Abstract:Cancer cells metastasize and are transported in the bloodstream, easily reaching any site in the body through the blood circulation. A method designed to assess the number of circulating tumor cells (CTCs) should be validated as a clinical tool for predicting the response to therapy and monitoring disease progression in patients with cancer. Although CTCs are detectable in many cases, they remain unavailable for clinic usage due to their high testing cost, tedious operation and poor clinical relevance. Herein, we developed a regeneratable microchip for isolating CTCs that is available for robust cell heterogeneity assays on-site without the need for a sterile environment. The ivy-like hierarchical roughened zinc oxide (ZnO) nanograss interface was synthesized and directly integrated into the microfluidic devices and enables effective CTC capture and flexible, nontoxic CTC release during incubation in a mildly acidic solution, thus enabling cellular and molecular analyses. The microchip can be regenerated and recycled to capture CTCs with the remaining ZnO without affecting the efficiency, even after countless cycles of cell release. Moreover, microbial infection is avoided during its storage, distribution and even in open-space usage, which ideally appeals to the demands of point-of-care (POC) and home testing and meets to the requirements for blood examinations in undeveloped or resource-limited settings. Furthermore, the findings generated using this platform based on the cocktail of anti-epithelial cell adhesion molecule (EpCAM) and anti-vimentin antibodies indicate that CTC capture was more precise and reasonable for patients with advanced cancer.

Abstract:CD45RA+ cells have been described to be less responsive to CD3/T cell receptor (TcR)‐mediated activation than CD45R0+ T cells. To analyze the underlying mechanism of the differential responses we compared CD3/TcR‐triggered tyrosine phosphorylation in the two subsets and studied the role of co‐stimulatory signals provided either by accessory cells or pharmacologic activation of protein kinase C by phorbol ester. Stimulation of purified CD45RA+ and CD45R0+ T cells with CD3/TcR antibodies induced similar patterns and intensities of tyrosine phosphorylation in the two subsets, but no proliferation. If accessory cells were used as the source of co‐stimulatory signals, strong expression of the 55‐kDa chain of the interleukin‐2 (IL‐2) receptor (CD25), significant IL‐2 production and vigorous proliferation were observed in CD45R0+ cells, whereas CD45RA+ cells responded weakly. However, when CD3/TcR‐mediated triggering was combined with activation of protein kinase C by phorbol ester, CD45RA+ cells responded strongly. These data indicate that the transmembrane signaling capacity of the T cell receptor expressed by CD45RA+ and CD45R0+ cells is similar and, therefore, is presumably not responsible for the differential reactivities of the two subsets. It is more likely that co‐stimulatory signals determine whether CD3/TcR‐initiated activation results in strong or weak responses.

Abstract:Circulating tumor cells (CTCs) in blood is the direct cause of tumor metastasis. The isolation and detection of CTCs in whole blood is very important and of clinical value in early diagnosis, postoperative review and personalized treatment. It is difficult to separate all types of CTCs that efficiently rely on a single path due to cancer cell heterogenicity. Here, we designed a new kind of "filter chip" for the retention of CTCs with very high efficiency by integrating the effects of cell size and specific antigens on the surface of tumor cells. The "filter chip" consists of a semicircle arc and arrays and can separate large-scale CTC-microspheres, which combined with CTCs automatically. We synthesized interfacial zinc oxide coating with nanostructure on the surface of the microsphere to increase specific surface area to enhance capturing efficiency of CTCs. Microspheres, trapped in the arrays, would entrap CTCs, too. The combination of the three kinds of strategies resulted in above 90% of capture efficiency for different tumor cell lines. Furthermore, it is easy to find and isolate the circulating tumor cells from the chip as tumor cells would be fixed inside the structure of "filter chip". To avoid the high background contamination when a few CTCs are surrounded by millions of nontarget cells, a digital detection method was applied to improve detection sensitivity. CTCs in whole blood were specifically labeled by the antibody-DNA conjugates and were detected via the DNA of the conjugates with a signal amplification. The strategy of antibody-functional microspheres integrated microchip for cell sorting and the detection of CTCs may find broad implications that favor fundamental cancer biology research, the precise diagnosis and monitoring of cancer in the clinics.

