Abstract:Changes in the microbial community are considered important factors affecting the yield and quality of ginseng cultivated in farmland, while the characteristics of the change in the microbial community of the soil before and after cultivation of ginseng in farmland are not well understood. Here, we aimed to fill this research gap by comparing the differences in soil nutrients and microbial communities between continuously cultivated ginseng soils for 1-, 2-, 3- and 5-year-old ginseng soils and corresponding uncultivated ginseng soils. The soil properties (pH, total soil organic carbon (TOC) content and total nitrogen (TN) content) of aged ginseng soil (3- and 5-year-old) were significantly reduced compared with those of the control soil after ginseng was cultivated in farmland. With the exception of 2-year-old ginseng, soil bacterial richness and diversity were higher in different ages of cultivated ginseng soils than in the control soil, while fungal diversity decreased in aged ginseng soil and increased in young ginseng soil (1- and 2-year-old). Ascomycota and Basidiomycota showed a consistent increasing/decreasing trend in cultivated ginseng soils of all ages compared to the control. In addition, ginseng cultivation resulted in a substantial decrease in the complexity of soil bacterial networks but a slight decrease in the complexity of fungal networks. The results from the linear discriminant size effect (LEfSe) analysis showed that beneficial microorganisms were biomarkers of the young ginseng soil, including Nitrosospira , Cephalotrichum and Humicola , while harmful microorganisms were biomarkers of aged ginseng, such as Plectosphaerella , Cladosporium and Alternaria . PICRUSt2 analysis revealed that the increase in the incidence of ginseng disease with continuous cultivation time may be related to the decreases in amino acid synthesis and physiological metabolism and the increase in fungal saprophytes. Soil pH was the most common and best predictor of bacterial and fungal community structure, followed by TOC and ammonium nitrogen (NH 4 + -N) contents. Meanwhile, the decreased of soil pH, TOC and AK may be the driving factors for the growth of ginseng soil pathogens. Overall, this study revealed the changing ginseng rhizosphere soil microbial community at different continuous cultivation times, provided a theoretical basis for ginseng cultivation, and promoted the development of medicinal plants such as ginseng.

Abstract:Introduction Microbial communities are crucial for plant health and productivity. However, the influence of cultivation age on the ecological processes in assembling plant microbiomes at various ecological niches remains unclear. Methods We selected 12 samples from ginseng farmlands with different cultivation years (N4: 4 years old, N6: 6 years old). We used soil physicochemical properties, enzyme activities, and high-throughput sequencing (16S rDNA and ITS) to examine the rhizoplane (RP), rhizosphere (RS), and bulk soil (BS). Results Our results indicated that cultivation years significantly affect the soil microbiome’s diversity and community composition across different ecological niches. The BS microbiome experienced the largest effect, while the RS experienced the smallest. N6 showed a greater impact than N4. This effect was more pronounced on the fungal communities than the bacterial communities of various ecological niches and can be closely related to the soil’s physicochemical properties. In N4 soils, we observed an upward trend in both the number of ASVs (amplicon sequence variations) and the diversity of soil microbial taxa across various ecological niches. In N4RP, the bacteria Sphingomonas , known for degrading toxic soil compounds, was present. All ecological niches in N4 showed significant enrichment of Tetracladium fungi, positively associated with crop yield (N4RP at 6.41%, N4RS at 11.31%, and N4BS at 3.45%). In N6 soils, we noted a stark decline in fungal diversity within the BS, with a 57.5% reduction in ASVs. Moreover, Sphingomonas was abundantly present in N6RS and N6BS soils. The relative abundance of the pathogen-inhibiting fungus Exophiala in N6RP and N6RS reached 34.18% and 13.71%, respectively, marking increases of 4.9-fold and 7.7-fold. Additionally, another pathogeninhibiting fungus, Humicola , showed significant enrichment in N6BS, with a 7.5-fold increase. The phenolic acid-producing fungus Pseudogymnoascus in N6RP, N6RS, and N6BS showed increases of 2.41-fold, 2.55-fold, and 4.32-fold, respectively. We hypothesize that functional genes related to the metabolism of terpenoids and polyketides, as well as signaling molecules and interactions, regulate soil microbial taxa in ginseng from different cultivation years. Discussion In conclusion, our study enhances understanding of plant-microbe interactions and aids the sustainable development of medicinal plants, particularly by addressing ginseng succession disorder.

