Abstract:Covalent adaptable networks are polymers that can alter the arrangement of network connections by bond exchange reactions where an active unit attaches to an existing bond then kicks off its pre-existing peer to form a new bond. When the polymer is stretched, bond exchange reactions lead to stress relaxation and plastic deformation, or the so-called reforming. In addition, two pieces of polymers can be rejoined together without introducing additional monomers or chemicals on the interface, enabling welding and reprocessing. Although covalent adaptable networks have been researched extensively in the past, knowledge about the macromolecular level network alternations is limited. In this study, molecular dynamics simulations are used to investigate the macromolecular details of bond exchange reactions in a recently reported epoxy system. An algorithm for bond exchange reactions is first developed and applied to study a crosslinking network formed by epoxy resin DGEBA with the crosslinking agent tricarballylic acid. The trace of the active units is tracked to show the migration of these units within the network. Network properties, such as the distance between two neighboring crosslink sites, the chain angle, and the initial modulus, are examined after each iteration of the bond exchange reactions to provide detailed information about how material behaviors and macromolecular structure evolve. Stress relaxation simulations are also conducted. It is found that even though bond exchange reactions change the macroscopic shape of the network, microscopic network characteristic features, such as the distance between two neighboring crosslink sites and the chain angle, relax back to the unstretched isotropic state. Comparison with a recent scaling theory also shows good agreement.
Response of Biopolymer Networks Governed by the Physical Properties of Cross-Linking Molecules.

Abstract:In this study, we examine how the physical properties of cross-linking molecules affect the bulk response of bio-filament networks, an outstanding question in the study of biological gels and the cytoskeleton. We show that the stress-strain relationship of such networks typically undergoes linear increase - strain hardening - stress serration - total fracture transitions due to the interplay between the bending and stretching of individual filaments and the deformation and breakage of cross-linkers. Interestingly, the apparent network modulus is found to scale with the linear and rotational stiffness of the crosslinks to a power exponent of 0.78 and 0.13, respectively. In addition, the network fracture energy will reach its minimum at intermediate rotational compliance values, reflecting the fact that most of the strain energy will be stored in the distorted filaments with rigid cross-linkers while the imposed deformation will be "evenly" distributed among significantly more crosslinking molecules with high rotational compliance.

Abstract:Soft materials featuring dynamic networks represent a burgeoning frontier in materials science, offering multifaceted applications spanning soft robotics, biomaterials, and flexible electronics. Unraveling the time-dependent constitutive behavior of these materials, rooted in dynamic networks, stands as a pivotal pursuit for engineering advancements. Herein, we fabricate a tough and extreme-temperature-tolerant glycerogel with a polymer network crosslinked by metal-coordination crosslinkers and conduct a thorough analysis of its intricate mechanical responses across monotonic loading, relaxation, creep, and cyclic tests. We then develop a physically grounded constitutive model integrating the dynamics of crosslinker association/dissociation and polymer chain diffusion, furnishing a holistic framework to elucidate their interplay. We employ a statistical description, using density functions of chains in terms of end-to-end vectors, to characterize network reconfiguration. The evolution of chain density under external load, mediated by crosslinker kinetics and chain diffusion in a viscous medium, leads to intriguing variations in elastic energy and stress responses. Through meticulous experimental validation and numerical simulations, we demonstrate the efficacy of the model in forecasting the mechanical behavior of dynamic polymer networks under diverse loading scenarios, encompassing strain rate effects, stress relaxation, Mullins effect, and self-recovery phenomena. Our findings provide valuable insights into the design and optimization of dynamic network-based materials for diverse applications in biomedical and engineering fields.

