Abstract:Context: Use of herbal medicine throughout the world is increasing. Plants still remaining the primary source of supply of many important drugs used in modem medicine. Spinacia Oleracea i,e spinach leaves contain more number of anti-inflammatory phytochemcicals such as carbohydratie, tanins and phenolic compounds, saponins, flavinoids and steroid. Better collagenation seen under the influence of the flavinoids, phenolic compound and saponins which is responsible for the free radical scavenging activity and are believed to be some of the most important component for wound healing and antioxident activity. Spinacia Oleracea scavenges free oxygen redicals and increases the catalase level in granulation tissue. Plants still remaining the primary source of supply of many important drugs used in modem medicine. Considering its medicinal value and availability in our country this study was undertaken to evaluate the antiinflammatory effect of the Spinacia Oleracea leaf extract in rat models. Material and Methods: The experiments were carried out on 30 (thirty) Swiss male albino rats. They were collected from the ICDDRB, Dhaka. The rats were of male weighing between 150- 200gm which were divided randomly into 5 groups each having 6 rats. Groups were labeled as group-I, group-II, group-III, group-IV and group-V. The anti inflammatory effect of Spinacia Oleracea leaf extract in experiment rat were evaluated and compared with the anti inflammatory effects of aspirin and hydrocortisone. The study was prospective experimental type and was conducted in the department of Pharmacology, Dhaka Medical College, Dhaka, from July 2011 to June 2012. Result : Administration of mathanolic extract and water extract of Spinacia Oleracea leaf at a dose of 200mg/kg body weight orally produced a significant (P<0.05) anti-inflammatory effect, and the percentage of inhibition of oedema formation was 28.75% and 40.79% respectively. There were highly significant (P<0.05) percentage of inhibition of oedema formation was observed in aspirin (40.52%) and in hydrocortisone (47.71%). Abstract:OBJECTIVE:To screen, isolate and identify the compounds with anti-inflammatory activity from Dalbergia odorifera T. Chen.METHODS:Lipopolysaccharide (LPS) was used to stimulate RAW264.7 macrophages, and this model was further used to screen and separate active compounds from Dalbergia odorifera T. Chen, followed by compound chemical structure identification using mass spectrometry and nuclear magnetic resonance. Griess assay was performed to measure the released nitric oxide (NO), and ELISA was applied to detect TNF-α content.RESULTS:Compound I (sativanone) showed strong anti-inflammatory activity (IC50;=12.48 g/mL). Isoliquiritigenin (compound III), naringenin (compound IV) and liquiritigenin (compound V) had moderate anti-inflammatory activity, with IC50; value at 18.33, 42.59, 29.43 g/mL, respectively. Compound II and compound VI had no anti-inflammatory activity with concentration at 50 g/mL. Sativanone, isoliquiritigenin, liquiritigenin and naringenin at their concentrations ranging from 3.125-50 g/mL, could inhibit NO release in RAW264.7 cells stimulated by LPS. Sativanone (compound I) could also inhibit TNF-α secretion from LPS stimulated RAW264.7 cells.CONCLUSION:Liquiritigenin, isoliquiritigenin, naringenin and sativanone isolated from Dalbergia odorifera T. Chen, could reduce NO release from LPS-stimulated macrophage cell line (RAW264.7), at a dose-dependent manner. What's more, sativanone play an anti-inflammatory role also through inhibiting inflammatory cytokine TNF-α.

