Abstract:The misuse and abuse of antibiotics have given rise to a severe problem of the drug resistance of bacteria. Solving this problem has been a vitally important task in the modern medical arena. In this work, an antimicrobial peptide (AMP), BF2b, and gold nanorods (AuNRs) were used to develop a specific drug delivery system for killing methicillin-resistant Staphylococcus aureus (MRSA). On the one hand, BF2b has unique anti-bacterial performance and has a lower tendency than traditional antibiotics to engender the drug resistance of bacteria. On the other hand, AuNRs have diverse distinct properties, such as photo-thermal conversion, which can be employed for photo-thermal sterilization. We aimed to integrate the anti-bacterial activity of BF2b and the photo-thermal sterilization of AuNRs to kill drug-resistant bacteria. Fourier-transform infrared spectroscopy, microBCA and zeta potential measurements were utilized to characterize the product, AuNR@PEG/BF2b. Transmittance electron microscopy, UV-vis spectroscopy and photothermal conversion measurement were conducted to verify the stability and photothermal conversion capacity of AuNR@PEG/BF2b. Cell viability and hemolysis assay were carried out to test the biocompatibility of AuNR@PEG/BF2b. Finally, the in vitro and in vivo experiments were performed to demonstrate the excellent bactericidal activity of AuNR@PEG/BF2b.

Abstract:The abuse of antibiotics has led to bacterial drug resistance, which hinders traditional treatments for infection. It is a vitally important task to develop new methods for combating bacterial infection. In this work, we design a multifunctional peptide (BrEK) that consists of three functional building blocks: an antimicrobial sequence (BF2b), a gelatinase B responsive linker, and a zwitterionic antifouling block. The BrEK peptide is conjugated onto gold nanorods (AuNRs) via a cysteine on one end of the peptide sequence, with excellent photothermal conversion ability under near-infrared (NIR) light. The obtained AuNR@BrEK are tested in vitro and in vivo for antibiotic capacity against methicillin-resistant Staphylococcus aureus (MRSA). The multifunctional peptide coating provides the nanocomposite excellent biocompatibility, relative long circulation in blood, infection microenvironment responsive targeting ability, and unique antibacterial performance. Ultimately, we aim to eliminate drug resistance bacteria by (1) integrating infection microenvironment responsiveness for efficient targeting, (2) capitalizing on antibacterial capacity of BF2b peptides, and (3) developing photothermal sterilization. Transmittance electron microscopy, dynamic light scatter, and zeta potential measurements are introduced to characterize the synthesis of AuNR@BrEK. The evaluation of particle stability is conducted by Fouriertransform infrared spectroscopy and UV?vis spectroscopy. Photothermal conversion capacity of AuNR@BrEK is examined via photothermal effect measurement. Cell viability and hemolysis assay are conducted to further assess the biocompatibility of AuNR@BrEK (200 mu g/mL). Lastly, results from the in vitro and in vivo experiments indicate the outstanding antibacterial activity of AuNR@BrEK (10 mg/kg). Such multifunctional gold nanorods represent an important step forward in the goal to combat bacterial resistance.

Methods: In this study, we designed a pH-responsive cationic polymer sodium nitroside dihydrate/branched polyethylenimine–indocyanine green@polyethylene glycol (SNP/PEI-ICG@PEG) nanoplatform using the electrostatic adsorption method and Schiff's base reaction. Relevant testing techniques were applied to characterize and analyze SNP/PEI-ICG@PEG, proving the successful synthesis of the nanomaterials. In vivo and in vitro experiments were performed to evaluate the antimicrobial properties of SNP/PEI-ICG@PEG. Results: The morphology and particle size of SNP/PEI-ICG@PEG were observed via TEM. The zeta potential and UV-visible (UV‐vis) results indicated the synthesis of the nanomaterials. The negligible cytotoxicity of up to 1 mg/mL of SNP/PEI-ICG@PEG in the presence or absence of light demonstrated its biosafety. Systematic in vivo and in vitro antimicrobial assays confirmed that SNP/PEI-ICG@PEG had good water solubility and biosafety and could be activated by near-infrared (NIR) light and synergistically treated using four therapeutic modes, photodynamic therapy (PDT), gaseous therapy (GT), mild photothermal therapy (PTT, 46 °C), and cation. Ultimately, the development of Gram-positive (G + ) Staphylococcus aureus ( S. aureus ) and Gram-negative (G − ) Escherichia coli ( E. coli ) were both completely killed in the free state, and the biofilm that had formed was eliminated. Conclusion: SNP/PEI-ICG@PEG demonstrated remarkable efficacy in achieving controlled multimodal synergistic antibacterial activity and biofilm infection treatment. The severity of infection primarily relies on the virulence of bacteria and may involve biofilm formation. Using a Bacterial Protein to Selectively Target Bacterial Biofilms: Treatment of S. epidermidis Biofilms with Targeted Photothermal Gold Nanoparticles

