Abstract:Boron (B) is essential for plant growth, and B deficiency causes severe losses in crop yield. Here we isolated and characterized a rice (Oryza sativa L.) mutant named dwarf and tiller-enhancing 1 (dte1), which exhibits defects under low-B conditions, including retarded growth, increased number of tillers and impaired pollen fertility. Map-based cloning revealed that dte1 encodes a NOD26-LIKE INTRINSIC PROTEIN orthologous to known B channel proteins AtNIP5;1 in Arabidopsis and TASSEL-LESS1 in maize. Its identity was verified by transgenic complementation and RNA-interference. Subcellular localization showed DTE1 is mainly localized in the plasma membrane. The accumulation of DTE1 transcripts both in roots and shoots significantly increased within 3h of the onset of B starvation, but decreased within 1h of B replenishment. GUS staining indicated that DTE1s are expressed abundantly in exodermal cells in roots, as well as in nodal region of adult leaves. Although the dte1 mutation apparently reduces the total B content in plants, it does not affect in vivo B concentrations under B-deficient conditions. These data provide evidence that DTE1 is critical for vegetative growth and reproductive development in rice grown under B-deficient conditions.
Hexagonal crystal structure of the A-DNA octamer d(GTGTACAC) and its comparison with the tetragonal structure: correlated variations in helical parameters.

Abstract:The alternating DNA octamer d(GTGTACAC) has been grown in a novel hexagonal crystal form. The structure has been determined and refined to a 2-A resolution, with 51 water molecules. The A-DNA conformation is a variant of that observed for the tetragonal form of the same sequence (Jain et al., 1989) containing a bound spermine. The crystals belong to the space group P6(1)22, a = b = 32.40 A and c = 79.25 A, with one strand in the asymmetric unit. The new hexagonal structure was solved by rotation and translation searches in direct space and refined to a final R value of 12.7% by using 1561 unique reflections greater than 1.5 sigma (I). The electron density clearly shows that the penultimate A7 sugar had flipped into the alternative C2'-endo pucker. This dent in the molecule can be attributed to close intermolecular contacts. In contrast, in the tetragonal structure, the DNA is distorted in the central TA step, where the A5 backbone bonds C4'-C5' and O5'-P assume trans conformations. The hexagonal double helix more closely resembles the fiber diffraction A-DNA, compared to the tetragonal form. For instance, the tilt angle is higher (16 degrees vs 10 degrees), which is correlated with a larger displacement from the helix axis (3.5 vs 3.3), a lower rise per residue (2.9 vs 3.2), and a smaller major-groove width (6.1 vs 8.7), thus indicating that the variations in these global helical parameters are correlated. The propeller twist angles in both forms are higher for the G-C base pairs (15.3 degrees, 12.14 degrees) than for the A-T base pairs (10.8 degrees, 9.1 degrees), which is the reverse of the expected order. Unlike the tetragonal structure, the hexagonal crystal structure interestingly does not contain a bound spermine molecule. Our analysis reveals that the conformational differences between the tetragonal and hexagonal forms are not entirely due to the spermine binding, and crystal packing seems to play an important role.
BnaC4.BOR2 mediates boron uptake and translocation in Brassica napus under boron deficiency

Abstract:Boron (B) is an essential microelement in plant growth and development. However, the molecular mechanisms underlying B uptake and translocation in Brassica napus are poorly understood. Herein, we identified a low‐B (LB)‐inducible gene, namely BnaC4.BOR2, with high transcriptional activity in root tips, stele cells, leaves, and floral organs. The green fluorescence protein labelled BnaC4.BOR2 protein was localised to the plasma membrane to demonstrate the B efflux activity in yeast and Arabidopsis. BnaC4.BOR2 knockout considerably reduced B concentration in the root and xylem sap, and altered B distribution in different organs at low B supply, exacerbating B sensitivity at the vegetative and reproductive stages. Additionally, the grafting experiment showed that BnaC4.BOR2 expression in the roots contributed more to B deficiency adaptability than that in the shoots. The pot experiments with LB‐soil revealed B concentration in leaves and siliques of BnaC4.BOR2 mutants were markedly reduced, showing an obvious B‐deficient phenotype of 'flowering without seed setting' and a considerable reduction in seed yield in B‐deficient soil. Altogether, the findings of this study highlight the crucial role of BnaC4.BOR2 in B uptake and translocation during B. napus growth and seed yield under LB conditions.

