Abstract:In this paper, an in-host HIV infection model with latent reservoir, delayed CTL immune response and immune impairment is investigated. By using suitable Lyapunov functions and LaSalle's invariance principle, it is shown that when time delay is equal to zero, the immunity-inactivated reproduction ratio is a threshold determining the global dynamics of the model. By means of the persistence theory for infinite dimensional systems, it is proven that if the immunity-inactivated reproduction ratio is greater than unity, the model is permanent. Choosing time delay as the bifurcation parameter and analyzing the corresponding characteristic equation of the linearized system, the existence of a Hopf bifurcation at the immunity-activated equilibrium is established. Numerical simulations are carried out to illustrate the theoretical results and reveal the effects of some key parameters on viral dynamics.
Mathematical analysis of an age-structured HIV-1 infection model with CTL immune response

Abstract:In this paper, an age-structured HIV-1 infection model with CTL immune response is investigated. In the model, we consider the infection age (i.e. the time that has elapsed since an HIV virion has penetrated the cell) of infected $T$ cells. The asymptotic smoothness of the semi-flow generated by the system is established. By calculation, the immune-inactivated reproduction rate $\mathscr{R}_0$ and the immune-activated reproduction rate $\mathscr{R}_1$ are obtained. By analyzing the corresponding characteristic equations, the local stability of an infection-free steady state and a CTL-inactivated infection steady state of the model is established. By using the persistence theory for infinite dimensional system, the uniform persistence of the system is established when $\mathscr{R}_1>1$. By means of suitable Lyapunov functionals and LaSalle's invariance principle, it is shown that if $\mathscr{R}_0<1$, the infection-free steady state is globally asymptotically stable; if $\mathscr{R}_1<1< \mathscr{R}_0$, sufficient conditions are derived for the global stability of the CTL-inactivated infection steady state; if $\mathscr{R}_1>1$, sufficient conditions are obtained for the global attractivity of the CTL-activated infection steady state. Numerical simulations are carried out to illustrate the feasibility of the theoretical results.
Regional differences in the frequency of slow waves in smooth muscle of the guinea-pig stomach.

Abstract:The frequency of slow waves recorded from circular muscle bundles with attached longitudinal muscle (intact muscle) was compared with that of slow potentials recorded from isolated circular muscle bundles (isolated muscle) from the guinea-pig stomach. In intact muscle preparations, slow waves were generated in the corpus, antrum and pylorus with a higher frequency in the corpus (about 5 min(-1)) than the other regions (about 2 min(-1) in antrum, about 1.5 min(-1) in pylorus). The resting potential amplitude was graded across the stomach, at about -50 mV in the fundus, -60 mV in the corpus, -65 mV in the antrum and -70 mV in the pylorus. A similar distribution of resting membrane potential and slow potential frequency was also observed in isolated muscle bundles from the different regions. Caffeine (1 mM) abolished slow waves in some corpus preparations and inhibited the 2nd component of slow waves in the antrum and pylorus, and also abolished slow potentials in isolated muscle preparations from any region of the stomach. This suggests that myenteric interstitial cells of Cajal (ICC-MY) are heterogeneously distributed in the stomach (pylorus, antrum and part of the corpus regions), with a homogeneous distribution of muscular interstitial cells of Cajal (ICC-IM) within the circular muscle bundles. The frequency of slow potentials in smooth muscle isolated from any region of the stomach changed linearly in response to membrane potential changes produced by either current injection or high potassium solutions. The frequency of slow potentials after setting the membrane potential at -60 mV was larger in the corpus than the antrum, suggesting that the high frequency discharge of corpus muscle is produced by the low membrane potential and additional unidentified factors. We suggest that the regional difference in slow wave discharge is produced mainly by ICC-IM, and the role of ICC-MY may be little, if any.
Mathematical modeling of escape of HIV from cytotoxic T lymphocyte responses

