acids, which are both typical chemotaxonomic characteristics of the genus Anoxybacillus. The mean DNA G+C content of strain 3nP4(T) was 39.2±0.95 mol% (HPLC). A distinctive characteristic of the novel isolate was its extreme reliance on vitamin mixture or yeast extract for growth. Based on data from this taxonomic study using a polyphasic approach, strain 3nP4(T) is considered to represent a novel species of the genus Anoxybacillus, for which the name Anoxybacillus vitaminiphilus sp. nov. is proposed. The type strain is 3nP4(T) ( = CGMCC 1.8979(T) = JCM 16594(T)).

Abstract:A Gram-stain-negative, non-endospore-forming, motile, short rod-shaped strain, designated SYSU G07232 T , was isolated from a hot spring microbial mat, sampled from Rehai National Park, Tengchong, Yunnan Province, south-western China. Strain SYSU G07232 T grew at 25–50 °C (optimum, 37 °C), at pH 5.5–9.0 (optimum, pH 6.0) and tolerated NaCl concentrations up to 1.0 % (w/v). Phylogenetic analysis based on the 16S rRNA gene sequences revealed that strain SYSU G07232 T showed closest genetic affinity with Chelatococcus daeguensis K106 T . The genomic features and taxonomic status of this strain were determined through whole-genome sequencing and a polyphasic approach. The predominant quinone of this strain was Q-10. Major cellular fatty acids comprised C 19 : 0 cyclo ω 8 c and summed feature 8. The whole-genome length of strain SYSU G07232 T was 4.02 Mbp, and the DNA G+C content was 69.26 mol%. The average nucleotide identity (ANIm ≤84.85 % and ANIb ≤76.08 %) and digital DNA–DNA hybridization (≤ 21.9 %) values between strain SYSU G07232 T and the reference species were lower than the threshold values recommended for distinguishing novel prokaryotic species. Thus, based on the provided phenotypic, phylogenetic, and genetic data, it is proposed that strain SYSU G07232 T (=KCTC 8141 T =GDMCC 1.4178 T ) be designated as representing a novel species within the genus Chelatococcus , named Chelatococcus albus sp. nov.

Abstract:Two aerobic, non-motile and non-sporulating bacteria, strains LY1( T) and L462, were recovered from a hot spring located in the Qianshan area of north-eastern China. The novel strains had Gram-negative cell walls and grew at 30-66 degrees C (optimum 55-60 degrees C) and pH 5.5-10.0 (optimum pH 8.0-8.5). 16S rRNA gene sequence similarity analysis revealed that strains LY1( T) and L462 were most closely related to Meiothermus ruber ATCC 35948(T), M. taiwanensis WR-30(T) and M. cerbereus GY-1(T), with 97.1-98.4 % sequence similarity. Phylogenetic analysis indicated that the new isolates represented a novel species by forming a distinctive lineage within genus Meiothermus, which was confirmed by DNA-DNA reassociation values. Strain LY1( T) was obviously different from its closest relatives in a number of phenotypic characteristics, such as the inability to hydrolyse casein or to assimilate melibiose, trehalose, sucrose and lactose. Strain L462 was distinct in its ability to reduce nitrate. In addition, the novel strains were distinct in having larger amounts of anteiso-C(17 : 0) than their closest phylogenetic neighbours. On the basis of this polyphasic taxonomic characterization, the name Meiothermus cateniformans sp. nov. is proposed for the novel species, currently represented by the type strain LY1( T) (=CGMCC 1.6990(T) =JCM 15151(T)) and strain L462 (=CGMCC 1.6989 =JCM 15150).
Rhodococcus biphenylivorans sp. nov., a polychlorinated biphenyl-degrading bacterium

