Abstract:Sarcoptes scabiei var. cuniculi are cutaneous mites of companion rabbits. In rabbits, sarcoptic mange is characterized by intense pruritus, alopecia, erythema, scales, and crusts around the head, neck, trunk, feet, and genitals. The goal of this study was to evaluate the effectiveness of oral fluralaner in pet rabbits naturally infested by S scabiei var. cuniculi. Twelve privately owned pet rabbits with a definitive diagnosis of sarcoptic mange were included in the study if they had compatible clinical signs and positive superficial skin scrapings for S scabiei. Clinical and parasitological evaluations were performed on days 0, 7, 14, 21, 30, 60, and 90 after receiving a single oral dose of 25 mg/kg of fluralaner. On day 7, 5 of 12 rabbits had positive skin scrapings, while 8 of 12 had clinical signs (alopecia and erythema) still present. By day 14 none of the rabbits had positive skin scrapings and only 1 of 12 had persistent clinical signs. By day 21 none of the rabbit had positive skin scrapings nor had clinical signs. A reoccurrence of the clinical signs or positive skin scrapings was not observed for the duration of the study (day 90). A single oral dose of fluralaner was effective for the treatment of naturally occurring sarcoptic mange in rabbits over a 90-day period.

Abstract:Various kinds of pets have been known to contract the ectoparasite Sarcoptes scabiei . Current acaricides are becoming less effective because of the resistance developed by the mite besides their adverse effects on the general activity and reproductive performance of domestic pets. For this reason, the present study aims to discover a novel and safe approach using silver and gold nanoparticles to fight Sarcoptic mange in rabbits as well as to explain their mechanism of action. 15 pet rabbits with clinical signs of Sarcoptic mange that were confirmed by the microscopic examination were used in our study. All rabbits used in this study were assessed positive for the presence of different developing stages of S. scabiei . Three groups of rabbits (n = 5) were used as follows: group (1) didn't receive any treatment, and group (2 and 3) was treated with either AgNPs or GNPs, respectively. Both nanoparticles were applied daily on the affected skin areas via a dressing and injected subcutaneously once a week for 2 weeks at a dose of 0.5 mg/kg bwt. Our results revealed that all rabbits were severely infested and took a mean score = 3. The skin lesions in rabbits that didn't receive any treatments progressed extensively and took a mean score = of 4. On the other hand, all nanoparticle-treated groups displayed marked improvement in the skin lesion and took an average score of 0–1. All NPs treated groups showed remarkable improvement in the microscopic pictures along with mild iNOS, TNF-α, and Cox-2 expression. Both nanoparticles could downregulate the m-RNA levels of IL-6 and IFγ and upregulate IL-10 and TGF-1β genes to promote skin healing. Dressing rabbits with both NPs didn't affect either liver and kidney biomarkers or serum Ig levels indicating their safety. Our residual analysis detected AgNPs in the liver of rabbits but did not detect any residues of GNPs in such organs. We recommend using GNPs as an alternative acaricide to fight rabbit mange.

Abstract:Four Rocky Mountain bighorn sheep (Ovis canadensis canadensis) were experimentally inoculated with Psoroptes sp. of bighorn sheep origin by placing approximately 50 mites into each ear. Two sheep developed reproducing mite colonies and lesions consisting of alopecia, and exudative dermatitis with scabs on ears, face and neck. Three months after inoculation, all four sheep were treated with an in-feed formulation of ivermectin at a dosage of 1 mg/kg of body weight in a pelleted ration daily for seven consecutive days. Mites could not be detected on any of the sheep 1, 2, or 3 mo after treatment, and lesions on the affected sheep resolved within 6 to 8 wk after treatment. Thus, under these conditions, ivermectin was completely effective in eliminating the mites; it also was palatable and nontoxic. Use of ivermectin in pelleted feed for field treatment of psoroptic mange is encouraged.

