Abstract:Premise Seed germination involves risk; post‐germination conditions might not allow survival and reproduction. Variable, stressful environments favor seeds with germination that avoids risk (e.g., germination in conditions predicting success), spreads risk (e.g., dormancy), or escapes risk (e.g., rapid germination). Germination studies often investigate trait correlations with climate features linked to variation in post‐germination reproductive success. Rarely are long‐term records of population reproductive success available. Methods Supported by demographic and climate monitoring, we analyzed germination in the California winter‐annual Clarkia xantiana subsp. xantiana. Sowing seeds of 10 populations across controlled levels of water potential and temperature, we estimated temperature‐specific base water potential for 20% germination, germination time weighted by water potential above base (hydrotime), and a dormancy index (frequency of viable, ungerminated seeds). Mixed‐effects models analyzed responses to (1) temperature, (2) discrete variation in reproductive success (presence or absence of years with zero seed production by a population), and (3) climate covariates, mean winter precipitation and coefficient of variation (CV) of spring precipitation. For six populations, records enabled analysis with a continuous metric of variable reproduction, the CV of per‐capita reproductive success. Results Populations with more variable reproductive success had higher base water potential and dormancy. Higher base water potential and faster germination occurred at warmer experimental temperatures and in seeds of populations with wetter winters. Conclusions Geographic variation in seed germination in this species suggests local adaptation to demographic risk and rainfall. High base water potential and dormancy may concentrate germination in years likely to allow reproduction, while spreading risk among years.

Abstract:Abstract Seed germination responses for most narrow-range endemic species are poorly understood, imperilling their conservation management in the face of warming and drying terrestrial ecosystems. We quantified the realized microclimatic niches and the hydrothermal germination thresholds in four threatened taxa (Tetratheca erubescens, Tetratheca harperi, Tetratheca paynterae subsp. paynterae and Tetratheca aphylla subsp. aphylla) that are restricted to individual Banded Ironstone Formations in Western Australia. While T. aphylla subsp. aphylla largely failed to germinate in our trials, all other species demonstrated extended hydrothermal time accumulation (186–500°C MPa days), cool minimum temperatures (7.8–8.5°C), but broad base water potential thresholds (−2.46 to −5.41 MPa) under which germination occurred. These slow germination dynamics are suggestive of cool and wet winter months, where soil moisture is retained to a greater capacity in local microsites where these species occur, rather than the warmer and drier conditions in the surrounding arid environment. Hydrothermal time-to-event modelling showed that each species occupied unique hydrothermal germination niches, which correspond with the microclimatic differences the species are exposed to. Our results provide a baseline understanding for environmental and germination thresholds that govern the recruitment, and ultimately the population structure and persistence, of these short-range endemic plants. In addition, our results can aid future conservation, as well as restoration actions such as translocation to bolster population numbers and to mitigate against losses due to anthropogenic disturbance and global environmental change.

Abstract:Climate variability is expected to increase due to climate change, with projected increases in temperature and erratic rainfall patterns. These changes will alter the environmental cues sensed by seeds, and therefore will impact plant recruitment. This study investigated the effects of seed functional traits (germinability, germination time, synchrony and seed mass) on germination responses of several sub-tropical native Australian plant species under different environmental factors (water stress, salinity and pH). The effect of a hot water pre-treatment was also tested on Fabaceae seeds with known physical dormancy. Seed traits, environmental factors and seed pre-treatments had significant effects on final germination percentage and germination time. Seed mass and time to 50% germination (t 50 ) were also positively correlated. In contrast, pH did not affect germination and there was no interaction between pH and any of the measured seed functional traits. Some species showed a high thermal tolerance to germination and germination was indifferent to light conditions for all species. Results showed that certain seed functional traits interact with environmental factors to influence germination percentage and time. These findings highlight the importance of considering seed functional traits when determining a species germination response under a changing climate. In addition, the findings provide important knowledge to better guide seed-based land restoration programmes.