Abstract:Elastomers and hydrogels with dynamic network structures have received considerable research interest due to their high strength and toughness, and self-healing ability. While the general mechanical responses of dynamic polymer networks have been investigated, the study on their fatigue behaviors is still in its infancy. In this work, we employ experimental and theoretical approaches to gain a comprehensive understanding of the mechanical response of an elastomer with physical crosslinkers under fatigue tests up to 10,000 cycles. The constitutive model characterizes the network configuration using the density distribution function of the polymer chains, which is represented by a state variable denoting the stress-free configuration of each individual chain. The model takes into account the reversible dissociation/association of dynamic bonding facilitated by the physical crosslinkers. As a result, polymer chains can repeatedly release the involved strain energy and transform between different stress-free configurations. A multidispersed reaction process is further employed to describe the state transition of polymer chains, enabling the model to elucidate the progressive damage in long-term relaxation and fatigue tests by recording and updating the density distribution function of chain state. In addition, the reversible transition of the polymer chain between different state configurations enables the model to capture the rate-dependent stress response and relaxation behaviors of physically crosslinked elastomers and gels. Remarkably, our microscopic model, which requires only five parameters, offers an efficient approach to describe the mechanical responses of elastomers and gels with dynamic bonds.
Statistical mechanical constitutive theory of polymer networks: The inextricable links between distribution, behavior, and ensemble

Abstract:A fundamental theory is presented for the mechanical response of polymer networks undergoing large deformation which seamlessly integrates statistical mechanical principles with macroscopic thermodynamic constitutive theory. Our formulation permits the consideration of arbitrary polymer chain behaviors when interactions among chains may be neglected. This careful treatment highlights the naturally occurring correspondence between single-chain mechanical behavior and the equilibrium distribution of chains in the network, as well as the correspondences between different single-chain thermodynamic ensembles. We demonstrate these important distinctions with the extensible freely jointed chain model. This statistical mechanical theory is then extended to the continuum scale, where we utilize traditional macroscopic constitutive theory to ultimately retrieve the Cauchy stress in terms of the deformation and polymer network statistics. Once again using the extensible freely jointed chain model, we illustrate the importance of the naturally occurring statistical correspondences through their effects on the stress-stretch response of the network. We additionally show that these differences vanish when the number of links in the chain becomes sufficiently large enough, and discuss why certain methods perform better than others before this limit is reached.

Abstract:A new class of polymers characterized by dynamic cross-links is analyzed from a mechanical point of view. A thermodynamically consistent model is developed within the Lagrangian framework for polymers that can rearrange their internal cross-links. Such a class of polymers has the capability to reset their internal microstructure and the microscopic remodeling mechanism leads to a behavior similar to that of an elastic fluid. These materials can potentially be used in several fields, such as in biomechanics, smart materials, morphing materials to cite e few. However, a comprehensive understanding is necessary before we can predict their behavior and perform material design for advanced technologies. The proposed formulation-following a statistical approach adapted from classical rubber elasticitye is based on the evolution of the molecular chains' end-to-end distance distribution function. This distribution is allowed here to evolve with time, starting from an initial stress-free state and depending on the deformation history and the cross-link attachment/detachment kinetics. Some simple examples are finally presented and discussed to illustrate the capability and generality of the developed approach.
Statistical field theory for nonlinear elasticity of polymer networks with excluded volume interactions

Abstract:Polymer networks formed by cross linking flexible polymer chains are ubiquitous in many natural and synthetic soft-matter systems. Current micromechanics models generally do not account for excluded volume interactions except, for instance, through imposing a phenomenological incompressibility constraint at the continuum scale. This work aims to examine the role of excluded volume interactions on the mechanical response. The approach is based on the framework of the self-consistent statistical field theory of polymers, which provides an efficient mesoscale approach that enables the accounting of excluded volume effects without the expense of large-scale molecular modeling. A mesoscale representative volume element is populated with multiple interacting chains, and the macroscale nonlinear elastic deformation is imposed by mapping the end-to-end vectors of the chains by this deformation. In the absence of excluded volume interactions, it recovers the closed-form results of the classical theory of rubber elasticity. With excluded volume interactions, the model is solved numerically in three dimensions using a finite element method to obtain the energy, stresses, and linearized moduli under imposed macroscale deformation. Highlights of the numerical study include: (i) the linearized Poisson's ratio is very close to the incompressible limit without a phenomenological imposition of incompressibility; (ii) despite the harmonic Gaussian chain as a starting point, there is an emergent strain-softening and strain-stiffening response that is characteristic of real polymer networks, driven by the interplay between the entropy and the excluded volume interactions; and (iii) the emergence of a deformation-sensitive localization instability at large excluded volumes.

