Abstract:Chemosensory proteins (CSPs) are ubiquitous soluble small proteins isolated from sensory organs thought to be involved in chemical communication. Here we report the first cDNA of CSPs, called Ac-ASP3, cloned and characterized from antennas of adult worker bees in Chinese honeybee, Apis cerana cerana. The Ac-ASP3 cDNA comprises 2 exons, with an ORF of 393-bp encoding 130 aa. Protein signature analyses show that the protein consists of four conserved cysteines and a signal peptide with 19 aa in the N-terminal sequence. The deduced protein sequence shares high homology with Am-ASP3 of Apis mellifera and low similarity with other species of insects. Immunocytochemical localization shows that Ac-ASP3 is only specifically expressed on the antenna contact chemosensilla such as sensilla trichodea B and sensilla basiconica, whereas Ac-ASP3 is scarcely expressed on olfactory chemosensilla such as sensilla placodea. Real-time PCR of Ac-ASP3 transcripts shows that Ac-ASP3 is highly expressed on wings and legs, but expression is lower on antenna. Temporal expression patterns suggest that Ac-ASP3 is expressed during the period of pupa and adults from 1-d to 6-d stages when bees act as house bees, cleaning the comb and taking care of the queen and larvae in comb. The above evidence suggests that Ac-ASP3 is unique in species and is generally not involved in olfaction during searching for honey and pollen. Rather, the protein seems to function in recognition of chemosensory substances on bees’ cuticle and mechanical movement of antenna.

Abstract:The spatio-temporal expressed profiles of two kinds of chemical communication-related protein genes, the odorant-binding protein of Ac-ASP2 and chemosensory protein of Ac-ASP3, were identified by real-time polymerase chain reaction (PCR). Results obtained using the 2 -D DCt method showed that Ac-ASP2 was a gene coding antenna-specific protein that did not express in larvae and pupae, but had discontinuous high abundance periods at 1, 9, 15, 27 and 30 days. The expressing abundance at such periods was at least ten times higher than that at other periods. From the distribution ofAc-ASP3 mRNA observed in different tissues, the transcript levels seemed to be higher in the wings, abdomen and thorax (of order � 10 6 ), and lower in the legs, antennae and head (of order � 10 5 ). From highest to lowest, the original copy number was found in the various body parts in the following order: wings, abdomen, thorax, legs, antenna, and head. The results suggest that Ac-ASP3 has an intimate relation with the chemosensory behaviour of wings and abdomen in Apis cerana cerana.

Candidate Odorant Binding Proteins and Chemosensory Proteins in the Larval Chemosensory Tissues of Two Closely Related Noctuidae Moths, Helicoverpa Armigera and H. Assulta

Abstract:In order to acquire enough nutrients and energy for further development, larvae need to invest a large portion of their sensory equipments to identify food sources. Yet, the molecular basis of odor-driven behavior in larvae has been poorly investigated. Information on olfactory genes, particularly odorant binding proteins (OBPs) and chemosensory proteins (CSPs) which are involved in the initial steps of olfaction is very scarce. In this study, we have identified 26 OBP and 21 CSP genes from the transcriptomes of Helicoverpa armigera larval antennae and mouthparts. A comparison with the 34 OBP and 18 CSP genes of the adult antenna, revealed four novel OBPs and seven novel CSPs. Similarly, 27 OBPs (six novel OBPs) and 20 CSPs (6 novel CSPs) were identified in the transcriptomes of Helicoverpa assulta larval antennae and mouthparts. Tissue-specific profiles of these soluble proteins in H. armigera showed that 6 OBP and 4 CSP genes are larval tissue-specific, 15 OBPs and 13 CSPs are expressed in both larvae and adult, while the rest are adult- specific. Our data provide useful information for functional studies of genes involved in larval foraging.
Identification and Characterization of Chemosensory Genes in the Antennal Transcriptome of Spodoptera Exigua