Abstract:Understanding plant responses to temperature is critical for predicting their vulnerability to global warming and for planning management responses. Germination is a key life‐stage, strongly regulated by temperature, that affects the potential for plant populations to persist. Here, we compared the thermal germination niches of three unrelated, declining woodland forb species – Arthropodium fimbriatum (Asparagaceae), Bulbine bulbosa (Asphodelaceae), Microseris walteri (Asteraceae) – across common temperature and precipitation gradients, to characterize the relationships with home‐site climate, and associated implications for ecological restoration in a changing climate. Open‐pollinated seed were sampled from 14–15 populations per species across an aridity gradient in south‐eastern Australia. Germination responses for each population were tested in controlled temperature cabinets under five temperature regimes encompassing contemporary and projected future temperatures. Optimum germination temperature and thermal germination niche were characterized and assessed for associations with home‐site climate and potential germination under projected climate change. The three species showed significant intra‐specific variation in the thermal germination niche. Optimum germination temperature was correlated with home‐site climate, suggesting adaptive variation among populations in germination requirements. However, the pattern of variation in optimum germination across the environmental gradients was not always consistent among the species. Future temperatures projected under climate change tended to be outside the current thermal germination niche for all species, indicating potential benefits of incorporating pre‐adapted populations in ecological restoration. Climate‐related intra‐specific variation in the thermal germination niche provides evidence for local adaptation to climate in all three forb species and suggests potential differences among populations in vulnerability to global warming. These results emphasize the importance of understanding the extent of intra‐specific variation in key life history traits to better manage and conserve populations and restore their ecosystems as climates change.

Distinct germination response of endangered and common arable weeds to reduced water potential

Abstract:Arable weeds are one of the most endangered species groups in Europe. Modern agriculture and intensive land-use management are the main causes of their dramatic decline. However, besides the changes in land use, climate change may further challenge the adaptability of arable weeds. Therefore, we investigated the response pattern of arable weeds to different water potential and temperature regimes during the phase of germination. We expected that endangered arable weeds would be more sensitive to differences in water availability and temperature than common arable weeds. To this end, we set up a climate chamber experiment where we exposed seeds of five familial pairs of common and endangered arable weed species to different temperatures (5/15, 10/20 °C) and water potentials (0.0 to -1.2 MPa). The results revealed a significant relationship between the reaction of arable weed species to water availability and their Red List status. The effects of reduced water availability on total germination, mean germination time and synchrony were significantly stronger in endangered than in common arable weeds. Therefore, global climate change may present a further threat to the survival of endangered arable weed species.

Abstract:We investigated the effects of soil water potential (SWP) and temperature on seed germination of six coexisting species of an inter-tropical desert. These species include three life-forms; the shrubs Cercidium praecox andProsopis laevigata ; the columnar succulents Neobuxbaumia tetetzo and Pachycereus hollianus; and the arborescent semi-succulents Beaucarnea gracilis and Yucca periculosa. In the six species germinability increased and germination time and speed of germination (t50) decreased as SWP decreased. The SWP treatments were 0 MPa, −0·12 MPa, −0·2 MPa, −0·41 MPa and −0·66 MPa. There was, however, a SWP threshold below which germination time and t50increased and germinability decreased. The shrubs had the highest germinability whereas the columnar succulents had the lowest. The shrubs also had shorter germination time andt50 than arborescent semi-succulents whereas seeds of the columnar succulents were indeterminate. In all species except P. laevigata germinability increased and the germination time and t50decreased as temperature increased. The temperature treatments were 12°C, 20°C and 26°C. The shrubs had the shortest t50and germination time and the highest germinability at all temperatures. Arborescent semi-succulents had the lowest germinability and longest germination time and t50at the three temperatures treatments. Our results support the hypothesis that in desert environments different plant life-forms utilise different germination strategies to persist.