Correlation of cultivation time of Panax ginseng with metabolic profiles of nine ginsenosides and mRNA expression of genes encoding major biosynthetic enzymes

Abstract:Ginseng (Panax ginseng C. A. Meyer) is one of the most important Chinese medicinal herbs. This medicinal plant produces numerous bioactive substances, including triterpene saponins of the ginsenoside-type. The endogenous levels of all types of ginsenosides largely determine the therapeutic quality of P. ginseng. Here, a field investigation was conducted to compare the changes in ginsenoside composition and mRNA expression levels of some key genes in the biosynthetic pathway of P. ginseng seedlings cultivated for different times (1, 2, 3 or 4 years). After establishing a high performance liquid chromatography method to simultaneously determine nine different ginsenosides, we measured the variations of ginsenoside contents in P. ginseng seedlings collected from our field base in Jilin province, China. The levels of total ginsenosides obviously increased with longer cultivation times and reached peak values in the 4-year seedlings. In light of the changes of ginsenosides composition, protopanaxatriol (PPT)-type ginsenosides displayed a synchronized change with total ones, while the peak point of protopanaxadiol (PPD)-type ginsenoside contents appeared in the 3-year plants. Transcript analyses of squalene synthase (SS), squalene epoxidase (SQE1) and dammarenediol synthase (DS), which are the rate-limiting enzymes in the ginsenoside biosynthetic pathway, showed that the gene expression levels of PgSS and PgSQE1 were highest in the 2-year seedlings, and could indirectly control saponins. While the PgDS gene was highly expressed in 4-year plants, and another important gene pleiotropic drug resistance (PgPDR) has a consistent trend. The further correlation analysis between gene expressions and metabolic changes showed that PgDS transcript level significantly correlated with PPT-type ginsenosides, Rg1, Re, and Rf. PgPDR expression levels positively correlated with total ginsenoside, Rg1, Re, and Rb1 (p < 0.01) and Rg3 negatively correlated with PgDS and PgPDR genes expression (p < 0.01). However, the PgSS gene expression was negatively correlated with Rb1 (p < 0.05). In conclusion, we propose that PPT-type ginsenoside and PPD or dammarenediol-II compounds have a close relationship. Particularly, we highlight recent reports on functional characterization of key genes dedicated to the production of ginsenosides in P. ginseng seedling with cultivated time.

Abstract:Soil is the material basis of ginseng survival, and revealing the correlation between soil and ginsenoside has far-reaching significance for the sustainable development of ginseng industry. In this study, the content of ginsenosides in 3-year-old ginseng roots, the physicochemical properties of rhizosphere soil and the microbial community composition were studied. The results showed that the contents of total saponins in different months were significantly different. The richness and diversity of soil microbial community decreased with the extension of planting time. The activities of complexed iron, organic matter and protease in soil had significant effects on α-diversity of soil microbial community. Functional gene analysis showed that carbon sequestration, protein translation, nitrogen metabolism, transcription factors and chlorophyll metabolism were the main functions of soil bacterial community. The results of correlation analysis and redundancy analysis showed that pH, available potassium, organic matter, complexed iron, Firmicutes and Acidobacteria were the key factors affecting ginsenoside accumulation. The changes of soil physical and chemical properties affect the abundance of soil microorganism, and the accumulation of ginsenoside in ginseng is affected by soil microorganism. A co-regulatory network of Physicochemical Properties-Microbe-Ginsenoside was established. To provide theoretical support for the cultivation of ginseng.