Abstract:Biofilm-related infections are associated with high mortality and morbidity, combined with increased hospital stays and overall treatment costs. Traditional antibiotics play an essential role in controlling biofilms; however, they are becoming less effective due to the emergence of drug-resistant bacterial strains. The need to treat biofilms on medical implants is particularly acute, and one persistent challenge is selectively directing nanoparticles to the biofilm site. Here, we present a protein-based functionalization strategy that targets the extracellular matrix of biofilms. The protein, derived from the extracellular Staphylococcus epidermidis autolysin, directs nanoparticles to S. epidermidis cell wall components, which are not expected to be present in mammalian tissues. This functionalization is applied to a gold nanoparticle (AuNP) core, along with elastin-like polypeptides (ELPs), which generate a robust photothermal response. In addition to specific biofilm targeting, the particles exhibit low protein binding, and the photothermal conversion can be easily modulated by changing the ELP transition temperature (Tt). These functionalized AuNPs strongly interact with biofilms under static and flow conditions but exhibit relatively weak interactions with serum-coated surfaces. NIR irradiation resulted in a 10,000-fold improvement in killing efficiency compared to untreated controls (p < 0.0001). The novel targeting strategy utilized in this work represents a general and versatile approach to targeting drug-resistant bacterial infections and could be readily expanded to other anti-biofilm nanoparticle platforms.

Abstract:With the ever-growing threat of bacterial infections, especially for multidrug-resistant microbial infections, the development of highly effective treatment modalities to inhibit the infections is challenging. Although silver nanoparticles have been intensively applied as antimicrobial agent for decades, the therapeutic efficacy toward multidrug-resistant bacteria is still unsatisfactory. Here, we show that near-infrared (NIR) laser-excited silver triangular nanoparticles (Tri-Ag) can efficiently kill Gram-negative Escherichia coli and Gram-positive Staphylococcus aureus both in vitro and in vivo. Notably, multidrug-resistant bacterial clinical isolates, including methicillin-resistant S. aureus and extended spectrum β-lactamase E. coli strain were significantly inhibited by the combined treatment of the Tri-Ag with NIR laser irradiation due to their synergistic antibacterial ability. Taking the advantage of its strong near-infrared absorbance, photothermal treatment is also conducted with Tri-Ag, achieving a remarkable synergistic antibacterial effect to inhibit various bacteria at a rather low concentration of this agent. Given the above advantages, the combination therapy of Tri-Ag with assistance of NIR laser may find potential applications to strengthen the antimicrobial arsenal for fighting bacterial infections.

Abstract:The development of non-antibiotic strategies for bacterial disinfection is of great clinical importance. Among recently developed different antimicrobial strategies, nanomaterial-mediated approaches, especially the photothermal way and reactive oxygen species (ROS)-generating method, show many significant advantages. Although promising, the clinical application of nanomaterials is still limited, owing to the potential biosafety issues. Further improvement of the antimicrobial activity to reduce the usage, and thus reduce the potential risk, is an important way to increase the clinical applicability of antibacterial nanomaterials. In this paper, an antimicrobial nanostructure with both an excellent photothermal effect and peroxidase-like activity was constructed to achieve efficient synergistic antimicrobial activity. The obtained nano-antimicrobial agent (ZIF-8@PDA@Pt) can not only efficiently catalyze the production of ROS from H 2 O 2 to cause damage to bacteria but also convert the photon energy of near-infrared light into thermal energy to kill bacteria, and the two synergistic effects induced in a highly efficient antimicrobial activity. This study not only offers a new nanomaterial with efficient antibacterial activity but also proposes a new idea for constructing synergistic antibacterial properties.