Abstract:Vascular tissues essentially regulate water, nutrient, photo-assimilate, and phytohormone logistics throughout the plant body. Boron (B) is crucial for the development of the vascular tissue in many dicotyledonous plant taxa and B deficiency particularly affects the integrity of phloem and xylem vessels, and, therefore, functionality of long-distance transport. We hypothesize that changes in the plants’ B nutritional status evoke differential responses of the vasculature and the mesophyll. However, direct analyses of the vasculature in response to B deficiency are lacking, due to the experimental inaccessibility of this tissue. Here, we generated biochemical and physiological understanding of B deficiency response reactions in common plantain (Plantago major L.), from which pure and intact vascular bundles can be extracted. Low soil B concentrations affected quantitative distribution patterns of various phytohormones, sugars and macro-, and micronutrients in a tissue-specific manner. Vascular sucrose levels dropped, and sucrose loading into the phloem was reduced under low B supply. Phytohormones responded selectively to B deprivation. While concentrations of abscisic acid and salicylic acid decreased at low B supply, cytokinins and brassinosteroids increased in the vasculature and the mesophyll, respectively. Our results highlight the biological necessity to analyze nutrient deficiency responses in a tissue- rather organ-specific manner.

Circadian Rhythm and Nitrogen Metabolism Participate in the Response of Boron Deficiency in the Root of Brassica napus

Abstract:Boron (B) deficiency has been shown to inhibit root cell growth and division. However, the precise mechanism underlying B deficiency-mediated root tip growth inhibition remains unclear. In this study, we investigated the role of BnaA3.NIP5;1, a gene encoding a boric acid channel, in Brassica napus (B. napus). BnaA3.NIP5;1 is expressed in the lateral root cap and contributes to B acquisition in the root tip. Downregulation of BnaA3.NIP5;1 enhances B sensitivity in B. napus, resulting in reduced shoot biomass and impaired root tip development. Transcriptome analysis was conducted on root tips from wild-type B. napus (QY10) and BnaA3.NIP5;1 RNAi lines to assess the significance of B dynamics in meristematic cells during seedling growth. Differentially expressed genes (DEGs) were significantly enriched in plant circadian rhythm and nitrogen (N) metabolism pathways. Notably, the circadian-rhythm-related gene HY5 exhibited a similar B regulation pattern in Arabidopsis to that observed in B. napus. Furthermore, Arabidopsis mutants with disrupted circadian rhythm (hy5/cor27/toc1) displayed heightened sensitivity to low B compared to the wild type (Col-0). Consistent with expectations, B deficiency significantly disrupted N metabolism in B. napus roots, affecting nitrogen concentration, nitrate reductase enzyme activity, and glutamine synthesis. Interestingly, this disruption was exacerbated in BnaA3NIP5;1 RNAi lines. Overall, our findings highlight the critical role of B dynamics in root tip cells, impacting circadian rhythm and N metabolism, ultimately leading to retarded growth. This study provides novel insights into B regulation in root tip development and overall root growth in B. napus.
Natural variation in the Arabidopsis thaliana root and shoot response to boron deficiency reveals sensitive and responsive phenes and phylogenetic and geographic clustering of boron efficiency adaptations

Abstract:Boron (B) is an essential plant micronutrient with a crucial role in cell wall stability. To improve plant tolerance to suboptimal B-availability, it is crucial to understand the mechanisms plants have evolved to tolerate B-limited conditions. We assessed temporal physiological, ionomic and molecular responses to B-deficiency across a diversity panel of 185 Arabidopsis thaliana accessions. Plants were characterised in soil-substrate in an automated phenotyping system to track shoot performance, and on agar plates to track root traits. Accessions varied widely in their response to B-deficiency. Rosette area decreased by 50% under B-deficiency across all accessions on average but by less than 15% in B-efficient accessions alone. Most root traits suffered under B-deficiency but lateral root length increased in 20% of accessions. The B-uptake and B transporter expression response of B-efficient and B-inefficient accessions was similar, suggesting increased B-use rather than B-uptake efficiency in efficient accessions. Phylogenetic analysis demonstrated enrichment of B-efficient accessions within a clade containing accessions originating from areas characterised as B-deficient. K-means clustering revealed increased lateral root length was associated with efficient shoot growth under B-deficiency. The results demonstrate different response strategies to B-deficiency, indicating target traits to improve the performance of plants grown under B-limiting conditions.