Abstract:A Gram-positive, aerobic, non-motile and rod–coccus shaped novel actinobacterial strain, designated as TG9 T , was isolated from a polychlorinated biphenyl (PCB)-contaminated sediment in Taizhou city, Zhejiang province, eastern China. The isolate was observed to grow at 10–45 °C (optimum 28–32 °C), pH 5.0–11.0 (optimum pH 7.0–8.0) and with 0–9.0 % (w/v) NaCl (optimum 0–3.0 %). Comparison of the 16S rRNA gene sequences of strain TG9 T and other members of the genus Rhodococcus showed that strain TG9 T shared highest similarities with Rhodococcus pyridinivorans DSM 44555 T (99.4 %), R. rhodochrous DSM 43241 T (99.2 %), R. gordoniae DSM 44689 T (99.2 %) and R. artemisiae DSM 45380 T (98.2 %). However, low levels of DNA–DNA relatedness (15–48 %), which are below the 70 % limit for prokaryotic species identification, were obtained by DNA–DNA hybridization. Strain TG9 T was found to contain meso-diaminopimelic acid as the diagnostic diamino acid and arabinose and galactose in the whole-cell hydrolysate. Mycolic acids were found to be present. The major fatty acids were identified as C 16:0 , C 16:1 ω 7 c and/or iso-C 15:0 2-OH, 10-methyl C 18:0 and C 18:1 ω 9 c . The only menaquinone detected was MK-8 (H 2 ). The major polar lipids detected were diphosphatidylglycerol, phosphatidylethanolamine, phosphatidylinositol, glycolipid and traces of some unknown lipids. The genomic DNA G+C content of strain TG9 T was determined to be 62.8 %. The combined phenotypic and genotypic data show that the strain represents a novel species of the genus Rhodococcus for which the name Rhodococcus biphenylivorans sp. nov. is proposed, with the type strain TG9 T (=CGMCC 1.12975 T = KCTC 29673 T = MCCC 1K00286 T ).
Rufibacter psychrotolerans sp. nov., a Cold-Tolerating Novel Species Isolated from Desert Soil

Abstract:Strain SYSU D00308 T , a short-rod-shaped bacterium, was isolated from a sandy soil collected from the Gurbantunggut Desert, Xinjiang, PR China. Strain SYSU D00308 T was Gram-stain-negative, aerobic, pink-pigmented, non-motile, catalase- and oxidase-positive. The strain grew at 4–37 °C, pH 5.0–8.0 and 0–1.5% (w/v) NaCl. 16S rRNA gene sequencing analyses demonstrated that strain SYSU D00308 T belonged to the genus Rufibacter and exhibited the highest sequence similarity (97.4%) to Rufibacter glacialis MDT1-10-3 T . Summed features 3, 4, and iso-C 15:0 were the major fatty acids, and menaquinone 7 (MK-7) was the sole respiratory menaquinone. The polar lipid profiles comprised phosphatidylethanolamine, an unidentified glycolipid, an unidentified phospholipid, two unidentified aminophospholipids, and two unidentified lipids. The genome size and DNA G + C content of strain SYSU D00308 T were 5,176,683 bp and 54.8%, respectively. The average nucleotide identity (ANI) and digital DNA–DNA hybridization (dDDH) values between SYSU D00308 T and members of the genus Rufibacter were 77.7–81.8% and 20.4–23.4% respectively, which were less than the corresponding thresholds (ANI: 95–96%; dDDH: 70%) for prokaryotic species definition. According to the genotypic, phenotypic and phylogenetic characteristics, strain SYSU D00308 T represents a novel species of the genus Rufibacter . We propose the name, Rufibacter psychrotolerans sp. nov., with SYSU D00308 T (= CGMCC 1.18621 T = KCTC 82275 T = MCCC 1K04970 T ) as the type strain.

