Abstract:Modern viticultural areas are being confronted with the negative impacts of global warming on yield and fruit composition, with especially adverse effects on anthocyanin synthesis. Novel and sustainable tools, such as biostimulants, may represent a viable alternative to traditional cultural practices, thus promoting eco-friendly strategies to enhance the yield, fruit quality and abiotic stress tolerance of grapevines. 'Crimson Seedless' is a late-season red table grape variety, and due to climatic warming, its berries are frequently failing to acquire the commercially acceptable red color. Canopy applications of different biostimulants, namely, Kelpak®, Sunred®, Cytolan®, LalVigne™ Mature as well as Ethrel® Top, were tested on grapevine cv. Crimson Seedless grown under semi-arid Mediterranean conditions in order to evaluate their effects on yield and fruit quality. Some of the products were sprayed in canopies at labeled doses, and some were applied at doses reported in other studies. For the control treatment, canopies were sprayed with water. Sampling started at veraison and was repeated at 10-day intervals to measure the evolution of berry weight, length and diameter, as well as the total soluble solids and titratable acidity of the juice. The grapes were harvested when the berries of one of the treatments attained the commercially acceptable color. The greatest improvements in the red berry color were achieved with Sunred® (at a dose of 4 L ha-1) and Ethrel® Top (250 ppm plus glycerol at 1%), each applied at veraison and 10 days later. The different applications had varying effects on productivity and qualitative parameters. Only Sunred® improved the accumulation of anthocyanin and the overall acceptability of table grapes by consumers. The obtained results clearly demonstrate that applying Sunred® can improve the yield and qualitative parameters of the red table grape variety 'Crimson Seedless', indicating that this biostimulant could be a viable alternative to the most widely used plant growth regulator, ethephon.

Abstract:The influence of red light-emitting diodes (LEDs) irradiation (454 LUX) on the quality and physiology of postharvest table grapes fruit stored at (22.5 ± 2.5 ℃) for 6 days after harvest were explored. The results exhibited that red-light irradiation exposure was remarkably efficient in decreasing the weight loss of the fruit. The rachis browning and chlorophyll degradation were significantly inhibited when exposed to red-light. During storage, color, total soluble solids, firmness, titratable acidity, malondialdehyde, hydrogen peroxide, and superoxide levels reduced to levels lower than under control. In addition, red-light irradiation retained higher membrane permeability, bioactive compound, leading to higher fruit antioxidant capacity. Further research has shown that treatment with red-light enhanced antioxidant enzymes activity. Red-light treatment maintained significantly higher levels of individual phenolic compounds than control during storage. These results indicate that irradiating table grapes with red-light will help in extending postharvest shelf-life and improve commercially grown grapes’ quality.

Abstract:Grape polyphenols contributing to more than half of the global polyphenol market were well studied; however, how melatonin (MLT), a potential plant hormone, and abscisic acid (ABA) affects polyphenols profile is still poorly understood. To explore whether these hormones are involved in polyphenolic biosynthesis, grape (Vitis vinifera cv. Kyoho) was exposed to MLT, ABA, and NDGA (nordihydroguaiaretic acid, an ABA biosynthesis inhibitor) treatments, and 16 polyphenols were identified from grape extracts by high performance liquid chromatography quadrupole time of flight mass spectrometry (HPLC-Q-TOF-MS). Both exogenous MLT and ABA significantly enhanced the biosynthesis of each flavonol and flavanol component, especially catechin, which was almost increased double by 200 mu M of MLT treatment. Furthermore, the expression of genes involved in flavonoid biosynthesis, including 4-coumaroyl-CoA synthase, chalcone synthase, flavonoid 3'-hydroxylase, anthocyanin 3'-methyltransferase, flavonol synthase, flavonoid-3-O-glucosyltransferase, and flavonoid 3',5'-methyltransferase were highly up-regulated as well but were down-regulated by NDGA. The present study provided new insights for improving flavonoids accumulation in agricultural production and its underlying mechanism.
Application timing and concentration of abscisic acid improve color of ‘Benitaka’ table grape

