Abstract:Chemical investigation of a co-culture of two endophytic fungi Aspergillus fumigatus D and Fusarium oxysporum R1 from two traditional medicinal plants, Edgeworthia chrysantha Lindl. and Rumex madaio Makino, led to isolation of a new amide 1 and six known compounds, including neovasinin (2), neovasifuranone B (3), N-(2-phenylethyl)acetamide (4), α-linolenic acid (5), α-elaeostearic acid (6), palmitoleic acid (7). On the basis of extensive spectroscopic analysis including 1D and 2D NMR, HR-ESI-MS and optical rotation measurement as well as comparison of literature data, chemical structure of 1 was unambiguous elucidated as (S, E)-methyl-2-(2,4-dimethylhex-2-enamido)acetate. Bioassay results indicated that none of these compounds exhibited strong inhibitory effect on three human pathogens Escherichia coli, Staphyloccocus aureus and Candida albicans with MIC values of ≥25 μM.

Abstract:Six new xanthone dimers, diaporxanthones A-F (1-6), and an unusual xanthone monomer diaporxanthone G (7), in addition to seven known analogues (8-14), were isolated and identified from endophytic Diaporthe goulteri L17 harbored in the fruits of the salt-tolerant plant Vitex trifolia. The chemical structures of these metabolites were elucidated on the basis of nuclear magnetic resonance, high-resolution electrospray ionization mass spectrometry, and reported data in the literature. Their absolute configurations were established by single-crystal X-ray diffraction analysis together with time-dependent density functional theory electronic circular dichroism calculations. Among these compounds, compounds 1 and 6 exhibited moderate antifungal activities against Nectria sp. and Colletotrichum musae and compound 4 showed significant cytotoxicity against all selected five cancer cell lines.

Abstract:Endophytic fungi in medicinal plants are important sources for microorganisms which can produce a variety of bioactive compounds.In this study,an endophytic fungus TMSF169 with antifungal activity was isolated from Ginkgo biloba in Tianmushan Nature Reserve,Zhejiang province.TMSF169 was identified as Penicillium commune based on morphology and ITS rDNA sequence analysis.An antifungal and herbicidal compound TMSF169A was isolated and purified from the fermentation broth of TMSF169 through normal-phase silica gel column chromatography and Sephadex LH-20 gel column chromatography,traced by ultraviolet light or iodine vapor with bioassay-guided fractionation.TMSF169A was elucidated as cyclopaldic acid based on mass spectrometry and nuclear magnetic resonance spectroscopy.Antifungal activity assays showed that the compound had inhibitory to a series of plant pathogenic fungi.The metabolite was also pathogenic to Commelina communis,Digitaria sanguinalis,Amaranthus retroflexus,Alternanthera philoxeroides and Oryza sativa,but not sensitive to Sesamum indicum and Glycine max.All the results demonstrated that the strain TMSF169 deserves to develop further.
Polyhydroxylated sesquiterpenes and ergostane glycosides produced by the endophytic fungus Xylaria sp. from Azadirachta indica

Abstract:The investigation of the metabolites from the endophytic fungus Xylaria sp. YM 311647 in solid fermentation resulted in the isolation of six undescribed compounds, namely xylarioxides A–F, respectively. These included one eremophilane sesquiterpene, three guaiane sesquiterpene glycosides, and two ergostane glycosides. The structures of the compounds were determined by extensive analyses of spectroscopic data, including 1D and 2D NMR, as well as HRESIMS data. The stereochemistry of xylarioxide A was confirmed by X-ray crystallographic analysis. All of the isolated compounds were assayed for their antifungal activities against seven phytopathogenic fungi and two human pathogenic fungi. Among them, xylarioxides A, E and F showed potent activities against the tested phytopathogens. Particularly, xylarioxide E exhibited the highest activity against Gibberella saubinetii, Curvularia lunata, and Colletotrichum gloeosporioides with MIC values of 4, 4, and 8 μg/mL, respectively, which were comparable to the positive control of nystatin. Interestingly, guaiane sesquiterpene glycosides have been rarely reported from fungal sources. Additionally, xylarioxide E represented an unusual naturally occurring 3,4-seco-steroidal glycoside with a seven-membered lactone in ring A.