Abstract:A Gram-stain-negative, non-motile, aerobic, rod-shaped bacterium, designated 15182T, was isolated from a saline lake in China. The novel strain 15182T was able to grow at 10-40 °C (optimum, 37 °C), pH 7.0-8.0 (optimum, 7.5) and with 0.5-4 % NaCl (optimum, 2-3 %, w/v). The phylogenetic analysis based on 16S rRNA gene sequences revealed that strain 15182T was most closely related to the genus Rhodohalobacter by sharing the highest sequence similarity of 97.0 % with Rhodohalobacter halophilus JZ3C29T. Chemotaxonomic analysis showed that the sole respiratory quinone was menaquinone 7, the major fatty acids included C16 : 0 N alcohol and C16 : 1ω11c. The major polar lipids included diphosphatidylglycerol, phosphatidylglycerol, phosphatidylethanolamine, four uncharacterized glycolipids, one uncharacterized phospholipid and two uncharacterized lipids. The genomic DNA G+C content of the strain 15182T was 42.4 mol%. The average nucleotide identity value between 15182T and R. halophilus JZ3C29T was 75.4 %, and the in silico DNA-DNA hybridization value of the two strains was 19.1 %. On the basis of its phenotypic, chemotaxonomic, genotypic and genomic characteristics presented in this study, strain 15182T is suggested to represent a novel species in the genus Rhodohalobacter, for which the name Rhodohalobacter barkolensis sp. nov. is proposed. The type strain is 15182T (=KCTC 62172T=MCCC 1K03442T). An emended description of the genus Rhodohalobacter is also presented.
Polyphasic Taxonomic Characterisation of a Novel Strain As Pararhizobium Haloflavum Sp. Nov., Isolated from Soil Samples Near a Sewage Treatment Tank

Abstract:A Gram-stain negative, aerobic, motile and ovoid- to rod-shaped bacteria strain, designated XC0140T, was isolated from soil samples near the sewage treatment tank of a chemical factory in Zhejiang Province, China, and subjected to polyphasic taxonomic investigation. Strain XC0140T grew at 10–37 °C and pH 6.0–9.0 (optimum, 35 °C and pH 7.5) and with 0–17% (w/v) NaCl (optimum, 1%). According to phylogenetic analysis based on 16S rRNA gene sequences, strain XC0140T was assigned to the genus Pararhizobium with high 16S rRNA gene sequence similarity of 95.97% to “Pararhizobium helanshanense CCNWQTX14T”, followed by Pararhizobium sphaerophysae CCNWGS0238T (95.95%). Chemotaxonomic analysis showed that strain XC0140T contains ubiquinone-10 as the predominant respiratory quinone and possessed summed feature 8 (comprising C18: 1 ω7c and/or ω6c), 11-methyl C18:1 ω7c, C18: 0 and C16: 0 as predominant forms of fatty acids. The polar lipids of strain XC0140T consisted of seven phospholipids (PL), two aminolipids (AL), one glycolipid (GL) and three unidentified lipids (L1, L2 and L3). The DNA G+C content was 62.7 mol%. Based on the polyphasic taxonomic characterization, strain XC0140T is considered to represent a novel species of the genus Pararhizobium, for which the name Pararhizobium haloflavum sp. nov. is proposed. Noncanonical Wnt Signaling Promotes Obesity-Induced Adipose Tissue Inflammation and Metabolic Dysfunction Independent of Adipose Tissue Expansion

