Abstract:Despite the effects of ocean acidification (OA) on seagrasses have been widely investigated, predictions of seagrass performance under future climates need to consider multiple environmental factors. Here, we performed a mesocosm study to assess the effects of OA on shallow and deep Posidonia oceanica plants. The experiment was run in 2021 and repeated in 2022, a year characterized by a prolonged warm water event, to test how the effects of OA on plants are modulated by thermal stress. The response of P. oceanica to experimental conditions was investigated at different levels of biological organization. Under average seawater temperature, there were no effects of OA in both shallow and deep plants, indicating that P. oceanica is not limited by current inorganic carbon concentration, regardless of light availability. In contrast, under thermal stress, exposure of plants to OA increased lipid peroxidation and decreased photosynthetic performance, with deep plants displaying higher levels of heat stress, as indicated by the over-expression of stress-related genes and the activation of antioxidant systems. In addition, warming reduced plant growth, regardless of seawater CO2 and light levels, suggesting that thermal stress may play a fundamental role in the future development of seagrass meadows. Our results suggest that OA may exacerbate the negative effects of future warming on seagrasses.

In spite of previous publications of replantations based on a single artery and vein, it is now accepted that multiple anastomoses increase the success rate. We present herein the case of a successfully replanted avulsion in a child who caught her hair in a go-kart motor belt, and we discuss the mechanism of injury and number of anastomoses.

Abstract:Seagrass meadows play vital ecological roles in the marine ecosystem. Global climate change poses considerable threats to seagrass survival. However, it is unclear how seagrass and its associated bacteria will respond under future complex climate change scenarios. This study explored the effects of ocean warming (+2 °C) and ocean acidification (-0.4 units) on seagrass physiological indexes and bacterial communities (sediment and rhizosphere bacteria) of the seagrass Thalassia hemprichii during an experimental exposure of 30 days. Results demonstrated that the synergistic effect of ocean warming and ocean acidification differed from that of one single factor on seagrass and the associated bacterial community. The seagrass showed a weak resistance to ocean warming and ocean acidification, which manifested through the increase in the activity of typical oxidoreductase enzymes. Moreover, the synergistic effect of ocean warming and ocean acidification caused a significant decrease in seagrass's chlorophyll content. Although the bacterial community diversity exhibited higher resistance to ocean warming and ocean acidification, further bacterial functional analysis revealed the synergistic effect of ocean warming and ocean acidification led to significant increases in SOX-related genes abundance which potentially supported the seagrass in resisting climate stress by producing sulfates and oxidizing hydrogen sulfide. More stable bacterial communities were detected in the seagrass rhizosphere under combined ocean warming and ocean acidification. While for one single environmental stress, simpler networks were detected in the rhizosphere. In addition, the observed significant correlations between several modules of the bacterial community and the physiological indexes of the seagrass indicate the possible intimate interaction between seagrass and bacteria under ocean warming and ocean acidification. This study extends our understanding regarding the role of seagrass associated bacterial communities and sheds light on both the prediction and preservation of the seagrass meadow ecosystems in response to global climate change.

Abstract:Since the Industrial Revolution, increasing atmospheric CO2 concentrations have had a substantial negative impact influence on coastal ecosystems because of direct effects including ocean acidification and indirect effects such as extreme rainfall events. Using a two-factor crossover indoor simulation experiment, this study examined the combined effects of acidification and hyposaline stress on Thalassia hemprichii. Seawater acidification increased the photosynthetic pigment content of T. hemprichii leaves and promoted seagrass growth rate. Hyposaline stress slowed down seagrass growth and had an impact on the osmotic potential and osmoregulatory substance content of seagrass leaves. Acidification and salinity reduction had significant interaction effects on the photosynthesis rate, photosynthetic pigment content, chlorophyll fluorescence parameters, and osmotic potential of T. hemprichii, but not on the growth rate. Overall, these findings have shown that the hyposaline stress inhibitory effect on the T. hemprichii physiological performance and growth may be reduced by acidification.

