Abstract:Achieving nanometer-scale resolution remains challenging in expansion microscopy due to photon loss. To address this concern, here we develop a multi-color expansion stimulated emission depletion technique based on small-molecule probes to realize high labeling density and intensity. Our method substantially lowers the barrier to visualizing diverse intracellular proteins and their interactions in three dimensions. It enables us to achieve sub-10-nm resolution in structures such as microfilaments, lysosomes, and mitochondria, providing new insights into cell biology.

Abstract:Lysosomes and mitochondria play an important role in maintaining cell homeostasis. Visualizing the long‐term activities of lysosomes and mitochondria on the nanometer scale in live cells is essential for further understanding their functions but remains challenging due to the limitations of existing fluorescent probes, such as aggregation‐caused quenching (ACQ) effect, limited signal‐to‐noise ratio from fluorescence "always on" in the process of targeting organelle and poor photobleaching resistance. Herein, two efficient red‐emitting aggregation‐induced emission (AIE) luminogens are reported, which showed "off‐on" fluorescence characteristic and specific lysosomes as well as mitochondria targeting capability. Owing to their AIE characteristics, a Stokes' shift larger than 100 nm, good biocompatibility, and excellent photostability, the AIE luminogens have been successfully utilized for high fidelity imaging of lysosomes and mitochondria. By virtue of these two probes, stimulated emission depletion (STED) images of dynamic lysosomal fusion and mitochondrial fission with a high resolution of 65.6 nm are obtained. Furthermore, the interactions between lysosomes and mitochondria in the process of mitophagy are recorded. This study also provides practical guidance for designing specific organelle targeting probes to support live cell dynamic super‐resolution imaging.

Abstract:Characterizing the long-term nanometer-scale interactions between lysosomes and mitochondria in live cells is essential for understanding their functions but remains challenging due to limitations of the existing fluorescent probes. Here, we develop cell-permeable organic fluorescent probes for lysosomes with excellent specificity and high photostability. We also use an existing Atto 647N dye with high brightness and excellent photostability to achieve specific labeling of mitochondria in live cells. Using these probes, we obtain dual-color structured illumination microscopy (SIM) images of dynamic physical lysosome-mitochondrion interactions in live cells at an ~90-nm resolution over a long time course of ~13 min. We successfully record the consecutive dynamic processes of lysosomal fusion and fission, as well as four types of physical lysosome-mitochondrion interactions by super-resolution imaging. Our probes provide an avenue for understanding the functions and the dynamic interplay of lysosomes and mitochondria in live cells.

Abstract:Background: The interaction between lysosomes and mitochondria includes not only mitophagy but also mitochondrion–lysosome contact (MLC) that enables the two organelles to exchange materials and information. In our study, we synthesised a biosensor with fluorescence characteristics that can image lysosomes for structured illumination microscopy and, in turn, examined morphological changes in mitochondria and the phenomenon of MLC under pathological conditions. Methods: After designing and synthesising the biosensor, dubbed CNN, we performed an assay with a Cell Counting Kit-8 to detect CNN’s toxicity in relation to H9C2 cardiomyocytes. We next analysed the co-localisation of CNN and the commercial lysosomal probe LTG in cells, qualitatively analysed the imaging characteristics of CNN in different cells (i.e. H9C2, HeLa and HepG2 cells) via structured illumination microscopy and observed how CNN entered cells at different temperatures and levels of endocytosis. Last, we treated the H9C2 cells with mannitol or glucose to observe the morphological changes of mitochondria and their positions relative to lysosomes. Results: After we endocytosed CNN, a lysosome-targeted biosensor with a wide, stable pH response range, into cells in an energy-dependent manner. SIM also revealed that conditions in high glucose induced stress in lysosomes and changed the morphology of mitochondria from elongated strips to round spheres. Conclusion: CNN is a new tool for tracking lysosomes in living cells, both physiologically and pathologically, and showcases new options for the design of similar biosensors.

Abstract:Lysosomes are multifunctional organelles involved in macromolecule degradation, nutrient sensing and autophagy. Live imaging has revealed lysosome subpopulations with dynamics and characteristic cellular localization. An as-yet unanswered question is whether lysosomes are spatially organized to coordinate and integrate their functions. Combined with super-resolution microscopy, we designed a small organic fluorescent probe, TPAE, that targeted lysosomes with a large Stokes shift. When we analyzed the spatial organization of lysosomes against mitochondria in different cell lines with this probe, we discovered different distance distribution patterns between lysosomes and mitochondria during increased autophagy flux. By using SLC25A46 mutation fibroblasts derived from patients containing highly fused mitochondria with low oxidative phosphorylation, we concluded that unhealthy mitochondria redistributed the subcellular localization of lysosomes, which implies a strong connection between mitochondria and lysosomes.