Abstract:Human immunodeficiency virus (HIV-1 or simply HIV) induces a persistent infection, which in the absence of treatment leads to AIDS and death in almost all infected individuals. HIV infection elicits a vigorous immune response starting about 2-3 weeks post infection that can lower the amount of virus in the body, but which cannot eradicate the virus. How HIV establishes a chronic infection in the face of a strong immune response remains poorly understood. It has been shown that HIV is able to rapidly change its proteins via mutation to evade recognition by virus-specific cytotoxic T lymphocytes (CTLs). Typically, an HIV-infected patient will generate 4-12 CTL responses specific for parts of viral proteins called epitopes. Such CTL responses lead to strong selective pressure to change the viral sequences encoding these epitopes so as to avoid CTL recognition. Here we review experimental data on HIV evolution in response to CTL pressure, mathematical models developed to explain this evolution, and highlight problems associated with the data and previous modeling efforts. We show that estimates of the strength of the epitope-specific CTL response depend on the method used to fit models to experimental data and on the assumptions made regarding how mutants are generated during infection. We illustrate that allowing CTL responses to decay over time may improve the fit to experimental data and provides higher estimates of the killing efficacy of HIV-specific CTLs. We also propose a novel method for simultaneously estimating the killing efficacy of multiple CTL populations specific for different epitopes of HIV using stochastic simulations. Lastly, we show that current estimates of the efficacy at which HIV-specific CTLs clear virus-infected cells can be improved by more frequent sampling of viral sequences and by combining data on sequence evolution with experimentally measured CTL dynamics.

The Effects of Latent Infection on the Dynamics of HIV

Abstract:One way in which the human immunodeficiency virus (HIV-1) replicates within a host is by infecting activated CD4+ T-cells, which then produce additional copies of the virus. Even with the introduction of antiretroviral drug therapy, which has been very successful over the past decade, a large obstacle to the complete eradication of the virus is the presence of viral reservoirs in the form of latently infected CD4+ T-cells. We consider a model of HIV infection that describes T-cell and viral interactions, as well as, the production and activation of latently infected T-cells. Upon determining equilibrium states of the latent cell model, the local and global asymptotic behavior of solutions is examined, and the basic reproduction number of the system is computed to be strictly less than that of the corresponding three-component model, which omits the effects of latent infection. In particular, this implies that a wider variety of parameter values will lead to viral eradication as $t \to \infty$ due to the appearance of latent CD4+ T-cells. With this realization we discuss possible alternative notions for eradication and persistence of infection other than traditional tools. These results are further illustrated by a number of numerical simulations.

Stability analysis for a HIV model with cell-to-cell transmission, two immune responses and induced apoptosis

Abstract:In this paper, a dynamic HIV model with cell-to-cell transmission, two immune responses, and induced apoptosis is proposed and studied. First, the non-negativity and boundedness of the solutions of the model are given, and then the exact expression of the basic reproduction number $ R_{0} $ is obtained by using the next generation matrix method. Second, criteria are obtained for the local stability of the disease-free equilibrium, immune response-free equilibrium, and the infected equilibrium with both humoral and cellular immune responses. Furthermore, the threshold conditions are also derived for the global asymptotic stability of the disease-free equilibrium, immune response-free equilibrium, and the infected equilibrium with both humoral and cellular immune responses by constructing the suitable Lyapunov function. Finally, some numerical simulations are conducted to verify the theoretical results; the numerical simulation results show that the increase of apoptosis rate had a positive role in the control of viral infection.

Abstract:The human immunodeficiency virus (HIV) interacts with the immune cells within the human body, where the environment is uncertain and noisy. Stochastic models can successfully encapsulate the effect of such a noisy environment compared to their deterministic counterparts. The human immune system is complex but well-coordinated with various immune cells like C D 4T cells, dendritic cells, and cytotoxic T-lymphocyte (CTL) cells, among many others. The CTL can kill the antigenic cells after its recognition. However, the efficacy of CTL in removing the infected C D 4T cells is progressively compromised in HIV-infected individuals. This paper considers a noise-induced HIV-immune cell interaction model with immune impairment. A multiplicative white noise is introduced in the infection rate parameter to represent the fluctuations around the average value of the rate parameter as a causative effect of the noise. We analyzed the deterministic and stochastic models and prescribed sufficient conditions for infection eradication and persistence. It is determined under what parametric restrictions the asymptotic solutions of the noise-induced system will be a limiting case of the deterministic solutions. Simulation results revealed that the solutions of the deterministic system either converge to a CTL-dominated interior equilibrium or a CTL-free immunodeficient equilibrium, depending on the initial values of the system. Stochastic analysis divulged that higher noise might be helpful in the infection removal process. The extinction time of infected C D 4T cells for some fixed immune impairment gradually decreases with increasing noise intensity and follows the power law.
Mutational analysis of the necdin gene in patients with congenital isolated hypogonadotropic hypogonadism.