Abstract:Protein stabilization upon ligand binding has frequently been used to identify ligands for soluble proteins. Methods such as differential scanning fluorimetry (DSF) and differential static light scattering (DSLS) have been employed in the 384-well format and have been useful in identifying ligands that promote crystallization and 3D structure determination of proteins. However, finding a generic method that is applicable to membrane proteins has been a challenge as the high hydrophobicity of membrane proteins and the presence of detergents essential for their solubilization interfere with fluorescence-based detections. Here the authors used MsbA (an adenosine triphosphate binding cassette transporter), CorA (a Mg++ channel), and CpxA (a histidine kinase) as model proteins and show that DSLS is not sensitive to the presence of detergents or protein hydrophobicity and can be used to monitor thermodenaturation of membrane proteins, assess their stability, and detect ligand binding in a 384-well format.
Implication of apoptosis and oxidative stress in mitigation of ivermectin long-term hazards by zinc nanoparticles in male rabbits

Abstract:Ivermectin is the medication of choice for treating human onchocerciasis and is used in veterinary medicine to treat a variety of ectoparasites and endoparasites. This study was designed to investigate the effects of zinc nanoparticles (ZnNPs) on the fertility of male rabbits exposed to experimental ivermectin (IVM) intoxication. A total of 72 mature male rabbits were equally divided into 4 groups (n = 18). The first group (CTR) served as control; the second group (IVM) received subcutaneous injection of IVM (0.2 mg/kg body weight); the third group (ZnNPs) fed on zinc nanoparticles (60 mg/kg diet); and the fourth group (ZnNPs + IVM) were administered IVM and zinc nanoparticles at the same doses. The experiment lasted for 9 weeks. Results revealed that IVM-intoxicated rabbits showed impaired growth performance parameters, including body weight, total body weight gain (TBWG), total feed intake (TFI), and feed conversion ratio (FCR). Moreover, carcass characteristic and fertility parameters (including semen quality parameters and testosterone levels) were also impaired after IVM administration. Additionally, testicular malondialdehyde (MDA) and antioxidant (reduced glutathione, superoxide dismutase, catalase) levels as well as the histopathology and immunohistochemical expression of caspase 3 and PCNA in the testes and epididymis were detrimentally affected. On the contrary, ZnNP administration efficiently improved most of these parameters in IVM-intoxicated rabbits. In conclusion, ZnNPs exhibited promising ability for improving the growth and fertility status of rabbits and reducing the deleterious effects of IVM possibly through the suppression of apoptotic and oxidative pathways.
The effects of acetazolamide on visual function in retinitis pigmentosa.

Abstract:PURPOSE To study the effects of acetazolamide on central and peripheral visual function in patients with retinitis pigmentosa (RP) who showed no evidence of macular edema. METHODS Thirteen patients with retinitis pigmentosa participated in a preliminary study. Measures of central and peripheral visual function were obtained before and after an 8 wk period on acetazolamide. An additional 10 patients participated in a cross-over study. They were placed on a placebo for an 8 wk period, then on acetazolamide for a second 8 wk period. RESULTS None of the patients in the preliminary study showed significant changes in visual acuity, color vision, foveal cone pathway sensitivities, focal electroretinogram (ERG) amplitudes, or in any ERG parameter. Three patients, however, showed significant changes in visual field area and in dark-adapted thresholds. None of the patients in the cross-over study showed significant increases in visual field area. CONCLUSIONS Given the results and the reports of side-effects, it is difficult to justify using acetazolamide to improve retinal function in RP patients who show no evidence of cystoid macular edema.
Preliminary detection of mites and coccidia with their zoonotic potential in meat-farmed rabbits in three districts in Selangor, Malaysia

Abstract:Mites infestation and gastrointestinal parasites including coccidia are common problems reported in pets, petting farms and farmed practices. Sarcoptes sp. and Cheyletiella sp. could be a potential zoonosis from rabbits to human. Detection of mites and coccidia with their zoonotic potential in meat-farmed rabbits from three (3) commercial farms in Selangor were investigated. Tape impression, fur pluck, skin scraping and ear swab tests were used for mites detection and faecal samples was used for coccidia examination by using McMaster's technique and the identification of Eimeria spp. was further analysed by sporulation technique. The overall prevalence of mites and Eimeria spp. (oocysts) in rabbits were 51.85% ± 0.38 (standard deviation; S.D.) and 76.47% ± 0.42 respectively. Sarcoptes scabiei was the most frequent mite found (25.92% ± 0.44), followed by Cheyletiella parasitovorax and Psoroptes cuniculi. Nine Eimeria spp. were identified and the oocysts of E. perforans shows the highest prevalence (64.71% ± 3.97) followed by E. exigua, E. coecicola, E. magna, E. flavescens, E. irresidua, E. intestinalis, E. media and E. stiedai. There was a significant difference (p = 0.013) where large-scale farm has a higher prevalence of coccidia than small scale farms apparently due to the excessive stocking density as coccidia are easily transmitted among rabbits through ingestion of sporulated oocysts. In conclusion, mites and coccidia are commonly present in the commercial rabbit farms, thus control and preventive measures should be executed to reduce the incidence of parasites. The zoonotic mites Sarcoptes scabiei and Cheyletiella parasitovorax detected in this study could be regarded as a public health concern especially when handling the rabbit.