Near-Infrared-Controlled Nanoplatform Exploiting Photothermal Promotion of Peroxidase-like and OXD-like Activities for Potent Antibacterial and Anti-biofilm Therapies.

Abstract:Nanozymes that mimic peroxidase (POD) activity can convert H2O2 into bactericidal free radicals, which is referred to as chemodynamic therapy (CDT). High glutathione (GSH) levels in the infectious tissue severely limit the performance of CDT. Herein, we report a near-infrared-controlled antibacterial nanoplatform that is based on encapsulating tungsten sulfide quantum dots (WS2QDs) and the antibiotic vancomycin in a thermal-sensitive liposome. The system exploits the photothermal sensitivity of the WS2QDs to achieve selective liposome rupture for the targeted drug delivery. We determined that WS2QDs show a strong POD-like activity under physiological conditions and the oxidase-like activity, which can oxidate GSH to further improve the CDT efficacy. Moreover, we found that increased temperature promotes multiple enzyme-mimicking activities of WS2QDs. This platform exerts antibacterial effects against Gram-positive Mu50 (a vancomycin-intermediate Staphylococcus aureus reference strain) and Gram-negative Escherichia coli and disrupts biofilms for improved penetration of therapeutic agents inside biofilms. In vivo studies with mice bearing Mu50-caused skin abscess revealed that this platform confers potent antibacterial activity without obvious toxicity. Accordingly, our work illustrates that the photothermal and nanozyme properties of WS2QDs can be deployed alongside a conventional therapeutic to achieve synergistic chemodynamic/photothermal/pharmaco therapy for powerful antibacterial effects.

Effective reduction of biofilm through photothermal therapy by gold core@shell based mesoporous silica nanoparticles

Abstract:Bacterial biofilms can initiate chronic infections that become difficult to eradicate. There is an unmet need for effective therapeutic strategies that control and inhibit the growth of these biofilms. Herein, light sensitive mesoporous silica nanoparticles (MSNs) with photothermal (PTT) and antimicrobial combined capabilities have been developed. These nanosystems have high therapeutic potential to affect the bacterial biofilm architecture and subsequently inhibit its growth. Nucleation of gold nanorods followed by the growth of a silica shell leads to a core@shell design (AuNR@MSN) with PTT properties. Incorporation of nitrosothiol groups (-SNO) with a heat liable linker, enables an enhanced nitric oxide release upon photothermal stimulation with near infrared radiation. Further loading of an antimicrobial molecule such as the levofloxacin (LEVO) antibiotic creates a unique nanoassembly with potential therapeutic efficacy against Staphylococcus aureus bacterial biofilms. A dispersion rate of the bacterial biofilm was evident when light stimuli is applied because impregnation of the nitrosothiol functionalized nanosystem with the antibiotic LEVO led to ca. 30% reduction but its illumination with near infrared (NIR) irradiation showed a biofilm reduction of ca. 90%, indicating that localized antimicrobial exposure and PTT improves the therapeutic efficacy. These findings envision the conception of near-infrared-activated nanoparticle carriers capable of combined therapy upon NIR irradiation, which enables photothermal therapy, together with the release of levofloxacin and nitric oxide to disrupt the integrity of bacterial biofilms and achieve a potent antimicrobial therapy.