Abstract:CONTEXT Necdin activates GNRH gene expression and is fundamental for the development, migration, and axonal extension of murine GNRH neurons. In humans, necdin plays a potential role in the hypogonadotropic hypogonadism phenotype in patients with Prader-Willi syndrome. AIM To investigate necdin gene (NDN) variants in patients with isolated hypogonadotropic hypogonadism (IHH). PATIENTS AND METHODS We studied 160 Brazilian patients with IHH, which includes 92 with Kallmann syndrome and 68 with normosmic IHH. Genomic DNA was extracted and the single NDN exon was amplified and sequenced. To measure GNRH transcriptional activity, luciferase reporter plasmids containing GNRH regulatory regions were transiently transfected into GT1-7 cells in the presence and absence of overexpressed wild-type or mutant necdin. RESULTS A heterozygous variant of necdin, p.V318A, was identified in a 23-year-old male with Kallmann syndrome. The p.V318A was also present in affected aunt and his father and was absent in 100 Brazilian control subjects. Previous FGFR1 gene analysis revealed a missense mutation (p.P366L) in this family. Functional studies revealed a minor difference in the activation of GNRH transcription by mutant protein compared with wild type in that a significant impairment of the necdin protein activity threshold was observed. CONCLUSION A rare variant of necdin (p.V318A) was described in a family with Kallmann syndrome associated with a FGFR1 mutation. Familial segregation and in vitro analysis suggested that this non-synonymous variant did not have a direct causative role in the hypogonadism phenotype. NDN mutations are not a frequent cause of congenital IHH.
A mathematical model for HIV dynamics with multiple infections: implications for immune escape

Abstract:Multiple infections enable the recombination of different strains, which may contribute to viral diversity. How multiple infections affect the competition dynamics between the two types of strains, the wild and the immune escape mutant, remains poorly understood. This study develops a novel mathematical model that includes the two strains, two modes of viral infection, and multiple infections. For the representative double-infection case, the reproductive numbers are derived and global stabilities of equilibria are obtained via the Lyapunov direct method and theory of limiting systems. Numerical simulations indicate similar viral dynamics regardless of multiplicities of infections though the competition between the two strains would be the fiercest in the case of quadruple infections. Through sensitivity analysis, we evaluate the effect of parameters on the set-point viral loads in the presence and absence of multiple infections. The model with multiple infections predict that there exists a threshold for cytotoxic T lymphocytes (CTLs) to minimize the overall viral load. Weak or strong CTLs immune response can result in high overall viral load. If the strength of CTLs maintains at an intermediate level, the fitness cost of the mutant is likely to have a significant impact on the evolutionary dynamics of mutant viruses. We further investigate how multiple infections alter the viral dynamics during the combination antiretroviral therapy (cART). The results show that viral loads may be underestimated during cART if multiple-infection is not taken into account.

Abstract:A synthetic tetrasaccharide (TS4), structurally related to blood groups, inhibited the proliferation of C6 glioma cells in culture and the growth of tumors formed after intracerebral transplantation of C6 cells. TS4-treated tumors were substantially smaller than controls, as expected from TS4 cytostatic action on C6 glioma cells in culture. However, in vivo treatment also caused extensive tumor destruction. This effect appeared to be caused indirectly, either by activation of natural killer cells, cytotoxic lymphocytes, or by inhibition of tumor vascularization. Enhanced antigenicity of TS4-treated glioma may be related to the increased expression of connexin 43 observed in glioma cell cultures treated with the oligosaccharide. Because concentrations of up to 20 mg/ml of TS4 were not toxic for normal neuronal or glial cells, specific oligosaccharides such as TS4 offer the possibility of selective tumor treatment.
Multistability in a Model for CTL Response to HTLV-I Infection and Its Implications to HAM/TSP Development and Prevention

Abstract:Human T-cell leukaemia/lymphoma virus type I (HTLV-I) is a retrovirus that has been identified as the causative agent of HTLV-I-associated myelopathy/tropical spastic paraparesis (HAM/TSP) and other illnesses. HTLV-I infects primarily CD4+ T cells and the transmission occurs through direct cell-to-cell contact. HAM/TSP patients harbor higher proviral loads in peripheral blood lymphocytes than asymptomatic carriers. Also, HAM/TSP patients exhibit a remarkably high number of circulating HTLV-I-specific CD8+ cytotoxic T lymphocytes (CTLs) in the peripheral blood. While CTLs have a protective role by killing the infected cells and lowering the proviral load, a high level of CTLs and their cytotoxicity are believed to be a main cause of the development of HAM/TSP. A mathematical model for HTLV-I infection of CD4+ T cells that incorporates the CD8+ cytotoxic T-cell (CTL) response is investigated. Our mathematical analysis reveals that the system can stabilize at a carrier steady-state with persistent viral infection but no CTL response, or at a HAM/TSP steady-state at which both the viral infection and CTL response are persistent. We also establish two threshold parameters R0 and R1, the basic reproduction numbers for viral persistence and for CTL response, respectively. We show that the parameter R1 can be used to distinguish asymptomatic carriers from HAM/TSP patients, and as an important control parameter for preventing the development of HAM/TSP.

