The use of elemental sulfur (S0) instead of sulfate (SO42-) as electron acceptor reduces electron donor requirements four-fold, lowering process costs, and expanding the range of operating conditions to a more acidic pH. We previously reported autotrophic S0 reduction using an industrial mesophilic granular sludge as inoculum under thermoacidophilic conditions. Here, we examined the effect of pH on the S0 reduction performance of the same inoculum, in a gas-lift reactor run at 30°C under neutral (pH 6.9) and acidic (pH 3.8) conditions, continuously fed with mineral media and H2 and CO2. Steady-state volumetric sulfide production rates (VSPR) dropped 2.5-fold upon transition to acidic pH, from 1.79 ± 0.18 g S2-·L-1·d-1 to 0.71 ± 0.07 g S2-·L-1·d-1. Microbial community composition was analyzed using 16S rRNA gene amplicon sequencing. At neutral pH (6.9), the high relative abundance of the S0-reducing genus Sulfurospirillum, previously known only for heterotrophic members, combined with the presence of Acetobacterium and detection of acetate, suggests an important role for heterotrophic S0 reduction facilitated by acetogenesis. Conversely, at acidic pH (3.9), S0 reduction appeared autotrophic, as indicated by the high relative abundance of Desulfurella.

The detection of acetic acid and the relative abundance of Acetobacterium at pH 6.9 pointed towards acetogenesis, explaining the dominance of the heterotrophic genus Sulfurospirillum in this H2 and CO2-fed bioreactor.

Abstract:Sulfur is not only one of the most abundant elements on the Earth, but it is also essential to all living organisms. As life likely began and evolved in a hydrogen sulfide (H2S)-rich environment, sulfur metabolism represents an early form of energy generation via various reactions in prokaryotes and has driven the sulfur biogeochemical cycle since. It has long been known that H2S is toxic to cells at high concentrations, but now this gaseous molecule, at the physiological level, is recognized as a signaling molecule and a regulator of critical biological processes. Recently, many metabolites of H2S, collectively called reactive sulfur species (RSS), have been gradually appreciated as having similar or divergent regulatory roles compared with H2S in living organisms, especially mammals. In prokaryotes, even in bacteria, investigations into generation and physiology of RSS remain preliminary and an understanding of the relevant biological processes is still in its infancy. Despite this, recent and exciting advances in the fields are many. Here, we discuss abiotic and biotic generation of H2S/RSS, sulfur-transforming enzymes and their functioning mechanisms, and their physiological roles as well as the sensing and regulation of H2S/RSS.
The enrichment of a specialized elemental sulfur–producing bacterium under extremely acidic condition

Abstract:In the gas biodesulfurization process, converting sulfides into elemental sulfur (S0) for reuse holds significant environmental, economic, resource, and social values. While the influence of pH on microbial community structure and enzyme activity is recognized, its effect on S0 yield has been underestimated. This study delves into how pH fluctuations affect the S0 yield in a biodesulfurization system. Under extremely acidic condition (pH < 1), the selectivity of S0 reached 90% even at high O2/H2S supply ratio of 1.8–2.1. Metagenomic analyses showed that as the pH decreased from neutral–alkaline to extremely acidic range, the dominant bacterium gradually shifted from Acidithiobacillus (37.0%), Acidiphilium (1.5%) and Thiomonas (1.2%), which possess complete sulfur metabolism (from sulfide to sulfate) genes, to Mycobacterium (79.2%) possessing the S0–synthesis gene (sqr) without the S0–oxidizing genes (ETHE1, sor, hdr, dsr and the Sox and S4I complex genes), resulting in a robust S0 yield in extremely acidic period. Various pathways for respiratory sulfur oxidation were present in different pH periods. The extremely acidic condition facilitated the selection of specific microorganisms by creating a highly selective environment, promoting the growth of acidophilic or acid-tolerant microorganisms with unique traits and altering the sulfur–metabolic pathways. These finding filled a knowledge gap regarding the impact of pH on sulfur–metabolic pathways and offered a new strategy for gaining S0.