Abstract:Panax ginseng C.A. Meyer is one of the most important medicinal plants in Northeast China, and ginsenosides are the main active ingredients found in medicinal ginseng. The biosynthesis of ginsenosides is regulated by environmental factors and the expression of key enzyme genes. Therefore, in this experiment, ginseng in the leaf opened stage, the green fruit stage, the red fruit stage, and the root growth stage was used as the test material, and nine individual ginsenosides and total saponins (the sum of the individual saponins) were detected by HPLC (High Performance Liquid Chromatography). There was a trend of synergistic increase and decrease, and saponin accumulation and transfer in different tissues. The expression of key enzyme genes in nine synthetic pathways was detected by real-time PCR, and the correlation between saponin content, gene expression, and ecological factors was analyzed. Correlation analysis showed that in root tissue, PAR (Photosynthetically Active Radiation) and soil water potential had a greater impact on ginsenoside accumulation, while in leaf tissue, temperature and relative humidity had a greater impact on ginsenoside accumulation. The results provide a theoretical basis for elucidating the relationship between ecological factors and genetic factors and their impact on the quality of medicinal materials. The results also have guiding significance for realizing the quality of medicinal materials.

Abstract:Abstract Background Continuous cropping of ginseng ( Panax ginseng Meyer) cultivated in farmland for an extended period gives rise to soil-borne disease. The change in soil microbial composition is a major cause of soil-borne diseases and an obstacle to continuous cropping. The impact of cultivation modes and ages on the diversity and composition of the P. ginseng rhizosphere microbial community and technology suitable for cropping P. ginseng in farmland are still being explored. Methods Amplicon sequencing of bacterial 16S rRNA genes and fungal ITS regions were analyzed for microbial community composition and diversity. Results The obtained sequencing data were reasonable for estimating soil microbial diversity. We observed significant variations in richness, diversity, and relative abundances of microbial taxa between farmland, deforestation field, and different cultivation years. The bacterial communities of LCK (forest soil where P. ginseng was not grown) had a much higher richness and diversity than those in NCK (farmland soil where P. ginseng was not grown). The increase in cultivation years of P. ginseng in farmland and deforestation field significantly changed the diversity of soil microbial communities. In addition, the accumulation of P. ginseng soil-borne pathogens ( Monographella cucumerina , Ilyonectria mors - panacis , I. robusta , Fusarium solani , and Nectria ramulariae ) varied with the cropping age of P. ginseng . Conclusion Soil microbial diversity and function were significantly poorer in farmland than in the deforestation field and were affected by P. ginseng planting years. The abundance of common soil-borne pathogens of P. ginseng increased with the cultivation age and led to an imbalance in the microbial community.

Abstract:American ginseng (Panax quinquefolium L.) is a perennial herbaceous plant widely cultivated in China, Korea, the United States, and Japan due to its multifunctional properties. In northwest China, transplanting after 2-3 years has become the main mode of artificial cultivation of American ginseng. However, the effects of the cultivation process on the chemical properties of the soil and bacterial community remain poorly understood. Hence, in the present study, high-throughput sequencing and soil chemical analyses were applied to investigate the differences between bacterial communities and nutrition driver factors in the soil during the cultivation of American ginseng. The responses of soil nutrition in different ecological niches were also determined with the results indicating that the cultivation of American ginseng significantly increased the soluble nutrients in the soil. Moreover, the bacterial diversity fluctuated with cultivation years, and 4-year-old ginseng roots had low bacterial diversity and evenness. In the first two years of cultivation, the bacterial community was more sensitive to soil nutrition compared to the last two years. Proteobacteria, Actinobacteria, Gemmatimonadetes, Acidobacteria, Firmicutes, and Bacteroidetes dominated the bacterial community regardless of the cultivation year and ecological niche. With the increase of cultivation years, the assembly of bacterial communities changed from stochastic to deterministic processes. The high abundance of Sphingobium, Novosphingobium, and Rhizorhabdus enriched in 4-years-old ginseng roots was mainly associated with variations in the available potassium (AK), total phosphorus (TP), total potassium (TK), and organic matter (OM).