Abstract:Plants are a rich source of antioxidants that are produced naturally. Therefore, this study was aimed to evaluate the effect of the plant Ricinodendron heudelotii (Baill.) in the attenuation of paracetamol (PCM) hepatotoxicity in Wistar rats. Twenty-four male albino Wistar rats weighing between 200 and 250 g were divided into four groups, with six rats each. Group 1 served as the control group, receiving just distilled water. Groups 2 and 3 received orally 250 mg/kg bwt/day PCM and 300 mg/kg bwt/day methanol extract of Ricinodendron heudelotii (Baill.) leaves for two weeks, respectively. For group 4, the Ricinodendron heudelotii (Baill.) leaf extract was pre-administered for 1 week before receiving 300 mg/kg bwt/day Ricinodendron heudelotii (Baill.) leaves extract and 250 mg/kg bwt/day PCM for 2 weeks. As a marker of liver damage, serum aspartate aminotransferase (AST) and alanine aminotransferase (ALT) were measured. Liver tissue reduced glutathione (GSH) concentration, superoxide dismutase (SOD), glutathione-S-transferase (GST), and catalase activities were utilized to determine antioxidant state, while malondialdehyde (MDA) concentration was employed as a lipid peroxidation indicator. When compared to the control group, the activities of serum AST, ALT, SOD, and MDA levels were considerably ( p < 0.05) higher in the PCM group, although GSH level and GST and catalase activities were significantly lower. In comparison to the PCM group, co-administration of PCM with Ricinodendron heudelotii (Baill.) extract decreased serum AST and ALT activities. This study shows that the leaf extracts of Ricinodendron heudelotii (Baill.) protects Wistar rats' livers from PCM-induced oxidative stress by increasing antioxidant enzymes.

Abstract:Hyperkalemic RTA in young children usually indicates renal or adrenal insufficiency, adrenal hyperplasia or a form of tubular resistance to mineralocorticoid. NH4Cl used to evaluate acidifying ability may augment pre-existing hyperkalemia to potentially dangerous cardiotoxic levels. An infant with profound neonatal metabolic alkalosis secondary to maternal psychogenic vomiting was found to have hyperkalemic RTA at two months of age. Lab values included, in mEq/L: Na 138, Cl 105, K 5.8, HCO3 18. BUN, blood sugar, serum creatinine and cortisol levels and BP were normal. Serum aldosterone (225 ng/ml) and plasma renin activity (20.1 ng/ml/hr) were high. Neither Florinef .1 mg/day nor Florinef + 4 days of supplemental NaCl 6 mEq/kg/day corrected the hyperkalemia, but renin and aldosterone levels dropped during this time. After Furosemide (F) 1 mg/kg IV:Fractional excretion of K rose and urine pH dropped within one hour after Furosemide and serum K and HCO3 were normal after 4 hours. Twelve mEq/kg/day of NaHCO3 were sufficient to correct the hyperkalemic acidosis. We conclude that, as in adults (Kurtzman KI, 1983 and Arruda KI, 1983), Furosemide stimulates acid secretion in the infant and is useful in the evaluation of RTA without the risk of worsening hyperkalemia.

Abstract:Ricinus communis (castor plant) is a potent medicinal plant, which is commonly used in the treatment of various ailments. The present study was conducted to appraise the cytotoxicity and mutagenicity of R. communis along with antioxidant and antimicrobial activities. Cytotoxicity was evaluated by hemolytic and brine shrimp assays, whereas Ames test (TA98 and TA100) was used for mutagenicity evaluation. Plant different parts were extracted in methanol by shaking, sonication and Soxhlet extraction methods. The R. communis methanolic extracts showed promising antioxidant activity evaluated as through total phenolic contents (TPC), total flavonoid content (TFC), DPPH free radical inhibition, reducing power and inhibition of linoleic acid oxidation. R. communis seeds, stem, leaves, fruit and root methanolic extracts showed mild to moderate cytotoxicity against red blood cells (RBCs) of human and bovine. Brine shrimp lethality also revealed the cytotoxic nature of extracts with LC50 in the range of 0.22-3.70 (µg/mL) (shaking), 1.59-60.92 (µg/mL) (sonication) and 0.72-33.60 (µg/mL) (Soxhlet), whereas LC90 values were in the range of 345.42-1695.81, 660.50-14,794.40 and 641.62-15,047.80 µg/mL for shaking, sonication and Soxhlet extraction methods, respectively. R. communis methanolic extracts revealed mild mutagenicity against TA98 (range 1975 ± 67 to 2628 ± 79 revertant colonies) and TA100 (range 2773 ± 92 to 3461 ± 147 revertant colonies) strains and these values were 3267 ± 278 and 4720 ± 346 revertant colonies in case of TA98 and TA100 positive controls, respectively. R. communis methanolic extracts prevented the H2O2 and UV to Plasmid pBR322 DNA oxidative damage. Results revealed that R. communis is a potential source of bioactive compounds and in future studies the bioactive compounds will be identified by advanced spectroscopic techniques.