Abstract:Solutions are being sought to ameliorate the impacts of anthropogenic climate change. Seagrass may be a solution to provide refugia from climate change for marine organisms. This study aimed to determine if the seagrass Zostera muelleri sub spp. capricorni benefits the Sydney rock oyster Saccostrea glomerata, and if these benefits can modify any anticipated negative impacts of ocean acidification. Future and ambient ocean acidification conditions were simulated in 52 L mesocosms at control (381 μatm) and elevated (848 μatm) CO2 with and without Z. muelleri. Oyster growth, physiology and microbiomes of oysters and seagrass were measured. Seagrass was beneficial to oyster growth at ambient pCO2, but did not positively modify the impacts of ocean acidification on oysters at elevated pCO2. Oyster microbiomes were altered by the presence of seagrass but not by elevated pCO2. Our results indicate seagrasses may not be a panacea for the impacts of climate change.

Abstract:To investigate the ecological impacts of macroalgae in the framework of shifting global CO2 concentrations, we conducted a study utilizing Ulva fasciata and Sargassum horneri specimens sourced from the Ma'an Archipelago in Zhejiang Province on how ocean acidification (OA) and temperature changes interact to affect the photosynthetic physiological responses of macroalgae. The results of the study showed that OA reduced the tolerance of U. fasciata to bright light at 20 °C, resulting in more pronounced photoinhibition, while 15 °C caused significant inhibition of U. fasciata, reducing its growth and photosynthetic activity, but OA alleviated the inhibition and promoted the growth of the alga to a certain extent. The tolerance of S. horneri to bright light was also reduced at 20 °C; the inhibition was relieved at 15 °C, and the OA further improved the algal growth. The Relative Growth Rate (RGR), photosynthetic pigment content, and the release of the dissolved organic carbon (DOC) of U. fasciata were mainly affected by the change in temperature; the growth of the alga and the synthesis of metabolites were more favored by 20 °C. A similar temperature dependence was observed for S. horneri, with faster growth and high metabolism at 15 °C. Our results suggest that OA reduces the tolerance of macroalgae to high light at suitable growth temperatures; however, at unsuitable growth temperatures, OA effectively mitigates this inhibitory effect and promotes algal growth.

Abstract:Quantifying the strength of non-trophic interactions exerted by foundation species is critical to understanding how natural communities respond to environmental stress. In the case of ocean acidification (OA), submerged marine macrophytes, such as seagrasses, may create local areas of elevated pH due to their capacity to sequester dissolved inorganic carbon through photosynthesis. However, although seagrasses may increase seawater pH during the day, they can also decrease pH at night due to respiration. Therefore, it remains unclear how consequences of such diel fluctuations may unfold for organisms vulnerable to OA. We established mesocosms containing different levels of seagrass biomass (Zostera marina) to create a gradient of carbonate chemistry conditions and explored consequences for growth of juvenile and adult oysters (Crassostrea gigas), a non-native species widely used in aquaculture that can co-occur, and is often grown, in proximity to seagrass beds. In particular, we investigated whether increased diel fluctuations in pH due to seagrass metabolism affected oyster growth. Seagrasses increased daytime pH up to 0.4 units but had little effect on nighttime pH (reductions less than 0.02 units). Thus, both the average pH and the amplitude of diel pH fluctuations increased with greater seagrass biomass. The highest seagrass biomass increased oyster shell growth rate (mm day-1) up to 40%. Oyster somatic tissue weight and oyster condition index exhibited a different pattern, peaking at intermediate levels of seagrass biomass. This work demonstrates the ability of seagrasses to facilitate oyster calcification and illustrates how non-trophic metabolic interactions can modulate effects of environmental change.