Harnessing the potential of the microbial sulfur cycle for environmental biotechnology

Abstract:The sulfur cycle is a complex biogeochemical cycle characterized by the high variability in the oxidation states of sulfur. While sulfur is essential for life processes, certain sulfur compounds, such as hydrogen sulfide, are toxic to all life forms. Micro-organisms facilitate the sulfur cycle, playing a prominent role even in extreme environments, such as soda lakes, acid mine drainage sites, hot springs, and other harsh habitats. The activity of these micro-organisms presents unique opportunities for mitigating sulfur-based pollution and enhancing the recovery of sulfur and metals. This review highlights the application of sulfur-oxidizing and -reducing micro-organisms in environmental biotechnology through three illustrative examples. Additionally, it discusses the challenges, recent trends, and prospects associated with these applications.

Abstract:It is now well established that microbial sulphate-reduction can proceed in environments with a pH<5. This review summarizes existing reports on sulphate reduction at low pH and discusses possible pH effects on sulphate-reducing bacteria. Microbial sulphate reduction has been observed in acidic lakes, wetlands, mesocosms, acidic sulphate soils and bioreactors. Possible inhibitory factors include the metabolites H(2)S and organic acids, which can be toxic depending on pH. Metal sulphide precipitation and competition with other bacteria, namely iron-reducing bacteria, can inhibit sulphate reduction. Theoretical considerations show that normal sulphate reduction rates are too low to maintain a neutral micro niche in an acidic environment. The first acidotolerant sulphate-reducing bacteria have been isolated recently.

Abstract:Elemental sulfur (S 0 ) plays a vital role in the coupled cycling of sulfur and iron, which in turn affects the transformation of carbon and various pollutants. These processes have been well characterized under static anoxic or oxic conditions, however, how the natural redox fluctuations affect the bio-mediated sulfur cycling and coupled iron cycling remain enigmatic. The present work examined S 0 disproportionation as driven by natural microbial communities under fluctuating redox conditions and the contribution of S 0 disproportionation to ferrihydrite transformation. Samples were incubated at either neutral or alkaline pH values, applying sequential anaerobic, aerobic and anaerobic conditions over 60 days. Under anaerobic conditions, S 0 was found to undergo disproportionation to sulfate and sulfide, which subsequently reduced ferrihydrite at both pH 7.4 and 9.5. Ferrihydrite promoted S 0 disproportionation by scavenging biogenic sulfide and maintaining a suitable degree of sulfate formation. After an oxic period, during the subsequent anoxic incubation, bioreduction of sulfate occurred and the biogenic sulfide reduced iron (hydr)oxides at a rate approximately 25 % lower than that observed during the former anoxic period. A 16S rDNA-based microbial community analysis revealed changes in the microbial community in response to the redox fluctuations, implying an intimate association with the coupled cycling of sulfur and iron. Microscopic and spectroscopic analyses confirmed the S 0 -mediated transformation of ferrihydrite to crystalline iron (hydr)oxide minerals such as lepidocrocite and magnetite and the formation of iron sulfides precipitated under fluctuating redox conditions. Finally, a reaction mechanism based on mass balance was proposed, demonstrating that bio-mediated sulfur transformation maintained a sustainable redox reaction with iron (hydr)oxides under fluctuating anaerobic-aerobic-anaerobic conditions tested in this study. Altogether, the finding of our study is critical for obtaining a more complete understanding of the dynamics of iron redox reactions and pollutant transformation in sulfur-rich aquatic environments.