Abstract:The background barrier restricts the dimensionality of live-cell single-molecule localization super-resolution imaging. Ideally, a probe exclusively turned on by its target, without any nonspecific fluorescence signals from off-target molecules, constitutes a practical solution to surpass this barrier. Yet, few such fluorophores have been developed. A lysosome with a unique acidic lumen was chosen as the target for demonstrating the concept advantage. A representative lyso-tracker Lyso-R (piperazine rhodamine) with high brightness has been spirocyclized with o-phenylenediamine to form Lyso-Ropa. This probe shifted its bright-dark spirocyclization balance to a strong acidity domain (pKa = -0.18). Consequently, under no-wash conditions, Lyso-Ropa showed almost undetectable background photons (only one-sixtieth of that of Lyso-R) in a neutral cellular environment, and it formed sparsely brightened molecules at a low ratio (∼1 × 10-3%) in lysosomes. This background-free probe enabled super-resolution imaging and modeling of live-cell lysosomes in four dimensions at 2 s resolution, with quantitative determination of lysosomal volume expansion and deformation at starvation. Our molecular approach sheds new light on surpassing the background barrier for multidimensional super-resolution imaging.

Abstract:Eating attitudes and behaviors, body image, and psychological functioning were evaluated in 98 female college students: 36 African-Americans, 34 Asian-Americans, and 28 Caucasians. African-Americans had significantly higher body mass index than either Asian-American or Caucasians. In contrast, Caucasians reported greater levels of disordered eating and dieting behaviors and attitudes and greater body dissatisfaction than did Asian-Americans and African-Americans who differed little on these measures. The nature of variability in these eating behaviors and attitudes and body image was also examined within each of the three groups. A generally consistent pattern emerged within each racial group: low self-esteem and high public self-consciousness were associated with greater levels of problematic eating behaviors and attitudes and body dissatisfaction. A history of being teased about weight and size was associated with problematic eating behaviors and attitudes and body dissatisfaction in African-Americans and Caucasians but not in Asian-Americans. The findings suggest that there exist important racial differences on various aspects of eating, dieting, and body image in college women. Contrary to hypothesis, the degree of acculturation and assimilation within the African-American and Asian-American groups was unrelated to variability in these domains.

Abstract:Nowadays, more and more studies have linked the abnormal expression of active molecules in organelles with the occurrence of diseases, so there is an urgent need to develop tools for detecting active molecules in specific organelles. However, the recognition receptors of most organelle-targeting probes currently developed always remain active, which easily causes them to react with the analyte in the cytoplasm, thus misjudging the role of the analyte in the physiological and pathological processes. Therefore, it is of great significance to develop a new strategy for the design of probes capable of high-fidelity imaging of the analyte in specific organelles. Herein, we propose a new strategy that the activation of recognition receptors that can be triggered by the microenvironment of targeting organelles. Based on this strategy, we develop a novel lysosome-targeting fluorescent probe (Lyso-SO2) for imaging of sulfur dioxide (SO2) with high-fidelity in lysosomes. The inert probe is activated by the acidic environment in the lysosome and then responds quickly (<2 s) and sensitively (LOD = 0.34 μM) to SO2. This paradigm by taking full advantage of the features of the organelle microenvironment provides a promising methodology for developing organelle-targeting probes for high-fidelity imaging.

Abstract:Zinc (Zn) based fluorescent metal complexes have gained increasing attention due to their non-toxicity and high brightness with marked fluorescence quantum yield (QY). However, they have rarely been employed in super-resolution microscopy (SRM) to study live cells and in vivo dynamics of lysosomes. Here, we present an NIR emissive highly photostable Zn-complex as a multifaceted fluorescent probe for the long-term dynamical distribution of lysosomes in various cancerous and non-cancerous cells in live condition and in vivo embryogenic evolution in Caenorhabditis elegans (C. elegans). Apart from the normal fission, fusion, and kiss & run, the motility and the exact location of lysosomes at each point were mapped precisely. A notable difference in the lysosomal motility in the peripheral region between cancerous and non-cancerous cells was distinctly observed. This is attributed to the difference in viscosity of the cytoplasmic environment. On the other hand, along with the super-resolved structure of the smallest size lysosome (∼77 nm) in live C. elegans, the complete in vivo embryogenic evolution of lysosomes and lysosome-related organelles (LROs) was captured. We were able to capture the images of lysosomes and LROs at different stages of C. elegans, starting from a single cell and extending to a fully matured adult animal.

