Abstract:Plant-derived terpenes are effective in treating chronic dysentery, rheumatism, hepatitis, and hyperlipemia. Thus, understanding the molecular basis of terpene biosynthesis in some terpene-abundant Chinese medicinal plants is of great importance. Abundant in mono- and sesqui-terpenes, Rhodomyrtus tomentosa (Ait.) Hassk, an evergreen shrub belonging to the family Myrtaceae, is widely used as a traditional Chinese medicine. In this study, (+)-α-pinene and β-caryophyllene were detected to be the two major components in the leaves of R. tomentosa, in which (+)-α-pinene is higher in the young leaves than in the mature leaves, whereas the distribution of β-caryophyllene is opposite. Genome-wide transcriptome analysis of leaves identified 138 unigenes potentially involved in terpenoid biosynthesis. By integrating known biosynthetic pathways for terpenoids, 7 candidate genes encoding terpene synthase (RtTPS1-7) that potentially catalyze the last step in pinene and caryophyllene biosynthesis were further characterized. Sequence alignment analysis showed that RtTPS1, RtTPS3 and RtTPS4 do not contain typical N-terminal transit peptides (62-64aa), thus probably producing multiple isomers and enantiomers by terpenoid isomerization. Further enzyme activity in vitro confirmed that RtTPS1-4 mainly produce (+)-α-pinene and (+)-β-pinene, as well as small amounts of (-)-α-pinene and (-)-β-pinene with GPP, while RtTPS1 and RtTPS3 are also active with FPP, producing β-caryophyllene, along with a smaller amount of α-humulene. Our results deepen the understanding of molecular mechanisms of terpenes biosynthesis in Myrtaceae.

Abstract:Dendrobium catenatum is a widely cultivated Chinese orchid herb rich in abundant secondary metabolites, such as terpenes. However, terpene distribution and characterization of terpene biosynthesis-related genes remain unknown in D. catenatum. In this study, metabolic profiling was performed to analyze terpene distribution in the root, stem, leaf, and flower of D. catenatum. A total of 74 terpene compounds were identified and classified. Clustering analysis revealed that terpene compounds exhibited a tissue-specific accumulation, including monoterpenes in the flowers, sesquiterpenes in the stems, and triterpenes in the roots. Transcriptome analysis revealed that the ‘terpenoid backbone biosynthesis’ pathway was only significantly enriched in root vs. flower. The expression of terpene biosynthesis-related genes was spatiotemporal in the flowers. Prenylsynthase-terpene synthases (PS-TPSs) are the largest and core enzymes for generating terpene diversity. By systematic sequence analysis of six species, 318 PS-TPSs were classified into 10 groups and 51 DcaPS-TPSs were found in eight of them. Eighteen DcaPS-TPSs were regulated by circadian rhythm under drought stress. Most of the DcaPS-TPSs were influenced by cold stress and fungi infection. The cis-element of the majority of the DcaPS-TPS promoters was related to abiotic stress and plant development. Methyl jasmonate levels were significantly associated with DcaTPSs expression and terpene biosynthesis. These results provide insight into further functional investigation of DcaPS-TPSs and the regulation of terpene biosynthesis in Dendrobium.
Chemical Compositions and Enantiomeric Distributions of Foliar Essential Oils of Chamaecyparis lawsoniana (A. Murray bis) Parl, Thuja plicata Donn ex D. Don, and Tsuga heterophylla Sarg