Abstract:The seasonal timing of life history transitions is often critical to fitness, and many organisms rely upon environmental cues to match life cycle events with favorable conditions. In plants, the timing of seed germination is mediated by seasonal cues such as rainfall and temperature. Variation in cue responses among species can reflect evolutionary processes and adaptation to local climate and can affect vulnerability to changing conditions. Indeed, climate change is altering the timing of precipitation, and germination responses to such change can have consequences for individual fitness, population dynamics, and species distributions. Here, we assessed responses to the seasonal timing of germination‐triggering rains for eleven species spanning the Streptanthus/Caulanthus clade (Brassicaceae). To do so, we experimentally manipulated the onset date of rainfall events, measured effects on germination fraction, and evaluated whether responses were constrained by evolutionary relationships across the phylogeny. We then explored the possible consequences of these responses to contemporary shifts in precipitation timing. Germination fractions decreased with later onset of rains and cooler temperatures for all but three Caulanthus species. Species' germination responses to the timing of rainfall and seasonal temperatures were phylogenetically constrained, with Caulanthus species appearing less responsive. Further, four species are likely already experiencing significant decreases in germination fractions with observed climate change, which has shifted the timing of rainfall towards the cooler, winter months in California. Overall, our findings emphasize the sensitivity of germination to seasonal conditions, underscore the importance of interacting environmental cues, and highlight vulnerability to shifting precipitation patterns with climate change, particularly in more northern, mesic species.

Abstract:Plant abundance and distribution are regulated by subtle changes in ecological factors, which are becoming more frequent under global climate change. Species with a higher tolerance to such changes, especially during early lifecycle stages, are highly likely to endure climate change. This study compared the germination adaptability of Halopeplis amplexicaulis and H. perfoliata, which differ in life-form and grow in different environments. Optimal conditions, tolerances and the biochemical responses of seeds to osmotic stresses were examined. Seeds of H. perfoliata germinated in a wider range of temperature regimes and were more tolerant to osmotic stresses than H. amplexicaulis seeds. Neither NaCl nor PEG treatment invoked the H2O2 content in germinating seeds of the tested species. Consequently, unaltered, or even decreased activities of H2O2 detoxification enzymes and non-enzymatic antioxidants were observed in germinating seeds in response to the aforementioned stresses. High and comparable levels of recovery from isotonic treatments, alongside a lack of substantial oxidative damage indicated that the osmotic stress, rather than the ionic toxicity, may be responsible for the germination inhibition. Hence, rainy periods, linked to water availability, may act as a key determinant for germination and H. perfoliata could be less affected by global warming owing to better germinability under high temperatures compared with H. amplexicaulis. Such studies involving biochemical analysis coupled with the germination ecology of congeneric species, which differ in life-form and occurrence are scarce, therefore are important in understanding the impacts of global changes on species abundance/distribution.

Abstract:Dormancy in seeds is a key persistence mechanism for many flowering plants. Physically dormant (PY) seeds have water impermeable seed coats, and in fire-prone systems a common mechanism for dormancy release is fire-induced soil heating. However, the thermal thresholds innate to seeds with PY may be influenced by vegetation, climate, and fire regimes, varying substantially between populations of the same species. To investigate intraspecific variation of thermal thresholds in PY seeds, we sampled obligate seeding Acacia pulchella (Fabaceae) which produces PY seeds. Sampling was undertaken from 13 populations across a climate gradient of rainfall and temperature, and between two vegetation communities in fire-prone Mediterranean-type ecosystems of south-west Western Australia. To test a range of weather and fire-induced soil heating dormancy-break scenarios, we conducted dry heat shock experiments between 40 and 140 °C for 10 min and scored germination for 16 weeks. We created population-specific thermal performance curves and extracted the dormancy release temperature at which 50 % of the seeds had germinated (DRT50), the optimum dormancy-breaking temperature to stimulate maximum germination (T0), and the lethal temperature at which 50 % of the seeds were killed (LT50). Generalised linear models were used to examine relationships between thermal thresholds and possible vegetation, climate, and fire regime drivers of intraspecific variation in seed traits. We found that thermal thresholds differed between vegetation communities, with thresholds consistently higher in forest-type ecosystems compared to open woodland, and the influence of climate varied significantly between the two communities. Seeds from Jarrah Forest populations had a DRT50 16.0 °C higher, a T0 9.7 °C higher, and LT50 7.8 °C higher than seeds from Banksia woodlands. A high rate of non-dormancy was identified in one population that had lost fire in its system and displayed significant germination after both summer and fire-related temperatures. The PY thermal thresholds modelled here provide insight into the strong influence of variable soil heating as a function of vegetation and fuel dynamics in fire-prone environments. Our findings highlight the significant intraspecific variation for this species and suggest that fire-induced soil heating generated by vegetation characteristics may be an overlooked element of fire regimes shaping seed traits.