Abstract:Nitric oxide (NO), as a special biological small molecule, plays an important role in physiology and pathology. Studies have revealed a strong correlation between human illness and the amount of NO in lysosomes. In this research, we synthesized a BODIPY-based fluorescent probe BML that can be localized in the lysosome to detect NO. The secondary amine in the BML structure can be N- nitrosation reaction with NO in the lysosome, preventing the initial photo-induced electron transfer (PET) process and leading to the creation of the highly luminescent BML-NO. The reaction mechanism was proved in the high-resolution mass spectrometry and DFT simulation calculations. Additionally, BML was discovered to have some evident benefits, such as fast detection time (3.5 min), low detection limit (LOD = 10.3 nM), and the resistance to interference and acidity, which are essential for the detection of NO in lysosomes. Furthermore, the BML was effectively employed to detect endogenous and exogenous nitric oxide in living cells and zebrafish.

Abstract:Reactive oxygen species (ROS) are extremely important for various biological functions. Lysosome plays key roles in cellular metabolism and has been known as the stomach of cells. The abnormalities and malfunctioning of lysosomal function are associated with many diseases. Accordingly, the quantitative monitoring and real-time imaging of ROS in lysosomes are of great interest. In recent years, with the advancement of fluorescence imaging, fluorescent ROS probes have received considerable interest in the biomedical field. Thus far, considerable efforts have been undertaken to create synthetic fluorescent probes for sensing ROS in lysosomes; however, specific review articles on this topic are still lacking. This review provides a general introduction to fluorescence imaging technology, the sensing mechanisms of fluorescent probes, lysosomes, and design strategies for lysosome-targetable fluorescent ROS probes. In addition, the latest advancements in organic small-molecule fluorescent probes for ROS detection within lysosomes are discussed. Finally, the main challenges and future perspectives for developing effective lysosome-targetable fluorescent ROS probes for biomedical applications are presented.

Abstract:Combining probes with advanced imaging techniques has enabled single-organelle localization-based superresolution imaging, revolutionizing our ability to investigate subcellular molecules with unprecedented precision. This review presents an in-depth discussion on the design and applications of small-molecule probes for specific small and large molecules, alongside advancements in super-resolution imaging. This review discusses super-resolution imaging, emphasizing its operational principles, technological progress, and unique benefits. We also outline innovative probe design strategies that may enhance our understanding of subcellular molecular dynamics and their single-organelle localization. Probe design strategies for diverse intra-organelle molecules are also provided, stressing the need to consider elements such as photostability, signal-to-noise ratio, and targeted binding. Challenges associated with this field, potential solutions, and an outlook are also discussed in detail. By integrating probe design with state-of-the-art imaging technologies based on single-organelle localization, this review provides a path toward remarkable revelations in single-organelle functionalities at the most intricate level of molecular interactions.
Aggregation-induced Emission Luminogen-Assisted Stimulated Emission Depletion Nanoscopy for Super-Resolution Mitochondrial Visualization in Live Cells

Abstract:Aggregation-induced emission luminogens (AIEgens) are fluorescent agents that are ideal for bioimaging and have been widely used for organelle targeting, cellular mapping, and tracing. Owing to their promising characteristics, AIEgen-based nanoparticles have recently been used for the stimulated emission depletion (STED) super-resolution imaging of fixed cells. In the present study, and for the first time, we used an AIEgen for dynamic STED nanoscopic imaging of a specific organelle in live cancer cells. TPA-T-CyP is a synthetic red&NIR-emitting luminogen with AIE features that can spontaneously and specifically aggregate on mitochondria without the need for encapsulation or surface modification. The STED efficiency of aggregated TPA-T-CyP can reach more than 80%, and super-resolution imaging of TPA-T-CyP-stained mitochondria in live HeLa cells is possible, with a lateral spatial resolution of 74 nm. We found that TPA-T-CyP enabled the dynamic visualization of mitochondria, and the motion, fusion, and fission of mitochondria were clearly observable on a super-resolution scale. AIEgen-based super-resolution organelle visualization has great potential for many basic biomedical studies.