Abstract:The enumeration of circulating tumor cells (CTCs) has proved valuable for early detection and prognosis in cancer treatment. In this work, an optimized design of a microfluid-based immunomagnetic chip is proposed to separate CTCs from whole blood. Unlike most reported immimomagnetic-based methods in which the magnetic field is applied perpendicular to the flow direction, here, the magnetic field is applied parallel to the flow direction, which makes the hydrodynamic force not the dominant force to hamper cell capture. Experimental validation showed that this novel chip can be operated at a flow rate of up to 6 ml/h with 90% CTC capture efficiency.

Abstract:A B S T R A C T Despite the recent advancement of biotechnology and pharmaceutical research, cancers remain the leading cause of human mortality. It is vital to diagnose cancers at an early stage when treatment can dramatically improve prognosis. So far, low-cost and easy to operate devices, which allow efficient isolation and sensitive detection of circulating tumor cells (CTCs) for routine blood screening, remain lacking. This talk will introduce a novel micro fluidic platform which can isolate CTCs from the real blood sample in 30 minutes: this system includes a high throughput blood cell separation chip which can separate white blood cells with CTCs from red blood cells and platelets by inertial and suction actions; a nano structured surface which can allow higher retention rate of CTCs on the surface for sample enrichment by 100 folds from 1/107 upto 1/105 CTCs/WBCs.and the enriched sample will go through a final cells self-assembly process into a densed monolayer on a cell assembly chip for in parallel inspection at high speed. As a result, the CTCs can be identified in 30 minutes by the integration of these three chips altogether. Isolated CTCs will still be in vital and can be further characterized and cultivated for the identification of cancer stem cells for prognosis

Abstract:Circulating tumor cells (CTCs) serve as crucial indicators for tumor occurrence, progression, and prognosis monitoring. However, achieving high sensitivity and high purity capture of CTCs remains challenging. Additionally, in situ capture and synchronous clearance hold promise as methods to impede tumor metastasis, but further exploration is needed. In this study, biomimetic cell membrane-coated magnetic nanoparticles (NPs) were designed to address the issue of nonspecific adsorption of capture probes by the immune system during blood circulation. Membranes from human breast cancer cells (tumor cell membranes, TMs) and leukocytes (white blood cell membranes, WMs) were extracted and fused to form a hybrid membrane (HM), which was further modified onto the surface of porous magnetic NPs loaded with indocyanine green (ICG). The incorporation of TM enhanced the material's target specificity, thus increasing capture efficiency, while WM coating reduced interference from homologous white blood cells (WBCs), further enhancing capture purity. Additionally, in conjunction with our novel inverted microfluidic chip, this work introduces the first use of polymer photonic crystals as the capture interface for CTCs. Besides providing an advantageous surface structure for CTC attachment, the 808 nm photonic bandgap effectively amplifies the 808 nm excitation light at the capture surface position. Therefore, upon capturing CTCs, the ICG molecules in the probes facilitate enhanced photothermal (PTT) and photodynamic (PDT) synergistic effects, directly inactivating the captured CTCs. This method achieves capture efficiency and purity exceeding 95% and permits in situ inactivation post-capture, providing an important approach for future research on impeding tumor metastasis in vivo.