Abstract:Understanding how seed functional traits interact with environmental factors to determine seedling recruitment is critical to assess the impact of climate change on ecosystem restoration. This study focused on the effects of environmental factors on the mother plant during early plant life history stages and during seed development. Desmodium brachypodum A. Gray (large tick trefoil, Fabaceae) was used as a model species. Firstly, this study analyzed seed germination traits in response to temperature and moisture stress. Secondly, it investigated how seed burial depth interacts with temperature and soil moisture to influence seedling emergence traits. Finally, it determined if contrasting levels of post-anthesis soil moisture could result in changes in D. brachypodum reproductive biology and seed and seedling functional traits. The results showed that elevated temperature and moisture stress interacted to significantly reduce the seed germination and seedling emergence (each by >50%), while the seed burial improved the seedling emergence. Post-anthesis soil moisture stress negatively impacted the plant traits, reducing the duration of the reproductive phenology stage (by 9 days) and seed production (by almost 50%). Unexpectedly, soil moisture stress did not affect most seed or seedling traits. In conclusion, elevated temperatures combined with low soil moisture caused significant declines in seed germination and seedling emergence. On the other hand, the reproductive output of D. brachypodum had low seed variability under soil moisture stress, which might be useful when sourcing seeds from climates with high variability. Even so, a reduction in seed quantity under maternal moisture stress can impact the long-term survival of restored plant populations.

Comprehensive Morphological and Molecular Insights into Drought Tolerance Variation at Germination Stage in Brassica Napus Accessions

Abstract:Drought constitutes a noteworthy abiotic stressor, detrimentally impacting seed germination, plant development, and agricultural yield. In response to the threats imposed by climate change and water paucity, this study examined the morphological divergence and genetic governance of drought resilience traits at the germination stage in 196 rapeseed (Brassica napus L.) lines under both normal (0 MPa) and drought-induced stress (−0.8 MPa) scenarios. Our study showed that the composite drought tolerance D value is a reliable index for identifying drought resilience. Through a genome-wide association study (GWAS), we uncovered 37 significant SNP loci and 136 putative genes linked to drought tolerance based on the D value. A key discovery included the gene BnaA01g29390D (BnNCED3), encoding 9-cis-epoxycarotenoid dioxygenase, which exhibited significantly heightened expression levels in drought-resistant accessions (p < 0.01), underscoring its potential as a positive drought stress regulator and a suitable candidate for genetically enhancing drought resilience. Moreover, we pinpointed four stress-reactive transcription factors (BnaA07g26740D, BnaA07g26870D, BnaA07g26910D, and BnaA07g26980D), two E3 ubiquitin-protein ligases (BnaA05g22900D and BnaC06g28950D), two enzymes (BnaA01g29390D and BnaA03g48550D), and two photosystem-associated proteins (BnaA05g22950D and BnaC06g28840D) as vital components in drought response mechanisms. The construction of a regulatory network reveals an ABA-dependent pathway (NCED3/RGLG5/IDD14) that contributes to drought tolerance in rapeseed seedlings, alongside the involvement of a drought avoidance strategy (APRR6/PHYB). The SNPs and genes unveiled in this study offer a substantial theoretical foundation for subsequent investigations targeting genetic improvement for drought resilience during seed germination in rapeseed.
Experimental climate warming enforces seed dormancy in South African Proteaceae but seedling drought resilience exceeds summer drought periods