Abstract:Chronic infections caused by Pseudomonas aeruginosa pose severe threats to human health. Traditional antibiotic therapy has lost its total supremacy in this battle. Here, nanoplatforms activated by the clinical microenvironment are developed to treat P. aeruginosa infection on the basis of dynamic borate ester bonds. In this design, the nanoplatforms expose targeted groups for bacterial capture after activation by an acidic infection microenvironment, resulting in directional transport delivery of the payload to bacteria. Subsequently, the production of hyperpyrexia and reactive oxygen species enhances antibacterial efficacy without systemic toxicity. Such a formulation with a diameter less than 200 nm can eliminate biofilm up to 75%, downregulate the level of cytokines, and finally promote lung repair. Abstract:The emphasis is on how the atomic‐precision size effect of indocyanine green‐gold nanoclusters impacts reactive oxygen species generation and antibacterial efficacy toward Staphylococcus aureus. Near‐infrared light activates indocyanine green‐gold nanoclusters, disrupts quorum sensing, and reduces the expression of virulence factors and biofilm formation genes, resulting in effective antibacterial action. Persistent biofilm infections pose a critical health threat with their relentless presence and amplified antibiotic resistance. Traditional antibacterial photodynamic therapy can inhibit bacteria extracellularly but struggles to control biofilm formation and virulence. Thus, there is an urgent need to develop photosensitizers, such as ultra‐small gold nanoclusters (AuNCs), that can penetrate biofilms and internalize into bacteria. However, AuNCs still face the challenge of insufficient reactive oxygen species (ROS) production and limited near‐infrared light absorption. This study develops a model of indocyanine green (ICG)‐sensitized AuNCs with atomic‐precision size effect. This approach achieved near‐infrared light absorption while inhibiting radiation transitions, thereby regulating the generation of ROS. Notably, different‐sized AuNCs (Au10NCs, Au15NCs, Au25NCs) yielded varied ROS types, resulting from different energy level distributions and electron transfer rates. ICG‐Au15NCs achieved a treatment efficacy of 99.94% against Staphylococcus aureus infections in vitro and significantly accelerated wound healing in vivo. Moreover, this study highlights the unique role of ICG‐AuNCs in suppressing quorum sensing, virulence, and ABC transporters compared to their larger counterparts. This strategy demonstrates that atomic‐precision size effect of AuNCs paves the way for innovative approaches in antibacterial photodynamic therapy for infection control.

Abstract:The aim of the described research is to destroy MRSA with the help of MFBA‐functionalized AuNRs using PTT. CTAB molecules used in the synthesis of AuNR (CTAB@AuNR) were removed from the surface due to their toxic activity and replaced with MFBA, which provides selectivity (MFBA@AunR). Next, MRSA viability was investigated using different concentrations of CTAB@AuNR and MFBA@AuNR, and finally the effect of both treatments in contact with MRSA bacteria was evaluated. Additionally, MTS toxicity testing of substances was performed. Methicillin‐resistant Staphylococcus aureus (MRSA) is one of the deadliest pathogenic bacteria. Using photothermal therapy (PTT) to eradicate MRSA bacteria easily and effectively, it has directed this bacterium to be destroyed. Gold nanorods (AuNRs), which are nanoparticles that provide PPT, were synthesized and removed from the CTAB molecule reduce the toxic effect caused by CTAB. Subsequently, mercaptophenylboronic acid (MFBA) coated AuNRs were synthesized and used in photothermal therapy to develop a targeting agent to selectively eliminate MRSA. A decrease in cytotoxic effect of CTAB@AuNRs after conjugation with MFBA was also demonstrated by the MTS cell viability test. We found that at the end of 48 hours and 72 hours of interaction, IC50 values of MFBA@ AuNR increased by approximately 50 % compared to CTAB@AuNR. Also, it shows that the cytotoxicity of AuNRs conjugated with MFBA was reduced. Herein, photothermal efficiency was achieved with MFBA@AuNRs targeting MRSA. The purpose of using and modifying gold nanorods is to reduce the toxicity of AuNRs and to examine their efficacy on resistant pathogenic bacteria strains by taking advantage of photothermal therapy properties.