Abstract:Polymer network models construct the constitutive relationships of a broader polymer network from the behavior of a single polymer chain (e.g. viscoelastic response to applied forces, applied electromagnetic fields, etc.). Network models have been used for multiscale phenomena in a variety of contexts such as rubber elasticity, soft multifunctional materials, biological materials, and even the curing of polymers. For decades, a myriad of polymer network models have been developed with differing numbers of chains, arranged in different ways, and with differing symmetries. To complicate matters further, there are also competing assumptions for how macroscopic variables (e.g. deformation) are related to individual chains within the network model. In this work, we propose a simple, intuitive assumption for how the network locally rotates relative to applied loading (e.g. stresses, external fields) and show that this assumption unifies many of the disparate polymer network models--while also recovering one of the most successful models for rubber elasticity, the Arruda-Boyce $8$-chain model. The new assumption is then shown to make more intuitive predictions (than prior models) for stimuli-responsive networks with orientational energies (e.g. electroactive polymers), which is significant for shape morphing and designing high degree of freedom actuators (e.g. for soft robotics). Lastly, we unveil some surprising consequences of the new model for the phases of multistable biopolymer and semi-crystalline networks.

Abstract:Mucus linings serve as the body's first line of defense at exposed surfaces of the eye and respiratory, gastrointestinal, and cervicovaginal tracts. The high viscoelasticity and adhesivity of mucus traps and limits exposure to foreign pathogens,[1,2] toxins,[3] and environmental ultrafine particles,[1,4] which are all typically removed by normal mucus clearance mechanisms. Numerous studies have demonstrated that human mucus also strongly immobilizes conventional synthetic nanoparticles,[5-7] and therefore represents a hurdle for localized drug and gene delivery at mucosal surfaces, such as aerosol-based gene carriers for cystic fibrosis gene therapy.[8] To increase the bioavailability of cargo therapeutics, it is important that carrier particles rapidly penetrate mucus to avoid being shed.
From statistical polymer physics to nonlinear elasticity

Abstract:A polymer-chain network is a collection of interconnected polymer-chains, made themselves of the repetition of a single pattern called a monomer. Our first main result establishes that, for a class of models for polymer-chain networks, the thermodynamic limit in the canonical ensemble yields a hyperelastic model in continuum mechanics. In particular, the discrete Helmholtz free energy of the network converges to the infimum of a continuum integral functional (of an energy density depending only on the local deformation gradient) and the discrete Gibbs measure converges (in the sense of a large deviation principle) to a measure supported on minimizers of the integral functional. Our second main result establishes the small temperature limit of the obtained continuum model (provided the discrete Hamiltonian is itself independent of the temperature), and shows that it coincides with the $\Gamma$-limit of the discrete Hamiltonian, thus showing that thermodynamic and small temperature limits commute. We eventually apply these general results to a standard model of polymer physics from which we derive nonlinear elasticity. We moreover show that taking the $\Gamma$-limit of the Hamiltonian is a good approximation of the thermodynamic limit at finite temperature in the regime of large number of monomers per polymer-chain (which turns out to play the role of an effective inverse temperature in the analysis).

Abstract:Disordered networks of semiflexible filaments are common support structures in biology. Familiar examples include fibrous matrices in blood clots, bacterial biofilms, and essential components of cells and tissues of plants, animals, and fungi. Despite the ubiquity of these networks in biomaterials, we have only a limited understanding of the relationship between their structural features and highly strain-sensitive mechanical properties. In this work, we perform simulations of three-dimensional networks produced by the irreversible formation of crosslinks between linker-decorated semiflexible filaments. We characterize the structure of networks formed by a simple diffusion-dependent assembly process and measure their associated steady-state rheological features at finite temperature over a range of applied prestrains that encompass the strain-stiffening transition. We quantify the dependence of network connectivity on crosslinker availability and detail the associated connectivity dependence of both linear elasticity and nonlinear strain stiffening behavior, drawing comparisons with prior experimental measurements of the crosslinker concentration-dependent elasticity of actin gels.