Abstract:As part of our continuing interest in the essential oil compositions of gymnosperms, particularly the distribution of chiral terpenoids, we have obtained the foliar essential oils of Chamaecyparis lawsoniana (two samples), Thuja plicata (three samples), and Tsuga heterophylla (six samples) from locations in the state of Oregon, USA. The essential oils were obtained via hydrodistillation and analyzed by gas chromatographic techniques, including chiral gas chromatography—mass spectrometry. The major components in C. lawsoniana foliar essential oil were limonene (27.4% and 22.0%; >99% (+)-limonene), oplopanonyl acetate (13.8% and 11.3%), beyerene (14.3% and 9.0%), sabinene (7.0% and 6.5%; >99% (+)-sabinene), terpinen-4-ol (5.0% and 5.3%; predominantly (+)-terpinen-4-ol), and methyl myrtenate (2.0% and 5.4%). The major components in T. plicata essential oil were (−)-α-thujone (67.1–74.6%), (+)-β-thujone (7.8–9.3%), terpinen-4-ol (2.7–4.4%; predominantly (+)-terpinen-4-ol), and (+)-sabinene (1.1–3.5%). The major components in T. heterophylla essential oil were myrcene (7.0–27.6%), α-pinene (14.4–27.2%), β-phellandrene (6.6–19.3%), β-pinene (6.4–14.9%; >90% (−)-β-pinene), and (Z)-β-ocimene (0.7–11.3%). There are significant differences between the C. lawsoniana essential oils from wild trees in Oregon and those of trees cultivated in other geographical locations. The essential oil compositions of T. plicata are very similar, regardless of the collection site. There are no significant differences between T. heterophylla essential oils from the Oregon Coastal Range or those from the Oregon Cascade Range. Comparing essential oils of the Cupressaceae with the Pinaceae, there are some developing trends. The (+)-enantiomers seem to dominate for α-pinene, camphene, sabinene, β-pinene, limonene, terpinen-4-ol, and α-terpineol in the Cuppressaceae. On the other hand, the (−)-enantiomers seem to predominate for α-pinene, camphene, β-pinene, limonene, β-phellandrene, terpinen-4-ol, and α-terpineol in the Pinaceae.

Abstract:Introduction Daphne pontica L. is an evergreen shrub that is recorded as an anti‐diarrheic plant in Turkish folk medicine. Previous studies on D. pontica have reported, albeit slightly, the isolation of daphnane diterpenoids, but no systematic phytochemical analysis of daphnane diterpenoids has been conducted. Objective This study aimed to comprehensively investigate daphnane diterpenoids in the extracts from the different parts (stems, leaves, and fruits) of D. pontica. Methods An ultra‐high‐performance liquid chromatography coupled with Q‐Exactive hybrid quadrupole Orbitrap mass spectrometer (UHPLC‐Q‐Exactive‐Orbitrap MS) was used for the qualitative analysis of D. pontica. The stems, leaves, and fruits of D. pontica were extracted with diethyl ether. Each extract was then pretreated by a solid phase extraction cartridge and subjected to LC–MS/MS analysis. Detected daphnane diterpenoids were tentatively identified by comparison with an in‐house daphnane library, and their chemical structures were estimated in detail by MS/MS fragmentation evaluation. Results A total of 33 kinds of daphnanes were identified from the different parts of D. pontica, and were classified into three subtypes: daphnane orthoester, polyhydroxy daphnane, and macrocyclic daphnane orthoester. Among them, six daphnanes were postulated to be previously unreported compounds based on MS/MS fragmentation elucidation. Furthermore, the three plant parts showed similar daphnane diterpenoid profiles, with the stems containing the most abundant daphnane diterpenoids. Conclusion This is the first study to perform qualitative analysis of daphnane diterpenoids systematically and comprehensively in different parts of D. pontica. The results revealed that D. pontica is a plant resource rich in a variety of daphnane diterpenoids.

Abstract:Drimane‐type sesquiterpenes (DTSs) are significant terpenoid natural products characterized by their unique C15 bicyclic skeleton. They are produced by various organisms including plants, fungi, bacteria and marine organisms, and exhibit a diverse array of bioactivities. These bioactivities encompass antifeedant, anti‐insecticidal, anti‐bacterial, anti‐fungal, anti‐viral and anti‐proliferative properties. Some DTSs contribute to the pungent flavor found in herb plants like water pepper, while others serve as active components responsible for the anti‐cancer activities observed in medicinal mushrooms such as (‒)‐antrocin from Antrodia cinnamomea. Recently, DTS synthases have been identified in various organisms, biosynthesizing drimenol, drim‐8‐ene‐11‐ol and (+)‐albicanol, which all possess the characteristic drimane skeleton. Interestingly, despite these enzymes producing chemical molecules with a drimane scaffold, they exhibit minimal amino acid sequence identity across different organisms. This Concept article focuses on the discovery of DTS synthases and the tailoring enzymes generating the chemical diversity of drimane natural products. We summarize and discuss their key features, including the chemical mechanisms, catalytic motifs and functional domains employed by these terpene synthases to generate DTS scaffolds.