Conclusion In pet rabbits with Cheyletiella parasitivorax infestations both topical selamectin and injectable ivermectin have been recognised to be a safe and effective treatment. There is currently insufficient evidence that one is more effective than the other and therefore veterinarians should consider their own clinical experience, client preference and use the licensed product first (in the UK, ivermectin (Xeno, Dechra) spot-on). However, this Knowledge Summary does not comment on the effectiveness of topical ivermectin in treating cheyletiellosis. If injectable ivermectin is , topical selamectin can be used under the cascade as an alternative treatment, as it is licensed for the same indication in dogs and cats. It is intensely itchy and caused by the highly contagious mite Sarcoptes scabiei. There are many theories regarding the increased prevalence, including hypotheses related to global warming, increased poverty, reduced treatment availability and resistance. The standard treatments in the UK include topical malathion, topical permethrin and oral ivermectin. All have had significant supply issues as reported by the British Association of Dermatologists. There have also been concerns of permethrin resistance. Oral ivermectin is expensive, with four tablets listed at £116.84 in the British National Formulary (BNF). Finally, oral ivermectin is not licensed in children weighing < 15 kg (although data seem to suggest this is safe). One option that is less used but reported is topical ivermectin. This was in the 2018 Cochrane review that assessed the efficacy of topical permethrin, and oral and topical (as 1% lotion) ivermectin. This showed no difference in efficacy and few or mild adverse events. These findings are also replicated in recent studies and there are case reports of topical ivermectin to treat neonatal scabies that seems resistant to permethrin. In our centre, we have used topical ivermectin (as the 10 mg g−1 cream Soolantra) to treat 10 patients, with great success. The first was a 14-month-old child who weighed 9.2 kg. She had previously had three courses of permethrin and two courses of malathion with ongoing signs and symptoms of scabies, including burrows. The entire family had been treated every time and the entire family (except this patient) was treated with oral ivermectin during the final treatment before topical ivermectin. The patient was treated top to toe with two applications of topical ivermectin cream 1 week apart, with resolution after one application. Subsequently we have had success using the same protocol to treat a 21-year-old man (with no response to four courses of permethrin) and his 53-year-old mother (with no response to three courses of permethrin). We have also treated a 31-year-old man, a 48-year-old man and his partner (all of whom had not responded to one course of permethrin), and a 68-year-old man and his family who had no previous treatment. Our patients have not experienced any adverse effects and there have been no treatment failures. We report on 10 patients who we treated successfully with topical ivermectin. Topical ivermectin provides an exciting option for treatment and, of note, one tube is listed at £27.43 in the BNF, making it significantly cheaper than the oral alternative.

Abstract:Murine fur mites are commonly excluded in modern research animal programs, yet infestations continue to persist due to challenges in detection and control. Because all diagnostic methods and treatment options have limitations, programs must make many operational decisions when trying to eradicate these ectoparasites. The primary aim of this study was to assess various durations of treatment time with an ivermectin-compounded diet in eliminating Radfordia affinis in mice as determined by PCR testing and pelt examination. A shorter treatment duration would be highly advantageous as compared with the current regimen of 8 wk as it would minimize cost and time for animal management programs, impediments to research, and ivermectin drug effects on infested animals. Five experimental groups of R. affinis -positive mice received dietary ivermectin for 0, 2, 4, 6, or 8 wk. A fur mite-negative, naïve mouse was added to each group every 8 wk to perpetuate the infestation and amplify any remaining populations of fur mites. At 16 wk after the respective treatment end, PCR testing was performed for all treated groups in conjunction with the positive control group (no treatment). Visual examination of pelts for mites and eggs via direct microscopy was also performed at each time point. All treated mice were free of R. affinis at 16 wk after the end of treatment as confirmed by both PCR testing and pelt examination. These findings indicate that a dietary ivermectin treatment duration of as little as 2 wk is effective in eliminating R. affinis , making successful eradication initiatives more achievable.