Abstract:Acidophilic sulphate‐reducing bacteria (aSRB) perform dissimilatory sulphate reduction under low pH and high metal conditions. They play key roles in the sulphur and carbon cycles in natural acidic environments, emphasized by their interactions with other microbial groups, which create an intricate ecological network. Potential applications of aSRB and their products include treating acid mine drainage, technological uses of metal sulphides, and applications as biosignatures. Acidophilic sulphate‐reducing bacteria (aSRB) are widespread anaerobic microorganisms that perform dissimilatory sulphate reduction and have key adaptations to tolerate acidic environments (pH <5.0), such as proton impermeability and Donnan potential. This diverse prokaryotic group is of interest from physiological, ecological, and applicational viewpoints. In this review, we summarize the interactions between aSRB and other microbial guilds, such as syntrophy, and their roles in the biogeochemical cycling of sulphur, iron, carbon, and other elements. We discuss the biotechnological applications of aSRB in treating acid mine drainage (AMD, pH <3), focusing on their ability to produce biogenic sulphide and precipitate metals, particularly in the context of utilizing microbial consortia instead of pure isolates. Metal sulphide nanoparticles recovered after AMD treatment have multiple potential technological uses, including in electronics and biomedicine, contributing to a cost‐effective circular economy. The products of aSRB metabolisms, such as biominerals and isotopes, could also serve as biosignatures to understand ancient and extant microbial life in the universe. Overall, aSRB are active components of the sulphur and carbon cycles under acidic conditions, with potential natural and technological implications for the world around us.

Abstract:Biochemical oxidation and reduction are the principle of biological water and wastewater treatment, in which electron donor and/or acceptor shall be provided. Elemental sulfur (S0) as a non-toxic and easily available material with low price, possesses both reductive and oxidative characteristics, suggesting that it is a suitable material for water and wastewater treatment. Recent advanced understanding of S0-respiring microorganisms and their metabolism further stimulated the development of S0-based technologies. As such, S0-based biotechnologies have emerged as cost-effective and attractive alternatives to conventional biological methods for water and wastewater treatment. For instance, S0-driven autotrophic denitrification substantially lower the operational cost for nitrogen removal from water and wastewater, compared to the conventional process with exogenous carbon source supplementation. The introduction of S0 can also avoid secondary pollution commonly caused by overdose of organic carbon. S0 reduction processes cost-effectively mineralize organic matter with low sludge production. Biological sulfide production using S0 as electron acceptor is also an attractive technology for metal-laden wastewater treatment, e.g. acid mine drainage. This paper outlines an overview of the fundamentals, characteristics and advances of the S0-based biotechnologies and highlights the functional S0-related microorganisms. In particular, the mechanisms of microorganisms accessing insoluble S0 and feasibility to improve S0 bio-utilization efficiency are critically discussed. Additionally, the research knowledge gaps, current process limitations, and required further developments are identified and discussed.

Abstract:Acid mine drainage (AMD) pollutes natural waters, but some impacted systems show natural attenuation. We sought to identify the biogeochemical mechanisms responsible for the natural attenuation of AMD. We hypothesized that biogenic sulfide-mediated iron reduction is one mechanism and tested this in an experimental model system. We found sulfate reduction occurred under acidic conditions and identified a suite of sulfate-reducing bacteria (SRB) belonging to the groups Desulfotomaculum, Desulfobacter, Desulfovibrio, and Desulfobulbus. Iron reduction was not detected in microcosms when iron-reducing bacteria or SRB were selectively inhibited. SRB also did not reduce iron enzymatically. Rather, the biogenic sulfide produced by SRB was found to be responsible for the reduction of iron at low pH. Addition of organic substrates and nutrients stimulated iron reduction and increased the pH. X-ray diffraction and an electron microprobe analysis revealed that the polycrystalline, black precipitate from SRB bioactive samples exhibited a greater diversity of iron chalcogenide minerals with reduced iron oxidation states, and minerals incorporating multiple metals compared to abiotic controls. The implication of this study is that iron reduction mediated by biogenic sulfide may be more significant than previously thought in acidic environments. This study not only describes an additional mechanism by which SRB attenuate AMD, which has practical implications for AMD-impacted sites, but also provides a link between the biogeochemical cycling of iron and sulfur.