Abstract:The efficient isolation and the accurate phenotype discrimination of circulating tumor cells (CTCs) are expected to provide much valuable information for the understanding of tumor metastasis and to play an important role in personalized treatment of cancer patients in the future. In this study, we developed a novel, rapid, and simple method for efficient capture and accurate identification of CTCs using aptamer conjugated magnetic beads and surface-enhanced Raman scattering (SERS) imaging technique. Using aptamer conjugated magnetic beads, rare target cancer cells can be captured efficiently from buffer and whole blood sample with capture efficiency of 73 % and 55 %, respectively. Meanwhile, captured cancer cells were labeled by specific SERS probes and can be identified readily and accurately by SERS imaging technique. Results of our experiment demonstrate the potential feasibility of aptamer conjugated magnetic beads coupled with SERS imaging technique for the efficient capture and accurate discrimination of CTCs in clinical whole blood sample. Abstract:Background: Circulating tumor cells (CTCs) are considered as a useful biomarker for early cancer diagnosis, which play a crucial role in metastatic process. Unfortunately, the tumor heterogeneity and extremely rare occurrence rate of CTCs among billions of interfering leukocytes seriously hamper the sensitivity and purity of CTCs isolation. Methods: To address these, we firstly used microfluidic chips to detect the broad-spectrum of triple target combination biomarkers in CTCs of 10 types of cancer patients, including EpCAM, EGFR and Her2. Then, we constructed hybrid engineered cell membrane-camouflaged magnetic nanoparticles (HE-CM-MNs) for efficient capture of heterogeneous CTCs with high-purity, which was enabled by inheriting the recognition ability of HE-CM for various CTCs and reducing homologous cell interaction with leukocytes. Compared with single E-CM-MNs, HE-CM-MNs showed a significant improvement in the capture efficiency for a cell mixture, with an efficiency of 90%. And the capture efficiency of HE-CM-MNs toward 12 subpopulations of tumor cells was ranged from 70 to 85%. Furthermore, by using HE-CM-MNs, we successfully isolated heterogeneous CTCs with high purity from clinical blood samples. Finally, the captured CTCs by HE-CM-MNs could be used for gene mutation analysis. Conclusions: This study demonstrated the promising potential of HE-CM-MNs for heterogeneous CTCs detection and downstream analysis.
Playing the ‘trust game’ with robots: Social strategies and experiences

Abstract:We present the results of a pilot study that investigates if and how people judge the trustworthiness of a robot during social Human-Robot Interaction (sHRI). Current research in sHRI has observed that people tend to interact with robots socially. However, results from neuroscience suggests people use different cognitive mechanisms interacting with robots than they do with humans, leading to a debate about whether people truly perceive robots as social entities. Our paper focuses on one aspect of this debate, by examining trustworthiness between people and robots using behavioral economics' `Trust Game' scenario. Our pilot study replicates a trust game scenario, where a person invests money with a robot trustee in hopes they will receive a larger sum (trusting the robot to give more back), then gets a chance to invest once more. Our qualitative analysis of investing behavior and interviews with participants suggests that people may follow a human-robot (h-r) trust model that is quite similar to the human-human trust model. Our results also suggest a possible resolution to the sHRI and Neuroscience debate: people try to interact socially with robots, but due to lack of common social cues, they draw from social experience, or create new experiences by actively exploring the robot behavior.

Abstract:Circulating tumor cell (CTC) and their isolation is one potential clinically useful target within a “liquid biopsy”. Detection and characterization of CTCs provide a non-invasive approach for monitoring cancer therapy. In the present research, a magnet- deformability hybrid integrated microfluidic chip was designed to enumerate CTCs. Arrays of narrow gaps (patterned by micro-ellipse microposts) were designed to capture CTCs bonded with magnetic immune beads. Eleven samples, including Gastric of Colorectal, non-small-cell lung cancer (NSCLC), Ureter cancer, and Breast cancer, were used to validate the microfluidic device clinically. Clinical test results show a high capture efficiency (over 90% at 3ml/h) and high viability (96%) at high flow rate. Furthermore, comparisons of device performance with and without magnet beads were also conducted. This hybrid chip design demonstrated excellent performance in the clinical enumeration of CTCs, which is promising in the future cancer therapy evaluation.

