Abstract:We investigated the antiproliferative effect of genistein, and its antileukemia effect in combination with cytosine arabinoside (ara-C) in acute myeloid leukemia (AML). Optimal dosage of genistein as single agent and in combination with ara-C was first determined in vitro. Genistein demonstrated a dose- and time-dependent inhibition of cell proliferation, induction of apoptosis, and cell-cycle arrest at G(2)/M phase. Gene-expression profiles revealed mitogen-activated protein kinase (MAPK) signaling as one of the most affected biological pathways. Phosphatidylinositol 3 kinase, protein kinase A, protein kinase C, MAPK kinase 4, KIT, PIM1, and transforming growth factor-beta receptor 1, were significantly downregulated by genistein. To test whether genistein could augment the antiproliferation activity of ara-C, two groups of severe combined immunodeficient mice were inoculated with NB4 and HL-60 cells, respectively, followed by treatment with either genistein or combination of genistein and ara-C. The combination treatment significantly inhibited tumor growth, and improved survival of NB4 (p = 0.0031) and HL-60 (p = 0.0007) xenograft mice. Our present study highlighted the schedule-dependent synergistic antileukemia effect of genistein with chemotherapy in both in vitro and in vivo models. This novel combination could potentially be a promising regimen for treatment of AML.

Abstract:Combination therapy, a treatment modality that studies the effect of two or more drugs is the most common strategy used to overcome drug resistance in cancer. Cannabinoids have been proven to be potential anticancer agents. Terpenes have also been demonstrated to exhibit anticancer activity. The purpose of this study was to evaluate the effect of combinations of various cannabinoids and terpenes against drug resistant lung and breast cancer. Several cannabinoids and terpenes such as cannabidiol (CBD), cannabichromene (CBC), cannabidiolic acid (CBDA), cannabidivarin (CBDV), cannabinol (CBN), cannabigerol (CBG) and β-caryophyllene (BC), bisabolol, myrcene, linalool, limonene, geraniol, neralidol, α-pinene, α-terpineol, γ-terpinene respectively were screened for their anticancer activity against MDA MB 231 DOX RT, H1975 OSM RT, PDX Lung cancer cell lines using 2D cytotoxicity and 3D spheroids assay. It was found that the cytotoxic potential of combinations of cannabinoids with BC increased by 10 folds in comparison to their individual anticancer activity. The combinations that exhibit synergistic action were determined from combination index (CI) values using compusyn software. Other assays such as colony formation, wound healing, apoptotic assay, cell cycle analysis by flow cytometry elucidated the possible mechanism underlying the synergism of the combinations investigated. CBC+BC was proven to be the most potent combination from all the in-vitro studies and hence was further evaluated in in-vivo tumor xenografts. Briefly, 5 million MDA MB 231 DOX RT cells were injected by S.C route in the right flank region of BALB/c nude mice. The animals were treated after the tumor volume reached 180-200mm3. The animals were administered CBC (15mg/kg, i.p.), BC (100mg/kg, i.p.) and CBC+BC thrice a week for two weeks. The tumor volume in the combination group was reduced by 2 folds relative to individual treatments and 4 folds relative to control. Western blot analysis of tumor xenografts showed downregulation of apoptotic markers such as PARP, mTOR, pAMPK, Survivin, autophagy markers such as LC 3, Glypican 5 and migration markers such as vimentin and Integrin. Immune check point proteins such as PD-L1 and PD-1 were also downregulated by combination treatment. This was further validated by qPCR. In conclusion, CBC induces programmed cell death in tumors and induces autophagy whereas BC inhibits migration and thus this combination was able to overcome resistance than individual therapies in a synergistic fashion. Abstract:Background: Despite modern developments in its management, still major concerns remain about drug resistance in chemotherapy. Natural adjuvants combined with chemotherapy might be the answer. We examined the anti-cancer, anti-proliferative and synergistic effects of Sambucus nigra extract with cisplatin chemotherapy (CDDP) on MCF-7 and MDA-MB-231 human cancer cell lines. Methods: MCF-7 and MDA-MB-231 cell lines were cultured in DMEM culture media, containing 10% FBS and 100 U/ml penicillin/streptomycin. The anti-proliferative activity of SNA, CDDP and their synergic doses were determined using MTT method. Next, the apoptotic, metabolic, and cellular resistance gene expressions were measured through real-time quantitative PCR technique. To show the apoptosis effects and to diagnose cellular damages, an annexin V/propidium iodide (AV/PI) kit and malondialdehyde level were performed, respectively. Results: The synergic doses of SNA and CDDP in MCF-7 were 1.25μM CDDP+1.25μM SNA and on MDA-MB-231 was 2.5μM CDDP+2.5μM SNA. The results of real-time PCR showed that SNA induced apoptosis, disrupted metabolic pathways and reduced cellular drug resistance. In addition, the combination of SNA with CDDP compared with CDDP alone was able to change the expression of these genes and increase the rate of MDA and apoptosis generation (P<0.05). Conclusion: The outcomes of this investigation indicate that SNA, as a herbal supplement, may be a candidate for increasing the effect of CDDP therapy in the treatment of breast cancers. This synergy was not estrogen-dependent in the MDA-MB-231 cells, promoted apoptosis, cell damages, disorders of metabolism, and reduced the drug resistance in the cancer cells.