Sulfur species as redox partners and electron shuttles for ferrihydrite reduction by Sulfurospirillum deleyianum

Abstract:Iron(III) (oxyhydr)oxides can represent the dominant microbial electron acceptors under anoxic conditions in many aquatic environments, which makes understanding the mechanisms and processes regulating their dissolution and transformation particularly important. In a previous laboratory-based study, it has been shown that 0.05 mM thiosulfate can reduce 6 mM ferrihydrite indirectly via enzymatic reduction of thiosulfate to sulfide by the sulfur-reducing bacterium Sulfurospirillum deleyianum, followed by abiotic reduction of ferrihydrite coupled to reoxidation of sulfide. Thiosulfate, elemental sulfur, and polysulfides were proposed as reoxidized sulfur species functioning as electron shuttles. However, the exact electron transfer pathway remained unknown. Here, we present a detailed analysis of the sulfur species involved. Apart from thiosulfate, substoichiometric amounts of sulfite, tetrathionate, sulfide, or polysulfides also initiated ferrihydrite reduction. The portion of thiosulfate produced during abiotic ferrihydrite-dependent reoxidation of sulfide was about 10% of the total sulfur at maximum. The main abiotic oxidation product was elemental sulfur attached to the iron mineral surface, which indicates that direct contact between microorganisms and ferrihydrite is necessary to maintain the iron reduction process. Polysulfides were not detected in the liquid phase. Minor amounts were found associated either with microorganisms or the mineral phase. The abiotic oxidation of sulfide in the reaction with ferrihydrite was identified as rate determining. Cysteine, added as a sulfur source and a reducing agent, also led to abiotic ferrihydrite reduction and therefore should be eliminated when sulfur redox reactions are investigated. Overall, we could demonstrate the large impact of intermediate sulfur species on biogeochemical iron transformations.

Abstract:Sulfide is highly toxic, corrosive, malodorous and may cause many other hazards; therefore, it is necessary to take effective measures to control its emission to the environment. Many physicochemical processes have been devised to separate sulfur from wastewaters. Compared with those traditional methods, biological sulfide removal processes not only have high removal rates, but also generate elemental sulfur, thus recovering mineral resources from wastes. In the paper, the generation methods of elemental sulfur in biological sulfide removal process are reviewed and the characteristics of biologically produced sulfur are described, accordingly the recovery methods of elemental sulfur from wastewater are discussed.
Elemental sulfur-driven sulfidogenic process under highly acidic conditions for sulfate-rich acid mine drainage treatment: Performance and microbial community analysis

Abstract:Elemental sulfur-driven sulfidogenic process has been demonstrated to be more economical and energy-efficient than sulfate-driven sulfidogenic process when treating metal-laden wastewater. In previous studies, we observed that the polysulfide-involved indirect sulfur reduction ensured the superiority of sulfur over sulfate as the electron acceptor in the sulfidogenic process under neutral or weak-alkaline conditions. However, realizing high-rate sulfur reduction process for acid mine drainage (AMD) treatment without pH amelioration is still a great challenge because polysulfide cannot exist under acidic conditions. In this study, a laboratory-scale sulfur-packed bed reactor was therefore continuously operated with a constant sulfate concentration (~1300 mg S/L) and decreasing pH from 7.3 to 2.1. After 400 days of operation, a stable sulfide production rate (38.2 ± 7.6 mg S/L) was achieved under highly acidic conditions (pH 2.6-3.5), which is significantly higher than those reported in sulfate reduction under similar conditions. In the presence of high sulfate content, elemental sulfur reduction could dominate over sulfate reduction under neutral and acidic conditions, especially when the pH ≥ 6.5 or ≤ 3.5. The decreasing pH significantly reduced the diversity of microbial community, but did not substantially influence the abundance of functional genes associated with organic and sulfur metabolisms. The predominant sulfur-reducing genera shifted from Desulfomicrobium under neutral conditions to Desulfurella under highly acidic conditions. The high-rate sulfur reduction under acidic conditions could be attributed to the combined results of high abundance of Desulfurella and low abundance of sulfate-reducing bacteria (SRB). Accordingly, sulfur reduction process can be developed to achieve efficient and economical treatment of AMD under highly acidic conditions (pH ≤ 3.5).