Abstract:Sugar beet (Beta vulgaris L.) has a high demand for B, and B deficiency inhibits normal growth and productivity. However, there is a lack of information on how B deficiency affects the growth of beet at the transcriptome level, and the factors that govern B utilisation efficiency. This study aimed to identify the genes differentially expressed under B deficiency and those that underlie the mechanisms of efficient B use in two sugar beet cultivars. Accordingly, B-efficient (H, KWS1197) and B-inefficient (L, KWS0143) sugar beet cultivars were used, and two levels of boron were employed in the hydroponic experiments: B0.1 (0.1 μM B, deficiency) and B50 (50 μM B, CK). The results showed that B deficiency inhibited leaf growth, significantly reduced B concentration and B transfer coefficient, and increased peroxidase (POD) activity and malondialdehyde and proline content. The transcriptome data showed that the B-efficient variety exhibited more differentially expressed genes than the B-inefficient variety. Metabolic pathways were the most critical pathways involved in the B deficiency response. The expression of POD, bHLH, WRKY transcription factors, and nodulin26-like intrinsic protein (NIP5;1) were upregulated in the KWS1197 variety. In conclusion, the KWS1197 variety had physiological advantages and a highly efficient B utilisation molecular mechanism, contributing to a high B deficiency tolerance. This study provides a theoretical basis for the adaptation mechanism to B deficiency in sugar beets.

Abstract:Although boron deficiency (BD) is a drastic disorder to the agriculture crops, its effects on watermelon still unknown. Therefore, the present study investigates the effect of BD (0 and 0.5mg L-1) on different morphological, physiological and biochemical traits, and mineral uptake during early seedlings stage of watermelon. B-deficiency induced leaf chlorosis in watermelon initiated from the leaf margins and tips. Despite that, BD increased shoot length, and root and shoot dry weight of BD-watermelon seedlings. BD decreased leaf chlorophyll and carotenoid contents, and photosynthetic parameters without affecting quantum yield of PSII. BD significantly inhibited total soluble protein accumulation, while leaf proline content was unaffected. A significant increase in antioxidant enzyme activities in response to higher hydrogen peroxide (H2O2) generation could possibly reduced lipid peroxidation and promoted BD-watermelon plant growth. BD influenced mineral uptake variously: the induction of phosphorus (P) and zinc (Zn), and the preservation of potassium (K), calcium (Ca) and molybdenum (Mo) levels in the leaves could maintain watermelon growth; while, the inhibition of magnesium (Mg) uptake seemed to be responsible for suppressed leaf chlorophyll and photosynthesis. These results would further help in understanding the physiological pathways and mechanisms associated with BD especially in watermelon, and provide database for fertilizer recommendations in B-deficient areas.

Abstract:Nitric oxide (NO) mediates plant response to a variety of abiotic stresses; however, limited information is available on its effect on boron (B)-stressed watermelon plants. The present study investigates the mechanism through which NO protects watermelon seedlings from B deficiency and toxicity stresses. Five days old watermelon seedlings were exposed to B (0, 0.5 and 10 mg L-1) alone or with 75 mole of NO donor sodium nitroprusside (SNP) for 30 days. Both low and high B concentrations in the media altered nutrient accumulation and impaired various physiological processes of watermelon seedlings, leading to a significant reduction in biomass production. The plants exposed to B deficient or toxic concentrations had 66 and 69% lower shoot dry weight, respectively compared with optimum B levels. B toxicity-induced growth inhibition of watermelon seedlings was associated with high B translocation to shoot tissues, which caused lipid membrane peroxidation (12% increase) and chlorophyll destruction (25% reduction). In contrast, B deficiency accelerated generation of reactive oxygen species (ROS), specifically OH-1 and induced cellular oxidative injury. Exogenously applied SNP promoted leaf chlorophyll, photosynthesis and consequently biomass production in B-stressed watermelon seedlings by reducing B accumulation, lipid membrane peroxidation and ROS generation. It also activated antioxidant enzymes such as SOD, POD and APX, and protected the seedlings from ROS-induced cellular burst. (C) 2016 Published by Elsevier Masson SAS.