Abstract:Objective: To investigate the responses of Zostera marina seedlings to the individual and combined stresses of seasonal temperature increase and ocean acidification (OA) caused by global climate change and anthropogenic factors. This data will help in efforts to protect and restore seagrass beds in temperate coastal zones of China. Methods: A mesoscale experimental system was utilized to analyze stress response mechanisms at multiple levels - phenotype, transcriptome, and metabolome - during the seedling stage of Z. marina , a dominant temperate seagrass species in China. The study monitored the seedlings under varying conditions: increased seasonal temperature, OA, and a combination of both. Results: Findings revealed that under high-temperature conditions, carotenoid biosynthesis was stimulated through the upregulation of specific metabolites and enzymes. Similarly, the biosynthesis of certain alkaloids was promoted alongside modifications in starch, sucrose, and nitrogen metabolism, which improved the plant's adaptation to OA. Unique metabolic pathways were activated under OA, including the degradation of certain amino acids and modifications in the citric acid cycle and pyruvate metabolism. When subjected to both temperature and OA stresses, seedlings actively mobilized various biosynthetic pathways to enhance adaptability and resilience, with distinct metabolic pathways enhancing the plant's response under diversified stress conditions. In terms of growth, all treatment groups exhibited significant leaf length increase ( p < 0.05), but the weakest growth index was observed under combined stress, followed by the thermal treatment group. Conversely, growth under OA treatment was better, showing a significant increase in wet weight, leaf length, and leaf width ( p < 0.05). Conclusion: Seasonal temperature increase was found to inhibit the growth of Z. marina seedlings to some extent, while OA facilitated their growth. However, the positive effects of OA did not mitigate the damage caused by increased seasonal temperature under combined stress due to seedlings' sensitivity at this stage. Our findings elucidate differing plant coping strategies under varied stress conditions, contingent on the initial environment. This research anticipates providing significant data support for the adaptation of Z. marina seedlings to seasonal temperature fluctuations and global oceanic events like OA, propelling the effective conservation of seagrass beds.

Ocean Acidification Alleviates Dwarf Eelgrass (Zostera noltii) Lipid Landscape Remodeling under Warming Stress

Abstract:Coastal seagrass meadows provide a variety of essential ecological and economic services, including nursery grounds, sediment stabilization, nutrient cycling, coastal protection, and blue carbon sequestration. However, these ecosystems are highly threatened by ongoing climatic change. This study was aimed to understand how the dwarf eelgrass Zostera noltii leaf lipid landscapes are altered under predicted ocean warming (+4 °C) and hypercapnic (ΔpH 0.4) conditions. Warming and hypercapnic conditions were found to induce a severe reduction in the leaf total fatty acid, though the combined treatment substantially alleviated this depletion. The lipid discrimination revealed a significant increase in the relative monogalactosyldiacylglycerol (MGDG) content in both hypercapnic and warming conditions, allied to plastidial membrane stabilization mechanisms. Hypercapnia also promoted enhanced phosphatidylglycerol (PG) leaf contents, a mechanism often associated with thylakoid reinvigoration. In addition to changing the proportion of storage, galacto- and phospholipids, the tested treatments also impacted the FA composition of all lipid classes, with warming exposure leading to decreases in polyunsaturated fatty acids (PUFAs); however, the combination of both stress conditions alleviated this effect. The observed galactolipid and phospholipid PUFA decreases are compatible with a homeoviscous adaptation, allowing for the maintenance of membrane stability by counteracting excessive membrane fluidity. Neutral lipid contents were substantially increased under warming conditions, especially in C18 fatty acids (C18), impairing their use as substrates for fatty acylated derivatives essential for maintaining the osmotic balance of cells. An analysis of the phospholipid and galactolipid fatty acid profiles as a whole revealed a higher degree of discrimination, highlighting the higher impact of warming and the proposed stress alleviation effect induced by increased water-dissolved CO2 availability. Still, it is essential to remember that the pace at which the ocean is warming can overcome the ameliorative capacity induced by higher CO2 availability, leaving seagrasses under severe heat stress beyond their lipid remodeling capacity.
Effects of temperature and hypoxia on respiration, photorespiration, and photosynthesis of seagrass leaves from contrasting temperature regimes