Abstract:Nutrition can modulate host immune responses as well as promote anticancer effects. In this study, two nutritional supplements, namely gamma-tocotrienol (γT3) and Spirulina, were evaluated for their immune-enhancing and anticancer effects in a syngeneic mouse model of breast cancer (BC). Five-week-old female BALB/c mice were fed Spirulina, γT3, or a combination of Spirulina and γT3 (Spirulina + γT3) for 56 days. The mice were inoculated with 4T1 cells into their mammary fat pad on day 28 to induce BC. The animals were culled on day 56 for various analyses. A significant reduction (p < 0.05) in tumor volume was only observed on day 37 and 49 in animals fed with the combination of γT3 + Spirulina. There was a marked increase (p < 0.05) of CD4/CD127+ T-cells and decrease (p < 0.05) of T-regulatory cells in peripheral blood from mice fed with either γT3 or Spirulina. The breast tissue of the combined group showed abundant areas of necrosis, but did not prevent metastasis to the liver. Although there was a significant increase (p 0.05) differences in the expression of MIG-6, Cadherin 13, BIRC5, and Serpine1 upon combined feeding. This showed that combined γT3 + Spirulina treatment did not show any synergistic anticancer effects in this study model.

Abstract:Immune checkpoint inhibitors (ICPi) targeting the PD-1/PD-L1 pathway have shown marked success in patients with advanced melanoma. However, 60–70% of patients fail to respond, warranting a therapeutic intervention that could increase response rates. We and others have shown that S-adenosylmethionine (SAM), a universal methyl donor, has significant anticancer effects in numerous cancers previously; however, its effect on melanoma progression has not been evaluated. Interestingly, SAM was reported to be essential for T cell activation and proliferation and, thus, could potentially cooperate with ICPi and block melanoma progression. In this study, we examined the antitumor effects of SAM and ICPi alone and in combination in a well-established melanoma mouse model wherein syngeneic C57BL/6 mouse were subcutaneously (orthotopic) injected with B16-F1 cells. Treatment of mice with either SAM or anti-PD-1 antibody alone resulted in significant reduction in tumor volumes and weights; effects that were highest in mice treated with a combination of SAM+anti-PD-1. RNA-sequencing analysis of the primary tumors showed numerous differentially expressed genes (DEGs) following treatment with SAM+anti-PD-1, which was shown to downregulate cancer, MAPK, and tyrosine kinase pathways. Indeed, SAM+anti-PD-1 reversed the aberrant expression of some known melanoma genes. Tumor immunophenotyping revealed the SAM+anti-PD-1 combination was significantly more effective than either SAM or anti-PD-1 as the CD8+ T cells had higher activation, proliferation, and cytokine production compared to all other groups. This study shows that the combination of currently approved agents SAM and ICPi can effectively block melanoma via alteration of key genes/pathways implicated in cancer and immune response pathways, providing the rationale for the initiation of clinical trials with SAM and ICPi.