Abstract:The effective detection and release of circulating tumor cells (CTCs) are of great significance for cancer diagnosis and monitoring. The microfluidic technique has proved to be a promising method for CTCs isolation and subsequent analysis. However, complex micro-geometries or nanostructures were often constructed and functionalized to improve the capture efficiency, which limited the scale-up for high-throughput production and larger-scale clinical applications. Thus, we designed a simple conductive nanofiber chip (CNF-Chip)-embedded microfluidic device with a herringbone microchannel to achieve the efficient and specific capture and electrical stimulation-triggered rapid release of CTCs. Here, the most used epithelial cell adhesion molecule (EpCAM) was selected as the representative biomarker, and the EpCAM-positive cancer cells were mainly studied. Under the effects of the nanointerface formed by the nanofibers with a rough surface and the herringbone-based high-throughput microfluidic mixing, the local topographic interaction between target cells and nanofibrous substrate in the microfluidic was synergistically enhanced, and the capture efficiency for CTCs was further improved (more than 85%). After capture, the sensitive and rapid release of CTCs (release efficiency above 97%) could be conveniently achieved through the cleavage of the gold-sulfur bond by applying a low voltage (-1.2 V). The device was successfully used for the effective isolation of CTCs in clinical blood samples from cancer patients, indicating the great potential of this CNF-Chip-embedded microfluidic device in clinical applications.

Abstract:Efficient capture data on circulating tumor cells (CTCs) determines early-stage cancer diagnosis and contributes to timely clinical treatment. We present an enhanced method of capturing CTCs accomplished by a microfluidic device integrated with magnetic field to activate the kinetic motion of in-device magnetic beads. The device, consisting of a microfluidic chamber and two electrode chips applied with pulsatile alternating current in both, is designed based on simulations on the periodicity characteristic of magnetic beads as well as the effect of heat dissipation on cell culture medium manifest. Using MEMS technologies, the prototype is fabricated and assembled. The cell capture experiments based on active magnetic beads are achieved in separation of rare cancer cells (MCF7 cells) with low concentration. The capture rate is estimated up to 88 %, with great potential of dramatically improving detection efficiency in disease diagnostics.

Abstract:With the growing demands of the early, accurate, and sensitive diagnosis for cancers, the development of new diagnostic technologies becomes increasingly important. In this study, we proposed a strategy for efficient capture and sensitive detection of circulating tumor cells (CTCs) using an array nanochannel-ion channel hybrid coupled with an electrochemical detection technique. The aptamer probe was immobilized on the ion channel surface to couple with the protein overexpressed on the CTCs membrane. Through this special molecular recognition, CTCs can be selectively captured. The trapped CTCs cover the ion channel entrance efficiently, which will dramatically block the ionic flow through channels, resulting in a varied mass-transfer property of the nanochannel-ion channel hybrid. On the basis of the changed mass-transfer properties, the captured CTCs can be sensitively detected using the electrochemical linear sweep voltammetry technique. Furthermore, due to the amplified response of array channels compared to that of a single channel, the detection sensitivity can be enhanced greatly. The results showed that acute leukemia CCRF-CEM (a type of CTC) concentration as low as 100 cells mL-1 can be successfully captured and detected. The present method provides a simple, sensitive, and label-free technique for CTCs capture, detection, and release, which would hold great potential in the early clinical diagnosis and treatment of cancers.
Efficient Detection of Single Circulating Tumor Cell in Blood Using Raman Mapping Based on Aptamer-SERS Bio-Probe Coupled with Micropore Membrane Filtration.

Abstract:Rapid and accurate detection of rare circulating tumor cells (CTCs) in human blood still remains a challenge. We present a surface enhanced Raman spectroscopy (SERS) method based on aptamer-SERS bio-probe recognition coupled with micropore membrane filtration capture for the detection of CTCs at single cell level. The parylene micropore membrane with optimized micropore size installed on a filtration holder could capture bio-probe labeled CTCs by gravity in less than 10 s, and only with very less white blood cells (WBCs) residual. In order to facilitate the synthesis of the aptamer-SERS bio-probe, ethyl acetate dehydration method was established. The bio-probe can be rapidly synthesized within 2 h by binding SH-aptamer to 4- mercaptobenzoic acid (4-MBA) modified AuNPs with the help of ethyl acetate. The SERS bio-probe with selected specific aptamer could distinguish single human non-small cell lung cancer A549 cells from residual WBCs on membrane efficiently and reliably based on their Raman signal intensity difference at 1075 cm- 1. Through the filter membrane coupled with aptamer-SERS bio-probe system, even 20 A549 cells in blood solution simulating CTCs sample can be detected, which the recovery rate and recognition rate are more than 90%. This method is rapid, reliable and cost-effective, which indicates a good prospect in clinical application for CTCs detection.