Abstract:BackgroundScabies is caused by Sarcoptes scabiei burrowing into the stratum corneum of the host’s skin and is detrimental to the health of humans and animals. Vaccines are an attractive alternative to replace the acaricides currently used in their control.MethodsIn the present study, the S. scabiei chitinase-like protein 5 (SsCLP5) was characterized and recombinant SsCLP5 (rSsCLP5) was evaluated as a candidate vaccine protein for anti-mite protection in rabbits. The expression, characterization and immunolocalization of SsCLP5 were examined. Vaccination experiments were performed on three test groups (n = 12 per group) immunized with purified rSsCLP5. Control groups (n = 12 per group) were immunized with PBS, QuilA saponin or empty vector protein. After challenge, the inflammatory reaction and skin lesions were graded and rSsCLP5 indirect ELISA was used to detect antibody IgG levels in serum samples at the time of vaccination and post-challenge.ResultsThe results showed that rSsCLP5 had high immunoreactivity and immunogenicity. In S. scabiei, SsCLP5 had a wide distribution in the chewing mouthpart, legs and exoskeleton, especially the outer layer of the exoskeleton. Vaccination with rSsCLP5 resulted in 74.3% (26/35) of rabbits showing no detectable lesions after challenge with S. scabiei.ConclusionsOur data demonstrate that rSsCLP5 is a promising candidate for a recombinant protein-based vaccine against S. scabiei. This study also provides a method for studying scabies vaccine using rabbit as an animal model and a basis for screening more effective candidate proteins.

Abstract:CLINICAL sheep scab (Psoroptes ovis infestation) is a highly contagious ectoparasitic disease affecting sheep of all ages. The clinical signs include pruritus, serous exudation and eventually severe debilitation in untreated sheep. The failure to identify early lesions and/or improper treatment of sheep infested with P ovis, are real welfare concerns. Following the deregulation of the previous sheep scab control measures (Sheep Scab [National Dip] Order 1990) in July 1992, there has been an alarming increase in the number of outbreaks of sheep scab reported in the UK. There is now no statutory device to ensure that sheep are correctly plunge‐dipped for the control of P ovis infestation. Veterinary surgeons must, therefore, advise on the most appropriate treatment for each outbreak of sheep scab, depending on the severity of the infestation, the stage of pregnancy of the sheep and the handling facilities available.

[The influence of coccidiostats on the course of coccidiosis and production results in an industrial rabbit farm]

Abstract:The study was carried out on material consisting of 3375 rabbits of the White New Zealand breed. The total number of 9 species of coccidia were found, 8 of which were intestinal (E. perforans, E. media, E. magna, E. irresidua, E. coecicola, E. flavescens, E. piriformis oraz E. intestinalis) and E. stiedai inhabiting the liver. Three specirs - E . irresidua, E. coecicola and E. flavescens have been found in Poiand for the first time. Three of four coccidiostats examined i.e. Sacox, Cycostat and Baycox may be used to prevent coccidiosis in rabbit farm. Activity of Vetrocox is less effective and it should not be used in the prevention of coccidia infection. At the age of 90 days the best results were obtained by treatment of Sacox (257 g of increase body weight and 9 per cent lower mortality comparing with the control group), the second test result was after two doses of Baycox and after Cycostat treatment.
Wide host range of murine sarcoma virus

Abstract:Cultures of canine, rabbit, porcine, avian, feline, bovine, simian and human origin were tested for their susceptibility to the Kirsten murine sarcoma virus (Ki‐MSV). Data indicated that Ki‐MSV replicates and transforms the cultures of canine embryo, rabbit kidney, porcine kidney, feline embryo, bovine embryo and human tumor cells. These morphologically altered cells contained infectious virus and group‐specific (gs) complement‐fixing (CF) antigen characteristic of viruses of the murine sarcoma‐leukemia complex. The infected cultures produced type‐C virus particles. RNA‐dependent DNA polymerase activity was also demonstrable in the altered cells. The present results indicate that members of the murine sarcoma‐leukemia virus complex, particularly Ki‐MSV, exhibit a wider host range than was hitherto believed.