Bioactive metabolites produced by the endophytic fungus Phomopsis sp. YM355364

Abstract:A new compound, 16-acetoxycytosporone B (1), along with four known ones, dankasterone A (2), dankasterone B (3), 3beta,5alpha,9alpha-trihydroxy-(22E,24R)-ergosta-7,22-dien-6-one (4), and cyclonerodiol oxide (5), were isolated from Phomopsis sp. YM355364, an endophytic fungus of Aconitum carmichaeli. Their structures were characterized by spectral analysis. Compound 2 exhibited significant inhibitory activity against influenza A/Thailand/Kan353/2004(H5N1) pseudovirus with all IC50 value of 3.56 microM. Compounds 1, 2, and 4 showed either moderate or weak antifungal activities against four pathogenic fungi.

Endophytic Paraconiothyrium sp. from Zingiber officinale Rosc. Displays Broad-Spectrum Antimicrobial Activity by Production of Danthron

Abstract:The bioactivity spectrum of fungal endophytes isolated from Zingiber officinale was analyzed against clinical pathogens and against the phytopathogen Pythium myriotylum, which causes Pythium rot in ginger. One of the isolates GFM13 showed broad bioactivity against various pathogens tested including P. myriotylum. The spore suspension as well as the culture filtrate of the endophytic fungal isolate was found to effectively protect ginger rhizomes from Pythium rot. By molecular identification, the fungal endophyte was identified as Paraconiothyrium sp. The bioactive compound produced by the isolate was separated by bioactivity-guided fractionation and was identified by GC-MS as danthron, an anthraquinone derivative. PCR amplification showed the presence of non-reducing polyketide synthase gene (NR-PKS) in the endophyte GFM13, which is reported to be responsible for the synthesis of anthraquinones in fungi. This is the first report of danthron being produced as the biologically active component of Paraconiothyrium sp. Danthron is reported to have wide pharmaceutical and agronomic applications which include its use as a fungicide in agriculture. The broad-spectrum antimicrobial activity of danthron and the endophytic origin of Paraconiothyrium sp. offer immense applications of the study.

Abstract:The endophytic fungal strain ZJDH255 with antifungal activity was isolated from Podocarpus macrophyllus in Jiande of Zhejiang, and was identified as Nigrospora oryzae based on its morphological characters and ITS rDNA sequence. Bioassay showed that the crude extract of the ZJDH255 fermentation broth exhibited strong inhibitory activities against plant pathogenic fungi Botrytis cinerea and Colletotrichum orbiculare. Three antifungal compounds were separated from the crude extract through normal-phase silica gel column chromatography and Sephadex LH-20 column chromatography. They were identified as mellein, griseofulvin and dechlorogriseofulvin based on mass spectrometry and nuclear magnetic resonance spectroscopy.

Abstract:The mangrove ecosystem is a rich resource for the discovery of actinomycetes with potential applications in pharmaceutical science. Besides the genus Streptomyces, Micromonospora is also a source of new bioactive agents. We screened Micromonospora from the rhizosphere soil of mangrove plants in Fujian province, China, and 51 strains were obtained. Among them, the extracts of 12 isolates inhibited the growth of human lung carcinoma A549 cells. Strain 110B exhibited better cytotoxic activity, and its bioactive constituents were investigated. Consequently, three new isoflavonoid glycosides, daidzein-4-(2-deoxy--l-fucopyranoside) (1), daidzein-7-(2-deoxy--l-fucopyranoside) (2), and daidzein-4,7-di-(2-deoxy--l-fucopyranoside) (3) were isolated from the fermentation broth of strain 110B. The structures of the new compounds were determined by spectroscopic methods, including 1D and 2D nuclear magnetic resonance (NMR) and high-resolution electrospray ionization mass spectrometry (HR-ESIMS). The result of medium-changing experiments implicated that these new compounds were microbial biotransformation products of strain M. aurantiaca 110B. The three compounds displayed moderate cytotoxic activity to the human lung carcinoma cell line A549, hepatocellular liver carcinoma cell line HepG2, and the human colon tumor cell line HCT116, whereas none of them showed antifungal or antibacterial activities.