A terpene synthase supergene locus determines chemotype in Melaleuca alternifolia (tea tree)

Abstract:Leaf oil terpenes vary categorically in many plant populations, leading to discrete phenotypes of adaptive and economic significance, but for most species, a genetic explanation for the concerted fluctuation in terpene chemistry remains unresolved. To uncover the genetic architecture underlying multi-component terpene chemotypes in Melaleuca alternifolia (tea tree), a genome-wide association study was undertaken for 148 individuals representing all six recognised chemotypes. A number of single nucleotide polymorphisms in a genomic region of c. 400 kb explained large proportions of the variation in key monoterpenes of tea tree oil. The region contained a cluster of 10 monoterpene synthase genes, including four genes predicted to encode synthases for 1,8-cineole, terpinolene, and the terpinen-4-ol precursor, sabinene hydrate. Chemotype-dependent null alleles at some sites suggested structural variants within this gene cluster, providing a possible basis for linkage disequilibrium in this region. Genotyping in a separate domesticated population revealed that all alleles surrounding this gene cluster were fixed after artificial selection for a single chemotype. These observations indicate that a supergene accounts for chemotypes in M. alternifolia. A genetic model with three haplotypes, encompassing the four characterised monoterpene synthase genes, explained the six terpene chemotypes, and was consistent with available biparental cross-segregation data.
Pedoclimatic Conditions Influence the Morphological, Phytochemical and Biological Features of Mentha pulegium L.

Abstract:In this study, Mentha pulegium leaves and flowers harvested in three different Sicilian areas were investigated from a micromorphological, phytochemical and biological point of view. Light and scanning electron microscopy showed the presence of spherocrystalline masses of diosmin both in the leaf epidermal cells and in thin flower petals. Two different chemotypes were identified (I, kaempferide/rosmarinic acid; II, jaceidin isomer A). Phytochemical screening identified plant from collection site II as the richest in total phenolics (16.74 g GAE/100 g DE) and that from collection site I as the richest in flavonoids (46.56 g RE/100 g DE). Seventy-seven metabolites were identified both in flower and leaf extracts. Plant from site II showed the best antioxidant (0.90–83.72 μg/mL) and anti-inflammatory (27.44–196.31 μg/mL) activity expressed as half-maximal inhibitory concentration (IC50) evaluated by DPPH, TEAC, FRAP, ORAC, BSA denaturation and protease inhibition assays. These data were also corroborated by in vitro cell-based assays on lymphocytes and erythrocytes. Moreover, plant of site II showed the best antiangiogenic properties (IC50 33.43–33.60 μg/mL) in vivo on a chick chorioallantoic membrane. In conclusion, pedoclimatic conditions influence the chemotype and the biological activity of M. pulegium, with chemotype I showing the most promising biological properties.

Abstract:Wild aromatic plants present high intra- and inter-population chemical polymorphisms which are of great ecological and economic interest; however, the factors influencing the phytochemical diversity of aromatic plants remain still unexplored for many species. Here, Teucrium marum L. (Lamiaceae) has been studied, a plant endemic to the western Mediterranean islands, very well-known from the phytochemical point of view but scarcely investigated regarding the ecological factors that influence its phytochemical diversity within and among populations. The specific aims were to: 1) define the chemical composition variability of its essential oils; 2) determine its inter- and intra-population chemical variability; and 3) evaluate whether the elevation, climatic factors and/or the soil substrate determined the phytochemical variability of T. marum along a gradient from coastal to mountain wild populations on the island of Sardinia (Italy). Fresh T. marum aerial parts were randomly collected from ten individuals in six different localities in Sardinia: three coastal and three mountainous. Dried leaf samples were hydrodistillated using a classical Clevenger apparatus to obtain the corresponding essential oils. The composition of each essential oil was chemically characterised and analyzed by gas chromatography coupled to mass spectrometry. Ninety compounds were identified: among the others, the two compounds that mainly characterised the essential oils of the studied populations, dolichodial and (E)-β-caryophyllene, are of great economic interest. Statistical analyses showed significant differences in phytochemical essential oil composition among and within the studied populations, which clustered following a geographical pattern rather than a simple climatic or edaphic grouping. Taken together the results here shown shed light on the environmental and geographical conditions that determine the chemical variability of essential oils in T. marum, highlighting a clear coastal vs mountain clustering, which has an ecological and economic relevance, especially for the potential utilization of dolichodial as an environmental-friendly insecticide.