Abstract:Adipose tissue dysfunction plays a pivotal role in the development of insulin resistance in obese individuals. Cell culture studies and gain-of-function mouse models suggest that canonical Wnt proteins modulate adipose tissue expansion. However, no genetic evidence supports a role for endogenous Wnt proteins in adipose tissue dysfunction, and the role of noncanonical Wnt signaling remains largely unexplored. Here we provide evidence from human, mouse, and cell culture studies showing that Wnt5a-mediated, noncanonical Wnt signaling contributes to obesity-associated metabolic dysfunction by increasing adipose tissue inflammation. Wnt5a expression is significantly upregulated in human visceral fat compared with subcutaneous fat in obese individuals. In obese mice, Wnt5a ablation ameliorates insulin resistance, in parallel with reductions in adipose tissue inflammation. Conversely, Wnt5a overexpression in myeloid cells augments adipose tissue inflammation and leads to greater impairments in glucose homeostasis. Wnt5a ablation or overexpression did not affect fat mass or adipocyte size. Mechanistically, Wnt5a promotes the expression of proinflammatory cytokines by macrophages in a Jun NH2-terminal kinase–dependent manner, leading to defective insulin signaling in adipocytes. Exogenous interleukin-6 administration restores insulin resistance in obese Wnt5a-deficient mice, suggesting a central role for this cytokine in Wnt5a-mediated metabolic dysfunction. Taken together, these results demonstrate that noncanonical Wnt signaling contributes to obesity-induced insulin resistance independent of adipose tissue expansion.
Sabulibacter ruber gen. nov., sp. nov., a novel bacterium in the family Hymenobacteraceae, isolated from desert soil

Abstract:A novel Gram-stain-negative, aerobic, oxidase-positive, catalase-positive, non-motile, short rod-shaped, red-pigmented strain, designated as SYSU D00434T, was isolated from a dry sandy soil sample collected from the Gurbantunggut desert in Xinjiang, north-west PR China. Strain SYSU D00434T was found to grow at 4-37 °C (optimum, 28-30 °C), pH 6.0-8.0 (optimum, pH 7.0) and with 0-1.5 % (w/v) NaCl (optimum, 0-0.5 %). The predominant respiratory quinone was MK-7 and the major fatty acids (>10 %) were C16 : 1 ω5c, iso-C15 : 0, summed feature 3 (C16 : 1 ω6c and/or C16 : 1 ω7c) and summed feature 4 (anteiso-C17 : 1 B and/or iso-C17 : 1 I). The polar lipids consisted of phosphatidylethanolamine, two unidentified polar lipids, two unidentified aminolipids, two unidentified phospholipids and two unidentified glycolipids. The genomic DNA G+C content of strain SYSU D00434T was 50.6 mol%. Phylogenetic analyses based on 16S rRNA gene sequences indicated that strain SYSU D00434T belonged to the family Hymenobacteraceae, and shared a sequence similarity of less than 94.6 % to all validly named taxa. Based on the phenotypic, phylogenetic and chemotaxonomic properties, strain D00434T is proposed to represent a new species of a new genus, named Sabulibacter ruber gen. nov., sp. nov., within the family Hymenobacteraceae. The type strain is SYSU D00434T (=CGMCC 1.18624T=KCTC 82276T=MCCC 1K04975T).
Desertibaculum subflavum gen. nov., sp. nov., a novel member of the family Sneathiellaceae isolated from the Kumtag Desert soil

Abstract:A novel rod-shaped bacterium, designated as strain SYSU D60015T that formed yellowish colonies was isolated from a sandy soil collected from the Kumtag Desert in Xinjiang, China. Cells were Gram-stain-negative, oxidase-positive, catalase-negative and motile with a single polar flagellum. Growth optimum occurred between 28 and 37 °C, pH 7.0 and with 0-0.5% (W/V) NaCl. The predominant cellular fatty acids (> 5%) were summed feature 8 (C18:1 ω7c and/or C18:1 ω6c), C19:0 cyclo ω8c, C18:1 ω7c 11-methyl and C16:0. The polar lipid profile contained one phosphatidylethanolamine, one diphosphatidylglycerol, one phosphatidylglycerol, one unidentified phospholipid, three unidentified aminolipids, two unidentified aminophospholipids and seven unidentified lipids. The only respiratory quinone was ubiquinone-10. Based on 16S rRNA gene sequence phylogenetic analysis, strain SYSU D60015T was found to form a distinct linage within the family Sneathiellaceae, and had 16S rRNA gene sequence similarities of 90.8% to Taonella mepensis H1T, and 90.2% to Ferrovibrio denitrificans S3T. The genome of SYSU D60015T was 5.66 Mb in size with 68.2% of DNA G + C content. The low digital DNA-DNA hybridization (dDDH, 18.0%), average nucleotide identity (ANI, 77.5%) and amino acid identity (AAI, 56.0%) values between SYSU D60015T and Ferrovibrio terrae K5T indicated that SYSU D60015T might represent a distinct genus. Based on the phylogenetic, phenotypic, chemotaxonomic and genomic data, we propose Desertibaculum subflavum gen. nov., sp. nov. as a novel species of a new genus within the family Sneathiellaceae. The type strain is SYSU D60015T (= NBRC 112952T = CGMCC 1.16256T).
Paenibacillus thermotolerans sp. nov., isolated from a hot spring in Yunnan Province, south-west China