Abstract:Traditional chemotherapy and radiotherapy for cancer treatment face serious challenges such as drug resistance and toxic side effects. Complementary / Alternative medicine is increasingly being practiced worldwide due to its safety beneficial therapeutic effects. We hypothesized that a super combination (SC) of known phytochemicals used at bioavailable levels could induce 100% killing of breast cancer (BC) cells without toxic effects on normal cells and that microarray analysis would identify potential genes for targeted therapy of BC. Mesenchymal Stems cells (MSC, control) and two BC cell lines were treated with six well established pro-apoptotic phytochemicals individually and in combination (super cocktail), at bioavailable levels. The compounds were ineffective individually. In combination, they significantly suppressed BC cell proliferation (>80%), inhibited migration and invasion, caused cell cycle arrest and induced apoptosis resulting in 100% cell death. However, there were no deleterious effects on MSC cells used as control. Furthermore, the SC down-regulated the expression of PCNA, Rb, CDK4, BcL-2, SVV, and CD44 (metastasis inducing stem cell factor) in the BC cell lines. Microarray analysis revealed several differentially expressed key genes (PCNA, Rb, CDK4, Bcl-2, SVV, P53 and CD44) underpinning SC-promoted BC cell death and motility. Four unique genes were highly up-regulated (ARC, GADD45B, MYLIP and CDKN1C). This investigation indicates the potential for development of a highly effective phytochemical combination for breast cancer chemoprevention / chemotherapy. The novel over-expressed genes hold the potential for development as markers to follow efficacy of therapy.

Abstract:Dietary phytochemicals are currently being studied with great interest due to their ability to regulate the epigenome resulting in prevention of cancer. Some natural botanicals have been reported to have enhanced and synergistic impact on cancer suppression when administered at optimum concentrations and in-conjunction. Sulforaphane (SFN) is an isothiocyanate found in cruciferous vegetables and sodium butyrate (NaB) is a short-chain fatty acid produced by gut microbiota. They have been intensively explored due to numerous anti-cancerous properties and ability to modulate epigenetic machinery by inhibition of histone deacetylase (HDAC). Genistein (GE), present in soy, is a known DNA methyltransferase (DNMT) inhibitor. While combined chemoprotective epigenetic effects induced by SFN and GE have been investigated, the key impact of combinatorial SFN-NaB, GE-NaB, and SFN-GE-NaB bioactive components in regulation of various mechanisms are poorly defined. In the present study, we found that combinations of dietary compounds had synergistic effects in decreasing cellular viability at lower dosages than their single dosages in breast cancer cell lines. The respective combinations limited growth and increased apoptosis and necrosis in cancerous cells among which the tri-combination displayed the most significant impact. Additionally, the respective combinations of compounds arrested MDA-MB-231 and MCF-7 breast cancer cells at G2/M phase. Our further mechanistic evaluation revealed that respective di-combinations and tri-combination had higher impact in down-regulation of DNMTs (DNMT3A and DNMT3B), HDACs (HDAC1, HDAC6 and HDAC11), histone methyltransferases (EZH2 and SUV39H1) and histone acetyltransferases (GCN5, PCAF, P300 and CBP) levels as compared to singly administered compounds. We also found that these combinations exhibited global epigenetic changes by inhibition of DNMT and HDAC activity, histone H3 at lysine 27 methylation (H3K27me) and histone H3 at lysine 9 methylation (H3K9me) levels, and by induction of histone acetyltransferases activity. Collectively, our investigation indicates that combined SFN, GE and NaB is highly effective in inhibiting breast cancer genesis by, at least in part, regulating epigenetic modifications, which may have implications in breast cancer therapy.

Anti-Cancer Agents in Medicinal Chemistry

Abstract:Background: Due to its systemic toxicity, traditional chemotherapy of tumors is being taken into consideration. Herbal therapy, containing phytochemical polyphenol derivatives such as Curcumin (Cur), Ginger (Gin), Cloves (Clov) and Amygdaline (Amyg), is one of the numerous complementary and alternative approaches as an anti-cancer therapy and holds great promise for cancer chemo-prevention with fewer side effects. Aim: The current study was designated to assess anti-tumoral immunity and anti-cancer and chemo-preventive effectiveness of herbal extracts of Cur, Ginger, Clov and Amyg in Ehrlich Ascites Carcinoma (EAC)-challenging mice. Methods: Chemo-preventive efficacy of herbal extracts of Cur, Gin, Clov and Amyg were analyzed in vivo by examination of the apoptosis rate of EAC tumor cells by flow cytometry. The total numbers of EAC cells, splenocytes counts and leucocytes count with their differentials relative % in peripheral blood (PB) of EACchallenging mice were investigated. Results: EAC-challenging mice treated with herbal extracts of Cur, Gin, Clov and Amyg showed a marked decline in EAC tumor cell count and a noticeable increase in apoptosis rate of EAC tumor cells, a remarkable decrease in serum level of cancer antigen 125 (CA-125) with an obvious increase in the number of splenocytes comparing to that in EAC-challenging mice treated with PBS alone. Moreover, the data indicated an insignificant change in the total leucocytes count and their differentials relative % of eosinophil, neutrophils, monocytes and lymphocytes in EAC-challenging mice treated with Cur and Amyg, but these parameters were markedly increased in EAC-challenging mice injected with Gin and Clov compared to that in EAC-challenging mice treated with PBS alone. Conclusion: To conclude, the herbal extracts of Cur, Gin, Clov and Amyg may have anti-tumoral immunity and anti-cancer potency and potential to reduce the resistance to cancer conventional chemotherapy and exert cancer chemo-protective approaches with low adverse effects. Further research is necessary to determine the regimen's toxicity on various tissues and organs and to connect the diagnostic and therapeutic approaches used in the regimen's biomedical use.