Abstract:‘Benitaka’ is one of the most important colored table grapes grown in Brazil. However, the lack of a pink and uniform color of berry skins is frequently observed when this cultivar is grown in warm climates, where high temperatures may inhibit anthocyanin accumulation. The endogenous plant growth regulator, abscisic acid (S-ABA), the concentration of which increases in grape berry skins at the onset of maturation (veraison), seems to be involved in the regulation of anthocyanin accumulation. Recently, an efficient S-ABA production method was developed so that it could be used on crops, which led to the evaluation of different concentrations applied at or around veraison to colored table grapes. Thus, a trial was carried out during two consecutive seasons in a commercial vineyard of ‘Benitaka’ table grape trained on an overhead trellis system, located at Marialva, Paraná state, Brazil. A randomized complete-block design was used with vines as blocks and clusters as experimental units, with four replications and eight treatments, as follows: (a) control; (b) S-ABA 200mg/L applied 7 days after veraison (DAV); (c) S-ABA 400mg/L 7 DAV; (d) S-ABA 200mg/L 15 days before harvest (DBH); (e) S-ABA 400mg/L 15 DBH; (f) S-ABA 200mg/L 7 DAV+200mg/L 15 DBH; (g) S-ABA 400mg/L 7 DAV+400mg/L 15 DBH; (h) ethephon 500mg/L 7 DAV. In both seasons, S-ABA improved the color of ‘Benitaka’ grapes, especially when it was applied twice, i.e., 400mg/L 7 days after veraison and 400mg/L 15 days before harvest. Regardless of the concentrations or application timing, there is no alteration in the berries and clusters physical characteristics.

Abstract:Color and other quality parameters of "Redglobe" grape (Vitis vinifera L.) berries were evaluated after treatment with brassinosteroid (BR) analogs. Three BRs analogs (24-epibrassinolide, Triol, or Lactone) were applied at three concentrations (0.0, 0.4, or 0.8 mg⋅L-1), at the onset of veraison. A commercial formulation (B-2000®) was also applied, at a recommended rate of 0.06 mg⋅L-1. The tested BR analogs were effective improving berry color (evaluated as color index for red grapes, CIRG), increasing the levels of soluble solids and anthocyanins, and changing the types of anthocyanins present without altering other quality and yield parameters. The effects of BR analogs on color enhancement could be explained by an increase in soluble solids content and/or anthocyanin content. Treatment with 24-epibrassinolide (at 0.4 mg⋅L-1) or the commercial formulation tended to favor the production of dihydroxylated anthocyanins, which are responsible for the red and pink colors of grape berries. Results indicate that the use of BRs constitutes a potential tool in the production of table grapes. This is the first report of this enhancement effect in a productive context.

Abstract:The color and berry size of ‘Crimson Seedless’ play vital role in their marketability. In the present study, eight-year-old ‘Crimson Seedless’ was used as the experimental material to investigate the effect of plant growth regulators on the fruit weight, quality and color. Several dilutions of “Pro-Gibb” (20% gibberellic acid (A3)) and “Promalin” (1.8% 6-BA & 1.8% gibberellic acid (A4 + A7)) were applied to the ‘Crimson Seedless’ plants during the swelling period, and the results showed that the cluster weight and berry weight were significantly increased by 46.77% and 20.64% respectively when treated with 6000 times diluted solution of “Promalin”. Except for application of 15,000 times diluted solution of “Pro-Gibb”, 4000 times and 5000 times diluted solution of “Promalin”, there were no significant differences between control and other treatments in soluble solid content (SSC). The titratable acid content (TA) of the berries was also significantly reduced under various treatments of plant growth regulators. The Vc content of the berries under T2 treatment increased up to 6.40 mg/100 g compared with the control. The color of berries with 20,000 times diluted solution of “Pro-Gibb” treatment was pink, and the color of berry with other treatments was red. In conclusion, application of 6000 times diluted solution of “Promalin” improved fruit quality, texture and berry weight more effectively compared with other applied treatments. Overall, the results of the present study showed that optimized doses of the plant growth regulators could significantly improve the ‘Crimson Seedless’ fruit quality.