Abstract:Abstract In near-future climate change scenarios, elevated ocean temperatures with higher and more frequent peaks are anticipated than at present. Moreover, increased eutrophication and higher primary and secondary productivity will affect the oxygen levels of shallow-water coastal ecosystems, creating hypoxic conditions that can be experienced regularly, especially in dense vegetated systems. These climate-related events may impose detrimental effects on the primary productivity of seagrass. To evaluate such effects, this study combined gas exchange measurements with pulse amplitude-modulated fluorometry to assess the impact of short-time exposure to a range of water temperatures at ambient and low-oxygen levels on mitochondrial respiration, chlorophyll fluorescence (based on the Fv/F0 ratio), photosynthetic oxygen evolution, and photorespiration in leaf segments of the temperate seagrass Zostera marina and the tropical seagrass Thalassia hemprichii. We found that mitochondrial respiration in both Z. marina and T. hemprichii increased with higher temperatures up to 40°C and that low O2 caused significantly reduced respiration rates, particularly in T. hemprichii. Elevated water temperature had a clear negative effect on the Fv/F0 of both seagrass species, indicating damage or inactivation of the photosynthetic apparatus, even when light is not present. Moreover, damage to the photosynthetic apparatus was observed as an effect of elevated temperature combined with low O2 during darkness, resulting in subsequent lower photosynthesis in light. Photorespiration was present, but not promoted by increased temperature alone and will thus not further contribute to productivity losses during warmer events (when not carbon limited). This study demonstrates the negative impact of hypoxic stress and elevated temperatures on seagrass productivity, which may influence the overall health of seagrass plants as well as oxygen and carbon fluxes of shallow-water coastal ecosystems in warmer climate scenarios.

Abstract:Ocean warming (OW) and ocean acidification (OA), driven by rapid global warming accelerating at unprecedented rates, are profoundly impacting the stability of seagrass ecosystems. Yet, our current understanding of the effects of OW and OA on seagrass remains constrained. Herein, we investigated the response of eelgrass ( Zostera marina L.), a representative seagrass species, to OW and OA through comprehensive transcriptomic and metabolomic analyses. The results showed notable variations in plant performance under varying conditions: OW, OA, and OWA (a combination of both conditions). Specifically, under average oceanic temperature conditions for eelgrass growth over the past 20 years —from May to November—OA promoted the production of differentially expressed genes and metabolites associated with alanine, aspartate, and glutamate metabolism, as well as starch and sucrose metabolism. Under warming condition, eelgrass was resistant to OA by accelerating galactose metabolism, along with glycine, serine, and threonine metabolism, as well as the tricarboxylic acid (TCA) cycle. Under the combined OW and OA condition, eelgrass stimulated fructose and mannose metabolism, glycolysis, and carbon fixation, in addition to galactose metabolism and the TCA cycle to face the interplay. Our findings suggest that eelgrass exhibits adaptive capacity by inducing different metabolites and associated genes, primarily connected with carbon and nitrogen metabolism, in response to varying degrees of OW and OA. The data generated here support the exploration of mechanisms underlying seagrass responses to environmental fluctuations, which hold critical significance for the future conservation and management of these ecosystems.

Abstract:Extreme ocean events caused by global warming, such as marine heatwaves (MHWs) and ocean acidification (OA), are projected to intensify. A combination of extreme events may have severe consequences for marine ecosystems. Zostera marina was selected to understand how seagrass adapts to OA in extremely hot conditions. By combining morphology, transcriptomics, and metabolomics under mesoscale experimental conditions, we systematically investigated the response characteristics of Z. marina. Extremely high temperatures had a pronounced effect on growth, and the combined effect of OA mitigated the inhibitory effect of MHW. Both transcriptomic and metabolomic results showed that Z. marina resisted OA and MHW by upregulating the TCA cycle, glycolysis, amino acid metabolism, and relevant genes, as well as by activating the antioxidant system. The results of this study serve to improve our understanding of dual effects of factors of climate change on seagrass and may be used to direct future management and conservation efforts.