Abstract:Sulfur as a macroelement plays an important role in biochemistry in both natural environments and engineering biosystems, which can be further linked to other important element cycles, e.g. carbon, nitrogen and iron. Consequently, the sulfur cycling primarily mediated by sulfur compounds oxidizing microorganisms and sulfur compounds reducing microorganisms has enormous environmental implications, particularly in wastewater treatment and pollution bioremediation. In this review, to connect the knowledge in microbial sulfur metabolism to environmental applications, we first comprehensively review recent advances in understanding microbial sulfur metabolisms at molecular-, cellular- and ecosystem-levels, together with their energetics. We then discuss the environmental implications to fight against soil and water pollution, with four foci: (1) acid mine drainage, (2) water blackening and odorization in urban rivers, (3) SANI® and DS-EBPR processes for sewage treatment, and (4) bioremediation of persistent organic pollutants. In addition, major challenges and further developments toward elucidation of microbial sulfur metabolisms and their environmental applications are identified and discussed.

Abstract:Key PointsSulphate reducers have the unique ability to respire using sulphate as an electron acceptor. Sulphate-reducing bacteria and archaea are ubiquitous in the environment. The sulphate reducers that have been isolated and described thus far can be divided into seven phylogenetic lineages, five within the Bacteria and two within the Archaea. Most sulphate reducers belong to approximately 23 genera, with the Gram-negative bacteria belonging to the Deltaproteobacteria and the Gram-positive bacteria within the Clostridia.Sulphate reducers can thrive in a broad range of environmental conditions. They have been detected in shallow marine and freshwater sediments and in deep subsurface environments, such as oil wells, hydrothermal vents and mud volcanoes. They occur in environments with extremely low or high pH, extremely low or high temperature and high or low salt concentrations. They are also present in living organisms, such as ruminants and in the human intestinal tract. In marine worms, they form an intimate relationship with aerobic sulphide-oxidizing bacteria.Sulphate reducers play a key part in the carbon and sulphur cycles. They are extremely versatile with respect to the electron donors and electron acceptors that are used for growth. They can grow in a sulphate-dependent manner using hydrogen and a wide range of organic compounds. However, polymeric compounds (polysaccharides and proteins) are not typically used by sulphate reducers. Although named after their ability to respire using sulphate, other inorganic compounds can also be used as electron acceptors. Some sulphate reducers can even respire with oxygen.In environmental biotechnology, sulphate reducers have an important role. One unwanted effect is the production of hydrogen sulphide in anaerobic digesters that are used for waste and waste-water treatment and its role in the corrosion of iron and steel. However, sulphate reducers are also beneficially used in biotechnological processes to remove heavy metals and oxidized sulphur compounds from gas and water. In nature, sulphate reducers live in close vicinity with other microorganisms, which results in metabolic interactions with other anaerobes. In the presence of sulphate, they compete with methanogens and acetogens for common substrates, such as hydrogen and acetate. In the absence of sulphate, however, sulphate reducers grow acetogenically in syntrophy with methanogens. The complete genomes of different sulphate reducers have been, or are currently being, sequenced. pH-dependent biological sulfidogenic processes for metal-laden wastewater treatment: Sulfate reduction or sulfur reduction?

Abstract:Both biological sulfate reduction process and sulfur reduction process are attractive technologies for metal-laden wastewater treatment. However, the acidity stress of metal-laden wastewater could affect the sulfidogenic performance and the microbial community, weaken the stability, efficiency and cost-effectiveness of the biological sulfidogenic processes (BSP). In this study, long-term lab-scale trials were conducted with a sulfate-reducing bioreactor and a sulfate-reducing bioreactor to evaluate the effects of acidity on sulfidogenic activities and the microbial community of the BSP. In the 300-day trial, the sulfate-reducing bacteria (SRB)-driven BSP was stable in terms of sulfidogenic performance and microbial community with the decline of pH, while the sulfate-reducing bacteria (S0RB)-driven BSP achieved high-rate and low-cost sulfide production under neutral conditions but unstable under acidic conditions. With the decline of pH, the sulfide production rate (SPR) of the SRB-driven BSP stably increased from 30 to 83 mg S/L-h; while it decreased from 56 to 37 mg S/L-h in the S0RB-driven BSP with high fluctuation. The results of estimation were consistent with the thermodynamical calculations, in which the sulfur reduction process showed a better performance at pH 5–7, while the sulfate reduction process might gain more energy when pH<5. The stable sulfidogenic performance and microbial community diversity of the SRB-driven BSP could be attributed to the alkalinity produced in sulfate reduction to buffer the acidic stress. In comparison, the microbial community in the S0RB-driven BSP was significantly re-shaped by acidity stress, and the predominant sulfidogenic bacterium changed from Desulfovibrio at neutral condition, to Desulfurella at pH≤5.4. The stability of the microbial community significantly affected the SPR and the operational cost. Nevertheless, the organic consumption for sulfide production of the S0RB-driven BSP was still less than the SRB-driven BSP even in acidic conditions. Collectively, the S0RB-driven BSP was recommended under neutral or mild acid conditions, while the SRB-driven BSP was more suitable under fluctuating pH conditions, especially at low pH. Overall, this study presented the long-term performance of SRB- and S0RB-driven BSP under varying pH conditions, and provided guidance to determine the suitable BSP and operational cost for different metal-laden wastewater.