Abstract:A total of 43 compounds, including phenolic acids, flavonoids, lignans, and diterpene, were identified and characterized using UPLC‐ESI‐Q‐TOF‐MS coupled with UNIFI software. The identified flavonoids were mostly isomers of luteolin, apigenin, and quercetin, which were elucidated and distinguished for the first time in pepper cultivars. The use of multivariate data analytics for sample discrimination revealed that luteolin derivatives played the most important role in differentiating pepper cultivars. The content of phenolic acids and flavonoids in immature green peppers was generally higher than that of mature red peppers. The pepper extracts possessed significant antioxidant activities, and the antioxidant activities correlated well with phenolic contents and their molecular structure. In conclusion, the findings expand our understanding of the phytochemical components of the Chinese pepper genotype at two maturity stages. Moreover, a UPLC‐ESI‐Q‐TOF‐MS in negative ionization mode rapid methods for characterization and isomers differentiation was described.

Abstract:Hedychium spicatum Sm. is an endangered herb which grows preferably in humid tropics and subtropics known for its distinct camphoraceous flavour and medicinal properties. The essential oils of H. spicatum rhizomes from six different natural habitats at different altitudes in Uttarakhand, India were analyzed. Twenty compounds were identified, accounting for 87.4–98.5% of the essential oils, with 1,8-cineole (33.4–41.9%), α-terpinene (39.6%), camphor (31.4%), and linalool (29.9%), being the prevalent major constituents of all accessions. HS-ALM ( H. spicatum rhizomes Almora essential oil), HS-DDN ( H. spicatum rhizomes Dehradun essential oil), HS-PAU ( H. spicatum rhizomes Pauri essential oil), HS-TEH ( H. spicatum rhizomes Tehri essential oil), and HS-USN ( H. spicatum rhizomes Udham Singh Nagar essential oil) were dominated by oxygenated monoterpenes with 43.1–74.0%, respectively. In contrast, in HS-PIT ( H. spicatum rhizomes Pithoragarh essential oil), hydrogenated monoterpenes dominated 57.8% of the total oil constituents. All accessions were assessed for significant nematicidal potential against Meloidogyne incognita and insecticidal potential against Spodoptera litura . The proteins/enzymes used for validation using docking studies were acetylcholinesterase (PBD ID: IC2O) and carboxylesterase (PDB ID: 1CI8). The study produced significant outcomes, showing the binding energy estimation of tested oil components to active sites supported the structure-activity relationship with bioinformatics tools.

Monoterpene composition of essential oil from peppermint (Mentha x piperita L.) with regard to leaf position using solid-phase microextraction and gas chromatography/mass spectrometry analysis

Abstract:Monoterpene compounds of leaf pairs and flowers of Mentha x piperita have been studied by direct headspace sampling using solid-phase microextraction coupled with gas chromatography/mass spectrometry (SPME-GC/MS). The content of peppermint-characteristic compounds such as menthol, menthyl acetate, and neomenthol increased in a basipetal direction (older plant parts), whereas menthone and isomenthone showed higher levels in the acropetal direction (younger plant parts). Higher levels of menthofuran were found in peppermint flowers in contrast to the leaves. SPME sampling resulted in relatively higher amounts of high-volatile monoterpenes and lower detection of less volatile compounds such as menthol and menthone, compared to solvent-based samples from essential oil distillation.
Transcriptome analysis of transcription factors and enzymes involved in monoterpenoid biosynthesis in different chemotypes of Mentha haplocalyx Briq