Abstract:Boron (B) deficiency and consequent limitation of plant yield and quality, particularly of sugar beet (Beta vulgaris L.) has emerged as a maior problem,which is exacerbating due to cultivar dependent variability in B deficiency tolerance. Pertinently, the current study was designed to elucidate the physiological and molecular mechanisms of B deficiency tolerance of sugar beet varieties KWS1197 (B-efficient variety) and KWS0143 (B-inefficient variety). A hydroponic experiment was conducted employing two B levels B0.1 (0.1 μM L−1 H3BO3, deficiency) and B50 (50 μM L−1 H3BO3, adequacy). Boron deficiency greatly inhibited root elongation and dry matter accumulation; however, formation of lateral roots stimulated and average root diameter was increased. Results exhibited that by up-regulating the expression of NIP5-1, NIP6-1, and BOR2, and suppressing the expression of BOR4, cultivar KWS1197, in contrast to KWS0143, managed to transfer sufficient amount of B to the aboveground plant parts, facilitating its effective absorption and utilization. Accumulation of malondialdehyde (MDA) and reactive oxygen species (ROS) was also mellowed in KWS1197, as well as the oxidative damage to root cells via preservation of the antioxidant enzyme system. Additionally, the expression of essential enzymes for biosynthesis of phytohormone (PYR/PYL) and lignin (COMT, POX, and CCoAOMT) were found to be highly up-regulated in KWS1197. Deductively, through effective B absorption and transportation, balanced nutrient accumulation, and an activated antioxidant enzyme system, B-efficient cultivars may cope with B deficiency while retaining a superior cellular structure to enable root development.

Abstract:When faced with salinity stress, plants typically exhibit a slowdown in their growth patterns. Boron (B) is an essential micronutrient for plants that are known to play a critical role in controlling cell wall properties. In this study, we used the model plant Arabidopsis thaliana Col-0 and relevant mutants to explore how the difference in B availability may modulate plant responses to salt stress. There was a visible root growth suppression of Col-0 with the increased salt levels in the absence of B while this growth reduction was remarkably alleviated by B supply. Pharmacological experiments revealed that orthovanadate (a known blocker of H+-ATPase) inhibited root growth at no B condition, but had no effect in the presence of 30 μM B. Salinity stress resulted in a massive K+ loss from mature zones of A. thaliana roots; this efflux was attenuated in the presence of B. Supplemental B also increased the magnitude of net H+ pumping by plant roots. Boron availability was also essential for root halotropism. Interestingly, the aha2Δ57 mutant with active H+-ATPase protein exhibited the same halotropism response as Col-0 while the aha2-4 mutant had a stronger halotropism response (larger bending angle) compared with that of Col-0. Overall, the ameliorative effect of B on the A. thaliana growth under salt stress is based on the H+-ATPase stimulation and a subsequent K+ retention, involving auxin- and ROS-pathways.

Abstract:Boron is an essential micronutrient for plant growth as it participates in cell wall integrity. The growth and development of Acacia melanoxylon stem can be adversely affected by a lack of boron. To explore the mechanism of boron deficiency in A. melanoxylon stem, the changes in morphological attributes, physiological, endogenous hormone levels, and the cell structure and component contents were examined. In addition, the molecular mechanism of shortened internodes resulting from boron deficiency was elucidated through transcriptome analysis. The results showed that boron deficiency resulted in decreased height, shortened internodes, and reduced root length and surface area, corresponding with decreased boron content in the roots, stems, and leaves of A. melanoxylon . In shortened internodes of stems, oxidative damage, and disordered hormone homeostasis were induced, the cell wall was thickened, hemicellulose and water-soluble pectin contents decreased, while the cellulose content increased under boron deficiency. Furthermore, plenty of genes associated with cell wall metabolism and structural components, including GAUTs , CESAs , IRXs , EXPs , TBLs , and XTHs were downregulated under boron deficiency. Alterations of gene expression in hormone signaling pathways comprising IAA, GA, CTK, ET, ABA, and JA were observed under boron deficiency. TFs, homologous to HD1 s, NAC10 , NAC73 , MYB46s , MYB58 , and ERF92s were found to interact with genes related to cell wall metabolism, and the structural components were identified. We established a regulatory mechanism network of boron deficiency-induced shortened internodes in A. melanoxylon based on the above results. This research provides a theoretical basis for understanding the response mechanism of woody plants to boron deficiency.