Abstract:Despite the significant advances in screening methods for early diagnosis, breast cancer remains a global threat and continues to be the leading cancer diagnosed in women, requiring effective therapy. Currently, combination therapy has become the hallmark of breast cancer treatment due to the high incidence of tumor recurrence and disease progression after monotherapeutic treatments, including surgery, radiotherapy, endocrine therapy, and chemotherapy. Over the past decades, there has been considerable interest in studying the anticancer effect of bioactive phytochemicals from medicinal plants combined with these conventional therapies. The rationale for this type of therapy is to use combinations of drugs that work by different mechanisms, thereby decreasing the likelihood that cancer cells will develop resistance, and also reduce the therapeutic dose and toxicity of single treatments. Three agents have received great attention with regard to their anticancer properties: 1) piperine, a dietary phytochemical isolated from black pepper (Piper nigrum L.) and long pepper (Piper longum L.); 2) sulforaphane, an isothiocyanate mainly derived from cruciferous vegetables; and 3) thymoquinone, the active compound from black seed (Nigella sativa L.). This review focused on the combined effect of these 3 compounds on conventional cancer therapy with the objective of observing enhanced efficacy compared with single treatments. This review also highlights the importance of the nanoformulation of such bioactive phytochemicals that could enhance their bioavailability by providing an efficient targeted delivery system with a reduced systemic dose while resulting in a more efficient dosing at the target site.

Abstract:Systematically identifying synergistic combinations of targeted agents and immunotherapies for cancer treatments remains difficult. In this study, we integrated high-throughput and high-content techniques—an implantable microdevice to administer multiple drugs into different sites in tumors at nanodoses and multiplexed imaging of tumor microenvironmental states—to investigate the tumor cell and immunological response signatures to different treatment regimens. Using a mouse model of breast cancer, we identified effective combinations from among numerous agents within days. In vivo studies in three immunocompetent mammary carcinoma models demonstrated that the predicted combinations synergistically increased therapeutic efficacy. We identified at least five promising treatment strategies, of which the panobinostat, venetoclax and anti-CD40 triple therapy was the most effective in inducing complete tumor remission across models. Successful drug combinations increased spatial association of cancer stem cells with dendritic cells during immunogenic cell death, suggesting this as an important mechanism of action in long-term breast cancer control.

Abstract:Dietary compounds that possess the properties of altering epigenetic processes are gaining popularity as targets for cancer prevention studies. These compounds when administered at optimal concentrations and especially in combination can have enhanced effects in cancer prevention or therapy. It is important to study the interaction of two or more compounds in order to assess their role in enhancing prevention. Genistein (GEN), found in soy, has been extensively studied for its role as an epigenetic modifier especially as a DNA methyltransferase (DNMT) inhibitor and sulforaphane (SFN), found in cruciferous vegetables, is known as a histone deacetylase (HDAC) inhibitor. However, very little is known about the effects of these two compounds in conjunction in breast cancer prevention or therapy. In our current study, we determined that, at certain doses, the compounds have synergistic effects in decreasing cellular viability of breast cancer cell lines. Our results indicate that the combination of GEN and SFN is much more effective than their single doses in increasing the rate of apoptosis and lowering the colony forming potential of these cells. We determined that these compounds inhibit cell cycle progression to G2 phase in MDA-MB-231 and G1 phase in MCF-7 breast cancer cell lines. Additionally, we determined that the combination is effective as an HDAC and histone methyltransferase (HMT) inhibitor. Furthermore, we demonstrated that this combination downregulates the levels of HDAC2 and HDAC3 both at the mRNA and protein levels. We also found that these compounds have the potential to downregulate KLF4 levels, which plays an important role in stem cell formation. The combination of GEN and SFN is also effective in downregulating hTERT levels, which is known to be activated when KLF4 binds to its promoter region. Our hypothesis is further strengthened by in vivo studies, where the combination is administered to transgenic mice in the form of genistein and SFN-enriched broccoli sprouts. We have demonstrated that the combination is more effective in preventing or treating mammary cancer via extending tumor latency and reducing tumor volumes/sizes than either of these dietary components administered alone. These results are consistent with our in vitro study suggesting potential preventive and therapeutic effects of this novel dietary combinatorial approach against breast cancer.