Abstract:Two Gram-positive, rod-shaped, endospore-forming strains, YIM B05601 and YIM B05602 T , were isolated from soil sampled at Hamazui hot spring, Tengchong City, Yunnan Province, PR China. Phylogenetic analysis based on 16S rRNA gene sequences suggested that the two strains fell within the genus Paenibacillus , appearing most closely related to Paenibacillus alkalitolerans YIM B00362 T (96.9 % sequence similarity). Genome-based phylogenetic analysis confirmed that strains YIM B05601 and YIM B05602 T formed a distinct phylogenetic cluster within the genus Paenibacillus . The average nucleotide identity (ANI) and digital DNA–DNA hybridization (dDDH) values of strains YIM B05601 and YIM B05602 T with the related species P. alkalitolerans YIM B00362 T were within the ranges of 74.43–74.57 % and 12.1–18.5 %, respectively, which clearly indicated that strains YIM B05601, YIM B05602 T represented a novel species. Strains YIM B05601 and YIM B05602 T exhibited 99.6 % 16S rRNA gene sequence similarity. The ANI and dDDH values between the two strains were 99.8 and 100 %, respectively, suggesting that they belong to the same species. Optimum growth for both strains occurred at pH 7.0 and 45 °C. The diagnostic diamino acid in the cell-wall peptidoglycan of strains YIM B05601 and YIM B05602 T was meso -diaminopimelic acid. MK-7 was the predominant menaquinone. The polar lipids of strain YIM B05602 T were diphosphatidylglycerol, phosphatidylglycerol, phosphatidylethanolamine, phosphatidylmonomethylethanolamine, four unidentified glycolipids, an unidentified polarlipid and phosphatidylinositol mannoside. The major fatty acids of the two stains were iso-C 15 : 0 and anteiso-C 15 : 0 . Based on phylogenomic and phylogenetic analyses coupled with phenotypic and chemotaxonomic characterizations, strains YIM B05601 and YIM B05602 T could be classified as a novel species of the genus Paenibacillus , for which the name Paenibacillus thermotolerans sp. nov. is proposed. The type strain is YIM B05602 T (=CGMCC 1.60051 T =KCTC 43460 T =NBRC 115924 T ).

Abstract:A Gram-stain negative, pink-pigmented, strictly aerobic, non-motile and coccoid-shaped bacterial strain, designated TG-679T, was isolated from a deep-sea water sample collected from the South China Sea. Cells of strain TG-679T were catalase- and oxidase-positive, lacked bacteriochlorophyll a and carotenoid. Strain TG-679T was found to grow at 10-40 °C (optimum, 28 °C), pH 5.5-10.0 (optimum, pH 7.5) and with 0.5-2.0 % (w/v) NaCl (optimum, 1.5 %). Chemotaxonomic analysis of strain TG-679T indicated that the sole respiratory quinone was Q-10, the predominant cellular fatty acid was C18 : 1ω7c, and the major polar lipids consisted of phosphatidylethanolamine, phosphatidylmonomethylethanolamine, phosphatidylglycerol, phosphatidylcholine, an unidentified phospholipid and two unidentified glycolipids. The genomic DNA G+C content of strain TG-679T was 65.9 mol%. Phylogenetic analysis based on 16S rRNA gene sequences revealed that strain TG-679T constituted a separated branch in the family Rhodobacteraceae. Differential phenotypic properties, together with phylogenetic distinctiveness, demonstrated that strain TG-679T is clearly distinct from any validly published genus. Based on polyphasic taxonomic characterization, strain TG-679T is considered to represent a novel species of a novel genus, for which the name Meridianimarinicoccus roseus gen. nov., sp. nov. is proposed. The type strain of the species is TG-679T (=KCTC 62454T=MCCC 1K03496T).