Abstract:Rheological properties of soft, complex materials are essential to biological systems. Networks consisting of semiflexible polymers, which are determinants for dynamics and mechanics inside and outside cells, are described by theories that neglect molecular interactions between individual filaments. Using a surface modification approach, it is shown that such interactive forces significantly contribute to the viscoelastic properties of isotropic network structures. Established model theories, developed to capture the mechanical behavior of soft, complex materials composed of semiflexible polymers, assume that entropic interactions between filaments are primarily responsible for determining the mechanical response. In recent studies, the generally accepted tube model has been challenged in terms of this basic assumption about filament–filament interactions, but also because of its predictions regarding the frequency dependence of the elastic modulus in the intermediate frequency regime. A central question is how molecular interactions and friction between network constituents influence the rheological response of isotropic entangled networks of semiflexible polymers. It has been previously shown that friction forces between aligned pairs of actin filaments are not negligible. Here, the influence of friction forces and attractive interactions on network rheology is systematically investigated by means of targeted surface modification. It is shown that these forces have a qualitative and quantitative influence on the viscoelastic properties of semiflexible polymer networks and contribute to their response to nonlinear deformations. By comparing two polymer model systems with respect to their surface compositions, a possible explanation is given about the origin of acting forces on a molecular level.

Abstract:Live cells have ambiguous mechanical properties. They were often described as either elastic solids or viscoelastic fluids and have recently been classified as soft glassy materials characterized by weak power-law rheology. Nonlinear rheological measurements have moreover revealed a pronounced inelastic response indicative of a competition between stiffening and softening. It is an intriguing question whether these observations can be explained from the material properties of much simpler in-vitro reconstituted networks of biopolymers that serve as reduced model systems for the cytoskeleton. Here, we explore the mechanism behind the inelastic response of cells and biopolymer networks, theoretically. Our analysis is based on the model of the inelastic glassy wormlike chain that accounts for the nonlinear polymer dynamics and transient crosslinking in biopolymer networks. It explains how inelastic and kinematic-hardening type behavior naturally emerges from the antagonistic mechanisms of viscoelastic stress-stiffening due to the polymers and inelastic fluidization due to bond breaking. It also suggests a simple set of schematic constitutive equations which faithfully reproduce the rich inelastic phenomenology of biopolymer networks and cells.

Abstract:Although evidence has indicated that the presence of catch bond-like crosslinking proteins can significantly alter the viscoelastic response of bio-polymer networks, a detailed mechanistic understanding is still lacking. Here we report a computational investigation to examine how catch or slip-bond kinetics of crosslinking molecules affects the time-dependent behavior of realistic F-actin and collagen networks. Specifically, it was found that, under oscillating shear, the rate of cross-linkers getting ruptured and then reformed reaches the maximum at intermediate driving frequency, eventually leading to a locally peaked bulk loss modulus of the network. Interestingly, when a pre-stress was applied the position of such peak shifted to higher frequencies for networks with slip bond cross-linkers. In comparison, the peaked loss modulus of actin network constructed by catch bond-like crosslinking molecules was reached at a lower driving frequency because of the stabilization effect of pre-stress on such cross-linkers. Similarly, the appearance of catch bond cross-linkers was also found to greatly alter the hysteresis of networks under staircase of sinusoidal shear excitations, all in good agreement with experimental observations. Finally, we showed that stress relaxation in slip-bond collagen networks becomes faster under increasing imposed strain, in direct contrast to networks constructed by catch bond crosslinking proteins where the characteristic timescale for stress decay grows with the strain. By elucidating the mechanism by which crosslink kinetics profoundly affects the bulk behavior of biopolymer networks, our study provides useful insights for the development of future biomaterials and understanding the physical role of cytoskeleton in various cellular processes.

Abstract:The effect of network chain distribution on the mechanical behavior of elastomers is one of the long-standing problems in rubber mechanics. The classical theory of rubber elasticity is built upon the assumption of entropic elasticity of networks whose constitutive strands are of uniform length. The kinetic theories for vulcanization, computer simulations, and indirect experimental measurements all indicate that the microstructure of vulcanizates is made of polymer strands with a random distribution of length. The polydispersity in strand length is expected to control the mechanical strength of rubber as the overloaded short strands break at small deformations and transfer the load to the longer strands. The purpose of the contributions presented in this thesis is to present simple theories of rubber mechanics that take into account the length distribution of strands and its effect on elasticity and the onset of bulk failure in unfilled and filled elastomers. In unfilled system, the population of short chains is identified as the culprits for damage initiation. Upon deformation of a polydisperse network, shorter strands break at considerably smaller stretches compared to the longer ones. The network alteration continues concurrent with increasing deformation and controls the onset of mechanical failure. In the filled networks, the degradation in network mechanical behavior is assumed to be controlled by the adhesive failure of the short strands adsorbed onto the filler surface. The finite extensibility of the short adsorbed strands is a key determinant of mechanical strength.