Abstract:Women have been found to be at a higher risk of morbidity and mortality from type 2 diabetes mellitus (T2DM) and asthma. α-Glucosidase inhibitors have been used to treat T2DM, and arachidonic acid 15-lipoxygenase (ALOX15) inhibitors have been suggested to be used as treatments for asthma and T2DM. Compounds that inhibit both enzymes may be studied as potential treatments for people with both T2DM and asthma. This study aimed to determine potential anti-diabetic and anti-inflammatory bioactive hits from Coriaria intermedia Matsum. stem and Dracontomelon dao (Blanco) Merr. & Rolfe bark. A bioassay-guided fractionation framework was used to generate bioactive fractions from C. intermedia stem and D. dao bark. Subsequently, dereplication through ultra-high performance liquid chromatography-tandem mass spectrometry (UHPLC-MS/MS) and database searching was performed to putatively identify the components of one bioactive fraction from each plant. Seven compounds were putatively identified from the C. intermedia stem active fraction, and six of these compounds were putatively identified from this plant for the first time. Nine compounds were putatively identified from the D. dao bark active fraction, and seven of these compounds were putatively identified from this plant for the first time. One putative compound from the C. intermedia stem active fraction (corilagin) has been previously reported to have inhibitory activity against both α-glucosidase and 15-lipoxygenase-1. It is suggested that further studies on the potential of corilagin as an anti-diabetic and anti-inflammatory treatment should be pursued based on its several beneficial pharmacological activities and its low reported toxicity.

Abstract:Phytochemical study of Psychotria schlechtendaliana (Rubiaceae) aerial parts resulted in the isolation of alkaloids 4-N-oxide harmane (1) and strictosidinic acid (2), and the terpenoids sitosterol, estigmasterol, α-amyrin, β-amyrin and betulinic acid. The crude extract (CE), its fractions (hexane FH, chloroform FC, ethyl acetate FEA, hydromethanolic FHM, alkaloidal chloroform FCOH, alkaloidal aqueous FAq) and majority alkaloid (1) were investigated for their antiproliferative potential against nine human tumor cells lines and one non-tumoral human cell line (HaCat). CE, FH and FEA fractions exhibited strong growth inhibition for ovary cells (OVCAR-3, GI50 = 5.89; 1.36 and 6.59 μg mL-1, respectively) and FC, FH and FAq fractions showed potent activity on the growth of leukemia cell lines (K562, GI50 = 1.92; 7.23 and 8.81 μg mL-1, respectively). Compound 1 exhibited selective antiproliferative activity to breast cancer (MCF-7, GI50 = 32.7 μg mL-1) and was non-toxic to HaCat cells. To evaluate the anti-inflammatory effect, models of ear edema induced by croton oil and the enzyme myeloperoxidase (MPO) were used. FH and FC fractions exhibited antiinflammatory effect and reduce ear edema compared to the control group. These fractions showed results superior to those exhibited by indomethacin (75.1%), FH (97.5%) and FC (95.5%) fractions.