Abstract:A novel Fe(III)-reducing bacterium, designated GSS01(T), was isolated from a forest soil sample using a liquid medium containing acetate and ferrihydrite as electron donor and electron acceptor, respectively. Cells of strain GSS01(T) were strictly anaerobic, Gram-stain-negative, motile, non-spore-forming and slightly curved rod-shaped. Growth occurred at 16-40 °C and optimally at 30 °C. The DNA G+C content was 60.9 mol%. The major respiratory quinone was MK-8. The major fatty acids were C(16:0), C(18:0) and C(16:1)ω7c/C(16:1)ω6c. Strain GSS01(T) was able to grow with ferrihydrite, Fe(III) citrate, Mn(IV), sulfur, nitrate or anthraquinone-2,6-disulfonate, but not with fumarate, as sole electron acceptor when acetate was the sole electron donor. The isolate was able to utilize acetate, ethanol, glucose, lactate, butyrate, pyruvate, benzoate, benzaldehyde, m-cresol and phenol but not toluene, p-cresol, propionate, malate or succinate as sole electron donor when ferrihydrite was the sole electron acceptor. Phylogenetic analyses based on 16S rRNA gene sequences revealed that strain GSS01(T) was most closely related to Geobacter sulfurreducens PCA(T) (98.3% sequence similarity) and exhibited low similarities (94.9-91.8%) to the type strains of other species of the genus Geobacter. The DNA-DNA relatedness between strain GSS01(T) and G. sulfurreducens PCA(T) was 41.4 ± 1.1%. On the basis of phylogenetic analysis, phenotypic characterization and physiological tests, strain GSS01(T) is believed to represent a novel species of the genus Geobacter, and the name Geobacter soli sp. nov. is proposed. The type strain is GSS01(T) ( =KCTC 4545(T) =MCCC 1K00269(T)).

Abstract:Biological denitrification is a significant process in nitrogen biogeochemical cycle of terrestrial geothermal environments, and Thermus species have been shown to be crucial heterotrophic denitrifier in hydrothermal system. Five Gram-stain negative, aerobic and rod-shaped thermophilic bacterial strains were isolated from hot spring sediments in Tibet, China. Phylogenetic analysis based on 16S rRNA gene and whole genome sequences indicated that these isolates should be assigned to the genus Thermus and were most closely related to Thermus caldifontis YIM 73026T, and Thermus brockianus YS38T. Average nucleotide identity (ANI) and digital DNA-DNA hybridization (dDDH) values between the five strains and the type strains of the genus Thermus were lower than the threshold values (95% and 70%, respectively) recommended for bacterial species, which clearly distinguished the five isolates from other species of the genus Thermus and indicated that they represent independent species. Colonies are circular, convex, non-transparent. Cell growth occurred at 37–80 °C (optimum, 60–65 °C), pH 6.0–8.0 (optimum, pH 7.0) and with 0–2.0% (w/v) NaCl (optimum, 0–0.5%). Denitrification genes (narG, nirK, nirS, and norB genes) detected in their genomes indicated their potential function in nitrogen metabolism. The obtained results combined with those of morphological, physiological, and chemotaxonomic characteristics, including the menaquinones, polar lipids, and cellular fatty acids showed that the isolates are proposed as representing five novel species of the genus Thermus, which are proposed as Thermus hydrothermalis sp. nov. SYSU G00291T, Thermus neutrinimicus sp. nov. SYSU G00388T, Thermus thalpophilus sp. nov. SYSU G00506T, Thermus albus sp. nov. SYSU G00608T, Thermus altitudinis sp. nov. SYSU G00630T.