Abstract:(1) Background: To solve the problems of incomplete coloration and quality decline caused by unreasonable use of regulators in grapes, this study clarified the differences in the effects of a hormone-type growth regulator (AUT) and two commercial regulators on grape coloration and quality through field experiments. (2) Methods: The color indexes (brightness (L*), red/green color difference (a*), yellow/blue color difference (b*), and color index for red grapes (CIRG)) of grape fruit were measured using a CR-400 handheld color difference meter. The titratable acid content, total phenol content, and total sugar content were measured using anthrone colorimetry, folinol colorimetry, and NaOH titration, respectively, and the chalcone isomerase activity, phenylalanine ammoniase activity, dihydroflavol reductase activity, and anthocyanin content were measured using a UV spectrophotometer. (3) Results: The a*, total sugar and total phenol contents, and chalcone isomerase (CHI) and phenylalanine ammoniase (PAL) activities of grape fruit in the AUT treatment significantly increased, while the titratable acid content significantly decreased, compared to those in the CK treatment. The expressions of the differentially expressed genes (DEGs) trpB and argJ in AUT treatment were significantly up-regulated. The expressions of the differentially expressed metabolites (DEMs) phenylalanine and 4-oxoproline were significantly up-regulated, while those of 3,4-dihydroxybenzaldehyde and N-acetyl glutamate were significantly down-regulated. The CIRG significantly increased by 36.4% compared to that in the CK, indicating improved fruit coloration. (4) Conclusion: The AUT could shorten the color conversion period of grape fruit and improve the coloration, taste, and tolerance to saline and alkaline stresses.
Enhancing Red Table Grape Coloration Using Tsikoudia: A Novel and Sustainable Approach

Abstract:Achieving optimal coloration in red table grapes, especially in warm-climate regions, presents significant challenges due to high temperatures that inhibit anthocyanin biosynthesis. Conventional methods to enhance grape coloration, including the use of abscisic acid (ABA), ethephon, foliar nutrient supplementation, and viticultural practices like cluster trimming and girdling, have limitations related to cost, regulatory restrictions, and potential adverse effects on grapes quality. This study proposes the application of tsikoudia, a traditional Greek alcoholic beverage, as a novel, sustainable, and cost-effective alternative to conventional practices. Tsikoudia, applied during the veraison stage, significantly improved the coloration of 'Crimson Seedless' and 'Red Globe' grapes by enhancing anthocyanin accumulation and altering color parameters. Specifically, lightness (L*), chroma (C*), and hue angle (h), measured using the CIE-Lab color system, were reduced, while the Color Index for Red Grapes (CIRG) was increased. Additionally, total anthocyanin content, determined through spectrophotometric analysis, also showed an increase. These changes indicate a more intense red coloration. This research highlights the effectiveness of tsikoudia in improving grape coloration and contributes to the development of more sustainable viticultural practices.

Abstract:Grapes are important sources of phenolics, which are recognized to possess high biological value. In this work, we evaluated the total phenol content (TPC), total anthocyanin content (TAC), antioxidant activity (AA) and volatile composition of unstudied seedless table grape varieties (i.e., Autumn Crisp and Pristine as white cultivars, Scarlotta and Crimson as red cultivars and Adora and Melody as black cultivars). As a result, Adora and Melody exhibited higher TPCs, TACs and AA, measured by the 2,2-diphenyl-2-picrylhydrazil (DPPH) and photochemiluminescence (PCL) assays, than the rest of the varieties. A comparison between the two black cultivars proved that Adora possessed higher TPC (62.70 mg GAE 100 g −1 ), DPPH radical scavenging activity (IC 50 of 3.69 mg ml −1 ) and PCL antiradical activity (72.14 μg Trolox ml −1 ) than Melody (47.30 mg GAE 100 g −1 , IC 50 of 5.0 mg ml −1 and 42.36 μg Trolox ml −1 , respectively). The volatile composition, determined by solid phase microextraction (SPME)–gas chromatography/mass spectrometry (GC/MS), indicated a similar qualitative aroma profile for all varieties. However, quantitative differences were measured among them, which were reflected in distinct sensory perception by sensorial testing. From these results, Adora was selected for its antioxidant properties and flavor characteristics.