Abstract:Two hypotheses—that elevated night-time temperatures due to climate warming would enforce post-fire dormancy of Proteaceae seed due to low moisture, and that periods without rain during summer would exceed desiccation periods tolerated by Proteaceae seedlings—were tested empirically. Enforced dormancy, i.e., the inability to germinate due to an environmental restraint, was tested by measuring seed germination in 11 Proteaceae species in experimental mesocosms whose soils were artificially elevated by 1.4 and 3.5 °C above ambient by far-red wavelength filtered infrared lamps. Diminished totality of germination and velocities were observed in 91 and 64 %, respectively, of the Proteaceae species tested. Drought resilience was tested in one-year-old seedlings of 16 Proteaceae species by withholding water from potted plants during summer in a greenhouse. The most drought-resilient Proteaceae species displayed the lowest initial transpiration rates at field capacity, the smallest declines in transpiration rate with decreasing soil water content, and the lowest water losses by transpiration. Projected drought periods leading to the complete cessation of transpiration in all Proteaceae species greatly exceeded the number of days without rain per month during summer in the current distribution ranges of those species. It was therefore concluded that enforced seed dormancy induced by elevated night-time temperatures is the post-fire recruitment stage of Proteaceae that is most sensitive to climate warming.

Abstract:Ecological theory predicts that plant reproductive phenology in the Mediterranean regions is shaped by evolutionary processes driven by strong seasonality in precipitation–evaporation patterns. Here we tested, for the first time, the Mediterranean seed germination syndrome using experimental data occurring in the Mediterranean Basin. We show that species from lowland zonal habitats of the Mediterranean align with the Mediterranean germination syndrome hypothesis while habitat specialists deviate from the syndrome, showing nearly opposite germination requirements. Cumulatively, these results suggest that evolutionary pressures from local habitat‐related conditions override the macroclimatically imposed Mediterranean seed germination syndrome. Ecological theory predicts that plant reproductive phenology in the Mediterranean regions is shaped by evolutionary processes driven by strong seasonality in precipitation–evaporation patterns. Thus, it can be expected that seed germination phenology has adapted to maximise recruitment during the season of highest water availability. Cold‐cued and slow germination (i.e., the 'Mediterranean seed germination syndrome') has been hypothesised to be an adaptation to ensure that seedling emergence occurs in autumn/early winter, extending the growing season before the subsequent unfavourable summer drought. However, this hypothesis has been tested on individual species or local studies, without a proper synthesis for the whole Mediterranean region. Here we tested, for the first time, the Mediterranean seed germination syndrome using experimental data for 459 species (11,363 records, 59 families) occurring in the Mediterranean Basin. We performed a phylogenetically informed Bayesian meta‐analysis to model the effect on germination proportions of seven key experimental cues: mean incubation temperature, alternating temperature regime, light and dormancy‐breaking treatments (scarification, warm stratification and cold stratification) and the modulating role of seed mass on seed germination. We show that species from lowland zonal habitats of the Mediterranean align with the Mediterranean germination syndrome hypothesis, with their seeds responding positively to cool, constant temperatures and negatively to light. Yet, habitat specialists (i.e., species restricted to mountains, coasts and wetlands) deviate from the syndrome, showing nearly opposite germination requirements. Seed mass further influences the germination niche and phylogenetically related species exhibit similar germination responses. Cumulatively, these results suggest that evolutionary pressures from local habitat‐related conditions override the macroclimatically imposed Mediterranean seed germination syndrome.

Abstract:Plant regeneration strategy plays a critical role in species survival and can be used as a proxy for the evolutionary response of species to climate change. However, information on the effects of key plant traits and phylogenetic relatedness on seed germination is limited at large regional scales that vary in climate. To test the hypotheses that phylogenetic niche conservatism plays a critical force in shaping seed ecophysiological traits across species, and also drives their response to climatic fluctuation, we conducted a controlled experiment on seed germination and determined the percentage and rate of germination for 249 species in subtropical China under two temperature regimes (i.e., daily 25°C; daily alternating 25/15°C for each 12 hr). Germination was low with a skewed distribution (mean = 38.9% at 25°C, and 43.3% at 25/15°C). One fifth of the species had low (<10%) and slow (4–30 days) germination, and only a few (8%) species had a high (>80%) and rapid (1.2–6.6 days) germination. All studied plant traits (including germination responses) showed a significant phylogenetic signal, with an exception of seed germination percentage under the alternating temperature scenario. Generalized linear models (GLMs) and phylogenetic generalized estimation equations (GEEs) demonstrated that growth form and seed dispersal mode were strong drivers of germination. Our experimental study highlights that integrating plant key traits and phylogeny is critical to predicting seed germination response to future climate change.