Abstract:Panax ginseng C. A. Mey (PGCAM) is a herbaceous perennial belonging to the Araliaceae family, mainly including Mountain-Cultivated Ginseng (MCG) and Garden Ginseng (GG) on the market. We aimed to establish a rapid, accurate and effective method to distinguish 15-year-old MCG and GG using ultra-performance liquid chromatography-quadrupole time-of-flight-tandem mass spectrometry (UPLC-QTOF-MS/MS), and also explored the pharmacological mechanisms of the main components using the Integrative Pharmacology-based Network Computational Research Platform of Traditional Chinese Medicine (TCMIP V2.0; http://www.tcmip.cn/ ). Altogether, 23 potential quality markers were characterized to distinguish 15-year-old MCG and GG, including ginsenosides Ra2, Rg1, and Ra1, and malonyl-ginsenoside Ra3, etc. The contents of 19 constituents (mainly protopanaxadiol-type) were higher in MCG compared with that in GG, and four constituents (mainly carbohydrate compounds) were higher in GG. The 105 putative targets corresponding to 23 potential quality markers were mainly involved in 30 pathways, which could be divided into 10 models, such as immune regulation, systems (metabolic, nervous, cardiovascular, reproductive), blood-pressure regulation, as well as antitumor, antiaging, antibacterial and anti-inflammatory effects. Furthermore, the potential quality markers of MCG and GG could inhibit the proliferation of breast cancer by regulating the mRNA expression of PSA, S6K, MDM2, and P53 genes by acting on AR, MTOR, PI3K and other targets. The Integrative Pharmacology Strategy may provide an efficient way to identify chemical constituents and explore the pharmacological actions of TCM formulations.

Correlations among Soil Properties, Growth Characteristics, and Ginsenoside Contents in Wild-Simulated Ginseng with Different Ages

Abstract:Wild-simulated ginseng (WSG) is naturally cultivated in forest environment without any artificial facilities or chemicals. Soil property is one of the major factors affecting the growth and active compound synthesis of vegetation. Therefore, growth characteristics and ginsenoside contents of WSG can be affected by soil properties of the cultivation field. Therefore, the aim of this study was to investigate correlations among soil properties, growth characteristics, and ginsenoside contents of WSG with different ages using Spearman's coefficient analysis method. It was found that most of growth characteristics of WSG except for rhizome length were significantly increased in 7-year-old WSG for both the above-ground part and root part. Soil pH, and exchangeable cations (Ca, Mg) of 13-year-old WSG cultivation site were significantly higher than those of 4-year-old WSG. However, available phosphate of 4-year-old WSG soil was higher than that of 13-year old WSG soil. Contents of ginsenosides of 4-year-old WSG were higher than those of 13-year-old WSG in the above-ground part. Otherwise, in the root part, contents of ginsenosides of 13-year-old WSG were higher than those of 4-year-old WSG. In correlation analysis, growth characteristics of 4-year-old WSG were correlated with more ginsenoside types. Correlations between soil properties and ginsenoside contents in leaves and roots of WSG with the same age also differed. These results suggest that soil properties play essential role in growth and ginsenoside synthesis of WSG. Based on results of this study, growth characteristics, soil properties, and ginsenoside contents of WSG in different WSG cultivation sites need to be further investigated to identify the most suitable cultivation site for WSG.