Abstract:Soil salinity is a major environmental constraint affecting agricultural production systems. Boron (B) is an essential micronutrient for plants. This work was aimed to reveal the mechanistic basis of amelioration of detrimental effects of salinity on plant performance by boron. Barley ( Hordeum vulgare ) plants were exposed to different boron (H 3 BO 3 , from 0 to 1 mM) and salt (NaCl, 0 and 100 mM) treatments demonstrating salt-tolerant phenotype in the presence of B. This was manifested by the significant increase in leaf and root fresh weight, enhanced CO 2 assimilation, intercellular CO 2 concentration, and stomatal conductance. A significant reduction in root and shoot Na content combined with higher amounts of K + in the presence of B provided more optimal tissue K + /Na + ratios. At the cellular level, B-treated roots showed stronger stimulation of H + -ATPase by NaCl (using net H + flux as a proxy) and could maintain more negative membrane potential. This enhanced cytosolic potassium retention (by reducing GORK-mediated K + loss) and provided a driving force for the operation of SOS1 Na + /H + exchanger explaining whole-plant ionomics data. Boron also affected the sensitivity of K + - and Ca 2+ - permeable ion channels to reactive oxygen species. Significantly less root K + loss but more root Ca 2+ uptake was observed for +B plants in response to H 2 O 2 , indicating the desensitization of K + - permeable channels and the cell type-specific Ca 2+ - permeable transporters to H 2 O 2 , and the role of B in their regulation. Overall, B availability resulted in better salt tolerance in barley, which could be a suitable alternative to crop breeding aimed at enhancing salt tolerance.

Abstract:Boron (B) is an essential microelement for plants, and its deficiency can lead to impaired development and function. Around 50% of arable land in the world is acidic, and low pH in the soil solution decreases availability of several essential mineral elements, including B, magnesium (Mg), calcium (Ca), and potassium (K). Plants take up soil B in the form of boric acid (H 3 BO 3 ) in acidic soil or tetrahydroxy borate [B(OH) 4 ] - at neutral or alkaline pH. Boron can participate directly or indirectly in plant metabolism, including in the synthesis of the cell wall and plasma membrane, in carbohydrate and protein metabolism, and in the formation of ribonucleic acid (RNA). In addition, B interacts with other nutrients such as Ca, nitrogen (N), phosphorus (P), K, and zinc (Zn). In this review, we discuss the mechanisms of B uptake, absorption, and accumulation and its interactions with other elements, and how it contributes to the adaptation of plants to different environmental conditions. We also discuss potential B-mediated networks at the physiological and molecular levels involved in plant growth and development.

Involvement of boron transporter BOR1 in growth under low boron and high nitrate conditions in Arabidopsis thaliana

Abstract:BOR1 is an efflux transporter of boron (B), responsible for loading B into the xylem. It has been reported that nitrate (NO3‐) concentrations significantly influence B concentrations in leaves and BOR1 mRNA accumulation in roots. Here, to unravel the interactive effects of B and NO3‐ on plant growth and the function of BOR1 under the combination of B and NO3‐, seedling growth was analyzed in Col‐0 and bor1 mutants. The growth of bor1 mutants was negatively affected by high NO3‐ but neither by potassium chloride (KCl) nor ammonium (NH4+) under low B conditions, suggesting the involvement of BOR1 in growth under high NO3‐. Mutants of bor2 and bor4 did not exhibit such growth responses, suggesting that this effect was specific to BOR1 among the BORs tested. Under low B conditions, loss of the BOR1 function led to a more significant decrease in B concentrations in the presence of high NO3‐ compared to normal NO3‐. Additionally, grafting experiments demonstrated that these effects of NO3‐ occurred when BOR1 is absent in roots. High NO3‐ treatment elevated BOR1 mRNA accumulation while the BOR1 protein accumulation was down regulated. These apparent opposite responses indicated that the transcriptional and (post‐)translational regulations follow different patterns. Our work provides evidence of a novel regulation of BOR1 and another B transport system by both B and NO3‐ in an interactive manner.