Abstract:Coriandrum sativum has been used in traditional medicines as an anti-inflammatory, analgesic, and antibacterial agent. It is recently been shown to have antioxidant activities. This study evaluated the effects of Flavonoid Rich Fraction of Coriandrum sativum Leaf (FRFCSL) on Lipid Profile, Nitric Oxide, Ang II and Cardio Histopathology in L-NAME intoxicated Experimental Rats. Standard analytical method and DPPH radical scavenging activity were employed for the phytochemicals and antioxidant analysis respectively. The acute toxicity study showed the extract and flavonoids fraction are nontoxic at 5000mg/kg body weight. Thirty rats were divided into five groups of six (6) rats each. Group 1 was administered water and feed only, Group 2 was administered 40mg/kg N-Nitro-L-Arginine Methyl Ester (L-NAME) without treatment, Group 3 through 5 were administered 40mg/kg N-Nitro-L-Arginine Methyl Ester (L-NAME) with concomitant administration of Captopril (20mg/kg b.w), FRFCSL (200mg/kg b.w) and FRFCSL (400mg/kg b.w) respectively for 21 days. In phytochemical screening, presence of flavonoids, saponins, tannins, alkaloids, cardiac glycosides, steroids were confirmed as part of secondary metabolites in the extract. The flavonoid fraction showed increase % DPPH radical inhibition in concentration-dependent manner. FRFCSL treatment significantly (p<0.05) and dose-dependently lowered low density lipoprotein and triglycerides. Also It significantly (p<0.05) prevented L-NAME induced decrease in serum angiotensin II, high density lipoprotein and serum nitric oxide concentrations compared to the untreated rats. The Flavonoid Rich Fraction of Coriandrum Sativum Leaf showed great potential as antihypertensive, antihyperlipidemic and cardioprotective agent in rats thus confirming its usefulness in traditional health therapy and potential for antihypertensive drug development.

Abstract:OBJECTIVE:To study the bioactive constituents of the fresh rhizome of Pinellia ternata, and provide the scientific basis for quality control.METHOD:Various chromatographic techniques were used to separate and purify the chemical constituents, and their chemical structures were determined on the basis of physical-chemical properties and spectroscopic analysis. The inhibitory effects of the isolated compounds on tumor necrosis factor (TNF)-alpha production in the peritoneal macrophages of mice stimulated with lipopolysaccharide (LPS) were assayed in vitro by microplate colorimetric method.RESULT:Nine compounds were isolated and identified as (E)-p-coumaryl alcohol (1), 3, 4-dihydroxycinnamyl alcohol (2), ferulic acid (3), lariciresinol (4), erythro-guaiacylglycerol-beta-O-4'-sinapyl ether (5), dehydrodiconiferyl alcohol (6) , isolariciresinol (7) , sachaliside 1 (8) and coniferin (9). The inhibitory effect of compounds 1, 2, 3, 8, and 9 were 24.1% , 57.6% , 40.2% , 82.7% , and 62.0% , respectively, against the TNF-alpha production in the peritoneal macrophages of mice stimulated with LPS at a concentration of 10(-5) mol L(-1) in vitro.CONCLUSION:The compounds 1, 2 and 4-8 from genera Pinellia Ten. and compound 9 from P. ternata were isolated for the first time. The compounds 1, 2, 8 and 9 were phenylpropanoids, and 4-7 were lignanoids. The anti-inflammatory effects of the rhizome of P. ternata might relate at the least to compounds 1, 2, 3, 8 and 9.

Abstract:The present study highlights the antioxidant and anti-inflammatory activities from Anadenanthera colubrina leaves ethanolic extract (EEAc) and its phytochemical profile. The chemical profile of EEAc was determined and in vitro free radical scavenging potential, cell viability in RAW 264.7 and in vitro and in vivo anti-inflammatory activity were evaluated. The analysis of EEAc showed several phenolic compounds such as tannins and phenolic acids. The high antioxidant potential observed is possibly due to its high phenolic content. The EEAc (500 mg kg−1) showed an in vivo effect at the same level of dexamethasone; in vitro, at 50 μg mL−1, inhibited approximately 80% of nitric oxide production concentration, showed an inhibition of more than 50% of TNF-α production and presented high cell viability. The results show that A. colubrina leaves are an important source of phytochemicals that possesses antioxidant and anti-inflammatory properties.