A Combined Finite Element-Langevin Dynamics (Fem-Ld) Approach for Analyzing the Mechanical Response of Bio-Polymer Networks

Abstract:A Langevin dynamics based formulation is proposed to describe the shape fluctuations of biopolymer filaments. We derive a set of stochastic partial differential equations (SPDEs) to describe the temporal evolution of the shape of semiflexible filaments and show that the solutions of these equations reduce to predictions from classical modal analysis. A finite element formulation to solve these SPDEs is also developed where, besides entropy, the finite deformation of the filaments has been taken into account. The validity of the proposed finite element-Langevin dynamics (FEM-LD) approach is verified by comparing the simulation results with a variety of theoretical predictions. The method is then applied to study the mechanical behavior of randomly cross-linked F-actin networks. We find that as deformation progresses, the response of such networks undergoes transitions from being entropy dominated to being governed by filament bending and then, eventually, to being dictated by filament stretching. The levels of macroscopic stress at which these transitions take place were found to be around 1% and 10%, respectively, of the initial bulk modulus of the network, in agreement with recent experimental observations.
Crosslinker mobility weakens transient polymer networks

Abstract:Transient networks comprised of polymers connected by short-lived bonds are a common design theme for both biological and synthetic materials. Transient bonds can provide mechanical rigidity, while still allowing for visco-elastic flows on timescales longer than the bond lifetime. In many biological polymer networks such as the actin cytoskeleton, the short-lived bonds are formed by accessory proteins that diffuse away after unbinding. By contrast, bonds in synthetic networks, such as the pendant groups of telechelic polymers, can only rebind in the same location. Using a recently developed theoretical model of the fracturing of visco-elastic materials, we here investigate the effect of linker mobility on the bond dynamics of a network under stress. We find that although mean field properties such as the average bond linker lifetime are barely affected by bond mobility, networks cross linked by mobile bonds fracture more readily due to 'leaking' of linkers from crack areas to less stressed regions within the network. We propose a theoretical model to describe the redistribution of mobile linkers, which we validate by simulations. Our work offers insight into a potential trade-off that cells face, between fracture strength versus the modularity and tight dynamic control offered by mobile linkers.

Mechanics and thermodynamics of contractile entropic biopolymer networks

Abstract:Contractile biopolymer networks, such as the actomyosin meshwork of animal cells, are ubiquitous in living organisms. The active gel theory, which provides the thermodynamic framework for these materials, has been mostly used in conjunction with the assumption that the microstructure of the biopolymer network is based on rigid rods. However, experimentally, crosslinked actin network exhibits entropic elasticity. Here we combine an entropic elasticity kinetic theory, in the spirit of the Green and Tobolsky model of transiently crosslinked networks, with an active flux modelling biological activity. We determine this active flux using Onsager reciprocal relations and interpret the corresponding microscopic dynamics. We obtain a closed-form model of the macroscopic mechanical behaviour. We show how this model can be written using the framework of multiplicative strain gradient decomposition, which is convenient for the resolution of such problems.

Abstract:Statistical mechanics is an important tool for understanding polymer electroelasticity because the elasticity of polymers is primarily due to entropy. However, a common approach for the statistical mechanics of polymer chains, the Gaussian chain approximation, misses key physics. By considering the nonlinearities of the problem, we show a strong coupling between the deformation of a polymer chain and its dielectric response; that is, its net dipole. When chains with this coupling are cross-linked in an elastomer network and an electric field is applied, the field breaks the symmetry of the elastomer's elastic properties, and, combined with electrostatic torque and incompressibility, leads to intrinsic electrostriction. Conversely, deformation can break the symmetry of the dielectric response leading to volumetric torque (i.e., a couple stress or torque per unit volume) and asymmetric actuation. Both phenomena have important implications for designing high-efficiency soft actuators and soft electroactive materials; and the presence of mechanisms for volumetric torque, in particular, can be used to develop higher degree of freedom actuators and to achieve bioinspired locomotion.