Abstract:Background: The main active ingredients of Mentha haplocalyx Briq. essential oils are monoterpenes. According to the component of essential oils, M. haplocalyx can be divided into different chemotypes. Chemotype variation is widespread in Mentha plants but its formation mechanism is unclear. Methods: We selected the stable chemotype l-menthol, pulegone, and carvone of M. haplocalyx for transcriptome sequencing. To further investigate the variation of chemotypes, we analyzed the correlation between differential transcription factors (TFs) and key enzymes. Results: Fourteen unigenes related to monoterpenoid biosynthesis were identified, among which (+)-pulegone reductase (PR) and (-)-menthol dehydrogenase (MD) were significantly upregulated in l-menthol chemotype and (-)-limonene 6-hydroxylase was significantly upregulated in carvone chemotype. In addition, 2,599 TFs from 66 families were identified from transcriptome data and the differential TFs included 113 TFs from 34 families. The families of bHLH, bZIP, AP2/ERF, MYB, and WRKY were highly correlated with the key enzymes PR, MD, and (-)-limonene 3-hydroxylase (L3OH) in different M. haplocalyx chemotypes (r > 0.85). The results indicate that these TFs regulate the variation of different chemotypes by regulating the expression patterns of PR, MD, and L3OH. The results of this study provide a basis for revealing the molecular mechanism of the formation of different chemotypes and offer strategies for effective breeding and metabolic engineering of different chemotypes in M. haplocalyx.
Characterization of two closely related citrus cultivars using UPLC-ESI-MS/MS-based widely targeted metabolomics

Abstract:Citrus cultivars are widely spread worldwide, and some of them only differ by specific mutations along the genome. It is difficult to distinguish them by traditional morphological identification. To accurately identify such similar cultivars, the subtle differences between them must be detected. In this study, UPLC-ESI-MS/MS-based widely targeted metabolomics analysis was conducted to study the chemical differences between two closely related citrus cultivars, Citrus reticulata ‘DHP’ and C . reticulata ‘BZH’. Totally 352 metabolites including 11 terpenoids, 35 alkaloids, 80 phenolic acids, 25 coumarins, 7 lignans, 184 flavonoids and 10 other compounds were detected and identified; Among them, 15 metabolites are unique to DHP and 16 metabolites are unique to BZH. Hierarchical cluster analysis (HCA), principal component analysis (PCA), and orthogonal signal correction and partial least squares-discriminant analysis (OPLS-DA) can be used to clearly discriminate between DHP and BZH. 93 metabolites including 36 down-regulated and 57 up-regulated are significantly different in DHP and BZH. They are mainly involved in the biosynthesis of flavonoids, flavones, flavonols, and isoflavonoids. In addition, the relative content levels of flavonoids, alkaloids, and terpenoids are much higher in the peel of DHP than that of BZH, the presence of which may correlate with the quality difference of the peels. The results reported herein indicate that metabolite analysis based on UPLC-ESI-MS/MS is an effective means of identifying cultivars with different genotypes, especially those that cannot be distinguished based on traditional identification methods.

Abstract:Glycolipid metabolism disorders (GLMDs) have been reported to be involved in dysfunctions of a few key enzymes such as ATP-citrate lyase (ACL) and death-associated protein kinase (DAPK)-related apoptosis inducing protein kinase-2 (DRAK2). Exploring new and effective small molecule ligands for these enzymes, natural products are expected to have the advantage over synthesized compounds. In our preceding work, a number of C50 terpenoids with bioactivities against ACL have been found from Pseudotsuga forrestii (a ‘vulnerable’ Yunnan Douglas fir) by the guidance of an LC-MS/MS-based molecular ion networking strategy. The combination of the rest portions of the EtOAc fraction from this precious plant sample did not predict any C50 compounds, but it was detected to be rich in C40/C30/C20 terpenoids and preliminarily exhibited DRAK2 inhibition. In this follow-up study, ten undescribed terpenoids (1−10) and 25 structurally related known compounds were further isolated and characterized. The new structures and their absolute configurations were determined by spectroscopic methods and/or X-ray diffraction analyses. 7,13-Friedo-lanostanes 1 and 2 feature intriguing skeletons with an unusual α, β-unsaturated spirolactone unit formed between C-13 and C-23 via oxygen-bridge or with an 8(14→13)-abeo moiety. Triterpene-monoterpene hybrid 3 could be regarded as a unique tetraterpene formed by intermolecular [4 + 2] Diels-Alder cycloaddition between a rearranged lanostane (dienophile) and a β-myrcene (diene). Methyl 15-methoxy-8,11,13-abietatrien-18-yl-succinate (9) and 3-oxo-24(E)-lanosta-8,24-dien-26-oic acid (35) were confirmed to have DRAK2 inhibitory effects with IC50 values of 6.6 and 4.1 μM, respectively. A molecular docking study disclosed alternative H-bonds/interacting amino acid residues between compounds 9/35 and the DRAK2 enzyme when comparing with those of the synthesized inhibitors reported in literature. The above findings could support the important role of protecting plant species diversity in support of chemical diversity and potential sources of new therapeutic agents for the treatment of GLMDS.
Phytochemical variations in Stachys lavandulifolia populations and the role of ecological and edaphic factors