Hepatoprotective Effects of Fractions of Adansonia Digitata Leaves on Carbon Tetrachloride (CCL4)- Induced Toxicity in Wistar Rats

Abstract:Phytochemicals, naturally occurring compounds in plants, offer health benefits to humans. This study aimed to determine the effects of fractions of ethanol extract of Adansonia digitata leaves on carbon tetrachloride (CCl4)-induced toxicity in wistar rats. The extraction was done using absolute ethanol, followed by fractionation with different solvent combination via column chromatography. Elution of extract was done with solvent system by gradually increasing polarity beginning from n-hexane, chloroform, ethyl acetate, methanol, ethanol and finally water. In total, 22 fractions were collected in 200 mL beaker each. The fractions were subjected to total antioxidant analysis using 1, 1-diphenyl-2-picryhydrazyl (DPPH) assay. Total phenolics and total flavonoids content were analyzed by Folin-Ciocalteu. GC-MS, FTIR and HPLC analysis were also carried out. Thirty-six male albino rats were distributed into six groups of six (n = 6). The negative control group received CCl4 only. The standard control group was administered 2 CCl4/kg body weight + 25mg/kg body weight silymarin followed by different doses of ethyl acetate:ethanol fraction-20 mg/kg, 40 mg/kg and 50 mg/kg for 21 days. Induction and treatment were carried out in the beginning of a new week. The animals were fasted for 24 hours, sacrificed and blood samples were collected for biochemical analysis. The administered fraction led to the statistically insignificant (p> 0.05) and statistically significant (p 0.05) in the treated rats as compared to the untreated rats. The phytochemical analysis of the extract revealed the presence of antioxidant and phenolics. From the total antioxidant capacity, fraction 6b (ethyl acetate: ethanol) was selected for GC-MS, FTIR and HPLC analysis. The GC-MS analysis of fraction 6b revealed the presence of 14 compounds from which Bis(2-ethylhexyl) phthalate had the highest percentage constituent (48.30%) while while 2-Methyl-Z,Z-3,13-octadecadienol had the lowest relative abundance of 0.14 %. Dichloroacetic acid, tridecyl este, d-Glycero-d-ido-heptose, Phthalic acid, ethyl pentyl ester, Trifluoroacetoxy hexadecane, 2-Hexene, 6-nitro-, 2-Tetradecanol, 3,5-Dimethyl-2-octanol, Cyclopropanecarboxylic acid, 7,11-Hexadecadienal, 9-Octadecenamide, Squalene and E, E-1,9,17-Docasatriene were the other compounds found in the fraction. FTIR spectral analysis of the fraction showed peaks of various functional groups. The HPLC results showed the presence of quercetin in the fraction.

Abstract:Background: The fruit of Schinus terebinthifolia Raddi is used in ethnomedicine for the treatment of different ailments. Materials and Methods: We studied the phytochemical profile of S. terebinthifolia fruit and evaluated its properties, including its antioxidant, anti-inflammatory, analgesic, cytotoxic and antimicrobial activity. Results: Phytochemical investigation of S. terebinthifolia fruit showed an estimated total phenolic content of 67 mg of gallic acid equivalent (GAE)/g, with respect to gallic acid, while the flavonoid content was 26 mg of quercetin equivalent (QE)/g, in terms of quercetin. Bioassay fractionation of methanolic extract of the fruit resulted in the isolation of a new molecule for the first time from this natural source: 4′,methoxy-6-vinyl-7- O-rhamnosyl-dihydromyricetin or 4′,methoxy-6-vinyl-7- O-rhamnosyl-ampelopsin (Compound 8). Seven other phenolic compounds were isolated, such as (1) gallic acid; (2) afzelin; (3) quercetin 3- O- β-D-glucopyranoside; (4) genistein 7- O- α-l-rhamnopyranoside; (5) ellagic acid; (6) kaempferol and (7) quercetin. The new compound was ascertained using UV, NMR and MS spectra and acid hydrolysis analysis. Significant antioxidant, cytotoxic, anti-inflammatory, analgesic and antimicrobial properties were observed in the alcohol extract, as well as proliferation inhibition in MCF-7 human cancer cell lines. Its activity in elevating glutathione levels in diabetic rats could be due to its anti-inflammatory and immunomodulatory properties. Conclusion: The new compound shows immunomodulatory effects that can be utilized to augment host responses, which is important in both cancer and infectious diseases and thus worth further pharmacological investigation.