It is primarily involved in pectin crosslinking in the rhamnogalacturonan II region to ensure cell wall integrity. In plants grown under limited soil B availability, seed yield and quality are often compromised. However, excessive B accumulation can also be toxic to plants. Therefore, plants must maintain optimal tissue concentrations of B by regulating transport processes.

Abstract:Summary Boron (B) is crucial for plant growth and development. B deficiency can impair numerous physiological and metabolic processes, particularly in root development and pollen germination, seriously impeding crop growth and yield. However, the molecular mechanism underlying boron signal perception and signal transduction is rather limited. In this study, we discovered that CPK10, a calcium‐dependent protein kinase in the CPK family, has the strongest interaction with the boron transporter BOR1. Mutations in CPK10 led to growth and root development defects under B‐deficiency conditions, while constitutively active CPK10 enhanced plant tolerance to B deficiency. Furthermore, we found that CPK10 interacted with and phosphorylated BOR1 at the Ser689 residue. Through various biochemical analyses and complementation of B transport in yeast and plants, we revealed that Ser689 of BOR1 is important for its transport activity. In summary, these findings highlight the significance of the CPK10‐BOR1 signaling pathway in maintaining B homeostasis in plants and provide targets for the genetic improvement of crop tolerance to B‐deficiency stress.

Abstract:Toxic boron (B) concentrations cause impairments in several plant metabolic and physiological processes. Recently we reported that B toxicity led to a decrease in the transpiration rate of Arabidopsis plants in an ABA-dependent process within 24 h, which could indicate the occurrence of an adjustment of whole-plant water relations in response to this stress. Since plasma membrane intrinsic protein (PIP) aquaporins are key components influencing the water balance of plants because of their involvement in root water uptake and tissue hydraulic conductance, the aim of the present work was to study the effects of B toxicity on these important parameters affecting plant water status over a longer period of time. For this purpose, transpiration rate, water transport to the shoot and transcript levels of genes encoding four major PIP aquaporins were measured in Arabidopsis plants treated or not with a toxic B concentration. Our results indicate that, during the first 24 h of B toxicity, increased shoot ABA content would play a key role in reducing stomatal conductance, transpiration rate and, consequently, the water transport to the shoot. These physiological responses to B toxicity were maintained for up to 48 h of B toxicity despite shoot ABA content returning to control levels. In addition, B toxicity also caused the down-regulation of several genes encoding root and shoot aquaporins, which could reduce the cell to cell movement of water in plant tissues and, consequently, the water flux to shoot. All these changes in the water balance of plants under B toxicity could be a mechanism to prevent excess B accumulation in plant tissues.

Abstract:Boron (B) is essential for plant growth. However, the molecular mechanism of B transport in rapeseed ( Brassica napus L.) is unknown well. Here, we report that B transporter BnaA4.BOR2 is involved in the transport of B from root to shoot and its distribution in shoot cell wall and flower in rapeseed. The results of GUS staining and in-situ PCR analysis showed that BnaA4.BOR2 is mainly expressed in cortex and endodermis of root tip meristem zone and endodermis of mature zone. BnaA4.BOR2 was mainly localized in plasma membrane and showed B transport activity in yeast. Overexpression of Bna4.BOR2 could rescue the phenotype of Arabidopsis mutant bor2-2 under low-B condition. Furthermore, knockout of BnaA4.BOR2 could significantly enhance the sensitivity of rapeseed mutants to B deficiency, including inhibition of root elongation and biomass decrease of roots and shoots. The B concentration in xylem sap of BnaA4.BOR2 mutants was significantly decreased under B deficiency, which resulted in significantly lower B concentrations in shoot cell wall at seedling stage and flower organ at reproductive stage compared to that of wild-type QY10. The growth of BnaA4.BOR2 mutants were severely inhibited, exhibiting a typical B-deficient phenotype of "flowering without seed setting", leading to a sharp decrease in seed yield in B deficient soil. Taken together, these results indicate that BnaA4.BOR2 is critical for rapeseed growth and seed yield production under low B level, which is mainly expressed in cortex and endodermis, and contributed to the transport of B from roots to shoots and its distribution in shoot.