The Complexity of Modulating Anthocyanin Biosynthesis Pathway by Deficit Irrigation in Table Grapes

Abstract:Deficit irrigation (DI) is an irrigation scheduling technique that is used in grapes to improve red color development; however, results are not always satisfactory in table grapes. The red color in grapes is mainly due to the plant pigment anthocyanin. In the present study, the anthocyanin biosynthesis in Scarlet Royal grapes (Vitis vinifera L.) grown in the San Joaquin and Coachella Valleys, and subjected to two different DI strategies was investigated. The objective of this study was to identify potential regulatory factors that may lead to potential treatments to improve red color in table grapes, especially under warm climate conditions. In both locations, DI induced the expression of several genes involved in three major pathways that control the red color in table grapes: anthocyanin biosynthesis, hormone biosynthesis, and antioxidant system. DI at veraison induced anthocyanin accumulation and enhanced red color in berries at harvest time. However, anthocyanin accumulation was lower at the Coachella Valley compared to the San Joaquin Valley. The lower level of anthocyanin was associated with lower expression of critical genes involved in anthocyanin biosynthesis, such as flavonoid-3-O-glucosyltransferase (UFGT), myb-related regulatory gene (R2R3-MYB) (MYBA1), basic helix-loop-helix (bHLH) (MYCA1) and the tryptophan-aspartic acid repeat (WDR or WD40) proteins (WDR1). Further, gene expression analysis revealed the association of ABA biosynthesis gene 9-cis-epoxycarotenoid dioxygenase (NCED1), 1-aminocyclopropane-1-carboxylic acid oxidase (ACO3), and the gibberellic acid (GA) catabolic gene GA2 oxidase (GA2ox1) in the induction of anthocyanin biosynthesis. An increase in the chalcone synthase gene (CHS2) was observed in response to DI treatments in both sites. However, CHS2 expression was higher in Coachella Valley after ending the DI treatment, suggesting the involvement of environmental stress in elevating its transcripts. This data was also supported by the lower level of antioxidant gene expression and enzyme activities in the Coachella Valley compared to the San Joaquin Valley. The present data suggested that the lack of grape red coloration could partially be due to the lower level of antioxidant activities resulting in accelerated anthocyanin degradation and impaired anthocyanin biosynthesis. It seems that under challenging warmer conditions, several factors are required to optimize anthocyanin accumulation via DI, including an active antioxidant system, proper light perception, and hormonal balance.
Improving Cuticle Thickness and Quality Traits in Table Grape cv. 'Italia' Using Pre-Harvest Treatments

Abstract:Table grape viticulture, due to the impact of climate change, will have to face many challenges in the coming decades, including resistance to pathogens and physiological disorders. Our attention was focused on fruit cracking due to its ubiquitous presence in several species. This study explores the effects of three different treatments on the epidermis and cuticle of table grape berries by evaluating the impact of the girdling technique on various fruit quality parameters, including cuticle thickness, sugar content, acidity, color, bunch weight, and rheological properties. The treatments were (1) calcium chloride (CaCl2), (2) calcium chloride + salicylic acid (CaCl2 + SA), and (3) calcium chloride + Ascophyllum nodosum (CaCl2 + AN), with and without girdling, plus an untreated control. This research was conducted over the 2021–2022 growing season in a commercial vineyard in Licodia Eubea, Sicily, Italy. The results indicate significant variations in cuticle thickness and other qualitative traits throughout the growth and ripening phases, with notable differences depending on the treatment used. This study's findings suggest that specific treatments can influence the structural integrity of the grape cuticle, potentially impacting the fruit's susceptibility to cracking and overall marketability. The findings provide valuable insights into the role of chemical treatments and cultural techniques in enhancing fruit quality and resistance to environmental stresses in table grape cultivation.

The grapevine VviPrx31 peroxidase as a candidate gene involved in anthocyanin degradation in ripening berries under high temperature