Abstract:Prompted by the pressing necessity to conquer phytopathogenic infections, the antimicrobial compounds were characterized with bioassay-guided method from the ethanol extract derived from the solid-substrate fermentation of Aspergillus sp. IFB-YXS, an endophytic fungus residing in the apparently healthy leave of Ginkgo biloba L. The aim of this work was to evaluate the antimicrobial activity and mechanism(s) of these bioactive compounds against phytopathogens. Among the compounds, xanthoascin (1) is significantly inhibitory on the growth of the phytopathogenic bacterium Clavibacter michiganense subsp. Sepedonicus with a minimum inhibitory concentration (MIC) value of 0.31μg/ml, which is more potent than streptomycin (MIC 0.62μg/ml), an antimicrobial drug co-assayed herein as a positive reference. Moreover, terphenyl derivatives 3, 5 and 6 are also found to be active against other phytopathogens including Xanthomonas oryzae pv. oryzae Swings, Xanthomonas oryzae pv. oryzicola Swings, Erwinia amylovora and Pseudomonas syringae pv. lachrymans etc. The antibacterial mechanism of xanthoascin (1) was addressed to change the cellular permeability of the phytopathogens, leading to the remarkable leakage of nucleic acids out of the cytomembrane. The work highlights the possibility that xanthoascin (1), an analogue of xanthocillin which is used to be an approved antibiotic, may find its renewed application as a potent antibacterial agrichemical. This study contributes to the development of new antimicrobial drugs, especially against C. michiganense subsp. Sepedonicus.

Abstract:Twelve compounds, including four undescribed cytochalasins, xylariachalasins A-D (1-4), four undescribed polyketides (5-8), and four known cytochalasins (9-12), were isolated from the mangrove endophytic fungus Xylaria arbuscula QYF. Their structures and absolute configurations were established by extensive spectroscopic analyses (1D and 2D NMR, HRESIMS), electronic circular dichroism (ECD) calculations, 13C NMR calculation and DP4+ analysis, single-crystal X-ray diffraction, and the modified Mosher ester method. Compounds 1 and 2 are rare cytochalasin hydroperoxides. In bioactivity assays, Compound 2 exhibited moderate antimicrobial activities against Staphylococcus aureus and Candida albicans with MIC values of 12.5 μM for both Compound 10 exhibited significant cytotoxic activity against MDA-MB-435 with an IC50 value of 3.61 ± 1.60 μM.

Abstract:A new endophytic strain of Paraconiothyrium brasiliens LT161 was isolated from the healthy stems of Cinamonum camphora collected from Nanjing, China, and was identified based on morphological characteristics and internal transcribed spacer (ITS) sequence analysis. The culture filtrate and its ethyl acetate extract of strain LT161 showed strong growth inhibition activity in vitro against fungal phytopathogens such as Rizoctonia solani, Alternaria alternate, Glomerella glycines, Phytophthora capsici, Fusarium oxysporum, Fusarium graminearum and Cryphonectria parasitica. The bioactive metabolites in the filtrate were relatively thermally stable, but sensitive to strong alkaline conditions. Simulation test within the dish indicated that the bioactive composition of strain LT161 pose considerable ability in controlling rice sheath blight disease, Fusarium root rot and wheat Alternaria leaf blight. The antifungal compounds in the filtrate of strain LT161 might be novel based on liquid chromatography/mass spectrometry (LC/MS) analysis and comparison to the Syngenta natural product dereplication database. These results suggest that the strain LT161 could be a promising candidate for producing leading compound of pesticide development or used directly as biological control agent in the sustainable agriculture system. [Isolation and identification of an endophytic fungus of Polygonatum cyrtonema and its antifungal metabolites].