Differences in the Quality, Yield, and Soil Microecology of Ginseng in Different Planting Environments

Abstract:The quality and yield of ginseng are significantly affected by ecological factors, especially soil, which is a complex environment. This study compared the quality, yield, and soil differences of ginseng in a Mongolian oak forest (MOFG), birch forest (BFG), mixed wood forest (MWFG), poplar forest (PFG), farmland (FlG), and a felling forest (DfG). Based on these differences, the relationship between the three was explored, as was the most suitable forest type for developing understory ginseng cultivation. The soil bacteria diversity of DfG was similar to that of understory ginseng, and its ginsenoside content was at the same level as that of understory ginseng. The soil pH of FlG was significantly lower than that of the other five cultivation methods, and the soil bacteria were also different from them, especially those related to the nitrogen cycle. Ginsenoside Rg2 has a significant correlation with many soil factors and is sensitive to soil environmental differences. In conclusion, there were significant differences in the quality, yield, and soil environments between the different planting methods. MOFG was the most suitable for planting with excellent quality and high yield, while BFG was the worst; as such, planting in BFG should be avoided.

Abstract:Chinese ginseng (Panax ginseng Meyer) is a medicinally important herb and plays crucial roles in traditional Chinese medicine. Pharmacological analyses identified diverse bioactive components from Chinese ginseng. However, basic biological attributes including domestication and selection of the ginseng plant remain under-investigated. Here, we presented a genome-wide view of the domestication and selection of cultivated ginseng based on the whole genome data. A total of 8,660 protein-coding genes were selected for genome-wide scanning of the 30 wild and cultivated ginseng accessions. In complement, the 45s rDNA, chloroplast and mitochondrial genomes were included to perform phylogenetic and population genetic analyses. The observed spatial genetic structure between northern cultivated ginseng (NCG) and southern cultivated ginseng (SCG) accessions suggested multiple independent origins of cultivated ginseng. Genome-wide scanning further demonstrated that NCG and SCG have undergone distinct selection pressures during the domestication process, with more genes identified in the NCG (97 genes) than in the SCG group (5 genes). Functional analyses revealed that these genes are involved in diverse pathways, including DNA methylation, lignin biosynthesis, and cell differentiation. These findings suggested that the SCG and NCG groups have distinct demographic histories. Candidate genes identified are useful for future molecular breeding of cultivated ginseng.

Abstract:Abstract Background Ginseng red skin root syndrome (GRS) is one of the most common ginseng ( Panax ginseng Meyer) diseases. It leads to a severe decline in P. ginseng quality and seriously affects the P. ginseng industry in China. However, as a root disease, the characteristics of the GRS rhizosphere microbiome are still unclear. Methods The amplicon bacterial 16 S rRNA genes and fungal ITS (Internal Transcribed Spacer) regions Illumina sequencing technology, combined with microbial diversity and composition analysis based on R software, was used to explore the relationship between soil ecological environment and GRS. Results There were significant differences in the diversity and richness of soil microorganisms between the rhizosphere with different degrees of disease, especially between healthy P. ginseng (HG) and heavily diseased groups. The variation characteristics of microbial abundance in different taxa levels were analyzed. The interaction network of rhizosphere microorganisms of P. ginseng under GRS background was established. We also found that different P. ginseng rhizosphere microbial communities have multiple changes in stability and complexity through the established interaction network. Microbes closely related to potential pathogenic fungi were also identified according to the interaction network, which provided clues for looking for biological control agents. Finally, the Distance-based redundancy analysis (dbRDA) results indicated that total phosphorus (TP), available potassium (AK), available phosphorus (AP), catalase (CAT), invertase (INV) are the key factors that influence the microbial communities. Moreover, the content of these key factors in the rhizosphere was negatively correlated with disease degrees. Conclusions In this study, we comprehensively analyzed the rhizosphere characteristics of P. ginseng with different levels of disease, and explored the interaction relationship among microorganisms. These results provide a basis for soil improvement and biological control of field-grown in the future.