Abstract:Global coral reef degradation has precipitated phase shifts toward macroalgal-dominated communities. Despite the negative repercussions for reefscapes, higher abundances of primary producers have the potential to positively impact the physicochemical environment and mitigate negative impacts of ocean acidification (OA). In this study, we investigated the influence of macroalgal (cf. Sargassum vulgare ) density on coral ( Acropora millepora and A. hemprichii ) calcification rates under current and future OA conditions. Corals were resistant to OA up to ~ 1100 μatm, with no changes in calcification rates. However, the presence of (low and high density) algae reduced calcification rates by ~ 41.8%, suggesting either a chemical defense response due to algal metabolites or potential physical impacts from shading or abrasion. Documented beneficial buffering effects of macroalgae in OA may also elicit negative impacts on coral calcification, suggesting further work is needed to elucidate how species interactions influence responses to projected climate change.

Abstract:Coastal water temperature anomalies are expected to intensify during the current period of global climate change. Sudden water temperature elevation (e.g., heat waves) or decrease (e.g., summer upwelling) can have a far-reaching influence on seagrasses in temperate regions. We examined the influence of anomalously elevated and decreased water temperatures on the growth of Zostera marina plants held under two different levels of irradiance. Core samples of wild Z. marina shoots were cultured under either suddenly elevated (from the in situ level of 20 to 27 °C during the fall experiment) or decreased (from the in situ level of 27 to 20 °C during the summer experiment) temperature. We included light as an experimental factor because the light requirements of seagrasses increase when plants are under thermal stress. The growth of Z. marina shoots under anomalously elevated water temperature conditions, which simulated marine heat waves, was significantly declined, whereas the plant growth under suddenly decreased water temperature conditions, which simulated summer upwelling events, was maintained or slightly increased. In the fall 2014 experimental period, Z. marina shoots under the suddenly elevated water temperature condition (27 °C) showed significant decreases in shoot density, biomass, and leaf productivity by ca. 40–80%, whereas plants held under the ambient water temperature condition (20 °C) maintained or slightly increased their growths. In the summer 2015 experimental period, Z. marina under the sudden temperature reduction conditions (20 °C) maintained relatively high shoot density and leaf productivity, whereas plants under the in situ high water temperature condition (27 °C) showed significantly reduced growths. The growth of Z. marina shoots, which were cultured in the optimal temperature condition (20 °C), was enhanced by increasing underwater irradiance. However, the adverse effects of high water temperature on Z. marina growth could not be reversed by increasing irradiance levels at the end of the experiments. According to the results of this study, the intensified sea surface water temperature anomalies induced by global climate change will alter the growth and distribution of temperate seagrasses and, consequently, the structure and function of the coastal seagrass ecosystems in the temperate region.

Abstract:Current rates of ocean warming are predicted to exacerbate ongoing declines in seagrass populations. Above-ground responses of seagrass to increasing temperatures have been studied from a direct physiological perspective while indirect effects, including changes to microbially-mediated below-ground processes, remain poorly understood. To test potential effects of increased temperature on seagrass growth and associated microbial communities, we sampled seagrass beds experiencing ambient and elevated water temperatures at Lake Macquarie, Australia. Sites with warmer water were associated with a plume from a power station discharge channel with temperatures analogous to conditions predicted by 2100 under current rates of ocean warming (+3°C). The microbial community composition in both sediments and leaf tissues varied significantly between warm and ambient water temperatures with higher relative abundances of putative sulphate-reducing bacteria such as Desulfocapsaceae, Desulfobulbaceae and Desulfosarcinaceae in sedimentary communities in warm water. Above-ground biomass and seagrass growth rates were greater at warm sites while below-ground biomass and detrital decomposition rates showed no difference suggesting potential buffering of temperature effects below-ground. These findings suggest a 3°C rise in temperate regions is unlikely to induce mortality in seagrass however, it may shift microbial communities towards more homogenous structure and composition.