Abstract:BACKGROUND: Circulating tumor cells (CTCs) are detectable in peripheral blood of metastatic lung cancer patients. In this article, we evaluate a new CTC separation method based on a combination of anti-EpCAM and immunomagnetic beads with the aim to detect CTCs more conveniently and specifically.METHODS: Lung cancer cells were magnetically labeled by anti-EpCAM magnetic beads, and subsequently captured by magnetic separation using our novel device. Isolated lung cancer cells were identified by pathomorphological by hematoxylin-eosin staining protocol. The system was used to detect CTCs in 2 ml blood. Blood samples of healthy donors spiked with lung cancer cell line A549 cells were used to determine the sensitivity and specificity of the method. Prevalence of CTCs was examined in samples from 56 patients with lung cancer.RESULTS: Regression analysis of number of recovered versus spiked A549 cells yielded a coefficient of determination of R(2) = 0.996 (P < 0.001). The average recovery was 68% or more at each spiking level. The coefficient of variation increased as the number of spiked cells decreased, ranging from 6.4% (1,000-cell spike) to 18.4% (50-cell spike). Forty-nine of the fifty-six patients (87.5%) were found to have CTCs in peripheral blood. None of the 2 ml peripheral blood samples of the 20 healthy subjects analyzed were found to have CTCs.CONCLUSIONS: This novel turbulence device provides a new tool allowing for feasible and specific detection of CTCs in lung cancer patients. It is likely clinically useful in diagnosis and monitoring of lung cancer and may have a role in clinical decision making.

Abstract:Detection of circulating tumor cells (CTCs) has provided a noninvasive and efficient approach for early diagnosis, treatment, and prognosis of cancer. However, efficient capture of CTCs in the clinical environment is very challenging because of the extremely rare and heterogeneous expression of CTCs. Herein, we fabricated a multimarker microfluidic chip for the enrichment of heterogeneous CTCs from peripheral blood samples of breast cancer patients. The multimarker aptamer cocktail DNA nanostructures (TP-multimarker) were modified on a deterministic lateral displacement (DLD)-patterned microfluidic chip to enhance the capture efficiency through the size selection effect of DLD arrays and the synergistic effect of multivalent aptamers. As compared to a monovalent aptamer-modified chip, the multimarker chip exhibits enhanced capture efficiency toward both high and low epithelial cell adhesion molecule expression cell lines, and the DNA nanostructure-functionalized chip enables the accurate capture of different phenotypes of CTCs. Abstract:Highly crystalline metastable bismuth titanate (Bi₂₀TiO₃₂) nanosheets are prepared via a simple green wet chemical route for the first time. The Bi₂₀TiO₃₂)photocatalysts were characterized by transmission electron microscopy (TEM), field emission scanning electron microscopy (FESEM), energy dispersive spectrum analysis (EDS), X-ray diffraction (XRD), N₂ adsorption-desorption (BET), and UV-vis diffuse reflectance spectroscopy (DRS). Inspiringly, Bi₂₀TiO₃₂ nanosheets showed high photocatalytic activity for the degradation of nonbiodegradable azo dye under simulated sunlight and visible-light irradiation. The experimental results showed that the photocatalytic activity of the Bi₂₀TiO₃₂ nanosheets was superior to the commercial Degussa P25 TiO₂, and demonstrated that the morphology and crystal structure have a distinct effect on the photocatalytic activity. The reasons for the high photocatalytic activity and the formation mechanism of Bi₂₀TiO₃₂ nanosheets are also discussed.