Abstract:Fluorescence probes have great potential to empower bioimaging, precision clinical diagnostics and surgery. However, current probes are limited to in vivo high-contrast diagnostics, due to the substantial background interference from tissue scattering and nonspecific activation in blood and normal tissues. Here, we developed a kind of cell endocytosis-activated fluorescence (CEAF) probe, which consists of a hydrophilic polymer unit and an acid pH-sensitive small-molecule fluorescent moiety that operates in the "tissue-transparent" second near-infrared (NIR-II) window. The CEAF probe stably presents in the form of quenched nanoaggregates in water and blood, and can be selectively activated and retained in lysosomes through cell endocytosis, driven by a synergetic mechanism of disaggregation and protonation. In vivo imaging of tumor and inflammation with a passive-targeting and affinity-tagged CEAF probe, respectively, yields highly specific signals with target-to-background ratios over 15 and prolonged observation time up to 35 hours, enabling positive implications for surgical, diagnostic and fundamental biomedical studies.

Abstract:The development of methods for real-time monitoring the level of Cys in biological system is a topic of continued importance. Furthermore, the probes emitting in the NIR region are favorable for cell imaging because of their low phototoxcity and high signal-to-noise ratio (S/N) in living cells. In the paper, we have developed a novel lysosome-targeting NIR xanthene-benzothiozolium dye BHCy-OH by taking advantage of the heteroatom replacement strategy to regulate the absorption and emission wavelengths of the known semi-heptamethine dye. Subsequently, the NIR probe BHCy-A with high sensitivity for Cys has been synthesized. The dye BHCy-OH and the Cys-selective probe BHCy-A emitted fluorescence at 735 nm in PBS buffer, and are lysosome-targetable in living cells. Because the lysosome-targeting abilities of them do not demand proton participation, we believe that they are very useful for investigation the function of lysosomes and for interpreting the function of lysosomal Cys in living-cell imaging.

Abstract:High-fidelity imaging and long-term visualization of lysosomes are pivotal factors in the functional assessment of lysosomes, which perform an instrumental role in the physiological activity of cells. However, commercial probes have great limitations in lysosome exploration resulting from the aggregation-caused quenching effect as well as photobleaching instability and small Stokes shift. Therefore, we constructed a novel probe named TTAM with triphenylamine as the matrix and morpholine ring as the targeting group. In contrast with commonly accessible Lyso-tracker Red, TTAM has the merits of aggregation-induced emission effect, extremely high quantum yields (51.57 % solid-state) as well as fluorescence intensity, significant photostability, and high resolution. These properties make it ideal for imaging and activity monitoring lysosomes, which provides a powerful condition for bio-imaging.

The nanoprobe comprised a 16 nm spherical gold nanoparticle as the core and an enzyme-responsive oligomer of fluorescein-conjugated oligo(4-vinyl-phenyl phosphate) as the shell. Due to quenching of the core gold nanoparticle, the nanoprobe was non-fluorescent. After incubation with cancer cells, the nanoprobe was rapidly internalized via scavenger receptor-mediated endocytosis and significantly shuffled into the lysosome. The nanoprobe specifically lighted up the lysosome owing to lysosome-induced fluorescence enhancement. Specifically, digestive inclusions in the lysosome hydrolyzed and released gold-quenched fluorescein molecules, leading to significant augmentation of fluorescence. On account of specific lysosomal labeling, the nanoprobe effectively facilitated imaging of a 4–6 μm anterograde trafficking event of the lysosome from the perinuclear region to the cell surface when an acidic extracellular environment developed. Our findings collectively highlight the use of nanoprobes for lysosomal imaging.
Super-Resolution Fluorescence Imaging of Organelles in Live Cells with Photoswitchable Membrane Probes

Abstract:Imaging membranes in live cells with nanometer-scale resolution promises to reveal ultrastructural dynamics of organelles that are essential for cellular functions. In this work, we identified photoswitchable membrane probes and obtained super-resolution fluorescence images of cellular membranes. We demonstrated the photoswitching capabilities of eight commonly used membrane probes, each specific to the plasma membrane, mitochondria, the endoplasmic recticulum (ER) or lysosomes. These small-molecule probes readily label live cells with high probe densities. Using these probes, we achieved dynamic imaging of specific membrane structures in living cells with 30–60 nm spatial resolution at temporal resolutions down to 1–2 s. Moreover, by using spectrally distinguishable probes, we obtained two-color super-resolution images of mitochondria and the ER. We observed previously obscured details of morphological dynamics of mitochondrial fusion/fission and ER remodeling, as well as heterogeneous membrane diffusivity on neuronal processes.