Abstract:Anthocyanin levels decline in some red grape berry varieties ripened under high-temperature conditions, but the underlying mechanism is not yet clear. Here we studied the effects of two different temperature regimes, representing actual Sangiovese (Vitis vinifera L.) viticulture regions, on the accumulation of mRNAs and enzymes controlling berry skin anthocyanins. Potted uniform plants of Sangiovese were kept from veraison to harvest, in two plastic greenhouses with different temperature conditions. The low temperature (LT) conditions featured average and maximum daily air temperatures of 20 and 29 °C, respectively, whereas the corresponding high temperature (HT) conditions were 22 and 36 °C, respectively. The anthocyanin concentration at harvest was much lower in HT berries than LT berries although their profile was similar under both conditions. Under HT conditions, the biosynthesis of anthocyanins was suppressed at both the transcriptional and enzymatic levels, but peroxidase activity was higher. This suggests that the low anthocyanin content of HT berries reflects the combined impact of reduced biosynthesis and increased degradation, particularly the direct role of peroxidases in anthocyanin catabolism. Overexpression of VviPrx31 decreased anthocyanin contents in Petunia hybrida petals under heat stress condition. These data suggest that high temperature can stimulate peroxidase activity thus anthocyanin degradation in ripening grape berries.
Quantitative Proteomics Analysis of ABA- and GA3-Treated Malbec Berries Reveals Insights into H2O2 Scavenging and Anthocyanin Dynamics

Abstract:Abscisic acid (ABA) and gibberellic acid (GA3) are regulators of fruit color and sugar levels, and the application of these hormones is a common practice in commercial vineyards dedicated to the production of table grapes. However, the effects of exogenous ABA and GA3 on wine cultivars remain unclear. We investigated the impact of ABA and GA3 application on Malbec grapevine berries across three developmental stages. We found similar patterns of berry total anthocyanin accumulation induced by both treatments, closely associated with berry H2O2 levels. Quantitative proteomics from berry skins revealed that ABA and GA3 positively modulated antioxidant defense proteins, mitigating H2O2. Consequently, proteins involved in phenylpropanoid biosynthesis were downregulated, leading to decreased anthocyanin content at the almost ripe stage, particularly petunidin-3-G and peonidin-3-G. Additionally, we noted increased levels of the non-anthocyanins E-viniferin and quercetin in the treated berries, which may enhance H2O2 scavenging at the almost ripe stage. Using a linear mixed-effects model, we found statistical significance for fixed effects including the berry H2O2 and sugar contents, demonstrating their roles in anthocyanin accumulation. In conclusion, our findings suggest a common molecular mechanism by which ABA and GA3 influence berry H2O2 content, ultimately impacting anthocyanin dynamics during ripening.

Abstract:Abstract The effect of elicitors on secondary metabolism in vines is receiving much interest, since it has been shown that they are able to increase the accumulation of phenolics, especially anthocyanins. This research aims to investigate the biochemical and molecular effects of the application of a commercial yeast derivative ( Saccharomyces cerevisiae ) on the accumulation of anthocyanins in potted Sangiovese vines. Experiments were performed on three consecutive years and the yeast derivative was applied at the beginning and at the end of veraison. Technological ripening, accumulation of anthocyanins and expression of the main genes involved in their biosynthesis were assessed. Technological ripening proceeded in a similar way in both treated and untreated berries in the three years. A significant increase in the concentration of anthocyanins was instead detected, following the induction by the yeast derivative of the expression of the genes involved in their biosynthesis. The research highlights the possibility of applying a specific inactivated yeast to increase the anthocyanin concentration even under the current climate change conditions, in Sangiovese, a cultivar extremely sensitive to high temperatures.