Abstract:In this paper, we present a stochastic HIV model, including logistic growth, CTL immune response and logarithmic mean–reverting Ornstein–Uhlenbeck process. Firstly, we certify that there exists a unique global positive solution by rigorous mathematical analysis. Then, by constructing a suitable Lyapunov function and using the strong law of large numbers and Fatou's lemma, we obtain the existence of the stationary distribution when stochastic reproduction number is greater than one. Moreover, we derive an approximate expression for the probability density function of the stochastic model around the infected equilibrium of its corresponding deterministic model. In addition, a critical condition for virus extinction is established. Finally, through numerical simulations, we investigate the effects of noise intensity and the logistic growth on model behavior, and we also explore the influence of key parameters on virus persistence and extinction.

Abstract:In this paper, we study the global well-posedness and global dynamics of a reaction–diffusion HIV infection model with the chemotactic movement of CTLs (Cytotoxic T lymphocytes). We first show the global existence and uniform boundedness for solutions of the system with general functional incidences. Then, for the model with bilinear incidence rate, we discuss the existence conditions of the three equilibria (infection-free, chemokines-extinct, chemokines-acute equilibria) of the model and obtain the conclusion of the local asymptotic stability of these equilibria by analyzing the linearized system at these equilibria. Moreover, by constructing reasonable Lyapunov functionals, we investigate the global stability and attractivity of the equilibria. Applying the Lp−Lq estimate, Young's inequality, Gagiardo-Nirenberg inequality and the parabolic regularity theorem, we also discuss the convergence rates of the equilibria. Finally, some numerical simulations are conducted to verify the theoretical results.

Abstract:This paper explores the dynamics analysis of a human immunodeficiency virus (HIV) model with saturated cytotoxic T lymphocyte (CTL) immune response and Beddington–DeAngelis infection rate. There are two time delays in the model to describe the time needed for infection of cell and CTL immune response generation, respectively. We obtain two thresholds and three possible equilibria from the model. By analyzing the corresponding characteristic equations, we study the stabilities of equilibrium and the effect of delays on CTL immune response. The results indicate that when immune delay is present, the steady state of equilibrium is disrupted and leads to a Hopf bifurcation. Finally, we use sensitivity analyses to show the effect of parameters on thresholds and numerical simulations to illustrate the theoretical results.

Abstract:As the prevalence of viral infection in body, human T‐cell leukemia virus type 1 (HTLV‐1) is receiving increasing attention. Research on the corresponding virus models is of great significance to tackle the challenges of understanding HTLV‐1 development and treatment. This paper focuses on the dynamic analysis for a stochastic model with nonlinear cytotoxic T lymphocyte (CTL) response, which is driven by Ornstein–Uhlenbeck (OU) process to model the progression of HTLV‐1 in vivo. Rich dynamic behaviors such as the extinction of infected CD4+ T cells (ITCs), stationary distribution (SD), probability density, and finite‐time stability (FTS) of the model are established to reveal the interaction of cell populations. The optimal therapeutic strategy based on the cost‐benefit viewpoint is further obtained. Finally, illustrative numerical simulations are represented to corroborate the effectiveness of treatment and the ambient perturbation's impact that strengthening the noise strength can lead to rapid virus clearance.

Abstract:We study a viral infection model incorporating both cell-to-cell infection and immune chemokines. Based on experimental results in the literature, we make a standing assumption that the cytotoxic T lymphocytes (CTL) will move toward the location with more infected cells, while the diffusion rate of CTL is a decreasing function of the density of infected cells. We first establish the global existence and ultimate boundedness of the solution via a priori energy estimates. We then define the basic reproduction number of viral infection and prove (by the uniform persistence theory, Lyapunov function technique and LaSalle invariance principle) that the infection-free steady state is globally asymptotically stable if . When , then becomes unstable, and another basic reproduction number of CTL response becomes the dynamic threshold in the sense that if , then the CTL-inactivated steady state is globally asymptotically stable; and if , then the immune response is uniform persistent and, under an additional technical condition the CTL-activated steady state is globally asymptotically stable. To establish the global stability results, we need to prove point dissipativity, obtain uniform persistence, construct suitable Lyapunov functions, and apply the LaSalle invariance principle.