Abstract:The fruits of Amomum villosum are often considered a medicinal and food homologous material and have been found to have therapeutic effects in chronic enteritis, gastroenteritis, and duodenal ulcer. The aim of this study is to discover the anti-inflammatory active ingredients from dried ripe fruits of A. villosum and to elucidate the molecular mechanisms. We verified that the inhibitory activity of the ethyl acetate extract was superior to Dexamethasone (Dex), so we ultimately chose to study the ethyl acetate extract from the fruits of A. villosum. A total of 33 compounds were isolated from its ethyl acetate extract, including nine known diterpenoids (compounds 1-9), twelve known sesquiterpenoids (compounds 10-21), ten known phenolics (compounds 22, 23, 25-29, 31-33) and two new phenolics (24 and 30). On the basis of chemical evidences and spectral data analysis (UV, ECD, Optical rotation data, 1D and 2D-NMR, HR-ESI-MS, NMR chemical shift calculations), the structures of new compounds were elucidated. Among these compounds, isocoronarin D (5) was found to have good anti-inflammatory activity. Further research has found that isocoronarin D can down-regulate the protein levels of COX2 and NOS2, activate Nrf2/Keap1 and suppress NF-κB signaling pathway in LPS-induced RAW264.7 cells. In addition, isocoronarin D inhibited inflammasome assembly during inflammasome activation by hampering the binding of NLRP3 and ASC. Further evidence revealed that isocoronarin D suppressed the assembly of the NLRP3 inflammasome via blocking the formation of ASC specks. From these results, isocoronarin D may be the important bioactive compound of A. villosum and exhibits anti-inflammatory effects by regulating the NF-κB/Nrf2/NLRP3 axis in macrophages.
In vitro assessment on anti-inflammatory and anti-lipidemic properties of selected plant species

Abstract:Preliminary assessment for anti-inflammatory and anti-lipidemic properties was done with different solvent extracts derived from Urtica urens and Polygonum chinense leaves through in vitro experimentation. To evaluate anti-inflammatory properties, the stability of human red blood cells membranes and the denaturation activity of proteins were assessed. For anti-lipidemic effects, an assay was conducted to measure the inhibition of HMG-CoA reductase. The results of membrane stabilization showed IC50 values of 480.96 ± 0.02 and 319.41 ± 0.19 μg/mL for ethyl acetate extract of U. urens and P. chinense, respectively. The standard drug Diclofenac sodium exhibited IC50 value of 240.37 ± 0.04 μg/mL. For protein denaturation, IC50 values were determined as 221.75 ± 0.2 and 315.76 ± 0.19 μg/mL for U. urens and P. chinense, respectively. The IC50 value of the standard drug was calculated as 126.7 ± 0.34. The IC50 values towards HMG-CoA reductase inhibition were subsequently determined as 29.84 ± 0.35 μg/mL for U. urens and 24.34 ± 0.04 μg/mL for P. chinense against the standard drug Diclofenac sodium (7.52 ± 0.43 μg/mL). GC-MS chromatograms revealed the presence of bioactive compounds in ethyl acetate extract of P. chinense leaves. This work is substantiation for the traditional therapeutic utilization of these extracts.