Abstract:Estrogen receptor-negative [ER(−)] mammary cancer is the most aggressive type of breast cancer (BC) with higher rate of metastasis and recurrence. In recent years, dietary prevention of BC with epigenetically active phytochemicals has received increased attention due to its feasibility, effectiveness, and ease of implementation. In this regard, combinatorial phytochemical intervention enables more efficacious BC inhibition by simultaneously targeting multiple tumorigenic pathways. We, therefore, focused on investigation of the effect of sulforaphane (SFN)-rich broccoli sprouts (BSp) and withaferin A (WA)-rich Ashwagandha (Ash) combination on BC prevention in estrogen receptor-negative [ER(−)] mammary cancer using transgenic mice. Our results indicated that combinatorial BSp + Ash treatment significantly reduced tumor incidence and tumor growth (~ 75%) as well as delayed (~ 21%) tumor latency when compared to the control treatment and combinatorial BSp + Ash treatment was statistically more effective in suppressing BC compared to single BSp or Ash intervention. At the molecular level, the BSp and Ash combination upregulated tumor suppressors (p53, p57) along with apoptosis associated proteins (BAX, PUMA) and BAX:BCL-2 ratio. Furthermore, our result indicated an expressional decline of epigenetic machinery HDAC1 and DNMT3A in mammary tumor tissue because of combinatorial treatment. Interestingly, we have reported multiple synergistic interactions between BSp and Ash that have impacted both tumor phenotype and molecular expression due to combinatorial BSp and Ash treatment. Our RNA-seq analysis results also demonstrated a transcriptome-wide expressional reshuffling of genes associated with multiple cell-signaling pathways, transcription factor activity and epigenetic regulations due to combined BSp and Ash administration. In addition, we discovered an alteration of gut microbial composition change because of combinatorial treatment. Overall, combinatorial BSp and Ash supplementation can prevent ER(−) BC through enhanced tumor suppression, apoptosis induction and transcriptome-wide reshuffling of gene expression possibly influencing multiple cell signaling pathways, epigenetic regulation and reshaping gut microbiota.

Abstract:Cancer patients face increased susceptibility to invasive infections, primarily due to ulcerative lesions on mucosal surfaces and immune suppression resulting from chemotherapy. Pseudomonas aeruginosa (P. aeruginosa) bacteremia is notorious for its rapid progression into fatal sepsis, posing a significant threat to cancer patients, particularly those experiencing chemotherapy-induced neutropenia. This bacterial infection contributes significantly to morbidity and mortality rates among such individuals. Our latest report showed the mutually beneficial effects of postbiotic butyrate on 1,25-dihydroxyvitamin D3 (1,25D3)-controlled innate immunity during Salmonella colitis. Hence, we investigated the impact of butyrate and 1,25D3 on chemotherapy-induced gut-derived P. aeruginosa sepsis in mice. The chemotherapy-induced gut-derived P. aeruginosa sepsis model was established through oral administration of 1 × 107 CFU of the P. aeruginosa wild-type strain PAO1 in C57BL/6 mice undergoing chemotherapy. Throughout the infection process, mice were orally administered butyrate and/or 1,25D3. Our observations revealed that the combined action of butyrate and 1,25D3 led to a reduction in the severity of colitis and the invasion of P. aeruginosa into the liver and spleen of the mice. This reduction was attributed to an enhancement in the expression of defensive cytokines and antimicrobial peptides within the cecum, coupled with decreased levels of zonulin and claudin-2 proteins in the mucosal lining. These effects were notably more pronounced when compared to treatments administered individually. This study unveils a promising alternative therapy that involves combining postbiotics and 1,25D3 for treating chemotherapy-induced gut-derived P. aeruginosa sepsis.