Abstract:A Gram-reaction-negative, rod-shaped, motile, neutrophilic bacterium, designated strain BH3(T), was isolated from wastewater of a sequential batch reactor treating wastewater taken from a leather plant. The isolate grew in 0-8 % (w/v) NaCl, at pH 6-9 and at 4-45 °C. Chemotaxonomic analysis showed that strain BH3(T) had characteristics typical of members of the genus Sphingomonas, such as the presence of sphingolipids, Q-10 and 2-hydroxymyristic acid and the absence of 3-hydroxy fatty acids. The presence of C(18 : 1)ω7c (39.2 %) and C(16 : 0) (11.2 %) as major fatty acids, C(14 : 0) 2-OH (20.6 %) as the major 2-hydroxy fatty acid and homospermidine as the major polyamine indicated that strain BH3(T) belonged to the genus Sphingomonas sensu stricto. The genomic DNA G+C content of strain BH3(T) was 65.6 mol%. 16S rRNA gene sequence similarities between the isolate and closely related members of the genus Sphingomonas sensu stricto ranged from 92.6 to 97.3 %, the highest sequence similarities being to Sphingomonas melonis DSM 14444(T) (97.3 %) and Sphingomonas aquatilis DSM 15581(T) (97.3 %). Based on its phenotypic characteristics and the results of DNA-DNA hybridization studies and 16S rRNA gene sequence comparisons, strain BH3(T) represents a novel species of the genus Sphingomonas sensu stricto, for which the name Sphingomonas rubra sp. nov. is proposed. The type strain is BH3(T) ( = CGMCC 1.9113(T) = JCM 16230(T)).

Abstract:A novel facultatively anaerobic, Gram-stain-negative bacterium, designated strain HT7-2(T), was isolated from Ulva prolifera collected from the intertidal zone of Qingdao sea area, China, during its bloom. Cells were rod-shaped (1.9-3.5x0.4-0.6 mu m), non-sporulating and motile by gliding. Strain HT7-2(T) was able to grow at 4-50 degrees C (optimum 40-42 degrees C), pH 5.5-8.5 (optimum pH 7.0), 0-8 % (w/v) NaCl (optimum 2-3 %) and 0.5-10% (w/v) sea salts (optimum 2.5 %). The genomic DNA G+C content was 38.8 mol%. The phylogenetic analysis based on 16S rRNA gene sequences revealed that strain HT7-2(T) belonged to the genus Maribacter with sequence similarity values of 94.5-96.6 %, and was most closely related to Maribacter aestuarii GY20(T) (96.6 %). Chemotaxonomic analysis showed that the main isoprenoid quinone was MK-6 and the major fatty acids were iso-C-15:0 and unknown equivalent chain-length 13.565. The polar lipids of strain HT7-2(T) consisted of one phosphatidylethanolamine, four unidentified lipids and one unidentified aminolipid. On the basis of the phenotypic, phylogenetic and chemotaxonomic characteristics, strain HT7-2(T) (=CGMCC 1.12207(T)=JCM 18466(T)) is concluded to represent a novel species of the genus Maribacter, for which the name Maribacter thermophilus sp. nov. is proposed. An emended description of the genus Maribacter is also proposed.