Abstract:Chemical senses are crucial for insect behaviors such as host preference, mate choice and oviposition site selection. Various protein families are involved in these processes, including odorant receptors (ORs), ionotropic receptors (IRs), gustatory receptors (GRs), chemosensory proteins (CSPs), odorant binding proteins (OBPs) and sensory neuron membrane proteins (SNMPs). To better understand the olfactory mechanism in Spodoptera exigua, we conducted transcriptome analysis of adult antennae and identified a total of 157 candidate chemosensory genes encoding 51 ORs, 20 IRs, 7 GRs, 32 CSPs, 45 OBPs and 2 SNMPs. Quantitative real time PCR (qPCR) analysis of the tissue- and sex-specific expression profiles of ORs, GRs and IRs revealed that these genes could be detected in at least one tissue tested. SexiOR6, 11, 13 and 16, which were predicted to be pheromone receptors based on phylogenetic analysis, exhibited male-specific antennae expression. SexiOR18, 26, 28, 30, 34, 39, and 40 exhibited female-biased expression. SexiGR1, SexiGR2 and SexiGR3, are predicted carbon dioxide receptors, and the former was expressed specifically in antennae, and the latter two were expressed both in antennae and labial palps. SexiIRs had diverse expression profiles. SexiIR8a and SexiIR25a were quite conserved and expressed at high levels. This work will greatly facilitate the understanding of olfactory system in S. exigua and provides valuable information for further functional studies of the chemoreception mechanism in Lepidopteran moths.
Identification and expression profiling of candidate chemosensory membrane proteins in the band-winged grasshopper, Oedaleus asiaticus

Abstract:The band-winged grasshopper Oedaleus asiaticus (Orthoptera: Acridoidea) is an important insect pest in north China. Chemosensory membrane proteins are believed to be crucial factors in direct interactions with odorants in the chemosensory process. However, there is much limited information on the chemosensory system in this pest. In this study, a total of 69 candidate chemosensory membrane protein genes, including 60 olfactory receptors (ORs), 6 ionotropic receptors (IRs) and 3 sensory neuron membrane proteins (SNMPs), were identified for the first time from the antennal transcriptomes of O. asiaticus. Blastp match and phylogenetic analysis demonstrated that these chemosensory membrane proteins were the closest to their orthologous species, Locusta migratoria. The qRT-PCR analysis revealed that all tested 14 OR and two SNMP genes (OasiSNMP1 and OasiSNMP2a) were strongly expressed in adult antennae, and nearly all tested genes (15/16) displayed significant differences in the expression levels between both sexes. Moreover, two IR genes (OasiIR25a and OasiIR76b) had uniquely high expression levels in the antennae, labial palps and maxillary palps, while three IR genes (OasiIR1, OasiIR2 and OasiIR3) were highly expressed in most tested tissues (heads without antennae and mouthparts, labial palps, maxillary palps, labia without labial palps, thoraxes, tarsi, and abdomens) except for antennae, labra, and wings but OasiIR5a was just faintly expressed in the antennae, labia without labial palps, labial palps, maxillary palps and abdomen. Our results provide important molecular information for further investigation on the chemoreception mechanisms in O. asiaticus.
Identification and Functional Characterization of General Odorant Binding Proteins in Orthaga Achatina