Abstract:Sheep scab, caused by infestation with the mite Psoroptes ovis, is highly contagious, causing intense pruritus and represents a major welfare and economic concern. Disease control strategies rely upon chemotherapy, however, sustainability is questionable due to issues of chemical residues, eco-toxicity and acaricide resistance. Control by vaccination is supported by demonstration of protective immunity in sheep previously infested with P. ovis. We identified vaccine candidates for P. ovis based on: (1) antigens selected by their interaction with host signalling pathways and the host immune-response; and (2) those shown to be either immunogenic or involved in mite feeding. This resulted in the development and validation, in repeated immunisation and challenge trials, of a seven recombinant protein sub-unit cocktail vaccine. Sheep were inoculated on three occasions, 2 weeks apart, along with QuilA adjuvant. Vaccination resulted in highly significant reductions in both lesion size (up to 63%) and mite numbers (up to 56%) following challenge. Mean lesion size in vaccinates was significantly smaller than controls from 1 week post infestation (wpi) until the end of the experiment at 6 wpi. All antigens elicited serum IgG responses following immunisation and prior to infestation, whereas controls did not produce antigen-specific IgG during the pre-infestation period. Vaccinated animals showed an amnestic response, with levels of antigen-specific IgG against muGST, Pso o 1 and Pso o 2 increasing following infestation. This vaccine represents the greatest reduction in lesion size to date with a sheep scab vaccine, providing encouragement for future production of a commercially-viable means of immunoprophylaxis.

Eradication of porcine sarcoptic mange within a health declared production model

Abstract:The success of a large-scale eradication programme against sarcoptic mange was evaluated in piglet producing herds belonging to the health class LSO 2000. These farms are monitored to be free from sarcoptic mange, swine enzootic pneumoniae, swine dysentery and progressive atrophic rhinitis. The local veterinarians were instructed to eradicate sarcoptic mange from the herds by 2 ivermectin injections 14 days apart or 3 treatments with phoxim pour-on 6 days apart before the farms were able to join the health class. The study consisted of a questionnaire administered to the farmers, farm visits and follow-ups associated with the regular control of the health class. A questionnaire was sent to all of the 379 accepted farrowing farms. Altogether 323 farmers had initiated an eradication programme at least 7 months before the questionnaire was distributed and 96% of them believed that the mange eradication programme had been successful. In most of the successful herds (90%), the local veterinarian had treated the animals by the recommended methods, and the majority of the farmers (88%) had cleaned and sprayed the pens with insecticides on the days of treatment. Three out of the 7 unsuccessful farms had employed methods other than those recommended. Twenty-five herds were randomly selected for an intensified control. The rubbing index was < or = 0.1 in all herds visited, indicating freedom from mange. Sows or gilts, 15-20 animals per farm, were skin scraped and no Sarcoptes scabiei was found. The same animals were blood sampled and serum antibodies to S. scabiei were measured by an indirect ELISA. All except 1 farm had low optical density (OD) values in most of the samples. The owner of that farm had become uncertain about the mange status of the herd and had recently retreated all animals with acaricides. The number of complaints about mange in the finishing units buying health class feeder pigs was low. The study demonstrates that it is possible to run an eradication programme against sarcoptic mange, in which a large number of farmers together with their veterinarians participate. It is possible to eradicate mange not only at the herd but also at the population level. The ELISA test employed proved to be a possible tool to monitor the efficacy of mange-eradication programmes.

Abstract:Scabies is a prevalent ectoparasitic infectious disease, caused by the mite Sarcoptes scabiei. As a consequence of the infestation, localised cutaneous inflammation, pruritus and polymorphic skin lesions develop. The primary symptoms of scabies manifest as hypersensitivity-like reactions and immune responses, the precise mechanisms of which remain poorly defined. The objective of this study was to evaluate the effects of oral ivermectin treatment in patients with scabies on the systemic immune response and the patient's quality of life (QoL). Patients admitted to the dermatology outpatient clinic and diagnosed with scabies were administered oral ivermectin treatment following diagnosis at week 0 and 2. Laboratory tests were conducted to measure complete blood count (CBC), erythrocyte sedimentation rate (ESR), and C-reactive protein (CRP) levels before treatment and at week 4. The systemic immune-inflammation index (SII) was calculated using the platelet, neutrophil and lymphocyte counts. Additionally, data pertaining to the Dermatological Life Quality Index (DLQI) were recorded. In 119 patients (51 males) diagnosed with scabies, increases in ESR, CRP, and SII values and decreases in inflammatory cell counts and DLQI scores were observed one month after treatment with oral ivermectin. The results of the study showed that the use of oral ivermectin, a scabicidal agent, triggered the inflammatory response and improved the QoL of the patients.