Abstract:OBJECTIVE Endophytic fungi in medicinal plants produce a variety of bioactivity compounds. In this study, an endophytic fungus zjqy610 with antifungal activity was isolated from Polygonatum cyrtonema in Zhejiang Qingyuan Baishanzu Mountain nature reserve. METHODS Strain zjqy610 was identified as Penicillium canescens based on the morphology and its rDNA sequence analysis. Three antifungal compounds were isolated from the fermentation broth of zjqy610 through normal-phase silica gel column chromatography and gel (Sephadex LH-20) column chromatography, traced by ultraviolet light or iodine vapor with bioassay-guided fractionation. RESULTS These three compounds were elucidated as o-acetylbenzeneamidinocarboxylic acid (zjqy610B-g-3), griseofulvin (zjqy610D-4) and naphtho [1,2-b] furan-3-carboxylic acid, 4-hydroxy-5-methoxy-2-methyl-(zjqy610F-2) based on mass spectrometry and nuclear magnetic resonance spectroscopy. The antifungal activity assays showed that the three compounds had inhibitory to variety of plant pathogenic fungi. Compound zjqy610D-4 had strong antifungal activity against Botrytis cinerea, Colletotrichum orbiculare, Didymella bryoniae and Sclerotinia sclerotiorum. EC50 was 0.68, 0.38, 0.91 and 0.61 mg/L, respectively. CONCLUSION Zjqy610D-4 is deserved to develop an agricultural antibiotics.

Abstract:Chemical investigation of the culture extract of an endophyte Xylaria curta YSJ-5 from Alpinia zerumbet (Pers.) Burtt. et Smith resulted in the isolation of eight previously undescribed compounds including five eremophilane sesquiterpenes xylarcurenes A-E, one norsesquiterpene xylarcurene F, and two α-pyrone derivatives xylarpyrones A-B together with eight known related derivatives. Their chemical structures were extensively established based on the 1D- and 2D-NMR spectroscopic analysis, modified Mosher's method, electronic circular dichroism calculations, single-crystal X-ray diffraction experiments, and the comparison with previous literature data. All these compounds were tested for in vitro cytotoxic, anti-inflammatory, α-glucosidase inhibitory, and antibacterial activities. As a result, 6-pentyl-4-methoxy-pyran-2-one was disclosed to display significant antibacterial activity against Staphylococcus aureus and methicillin-resistant S. aureus with minimal inhibitory concentration value of 6.3 μg/mL.

Abstract:Five eremophilane sesquiterpenes including three new ones, named paraconions A−C (1−3), were isolated from an endophytic fungus, Paraconiothyrium sp. from Artemisia selengensis. The structures of these new compounds were established based on spectroscopic methods, including nuclear magnetic resonance (NMR), ultraviolet (UV), and infrared (IR) spectroscopy, as well as high resolution electrospray ionization mass spectrometry (HR‐ESI‐MS). An anti‐inflammatory assay indicated that paraconion B (2) inhibited lipopolysaccharide‐induced nitric oxide (NO) production in RAW 264.7 cells, with an IC50 value of 51.7 μM. The compounds discovered in this study will enrich the structural types of secondary metabolites of the endophytic fungus Paraconiothyrium sp. and can be developed as lead compounds of anti‐inflammatory drugs.

Abstract:Six new sesquiterpenes, fusarchlamols A-F ( 1 , 2 , 4 – 7 ); one new natural product of sesquiterpenoid, methyltricinonoate ( 3 ); and ten known compounds were found from Fusarium sp. cultured in two different media by the one strain many compounds strategy. The compounds ( 1 , 2 , and 4 – 11 ) were isolated from Fusarium sp. in PDB medium, and compounds ( 3–5 , 8 , and 10 – 17 ) were discovered from Fusarium sp. in coffee medium. Additionally, the configuration of 8 was first reported in the research by Mosher's method. The structures were established by 1D, 2D NMR, mass spectrometry, calculated ECD spectra and Mosher's method. Compounds 1 , 2 , 6 / 7 , 12 , and 16 indicated significant antifungal activities against the phytopathogen Alternaria alternata isolated from Coffea arabica with MICs of 1 μg/mL. The investigation on the anti-phytopathogen activity of metabolites can provide lead compounds for agrochemicals.