AuNC@NO was prepared by immobilizing a temperature-responsive NO donor onto gold nanocages (AuNCs) through thiol-gold interactions. AuNC@NO possesses stable and excellent photothermal conversion efficiency, as well as the characteristics of slow NO release at physiological temperature and on-demand quick NO release under NIR irradiation. Based on these features, AuNC@NO exhibits enhanced in vitro bactericidal and antibiofilm efficacy compared with AuNCs, which could achieve 4 orders of magnitude bacterial reduction and 85.4% biofilm elimination under NIR irradiation. In addition, we constructed an implant biofilm infection model and a subcutaneous biofilm infection model to evaluate the anti-infective effect of AuNC@NO. The in vivo results indicated that after 5 min of 0.5 W cm-2 NIR irradiation, NO release from AuNC@NO was significantly accelerated, which induced the dispersal of methicillin-resistant Staphylococcus aureus (MRSA) biofilms and synergized with photothermal therapy (PTT) to kill planktonic MRSA that had lost its biofilm protection. Meanwhile, the surrounding tissues showed little damage because of controlled photothermal temperature and toxicity. In view of the above-mentioned results, the AuNC@NO nanocomposite developed in this work reveals potential application prospects as a useful antibiofilm agent in the field of biofilm-associated infection treatment.

Abstract:A novel antibacterial strategy utilizes nanostructured "light traps" mediated PTT to enhance the physical puncture of bacteria by nanorod arrays, effectively eliminating biofilm infections without requiring additional photosensitivities. Combined with nitric oxide therapy, this method effectively prevents MRSA infection on the surface of titanium implants, providing a promising approach for sustainable antibacterial implant surfaces. The physical killing of bacteria based on surface topography has attracted much attention due to the sustainable and safe prevention of biofilm formation. However, the antibacterial efficiency of biomedical implants derived solely from nanostructures or microstructures is insufficient to combat bacteria against common infections, such as methicillin‐resistant Staphylococcus aureus with thick cell walls. Herein, photothermal therapy is carried out in the presence of nanorod arrays to mitigate infection of biomedical implants. Different from traditional photothermal therapy relying on a photosensitizer, the photothermal effect is mediated by light traps rendered by the nanorod arrays, and consequently, the photosensitizer is not needed. Finite element simulations and experiments are performed to elucidate the light‐to‐thermal conversion mechanism. This photothermal platform, in conjunction with thermosensitive nitric oxide therapy, is applied to treat titanium implant infection. The nanostructure‐mediated photothermal effect destroys bacterial cell walls by inhibiting peptidoglycan synthesis and increasing the membrane permeability by affecting fatty acid synthesis. Furthermore, the nanorods synergistically puncture the bacterial membrane easily as demonstrated by experiments and transcriptome analysis. The results provide insights into the development of efficient antibacterial treatment of implants by combining nanostructures and photothermal therapy.

Abstract:Wound treatment is largely influenced by pre-existing hypoxic microenvironments and biofilms, which can severely diminish the efficacy of phototherapy, suggesting the importance of multifunctional nanoplatforms for synergistic treatment of wound infections. Here, we developed a multifunctional injectable hydrogel (PSPG hydrogel) by loading photothermal sensitive sodium nitroprusside (SNP) into Pt-modified porphyrin metal organic framework (PCN) and in situ modification of gold particles to form a near-infrared (NIR) light-triggered all-in-one phototherapeutic nanoplatform. The Pt-modified nanoplatform exhibits a remarkable catalase-like behavior and promotes the continuous decomposition of endogenous H 2 O 2 into O 2 , thereby enhancing the photodynamic therapy (PDT) effect under hypoxia. Under dual NIR irradiation, PSPG hydrogel can not only produce hyperthermia (η=89.21%) but also generate reactive oxygen species and trigger NO release, contributing jointly to removal of biofilms and disruption of the cell membranes of methicillin-resistant Staphylococcus aureus (MRSA) and Escherichia coli ( E. coli ). In vivo experiments demonstrated a 99.9% reduction in bacterial burden on wounds. Additionally, PSPG hydrogel can accelerate MRSA-infected and P. aeruginosa -infected wound healing by promoting angiogenesis, collagen deposition, and suppressing inflammatory responses. Furthermore, in vitro and in vivo experiments revealed that PSPG hydrogel has good cytocompatibility. Overall, we proposed an antimicrobial strategy to eliminate bacteria through the synergistic effects of gas-photodynamic-photothermal killing, alleviating hypoxia in the bacterial infection microenvironment, and inhibiting biofilms, offering a new way against antimicrobial resistance and biofilm-associated infections. Statement of significance The NIR light-triggered multifunctional injectable hydrogel nanoplatform (PSPG hydrogel) based on Pt-decorated gold nanoparticles with sodium nitroprusside (SNP)-loading porphyrin metal organic framework (PCN) as inner templates can efficiently perform photothermal conversion (η=89.21%) to trigger NO release from SNP, while continuously regulating the hypoxic microenvironment at the bacterial infection site through Pt-induced self-oxygenation, achieving efficient sterilization and removal of biofilm by synergistic PDT and PTT phototherapy. In vivo and in vitro experiments demonstrated that the PSPG hydrogel has significant anti-biofilm, antibacterial, and inflammatory regulatory functions. This study proposed an antimicrobial strategy to eliminate bacteria through the synergistic effects of gas-photodynamic-photothermal killing, alleviating hypoxia in the bacterial infection microenvironment, and inhibiting biofilms.