Synergistic anticancer effects and reduced genotoxicity of silver nanoparticles and tamoxifen in breast cancer cells

Abstract:Nanotechnology is emerging as a promising tool to enhance traditional cancer treatments due to rising chemotherapy resistance and the severe side effects of toxic drugs. Silver nanoparticles (AgNPs) are widely acknowledged for their antimicrobial and antiproliferative properties. Given these AgNP characteristics, this research conducts a comprehensive nanotoxicological assessment of strategic combinations involving AgNPs (68 nm) commercial formulation and tamoxifen on MCF-7 and MDA-MB-231 breast tumor cells. Utilizing CompuSyn software, the combination index was determined, revealing a synergistic cytotoxic and antiproliferative effect in AgNPs and tamoxifen combinations (CI < 0.97). Furthermore, this combination impaired cell migration (the scratch zone expanded by over 270%) and significantly increased reactive oxygen species production (up to 96% for MDA-MB-231 and 52% for MCF-7 cells). Surprisingly, the genotoxic effect of these mixtures was minimal (below the allowable genotoxicity index of 1.5). Additionally, both breast tumor cell lines exhibited increased proapoptotic and oxidative stress gene expression following the combined treatment. The internalization of AgNPs into breast cancer cells was observed, enhancing their synergistic antiproliferative effect when combined with tamoxifen. These findings suggest the potential of combining AgNPs with chemotherapeutic agents for innovative studies in oncology therapy.

Especially, rheumatoid nodules as bilateral cystic rounded masses in the submucosal space at the junction of the anterior and medial thirds of each vocal fold are described in the literature as a cause of hoarseness [Murano E et al., J Voice 2012; 15: 441–450]. A diff erent aspect of bamboo nodes is their bamboojoint-like appearance as described above, although the histological fi ndings are often consistent with rheumatoid nodules [Speyer R et al., J Voice 2008; 22: 232– 237]. In most cases with known bamboo nodes that are described in the literature, a rheumatoid arthritis was diagnosed, but the lesions have also been described in patients with systemic lupus erythematosus, Hashimoto’s thyroiditis and pro gressive systemic sclerosis [Schwemmle et al. Rheumatol Int 2011; 16, Epub ahead of print]. Diff erent strategies of therapies are discussed in the literature: Conservative : Murano believes that bamboo nodes are a sign of the activity of the autoimmune disorder. Thus, he and others suggest that clinical treatment with steroids (systemic) should be tried fi rst [Murano E et al., J Voice 2001; 15: 441– 450]. In general, the aim of conservative therapy of rheumatoid nodules with local injection of prednisolone in the lesions is a reduction of mass and infl ammatory processes. Surgery: Perouse reported 19 cases treated by surgery with a favourable outcome [Perouse R et al., Rev Laryngol Otol Rhinol 2001; 122: 299–302]. The recommended extent of surgery was not described. To Introduction ▼ Vocal fold lesions related to autoimmune diseases are rheumatoid nodules, and, less frequently, bamboo nodes. The latter may also occur in the absence of systemic rheumatic disease. Mostly transverse, they are located in the middle third of the vocal cord and may be located unior bilateral. Raven et al. [Raven RW et al., Ann Rheum Dis 1948; 7: 63–75] were the fi rst to describe small submucous rheumatoid nodules in the larynx and epiglottis. In 1993, Hosako described a case of systemic lupus erythematosus (SLE) in which the submucosal lesions in the vocal folds did not resemble the rounded nodules. Instead, they had a stripe-like appearance perpendicular to the free vocal cord edge: “bamboo-joint like” [Hosako Y et al., Larynx Japan 1993; 6: 171–175]. Murano used the term “vocal fold bamboo nodes” to simplify the nomenclature of the same feature [Murano E et al., J Voice 2001; 15: 441–450]. Here, we present symptoms and clinical fi ndings in a girl with bamboo nodes. In the literature, bamboo nodes are only reported in adults. Here we fi rst describe a 13-year-old girl with bamboo nodes. To our knowledge, this is the youngest patient with a hoarseness and bamboo nodes ever reported; an associated autoimmune disorder has not been found so far.
Does the availability of either B cells or CD4+ cells limit germinal centre formation?