Abstract:Climate change models project an increase in the frequency and duration of drought globally. Changes in rainfall are expected to have particularly detrimental effects on seedlings due to their inability to reach deep water sources. We conducted a greenhouse experiment to test how the timing of watering affects seedling performance, physiology, and morphology in a keystone island endemic tree, Metrosideros polymorpha. Seedlings were grown for 60 days in one of five conditions: control treatment with daily watering to 100% field capacity (FC), a 28-day pulse drought with no water, and three groups maintained at 80% FC and watered every 2, 7, or 14 days. Seedling drought tolerance depended on the duration of drought; growth for plants watered to 80% FC every 2 or 14 days was comparable to 100% FC plants, but was reduced in the 28-day pulse drought and 7-day 80% FC. Survival was high in all treatments except the pulse drought. Stomatal conductance, chlorophyll content, and C:N ratio were positively related to height and biomass under drought. Traits associated with resource investment, fluorescence, and water use were plastic in their expression, shifting under drought compared to control conditions. M. polymorpha seedlings exhibit trait plasticity that allows them to tolerate drought, but its duration and intensity influence their survival and growth. Our results highlight the need for experimental studies manipulating temporal variation in water with explicit consideration of the role of trait plasticity in drought tolerance.

Abstract:• Background and Aims In water-limited landscapes, some plants build structures that enable them to survive with minimal water (drought resistance). Instead of making structures that allow survival through times of water limitation, annual plants may invoke a drought escape strategy where they complete growth and reproduction when water is available. Drought escape and resistance each require a unique combination of traits and therefore plants are likely to have a suite of trait values that are consistent with a single drought response strategy. In environments where condi- tions are variable, plants may additionally evolve phenotypically plastic trait responses to water availability. Invasive annual species commonly occur in arid and semi-arid environments and many will be subject to reduced water availability associated with climate change. Assessing intraspecific trait variation across environmental gradients is a valuable tool for understanding how invasive plants establish and persist in arid environments. • Methods In this study, we used a common garden experiment with two levels of water availability to determine how traits related to carbon assimilation, water use, biomass allocation and flowering phenology vary in California wild radish populations across an aridity gradient. • Key Results We found that populations from arid environments have rapid flowering and increased allocation to root biomass, traits associated with both drought escape and tolerance. Early flowering was associated with higher leaf nitrogen concentration and lower leaf mass per area, traits associated with high resource acquisition. While trait values varied across low- and high-water treatments, these shifts were consistent across populations, indicating no differential plasticity across the aridity gradient. • Conclusions While previous studies have suggested that drought escape and drought resistance are mutually exclusive drought response strategies, our findings suggest that invasive annuals may employ both strategies to succeed in novel semi-arid environments. As many regions are expected to become more arid in the future, investi-gations of intraspecific trait variation within low water environments help to inform our understanding of potential evolutionary responses to increased aridity in invasive species.

Abstract:Drought-induced plant mortality, resulting from either hydraulic failure or carbon starvation, is hypothesized to be modulated by the drought intensity. However, there is a paucity of research investigating the response strategies in desert shrubs under drought stress with different intensities. We transplanted potted Reaumuria soongorica (Pall.) Maxim. and Salsola passerina Bunge seedlings in the rain-out shelter, and implemented three water treatments: a control (well-watered, CK), a chronic drought (gradually less watered, CD), and a flash drought (not watered, FD). We then quantified plant physiological traits associated with water use and carbon assimilation. Both R. soongorica and S. passerina showed similar changes in water use and carbon characteristics under different drought treatments. Water use efficiency was not significantly changed, but embolism resistance was significantly lower in CD, and leaf specific conductivity and embolism resistance were significantly lower in FD compared to CK. Under the drought treatment, both shrubs had significantly lower hydraulic safety margins than CK, with FD being significantly lower than CD. Notably, FD had lower carbon assimilation and a lower leaf non-structural carbon concentration, but higher stem non-structural carbon concentration. The results of a principal component analysis showed that net photosynthetic rate, sapwood specific conductivity, embolism resistance, midday water potential, and leaf and stem soluble sugar concentration were the main axes of variation for R. soongorica traits. CK had the highest water use efficiency, net photosynthetic rate, and gas exchange rate, while FD had the lowest embolism resistance and highest osmoregulation. Midday water potential, leaf and stem soluble sugar concentration were the main axes of variation for S. passerina traits, and individual distribution under three water treatments was associated with drought tolerance traits. The findings suggest that species exhibit different response strategies for resistance to drought stress, with R. soongorica being drought-avoidant and S. passerina being drought-tolerant. These findings highlight the adaptive capacity of desert shrubs to water deficit and provide insights for assessing hydraulic failure and carbon starvation in desert shrubs.