Abstract:In this paper, an in-host HBV model with two infection routes including cell-virus infection and cell-cell infection, intracellular delay , virus replication delay and CTL immune response delay are investigated. Firstly, the positivity and boundedness of all solutions for the model with nonnegative initial values have been established. Next, two key parameters with significant biological importance, the virus reproductive number and the CTL immune reproductive number , are derived. Subsequently, the stability of equilibria is analyzed by constructing appropriate Lyapunov functionals and applying LaSalle's invariance principle. The results indicate that intracellular delay and virus replication delay do not affect the stability of the three equilibria. However, a positive CTL immune response delay can lead to stability switches at the endemic equilibrium. By treating the CTL immune response delay as a bifurcation parameter, certain conditions for these stability switches are determined. It is concluded that as the CTL immune response delay increases, the endemic equilibrium becomes unstable, resulting in a Hopf bifurcation in the system. Finally, numerical simulations further validate the theoretical findings.

Abstract:To discuss the influence of cytokine-enhanced rate and antibody delay on viral infection, in this paper, we propose a cytokine-enhanced HIV model with saturation incidence, antibody immune response and time delays. Multiple time delays, namely intracellular delay, virus replication delay and antibody delay, are included. First, we prove that the model is well-posed. Then, we obtain two basic reproductive numbers of the model. Next, the stability of the equilibria is investigated by using Lyapunov functions and LaSalle’s invariance principle. The results show that intracellular delay and virus replication delay do not impact on the stability of the three equilibria. However, if antibody immune response delay is positive, we determine some conditions for stability switches of the endemic equilibrium by using antibody immune response delay as a bifurcation parameter. It is concluded that with the increase of antibody immune response delay, the endemic equilibrium loses its stability and the system generates Hopf bifurcation. Finally, the numerical simulation results also show the correctness of the theoretical results.

Abstract:This work investigated a piecewise immunosuppressive infection model that assessed the effectiveness of implementing this therapeutic regimen once the effector cell count falls below a specific threshold level by introducing a threshold strategy. The sliding mode dynamics, global dynamics, and boundary equilibrium bifurcations of the Filippov system were examined based on the global dynamics of the two subsystems. Our primary findings indicate that the HIV viral loads and effector cell counts can be stabilized within the required predetermined level. This outcome depends on the threshold level, immune intensity, and the initial values of the system. Therefore, properly combining these key factors makes it possible to effectively curb the abnormal increase of virus and keep the effector cells at a reliable level. This approach maximizes the controllable range of the HIV. The proposed switching system incorporating pseudo-equilibrium exhibits three types of equilibriums that could be bistable or tristable. It means there is a possibility of controlling the virus after administering therapy if the immune intensity $ c $ is limited within the range of the post-treatment control threshold and the elite control threshold when $ {R_0} > {R_{{c_1}}} > {R_{{c_2}}} > 1 $.

Abstract:A dynamic model of non-lineal time-dependent ordinary differential equations (ODE) has been applied to the interactions of a HIV infection with the immune system cells. This model has been simplified into two compartments: lymph node and peripheral blood. The model includes CD4 T-lymphocytes in several states (quiescent Q, naive N and activated T), cytotoxic CD8 T-cells, B-cells and dendritic cells. Cytokines and immunoglobulins specific for each antigen (i.e. gp41 or p24) have been also included in the model, modelling the atraction effect of CD4 T-cells to the infected area and the reduction of virus concentration by immunoglobulins. HIV virus infection of CD4 T-lymphocytes is modelled in several stages: before fusion as HIV-attached (H) and after fusion as non-permissive / abortively infected (M), and permissive / latently infected (L) and permissive / actively infected (I). These equations have been implemented in a C++/Python interface application, called Immune System app, which runs Open Modelica software to solve the ODE system through a 4th order Runge-Kutta numerical approximation. Results of the simulation show that although HIV virus concentration in both compartments is lower than $10^{-10}$ virus/$\mu L$ after t=2 years, quiescent lymphocytes reach an equilibrium with a concentration lower than the initial conditions, due to the latency state, which serves as a reservoir in time of virus production. As a conclusion, this model can provide reliable results in other conditions, such as antiviral therapies.