Abstract:The olfactory system in insects are crucial for recognition of host plants and oviposition sites. General odorant binding proteins (GOBPs) are thought to be involved in detecting odorants released by host plants. Orthaga achatina (Lepidoptera: Pyralidae) is one of the most serious pests of camphor trees, Cinnamomum camphora (L.) Presl, an important urban tree species in southern China. In this study, we study the GOBPs of O. achatina. Firstly, two full-length GOBP genes (OachGOBP1 and OachGOBP2) were successfully cloned according to transcriptome sequencing results, and real-time quantitative PCR measurements showed that both GOBP genes were specifically expressed in the antennae of both sexes, proposing their important roles in olfaction. Then, both GOBP genes were heterologous expressed in Escherichia coli and fluorescence competitive binding assays were conducted. The results showed that OachGOBP1 could bind Farnesol (Ki = 9.49 μM) and Z11-16: OH (Ki = 1.57 μM). OachGOBP2 has a high binding affinity with two camphor plant volatiles (Farnesol, Ki = 7.33 μM; α-Phellandrene, Ki = 8.71 μM) and two sex pheromone components (Z11-16: OAc, Ki = 2.84 μM; Z11-16: OH, Ki = 3.30 μM). These results indicate that OachGOBP1 and OachGOBP2 differ in terms of odorants and other ligands. Furthermore, key amino acid residues that bind to plant volatiles were identified in GOBPs using 3-D structure modeling and ligand molecular docking, predicting the interactions between the GOBPs and the host plant volatiles.

Abstract:The rice water weevil, Lissorhoptrus oryzophilus Kuschel (Coleoptera: Curculionidae), is a serious pest species both in its original distribution region of northern America and its invaded regions of eastern Asia and southern Europe. The odorant-binding proteins (OBPs) and the chemosensory proteins (CSPs) play important roles in host and mate locating, thus might play a significant role in the success of the species as an invader, which has not been characterized yet. We identified 10 OBPs and 5 CSPs in L. oryzophilus and investigated the expression profiles of these genes in various tissues by quantitative real-time PCR. Five classic OBPs were predominantly expressed in the antennae. CSPs were expressed ubiquitously with particularly high transcript levels in antennae, legs, and wings. Three antenna-specific OBPs (LoOBP1, 8, 11) were up-regulated following 1-3 d of food deprivation and down-regulated afterward. These findings suggest most classic OBPs are likely involved in chemoreception whereas CSPs as well as the minus-C OBPs may have broader physiological functions, which in turn may help to understand the molecular aspects of chemical communication in this invasive insect.
Identification and Expression Specificity of Chemosensory Genes in the Male Reproductive System of Spodoptera Exigua

Abstract:Some chemosensory genes are found to be expressed specifically or highly in the male reproductive system, suggesting their importance in male reproductive physiology. In this paper, we identified chemosensory genes expressed in the male reproductive system of the moth pest Spodoptera exigua by transcriptome and proteome sequencing, and further determined expression patterns of these genes regarding antennae and different tissues of the male reproductive system (testes, vas deferens, seminal vesicles, ejaculatory ducts, male accessory glands) by the real-time quantitative PCR (qPCR). Transcriptome analysis showed that 46 chemosensory genes were expressed in the male reproductive system of S. exigua, including 18 odorant binding proteins (OBPs), 12 che-mosensory proteins (CSPs), 10 odorant receptors (ORs) and 6 other chemosensory genes. Further qPCR mea-surements revealed that 14 chemosensory genes were expressed specifically or highly in the male reproductive tissues, relative to that in male antennae. Of the 14 genes, OBP17, OBP22, OBP27 and GR43 were specifically expressed in the testes; OBP20 was specifically expressed in the vas deferens; and PBP4 was specifically expressed in the male accessory glands. In addition, proteome data of the spermatophore confirmed that at least 4 OBPs and one CSP were transmitted from males to females, suggesting their function in the reproductive physiology of mated females. The results provide insights into the functional diversity of chemosensory genes, and bases for exploring functions of these chemosensory genes in male reproductive physiology.