Abstract:Abstract Background and Aims Hotter drought- and biotically-driven tree mortality are expected to increase with climate change in much of the western United States, and species persistence will depend upon ongoing establishment under novel conditions or migration to track ecological niche requirements. High-elevation tree species may be particularly vulnerable to increasing water stress as snowpack declines, increasing the potential for adult mortality and simultaneous regeneration failures. Seedling survival will be determined by ecophysiological limitations in response to changing water availability and temperature. Methods We exposed seedlings from populations of Pinus longaeva, Pinus flexilis, and Pinus albicaulis to severe drought and concurrent temperature stress in common gardens testing timing of drought onset under two different temperature regimes. We monitored seedling functional traits, physiological function, and survival. Key Results The combined stressors of water limitation and extreme heat led to conservative water use strategies and declines in physiological function, with these joint stressors ultimately exceeding species’ tolerances and leading to complete episodic mortality across all species. Growing conditions were the primary determinant of seedling trait expression, with seedlings exhibiting more drought-resistant traits such as lower specific leaf area in the hottest, driest treatment conditions. Water stress-induced stomatal closure was also widely apparent. Under adequate soil moisture, seedlings endured prolonged exposure to high air and surface temperatures, suggesting broad margins for survival. Conclusions The critical interaction between soil moisture and temperature suggests that rising temperatures will exacerbate growing season moisture stress. Our results highlight the importance of local conditions over population- and species-level influences in shaping strategies for stress tolerance and resistance to desiccation at this early life stage. By quantifying some of the physiological consequences of drought and heat that lead to seedling mortality, we can better understand the future effects of global change on the composition and distribution of high-elevation conifer forests.

Abstract:Extreme drought events during post-fire regeneration are becoming increasingly frequent in Mediterranean-type ecosystems. Understanding how plants with different traits and origins respond to such conditions during early life stages is therefore critical for assessing the effect of climate change. Here, seedlings of three Cistus (semi-deciduous malacophylls from the Mediterranean Basin) and three Ceanothus (evergreen sclerophylls from California) species, two post-fire seeder genera with contrasting leaf traits, were subjected to complete water deprivation for 3 months in a common garden experiment. The leaf and plant structure and plant tissue water relations were characterized before the drought, and the functional responses (water availability, gas exchange and fluorescence) were monitored during the drought. Both genera exhibited contrasting leaf structure and tissue water relations traits, with higher leaf area and specific leaf area as well as higher osmotic potential at maximum turgor and turgor loss point in Cistus than Ceanothus. During drought, Ceanothus showed a more conservative use of water than Cistus, with a water potential less sensitive to decreasing soil moisture and a strong decline in photosynthesis and stomatal conductance in response to water deficit, but also a level of fluorescence more responsive to drought than Cistus. However, we could not find a different degree of drought resistance between the genera. This was particularly clear between Cistus ladanifer L. and Ceanothus pauciflorus DC., the two most functionally contrasting species, but at the same time, the two most drought-resistant. Our findings demonstrate that species with different leaf traits and functional responses to water stress may not differ in their degree of drought resistance, at least during the seedling stage. This underlines the need to take general categorizations by genus or functional types with caution and to deepen our knowledge about the Mediterranean-type species ecophysiology, especially during early life stages, in order to anticipate their vulnerability to climate change.