Abstract:Summary Shine Muscat(Vitis labruscana Bailey × V.vinifera L.Shine Muscat) is a diploid table grape cultivar resulted from cross breeding of‘Akitsu-21”(‘Steuben”[V.labruscana] × ‘Muscat of Alexandria”[V. vinifera]) with ‘Hakunan”(V.vinifera) released by National Institute of Fruit Tree Science (NIFTS) in Japan.It has a large berry size,bright yellow-green pericarp,crispy and juicy flesh,musky flavor,high soluble solid content and low acidity.Fruit ripening of Shine Muscat was in middle to late August with a long shelf life. The average mass is 10.0 g and 12.4 g for seeded and seedless fruit production,respectively.The seedless berries can be commercially produced by applying gibberellin A3(GA3)to flower and fruit clusters in full bloom and 10 to 1 5 days after full bloom,respectively.Under natural conditions,however,the berries of Shine Muscat are smaller,and grape clusters are relatively sparse.Besides,Shine Muscat grape has the problem of rusty spot incidence,which occurs at the maturation stage just before harvest.Tiny reddish-brown blotches appear on the surface of berries and considerably affected its production quality and commodity value. In this study,3-year-old Shine Muscat trees were selected as the material,and the effects of different concentrations and collocations of GA3 and N-(2-chloro-4-pyridyl)-N′-phenylurea (CPPU) treatments on fruit setting and fruit quality of Shine Muscat grape were studied. The results showed that co-treatment with GA3 and CPPU could significantly improve the fruit setting rate, fruit cluster density,yield stability of Shine Muscat grape;meanwhile,it could also improve the percentage of seedless fruit.The combined use of GA3 and CPPU could remarkably increase single fruit mass,spike mass,total soluble solid content(TSS) and fruit hardness.Higher concentration of CPPU in the early stage would result in higher fruit hollow rate.On the contrary,lower concentration of CPPU in the early stage had no significant effect on fruit enlargement.Meanwhile,higher concentration of CPPU in the later stage would decrease the content of fruit soluble solids. In sum,comprehensive consideration for the fruit cluster compactness,fruit size,sugar degree,aroma component and cost saving,the better co-treatments in production are as follows:1) 25 mg/L GA3 + 10 mg/L CPPU for one time at 5 days after full bloom,or 2) 25 mg/L GA3 +5 mg/L CPPU at 1 3 days right after full bloom,and 25 mg/L GA3 at 10 1 5 days after full bloom(treatment Ⅴ),or 3)25 mg/L GA3 +5 mg/L CPPU at 1 3 days right after full bloom,and 25 mg/L GA3 +5 mg/L CPPU at 10 1 5 days after full bloom (treatmentⅨ).The results will provide technical support on solving the key problems during cultivation of the new cultivar Shine Muscat grape.
From Farm to Fork: Irrigation Management and Cold Storage Strategies for the Shelf Life of Seedless Sugrathirtyfive Table Grape Variety

Abstract:Background: Sustainable water management for table grape has the primary goal of optimizing irrigation through Smart Irrigation (SI) approaches, particularly in Mediterranean regions. In addition, extending the shelf life of table grapes through effective cold storage practices is crucial to meet consumer demands year-round. This research examined the journey "from farm to fork" of Sugrathirtyfive variety (Autumn Crisp® brand), exploring the combined effects of Irrigation Volumes (IV), SO2-Generating Pads (SGPs) and Cold Storage Duration (CSD) on the quality of grapes. Methods: Normal Irrigation (NI—based on the farmer's experience) and SI (100% vine evapotranspiration restored) were supplied in 2023 to Sugrathirtyfive variety white table grape, trained to an overhead tendone system. Yield and quality parameters, berry texture, CIELAB colour coordinates, phenolic content, flavonoids, antioxidant activity and sensory attributes were evaluated on grapes subjected to different times and methods of cold storage. Results: SI grapes showed higher Total Soluble Solids (TSSs) and nutraceutical content, as well as improved CIELAB coordinates with interesting improved berry texture parameters. No differences emerged between single- or dual-release SGPs after 15 days (T1) and 40 days (T2) of CSD. Conclusions: Under our cold storage conditions (3 °C, 85% U.R.), 40 days represent the maximum temporal limit for the cold storage of Sugrathirtyfive variety, regardless of IV, provided they are refrigerated with the aid of SGPs.

Abstract:Phytohormones strongly influence growth, development and nutritional quality of agricultural products by modulating molecular and biochemical changes. The purpose of the present study was to investigate the influence of root restriction (RR) treatment on the dynamic changes of main phytohormones during the berry development and ripening of Summer Black early ripening seedless grape (Vitis vinifera x V. labrusca), and to analyze the changes in the biosynthesis and signal transduction pathways of phytohormones by transcriptomics. Enzyme-linked immunosorbent assay (ELISA) and Ultra Performance Liquid Chromatography-High Resolution Mass Spectrometry (UPLC-HRMS) were used to quantify the phytohormone levels, and RNA-Seq was used to analyze the transcript abundance. The results showed that 23 transcripts involved in the phytohormone biosynthesis and 34 transcripts involved in the signal transduction pathways were significantly changed by RR treatment. RR also increased abscisic acid, brassinosteroid, ethylene, jasmonic acid and salicylic acid levels, while decreasing auxin, cytokinin, and gibberellin contents. The results of the present study suggest that RR treatment can accelerate the grape ripening process, and specific candidate genes were identified for further functional analysis.
Breeding new seedless table grapevines for a more sustainable viticulture in Mediterranean climate