Abstract:Chemosensory proteins (CSP) afford insects the ability to recognize complicated environmental cues and are involved in diverse physiological processes. The planthoppers Laodelphax striatellus and Sogatella furcifera are serious specialist pests of rice throughout eastern Asia. In this study, we identified 21 putative CSP genes (namely LsCSP1–12 and SfCSP1–9) in the two planthoppers L. striatellus and S. furcifera, and profiled their transcript accumulation in different developmental stages and body parts by real-time quantitative PCR (qPCR). All CSPs had the characteristic typical features of the CSP family, with the exception of LsCSP10 and SfCSP2, which lacked the predicted signal peptide sequence. Among these putative CSP genes, only two (LsCSP9 and SfCSP5) were more highly expressed in the antennae, indicating that they might be involved in olfaction. On the contrary, eleven genes (LsCSP2, 3, 4, 6, 8, 12; SfCSP1, 4, 6, 7, 8) and one gene (SfCSP9) were more highly expressed in the legs and heads (without antennae), respectively. The remainders of the genes were ubiquitously expressed in the heads (without antennae) and the bodies (without the legs and heads). The broad expression patterns in different body parts indicate that the CSPs of these two delphacid species may be involved in diverse functions besides chemosensation. These findings increase the understanding of the chemosensory protein system of Hemiptera species.

Abstract:Cyrtorhinus lividipennis Reuter (Hemiptera: Miridae) is an important predatory natural enemy of planthopper and leafhopper eggs in Asian rice paddy fields. Cyrtorhinus lividipennis is known to rely largely on herbivoreinduced plant volatiles to identify eggs embedded in rice stem tissues for feeding and on pheromones for mating. However, exactly how C. lividipennis decode these chemical information is unclear. In most insects, the odorant-binding proteins (OBPs) and the chemosensory proteins (CSPs) are essential for seeking out food resources and mates. In this study, we identified 10 OBP and 5 CSP genes in C. lividipennis and investigated their expression patterns in various tissues of adult males and females by quantitative real-time PCR (qRT-PCR). Six OBP genes (ClivOBP1, 2, 4, 6, 9, and 10) were mainly expressed in the male antennae, whereas three genes (ClivOBP3, ClivOBP7, and ClivOBP8) had high expression in the female antennae. ClivCSP1 was predominantly expressed in the male antennae. These findings suggest that most ClivOBPs and ClivCSPs are likely involved in food-searching behavior. The recognition of the pheromone molecules provides the basis for further functional studies on the chemoreception system of C. lividipennis.

Abstract:In this study, we constructed a high-quality cDNA library from the antennae of the Chilo suppressalis (Walker) (Lepidoptera: Pyralidae). A total of 1,235 colonies with inserts greater than 0.7 kb were sequenced and analyzed. Homology searching coupled with bioinformatics analysis identified 15 and 7 cDNA sequences, respectively, encoding putative odorant-binding proteins (OBPs) and chemosensory proteins (CSPs). A phylogenetic tree of CsupCSPs showed that each CsupCSP has orthologs in Manduca sexta and Bombyx mori with strong bootstrapping support. One CSP was either very specific or more related to the CSPs of another species than to conspecific CSP. The expression profiles of the OBPs and CSPs in different tissues were measured by real-time quantitative PCR. The results revealed that of the 11 OBP genes, the transcript levels of CsupOBP1, CsupOBP5, and CsupOBP7 were higher in both male and female antennae than those in other tissues. And CsupCSP7 was highly expressed in both male and female antennae. Based on these results, the possible physiological functions of CsupOBPs and CsupCSPs were discussed. Abstract:Cyrtorhinus lividipennis Reuter (Hemiptera: Miridae) is an important egg predator of planthoppers which are destructive rice pests. The chemosensory genes in the mirid antennae play important roles in mating and prey-seeking behaviors. To gain a better understanding of the olfaction of C. lividipennis, we sequenced the antennal transcriptomes of the predator to identify the key olfaction genes. We identified 18 odorant binding proteins (OBPs), 12 chemosensory proteins (CSPs), 1 Niemann-Pick C2 protein (NPC2), 15 odorant receptors (ORs), 6 ionotropic receptors (IRs), 3 gustatory receptors (GRs) and 3 sensory neuron membrane proteins (SNMPs). Quantitative real-time PCR results showed that the relative transcript levels of three ClivORs (ClivOR6, 7 and 14) in the female antennae were 3 to 6 folds higher than that in the male antennae, indicating these genes were more related to oviposition site selection. The relative transcript levels of ClivCSP8 and ClivOR11 were 2.6 and 2.7 times higher in the male antennae than that of the female, respectively, indicating that these genes might be involved in mate searching. Moreover, the responses of dsorco treated predators to volatiles emitted from infested rice were significantly reduced, indicating these volatiles might serve as crucial cues in the host searching of C. lividipennis.