Abstract:Bacterial infection is a major threat to human health. However, many antibacterial agents currently used are severely limited due to drug-resistance, and the development of side effects. Herein, we have developed a non-antibiotic nanocomposite consisting of chitosan (ChS) coated silver nanoparticles (AgNPs) and graphene nanoribbon (GNR)-based nanowires for light-triggered eradication of bacteria. The presence of AgNP/ChS significantly enhanced the interactions of the GNR nanowires with Pseudomonas aeruginosa , a clinically common Gram-negative bacterium. Which enables the highly effective photothermal eradication of bacteria by GNR upon near-infrared light irradiation. The nanocomposite was shown to be applicable for the light-triggered eradication of bacterial biofilms and the inhibition of bacterial growth on medical patches used for abdominal-wall hernia surgery.

Abstract:Bacterial infections caused by antibiotic-resistant pathogens have become intractable problems to public health. Therefore, there is an imperious demand for developing new approaches to effectively kill antibiotic-resistant bacteria. In this work, we report a kind of bacteria-targeted polydopamine nanoparticle exhibiting great photothermal killing ability toward methicillin-resistant Staphylococcus aureus (MRSA) by nano-localized hyperpyrexia under low-power near-infrared (NIR) light irradiation. These bacteria-targeted nanoparticles (PDA-PEG-Van) are prepared by modifying polydopamine nanoparticles with thiol-poly(ethylene glycol) (mPEG-SH) and vancomycin (Van) molecules. The PEG shell endows the nanoparticles with excellent long-term circulation stability. Due to the multivalent hydrogen-bond interactions between vancomycin and the MRSA cell wall, the vancomycin-modified polydopamine nanoparticles can specifically target MRSA rather than mammalian cells. These bacteria-targeted nanoparticles are employed as a nano-localized heat source to kill MRSA via disrupting the bacterial cell wall and membrane under irradiation of low-power NIR light. More importantly, the surrounding healthy tissues suffer bare damage, owing to the absence of any targeting effect of PDA-PEG-Van toward mammalian cells and the low power of NIR light used in the therapeutic process. Given the above advantages, the bacteria-targeted polydopamine nanoparticles proposed in this work show tremendous potential to treat MRSA infections, because they can effectively limit localized heating in the infection sites to kill bacteria and cut down damage to healthy tissues.