Abstract:To determine whether the availability of either B cells or T cells regulates the number and size of germinal centres observed following antigenic challenge, irradiated PVG rats were reconstituted with limiting numbers of thoracic duct B cells (with excess T cells) or with limiting numbers of thoracic duct CD4+ cells (with sufficient B cells). Germinal centre formation was measured histologically in the spleens of reconstituted rats 7 days after immunization with 2 x 10(9) sheep erythrocytes (at the height of the germinal centre reaction). Although reconstitution with B cells and antigen alone, or with thymocytes and antigen alone, produced no germinal centres, saturating numbers of germinal centres on the order observed for normal, non-irradiated rats were observed in irradiated rats reconstituted with only 10(7) B cells and 5 x 10(6) CD4+ cells. Germinal centres in the spleens of minimally reconstituted rats were also comparable in size to those observed in normal rats. Reconstitution with either 4 x 10(7) thoracic duct lymphocytes (including 2 x 10(7) B cells, as well as CD8+ and CD4+ cells) or with 4 x 10(7) thoracic duct lymphocytes and 2 x 10(8) thymocytes also led to saturating numbers of germinal centres. It is concluded therefore that (i) CD4+ cells are sufficient for the T-cell contribution required for germinal centre formation, and (ii) some factor other than the availability of B cells or CD4+ cells normally limits germinal centre formation.

Abstract:Introduction/Objective. Large diversity in molecular mechanisms of cancer regulation allows some marketed pleiotropic non-oncological non-toxic pharmaceuticals to be used in oncology, which reduces duration and cost of novel anticancer treatment research. Up to date, there are no published results in vivo on anticancer effects of certain combinations of non-oncological pleiotropic drugs (disulfiram, metformin, deoxycholic acid, mebendazole) that influence MAPK/PI3K/AKT/mTOR/NF-?B signaling. Methods. The anticancer effects of certain aforementioned repurposed drugs combinations, <50 % LD50 (equivalent to usual human dose) were assessed by fibrosarcoma growth kinetics (measured daily in vivo by calipers) and tumor proliferation (Ki-67, PCNA), neoangiogenesis (CD34, CD31), glucose metabolism (GLUT1), NO metabolism (iNOS) and apoptosis (COX4, Cytochrome C) in hamsters, randomly allocated to control and experimental groups (6 animals per group). Animals were sacrificed 19 days after BHK-21/C13 tumor inoculation. Tumors were excised, measured and blood collected. Biophysical, pathohistological, toxicological, hematological and biochemical analyses were performed. Results. Disulfiram with metformin, disulfiram with deoxycholic acid and deoxycholic acid with metformin are the combinations that have shown significant antitumor effects on the fibrosarcoma growth kinetics and tumor immunohistochemical markers in hamsters (p<0.05). All used drugs in efficacious combinations can inhibit MAPK/PI3K/AKT/mTOR/NF-?B signaling. Addition of NF-?B stimulator mebendazole to effective two-drug combinations rescued cancer growth, indicating that these pathways may be responsible for antitumor action. Conclusion. Combinations of non-oncological drugs: disulfiram with metformin, disulfiram with deoxycholic acid and deoxycholic acid with metformin have the potential to be used as effective non-toxic adjuvant anticancer therapy in oncology.

Abstract:Breast cancer has a diverse aetiology characterized by the heterogeneous expression of hormone receptors and signalling molecules, resulting in varied sensitivity to chemotherapy. The adverse side effects of chemotherapy coupled with the development of drug resistance have prompted the exploration of natural products to combat cancer. Lactoferricin B (LfcinB) is a natural peptide derived from bovine lactoferrin that exhibits anticancer properties. LfcinB was evaluated in vitro for its inhibitory effects on cell lines representing different categories of breast cancer and in vivo for its suppressive effects on tumour xenografts in NOD-SCID mice. The different breast cancer cell lines exhibited varied levels of sensitivity to apoptosis induced by LfcinB in the order of SKBR3>MDA-MB-231>MDA-MB-468>MCF7, while the normal breast epithelial cells MCF-10A were not sensitive to LfcinB. The peptide also inhibited the invasion of the MDA-MB-231 and MDA-MB-468 cell lines. In the mouse xenograft model, intratumoural injections of LfcinB significantly reduced tumour growth rate and tumour size, as depicted by live imaging of the mice using in vivo imaging systems (IVIS). Harvested tumour volume and weight were significantly reduced by LfcinB treatment. LfcinB, therefore, is a promising and safe candidate that can be considered for the treatment of breast cancer.