Abstract:The high sensitivity of the olfactory system is essential for feeding and oviposition in moth insects, and some chemosensory proteins (CSPs) are thought to play roles in this system by binding and carrying hydrophobic odorants across the aqueous sensillar lymph. In this study, to identify the olfactory CSPs from a repertoire of 21 CSP members in the notorious rice pest Chilo suppressalis (Walker) (Lepidoptera: Pyralidae), tissue expression patterns were firstly examined by quantitative real-time polymerase chain reaction (qPCR). It showed that CSP2 was antennae specific and seven more CSPs (CSP1, 3, 4, 6, 15, 16, and 17) were antennae biased in expression, suggesting their olfactory roles; while other CSPs were multiple-tissue expressed and non-antennae biased, suggesting other functions for these genes. To further determine the ligand binding specificity, three putative olfactory genes (CSP1-3) were expressed in Escherichia coli cells, and binding affinity of these three recombinant CSP proteins were measured for 35 plant volatiles by the ligand binding assays. CSP1 and CSP2 exhibited high binding affinities (Ki ≤ 10.00 µM) for four (2-tridecanone, benzaldehyde, laurinaldehyde and 2-pentadecanone) and two (2-heptanol and (+)-cedrol) host plant volatiles, respectively; the three CSPs also showed moderate binding affinity (Ki = 10.01-20.00 µM) for 16 plant volatiles. Our study suggests that the three CSPs play essential roles in the perception of host plant volatiles, providing bases for the elucidation of olfactory mechanisms in this important pyralid pest.

Abstract:Insects use olfaction to detect exogenous odors and adapt to environments. In their olfaction systems, odorant-binding proteins (OBPs) are believed to be a key component. The unique OBP system of each species reflects the evolution of chemosensation of insects with habits. Here, we for the first time identified 15 OBPs, OasiOBP1-15, of a grasshopper, Oedaleus asiaticus, that lives in the grasslands of Northern China and is closely related to the locust, Locusta migratoria. OasiOBP9 and OasiOBP10 are specifically expressed in the antennae. Other OBPs are expressed in the antennae as well as other chemosensory organs, such as the mouthparts and wings. Significantly more OasiOBP7 was detected in male than female antennae, but there are 9 OBPs that were more expressed in female than male antennae by quantitative real-time PCR. Phylogenetic analysis indicated that most of the O. asiaticus OBPs are similar to those of L. migratoria, but some are substantially different. This indicates that the OBPs originally evolved in a common ancestor, but their unique chemosensory systems are adapted to different ecosystems.

Abstract:Studies of chemosensory genes are key to a better understanding of intra- and interspecific communications between insects and their environment and provide opportunities for developing environmentally friendly pesticides to target pest species. The bamboo locust Ceracris kiangsu Tsai (Orthoptera: Acrididae) is one of the most important bamboo leaf-eating insects in southern China. However, the genes underlying olfactory sensation are lacking in the bamboo locust. In this study, the transcriptomes of male and female C. kiangsu antennae were sequenced and analyzed. A total of 125 chemosensory genes, including 91 odorant receptors (ORs), 13 ionotropic receptors (IRs), 13 odorant-binding proteins (OBPs), six chemosensory proteins (CSPs), and two sensory neuron membrane proteins, were identified based on sequence alignment and phylogenetic analyses. The expression patterns of all candidate genes on the antennae of males and females, maxillary palps, tarsi, wings, and thoraxes-abdomens were confirmed by real-time quantitative PCR. The analyses demonstrated that most genes are highly expressed in the antennae, and 35 ORs, 7 IRs, 10 OBPs, and 1 CSP exhibit significantly male-biased expression patterns, indicating their potential functions in mating behavior and the recognition of female sex pheromones. In addition to the antennal-predominant genes, some were abundant in the maxillary palps and some in the non-olfactory tissues, suggesting their different functions in the olfactory system of C. kiangsu. Our research offers an extensive resource for investigating the chemoreception mechanism of C. kiangsu. Further studies of olfactory function will provide comprehensive methods and original strategies for integrated pest management.