Abstract:The growing demand for sustainable and environmentally friendly viticulture is leading to a multiplication of breeding programs aimed at obtaining vines that are resistant to powdery mildew (PM) and downy mildew (DM), the two most damaging vine diseases. In Puglia, the most important Italian region for the production of table grapes, an extensive crossing program was launched in 2015 with 113 crosses, including elite table varieties, seedless varieties, and resistant varieties. The main seedling production parameters were measured for each cross. In particular, berries harvested as well as the number of seeds and seedlings obtained were considered. Approximately 103,119 seedlings were obtained and subjected to marker-assisted selection for seedlessness using the marker VvAGL11 and for resistance to PM and DM with appropriate markers. Approximately one third (32,638) of the progenies were selected as putative seedless and seventeen thousand five hundred-nine (17,509) were transferred to the field for phenotypic evaluation, including 527 seedless individuals putatively resistant, of which 208 confirmed to be resistant to DM, 22 resistant to PM, and 20 individuals that combined resistance and seedlessness traits. The work discusses the effects of parental combinations and other variables in obtaining surviving progeny and pyramiding genes in table grapes and provides useful information for selecting genotypes and increasing the efficiency of breeding programs for seedless disease-resistant grapes.

Abstract:Protected cultivation is currently one of the main cultivation modes for grape production, but the long-term use of plastic film will have a certain negative impact on the light environment in vineyards, which in turn causes poor colouring, low sugar content and a lack of aroma in some red grape varieties. Supplementing light can be an effective way to mitigate these problems. In this study, vines of three red table grape varieties ('Summer Black', 'Xinyu' and 'Queen Nina') cultivated in a plastic greenhouse were supplemented with red, blue, white and red-blue light from veraison to harvest. All four supplemental light treatments increased the content of anthocyanins, sugars and volatile compounds in three grape varieties compared to CK (no supplemental lighting). Red-blue light treatment was the most favourable for the accumulation of anthocyanins and sugars, and the grapes treated with blue light had the highest content of volatile compounds. The grapes treated with red-blue light all obtained the highest composite scores via principal component analysis. For most of the sensory properties, the highest scores were obtained by the red-blue light-treated grapes. The results of this study will be useful in improving the colouring, sugar, and aroma content of grapes under protected cultivation.

Abstract:Grape berries contain a variety of metabolites, such as anthocyanins, sugars, fatty acids, and antioxidants. Endogenous phytohormones strongly influence these metabolites, which regulate berry quality improvement. In this study, we evaluated the effects of 2,4-epibrassinolide (EBR, brassinolide (BR)-like growth regulator), jasmonic acid (JA), and their signaling inhibitors brassinazole (Brz), and sodium diethyldithiocarbamate (DIECA) on berry quality and antioxidant ability. Overall, the pre-harvest application of 0.5 mg L -1 EBR and 100 μmol L -1 JA significantly influences the quality of the grape berry. Results showed that EBR was superior to other treatments at enhancing the content of different metabolites, including anthocyanins, fructose, glucose, and a variety of fatty acids, in grapes. EBR and JA also enhanced the synthesis of gibberellin 3 (GA 3 ), cytokinin (CTK), salicylic acid (SA), JA, methyl jasmonate (MeJA), BR, and abscisic acid (ABA), while inhibiting the synthesis of auxin (IAA). Most genes related to BR/JA and anthocyanins/sugars/fatty acids biosynthesis were up-regulated. The effects of Brz and DIECA on the grape berry quality were totally reversed throughout the study, as shown by EBR and JA. According to correlation analysis, EBR and JA have a beneficial positive interaction that promotes the formation of strong coherences in grape berries between ABA/IAA/ZT-fruit expansion, BR/JA/MeJA/GA 3 /ZR-biochemical characteristics development, JA/MeJA/ABA/GA 3 /SA/ZR-antioxidant capacity enhancement, and JA/MeJA/IAA/GA 3 /ZT/ZR-fatty acids accumulation. In this regard, we concluded that preharvest exogenous 0.5 mg L -1 EBR and 100 μmol L -1 JA is a successful way to improve grape berry quality.