Abstract:To explore the effect of ecological factors on the features of ginsenosides and amino acids of mountainous forest cultivated ginseng (MFCG) and distinguish it from garden ginseng (GG). The main active constituents of ginsenosides and primary metabolites-amino acids of MFCG were determined by the high-performance liquid chromatography with diode array detector (HPLC-DAD) technique; The climatic factors were obtained by Geographic Information System for Global Medicinal Plants (GMPGIS), and the topographic factors were recorded at the place of origin; Mineral elements in rhizosphere soils of MFCG were determined by inductively coupled plasma mass spectrometry (ICP-MS), inductively coupled plasma optical emission spectrometry (ICP-OES) and atomic absorption spectrometry and other rhizosphere soil factors, inclusive of pH and available N, P and K. The findings indicated that the warmer, relatively humid, and relatively shady environments are suitable for the accumulation of ginsenosides of MFCG, in addition, they were also influenced by its rhizosphere soil. Contrastingly, conditions of lower temperature, more precipitation, weaker light, and higher altitude are beneficial to the accumulation of amino acids; Additionally, mineral elements are conducive to the accumulation of amino acids, while soil acidification is unfavorable. Surprisingly, the present study found that the temperature could regulate ginsenosides and amino acids in an opposite direction, and the ecological environment has a greater effect on primary metabolites of amino acids than secondary metabolites of ginsenosides. Finally, the Ser, Arg, Gly, Thr, Pro, Cys, Ile, Leu, Phe, Tyr, and GABA in MFCG were lower than those in GG (p < 0.01); The dencichine (p < 0.05) and dencichine/GABA (p < 0.01) in MFCG were higher than those in GG. Among them, the average ratio of dencichine to GABA is about 6:1 in MFCG and 3:1 in GG in most instances. It is for the first time that the effects of ecological factors on the chemical composition of MFCG and its distinction from GG by amino acids were investigated.

Abstract:Background Ginseng ( Panax ginseng Mayer) is an important natural medicine. However, a long culture period and challenging quality control requirements limit its further use. Although artificial cultivation can yield a sustainable medicinal supply, research on the association between the transplantation and chaining of metabolic networks, especially the regulation of ginsenoside biosynthetic pathways, is limited. Methods Herein, we performed Liquid chromatography tandem mass spectrometry based metabolomic measurements to evaluate ginsenoside accumulation and categorise differentially abundant metabolites (DAMs). Transcriptome measurements using an Illumina Platform were then conducted to probe the landscape of genetic alterations in ginseng at various ages in transplantation mode. Using pathway data and crosstalk DAMs obtained by MapMan, we constructed a metabolic profile of transplantation Ginseng. Results Accumulation of active ingredients was not obvious during the first 4 years (in the field), but following transplantation, the ginsenoside content increased significantly from 6−8 years (in the wild). Glycerolipid metabolism and Glycerophospholipid metabolism were the most significant metabolic pathways, as Lipids and lipid-like molecule affected the yield of ginsenosides. Starch and sucrose were the most active metabolic pathways during transplantation Ginseng growth. Conclusion This study expands our understanding of metabolic network features and the accumulation of specific compounds during different growth stages of this perennial herbaceous plant when growing in transplantation mode. The findings provide a basis for selecting the optimal transplanting time.

Abstract:Abstract Background The resources of wild ginseng have been reducing sharply, and it is mainly dependent on artificial cultivation in China, Korea and Japan. Based on cultivation modes, cultivated ginseng include understory wild ginseng (the seeds or seedlings of cultivated ginseng were planted under the theropencedrymion without human intervention) and farmland cultivated ginseng (grown in farmland with human intervention). Cultivated ginseng, can only be planted on the same plot of land consecutively for several years owing to soilborne diseases, which is mainly because of the variation in the soil microbial community. In contrast, wild ginseng can grow for hundreds of years. However, the knowledge of rhizosphere microbe communities of the wild ginseng is limited. Result In the present study, the microbial communities in rhizosphere soils of the three types of ginseng were analyzed by high-throughput sequencing of 16 S rRNA for bacteria and internal transcribed spacer (ITS) region for fungi. In total, 4,381 bacterial operational taxonomic units (OTUs) and 2,679 fungal OTUs were identified in rhizosphere soils of the three types of ginseng. Among them, the shared bacterial OTUs was more than fungal OTUs by the three types of ginseng, revealing fungal communities were to be more affected than bacterial communities. In addition, the composition of rhizosphere microbial communities and bacterial diversity were similar between understory wild ginseng and wild ginseng. However, higher bacterial diversity and lower fungal diversity were found in rhizosphere soils of wild ginseng compared with farmland cultivated ginseng. Furthermore, the relative abundance of Chloroflexi, Fusarium and Alternaria were higher in farmland cultivated ginseng compared to wild ginseng and understory wild ginseng. Conclusions Our results showed that composition and diversity of rhizosphere microbial communities were significantly different in three types of ginseng. This study extended the knowledge pedigree of the microbial diversity populating rhizospheres, and provided insights into resolving the limiting bottleneck on the sustainable development of P. ginseng crops, and even the other crops of Panax .