Abstract:Loxostege sticticalis Linnaeus is an economically important agricultural pest, and the larvae cause great damage to crops, especially in Northern China. However, effective and environmentally friendly chemical methods for controlling this pest have not been discovered to date. In the present study, we performed HiSeq2500 sequencing of transcriptomes of the male and female adult antennae, adult legs and third instar larvae, and we identified 54 candidate odorant receptors (ORs), including 1 odorant receptor coreceptor (Orco) and 5 pheromone receptors (PRs), 18 ionotropic receptors (IRs), 13 gustatory receptors (GRs), 34 odorant binding proteins (OBPs), including 1 general odorant binding protein (GOBP1) and 3 pheromone binding proteins (PBPs), 10 chemosensory proteins (CSPs) and 2 sensory neuron membrane proteins (SNMPs). The results of RNA-Seq and RT-qPCR analyses showed the expression levels of most genes in the antennae were higher than that in the legs and larvae. Furthermore, PR4, OR1-4, 7-11, 13-15, 23, 29-32, 34, 41, 43, 47/IR7d.2/GR5b, 45, 7/PBP2-3, GOBP1, OBP3, 8 showed female antennae-biased expression, while PR1/OBP2, 7/IR75d/CSP2 showed male antennae-biased expression. However, IR1, 7d.3, 68a/OBP11, 20-22, 28/CSP9 had larvae enriched expression, and OBP15, 17, 25, 29/CSP5 were mainly expressed in the legs. The results shown above indicated that these genes might play a key role in foraging, seeking mates and host recognition in the L. sticticalis. Our findings will provide the basic knowledge for further studies on the molecular mechanisms of the olfactory system of L. sticticalis and potential novel targets for pest control strategies.
Molecular characterization of two sensory neuron membrane proteins from Chilo suppressalis (lepidoptera: Pyralidae)

Abstract:Sensory neuron membrane proteins (SNMPs) play an especially important role in insect pheromone communication. However, the SNMPs for the Asiatic rice borer, Chilo suppressalis (Walker) (Lepidoptera: Pyralidae), a notorious rice pest, remain uncharacterized. Here we report the cloning and characterization of two novel SNMPs from the C. suppressalis, CsupSNMP1 and CsupSNMP2. The CsupSNMP1 and CsupSNMP2 cDNAs contained open reading frames (ORFs) of 1,572 and 1,569 bp, encoding proteins of 523 and 522 amino acid residues, respectively. The amino acid identity between the two deduced CsupSNMPs was low (30% identity), but they shared a high degree of similarity to previously characterized SNMP1s or SNMP2s from other moth species, which is consistent with phylogenetic analysis in which CsupSNMP1 and CsupSNMP2 are clustered into two distinct groups based on their amino acid sequences. The expression patterns of CsupSNMPs in various adult tissues and in different developmental stages were determined by real-time quantitative polymerase chain reaction. The results showed that both CsupSNMP1 and CsupSNMP2 were abundantly expressed in the male and female antennae, reaching their maximum in the adult stage, suggesting the two genes are involved in the process of olfaction. Low levels of CsupSNMP2 also were expressed in nonolfactory tissues such as legs and wings, implying possible gustatory roles of the protein in the moth.