Abstract:AbstractThermoleptolyngbyais a newly proposed genus of thermophilic cyanobacteria that are often abundant in thermal environments. However, a vast majority ofThermoleptolyngbyastrains were not systematically identified, and genomic features of this genus are also sparse. Here, polyphasic approaches were employed to identify a thermophilicThermoleptolyngbyaA183 (FACHB-2491) isolated from hot spring Erdaoqiao, Ganzi prefecture, China. Morphological characterization was investigated by microscopy. Whilst the results of 16S rRNA were inconclusive, the phylogenetic reconstruction and secondary structures of 16S-23S ITS strongly suggested that A183 strain is a novel species withinThermoleptolyngbya. Moreover, genome-scale average nucleotide identity (ANI) confirmed the genetic divergence of A183 fromThermoleptolyngbyaO-77. Comparative genome analysis revealed inconsistent genome structures ofThermoleptolyngbyastrains. Further GO analysis showed that the specific genes were distributed in a wide range of functional categories. In addition, analysis of genes related to thermotolerance, signal transduction, and carbon/nitrogen/sulfur assimilation revealed the ability of this strain to adapt to inhospitable niches in hot springs.

Abstract:A Gram-negative, aerobic, neutrophilic, rod-shaped bacterium, designated strain CN85(T), was isolated from a sediment sample collected from the East China Sea and was subjected to a polyphasic taxonomic characterization. This isolate grew in the presence of 0.5-6.0% (w/v) NaCl and at 20-40 degrees C; optimum growth was observed with 3% (w/v) NaCl and at 35 degrees C. Chemotaxonomic analysis showed that Q-8 was the predominant respiratory quinone and that iso-C(15:0), iso-C(11:0) 3-OH, iso-C(17:1) omega 9c, iso-C(17:0), iso-C(11:0) and C(16:0) were the major fatty acids. The G+C content of the genomic DNA was 57.8 mol%. On the basis of 16S rRNA gene sequence analysis, the isolate was affiliated to the genus Microbulbifer. Strain CN85(T) exhibited most phylogenetic affinity with respect to the type strain of Microbulbifer maritimus (97.0% sequence similarity) and showed less than 97% sequence similarity with respect to other described Microbulbifer species with known 16S rRNA gene sequences. The DNA-DNA hybridization between strain CN805(T) and M. maritimus JCM 12187(T) was 44%. On the basis of phenotypic and genotypic data, strain CN85(T) represents a novel species of the genus Microbulbifer, for which the name Microbulbifer donghaiensis sp. nov. is proposed. The type strain is CN85(T) (=CGMCC 1.7063(T) =JCM 15145(T)).
Litoribacter ruber gen. nov., sp. nov., an alkaliphilic, halotolerant bacterium isolated from a soda lake sediment

Abstract:A novel alkaliphilic, halotolerant, rod-shaped bacterium, designated strain YIM CH208(T), was isolated from a soda lake in Yunnan, south-west China. The taxonomy of strain YIM CH208(T) was investigated by a polyphasic approach. Strain YIM CH208(T) was Gram-negative, strictly aerobic and non-motile and formed red colonies. Optimal growth conditions were 28°C, pH8.5 and 0.5-2.5 % NaCl. Phylogenetic analysis based on 16S rRNA gene sequence comparisons showed that the isolate formed a distinct line within a clade containing the genus Echinicola in the phylum Bacteroidetes and was related to the species Echinicola pacifica and Rhodonellum psychrophilum, with sequence similarity of 91.7 and 91.6 % to the respective type strains. The DNA G+C content was 45.1&emsp14;mol%. The major respiratory quinone was menaquinone-7 (MK-7). The predominant cellular fatty acids were iso-C(17 : 1)ω9c (19.9 %), C(15 : 0) 3-OH (12.1 %), iso-C(17 : 0) 3-OH (11.3 %), summed feature 3 (iso-C(15 : 0) 2-OH and/or C(16 : 1)ω7c; 10.7 %) and C(17 : 1)ω6c (8.7 %). On the basis of the phenotypic, chemotaxonomic and phylogenetic data, strain YIM CH208(T) represents a novel species of a new genus, for which the name Litoribacter ruber gen. nov., sp. nov. is proposed. The type strain of Litoribacter ruber is YIM CH208(T) (=ACCC 05414(T) =KCTC 22899(T)).