Abstract:Heavy-metal pollution has been established to affect ginseng quality. However, this effect is still unknown in ginseng of different ages, emphasizing the need to investigate the effects of heavy metals in soils on ginseng growth. Herein, we determined the content of heavy metals (Cu, Cd, Pb, Hg, and As) in ginseng of different ages (2 to 6-year-old) and the corresponding soil samples. Then, the total ginsenosides content of ginseng and rate-limiting enzyme (HMGR, SQE, CYP450) activity in the synthesis of ginsenosides were assessed. Results from 200 differently-aged Chinese ginseng showed that increased ginsenoside content in 3 to 5-year-old ginseng was paralleled by increased heavy metal element content in ginseng and its soil. The activity of rate-limiting enzymes increased in the first four years of ginseng growth and then exhibited a steady or downward trend. Further analysis suggested that heavy metal elements in soils could directly affect ginsenoside content. Moreover, we found that Cu significantly affected the rate-limiting enzyme CYP450 activity. Further principal component analysis and correlation analysis found that heavy metals could obviously inhibit ginseng growth during the 5th and 6th years. Heavy metal content in soils has huge prospects for predicting ginsenoside, Cu and As content in ginseng. This study provided support for ginseng cultivation, quality research and quality assessment.

Abstract:This study aimed to further analyze the chemical characteristics of mountainous forest cultivated ginseng (MFCG) and garden ginseng (GG) concerning their ginsenosides and oligosaccharides. Under our research conditions, the results demonstrated that the growth year of MFCG less than 12 years: Rg1 < 0.40%, Re/Rg1 > 1.00 and Re/Rb1 > 0.50; The growth year of MFCG over 12 years: Rg1 ≥ 0.40%, Re/Rg1 < 1.00 and Re/Rb1 < 0.50 in most cases. In general, Rg1, Rf, Rb1, Rc, Rb2, Re and Rd, the sum of major ginsenosides, Rg1/Re, protopanaxadiol (PPD), protopanaxatriol (PPT) and PPD/PPT in commercially available MFCG were higher than those in GG (P < 0.05 or P<0.01); The proportion of ginsenosides (with Rb1(%) as the reference) in the entire ginseng and different parts of MFCG was lower than that of GG (P < 0.01). In most cases, the highest ginsenoside content in the rhizome of GG was Ro, the highest ratio of ginsenosides in the rhizome of GG was Ro/Rb1, however, the rhizome of MFCG does not have this characteristic. Thus, it is for the first time that the ratio of ginsenoside (Rb1 as the reference) was used to explore the chemical characteristics used to distinguish MFCG from GG. Surprisingly, the study found that the Re/Rb1, Rg1/Rb1 and Ro/Rb1 in the entire ginseng and different parts of MFCG were lower than that of GG, in particular, Re/Rb1<1/2 in entire MFCG and Re/Rb1>1/2 in entire GG in most cases. The Ro and Ro/Rb1 in the rhizome can be used to identify MFCG and GG in most cases. In addition, the differences in the content and proportion of ginsenosides in different parts of ginseng can provide a reference for the identification of different parts of ginseng.