Abstract:Ocean acidification is one of the major global environmental problems facing humankind today, and it has far-reaching impacts on marine organisms and the entire marine ecosystem. Zostera japonica, an important supporting species of intertidal seagrass beds, exhibits high photosynthetic productivity and plays an important role in the carbon cycle of nearshore waters. However, little is known about the characteristics, processes, and mechanisms of its response to ocean acidification. In this study, we conducted a 120-day acidification experiment in Z. japonica; here, plants underwent four leaf regeneration cycles to reveal the response mechanism of Z. japonica to ocean acidification (OA). We found that acidification significantly affected the seedling stage of Z. japonica, impacting leaf regeneration cycles by altering physiological and molecular responses. In one leaf regeneration cycle, the short-term exposure to CO2 affected the seagrass parameters, such as the regulation of inorganic carbon uptake modes and the regulation of photosynthesis between the dark and light reactions, with the potential to affect the carbon sinks of the marine organisms. The long-term effects on the regulation of antioxidant enzymes and antioxidant metabolites, caused an improvement in the marine life adaptation to OA. In a comparison of the different leaf regeneration cycles, the response pattern of Z. japonica showed an offset of the acidification during the short cycles and an adaption to the acidification during the long cycles. This study revealed the response mechanism of Z. japonica to OA at different time scales and could provide a theoretical basis for accurately assessing the impact of OA on seagrass and the entire seagrass bed ecosystem.
Carbon economy of Mediterranean seagrasses in response to thermal stress

Abstract:Increased plant mortality in temperate seagrass populations has been recently observed after summer heatwaves, although the underlying causes of plant death are yet unknown. The potential energetic constrains resulting from anomalous thermal events could be the reason that triggered seagrass mortality, as demonstrated for benthic invertebrates. To test this hypothesis, the carbon balance of Posidonia oceanica and Cymodocea nodosa plants from contrasting thermal environments was investigated during a simulated heatwave, by analyzing their photosynthetic performance, carbon balance (ratio photosynthesis:respiration), carbohydrates content, growth and mortality. Both species were able to overcome and recover from the thermal stress produced by the six-week exposure to temperatures 4 °C above mean summer levels, albeit plants from cold waters were more sensitive to warming than plants from warm waters as reflected by their inability to maintain their P:R ratio unaltered. The strategies through which plants tend to preserve their energetic status varied depending on the biology of the species and the thermal origin of plants. These included respiratory homeostasis (P. oceanica warm-plants), carbon diversion from growth to respiration (C. nodosa cold-plants) or storage (P. oceanica warm-plants) and changes in biomass allocation (C. nodosa warm-plants). Findings suggest an important geographic heterogeneity in the overall response of Mediterranean seagrasses to warming with potential negative impacts on the functions and services offered by seagrass meadows including among others their capacity for carbon sequestration and carbon export to adjacent ecosystems.
Temperature Effects on Leaf and Epiphyte Photosynthesis, Bicarbonate Use and Diel O2 Budgets of the Seagrass Zostera marina L.

Abstract:Ocean warming along with nutrient enrichment are major stressors causing global seagrass decline. While the effects of global warming on metabolic parameters in seagrasses are well described, the effect of increasing temperature on the epiphytic overgrowth of seagrass leaves and the consequences for the seagrass plant are poorly understood. Here, we investigated the effects of elevating temperature on the photosynthetic efficiency of the seagrass species Zostera marina L. and its associated epiphytes, to explore how ocean warming might affect epiphytism in seagrasses. Gas exchange and final pH measurements on bare seagrass leaves, leaves with epiphytes, and epiphytes separated from seagrass leaves were used to quantify photosynthesis and respiration rates, and the inorganic carbon extraction capacity of leaves and epiphytes as a function of photon scalar irradiance and temperature (12, 17, 22, and 27°C). Seagrass without epiphytic biofilm had a high ability to exploit the incoming irradiance regardless of the light intensity and temperature, shown as continuously high light use efficiency and maximum net photosynthesis rates. The presence of epiphytic biofilm on the seagrass leaves impaired plant photosynthesis by increasing light requirements and reducing the photosynthetic efficiency (especially at 27°C). Epiphytes showed the lowest respiration rates in darkness and had the highest oxygen surplus over diel cycles up to 22°C, whereas bare leaves had the highest diel oxygen surplus at 27°C. Both bare leaves and epiphytes lost the ability to utilize bicarbonate at 27°C, and epiphytes also did not show use of bicarbonate at 12°C. Our results indicate a competitive advantage for epiphytes in cold CO 2 -rich environments, whereas seagrass with bare leaves could be less affected under elevated seawater temperatures.