Anti-Inflammatory Effects and Metabolomic Analysis of Ilex Rotunda Extracted by Supercritical Fluid Extraction

Abstract:Ilex rotunda is a famous medicinal plant with many ethnopharmacological uses. It is traditionally employed for treating inflammation and cardiovascular diseases. In this study, we established green technology to extract the leaves and twigs of I. rotunda. The obtained extracts and their fractions were evaluated for their anti-inflammatory potential. In cytokine assays, the extract, n-hexane (H), methylene chloride (MC), and EtOAc (E) fractions of the twigs of I. rotunda significantly inhibited lipopolysaccharide (LPS)-induced nitric oxide (NO), interleukin (IL)-6, and tumor necrosis factor (TNF)-α production in RAW264.7 macrophages. Furthermore, the extract, H, and MC fractions of the leaves of I. rotunda modulated cytokine expression by downregulating LPS-induced NO, IL-6, and TNF-α production in RAW264.7 macrophages. Western blotting analysis revealed that the extracts and fractions of the leaves and twigs of I. rotunda inhibited inflammatory cytokines by inactivating nuclear factor kappa B (NFκB) action by reducing the phosphorylation of transcript factor (p65) and nuclear factor-kappa B inhibitor alpha (IκBα) degradation, or by inactivating mitogen-activated protein kinase (MAPK) through the p38 or ERK signaling pathways via the active ingredients of the leaves and twigs of I. rotunda. Ultra-high-resolution liquid chromatography–Orbitrap mass analysis (UHPLC–ESI-Orbitrap-MS/MS)-based molecular networking, in cooperation with social open platform-guided isolation and dereplication, led to the identification of metabolites in this plant. Our findings indicate that the leaves and twigs of I. rotunda could be promising candidates for developing therapeutic strategies to treat anti-inflammatory diseases.

Abstract:Radix Tetrastigma (RT) is a medicinal plant and functional food in China, distributed in various places in the south of China. Radix Tetrastigma extract (RTE) from different origins were collected and analyzed for their anti-inflammatory effects. Different RTEs showed different abilities to suppress shape deformation, decrease the nitric oxide (NO) production, inducible nitric oxide synthase (iNOS) and cyclooxygenase-2 (COX-2) expression in RAW 264.7 cells. Subsequently, their bioactive components were compared, and results showed RTEs are rich in flavonoid (85.25-436.70 mg RE/g DW), polysaccharides (100.45-349.26 mg glucose/g DW), phenolic (12.92-225.40 mg GAE/g DW) and protein contents (4.429-7.719 mg/g DW). Principal component analysis (PCA) and correlation studies indicated that anti-inflammatory capacity could be more associated with total flavonoid contents. High-performance liquid chromatography (HPLC) and Ultraperformance liquid chromatography-time-of flight mass spectrometry (UPLC-TOF/MS) analysis were conducted, and results showed that rutin, isoquercitrin, kaempferol-3-O-rutinoside and astragalin were main flavonoid compounds, among them astragalin exhibited a prior protective effect, suggesting it might be responsible for RTE's excellent anti-inflammatory capacity.

Abstract:Chronic noncommunicable diseases (NCDs) are responsible for approximately 74% of deaths globally. Medicinal plants have traditionally been used to treat NCDs, including diabetes, cancer, and rheumatic diseases, and are a source of anti-inflammatory compounds. This study aimed to evaluate the anti-inflammatory effects of Rhus trilobata (Rt) extracts and fractions in lipopolysaccharide (LPS)-induced inflammation models in vitro and in vivo. The aqueous extract (RtAE) and five fractions (F2 to F6) were obtained via C18 solid-phase separation and tested in murine LPS-induced J774.1 macrophages. Key inflammatory markers, such as IL-1β, IL-6, TNF-α, and COX-2 gene expression were measured using RT-qPCR, and PGE2 production was assessed via HPLC-DAD. The in vivo effects were tested in an LPS-induced paw edema model in Wistar rats. Results showed that RtAE at 15 μg/mL significantly decreased IL-1β and IL-6 gene expression in vitro. Fraction F6 further reduced IL-1β, TNF-α, and IL-6 gene expression, COX-2 expression, and PGE2 production. In vivo, F6 significantly reduced LPS-induced paw edema, inflammatory infiltration, and IL-1β and COX-2 protein expression. Chemical characterization of F6 by UPLC/MS-QTOF revealed at least eight compounds with anti-inflammatory activity. These findings support the anti-inflammatory potential of RtAE and F6, reinforcing the medicinal use of Rt.