Abstract:The fungus Emericella sp. XL029 isolated from leaves of Panax notoginseng was investigated for agents with potential antibacterial and antifungal activities using a one strain-many compounds (OSMAC) strategy. Fifteen compounds, including seven undescribed structures, were obtained from this species. Their structures were confirmed by extensive spectroscopic data, single-crystal X-ray crystallography and quantum chemistry calculations. Emerlactam A exhibited better antibacterial activity against multidrug-resistant Enterococcus faecium and antifungal activity against Helminthosporium maydis, with an MIC value of 12.5 μg/mL. Quiannulatic acid displayed significant antibacterial activity against multidrug-resistant Enterococcus faecium and multidrug-resistant Enterococcus faecalis with MIC values of 1.56 μg/mL and 3.13 μg/mL, respectively. 5-alkenylresorcinol exhibited significant antifungal activity against all tested phytopathogenic fungi with MIC values ranging from 6.25 to 12.5 μg/mL.

Abstract:Three new metabolites, microsphaeropsones A-C (1-3) with a unique oxepino[2,3-b]chromen-6-one (ring-enlarged xanthone) skeleton, were isolated from the endophytic fungus Microsphaeropsis species, co-occurring with their putative biogenetic anthraquinoide precursors citreorosein (4) and emodin (5). From another Microsphaeropsis species, large amounts of fusidienol A (8 a), smaller amounts of emodin (5), the known aromatic xanthones 9 a and 9 b, the new 3,4-dihydrofusidienol A (8 b), and the new aromatic xanthone 9 c were isolated. The endophyte Seimatosporium species produced a new aromatic xanthone, seimatoxanthone A (10), and 3,4-dihydroglobosuxanthone A (12), closely related to alpha-diversolonic ester (13) from Microdiplodia sp.. The structures were determined mainly by extensive 1D and 2D NMR experiments and supported by X-ray single-crystal analysis of 1 and the oxidation product 7. The absolute configurations of the microsphaeropsones A-C (1-3) were established by comparison of the electronic and vibrational circular dichroism (ECD and VCD) spectra of 1 with time-dependent DFT (TDDFT) and DFT calculations by using either the solid-state structures or DFT-optimized geometries as inputs. Preliminary studies indicated that 1, 2, and enone 7 showed antibacterial, fungicidal, and algicidal properties.
Characteristics and Outcomes of Patients with Pneumocystis jirovecii Pneumonia Who Were Initiated on Antiretroviral Therapy While Hospitalized: A Preliminary Study

Abstract:Abstract Background Pneumocystis jirovecii pneumonia (PJP) is the most frequent and severe respiratory infection in patients with acquired immunodeficiency syndrome (AIDS) with associated 20% mortality. There have been conflicting data regarding the optimal time to initiate antiretroviral therapy (ART) in these patients with most data suggesting benefit for early initiation. The objectives of this study were to compare patients with PJP and AIDS who were initiated on ART while hospitalized compared with those who were not; and to evaluate the association between inpatient initiation of ART and survival. Methods We conducted a retrospective chart review of patients 18 years or older with PJP and AIDS who were not on ART prior to admission. We collected demographic, laboratory and clinical information. SPSS was used to compare the two groups: those who initiated ART while inpatient (ART) vs.. those who did not (NoART) Results Of the 64 patients included in this study, 25 [39%] were in the ART group, 19 [27%] required intensive care unit (ICU), and 16 [25%] required mechanical ventilation (MV). There were no differences in age, gender, race/ethnicity, and smoking between the ART and NoART groups. A higher percentage of patients in the ART group received corticosteroids (96% vs. 72%; P = 0.020), required MV (48% vs.10%; P = 0.001), and ICU admission (60% vs. 10%; P = 0.000) than in the NoART group respectively. There were no differences in the ART and NoART groups in regards to ICU stay (4 vs. 0.5 days; P = 1.000) and APACHE II scores (15.2 vs. 10.7; P = 0.17). A total of 9 (14%) patients died while in the hospital 6 (24%) in ART vs.. 3 (8%) in NoART (P = 0.137). Conclusion Patients with PJP pneumonia who were initiated on ART while inpatient were more likely to require ICU admission, corticosteroids, and mechanical ventilation. There were no differences in APACHE II scores, CD4 count and mortality between those who initiated ART while inpatients vs. those who did not. Further studies with larger sample size are needed to evaluate the association between inpatient initiation of ART and survival. Disclosures All authors: No reported disclosures.