Abstract:Recently, nitric oxide (NO) has received increasing interest in combat against bacteria-induced infections because of its ability to sensitize and enhance the antibacterial effectiveness of many therapeutic approaches such as antibiotics. However, high-efficient loading and controlled release of NO remain a big challenge. In the present work, a type of gold nanostar/hollow polydopamine Janus nanostructure (GNS/HPDA JNPs) with precise near infrared (NIR)-controlled NO release property was fabricated using a facile seed-mediated method. Upon NIR laser irradiation, the NO-releasing GNS/HPDA JNPs (GNS/HPDA-BNN6) exhibited a synergistic photothermal and NO antibacterial effect by significantly inhibiting the growth and biofilm formation of both Gram-negative and Gram-positive bacterial strains, including methicillin-resistant Staphylococcus aureus (MRSA). An in-depth mechanism study revealed that two pathways were mainly involved in the synergistic photothermal and NO antibacterial effect. In one pathway, the synergistic effect severely destroyed the bacterial membrane by causing leakage of intracellular components such as DNA. In another pathway, the synergistic effect largely disturbed bacterial metabolism by regulating relative metabolic genes, followed by enhancing ROS generation to cause intracellular GSH depletion and DNA damage. More importantly, the synergistic effect significantly diminished the drug resistance of MRSA by downregulating the expression of the drug-resistant gene mecA and some relative multidrug efflux pumps (e.g., SepA and Tet38). An in vivo evaluation using a rat model with MRSA-infected wounds indicated that the synergistic photothermal and NO effect of GNS/HPDA-BNN6 can effectively eliminate MRSA from wounds, thereby alleviating inflammation and promoting wound healing. STATEMENT OF SIGNIFICANCE: Multidrug-resistant (MDR) bacteria have become a big threat to mankind, and therefore, the development of innovative antibacterial agents with high antibacterial efficiency is urgently required. Nanomaterial-mediated nitric oxide (NO) therapy is a promising strategy to effectively combat MDR bacteria through a synergistic antibacterial effect. Here, a gold nanostar/hollow polydopamine Janus nanostructure with precise near infrared (NIR) light-controlled NO release property (GNS/HPDA-BNN6) was developed. Both in vitro and in vivo evaluations demonstrated that GNS/HPDA-BNN6 could effectively eliminate methicillin-resistant Staphylococcus aureus (MRSA) from infected wounds and promote wound healing through a synergistic photothermal and NO therapeutic effect. Remarkably, the synergistic effect significantly diminished the drug resistance of MRSA by downregulating the expression of some drug-resistant genes and multidrug efflux pumps.

Abstract:The ever-increasing threats of multidrug-resistant bacteria and their biofilm-associated infections have bred a desperate demand for alternative remedies to combat them. Near-infrared (NIR)-absorbing photothermal agent (PTAs)-mediated photothermal therapy (PTT) is particularly attractive for biofilm ablation thanks to its superiorities of noninvasive intervention, satisfactory antibacterial efficiency, and less likelihood to develop resistance. Herein, three butterfly-shaped aggregation-induced emission luminogens (AIEgens) with balanced nonradiative decay (for conducting PTT) and radiative decay (for supplying fluorescence in the NIR-II optical window) are rationally designed for imaging-assisted photothermal obliteration of bacterial biofilms. After being encapsulated into cationic liposomes, AIEgens-fabricated nanoparticles can eradicate a wide spectrum of biofilms formed by Gram-positive bacteria (methicillin-resistant Staphylococcus aureus and Enterococcus faecalis) and Gram-negative bacteria (Escherichia coli and Pseudomonas aeruginosa) upon an 808 nm laser irradiation. In vivo experiments firmly demonstrate that the NIR-II AIE liposomes with excellent biocompatibility perform well in both the P. aeruginosa biofilm-induced keratitis mouse model and the MSRA biofilm-induced skin infection mouse model.

Abstract:Biofilms are structured communities of microbial cells embedded in a self-produced matrix of extracellular polymeric substances. Biofilms are associated with many health issues in humans, including chronic wound infections and tooth decay. Current antimicrobials are often incapable of disrupting the polymeric biofilm matrix and reaching the bacteria within. Alternative approaches are needed. Here, we describe a unique structure of dextran coated gold in a gold cage nanoparticle that enables photoacoustic and photothermal properties for biofilm detection and treatment. Activation of these nanoparticles with a near infrared laser can selectively detect and kill biofilm bacteria with precise spatial control and in a short timeframe. We observe a strong biocidal effect against both Streptococcus mutans and Staphylococcus aureus biofilms in mouse models of oral plaque and wound infections respectively. These effects were over 100 times greater than that seen with chlorhexidine, a conventional antimicrobial agent. Moreover, this approach did not adversely affect surrounding tissues. We conclude that photothermal ablation using theranostic nanoparticles is a rapid, precise, and non-toxic method to detect and treat biofilm-associated infections.