Abstract:Rhus ruspolii Engl. plant is traditionally used in Ethiopia to treat various diseases. However, the biological and phytochemical properties of the leaves are not well documented. Hence, this study aimed to isolate phytochemicals from R. ruspolii leaves and evaluate their antibacterial and DPPH radical scavenging activities. GC-MS analysis identified 16 compounds from combined fractions 6-10. Chromatographic separation and NMR analysis resulted in the isolation and characterization of palmitic acid (7), 3,4-dihydroxybenzoic acid (17), cupressuflavone (18), amentoflavone (19), shikimic acid (20), avicularin (21), and myricetin-3-O-5''-acetylarabinofuranoside (22). The inhibition zones of extracts (100 mg/mL) and isolated compounds (5 mg/mL) ranged from 8.33 ± 0.50 to 16.33 ± 0.47 mm against all evaluated bacteria. Of all isolated compounds, compounds 18 and 21 showed good activity against Gram-negative (supported by in silico molecular docking studies) and Gram-positive bacteria, respectively. The lowest (49.1 %) and the highest (91.3 %) DPPH radicals were inhibited by combined fractions 6-10 and compound 17, respectively, at 62.5 μg/mL. The SwissADME online analysis showed compounds 17 and 20 have good solubility and permeability. The Pro Tox 3.0 online analysis revealed none of the isolated compounds are fatal if swallowed. Therefore, the findings of this study support the traditional use of the plant for treating bacteria diseases.

Abstract:Introduction Acorus tatarinowii Schott is a traditional Chinese medicine that aromatizes dampness, opens the body and pacifies the mind, and is used medicinally as the dried rhizome of Acorus tatarinowii, family Tenaxaceae. Research has confirmed that it has a strong anti-inflammatory activity, however, its mechanism of anti-inflammatory is still unclear, and we continue to investigate its chemical composition and anti-inflammatory mechanism. Methods The separation and purification of the EtOH extract was carried out by preparative thin layer chromatography and column chromatography. The structures of the isolated compounds were identified by NMR spectroscopy and comparing their spectral data with those previously reported in literature. CCK-8 method was used to evaluate the anti-inflammatory activity of all compounds. The MDA, NO, SOD, IL-10, IL-1β, IL-6 and TNF-α levels were analyzed using ELISA kits. TNF-α and Beclin-1 expression were examined by western blot and immunofluorescence. Results Ten compounds, named cinnamic acid(1), sinapic acid(2), isoferulic acid(3), α-asarone(4), ferulic acid(5), arbutin (6), caffeic acid (7), 3-(3-hydroxyl phenyl)propanol(8), ethyl caffeate(9) and β-asarone(10) were isolated from the rhizome of Acorus tatarinowii Schott. Their structures were elucidated on the basis of extensive spectroscopic analyses, as well as by comparison with literature data. The anti-inflammatory effects of the all compounds were evaluated in lipopolysaccharide(LPS)-induced PC12 cell models in our study. The results of activity test showed that compounds 3 and 6 had good inhibitory effects on 5 μg·mL−1 LPS-induced PC12 cells. The IC50 of compounds 3 and 6 against PC12 cells were 27.41 ± 0.09 µmol·L−1 and 30.19 ± 0.10 µmol·L−1, respectively. Moreover, compounds 3 and 6 showed a significant protective effect on inflammation by increasing the IL-10 and SOD levels, and reducing the TNF-α, IL-6, IL-1β, MDA and NO levels. The most important point is that their anti-inflammatory effect is related to the autophagy factor Beclin-1. Conclusion These findings imply that compounds 3 and 6 exerts anti-inflammatory effects by regulating TNF-α/Beclin-1 pathway, which provide a new basis for the anti-inflammatory activity of Acorus tatarinowii Schott.