Abstract:Biological sulfate reduction is increasingly replacing chemical unit processes in mining biotechnology. Sulfate reducing bacteria (SRB) can be used for treating ground‐ and surface waters contaminated with acid mine drainage (AMD), and for recovering metals from wastewater and process streams. Biologically produced H2S precipitates metals as metal sulfides, while biogenic bicarbonate alkalinity neutralizes acidic waters. This paper reviews various passive and active SRB‐based alternatives as well as some process design aspects, such as reactor types, process configurations, and choices of substrates for sulfate reduction. The latest developments of using various low‐cost substrates together with new bioprocess designs are increasing the uses and applications of SRB‐based bioreactors in AMD control and selective metal recovery. Bioprocesses based on sulfate reduction provide multiple options for the treatment of acid mine drainage as well as the recovery of metals from process waters. Various passive and active bioprocess alternatives are reviewed and recent design aspects are discussed.

Abstract:Antimony (Sb), a toxic metalloid, has serious negative effects on human health and its pollution has become a global environmental problem. Bio-reduction of Sb(V) is an effective Sb-removal approach. This work, for the first time, demonstrates the feasibility of autotrophic Sb(V) bio-reduction and removal coupled to anaerobic oxidation of elemental sulfur (S0). In the S0-based biological system, Sb(V) was reduced to Sb(III) via autotrophic bacteria by using S0 as electron donor. Meanwhile, S0 disproportionation reaction occurred under anaerobic condition, generating sulfide and SO42- in the bio-systems. Subsequently, Sb(III) reacted with sulfide and formed Sb(III)-S precipitate, achieving an effective total Sb removal. The precipitate was identified as Sb2S3 by SEM-EDS, XPS, XRD and Raman spectrum analyses. In addition, it was found that co-existing nitrate inhibited the Sb removal, as nitrate is the favored electron acceptor over Sb(V). In contrast, the bio-reduction of co-existing SO42- enhanced sulfide generation, followed by promoting Sb(V) reduction and precipitation. Illumina high-throughput sequencing analysis revealed that Metallibacterium, Citrobacter and Thiobacillus might be responsible for Sb(V) reduction and S0 disproportionation. This study provides a promising approach for the remediation of Sb(V)-contaminated water.

Abstract:The dissociation dynamics of the Ã-state of ammonia have been studied using a resonant multiphoton ionisation probe in a photoelectron spectroscopy experiment. The use of a resonant intermediate in the multiphoton ionisation process changes the ionisation propensity, allowing access to different ion states when compared with equivalent single photon ionisation experiments. Ionisation through the E' 1A1' Rydberg intermediate means we maintain overlap with the ion state for an extended period, allowing us to monitor the excited state population for several hundred femtoseconds. The vibrational states in the photoelectron spectrum show two distinct timescales, 200 fs and 320 fs, that we assign to the non-adiabatic and adiabatic dissociation processes respectively. The different timescales derive from differences in the wavepacket trajectories for the two dissociation pathways that resonantly excite different vibrational states in the intermediate Rydberg state. The timescales are similar to those obtained from time resolved ion kinetic energy release measurements, suggesting we can measure the different trajectories taken out to the region of conical intersection.