Abstract:Geographic location, environmental conditions, and edaphic factors are interconnected, and collectively influence the total content of some plant-based compounds. Mountain tea ( Stachys lavandulifolia Vahl) is a perennial plant in the Lamiaceae family, having prominent medicinal properties due to its unique chemical composition. The current research evaluated effective traits that shape the content of essential oil (EO) and phytochemical compounds in S. lavandulifolia populations (P). Samples were collected from 20 different habitats located in the west and northwest of Iran. The results indicated that the Heris (P3) and Sarab (P7) populations exhibited high levels of EOs and phytochemical properties. In the studied areas, principal component and cluster analyses considered environmental factors like climate and soil, thus revealing significant findings. Soil characteristics, including organic matter content, total nitrogen (N), potassium (K), and soil phosphorus (P) content, were identified as the most influential factors, with average annual rainfall following closely. The highest average annual temperatures largely contributed to distinguishing the areas, thus categorizing the habitats into three distinct groups. EO from 20 populations analyzed using GC/MS showed 41 compounds, the most important of which were α-pinene (1.21–33.29 %), α-bisabolol (0.71–53.05 %), and spathulenol (2.8–18.8 %), germacrene-D (1.13–12.18 %), β-phellandrene (0.09–16.72 %), myrcene (1.01–13.34 %), phytol (1.19–10.14 %), and viridiflorol (0.93–6.19 %). Grouping the S. lavandulifolia populations based on their main compounds in the EO divided them into two distinct habitats. The first habitat had eleven S . lavandulifolia populations, whereas the second had nine. Comparison of mean values among habitats showed that the first habitat was superior to the second, regarding sesquiterpene chemical compounds such as spathulenol, viridiflorol, α-muurolol, α-cadinol, and valeranone. The second habitat was more suitable than the first because of higher amounts of monoterpene chemical compounds, such as α-pinene, myrcene, and β-phellandrene. Overall, in this study, Heris (P3), Azarshahr (P4), and Sarab (P7) populations were favorable because of their EO yield and compounds. Our approach to identifying environmental and edaphic cues that shape the EO content, chemotype, or the presence of high amounts of specific compounds can help to select regions for sampling plant material while prioritizing desirable chemical profiles for direct use or in breeding programs.

Abstract:In order to investigate the differences in the chemical composition of Dendrobium officinale at different altitudes, we collected Dendrobium officinale from different altitudes in Guizhou Province, China, and firstly determined its polysaccharide content by using a sulphuric acid‐phenol colour development method with reference to the Chinese Pharmacopoeia, and further determined its metabolites by using widely targeted metabolomics, and explored the differences in the chemical composition of Dendrobium officinale at different altitudes using multivariate statistical analysis. We found that the polysaccharide content was higher in plants growing at 1122 m, a total of 902 secondary metabolites were detected by wildly targeted metabolomics, and amino acids and their derivatives were more highly expressed at 1122 m, while other metabolites were more highly expressed at 835 m. Furthermore, we found that the phenolic acid compound nerugein was only present in plants at 835 m, and two lipid compounds, namely Lyso PE 20:4 and its isomer, were only present at 1122 m. Taken together, these results may provide a basis for the selection and clinical application of D. officinale cultivated at different altitudes.