Abstract:Ginseng has been widely applied in clinical practice, but the cultivation age cannot be ignored as it influences the quality of ginseng and its products. In this work, different cultivation ages of fresh ginseng (FG) from four to seven years were analysed by UPLC-Q-TOF-MS/MS. Principal component analysis and supervised orthogonal partial least squared discrimination analysis, which belong to the normal method of multivariate statistical analysis, were applied to discover the characteristic components of FG at different cultivation ages. The components of new type of red ginseng (NRG) derived from FG at different cultivation ages were compared by HPLC analysis. The pharmacological anti-inflammatory activity was evaluated by ELISA and qPCR. The result showed that the characteristic components of both 6- and 7-year-old ginseng were ginsenoside Rb1, mal-ginsenoside Rb1, ginsenoside Rc, mal-ginsenoside Rc, mal-ginsenoside Rb1 isomer, and mal-ginsenoside Rb2. Moreover, the characteristic components of both 4- and 5-year-old ginseng were ADP-glucose and 3-hydroxyhexanoyl CoA. In addition, 6-year-old NRG has higher rare ginsenosides than 4-year-old NRG, which possesses great anti-inflammatory activity in vitro. The results reveal the ginsenoside transformation law of NRG processing and suggest that the cultivation age of FG influences the content of ginsenosides in NRG. Therefore, 6-year-old ginseng is more suitable for red ginseng processing and clinical use.

Abstract:Panax ginseng is a traditional medicine. Fresh ginseng is one of the most important industries related to ginseng development, and fresh ginseng of varying ages has different medicinal properties. Previous research has not systematically reported the correlation between changes in key enzyme activity with changes in ginsenoside content in fresh ginseng over time. In this study, for the first time, we use ginseng samples of varying ages in Ji’an and systematically reported the changes in the activity of seven key enzymes (HMGR, FPS, SS, SE, DS, CYP450, and GT). We investigated the content of ginsenoside and gene expression of these key enzymes. Ginsenoside content was measured using HPLC. HPLC, GC-MS, and LC-MS were combined to measure the enzyme activity of the key enzymes. Quantitative PCR was used in the investigation of gene expression. By analyzing the correlation between the enzyme activity and the transcription level of the key enzymes with ginsenoside content, we found that DS and GT enzyme activities are significantly correlated with the ginsenoside content in different ages of ginseng. Our findings might provide a new strategy to discriminate between ginseng of different years. Meanwhile, this research provides important information for the in-depth study of ginsenoside biosynthesis.
Untargeted Metabolomics and Soil Community Metagenomics Analyses Combined with Machine Learning Evaluation Uncover Geographic Differences in Ginseng from Different Locations

Abstract:Panax ginseng C.A. Meyer, known as the "King of Herbs," has been used as a nutritional supplement for both food and medicine with the functions of relieving fatigue and improving immunity for thousands of years in China. In agricultural planting, soil environments of different geographical origins lead to obvious differences in the quality of ginseng, but the potential mechanism of the differences remains unclear. In this study, 20 key differential metabolites, including ginsenoside Rb1, glucose 6-phosphate, etc., were found in ginseng from 10 locations in China using an ultra-high performance liquid chromatography-quadrupole time-of-flight mass spectrometry (UHPLC-QTOF-MS)-untargeted metabolomics approach. The soil properties were analyzed and combined with metagenomics technology to explore the possible relationships among microbial elements in planting soil. Through Spearman correlation analysis, it was found that the top 10 microbial colonies with the highest abundance in the soil were significantly correlated with key metabolites. In addition, the relationship model established by the random forest algorithm and the quantitative relationship between soil microbial abundance and ginseng metabolites were successfully predicted. The XGboost model was used to determine 20(R)-ginseng Rg2 and 2'(R)-ginseng Rg3 as feature labeled metabolites, and the optimal ginseng production area was discovered. These results prove that the accumulation of metabolites in ginseng was influenced by microorganisms in the planting soil, which led to geographical differences in ginseng quality.