Abstract:Abstract Background Geographical factors affect the nutritional, therapeutic and commercial values of fruits. Dragon fruit ( Hylocereus spp ) is a popular fruit in Asia and a potential functional food with diverse pharmacological attributes. Although it is produced in various localities, the information related to the altitudinal variation of dragon fruit nutrients and active compounds is scarce. Hence, this study aimed to investigate the variations in metabolite profiles of H. polyrhizus (variety Jindu1) fruit pulps from three different altitudes of China, including Wangmo (WM, 650 m), Luodian (LD, 420 m), and Zhenning (ZN, 356 m). Jindu1 is the main cultivated pitaya variety in Guizhou province, China. Results The LC-MS (liquid chromatography-mass spectroscopy)-based widely targeted metabolic profiling identified 645 metabolites, of which flavonoids (22.64%), lipids (13.80%), phenolic acids (12.40%), amino acids and derivatives (10.39%), alkaloids (8.84%), and organic acids (8.37%) were dominant. Multivariate analyses unveiled that the metabolite profiles of the fruit differed regarding the altitude. Fruits from WM (highest altitude) were prime in quality, with higher levels of flavonoids, alkaloids, nucleotides and derivatives, amino acids and derivatives, and vitamins. Fruits from LD and ZN had the highest relative content of phenolic acids and terpenoids, respectively. We identified 69 significantly differentially accumulated metabolites across the pulps of the fruits from the three locations. KEGG analysis revealed that flavone and flavonol biosynthesis and isoflavonoid biosynthesis were the most differentially regulated. It was noteworthy that most active flavonoid compounds exhibited an increasing accumulation pattern along with the increase in altitude. Vitexin and isovitexin were the major differentially accumulated flavonoids. Furthermore, we identified two potential metabolic biomarkers (vitexin and kaempferol 3-O-[2-O-β-D-galactose-6-O-a-L-rhamnose]-β-D-glucoside) to discriminate between dragon fruits from different geographical origins. Conclusion Our findings provide insights into metabolic changes in dragon fruits grown at different altitudes. Furthermore, they show that growing pitaya at high altitudes can produce fruit with higher levels of bioactive compounds, particularly flavonoids.

Abstract:Melaleuca alternifolia (tea tree), family Myrtaceae, is endemic to the northern rivers of NSW, Australia. Since 1925, the volatile components of the hydro- and steam-distilled oils of the leaves have been studied in detail. However, the less-volatile compounds have not been investigated. Using an ethanolic extract of the seedling leaves, the non-volatile components were studied using gas chromatography-mass spectrometry (GC/MS) and liquid chromatography- mass spectrometry (LC/MS). Four of these less-volatile components were isolated by preparative-HPLC from young seedling leaves and identified as the acylphloroglucinols 1-(2,6-dihydroxy-4-methoxy-3-methylphenyl)-2-methylpropan-1-one, callisalignone A, 1-(2,6-dihydroxy-4-methoxyphenyl)-3-methylbutan-1-one and pulverulentone B described here for the first time from M. alternifolia. These compounds change in concentration in the leaf sets as later seedling leaves mature on the seedling.

Impact of Edaphic and Climatic Factors on Thymus pulegioides Essential Oil Composition and Potential Prevalence of Chemotypes

Abstract:Intraspecific chemical polymorphism is characteristic of essential oil bearing Thymus pulegioides (Lamiaceae). Soil chemical composition and climatic conditions can influence not only quantitative and qualitative composition of essential oils, but also on prevalence of different chemotypes in space. The purpose of study was to determine the impact of edaphic and climatic factors on T. pulegioides essential oil composition and potential chemotypes prevalence. It were investigated 131 habitats of T. pulegioides in all ten climatic sub-districts of Lithuania. Essential oils were isolated by hydrodistillation and analysed by GC-FID and GC-MS. The content of humus was estimated by oxidation method, the content of mobile potassium (K2O) and mobile phosphorus (P2O5)—flame photometry using 0.2 M HCl solution, the soil pH—electrometrically and 15 elements of soil—by energy-dispersive X-ray fluorescence. The meteorological data (temperature, rainfall, photosynthetically active solar radiation and sunshine duration) were obtained from the meteorological bulletins. Results showed that humus in soil positively, manganese and cobalt—negatively influenced on the essential oil accumulation in T. pulegioides. Carvacrol was the most common compound in the essential oil of T. pulegioides; higher amounts of sulphur and mobile phosphorus, lower amounts of sodium in soil, higher rainfall in blooming period are favourable for this compound and for potential prevalence of carvacrol chemotype. Geraniol was frequent in the eastern and the central part of Lithuania, where the temperature in vegetation season is higher, and in soils characterised by higher and lower amounts of sodium and chlorine, respectively. Thymol, linalool and α-terpinyl acetate were rare. Edaphic and climatic conditions can differently influence on essential oil yield and chemical composition of species as well as on potential prevalence of different chemotypes of same species in space.
